Testo completoSyftet med följande arbete har varit att undersöka hur högläsningen fungerar i dagens skola genom observationer av hur läraren planerar inför högläsningen, genom bokvalet och läsplatsen. Undersökningen har även fokuserat på den interaktion som sker i klassrummet i samband med högläsningen samt hur läraren under högläsningen arbetar med bearbetningen av den lästa texten. Undersökningsmetoden har varit videoobservationer under 12 högläsningstillfällen i förskoleklass och årskurs 1. Valet av ålderskategori gjordes för att eleverna på dessa stadier ännu inte har fått flyt i sin egen läsning och då är högläsningen av läraren ett viktigt undervisningstillfälle.
Resultat i studien visar att interaktionen mellan lärare och elever skiljer sig åt från lärare till lärare men också mellan olika högläsningsstunder med samma lärare. Detta beror bland annat på valet av bok. Det förekom även skillnader mellan sätten att ställa frågor, där vissa frågor var så kallade öppna frågor som ledde till mer vardagliga samtal utifrån texten. Det var också vid dessa frågor som man kunde notera att flerstämmighet rådde i klassrummen. Det förekom även mer slutna frågor, oftast i samband med ordförståelseträning, och då gällde handuppräckning hos eleverna.

Therefore, the purpose of this essay is to examine prealgebran in math books for grades 1-3 where the basis must be laid before algebra. What does the research show? Are the teaching materials based on the research results? Do the teaching materials have what it takes for students to be algebraic? These questions are going to be answered in this essay. The books that are analyzed are common in Sweden and are: Favorite mathematics, Prima mathematics, Eldorado and the Math detectives. The result of this essay shows that none of the teaching materials have all the elements that the research show is important so the pupils can be algebraic.

Testo completoGlycerol is generated in large amounts during the production of biodiesel and it is becoming a potentially attractive susbstrate for microbial production of value-added products such as 1,3-propanediol (1,3-PD). This research aimed at studying the production of 1,3-PD using immobilized cells of K. pneumoniae BLh-1 on residual glycerol as the carbon source. First, we studied the effect of cell concentration and the diameter of the beads in the immobilization system. The results showed that the best combination is 100 mg biomass by mL of support and diameter of 3.40 mm, as presented in increased productivity. In the experiments to compare the production of 1,3-PD with immobilized cells and suspension cells the production of 1,3-PD was similar in both experiments. But the use of immobilized cells led to obtaining greater productivity 1,3-PD, due to the high cell density used, one of the advantages of using this technique. Experiments were conducted reuse of immobilized cells, to assess the stability of production of 1,3-PD for five batches, in the experiments with interval between batches to the production of 1,3-PD was more stable in relation the use of beads without interval between batches and at the end of the fifth batch of the calcium alginate beads were with its damaged structure. In experiments in bioreactors the production of 1,3-PD was approximately of 25 g L-1 in batch, continuous bioreactors reached steady state after 40 h of cultivation. The control of pH is essential in the production of 1,3-PD K. pneumoniae BLh-1, but this destabilizes the structure of the spheres of calcium alginate. Future studies will be necessary to seek new alternatives supports of the immobilization, for can control the pH of the experiments, because the immobilization cellular proved to be a viable alternative biotechnological production of 1,3-PD.

Testo completoReading comprehension is a central part of most school subjects. It is important for all pupils to be able to understand what they read, as it can enable them to develop knowledge within the school subjects (Eckeskog, 2015, p. 5-6). Although reading comprehension is of such importance in school, results from the Program for International Student Assessment (PISA) shows that Swedish pupils results in reading comprehension have fallen (Skolverket, 2013, p. 13). The purpose of this study is, therefore, to study how teachers in grades 1-3 describe that they plan and implement their read aloud teaching to develop the pupils’ reading comprehension. This will be answered by the following questions: How do teachers describe that they work with reading strategies in connection with the read aloud? How do the teachers, according to themselves, give the pupils the opportunity to work with the texts in the read aloud? And finally: What different types of texts do teachers focus on in their teaching in the development of the pupils’ reading comprehension? The study is based on the socioultural perspective. The data was collected through a qualitative method, semi structured interviews. The data was analyzed by coding to find interesting patterns in the participants descriptions. The results showed that the teachers use read aloud as a tool to develop the pupils’ reading comprehension. They read both fiction and non-fiction. Before, during and after the read aloud most teachers use some of the reading strategies: predict, clarify, summarize and questioning. According to the teachers, the pupils show their reading comprehension through text conversation, writing and working with pictures.

Testo completoSyftet med detta examensarbete var att undersöka hur lärare använder olika laborativa material i sin matematikundervisning i år 1-3 och hur detta överensstämmer med det sätt som upphovsmännen till materialen avser. Mina frågeställningar har bland annat varit: Vad är laborativa material?, Hur använder lärare laborativa material i matematikundervisningen i år 1-3?. För att söka svar på dessa frågor har jag genomfört en litteraturstudie, och en kvalitativ intervjustudie med åtta lärare som använder laborativa material i sin matematikundervisning.
Resultatet visar att det finns skillnader mellan hur lärare använder vissa laborativa material och hur materialens upphovsmän beskriver att materialen ska användas. Både resultatet från litteraturstudien och från intervjuundersökningen visar att det finns nackdelar med att använda laborativa material om materialen används på ett felaktigt sätt.

Testo completoWith global production exceeding 750 million tons in 2017, wheat is considered a staple food for the world's population. Wheat mapping and monitoring could then be a very effective tool for achieving the Sustainable Development Goals (SDG2-Zero Hunger). In Lebanon, wheat receives technical and financial support, yet many errors occur in estimating the wheat acreage due to absence of reliable agricultural census and lack of wheat mapping using satellite images. In addition, identifying the best rotation type and agricultural practices leads to identify the most efficient wheat-based cropping system in terms of productivity (protein production and net profit), efficiency (water and nitrogen use), as well as the economic risk on the farmer. Thus, The aim of the current study, which is conducted in the Bekaa plain of Lebanon, is to utilize remote sensing technology and crop modelling for supporting policy makers and end-users in making strategic decisions regarding one of the most food security-driving crop in the Mediterranean (i.e. winter wheat).The first part of the thesis evaluates the potential of optical data for early winter wheat mapping by allowing the transfer of knowledge from one year to another (2016 and 2017 in this study). For its high spatial and temporal resolutions, Sentinel-2 data are employed. Results show that when the developed approach was applied on Sentinel-2 time series of 2017 in using 2016 ground truth data, the overall accuracy reaches 87.0%, whereas, when implemented using 2017 ground truth data, the overall accuracy is 82.6% on 2016 data. The outputs are executed up to six weeks before harvest, as well as distinguishing winter wheat from similar cereals (barley and triticale).The second part of the thesis examines the ability of the SAR (Synthetic Aperture Radar) C-band data of the new radar satellite (Sentinel-1) regarding its ability to monitor winter wheat crop by identifying the economically important phenological phases that cannot be detected relying solely on NDVI derived from optical satellite Sentinel-2. Results show that VV polarization at incidence angle of 32°-34° is best for predicting heading, VH polarization at incidence angle of 43°-45° for predicting soft dough, and the ratio VV/VH at incidence angle of 32°-34° for predicting germination and harvesting.The third part of the thesis is dedicated to test, in contrasted biophysical and management conditions, the hypothesis that promoting wheat-fava bean rotation leads to a significantly better productivity and resources use efficiency, as well as, reducing economic risk than the promoted intensive wheat-wheat and wheat-potato rotations. The cropping simulation model “CropSyst” is used after being calibrated and validated by using experimental data for different wheat-based rotations combining different soil, climate and management options. The results show that there is no particular optimal scenario that can simultaneously ensure high productivity, reduce economic risk, and achieve high wheat- water- and nitrogen-use efficiency. However, the wheat-fava bean rotation cultivated with no wheat fertilization appears to be a better substitute to the wheat-wheat rotation in terms of protein production in both (low and high) Water Holding Capacity (WHC) soils (0.93 t/ha versus 0.8 t/ha in low WHC and 1.34 t/ha versus 1.17 t/ha in high WHC). This cropping system could achieve a higher net profit, showing high resource-use efficiency and good economic sustainability. Moreover, a very high profit could only be attained with the wheat-potato rotation (8640 US DOLL./ha and 12170 US DOLL./ha), yet with low input-efficiency and high economic risk

Testo completoMore than 20 million people are infected by Human T-cell Lymphotropic Virus type 1 (HTLV-1) worldwide and this will cause T-cell leukemia in 5% of them. Yet the molecular mechanisms that underlie the oncogenic potential of this virus remain largely unknown. Bovine Leukemia Virus (BLV) is closely related to HTLV1 and causes a very similar B-cell leukemia in cattle and sheep. As for HTLV1, the oncogenic mechanisms underlying BLV-induced leukemia remain poorly understood. In both diseases, leukemic cells harbor mainly one integrated provirus, yet the integration sites are very variable. As a consequence, it is generally assumed that the oncogenic effect of the provirus is largely mediated by the virally encoded Tax protein. Paradoxically, however, both HTLV1 and BLV proviruses are found to be epigenetically silenced in tumor cells. Thus Tax, as any other virally encoded protein, is not expressed in leukemic cells suggesting that other factors are involved in tumorigenesis. In this study we made three observations that might dramatically change the prevalent dogma of HTLV1 and BLV-induced leukemia. First, we demonstrated that the BLV provirus is not silent at all in tumor cells. A cluster of BLV-encoded microRNAs (miRNAs) is highly expressed, accounting for 40% of the miRNAs present in leukemic cells. This finding is the first description of retroviral-encoded miRNAs. BLV miRNAs are transcribed from five independent RNA Pol III units and are exceedingly conserved across BLV isolates (more than the protein coding genes), strongly supporting an essential yet still unknown function. Next we showed that – as HTLV1 – BLV strongly expresses antisense RNAs. High-throughput sequencing of RNA libraries (RNA-seq) from BLV associated tumors, as expected, showed no expression of viral mRNA from the 5’ LTR. However, it did reveal the presence of two novel non-coding antisense transcripts originating in the 3’ LTR of BLV. Finally, RNA-Seq analysis of HTLV-1 and BLV-induced tumors revealed that the viral 3’ LTR-driven antisense RNAs produced by both viruses interact with host genes localized in the vicinity of proviral integration. Enrichment analysis of affected host genes suggests a significant bias towards cancer-related genes. Host gene perturbations were also found at early stages post-infection in the BLV experimental model in sheep, suggesting that provirus-dependent cancer driver gene perturbations trigger initial amplification of the corresponding clones, requiring additional genetic and/or epigenetic changes to develop full blown leukemia. Overall, our findings reveal an unexpected role for BLV and HTLV antisense transcripts and contribute to the understanding of non-coding RNA-mediated mechanisms in leukemogenesis.

Exposure to indoor allergens has been shown to be associated with the occurrence of asthma. However, studies assessing multi-allergens simultaneously have not yet been adequate. This study examined a group of common indoor allergens at homes of asthmatic and healthy children in Taiwan to construct the pattern of their concentration distributions and for a comprehensive assessment of microbial exposure and their effects on respiratory outcomes of asthmatics.A representative group of asthmatic children (11-13 years old) from a typical metropolitan city of Taiwan was selected with a cross-sectional questionnaire survey. The healthy controls were matched by their age, gender, schools attending, and residence. Dust samples of the mattress were collected with standard vacuum (3 min/m2). After sieving and extract on, all samples were analyzed by different ELISA kits (Lymphocyte Culture Center, the University of Virginia) for various allergens. Health outcomes werMultivariate regression showed that the standard deviation of PFM values measured in the morning, afternoon, evening, and daily average of the 7 days and symptom scores of the subjects study were all positively associated with the Der p 2 concentration of the mattress. The degree of effect on respiratory health by exposure to Der p 2 allergens appeared to be stronger on the asthmatics than healthy children . The positive association between mite allergen concentrations and 7-day standard deviation of PFM vaThe study was the first investigation ever conducted in the subtropical region to evaluate the domestic concentrations of multi- allergens simultaneously with respiratory health outcomes of randomly-selected asthmatic and health children. EBNA 1 gene of Epstein-Barr Virus (EBV) is transcribed from a latent promoter Qp in Burkitt’s lymphoma cell line, Rael, which is different from the Cp that is pre-ferentially activated in lymphoblastoid cell lines. In order to understand the mechanism that modulates the Qp activity, Qp and its deletion or site-specific mutant constructs were transfected into Rael cell and promoter activity was determined. Results showed that the region between —125 to —50 was not important for the minimal promoter activity of Qp. On the other hand, deletion to —37 reduced the promoter activity significantly. This region includes a binding motif recognized by Smad 4, a mediator involved in the TGF- signaling pathway. The binding was also proved by EMSA. Therefore, a site-specific mutation was made within this region and was then tested for its promoter activity. Data indicated the promoter activity was reduced to a basal level, and slightly higher then the deletion (-37) mutant. To analyze the effect of TGF- on Qp, promoter construct containing —50 to +20(pGL2B-50)and its Smad4 binding site mutant, pGL2B-50m4 were co-transfected with Smad3 and Smad4-expressing vector into Rael cells. The promoter strength was measured by the luciferase activity. Results showed that the minimal Qp(-50 to +20)activity was down-regulated upon TGF- stimulation through the Smad4 binding site. The mechanisms involved in this transcriptional repression and its biological significance were discussed.

Our previous study has known that Gastrodia elata Bl. (GE) can decrease epileptic seizures and oxygen free radicals in Kainic acid (KA)-induced epileptic seizures Sprague-Dawley (SD) rats, suggesting antiepileptic mechanisms of GE results from the suppressive or scavenging effect of free radicals partly. In addition, GE also can inhibit proliferation and activation of microglia, and apoptosis in the rat brain of KA-treated rats. Several studies finding that activator protein-1 (AP-1) involves neuronal apoptosis and plays a regulatory role in long-term neuronal plasticity. Therefore, the aim of the present study is to investigate the role of AP-1 in antiepileptic mechanism of GE. A total of 15 male SD rats were studied, they were treated with oral administration of PBS solution, GE 1.0g/kg, GE 0.5 g/kg, and valproic acid (VA) 250 mg/kg for one week, respectively, prior to KA 12 mg/kg intra-peritoneal injection. Electroencephalogram (EEG) and electromyogram (EMG) recordings, and behavior were observed for 3 hrs after KA administration, then the rats brain were removed, and cerebral cortex and hippocampus regions of the brain tissue were separated, respectively. Finally, the activating pathways of AP-1, including extracellular signal regulated kinase (ERK), Jun-N-terminal kinase (JNK) and p38 kinase were analyzed by Western blotting. The results indicated that Both GE and VA can decrease wet dog shakes, and AP-1 was activated via mediated a JNK-phosphate (JNK-P) pathway was observed in the frontal cortex and hippocampus region. In conclusion, GE can decrease epileptic seizures in KA-treated rats, but enhanced AP-1 activity, suggesting that GE possibly accelerated generation of AP-1 to induce a negative feedback for self-protection. In addition, GE is via mediated JNK-P pathway to activate AP-1. (GE) can decrease epileptic seizures and oxygen free radicals in Kainic acid (KA)-induced epileptic seizures Sprague-Dawley (SD) rats, suggesting antiepileptic mechanisms of GE results from the suppressive or scavenging effect of free radicals partly. In addition, GE also can inhibit proliferation and activation of microglia, and apoptosis in the rat brain of KA-treated rats. Several studies finding that activator protein-1 (AP-1) involves neuronal apoptosis and plays a regulatory role in long-term neuronal plasticity. Therefore, the aim of the present study is to investigate the role of AP-1 in antiepileptic mechanism of GE. A total of 15 male SD rats were studied, they were treated with oral administration of PBS solution, GE 1.0g/kg, GE 0.5 g/kg, and valproic acid (VA) 250 mg/kg for one week, respectively, prior to KA 12 mg/kg intra-peritoneal injection. Electroencephalogram (EEG) and electromyogram (EMG) recordings, and behavior were observed for 3 hrs after KA administration, then the rats brain were removed, and cerebral cortex and hippocampus regions of the brain tissue were separated, respectively. Finally, the activating pathways of AP-1, including extracellular signal regulated kinase (ERK), Jun-N-terminal kinase (JNK) and p38 kinase were analyzed by Western blotting. The results indicated that Both GE and VA can decrease wet dog shakes, and AP-1 was activated via mediated a JNK-phosphate (JNK-P) pathway was observed in the frontal cortex and hippocampus region. In conclusion, GE can decrease epileptic seizures in KA-treated rats, but enhanced AP-1 activity, suggesting that GE possibly accelerated generation of AP-1 to induce a negative feedback for self-protection. In addition, GE is via mediated JNK-P pathway to activate AP-1.
