The gene cassettes found in class 1 integrons are generally promoterless units composed by an open reading frame (ORF), a short 5’ untranslated region (UTR) and a 3’ recombination site (attC). Fused gene cassettes are generated by partial or total loss of the attC from the first cassette in an array, creating a fusion with the ORF from the next cassette. These structures are rare and little is known about their mechanisms of mobilization and expression. The aim of this study was to evaluate the dynamic of mobilization and transcription of the gcu14-blaGES-1/aacA4 gene cassette array, which harbours a fused gene cassette represented by blaGES-1/aacA4. The cassette array was analyzed by Northern blot and real-time reverse transcription-polymerase chain reaction (RT-PCR) in order to assess the transcription mechanism of blaGES-1/aacA4 fused cassette. Also, inverse polymerase chain reactions (PCR) were performed to detect the free circular forms of gcu14, blaGES-1 and aacA4. The Northern blot and real time RT-PCR revealed a polycistronic transcription, in which the fused cassette blaGES-1/aacA4 is transcribed as a unique gene, while gcu14 (with a canonical attC recombination site) has a monocistronic transcription. The gcu14 cassette, closer to the weak configuration of cassette promoter (Pc), had a higher transcription level than blaGES-1/aacA4, indicating that the cassette position impacts the transcript amounts. The presence of ORF-11 at attI1, immediately preceding gcu14, and of a Shine-Dalgarno sequence upstream blaGES-1/aacA4 composes a scenario for the occurrence of array translation. Inverse PCR generated amplicons corresponding to gcu14, gcu14-aacA4 and gcu14-blaGES-1/aacA4 free circular forms, but not to blaGES-1 and aacA4 alone, indicating that the GES-1 truncated attC is not substrate of integrase activity and that these genes are mobilized together as a unique cassette. This study was original in showing the transcription of fused cassettes and in correlating cassette position with transcription.

A resistência a carbapenemases em bacilos gram-negativas (BGN) tornou-se um problema mundial, elevando a taxa de morbidade e mortalidade. O método automatizado foi utilizado para identificação e suscetibilidade dos isolados. E o método molecular foi aplicado na detecção de bacilos gram-negativas (BGN) produtoras de carbapenemase. Um total de 205 BGN foram recuperados a partir de diferentes amostras clínicas, resistentes a carbapenemases. Os genes de resistência blaKPC (57,5%), blaVIM (30,2%), blaGES (17%), blaNDM (15%) e blaSPM (2,4%) foram recuperados a partir de isolados clínicos. O gene blaIMP não foi detectado em nenhum dos isolados. É preocupante que Pseudomonas aeruginosa e Acinetobacter baumannii representem a maioria dos isolados (61,6%) recuperados de infecções de diferentes sítios, com alta resistência a carbapenêmicos, carregando simultaneamente os genes blaKPC, blaGES e blaVIM, assim como encontrados também em Enterobactericeae. A carbapenemase mais prevalente foi blaKPC e blaVIM, seguida de blaNDM dentre os isolados multidroga resistentes. Esses resultados são um ameaça à saúde pública, configurando altas taxas de resistência, limitando as opções terapêuticas.

A novel class 1 integron carrying a gene cassette array oforf28-orf28-blaGES-5was identified on pGES-GZ, within whichorf28encoded a hypothetical protein possibly correlated to fosfomycin resistance.

Wastes arising from human activities can reach water bodies and contribute significantly to the presence of antibiotic resistant bacterial populations in aquatic environments. The objective of this study was to evaluate the cultivable antibiotic resistant bacterial populations from a coastal lagoon impacted by agriculture and urbanization activities. Water samples were collected in low and peak season and characterized regarding physicochemical variables, microbiological indicators and the presence of antimicrobial residues. In order to analyze the presence of resistant bacterial populations, the samples were grown in the presence of nalidixic acid, ceftazidime, imipenem and tetracycline. Genes associated with β-lactamic resistance (blaCTX-M-like, blaGES-like, blaOXA-51, blaOXA-23-like, blaSHV-like, blaTEM-like and blaSPM-1), class I integron and efflux systems (tetA, tetB, acrA, acrB, tolC, adeA, adeB, adeR, adeS, mexB, mexD, mexF and mexY) were analyzed by conventional in vitro amplification. Although antimicrobials residues were below the detection limit, resistant bacteria and resistance determinants - blaGES, class I integron, adeS, acrA, acrB, tolC, mexB, mexF - were present at almost all points, in both seasons and for all antimicrobials assessed. The high numbers of resistant bacteria counts observed after the antibiotic treatment were positively correlated to the urbanization effects on the Lagoon. Some resistant populations were even higher in the low season samples, indicating the importance of a systematic evaluation of antibiotic resistance on water resources.

La resistencia bacteriana representa un problema para la salud pública, especialmente la producida por enzimas llamadas betalactamasas, que bloquean a un grupo importante de antibióticos, los betalactámicos, utilizados ampliamente para el tratamiento de infecciones en seres humanos. Las enterobacterias productoras de betalactamasas de espectro extendido (BLEEs) son las más preocupantes, pues son un grupo importante dentro de la etiología de las infecciones tanto severas como no severas. Los carbapenemes son antibióticos betalactámicos para el tratamiento de infecciones causadas por enterobacterias BLEE, que están resultando ineficientes por el aparecimiento de cepas resistentes a estos antibióticos. Ante la escasez de otras opciones terapéuticas, el presente estudio tuvo como objetivo caracterizar molecularmente carbapenemasas en enterobacterias y analizar su posible inhibición con secreciones peptídicas de un bufónido nativo ecuatoriano. Se caracterizaron 57 enterobacterias resistentes a carbapenemes de pacientes hospitalizados. Esta población fue caracterizada molecularmente, encontrándose los genes blaKPC, blaGES, blaVIM y blaIMP. Los genes con mayor prevalencia fueron blaKPC y blaGES, seguidos por blaVIM y blaIMP. Todos los aislados fueron inhibidos por el péptido. La concentración inhibitoria mínima fue 250 μg/ml para Klebsiella pneumoniae, Serratia marcescens, Enterobacter aerogenes y E. cloacae, 125 μg/ml para Morganella morganii, 62.5 μg/ml para Klebsiella oxytoca y Serratia rubidaea y 31.3 μg/ml para Escherichia coli. La presencia de genes para betalactamasas en elementos móviles facilita su propagación. Por esta razón la importancia de analizar nuevos compuestos antimicrobianos.

Introduction: The spread of Acinetobacter baumannii, resistant to most of the available antimicrobial agents, is a serious health problem. The high rate of carbapenem resistance among Acinetobacter baumannii isolates is considered as a threat to public health. In this study, we aimed to determine the antibiotic resistance and related genes in carbapenem-resistant Acinetobacter baumannii isolates. Methodology: Ninety six isolates of A. baumannii were included. Antimicrobial susceptibility was performed by Phoenix Automated System and disk diffusion method. Carbapenem resistane was characterized by scrneeing of resistance genes such as blaTEM, blaSHV, blaCTX-M1-2, blaPER, blaVEB, blaKPC, blaGES, blaNDM, blaVIM, blaIMP and blaOXA23-24-51-58 using multiplex polymerase chain reaction. Results: Resistance for the levofloxacin, gentamicin, amikacin, and tigecycline were determined as 96.9%, 93.7%, 72.9% and 45.8% respectively. Colistin was the only susceptible antibiotic against all clinical isolates. All isolates were defined as multidrug resistance and of these, 31.2% were extensively drug-resistant (sensitive only to colistin). BlaOXA-51­ and blaOXA-23 genes were detected in 100% strains while blaTEM was found in only 2% strains. There was no amplification for the blaSHV, blaCTX-M1-2, blaPER, blaVEB, blaKPC, blaGES blaNDM, blaVIM, blaIMP and blaOXA24-58 genes. Conclusions: The high frequency of blaOXA-23 and low frequency of blaTEM gene was observed that indicate prevalence of a variety of A. baumannii strains. The rates of resistance genes vary from region to region. Studies are required for the prevention and control of A. baumannii infection and to formulate the strategies of antibiotic usage.

SUMMARY In this brief communication we describe the occurrence of a KPC-producing Serratia marcescensisolate in a home-care patient from Recife, Brazil. The blaKPC, blaSPM, blaIMP, blaVIMblaOXA, blaCTX-M, blaSHV, blaTEM and blaGES genes were investigated by Polymerase Chain Reaction (PCR) and DNA sequencing. The isolate was positive for blaKPC-2 and blaTEM-1 and was resistant to aztreonam, cefepime, cefotaxime, imipenem, meropenem, gentamicin, ciprofloxacin and cefazidime, and susceptible only to amikacin, tigecycline and gatifloxacin. This is the first report in Brazil of KPC-producing S. marcescens clinical isolate outside of a hospital environment. Caregivers should be alert for the presence of this isolate in the community setting.

Abstract Objectives Carbapenem-resistant Pseudomonas aeruginosa (CR-PSA) imposes great limitations on empirical therapeutic choices, which are further complicated by metallo-β-lactamase production. This study evaluated in vitro antimicrobial synergy of ceftolozane/tazobactam in combination with aztreonam and fosfomycin against MDR PSA. Methods MICs were determined by broth microdilution and gradient strips. The effect of ceftolozane/tazobactam+aztreonam and ceftolozane/tazobactam+fosfomycin combinations were tested against 27 MDR PSA isolates carrying blaSPM-1 (n = 13), blaIMP (n = 4), blaVIM (n = 3), blaGES-1 (n = 2) and blaCTX-M-like (n = 2), and 3 isolates with no acquired β-lactamase production detected by gradient diffusion strip crossing (GDSC). Six genetically unrelated SPM-1-producing isolates were also evaluated by time–kill analysis (TKA). Results All CR-PSA isolates harbouring blaSPM-1, blaGES-1 and blaIMP-1 were categorized as resistant to ceftolozane/tazobactam, meropenem and fosfomycin, with 70% being susceptible to aztreonam. Synergism for ceftolozane/tazobactam+fosfomycin and ceftolozane/tazobactam+aztreonam combinations was observed for 88.9% (24/27) and 18.5% (5/27) of the isolates by GDSC, respectively. A 3- to 9-fold reduction in ceftolozane/tazobactam MICs was observed, depending on the combination. Ceftolozane/tazobactam+fosfomycin was synergistic by TKA against one of six SPM-1-producing isolates, with additional non-synergistic bacterial density reduction for another isolate. Aztreonam peak concentrations alone demonstrated a ≥3 log10 cfu/mL reduction against all six isolates, but all strains were within the susceptible range for the drug. No antagonism was observed. Conclusions In the context of increasing CR-PSA and the genetic diversity of resistance mechanisms, new combinations and stewardship strategies may need to be explored in the face of increasingly difficult to treat pathogens.

New and specified data on the composition of the Triassic tetrapod assemblages of the Timan-North Urals region, the Mezen syneclise and the eastern part of the Moscow syneclise have been given. The succession of the Triassic tetrapod communities known in this area includes eight assemblages, seven of which correspond to the contemporary communities of the terrestial vertebrates known from other regions of the East European platform and the Cisurals. The first assemblage, known from the basal Triassic of the Mezen and Moscow syneclises, is Early Induan in age. The second, third and fourth assemblages are dated by Early Olenekian age and typical for the middle part of the Early Tri-assic section of the Timan-North Urals region, the Mezen and Moscow syneclises. The Late Olenekian fifth and sixth assemblages characterize the upper part of the Lower Triassic of the Northern Cisurals, the Moscow and Mezen syneclises. The seventh assemblage characterizes the base of the Middle Triassic (Lower Anisian) of the Northern Cisurals and has no analogues in other regions of the platform and of the Cisurals. The eighth assemblage is known from the upper part of the Middle Triassic of the Northern Cisurals and is Late Ladinian in age.

Abstract Objectives To evaluate the efficacy of the recently launched β-lactam/β-lactamase inhibitor combinations ceftazidime/avibactam and ceftolozane/tazobactam against ESBL-producing Escherichia coli and Pseudomonas aeruginosa strains. Methods A series of ESBL-encoding genes (blaTEM, blaSHV, blaCTX-M, blaVEB, blaPER, blaGES and blaBEL) was cloned and expressed in E. coli or P. aeruginosa recipient strains. Cultures of E. coli TOP10 harbouring recombinant plasmids and therefore producing the different ESBLs tested were grown in order to perform measurements of catalytic activities, using benzylpenicillin, ceftazidime and ceftolozane as substrates. IC50s were additionally determined for clavulanic acid, tazobactam and avibactam. Results We showed here an overall better activity of ceftazidime/avibactam compared with ceftolozane/tazobactam toward ESBL-producing E. coli and P. aeruginosa. Several ESBLs of the GES, PER and BEL types conferred resistance to ceftolozane/tazobactam in E. coli and P. aeruginosa. For GES-6 and PER-1 producers, resistance to ceftolozane/tazobactam could be explained by a high hydrolysis of ceftolozane and a low activity of tazobactam as an inhibitor. On the other hand, PER-producing P. aeruginosa also exhibited resistance to ceftazidime/avibactam. Conclusions Altogether, the results show that the ESBL PER-1, which is widespread worldwide, may be a source of resistance to both ceftolozane/tazobactam and ceftazidime/avibactam. Excellent activity of ceftazidime/avibactam was highlighted for both ESBL-producing E. coli and ESBL-producing P. aeruginosa.

We present the entomological results of a geological and biological exploration in the Lutdesert, performed in 2014 by the Iranian National Institute for Oceanography and AtmosphericSciences (INIOAS) and the University of Tehran. Five localities were sampled, including theone considered as the the hottest spot on Earth. They yielded 15 insect taxa belonging to Blat-todea, Orthoptera, Coleoptera, Heteroptera, suggesting that in spite of being originally de-scribed as “abiotic” or “aphytic”, the Lut desert shelters a diverse insect fauna with someremarkable elements such as the endemic desert cockroache Leiopteroblatta monodi. Compar-ison with results obtained by Franco-Iranian expeditions in the sixties and seventies revealsseveral taxa in common, but also striking differences in the composition of the insect assem-blages, suggesting that the insect fauna in the Lut desert is potentially much more diverse thanpreviously expected.

Resistance to carbapenems due to carbapenemase-producing Enterobacteriaceae (CPE) is an increasing threat to human health worldwide. In recent years, CPE could be found only sporadically from livestock, but concern rose that livestock might become a reservoir for CPE. In 2019, the first GES carbapenemase-producing Escherichia coli from livestock was detected within the German national monitoring on antimicrobial resistance. The isolate was obtained from pig feces and was phenotypically resistant to meropenem and ertapenem. The isolate harbored three successive blaGES genes encoding for GES-1, GES-5 and GES-5B in an incomplete class-I integron on a 12 kb plasmid (pEC19-AB02908; Acc. No. MT955355). The strain further encoded for virulence-associated genes typical for uropathogenic E. coli, which might hint at an increased pathogenic potential. The isolate produced the third carbapenemase detected from German livestock. The finding underlines the importance CPE monitoring and detailed characterization of new isolates.

Vibration localization of bladed disk turns to be much complex when a number of blades are assembled into periodic groups. This work focused on natural frequency distribution and modal localization of bladed disks with grouped blades based on the lumped parameter models, and effects of the blade number in each group on natural frequency were studied. Then Monte Carlo method was applied to analyze the sensitivity of modal localization to the random mistuning of blade stiffness. The results show that the number of blades in each group influences the nature frequency of bladed disk with grouped blades, and modal localization of tuned bladed disk with grouped blades is found in the closely spaced modal region. Moreover, compared to the bladed disk with free blades, the modal localization of bladed disk with grouped blades is much less sensitive to random mistuning of blade stiffness.

Abstract Background ASPECT-NP, a phase 3, randomized, double-blind, multicenter trial, evaluated ceftolozane/tazobactam (C/T) 3 g q8h vs. meropenem 1 g q8h for 8–14 days in adults for treatment of ventilated nosocomial pneumonia. Baseline Gram-negative (GN) isolates from patients were tested for mechanisms of resistance. Methods Lower respiratory tract (LRT) isolates were sent to a central laboratory for organism identification and susceptibility. Of 664 total Enterobacteriaceae (ENT) and Pseudomonas aeruginosa (PsA) isolates, 351 (53%) were nonsusceptible to broad-spectrum cephalosporins and/or carbapenems and underwent whole-genome sequencing, quantitative RT–PCR, and western blot analysis. ENT isolates were tested for the presence of acquired β-lactamase genes and AmpC levels (selected species). PsA isolates were tested for acquired β-lactamase genes, AmpC (PDC) levels, efflux pump expression, and OprD loss. Results Of 262 ENT isolates, 114 (44%) were susceptible to C/T (MIC ≤2 µg/mL). An extended-spectrum β-lactamase (ESBL) gene was carried by 89 (78%) of the C/T-susceptible isolates. Of 148 C/T-nonsusceptible (C/T-NS) isolates, 87 (59%) were carbapenemase negative, and the majority 135 (91%) also carried an ESBL gene. The most common ESBL was blaCTX-M15 with blaOXA-1 and blaOXA-30. Klebsiella pneumoniae often displayed higher C/T MICs compared with other species carrying the same resistance genes. Among all C/T-NS isolates, 61 (41%) were carbapenemase positive, most commonly K. pneumoniae carrying blaOXA-48, blaNDM-1, and blaNDM-5. Of 89 PsA isolates, 58 (65%) were susceptible to C/T (MIC ≤4 µg/mL), despite elevated AmpC expression, efflux pumps, or loss of OprD; only 5 isolates had an acquired β-lactamase. Of the 31 C/T-NS PsA isolates, only 12 (39%) were carbapenemase positive and carried blaVIM or blaGES; isolates carrying blaGES had lower C/T MICs (8–32 µg/mL) compared with blaVIM (MIC > 128 µg/mL). PDC alleles were similar in isolates with high and low C/T MICs. Conclusion In baseline GN LRT isolates from ASPECT-NP, the most common ESBL detected in ENT was blaCTX-M15; carbapenemases were uncommon. There was no correlation of ESBL phenotype to C/T susceptibility among ENT, nor of PDC allele to C/T susceptibility among PsA. Disclosures All authors: No reported disclosures.

Mistuning in bladed disks usually increases the forced response of the maximum responding blade leading to shortened component life in turbine engines. This paper investigates mistuning using a transfer function approach where the frequency response functions (FRFs) are described by natural frequencies and antiresonant frequencies. Using this approach, antiresonant frequencies are shown to be a critical factor in determining the maximum blade response. Two insights are gained by formulating antiresonant frequencies as the eigenvalues of reduced system matrices: 1) Mistuning a particular blade has no effect on that blade's antiresonant frequencies. 2) Engine orders N and N/2, where N is the number of blades on the disk, tend to produce the highest maximum local response. Numerical examples are given using a spring-mass-oscillator model of a bladed disk. Pole-zero loci of mistuned bladed disks show that increased maximum blade response is often due to the damping of antiresonant frequencies. An important conclusion is that antiresonant frequencies can be arranged such that a mistuned bladed disk has a lower maximum blade response than a tuned bladed disk.

Pseudomonas aeruginosa, a bacterium commonly isolated from hospital settings, exhibits intrinsic resistance to a number of antibiotics and can acquire resistance during antibiotic therapy. Resistance towards carbapenems is increasing due to its overuse in the treatment of infections caused by extended-spectrum β-lactamase (ESBL) producing organisms. Nonetheless, carbapenems are essential for the treatment of high-risk infections and are one of the remaining weapons in the fight against “extreme drug resistance” of Gram-negative/positive bacilli. Herein, we describe a case report of infections caused by P. aeruginosa strains that carry blaVIM-2 and blaKPC-2 carbapenemase genes simultaneously, identified in five patients who were admitted to a high complexity health institution in Colombia. Molecular characterization included PCR screening for blaKPC, blaGES, blaOXA-48, blaIMP, blaNDM, and blaVIM carbapenemase and other resistance genes as well as analysis of the genetic relationships by genome macro-restriction and Pulsed-Field Gel Electrophoresis (PFGE) separation. In conclusion, these infections represent a major challenge to public health due to the risk of the infection spreading compounded by the fact that limited treatment options are available, thereby increasing the risk of increased morbidity and mortality.

Recently, a total of 32 carbapenem- and fluoroquinolone-resistant Acinetobacter baumannii (Ab) isolates was isolated from war-injured patients who were treated at German Bundeswehr Hospitals, and preliminarily typed by “DiversiLab” repetitive elements sequence-based (rep-) PCR. Core genome-based sequence typing was also used to provide more detailed epidemiological information. From the clusters observed by rep-PCR, selected Ab strains were subjected to Next Generation Sequencing (NGS) in order to compare them with international outbreak-associated Ab strains and to identify MLST (multi-locus sequence type) lineages, as well as to identify known resistance genes. Accordingly, NGS indicated higher diversity than rep-PCR, but also confirmed likely transmission events. The identified acquired carbapenem-resistant genes comprised blaOXA-23, blaOXA-72 and blaGES-12, as well as various other intrinsic and acquired resistance-associated genetic elements. All isolates clustered with the previously identified international clonal lineages IC1, IC2, IC6 and IC7, with corresponding Pasteur sequence types ST1, ST2, ST78 and ST25, respectively. In conclusion, the assessment confirmed a broad spectrum of resistance-associated genes in Ab isolated from war-injured patients from the Eastern Ukraine, and provided the first insights into locally abundant clonal lineages.

This paper describes measurements of the shape of a 900 mm long, hand-carved timber blade for a 500 W three-bladed horizontal axis wind turbine. Four blades were hand-carved in Nepal by reference to a master blade cut in Australia on a numerically controlled milling machine. A high definition three-dimensional scanner was used to determine the surface of one hand-carved blade as a series of profiles at 50 mm intervals along the blade's length. A surface model generated from these profiles was compared to the designed blade shape in terms of the three fundamental blade design parameters: chord, twist, and profile shape. The measured twist and chord were less that the design values, particularly in the hub region. This is consistent with the poor starting performance of the turbine when mounted with the remaining hand-carved blades. Assessment of the differences in profile shape would require a detailed computational analysis, which has not been undertaken.

Carbapenems are considered to be the last resort antibiotics for the treatment of infections caused by extended-spectrum beta-lactamase (ESBL)-producing strains. The purpose of this study was to assess antimicrobial resistance profile of Carbapenem-resistant Enterobacteriaceae (CRE) isolated from cattle faeces and determine the presence of carbapenemase and ESBL encoding genes. A total of 233 faecal samples were collected from cattle and analysed for the presence of CRE. The CRE isolates revealed resistance phenotypes against imipenem (42%), ertapenem (35%), doripenem (30%), meropenem (28%), cefotaxime, (59.6%) aztreonam (54.3%) and cefuroxime (47.7%). Multidrug resistance phenotypes ranged from 1.4 to 27% while multi antibiotic resistance (MAR) index value ranged from 0.23 to 0.69, with an average of 0.40. Escherichia coli (E. coli), Klebsiella pneumoniae (K. pneumoniae), Proteus mirabilis (P. mirabilis) and Salmonella (34.4, 43.7, 1.3 and 4.6%, respectively) were the most frequented detected species through genus specific PCR analysis. Detection of genes encoding carbapenemase ranged from 3.3% to 35% (blaKPC, blaNDM, blaGES, blaOXA-48, blaVIM and blaOXA-23). Furthermore, CRE isolates harboured ESBL genes (blaSHV (33.1%), blaTEM (22.5%), blaCTX-M (20.5%) and blaOXA (11.3%)). In conclusion, these findings indicate that cattle harbour CRE carrying ESBL determinants and thus, proper hygiene measures must be enforced to mitigate the spread of CRE strains to food products.

The global dissemination of carbapenemase-producing Enterobacteriaceae (CPE) is a major concern in public health. Due to the existence of the diversity of carbapenemases, development of an easily available, cost-effective multiplex detection assay for CPE is required worldwide. Using clinically available and reliable equipment, COBAS® z480 (Roche Diagnostics K.K., Tokyo, Japan), we developed a multiplex real-time PCR assay for the detection of two combinations of carbapenemases; first, blaNDM, blaKPC, and blaIMP (Set 1), and second, blaGES, blaOXA-48, and blaVIM (Set 2). We constructed standard curves for each carbapenemase gene using serial dilutions of DNA standards, then applied reference or clinical isolates with each carbapenemase gene to this assay. The multiplex assay showed satisfactory accuracy to detect CPE genes, with the correlation coefficients of greater than 0.99 for all genotypes. The assay appropriately differentiated the reference or clinical strains harboring each carbapenemase gene without cross reactivity. Lastly, the assay successfully detected multiple genes without false-positive reactions by applying six clinical isolates carrying both NDM and OXA-48-like carbapenemase genes. Major advantages of our assay include multiplicity, simple operation, robustness, and speed (1 h). We believe that the multiplex assay potentially contributes to early diagnosis of CPE with a diverse genetic background.

In this paper, the influence of blades number on the performance of pico wind turbine was investigated by using a small-motorized axial DC fan with a rated power of 4W. Fixed streaming air blower was used as a source of wind. Varying in wind speed was accomplished by changing the distance from the blower. A resistor equals to the turbine internal resistance was utilized as a load to collect the electrical power across the load at various wind speeds and for fans of different blades (1, 2, and 5). Values of the cut-in and cut-out speeds were extracted from the power plot. Rated power was recorded, as well. The results have shown that the rated power generated by turbine has decreased due to the reduction of blades number (i.e., reduction in solidity) from 2.6W for a 5-bladed turbine to 0.665W for a 2-bladed turbine and to 0.13W for a 1-bladed turbine. Moreover, the cut-in speed (initial electrical generating speed) has increased from 4.9m/s for 5-bladed to 8m/s for 2-bladed, then to 19.15m/s for 1-bladed. These results are explained by the balancing problems during rotation (polar asymmetrical rotor), and it is seen that the reduction of blades has made a sharp reduction in power coefficient.

AbstractThis paper demonstrates the results of analyses of Cladocera, pollen, plant macrofossil, lithological and radiocarbon data recovered from a mire located in the Rawka River valley in central Poland. These studies permit to recognise the development of hydrology phases in Kopanicha mire and give insight into Holocene fluvial dynamics of the system; radiocarbon dating partly allowed da-ting of the patterns of Holocene valley floor development of the Rawka River. The Kopanicha mire was formed during the Atlantic period. At that time, an oxbow lake formed, becoming a mire during the Subboreal and possibly Subatlantic periods. The high sensitivity of the oxbow lakes, fens, fauna, and flora remains to climate variations - especially to changes in water level connected with the ag-gradation-erosion cycle of rivers - allows the reconstruction of the palaeoecological changes that oc-curred in the mire. The frequency and timing of hydroclimatic oscillations at Kopanicha show strong similarities to records from other sites in Poland. Changes in Cladocera frequency and plant assem-blages were mostly influenced by the Rawka River which controlled the hydrological regime of the mire. The main factors controlling the presence of Cladocera taxa were the water level and the pres-ence of favourable conditions in the mire (e.g. pH, vegetation). Most of the changes occurred in re-sponse to climate changes, but some of them were connected with local factors.

(1) Background: Beta-lactams are the most frequently used antimicrobials, and are the first-line drugs in many infectious diseases, e.g., pneumonia, otitis media. Due to this fact, various bacteria have developed resistance to this group of drugs. (2) Methods: Eighty-seven Haemophilus parainfluenzae isolates were obtained from adults 18–70 years old in eastern Poland. The presence of 10 bla genes and 2 substitutions in ftsI reported as the most frequent in H. parainfluenzae were analyzed. (3) Results: Among 57 beta-lactam-resistant isolates, 63.2% encoded bla genes; blaTEM-1 predominated (54.4%), followed by blaOXA (19.3%), blaDHA (12.3%), blaSHV (10.5%), blaGES (7.0%), blaCMY (5.3%), blaVEB (1.8%) and blaROB-1 (1.8%). Lys-526 was the most common substitution in ftsI gene. The resistance genotypes were as follows: gBLNAS (17.5%), low-gBLNAR I (1.8%), low-gBLNAR II (1.8%), gBLNAR II (15.8%), gBLPAS (15.8%), gBLPAR (19.3%), gBLPBS I (8.8%) and gBLPBS II (1.8%); (4) Conclusions: This has been the first study to report on the high diversity of bla genes in H. parainfluenzae isolates in Poland. High sensitivity and specificity of benzylpenicillin test, as well as PCR of bla genes were shown, indicating that these methods may be useful as tools for the rapid screening of beta-lactamase prevalence and resistance to beta-lactams among H. parainfluenzae isolated from respiratory microbiota.

Abstract Background Infections caused by carbapenem-resistant Enterobacterales (CREs) are an emerging problem associated with high rates of morbidity and mortality. CREs are divided into two categories (carbapenemase-producing [CP] CRE and non-CP CRE). The most prevalent carbapenemase produced by Enterobacterales is Klebsiella pneumoniae carbapenemase (KPC) in Korea. Rapid identification of CREs is clinically important in infection control precaution. We compared the performance of two chromogenic media (chromID CARBA agar and CHROMagar KPC agar) for non-CP CREs or CP CREs with blaGES-5, blaNDM-1 or blaVIM-2 in a Korean hospital. Methods The study was carried out during a 3-month period from April to June 2017 during the surveillance program for CRE colonization. Antimicrobial susceptibility testing (AST) and polymerase chain reaction (PCR) were performed at the Korean Centers for Disease Control and Prevention. Results A total of 45 rectal swabs from 42 hospitalized patients were examined. Sensitivity of both chromID CARBA and CHROMagar KPC were 100% for CP CREs; and 50% and 100% for non-CP CREs, respectively. Specificity of chromID CARBA and CHROMagar KPC were 89.2% and 70.3% for CP CRE, respectively; and 76.9% and 66.7% for non-CP CRE, respectively. Conclusions The CHROMagar KPC is useful to monitor non-CP and CP CREs. The chromID CARBA is efficient for rapid detection of CP CREs requiring high contact precaution.

The paper deals with the results of an archeo-zoological study of bone materials from a num- ber of newly discovered settlements in Northwestern Crimea (Tarkhankut-Н2, Tarkhankut-Н8, Tarkhankut-18 and Tarkhankut-22а). According to the available archaeological evidence and ra- diocarbon determinations obtained on bone remains from cultural layers and semi-closed assem- blages, the materials in question belong to the early stage of the Babino culture (Tarkhankut-18, Tarkhankut-22а) and to the Sabatinovka (Tarkhankut-Н2, Tarkhankut-22а) and Belozerka (Tarkhankut-Н8, Tarkhankut-18) cultures. In addition, the Tarkhankut-22а settlement yielded some Early Iron Age materials. Altogether, 1211 bones were studied, and 284 of them proved to be identifiable. An attempt is made to consider the herd composition in its dynamics: during the final stage of the Middle Bronze Age the herd was heavily dominated by cattle, followed by small ru- minants, while the percentage of horse remains was insignificant. The Late Bronze Age witnessed an increase in the numbers of small ruminants, and the presence of horse became perceptible in the final of the Bronze Age. The authors compare their data with the information available for the Middle and Late Bronze Age settlements of Crimea and the synchronous cultures of the North Black Sea region. The analyzed materials point to the existence of pastoral stockbreeding with stone architecture represented by livestock enclosures.
