It was determined that the metal boards of conservative liquid based on the composition of amidoamine (which is synthesized in a ratio of 1:1 mol with 1,6-diaminohexane of soya acid oil) and cotton oil acid have higher protection effects on corrosion than conservative liquids that based on composition of aminoamine and other fatty acids. Abstract (sommario):
Dendritic nano-polymers are recently used in medical and industrial applications. Cationic dendritic polymers can be used for the modification of anionic cellulose fibers. This review deals with the chemical modification of cellulosic fibers with poly(amidoamine) type dendritic polymers. It has been shown that after modification, the physical and mechanical properties including moisture regain, breaking strength, breaking elongation, and Young’s modulus of the treated cotton fibers increase slightly. It is also a possible way to achieve saltfree dyeing with reactive dyes. Several researches concluded that when poly(amidoamine) increases in cellulose fiber, the mechanism of dye adsorption changes from Freundlich to Langmuir model. Moreover, dendritic polymers can be used as a template for inorganic nano-particles. Both poly(amidoamine) silver salts and nano-composites can display antimicrobial activity. On the other hand, the water and oil repellency results showed that poly(amidoamine) dendrimer containing fluorocarbon had better results than conventional ones in relation to performance and washing resistance. To improve wash and wear properties, poly(amidoamine) dendrimers can be modified and applied as a new material for wrinkle resistance of cotton.

Abstract (sommario):
Poly(amidoamine)-based liquid crystalline dendrimers (LCDs) with mesogenes on the surface were successfully prepared by the reaction of poly(amidoamine) dendrimers with 40-cyanobiphenyl hydrogen glutarate in the presence of a condensing agent, diphenyl(2,3- dihydro-2-thioxo-3-benzoxazolyl)phosphonate. The structures of LCDs were characterized by IR, 1H-NMR and MALDI-TOF mass spectroscopy and elemental analysis. These LCDs did not show a thermotropic liquid crystalline nature but exhibited a lyotropic liquid crystalline property in the 80 wt% DMF solutions of LCDs containing lithium bromide.

Abstract (sommario):
Abstract: Poly(amidoamine)s (PAAs) represent a promising material for drug delivery applications. They are synthetic functional polymers endowed with a combination of properties making them suitable for a variety of biomedical applications mostly related to drug delivery systems. PAAs can be obtained by stepwise polyaddition of primary or secondary aliphatic amines to bisacrylamides. The synthesis is carried out in water or protic solvent, with a mild base and at room temperature. In the first section of this thesis, a group of novel amphoteric PAAs homo- and copolymers containing secondary and tertiary amine groups in their main chain and different structures in the bisacrylamide segments were synthesized and evaluated as magnetic nanoparticles coating agents. In the second section of this thesis, a general method for preparing linear high polymers of peptides was investigated. Polymers with an average molecular weight included in the range 3000 – 40000 were obtained by Michael polyaddition reaction between 2,2-bisacrylamidoacetic acid or N,N’-bisacryloylpiperazine and glutathione (reduced and oxidised) or the RGD peptide. In preliminary biological tests these polymers showed a good values of zeta potential and negligible values of cytotoxicity and haemolytic activity. Further in vitro tests are in progress to investigate the biological properties of these polymers in comparison with the parent peptides. In the third section of this thesis, gene delivery properties of new hyperbranched PAAs, containing disulfide linkages in the main chain, were investigated in comparison with the relevant linear polymers. Structural characterization of the polymers synthesized and the reactivity of the monomers utilized were also determined. The results shown by these polymers in DNA transfection are very promising and encourage further studies in vitro with these biodegradable hyperbranched PAAs to test the effect, on transfection efficiency, of different terminal groups.

Abstract (sommario):
The multi-step syntheses of two water-soluble AB(_2) monomers, N-acryloyl-1,2- diaminoethane hydrochloride and N-acryloyl-1,2-daminoethane, are reported. The melt polymerisation at 210 ºC for 4 hours of N-acryloyl-1,2-diaminoethane hydrochloride gave, by Michael additions of the terminal ammonium salts (B(_2) groups) of a monomer molecule or growing oligomer with the unreacted vinyl double bond (A group) of another monomer molecule or growing oligomer, hyperbranched macromolecules. High degrees of conversion, determined using (^1)H NMR spectroscopy, were obtained and a degree of branching, determined using (^15)N NMR spectroscopy, was found to be equal or close to 1. However, this method proved difficult to reproduce and the alternative aqueous solution polymerisation at 100 ºC for 10 days of this monomer was investigated. The use of ampoules closed with Teflon taps as reactors lead to polymers displaying random signs of free radical polymerisation and high degrees of conversion due to such side-reaction, whereas, the alternative use of sealed Carius tubes lead to lower DP materials displaying no significant signs of free radical side-reactions. The kinetic study of this polymerisation allowed the determination of the rate constant associated with the oligomer growth and the MALDI analysis of these polymers revealed an evolution with time of the oligomer molecular weight distribution with time in agreement with the theoretical prediction. This last study also revealed that at least 70% of the molecules were not cyclised. On the other hand, the aqueous solution polymerisation of the corresponding free base monomer, N-acryloyl-1,2- diaminoethane, at 100 ºC for 90 minutes lead to hyperbranched macromolecules with high DP values. The kinetic study did not allow a precise determination of the rate constant associated with the oligomer growth due to the high reactivity of the free terminal amino groups. The MALDI analysis of these polymer samples showed that the evolution with time of the oligomer molecular weight distribution did not follow the theoretical prediction and that these macromolecules were cyclised.

Abstract (sommario):
The control of cell growth, proliferation and differentiation on biomaterials surfaces is of fundamental importance for regenerative medicine, prosthetics, or cell-based assays. The microfabrication of cell-on-chip platforms based on a new family of poly (amidoamine) hydrogels, are promising for in vitro and in vivo applications. Hydrogels present characteristics that mimics biological environments, such as the cross-linked nature of the extracellular matrix, the tissue properties (high water content), and the permeability to oxygen and metabolites. Hydrogels based on poly (amidoamine) results in an optically transparent, biocompatible and fully biodegradable substrate recommended for body implants that are minimally invasive, and naturally eliminated by human body. In my PhD work I intended to use microfabricated hydrogels for fine-tuning the contact guidance of cells. As microfabrication tools I set up reaction injection moulding for producing features down to 100 µm and developed a novel approach relying on electron beam lithography. This innovating microfabrication consists in the ability of directly writing patterns on already cross-linked hydrogels, with the capability of producing structures at sub-micrometric scale. The exposure to the electron beam produces particular modifications enabling the control of physico-chemical properties of irradiated area. I obtained a selective attachment of proteins as a function of the electron-beam dose; an exclusive adhesion and growth of neural cells on the exposed surfaces; and the control of neurite outgrowth guidance along a microfabricated network. These results offer new perspectives to build physiological microenvironments or cell-on-chip platforms, based on a novel class of microfabricated hydrogels.

Abstract Major Applications Dendrimers are ideal macromolecular standards for use in size exclusion chromatography, membrane porosity evaluations, Newtonian viscosity applications, and electron microscopy. The mathematically defined surface functionality (Z may range from 2, 3, or 4 to several thousand) manifested by dendrimers (Figure 2) provides well-defined nanoscale building blocks for constructing complex nanostructures based on either covalent bonding or self-assembly-type processes. On the other hand, PAMAM dendrimers may function as hosts for the encapsulation of pharmaceuticals, agricultural chemicals, metals, fragrances, etc. once they have been grown to an appropriate congestion state that exhibits nanocontainer properties as shown in Figure 3.

Background: Poly (amidoamine) dendrimers (PAMAMs) are widely used in drug delivery systems and gene transfection as drug carriers. They also exert several biological effects like modulating gene expression, particularly EGFR (ErbB1) signaling pathway, which raises the question of whether these polymers can also inhibit the phosphorylation of HER2 (ErbB2) in breast cancer. However, this area hasn’t been investigated before. Methods: In this study, we evaluated the anticancer effects of different generations and surface chemistries of PAMAMs on HER2 positive breast cancer cells (SkBr3 and ZR-75 cell lines). Cell viability and morphological changes were evaluated upon treatment with PAMAMs. In addition, their effect on colony formation in soft agar was assessed. Additionally, western blot was performed to understand the underlying mechanisms of action. Results: PAMAMs anticancer effects were found to follow a specific trend, as they were more significant in cationic polymers and in higher generations. Cationic PAMAMs reduced cell viability of HER2 positive breast cancer cells up to 5.1% in SkBr3 and to 28% in ZR75 (p<0.001), in a dose and time-dependent manner. Cationic polymers also resulted in changing the morphology in the examined cell lines, as well as inhibiting colony formation in soft agar compared to controls (p<0.001). The mechanism of action was found to be mediated by inhibiting the phosphorylation of erbB2 and JNK1/2/3. Conclusion: These anticancer effects of PAMAM dendrimers make them promising molecules, which can add benefit to current anti-HER2 treatments and be employed successfully in different biomedical applications.

Abstract Emulsifier concentration in SBM is an important factor of drilling fluid stability. Proper concentration of amidoamine emulsifier is imperative for controlling low fluid loss and maintaining emulsion stability. This study investigates the physical and chemical interactions between emulsifier and other additives and describes the processes by which emulsifier is depleted from the drilling fluid. Three main pathways of emulsifier consumption are identified: emulsifier adsorption on solids found in drilling fluids and low gravity solids (LGS), chemical degradation, and to stabilize the invert emulsion. Design of experiments model and analytical procedure based on 1H NMR (nuclear magnetic resonance) spectroscopy was used to quantify the required emulsifier concentration in Non-Aqueous Fluid system (NAF). Additionally, model systems were used to estimate the excess of emulsifier, evaluate the emulsifier losses due to alkaline hydrolysis at elevated temperature, and measure adsorption of emulsifier on barite and various LGS types. Calculations for emulsifier depletion based on model systems were correlated to performance of formulated drilling fluids for verification. Typical emulsifier requirement in high performance NAF is 8-12 pounds per barrel (ppb). Majority of the emulsifier is adsorbed on weighting agents (barite) and rheology modifiers (clays), which are used to formulate NAF, that contribute to their effective dispersion in the solution and control fluid rheology. The adsorption process is found to be sensitive to the emulsifier concentration, solids mineralogy, wetting agent and temperature. Analytical Langmuir-Freundlich isotherm was used to describe adsorption data and estimate the adsorption capacity of the system. The emulsifier degradation pathway is another important factor of emulsifier consumption; however, emulsifier degradation at 250°F is not significant. While NAF are generally run ‘rich’ to mitigate depletion and maintain fluid stability, adsorption onto minerals will become an issue especially at high LGS concentration. These results will be greatly beneficial in the further development of NAF drilling fluid formulations and will assist field engineers in understanding the effect excess emulsifier will have on the drilling fluid and enable them to more effectively control the fluid properties under variations in emulsifier and LGS concentration during drilling.
