The industrial sector, particularly chemical manufacturing, is a significant contributor to global greenhouse gas emissions, with traditional processes primarily fueled by fossil fuels and operating under high temperatures and pressures. Introducing electrosynthesis at an industrial scale offers a promising avenue for integrating renewable electricity in chemical manufacturing, thus accelerating the decarbonization of large-scale chemical processes. This is especially pertinent in the production of Nylon 6,6, a key polymer reliant on adiponitrile (ADN) as a crucial intermediate. The dominant thermochemical production of ADN is not only energy-intensive but also employs hazardous reagents, such as HCN, highlighting the need for more sustainable production methods such as the electrochemical hydrodimerization of acrylonitrile (AN). Our research focuses on improving the performance and fundamentally understanding the electrochemical production of ADN from AN, one of the largest electro-organic reaction practiced in industry. In this reaction, the addition of tetraalkylammonium (TAA) salts as supporting electrolytes in moderate concentrations can enhance the solubility of organic reactants. By manipulating the molecular size and concentration of TAA ions, we observed an improvement in ADN selectivity and production efficiency, primarily influenced by the mass transport of organic reactants to the electrical double layer (EDL).1,2 To better understand the local effects of TAA ions at the electrode/electrolyte interface, we combined an electrochemical flow cell with attenuated total reflection Fourier-transform infrared (FTIR) spectroscopy. This approach revealed that TAA ions significantly increase the local concentration of AN near the electrode, which is influenced by the applied cathodic potentials and correlates with improved ADN selectivity. Additionally, we investigated the reaction mechanism of ADN synthesis using kinetic isotope effect (KIE) studies and electron paramagnetic resonance (EPR) spectroscopy. Results from KIE studies suggest that hydrogen transfer to AN is a rate-determining step in ADN production. EPR spectroscopy further revealed the presence of alkyl radicals in the solution, suggesting that ADN production partially occurs through the coupling of free AN radicals in solution. These insights can help in the design of electrolytes and the understanding of molecular processes that govern the selectivity and efficiency of ADN production via AN electrohydrodimerization and can inform the development and optimization of other electro-organic manufacturing processes. In the era of Industrial Revolution 4.0 and Society 5.0, 4C skills (communication, collaboration, creaticity, and critical thinking) have a major role in the development of Generation Z competencies in the era of Industrial Revolution 4.0 adn Societ 5.0. However, among the four skills, critical thinking skills have the most significant role in helping Generation Z deal with complexity and rapid change. Rapidly evolving technology and social changes also require Generation Z to think critically, as it helps in understanding and navigating information as well as challenges and making the right descisions in diverse situasions. Critical thinking skills also play an inmortant role in helping Generation Z develop a healthy skepticism towards information in the digital world. This article aims to explore the role og critical thinking skills as an important strategy in the development of Generation Z competencies. The methodology involves analyzing current literature in the fields of education, sociologi, and psychology to analyze the challenges and strategies associated with developing critical thinking among Generation Z. ZUSAMMENFASSUNGDie diabetische Neuropathie ist eine klinisch manifeste oder subklinische Erkrankung der peripheren Nerven, die infolge eines Diabetes mellitus ohne andere Ursachen auftritt. Sie kann das somatische (diabetische sensomotorische Polyneuropathie, DSPN) und/oder das autonome Nervensystem (autonome diabetische Neuropathie, ADN) betreffen. Eine DSPN ist in 85–90 % an der Ätiologie des diabetischen Fußsyndroms beteiligt und hat damit einen erstrangigen Stellenwert in der Risikokonstellation für Fußulkus und Amputation. Wenn die auf Symptomen beruhende Verdachtsdiagnose einer diabetischen Neuropathie nicht durch die Basisuntersuchungen gesichert werden kann, sollen spezifische Untersuchungen (Elektroneurografie und vor allem bei Verdacht auf eine sog. Small-Fiber-Neuropathie eine Quantitative Sensorische Testung) durchgeführt werden. Eine frühzeitige Kontrolle der Stoffwechseleinstellung und bestehender Risikofaktoren (z. B. Rauchen, übermäßiger Alkoholkonsum, Bluthochdruck) kann die Progression einer diabetischen Neuropathie verhindern oder zumindest hinauszögern oder verlangsamen. Die Therapie der schmerzhaften diabetischen Polyneuropathie umfasst pharmakologische und nicht-pharmakologische Ansätze. Im Rahmen der analgetischen Pharmakotherapie sind wichtige Leitsätze zu beachten und individuelle Therapieziele anzustreben. Bei ADN lassen sich symptomatische Manifestationen von asymptomatischen nur durch Funktionstests unterscheiden. Grundsätzlich kann die ADN jedes autonom innervierte Organ betreffen, die Einteilung erfolgt entsprechend den betroffenen Organen und Funktionssystemen. Das stufenweise Vorgehen bei der Diagnostik entspricht grundsätzlich dem bei der sensomotorischen Neuropathie. Im Vordergrund stehen organspezifische Untersuchungen in Zusammenarbeit mit anderen Fachdisziplinen. Die Grundsätze der allgemeinen Behandlungsstrategien und Prävention bei der diabetischen sensomotorischen Neuropathie gelten gleichermaßen für die autonomen Neuropathien. Die symptomatische Pharmakotherapie an verschiedenen Organ- und Funktionssystemen ist in der Regel durch die entsprechenden Spezialisten im Rahmen der interdisziplinären Zusammenarbeit einzuleiten.

Un alto porcentaje de especies de aves son sexualmente monomórficas, por lo que la determinación sexual es importante tanto para la reproducción en cautiverio así como para el establecimiento de programas de conservación y manejo de especies en peligro. En el presente trabajo se aplicó una técnica de sexaje molecular, inicialmente descrita para el género Ara, a otros cuatro géneros de Psitácidos: Pionus, Pionites, Amazona y Aratinga. El método consistió en el análisis de patrones de restricción de ADN generados por las enzimas HaeJII y DdeI en una región de los genes CHD 1 Z y CHD1 W Individuos analizados mediante esta técnica fueron apareados y tuvieron descendencia. La técnica de sexaje molecular usada en este estudio fue eficiente, rápida, sencilla y no agresiva para los individuos analizados.

AbstractIn this work we present chemical abundances of heavy elements (Z>28) for a homogeneous sample of 1059 stars from HARPS planet search program. We also derive ages using parallaxes from Hipparcos and Gaia DR1 to compare the results. We study the [X/Fe] ratios for different populations and compare them with models of Galactic chemical evolution. We find that thick disk stars are chemically disjunt for Zn adn Eu. Moreover, the high-alpha metal-rich population presents an interesting behaviour, with clear overabundances of Cu and Zn and lower abundances of Y and Ba with respect to thin disk stars. Several abundance ratios present a significant correlation with age for chemically separated thin disk stars (regardless of their metallicity) but thick disk stars do not present that behaviour. Moreover, at supersolar metallicities the trends with age tend to be weaker for several elements.

A high proportion of renewable energy connected to the power grid has caused power quality problems. Voltage-sensitive loads are extremely susceptible to voltage fluctuations, causing power system safety issues and economic losses. Considering the uncertainty factor and the time-varying characteristic, a linearized random ZIP model (constant impedance (Z), constant current (I), and constant power (P)) with time-varying characteristics was proposed. In order to improve the voltage quality of the voltage-sensitive loads in the day-here stage in an active distribution network (ADN), a linearized two-stage active and reactive power coordinated stochastic optimization model was established. The day-ahead active and reactive power coordination optimization was to smooth the large voltage fluctuation and develop a reserve plan to eliminate the unbalanced power caused by the prediction error in the day-here optimization. In the day-here real-time redispatch, the voltage was further improved by the continuous reactive power compensation device. Finally, the simulation results on the IEEE-33 bus system showed that the control strategy could better eliminate the unbalanced power caused by the prediction error and obviously improve the voltage of sensitive loads in the real-time stage on the premise of maintaining economic optimality.

Celem badań było porównanie wybranych składników odżywczych ziarna czterech odmian pszenżyta ozimego (‘Cultivo’, ‘Grenado’, ‘Benetto’, ‘Tarzan’) uprawianych w konwencjonalnym i ekologicznym systemie produkcji pochodzących z doświadczenia polowego w latach 2010–2011. Oceną objęto zawartość: suchej masy, białka ogółem, tłuszczu surowego, włókna surowego, popiołu surowego i BAW oraz frakcji włókna (NDF, ADF, ADL, HCEL, CEL), a także Ca, P, K i Na. W ziarnie pszenżyta z uprawy ekologicznej uzyskano średnio o 16% mniej białka ogółem w porównaniu z ziarnem pochodzącym z uprawy konwencjonalnej. Ziarno pochodzące z uprawy ekologicznej zawierało istotnie więcej tłuszczu, popiołu, BAW oraz ADF i ADL. System uprawy nie wpłynął istotnie na zawartość składników mineralnych w ziarnie pszenżyta. Ziarno odmiany ‘Cultivo’ charakteryzowało się największą zawartością białka, tłuszczu surowego, popiołu surowego, wszystkich frakcji włókna (z wyjątkiem ADL), a także fosforu i potasu w porównaniu z ziarnem pozostałych odmian.

La obesidad es una enfermedad inflamatoria donde la genética determina cierto nivel de riesgo. Aun cuando existen estudios que reportan asociación entre polimorfismos de FTO (fat-mass associated gene) y adiposidad, existe limitada evidencia en población infantil chilena.Objetivo: determinar la asociación entre el polimorfismo rs9939609 del FTO y marcadores de adiposidad en población infantil chilena.Pacientes y Método: Estudio de corte transversal incluyó 361 participantes (de 6 a 11 años; 50% niñas). Los datos clínicos y la recolección de muestras de sangre se realizaron entre marzo y junio de 2008. El polimorfismo SNP (rs9939609), del gen FTO, se determinó utilizando ADN genómico extraído de leucocitos, utilizando el Mini Kit QIAamp DNA Blood (Qiagen GmbH, Hilden, Alemania). Los marcadores de adiposidad estudiados fueron, índice de masa corporal (IMC), masa grasa, perímetro de cintura (PC) y razón cintura/talla, y se compararon ajustados por sexo, edad y estadío de Tanner. La asociación entre el polimorfismo estudiado y los marcadores de obesidad se realizó mediante análisis de regresión lineal.Resultados: Al ajustar los marcadores por sexo, edad y estadío de Tanner se observó una asociación significativa entre el polimorfismo e indicadores de adiposidad. Por cada copia extra del alelo de riesgo se encontró un aumento de 2,47 kg de peso corporal, (IC 19 95%: 1,39 -3,55); 1,06 kg/m2 de IMC, (IC 95%: 0,56-1,54); 2,55 cm de PC, (IC 95%: 1,26-3,85) y en 1,98 % de masa grasa, (IC 95%: 0,78-3,19). Al convertir los marcadores de adiposidad a z-score, la razón perímetro de cintura/talla arrojó la mayor asociación con el alelo de riesgo de FTO.Conclusión: Este estudio indica asociación entre el polimorfismo rs9939609 del gen FTO con marcadores de adiposidad general y central en población infantil en Chile.

Objetivo: analizar la complejidad de la expresión génica en tejido adiposo de genes asociados con obesidad, mediante simulación computacional con diferentes herramientas bioinformáticas. Métodos: después de una búsqueda bibliográfica en PubMed, se seleccionaron 37 genes asociados con obesidad con fold change mayor a 1,5. A partir del cálculo de valores de los z-score obtenidos de experimentos de micromatrices de ADN de muestras de tejido adiposo de personas obesas y de control, se construyó una red de interacción con el programa Cytoscape 3.2. La información detallada sobre las características genómicas de estos genes se extrajo de las bases de datos Genome Browser de la UCSC y del NCBI. Utilizando herramientas de análisis de multivariado, se hizo un análisis de componentes principales y uno de agrupación. Resultados: la red construida mostró que los genes con mayor número de interacciones fueron: 1) el factor nuclear respiratorio (NRF1), 2) el canal activado de potasio activado por calcio alfa 1 (KCNMA1) y 3) la sintasa de ácidos grasos (FASN). Los que tuvieron mayores valores de expresión fueron: 1) el factor de crecimiento endotelial vascular A (VEGFA), 2) la dioxigenasa dependiente de alfa-cetoglutarato (FTO) y 3) el regulador de crecimiento neuronal 1 (NEGR1). Las proteínas IL6, BDNF y HLC tuvieron los mayores valores de interacción con IL6R, NRF1 y ACACB, respetivamente. Las categorías ontológicas más importantes se relacionaron con procesos metabólicos de lipoproteínas, el ciclo de los ácidos tricarboxílicos, la activación de las MAP-quinasas y la cascada JNK. Conclusiones: en su conjunto los resultados obtenidos de sobreexpresión diferencial de genes asociados con el metabolismo de lípidos en el tejido adiposo de personas obesas podría ser un criterio para discriminar a nivel de diagnóstico esta patología.

Des anticorps anti adn-z interagissent fortement "in situ" avec des segments heterochromatiques de chromosomes de lebus (primate). Ces segments constitues d'un adn satellite dont l'unite est repetee (1500 bp) presentent deux sites sensibles a la nuclease s1. Cette unite repetee est inseree dans un plasmide. Pour etudier la conformation de ces regions en fonction des contraintes topologiques, des sondes chimiques (diethylpyrocarbonate, tetraoxyde d'osmium, hydroxylamine) et des anticorps anti adn-z ont ete utilises. L'unite repetee inversee et l'unite directe la plus proche adoptent une structure cruciforme (sequence palindromique). Des variations de sequence nucleotidique dans la region riche en (ac) semblent indiquer un site actif de recombinaison, a faible densite de supertours, la sequence (ac)::(n) est en conformation z. La conformation z sur plus de 100 paires de bases (bp) dans la region riche en acalccaca est obtenue par augmentation de le densite de supertours ou par interaction avec les anticorps anti adn-z

La méthylation de l'ADN est l’une des marques épigénétiques les plus étudiées et est largement conservée. Mes travaux de thèse présentent le premier méthylome d'une diatomée marine P. tricornutum qui appartient à la famille des Stramenopiles. P. tricornutum présente une méthylation d’environ 6% qui est présente en mosaïque sur l’ensemble du génome. Une méthylation importante a été retrouvé chez les éléments transposables, en particulier les éléments amplifiés récemment de type Copia. L’analyse met en évidence plus de 320 gènes méthylés dans trois contextes génomiques différents : à proximité des éléments transposables, en grappes de gènes méthylés, et dans des gènes uniques. En outre, les gènes largement et complètement méthylés ont été trouvé fortement corrélés avec le silencing transcriptionnel et l'expression différentielle dans des conditions spécifiques. Enfin, il a été constaté que les gènes susceptibles d’avoir été acquis par transfert horizontal de gènes bactériens étaient préférentiellement insérés dans des régions riches en éléments transposables, ce qui suggère un mécanisme par lequel l'expression de gènes étrangers peut être tamponnée à la suite de leur insertion dans le génome. En résumé, P. tricornutum a une faible méthylation de l'ADN et une methylation relativement importante des éléments transposables et seulement quelques gènes méthylés. Ce premier méthylome d’une diatomée Stramenopile ajoute de manière significative à notre compréhension de l'évolution de la méthylation de l'ADN chez les eucaryotes. En ce qui concerne les modifications des histones, la distribution des marques H3K4me2, H3K9me2 et H3K27me3 a été examinée chez P. tricornutum. H3K4me2 est principalement associée à des gènes alors que les deux marques H3K9me2 et H3K27me3 ciblent principalement des éléments transposables. La répartition de H3K27me3 est inhabituelle et différente de ce qui a été observé chez les espèces modèles étudiées à ce jour. Les gènes marqués par H3K27me3 ont tendance à être faiblement exprimés et de façon différentielle. H3K27me3 et H3K9me2 ont tendance à co-marquer non seulement les éléments transposables méthylés, mais aussi des gènes fortement méthylés, ce qui semble être important pour le maintien du silencing des gènes différentiellement exprimés. L'analyse combinatoire de différentes marques d’histones et la méthylation de l'ADN nous a donné un aperçu du paysage de la chromatine chez les diatomées, et aidera à définir les caractéristiques structurales et fonctionnelles conservées
DNA methylation is the most extensively studied and widely conserved epigenetic mark. Here the first whole genome methylome from a stramenopile, the marine model diatom P. tricornutum is reported. In P. tricornutum, around 6% of the genome was methylated in a mosaic landscape. Extensive methylation in transposable elements (TEs), especially in recently amplified Copia-like elements was found. Over 320 genes were found methylated occurring in three different genomic contexts: in the proximity of TEs, in clusters of methylated genes, and in single genes. Furthermore, genes extensively and completely methylated correlated strongly with transcriptional silencing and differential expression under specific conditions. Finally, it was found that genes likely acquired by horizontal gene transfer from bacteria were preferentially inserted within TE-rich regions, suggesting a mechanism whereby the expression of foreign genes can be buffered following their insertion in the genome. In general, P. tricornutum has low DNA methylation with relatively extensive DNA methylation on TEs and a few methylated genes. This first Stramenopile methylome adds significantly to our understanding of the evolution of DNA methylation in eukaryotes. As for the histone modifications, genome wide distribution of H3K4me2, H3K9me2 and H3K27me3 were examined in P. tricornutum. H3K4me2 is mainly associated with genes while both H3K9me2 and H3K27me3 marks target mainly transposable elements (TEs). The distribution of H3K27me3 is unusual and different from what have been profiled in model species so far. The genes marked by H3K27me3 tend to be lowly and differentially expressed. H3K27me3 and H3K9me2 tend to co-mark not only methylated TEs but also heavily methylated genes, which appears to be important for maintaining the silencing of differentially expressed genes. The combinatorial analysis of different histone marks and DNA methylation gave us an overview of diatom chromatin landscapes, and will help to define conserved structural and functional features

Le but de cette thèse est de décrire le chemin métabolique permettant de remplacer l’adénine (A) par la 2-aminoadénine (Z) dans le génome de bactériophages Siphoviridae. La 2-aminoadénine et la thymine (T) forment la paire Z:T, liée par trois liaisons hydrogène. Avec la paire G:C classique, ils forment un ADN « ZTGC » qui est résistant aux enzymes de réstriction de l’hôte. En premier lieu, mes travaux se sont focalisés sur le cyanophage S-2L, décrit comme porteur de 2-aminoadénine. J’ai d’abord étudié la primase-polymérase, PrimPol, responsable de la réplication de l’ADN chez ces phages, et qui possède la capacité surprenante d’incorporer à la fois le dATP et le dZTP. J’ai ensuite caractérisé une triphosphatase dATP-spécifique, appelée DatZ, conservée chez tous les bactériophages Siphoviridae, responsable de la dégradation spécifique du dATP. J’ai également mis en évidence que PurZ, l’enzyme-clé pour la production de la diaminopurine est non seulement une ATPase mais aussi une dATPase. J’ai identifié une nucléotide pyrophosphatase, appelée MazZ, composant essentiel du chemin de biosynthèse de Z, qui convertit dGTP en dGMP, en générant ainsi un des substrats de PurZ. Structures cristallographiques de haute résolution de tous les 4 enzymes avec leurs ligands respectifs expliquent les spécificités observées dans les tests catalytiques – ou leur absence. Enfin, j’ai caractérisé une structure d’une ADN polymérase Z-spécifique de famille A, PolZ, trouvée dans le vibriophage φVC8, mais absente dans S-2L. J’ai résolu sa structure cristallographique en modes polymérase et exonuléase « couplé-ouvert » et « couplé-fermé », permettant de decrire son activité au niveau atomique
The subject for this thesis is to dissect the enzymatic pathway allowing a non-canonical base 2-aminoadenine, or diaminopurine (Z) to replace adenine (A) in the genomes of a number of Siphoviridae bacteriophages. 2-aminoadenine and thymine (T) form the Z:T pair bound by fully saturated triple hydrogen bond. Together with the standard G:C pair they form ZTGC-DNA which is resistant to host’s restriction enzymes. I first focus on cyanophage S-2L, the originally-described bearer of 2-aminoadenine. I identify a DNA primase-polymerase, PrimPol, responsible for its replication, with surprisingly similar activity towards dATP and dZTP. This prompted the characterization of a dATP-specific triphosphatase, DatZ. Its activity and conservation between the phages of interest explains the mechanism behind adenine removal. Secondly, I find that PurZ of phage S-2L’s, a key enzyme in diaminopurine production, is not only an ATPase but also a dATPase. I identify a nucleotide pyrophosphatase, MazZ, as an essential component of the conserved Z biosynthetic pathway, that converts dGTP into dGMP, thus generating one of the substrates of PurZ. High resolution crystallographic structures of all 4 enzymes with their respective ligands explain the specificities observed in catalytic tests - or lack thereof. Finally, I characterized the structure of a Z-specific family A DNA polymerase, PolZ, found in a related vibriophage φVC8 but absent in S-2L. Its crystallographic structure in polymerase-exonuclease “coupled-open” and “coupled-close” states offers an explanation for the observed specificity

Le développement d'un organisme complexe repose sur une séquence de décisions cellulaires conduisant à l'émergence de centaines de types cellulaires différents avec des fonctions spécifiques. Cette progression est marquée par une perte graduelle de la potence cellulaire, des changements significatifs du transcriptome et un remodelage étendu de la chromatine. Les embryons totipotents à 2 cellules (2C) se caractérisent par une signature transcriptionnelle unique et une organisation de la chromatine relâchée (hypométhylation globale, faible compaction de la chromatine, accessibilité accrue de la chromatine). Cependant, comment les embryons 2C préservent leur identité cellulaire stricte dans une conformation de chromatine permissive reste mal compris.La conversion spontanée des cellules souches embryonnaires de souris (mESCs) à un état semblable à celui des embryons 2C, appelé cellules 2C-like (2CLCs), fournit un modèle in vitro idéal pour étudier les caractéristiques de la totipotence, dont elles partagent plusieurs traits avec les embryons 2C. Pour étudier les interactions de la chromatine à l'échelle du génome, des expériences de capture de conformation des chromosomes (Hi-C) ont été menées dans les mESCs et les 2CLCs. Bien que l'architecture globale des chromosomes soit restée globalement stable entre les deux populations cellulaires, nous avons identifié de nouvelles larges régions d'interaction spécifiques aux 2CLCs, principalement situées vers une extrémité de nombreux chromosomes. Des expériences de ADN-FISH ont montré que ces interactions se localisent à la périphérie nucléaire des 2CLCs, associées à un changement de marque d'histone, passant d'actif dans les ESCs à répressif dans les 2CLCs, empêchant l'activation transcriptionnelle dans ces régions. La formation de ces interactions de la chromatine dépend des protéines de la famille Zscan4, un facteur de la chromatine exprimé spécifiquement au stade 2C. De manière intrigante, il a été démontré que Zscan4 se lie à des motifs prédisposés à adopter une conformation en Z-ADN, caractérisée par une double hélice gauche. Nous avons détecté la présence de Z-ADN dans les 2CLCs, et leur induction a significativement augmenté la proportion de 2CLCs affichant des conformations de chromatine similaires à celles des 2CLCs spontanés.En résumé, nous proposons un mécanisme en deux étapes dans lequel la formation de Z-ADN induit l'émergence de 2CLCs, suivie de leurs relocalisations à l'enveloppe nucléaire formant de grands domaines d'interactions dépendants de Zscan4, qui pourraient être cruciaux pour préserver la totipotence dans un environnement de chromatine globalement permissif
Mammalian development is characterized by a sequence of cell fate decisionsalong an irreversible pathway of restricted developmental potential and increasingcell specialization. With progressing development, a gradual restriction inpotency is accompanied by significant transcriptome modulation and drasticchromatin reprogramming. Totipotent 2-cell (2C) embryos are defined by aunique transcriptional signature and a relaxed chromatin organization (globalhypomethylation, poor chromatin organization and increased chromatinaccessibility). However, how 2C embryos safeguard their strict cell identity in arelaxed chromatin conformation remains poorly understood.The spontaneous conversion of mouse embryonic stem cells (mESCs) to a 2C-like state, called 2C like cells (2CLCs), provides a convenient in vitro modelsystem to study totipotent-like characteristics as they share several features with2C embryos. To investigate genome-wide chromatin interactions, chromosomeconformation capture experiments (Hi-C) were conducted in both mESCs and2CLCs. While the global chromosome architecture remained stable between thetwo cell populations, we identified new large interacting regions specific to2CLCs, located towards one end of numerous chromosomes. The formation ofthese chromatin interactions depends on Zscan4, a transcription factor expressedspecifically at the 2C stage. Intriguingly, Zscan4 binds to motifs predisposed toadopt a Z-DNA conformation, characterized by a left-handed double helix. Wedetected the presence of Z-DNA in 2CLCs, and inducing it significantly increasedthe proportion of 2CLCs displaying chromatin conformation akin to spontaneous2CLCs. Mechanistically, we propose that Z-DNA formation plays a role inaltering DNA replication timing, a process known to promote cell transitions.In summary, we propose a novel role for Zscan4 in forming 3D genomeinteraction, a process that may be critical in establishing and maintainingtotipotency

The subject of this thesis is to compare two specific methods of the determination of the tax base; the "specific" method and the "ad valorem" method. Generally, the specific method uses the acrage of a real estate for the determination of the tax base. On the other hand the ad valorem method uses the market value of the real estate for the determination of the tax base. Using various sources I have analyzed the tax systems in two countries using the above mentioned methods: the Czech Republic representing the specific method and Italy representing the "ad valorem" method. The thesis is focused on the differences between the two taxation methods thereof. It demonstrates the differences by the practical application of the methods to the taxation of two real estates in each country.

A. Fumigatus a été exposé à 91 mélanges d’amphothericine B, micafungine et nykkomycine Z. Le meilleur modèle est un modèle global de surface de réponse avec des équations polynomièles pour m et CI50. On voit une large région de synergie, sur le bord micafungine et nykkomycine Z (proportions normalisées équivalentes), et vers le milieu de la figure. On a simulé différents profiles de m et CI50 en variant systématiquement les paramètres des polynomes. Pour CI50, on a pu simuler des isoboles assymétriques et irrégulières, mais passant au maximum 2 fois la ligne d’additivité. Enfin, l’évolution dans le temps et la relation concentration-effet pour des données de microarrays ont ete fittés simultanément, avec comme sous-modèles respectifs un modèle de croissance / décroissance exponentiel et un modèle de Hill. Les données étaient une base de résultats Affymetrix HG-U95Av2 de 51 arrays, aprés exposition au cisplatine ou à l’oxaliplatine de cellules de carcinome ovarien humain A2780. 18 gènes ont été sélectionés. Une comparaison des paramètres permet de voir les gènes présentant des profiles différents
A. Fumigatus was exposed to 91 fixed ratio combinations of amphotericin B, micafungin, and nikkomycin Z. Effect was assessed with an XTT assay. The best model was an overall response surface including polynomial equations for m and CI50. There was a large zone of synergy, mostly at the nikkomycin Z / micafungin edge and into the center of the ternary plots. The effect of changing the values of the different polynomial parameters for m and CI50 was systematically simulated. For CI50, asymmetrical and irregular isobols could be simulated, and the additivity line could be crossed twice at maximum. Finally, time-course and concentration-effect of gene expressions from microarray studies were fitted simultaneously, the respective submodels being the exponential increase / decrease model and the Hill model. The dataset was an Affymetrix HG-U95Av2 51 arrays dataset, after exposure of A2780 human ovarian carcinoma cells to cisplatin and oxaliplatin, focusing on 18 selected genes. Comparisons of model parameters helped distinguish genes with various expression patterns and could also help in understanding the molecular mechanisms of the agents, and their timing

AbstractIn recent years, fungi have attracted avid interest from the research community. This interest stems from several motives, including their network creation capabilities and fundamental role in the ecosystem. Controlled laboratory experiments of fungal behaviors are crucial to further understanding their role and functionalities.In this paper, we propose a method for automating the quantification and observation of fungal spores. Our approach consists of four steps: 1) a Z-stack image acquisition of the sample is performed, 2) a detection algorithm is applied to all Z-planes, 3) clustering of spores detected in different Z-planes, 4) determination of the optimal Z-plane for each cluster through an ad-hoc focus measure. We compared the spore count obtained through the automated tool to a manual count and the count obtained by applying the detection algorithm to a single plane. The result is a highly automated, non-invasive tool to determine spore count, estimate each spore depth, and retrieve an all-in-focus image to analyze further.

A tanulmány alapjául egy 2023. június 13-án megrendezett esemény szolgált, amelyet a NAV Kelet-budapesti Adó- és Vámigazgatósága (a továbbiakban: Igazgatóság) rendezett, a 30 év alatti, többnyire Z generációhoz tartozó foglalkoztatottjainak meghívásával. A szerző az ott elhangzottakat, az ott végzett kérdőíves felmérés és fókuszcsoportos interjú eredményeit foglalja össze e tanulmányban. Rámutat, hogy e generáció képviselői mitől mások, mint a megelőző generációba tartozók, milyen munkahelyi preferenciákkal rendelkeznek. A Z generációt vizsgáló korábbi kutatási eredményeket alátámasztották az Igazgatóság által szerzett tapasztalatok is: a Z generációs fiatalok számára fontos a rugalmasság, anyagi megbecsültség és az előrelépési lehetőség, sikerorientáltság. A fiatal kollégák pozitívan értékelték az Igazgatóságon folyó mentor program gyakorlatát, a közösségbe való beilleszkedést és a munkahely kapcsán érzett stabilitást is. Számos javaslatot fogalmaztak meg a szervezetfejlesztéssel és a munkaerő megtartásával kapcsolatban. Bár néhány kívánalom túlmutat az Igazgatóság hatáskörén, azonban az e tanulmányban bemutatott érvek és jövőre vonatkozó gondolatok tanulságosak lehetnek mindenki számára, akik a Z generációs fiatalok gondolkodásmódját, értékeit kívánja megismerni.

Portal monitor radiasi (RPM) karena proses sertifikasi harus diuji sesuai SNI IEC 62244:2016. Standar ini memerlukan beberapa pengujian untuk diterapkan seperti uji mekanik yang dilakukan dengan uji getaran. Persyaratan yang harus dipenuhi adalah tidak ada alarm atau perubahan lain yang terjadi selama proses indikasi monitor yang bervariasi tidak lebih dari 15%, dan tidak ada kerusakan pada komponen monitor. RPM diuji dalam kondisi operasi dan terkena sumber radiasi. Sumber radioaktif diatur 50 cm dari titik referensi. Untuk uji frekuensi, RPM diberi beban harmonik 0,5 gn selama 10 menit dengan frekuensi yang dinaikkan secara bertahap dari 10 Hz menjadi 150 Hz kemudian menurun dari 150 Hz menjadi 10 Hz pada masing-masing arah ortogonal (x,y, dan z). Ketahanan selama pengiriman diuji dengan memberikan beban harmonik sebesar 2 gn selama 15 menit dalam tiga arah ortogonal (x,y, dan z). Frekuensi untuk setiap rentang adalah 10 Hz hingga 21 Hz, dan 22 Hz hingga 33 Hz. Hasil dari semua pengujian menunjukkan bahwa RPM memenuhi semua persyaratan.

A közszféra szervezetei – köztük a Nemzeti Adó- és Vámhivatal (NAV) is – komoly kihívásokkal néznek szembe a toborzás és a munkaerő megtartása terén. Folyamatosan nő a megüresedő álláshelyek száma, viszont ezzel párhuzamosan csökken a közszféra, mint munkahely iránti érdeklődés, főleg a fiatalok, a Z generációhoz tartozók körében. Mindez a NAV-on belül a pénzügyőri szolgálat ellátását is nehezíti, a pénzügyőri hivatás is utánpótlás-problémákkal küzd. Ez a tanulmány ennek körülményeit igyekszik körbejárni.

Con base a la temática del Módulo de “Diversidad Generacional”, y el interés en la investigación formativa en el aula, se utilizaron métodos, técnicas y herramientas, que llevaron a los estudiantes a la autopercepción de identidad generacional, para develar los rasgos identitarios y comportamientos de estos estudiantes con la finalidad de generar un Manifiesto Generacional y dejar documentada la experiencia, empleando el método de sistematización de experiencias a partir de la construcción colaborativa en el aula, con enfoque cualitativo con análisis de contenido y la técnica de triangulación de fuentes: actores, bibliografía y observación docente. El tipo de investigación es descriptivo, ya que buscó describir y caracterizar los rasgos identitarios y comportamientos de la generación Z en los estudiantes del curso de Gestión del Cambio. Para la experiencia se utilizaron diferentes técnicas como: Observación directa, trabajo colaborativo, debate, focus group, técnica Ikigai, revisión documental, narrativa testimonial, entrevista semiestructurada. Los instrumentos empleados fueron: Matrices de registros, Bitácora de la investigación, Guía de entrevista semiestructurada. La triangulación de fuentes reveló las coincidencias con la declaración del Manifiesto Generacional y su Plan de Acción para la concreción de este, develando los rasgos identitarios y comportamientos de la generación z en estudiantes universitarios del curso de Gestión del Cambio en licenciaturas de ADEN University Campus Panamá.

Acoustic Doppler Velocimeter (ADV) is a useful technique for measuring flow velocities with frequency variations of up to approximately 200 Hz in laboratory settings and in field applications. Although measuring velocity with ADV has advantages over other velocity measurement methods, this technique is sensitive to operating conditions: in addition to noise, the signal can contain spikes with large amplitudes, a disadvantage of ADV. In this study, the effect of bubbles on ADV signals is experimentally assessed in a laboratory setting. Bubbles can intersect the sampling volume and the acoustic beams creating spikes. The impact and amplitude of these spikes is a function of the bubble size and position when it crosses the ADV sampling volume and the acoustic beams. Bubbles that intersect the sampling volume generate spikes in all three velocity directions simultaneously; bubbles that intersect acoustic beams, which span between the sampling volume and the ADV receivers, impact the velocity data in one or two directions, and has a negligible effect in the third direction. Bubbles that intersect the X-direction acoustic beam create spikes in velocity data in both X- and Z-directions, but have no significant impact on the Y-direction; the Y- and X-directions have spikes and the Z-direction is not significantly impacted, when bubbles intersect the Y-direction acoustic beam. In addition, spikes increase the magnitude of the power spectra at high frequencies. Without bubbles, the autocorrelation in the time domain decreases in value as the time-lag increases, approaching zero after 5 seconds. The presences of bubbles cause a large peak in the autocorrelation at a zero time-lag, and no autocorrelation thereafter. Furthermore, the autocorrelation without bubbles permit turbulence length scales to be calculated because of the positive autocorrelation value; unless spikes are removed by using an appropriate filter when bubbles are present, turbulence length scales cannot be calculated because the autocorrelation is zero.

We put forward novel schemes wherein lasing is attained by coherent superpositions of two free electronic states in interfering interaction regions that suppress stimulated absorption without hampering stimulated emission. This can strongly enhance the gain of a free-electron laser. In the first scheme an electron beam with mean momentum k → 1 interacts with wiggler mode k → w 1 in region 1, changes its momentum (by deflection) to k → 2 and interacts with wiggler mode k → w 2 in region 2. The interactions in both regions add coherently, provided both wiggler beams are from the same coherent source, and the signal photon is delayed to allow interaction with the same electron in both regions. Absorption of the signal photon in regions 1 and 2 scatters the electron into a common final state | k → a > if k → a ≈ k → 1 − k → w 1 + q → = k → 2 − k → w 2 + q → . This is the condition for interference that can suppress absorption. The final states for emission, | k → e 1 > and | k → e 2 are then strongly orthogonal, whence there is no interference in emission. In another scheme, the wigglers are replaced by Cherenkov or Smith–Purcell effects. Coherently superposed electron momenta k → 1 and k → 2 propagate in two sequential structures with different dispersion relations. Interference in signal absorption at frequency ω and direction z by both structures occurs for a common final electronic state | k → a > satisfying k → a z ≃ k → 1 z + q 1 z ( ω ) ≃ k → 2 z + q 2 z ( ω ) , where q → 1 ( ω ) and q → 2 ( ω ) are the respective wavevectors. Here too interference in emission is prevented by the orthogonality of the respective final states k → e i = k → i − q → i ( ω ) , ( i = 1 , 2 ) .

A fundamental challenge for engineers when applying the MBSE (Model-based Systems Engineering) is to analyze complex design problems in an effective way and to represent a complex system with complete models. Aiming at providing an efficient design problem analyzing process and general modeling framework, this paper proposed an Axiomatic Design Theory (ADT) based System Engineering approach by mapping design domains, design axioms and a “Z” mapping process of ADT to complex system’s design processes, modeling and control processes and information loops respectively. Then a MBSE modeling framework that includes the Requirement Model, Functional Architecture and Physical Implementation Architecture was constructed. The presented method and modeling framework is able to guide system engineers to capture design problems effectively. After that, a case study was illustrated to demonstrate how the presented approach was applied. Last, the paper gave conclusions and future works that show lights on improving the MBSE modeling framework to function in a consistent way.

Abstract To precisely predict the fluctuation of wellbore properties, local overheating and guide the drilling trajectory in the wellbore during drilling operations, it is preferable to use cost-effective drilling microchips to collect real-time downhole data measurements while drilling. The drilling microchips usually consist of SoC, add-on memory, sensors and on-chip battery. However, the microchips can be subjected to mechanical shocks during the release and drilling operations which can induce interconnects and components failure and eventually leads to damage, loss of stored data and a low recovery rate. As the efficient performance of the drilling microchips can be constrained due to the shock loads, hence, it is essential to understand the peak response of the device when subjected to such suddenly applied stresses or designed response spectrum. The development and validation of simulation modeling approaches is the initial step in this direction. This paper will present the effects of varied shock loads and duration on the drilling microchips using two methods: response spectrum analysis and direct time stepping through mode superposition. It can be deduced that the sum of the effective modal masses for all computed eigenmodes is around 0.8947, 0.8912 and 0.9076 in z, y and x directions. Thus, these modes account for 89.47%, 89.12% and 90.76% of the dynamic forces in the z-direction, y-direction and x-direction respectively.

Abstract Reverse engineering of electronic hardware has been performed for decades for two broad purposes: (1) honest and legal means for failure analysis and trust verification; and (2) dishonest and illegal means of cloning, counterfeiting, and development of attacks on hardware to gain competitive edge in a market. Destructive methods have been typically considered most effective to reverse engineer Printed Circuit Boards (PCBs) – a platform used in nearly all electronic systems to mechanically support and electrically connect all hardware components. However, the advent of advanced characterization and imaging tools such as X-ray tomography has shifted the reverse engineering of electronics toward non-destructive methods. These methods considerably lower the associated time and cost to reverse engineer a complex multi-layer PCB. In this paper, we introduce a new anti–reverse engineering method to protect PCBs from non-destructive reverse engineering. We add high-Z materials inside PCBs and develop advanced layout algorithms, which create inevitable imaging artifacts during tomography, thereby making it practically infeasible for an adversary to extract correct design information with X-ray tomography.
