Abstract (sommario):

Abscisic acid (ABA) plays crucial roles in plant development and adaption to environmental stresses. The ABA-responsive element binding protein/ABRE-binding factor and ABA INSENSITIVE 5 (AREB/ABF/ABI5) gene subfamily members, which belong to the basic domain/leucine zipper (bZIP) transcription factors family, participate in the ABA-mediated signaling pathway by regulating the expression of their target genes. However, information about potato (Solanum tuberosum) AREB/ABF/ABI5 subfamily members remains scarce. Here, seven putative AREB/ABF/ABI5 members were identified in the potato genome. Sequences alignment revealed that these members shared high protein sequence similarity, especially in the bZIP region, indicating that they might possess overlapping roles in regulating gene expression. Subcellular localization analysis illustrated that all seven AREB/ABF/ABI5 members were localized in the nucleus. Transactivation activity assays in yeast demonstrated that these AREB/ABF/ABI5 members possessed distinct transcriptional activity. Electrophoretic mobility shift assays (EMSA) confirmed that all of these AREB/ABF/ABI5 members could have an affinity to ABRE in vitro. The expression patterns of these AREB/ABF/ABI5 genes showed that they were in response to ABA or osmotic stresses in varying degrees. Moreover, most AREB/ABF/ABI5 genes were induced during stolon swelling. Overall, these results provide the first comprehensive identification of the potato AREB/ABF/ABI5 subfamily and would facilitate further functional characterization of these subfamily members in future work.

Abstract (sommario):

The abscisic acid (ABA)-responsive element binding protein/ABRE-binding factor (AREB/ABF) subfamily members are essential to ABA signaling pathways and plant adaptation to various environmental stresses. Nevertheless, there are no reports on AREB/ABF in jute (Corchorus L.). Here, eight AREB/ABF genes were identified in the C. olitorius genome and classified into four groups (A–D) based on their phylogenetic relationships. A cis-elements analysis showed that CoABFs were widely involved in hormone response elements, followed by light and stress responses. Furthermore, the ABRE response element was involved in four CoABFs, playing an essential role in the ABA reaction. A genetic evolutionary analysis indicated that clear purification selection affects jute CoABFs and demonstrated that the divergence time was more ancient in cotton than in cacao. A quantitative real-time PCR revealed that the expression levels of CoABFs were upregulated and downregulated under ABA treatment, indicating that CoABF3 and CoABF7 are positively correlated with ABA concentration. Moreover, CoABF3 and CoABF7 were significantly upregulated in response to salt and drought stress, especially with the application of exogenous ABA, which showed higher intensities. These findings provide a complete analysis of the jute AREB/ABF gene family, which could be valuable for creating novel jute germplasms with a high resistance to abiotic stresses.

ABSTRACT Helminth infections are among the most potent stimulators of Th2-type immune responses and have been widely demonstrated to modify responsiveness to both nonparasite antigens and other infectious agents in a nonspecific manner in infected animals. We investigated the immunomodulatory properties of pseudocoelomic body fluid from adult Ascaris suum gastrointestinal helminths (ABF) and its defined allergen (ABA-1) by examining their effects on the immune response to a heterologous antigen, ovalbumin. Our results indicate that ABF has potent immunomodulatory activity and that the effects observed are consistent with skewing towards a Th2-type response rather than induction of anergy. Our findings show that the immunomodulatory activities of ABF are associated with components other than the major constituent and putative allergen, ABA-1. Furthermore, the allergic responses to ABA-1 are not a result of an intrinsic allergenicity of the protein but are more a reflection of the wider induction of a Th2 response by the infection. Importantly, the induction of interleukin-10 by ABF also suggests that T regulatory cells may play a role in immunomodulation of immune responses by parasitic helminths.

Rosaceae is an important family containing some of the highly evolved fruit and ornamental plants. Abiotic stress responses play key roles in the seasonal growth and development of plants. However, the molecular basis of stress responses remains largely unknown in Rosaceae. Abscisic acid (ABA) is a stress hormone involving abiotic stress response pathways. The ABRE-binding factor/ABA-responsive element-binding protein (ABF/AREB) is a subfamily of the basic domain/leucine zipper (bZIP) transcription factor family. It plays an important role in the ABA-mediated signaling pathway. Here, we analyzed the ABF/AREB subfamily genes in nine Rosaceae species. A total of 64 ABF/AREB genes were identified, including 18, 28, and 18 genes in the Rosoideae, Amygdaloideae, and Maloideae traditional subfamilies, respectively. The evolutionary relationship of the ABF/AREB subfamily genes was studied through the phylogenetic analysis, the gene structure and conserved motif composition, Ka/Ks values, and interspecies colinearity. These gene sets were clustered into four groups. In the Prunus ABF/AREB (PmABF) promoters, several cis-elements related to light, hormone, and abiotic stress response were predicted. PmABFs expressed in five different tissues, except PmABF5, which expressed only in buds. In the dormancy stages, PmABF1, 2, 5 and 7 showed differential expression. The expression of PmABF3, 4 and 6 was positively correlated with the ABA concentration. Except for PmABF5, all the PmABFs were sensitive to ABA. Several ABRE elements were contained in the promoters of PmABF1, 3, 6, 7. Based on the findings of our study, we speculate that PmABFs may play a role in flower bud dormancy in P. mume.

Basic leucine zipper (bZIP) transcription factors of the ABA-responsive element binding factor/ABA-responsive element binding proteins (ABF/AREB) subfamily have been implicated in abscisic acid (ABA) and abiotic stress responses in plants. However, the specific function of ABF/AREB transcription factors under saline–alkaline stress is unclear. Here, we identified four ABF/AREB transcription factors in tomato and found that SlAREB1 strongly responded to both ABA and saline–alkaline stress. To further explore the function of SlAREB1 under saline–alkaline stress, SlAREB1-overexpressing lines were constructed. Compared with wild-type plants, SlAREB1-overexpressing transgenic tomato plants showed reduced malondialdehyde content, increased the relative water content, and alleviated the degradation of chlorophyll under saline–alkaline stress. Importantly, SlAREB1 directly physically interacted with SlMn-SOD, which improved the activity of antioxidant enzymes and increased the scavenging of excess reactive oxygen species. Overall, the overexpression of SlAREB1 increased the antioxidant capacity of the transgenic tomato under saline–alkaline stress.

Abstract Previous studies have established that the mitochondrial high mobility group (HMG) protein, Abf2p, of Saccharomyces cerevisiae influences the stability of wild-type (ρ+) mitochondrial DNA (mtDNA) and plays an important role in mtDNA organization. Here we report new functions for Abf2p in mtDNA transactions. We find that in homozygous Δabf2 crosses, the pattern of sorting of mtDNA and mitochondrial matrix protein is altered, and mtDNA recombination is suppressed relative to homozygous ABF2 crosses. Although Abf 2p is known to be required for the maintenance of mtDNA in ρ+ cells growing on rich dextrose medium, we find that it is not required for the maintenance of mtDNA in ρ− cells grown on the same medium. The content of both ρ+ and ρ− mtDNAs is increased in cells by 50–150% by moderate (two- to threefold) increases in the ABF2 copy number, suggesting that Abf2p plays a role in mtDNA copy control. Overproduction of Abf 2p by ≥10-fold from an ABF2 gene placed under control of the GAL1 promoter, however, leads to a rapid loss of ρ+ mtDNA and a quantitative conversion of ρ+ cells to petites within two to four generations after a shift of the culture from glucose to galactose medium. Overexpression of Abf2p in ρ− cells also leads to a loss of mtDNA, but at a slower rate than was observed for ρ+ cells. The mtDNA instability phenotype is related to the DNA-binding properties of Abf 2p because a mutant Abf 2p that contains mutations in residues of both HMG box domains known to affect DNA binding in vitro, and that binds poorly to mtDNA in vivo, complements Δabf2 cells only weakly and greatly lessens the effect of overproduction on mtDNA instability. In vivo binding was assessed by colocalization to mtDNA of fusions between mutant or wild-type Abf 2p and green fluorescent protein. These findings are discussed in the context of a model relating mtDNA copy number control and stability to mtDNA recombination.

Two genes encoding the ASABF (Ascarissuumantibacterial factor)-type antimicrobial peptide, abf-1 and abf-2, were identified in Caenorhabditis elegans. Recombinant ABF-2 exhibited potent microbicidal activity against Gram-positive and Gram-negative bacteria, and yeasts. The tissue-specific distribution estimated by immunofluorescence staining and transgenic analysis of a gfp fusion gene (where GFP corresponds to green fluorescent protein) suggested that ABF-2 contributes to surface defence in the pharynx. abf-1 contains a single intron at a conserved position, suggesting that asabf and abf originated from a common ancestor. Both transcripts for abf-1 and abf-2 were detected as two distinct forms, i.e. spliced leader (SL)1-trans-spliced with a long 5′-untranslated region (UTR) and SL-less with a short 5′-UTR. A polycistronic precursor RNA encoding ABF-1 and ABF-2 was detected, suggesting that these genes form an operon. An ‘opportunistic operon’ model for regulation of abf genes, including the generation of short SL-less transcripts, is proposed. In conclusion, C. elegans should have an immune defence system due to the antimicrobial peptides. C. elegans can be a novel model for innate immunity. Furthermore, the combination of biochemical identification in Ascaris suum and homologue hunting in C. elegans should be a powerful method of finding rapidly evolved proteins, such as some immune-related molecules in C. elegans.

We measured forearm blood flow (ABF) bilaterally on six subjects during 15-min periods of leg exercise and the first 10 min of recovery. One forearm (control) was kept at about 33 degrees C skin temperature in all experiments. In experiments at ambient temperature (Ta) of 15 degrees C, the other arm (experimental) was kept at about 26, 33, and 40 degrees C, respectively, during three successive cycles of exercise and recovery. ABF in the 26 degrees C forearm was linearly related to and averaged 42% of control. The relation of ABF in the 40 degrees C forearm to control ABF showed a bend at control ABF of 4–5 ml X 100 ml-1 X min-1. Below the bend, experimental ABF average 213% of control. Above the bend, experimental ABF averaged 5.09 ml X 100 ml-1 X min-1 above control. In four subjects, after heating the experimental forearm to 40 degrees C, we measured ABF for 25–30 min at rest in Ta of both 15 and 25 degrees C. At 25 degrees C Ta, ABF in the heated forearms rose gradually, but control ABF showed little change. At 15 degrees C Ta, the effect on ABF of local heating to 40 degrees C was much reduced, apparently due to reflex vasoconstrictor signals.

Physiol. 84(1): 82–89, 1998.—Adrenal gland blood flow (ABF) increases during hypoxemia in fetal sheep, but regulation of ABF is poorly understood. The purpose of this study was to determine the effects of splanchnic nerve section on fetal ABF responses to hypoxemia using the fluorescent microsphere (FM) technique. At 125 days of gestation, 14 unanesthetized fetal sheep [bilateral splanchnicotomy (Splx, n = 6) and control (Cont, n = 8)] were injected with FM before and at 60 min of N2-induced hypoxemia (∼40% decrease in fetal arterial [Formula: see text]). Adrenal tissue and reference blood samples were digested and filtered, and FM dye was extracted for spectrometer analysis. Baseline whole, medullary, and cortical ABF for the Cont group were similar to published values using radioactive microspheres and did not differ from Splx values. Hypoxemia increased whole, medullary, and cortical ABF (mean ± SE) from baseline for the Cont group by 281 ± 35, 258 ± 31, and 496 ± 81% ( P < 0.05). The increase for the Splx group was attenuated compared with the Cont group ( P < 0.05) for whole and medullary ABF (139 ± 27 and 43 ± 27%) but not cortical ABF (326 ± 91%). We conclude that 1) the FM technique is valid for measuring fetal ABF and 2) in fetal sheep the splanchnic nerve is not necessary to maintain basal ABF but plays an important role in regulating the hypoxemia-induced increase in ABF through the medullary, but not cortical, ABF response.

-1949 [26][upps-01.gif] The aim of this master thesis is to investigate how W.E.A. Workers Educational Association, took part in the great changes of society and in the modernizing process in the period of 1920-1949. The examination is based upon three journals of educational material that were connected with W.E.A. The question at issue is to show how the people behind W.E.A. aimed to educate and indeed educated the workers to modern people and moreso to see how the articles in fact were written and to analyse in what way they turn to the workers and get their attention. I also want to examine if it is possible to observe a change in the upbringing-process through the time period and if the focus was altered in that very process. I have divided the articles into three parts: one moral, one physical and one educational which in their turn are subdivided on the basis of how the articles are written, not accordingly to their subject. The essay also contains a short review of the Swedish political history and what the modernisation in Sweden looked like between the 1880s and 1950s. Furthermore, I have studied the establishment of the W.E.A. as well as its development and cultural point of view. The results indicate that the W.E.A. intended to give a total education - the inner side through opinions, knowledge and moral, the outside through gymnastics, personal in addition to public hygiene. In these articles I have distinguished between three different ways in which the W.E.A. intended to educate. Firstly, a stupefying manner that manifested to what extent people ought to dislike certain phenomena. Secondly, a transmitting of opinions and values already accepted in the society and finally, the viewpoint that educating was to speak well about certain phenomena.

ABF -1 is a human class II bHLH transcription factor that is expressed predominantly in activated B cells and EBV immortalized cell lines. A portion of this study sought to characterize the homolog of ABF- l in Caenorhabditis e/egans. The nematode gene product, ceABF -1, is capable of forming heterodimers with E2A gene products and binding E box binding sites. HeLa cells transfected with ceABF-1 reveal that it is capable of blocking E2A mediated gene transcription. In order to maintain full repression capabilities, two conserved amino acid residues within helix I ofthe HLH domain are required. These results show a conserved mechanism of gene repression between invertebrates and vertebrates. This study also sought to analyze ABF-1 mediated regulation of both ld3 and cellular growth. Using a human ABF-1 stably transfected cell line, ID3 protein levels and transcript levels were shown to increase in response to overexpression of ABF-1 via western and northern blot, respectively. Flow cytometry analysis and Real-time PCR revealed that ABF-1 programs a slow down in the cell cycle, however this growth arrest is not mediated by ID3.

Abstract (sommario):

The basic helix-loop-helix (bHLH) family oftranscription factors consists of proteins involved in cellular proliferation and differentiation. The HLH structure plays a key role in protein-protein dimerization and with the DNA target sites, referred to as E boxes containing the consensus DNA sequence CANNTG. One class of mammalian class I bHLH proteins includes products of the E2A gene, which result from alternative splicing (E12, E47, and ITF), E2-2 and HEB. E2A proteins have also been detected in most cell lines with high levels of expression in lymphoid- and pancreatic cells. It has also been demonstrated that E2A is required for B cell maturation, T cell development and has been shown to function as tumor suppressors. To date, an E2A-interacting bHLH transcription factor largely restricted to activated B lymphocytes, called ABF -1, was isolated using the two-hybrid system. ABF -1 is the only B cell restricted bHLH protein isolated. ABF-1/E2A heterodimers have been detected in B lymphocytes. In these studies, the mapping of the ABF-1 promoter and the critical 5' regulatory elements that control ABF-1 gene expression were analyzed through 5' deletional analysis. 5' -DNA flanking pieces of the promoter region were created through PCR and inserted into a promoterless cloning vector containing the firefly luciferase reporter gene. RT -PCR analysis and anchored PCR was utilized to demonstrate the transcriptional activity of the - promoter region of the ABF-1 gene. Transient transfections were completed to determine critical regulatory sequences. The promoter location was confirmed through computer analysis of the nucleotide sequence and deletional analysis.

Abstract (sommario):

The human ABF-1 gene is expressed in activated B-cells and Epstein-Barr virus immortalized lymphoblastoid cell lines. ABF-1 represents the only member belonging to the basic helix-loop-helix (bHLH) family of transcription factors whose expression pattern is restricted to B cells. ABF-1 forms heterodirner complexes with E2A to modulate gene transcription. Here we report the isolation often novel ABF-1 splice products from a human B-cell plasmacytoma cell line. Analysis of the nucleotide sequence revealed approximately five clones harboring changes within the coding region of the reported full-length ABF-1 eDNA. In some cases, these changes resulted in the elimination of the coding region specifying the bHLH domain and likely affect DNAbinding and dirnerization of ABF-1. For the other retrieved splice products, the changes within the eDNA were localized to the 3' untranslated region (UTR), which likely indicate the potential for translational regulation of the ABF -1 gene.

The purpose of this master's thesis is to describe and analyse the origin and development of the library of Workers´ Educational Association in the community of Oskarström. This library is special because the organisation run it much longer than usual, until 1977, and to find an answer to why, we have the following questions: Why does the library of Workers´ Educational Association start their activity in Oskarström, and how does it develop, in relation to premises, librarians, financing, the stock of books and the numbers of books out on loan? What is the reason that the library in Oskarström is municipalized as late as 1977? The methods we use are hermeneutics and evaluation of the sources. The library is founded in 1914 and become important for the workers. Important sources of income are the subventions given by the state, municipality and other associations. The library acts as a public library during all its time. After the final reforms of the municipalities in Sweden, the new municipality Halmstad take over the library in 1974. The reason why the organisation runs it until 1977 is because the municipality wants to build new premises for the library, and these are not finished until that year.

This thesis describes and analyses how the leadership culture is constructed by the leadership discourse and leadership practices, in the Swedish non-profit organisation ABF, the Workers Education Association. The theoretical framework for the study is based on the practice theory by Swidler (2001) who claim that culture conceptualizes as practices in interaction with discourse. The empirical material consists of policy documents for the study of leadership discourse, and twelve qualitative interviews with leaders about their leadership practices. The empirical results show that the leadership discourse is based on three concepts: Carrying of idéas, Communicating and Developing. The leadership practice of the organisation is demonstrated through six leadership approaches: The communicative, The democratic, The stringent, The development focused, The situated and The positive. These approaches are used in internal and external activities. The conclusion of the study is that there is a clear stringency between the leadership discourse and leadership practices within the organisation which creates the leadership culture within ABF. The values and approaches expressed in the leadership discourse are also described in the interviews as practiced by the leaders.

Uppsatsen syftar till att skapa en bild om hur folkbildningsarbetet har pågått i Sandviken samt hur kvinnorna tar större plats i samhället. Det är ABF-Sandviken som folkbildare som står i centrum för undersökningen under perioden 1939-1968. Källmaterialet är studieprogram tryckta av ABF-Sandviken samt deras avrapporteringskort under tidsperioden. Frågeställningar som besvaras i uppsatsen är: Vilket folkbildningsarbete gjorde ABF-Sandviken? Fanns det någon ökning i antalet studiecirklar mellan åren 1939-1968? Hur står sig resultatet i förhållande till den skedda invånarökning i Sandviken vid samma tidsperiod? Hur såg könsfördelningen ut vid val av studiecirklar? Vid ett eventuellt överslag åt ett av könen, är det då nya studiecirkelämnen som har gjort överslaget möjligt? Hade studiecirklarna vid ABF-Sandviken någon koppling till samtiden? För att besvara dessa frågeställningar har jag använt mig av samtliga studieprogram och närvarolistor från ABF-Sandviken mellan 1939-1968. Jag har genom detta kommit fram till resultatet att det skedde en ökning av antalet studerande vid ABF-Sandviken på slutet av 1940-talet. Med ökningen av studerande så är en logisk förklaring att det även ökade antalet studiecirklar kopplade till ABF-Sandviken. Fördelningen mellan de olika könens val av studieämnen skiftar även under tidsperioden 1939-1968. Det börjar med ett överslag åt det manliga könet för att sedan under 1950- och 1960-talet göra en växling mot ett mer jämställt sökande. Dessa resultat har en grund i folkbildningsreformen år 1947 samt en jämställdhetsdebatt som blossade upp på 1950-talet. Folkbildningsreformen gjorde det möjligt att lättare få bidrag för studiecirklar och det gjorde en ökning in på 1950-talet och toppar på 1960-talet möjliga. Sedan med att fler kvinnor tog sig från den fasta positionen i hemmet och deltog mer i samhället blev deltagandet mellan könen mer jämställt. Studiecirkelämnen var påverkade av händelser i samtiden. Till exempel diskuterades andra världskriget och på en mer lokalnivå hur kommunen fungerade, och vart skulle man vända sig?

Dabei wird auf die ersten drei Bausteine des ISA-Modells eingegangen. Kapitel 6.2 stellt die Internationalisierungsmuster von zehn Unternehmen vor, dabei wird auf alle Bausteine des ISA-Modells eingegangen. Die Fallstudien in Kapitel 6.2 sind so ausgewählt, dass möglichst unterschiedliche Markteintrittsstrategien und deren Implementierung beschrieben werden können (siehe auch Abb. 6–9).

Abstract (sommario):

Recent studies show that nano-sized particles can be fluidized in the form of nanoparticle agglomerates. However, fluidization behavior such as the minimum fluidization velocity and fluidization regime differ significantly for different nanoparticles. Hence this paper is aimed to experimentally investigate the general fluidization characteristics of different nanoparticles. It is interesting to note that a fluidized bed of nanoparticle agglomerates is optically semi-transparent due to the extremely high porosity (typically over 99%) of the bed with respect to the primary materials of nanoparticles. Taking advantage of this unique feature, traditional optical measurement techniques are applied to visualize the flow structure as well as to measure the size of the fluidizing nanoparticle agglomerates. Based on measurements of four different nanoparticle materials, two types of fluidization behavior have been identified, which closely resemble those of classical Geldart Group A and Group B particles, respectively. It shows, however, that the bed of “Group A” nanoparticles expands as long as there is a flow through the bed, which is different from the classical fluidization of Geldart Group A particles where there is no bed expansion until reaching the minimum fluidization velocity. It is also noted that, based on the apparent density and size, the fluidization behavior of nanoparticle agglomerates do not precisely follow the Geldart classification. To differentiate these particles with very similar fluidization characteristics, terms the APF and ABF are introduced for the fluidization classification of nanoparticle agglomerates. Typical fluidization characteristics including bed expansion, bed pressure drop and hysteresis effects of both APF and ABF nanoparticles. The sizes of nanoparticle agglomerates also have been measured using an in-situ optical measurement system.

In this study, the thermo-mechanical finite element (FE) analysis of the 3D chip stacking packaging is accomplished by employing the commercial software, ANSYS®. After manufacturing process, the thickness of the deposited material becomes variable. For the most part, this is due to the uncertainty of the manufacture process. In analyzing the effect of thickness difference, the process modeling technique is adopted. The technique can be demonstrated by comparing simulation results and the designed experiment for established chip displacement measurements. The out-of-plane displacement of the fabricated chip is measured by the Twyman-Green (T/G) interferometer. According to the results simulated by the validated process modeling technique, the effect of the thickness difference of the ABF layer is insignificant. In thermo-mechanical FE analysis, the thermal expansion of ABF material can induce stress concentration at the copper via. Moreover, thermal expansion of the ABF material and copper via can also affect the reliability of the silicon chip. Based on the design concept, the effect of the copper via diameter is analyzed. Based on the results, the stress concentration phenomenon at the copper via improves as the diameter increases. However, a larger thermal expansion of the copper via can damage the chip structure because of the larger diameter.
