Successful and creative plant-breeding work in a stock-raising is based on the estimation of genetic potential of separate breeds, herds and individuals, that is impossible without thorough genetic- populations knowledge. Knowledge of features of caryotype gives an opportunity objectively to estimate the breeds of animals taking into account their population-cytogenetic features, that assists more complete idea about the evolution of breeds. However such important agricultural object, as a domestic sheep, remains cytogenetic poorly studied, especially in a population-cytogenetic aspect.In literature different breeds have small information about frequency and spectrum of the inherited anomalies and populations of sheep. Most chromosomal and genic anomalies of sheep in general not research, although for practice of plant-breeding work necessary knowledge of reasons of their appearance. Caryotype of sheep is presented by 54 chromosomes, from them 26 pairs of autsom and one pair of sexual chromosomes (ХХ or ХУ). Three pairs of large metacentric and 23 pairs of acrocentric chromosomes of different size enter in the complement of autsom. For sheep as well as for other animals characteristic chromosomal polymorphism as a numerical varying of chromosomes in caryotype (aneuploidi and poliploidi), morphological aberations and associations of separate chromosomes. Chromosomal anomalies of sheep are reason of forming of nonviable gamet, that results in death of embryos on the early stages, and, as a result, to the considerable economic losses in economies. The facts of chromosomal aberation educed for sheep testify to the necessity of cytogenetic control of tribal animals, especially rams, with the aim of exposure of animals-transmitters of undesirable changes in caryotype and exception of them from a plant-breeding process.

Mercury and its derivatives have become an alarming environmental problem, necessitating the search for effective antagonists, including homeopathic drugs, which are generally used in micro doses and are devoid of any palpable side-effects. On the basis of homeopathic similia principle, two potencies ofMercurius solubilis(Merc Sol-30 and Merc Sol-200) were tested by three administrative modes, i.e. pre-feeding, post-feeding and combined pre- and post-feeding, for their possible efficacy in ameliorating mercuric chloride-induced genotoxicity in mice. Healthy mice,Mus musculus, were intraperitoneally injected with 0.06% solution of mercuric chloride at the rate of 1 ml/100 g of body weight, and assessed for genotoxic effects through conventional endpoints. i.e. chromosome aberrations, micronuclei, mitotic index and sperm head abnormality, keeping suitable controls. Mercuric chloride-treated mice were divided into three sub-groups, which were orally administered with the drug prior to, after and both prior to and after injection of mercuric chloride, and their genotoxic effects were analysed at specific intervals of fixation. Mercuric chloride treatment generally produced more chromosome aberations, micronuclei and sperm head anomaly in mice, but the mitotic index appeared to be slightly reduced. While chromosome aberations, micronuclei and sperm head anomaly were generally reduced in the drug-fed series, the mitotic index showed an apparent increase. In most cases, the combined pre- and post-feeding mode appeared to show the maximum amelioration, followed by post-feeding and pre-feeding, in that order. The amelioration by Merc Sol-200 appeared to be slightly more pronounced. We conclude that potentized homeopathic drugs can serve as possible anti-genotoxic agents against specific environmental mutagens, including toxic heavy metals.

AbstractHigh resolution spectroscopy will be an important diagnostic tool in future X-ray astronomy. SPEKTROSAT, the follow-up mission to ROSAT will be equipped with a transmission grating spectrometer with a spectral resolution of 0.2 Å over the wavelength range between 6 Å and 300 Å. Third order aberations are minimized by a mounting of the grating elements according to the Rowland-torus geometry. The ROSAT mirror system will be slightly changed by a redesign of the optical stops in order to reduce the field of view. The Focal Instrumentation has to be modified for a better match with the spectroscopic requirements. Since SPEKTROSAT is almost identical to ROSAT, this concept offers a large amount of science at relatively low costs. Its limiting line sensitivity will be about 20 times better than that of the EINSTEIN-OGS.

Bacground/Aim. Mucosa-associated lymphoid tissue (MALT) lymphoma accounts for 5-17% non-Hodgkin lymphomas (NHL). The molecular pathogenesis of MALT lymphomas is not well-established. The aim of this study was to evaluate immunohistochemically determined nuclear coexpression of BCL10 and NF-kappaB (NF-?B) in tumor cells of gastric MALT lymphoma and its impact on the patogenesis and outcome of the disease. Methods. Medical records of 35 patients with newly diagnosed gastric MALT lymphoma were analyzed and biopsy specimens were immunostained for BCL10 and NF-kB expression (p65 subunit). Results. The median age of 35 patients diagnosed with gastric MALT lymphoma was 63.5 years (male/female = 21/14). Symptoms were present in 23/35 (65.7%) patients with the weight loss as the most common symptom. Gastric MALT lymphomas were usually localized in the stomach corpus and corpus and antrum (45.7% and 31.2%, respectively). H. pylori infection was confirmed in 20 out of 30 (66.7%) patients. Treatment options were as follows: immunochemotherapy in 10 (28.5%) patients, surgery in 9 (25.8%) patients, combined surgery and chemotherapy in 14 (40%) patients and supportive measures in 2 (5.7%) patients. Complete remission was achieved in 13 (37.1%) patients and partial remission in two (5.7%) patients. Sixteen (45.7%) patients had disease progression (p < 0.001). Cytoplasmatic expression of BCL10 in tumor cells was detected in 19 (54.3%) specimens. Nuclear expression was detected in no specimen. Cytoplasmic expression of NF-?B was present in 22 (65.7%) specimens, but nuclear expression was not detected in any specimens. Conclusion. Nuclear expressions (activation) of NF-?B p65 subunit and BCL10 were not detected in specimens of gastric MALT lymphoma. The correlation of nuclear coexpression of BCL10 and NF-?B in gastric MALT lymphoma was not established. These results indicate that other mechanisms and signal pathways are active in lymphogenesis of gastric MALT lymphoma, as that apoptotic inhibition is not the main, nor the only mechanism in tumorogenesis.

AbstractCystic echinococcosis (CE) due to Echinococcus granulosus is one of the most important helminthic diseases in Turkey where it constitutes a public health and economic problems. Its mean annual incidence in humans is 4.4/100 000 and the prevalence of the tapeworm agent in domestic animals ranges from 11.2 to 50.7 %. Since 1980s, albendazole has been used for treatment of the disease, and this benzimidazole drug has been considered to be of relatively low toxicity. However, prolonged albendazole therapy of CE became to be a common practice, and data on possible genotoxic effects of the medication in humans are lacking. This study has concerned 17 women and 11 men, in total 28 patients with liver cystic hydatid complaints, who were administered albendazole (15 mg/kg) preoperatively (2 weeks) and postoperatively (6 months). Genotoxic effects of albendazole were searched using Sister Chromatid Exchange (SCE), mitotic index (MI) and chromosomal aberations (CAs) methods, comparing lymphocyte chromosomes of treated patients and a control group of healthy individuals. The results indicated a significant increase of SCE frequencies and decrease of MI in the treated group (p < 0.001). Regarding CAs, any difference between the groups was not determined.

The subchronic dermal toxicity of aqueous solutions containing the diethanolamine salt of para-tertiary butyl benzoic acid was determined in Fischer 344 rats. Para-tertiary butyl benzoic acid (pt BBA) has important applications in the manufacture of resins, polymers, and corrosion inhibitors. Male and female rats were exposed topically 5 days/week for up to 13 weeks with dosing solutions that resulted in daily exposures of 0.0, 17.5, 35.0, 70.0, or 140.0 mg/kg pt BBA. These concentrations did not produce overt clinical signs of toxicity and did not cause irritation to dermal exposure sites. However, exposure to the two highest concentrations resulted in decreased weight gain. Exposure at all concentrations resulted in dosage-related increases in relative hepatic and renal weights. Exposure of males to the two highest concentrations caused decreased testis weight. Exposure-related pathologic changes were confined to three organ systems: cytoplasmic vacuolation in the liver; tubular dilation and papillary necrosis of the kidneys; and tubular degeneration in the testes. Accompanying aberations in clinical chemistry values suggested altered hepatic and renal function. In males exposed daily to 70.0 and 140.0 mg/kg pt BBA, the testicular effects were marked, but no effects were detected in rats exposed to 17.5 or 35.0 mg/kg of pt BBA.

A working model of an ocular micrometer has been designed for the MAHIS (Meridian Automated Horizontal Instrument by Sukcharev). An optical scheme of the micrometer includes the following devices: a)Artificial light marks in the focal plane of the objective. The marks have an increased sharpness and have a stable scale factor under defocusing. An amplitude-phase grid illuminated by coherent light from the second main point of the objective is used to make the marks.b)The observed stars and artificial marks are imaged on the CCD chip by means of an additional objective. This objective also corrects chromatic aberations of the main objective.c)A concentric meniscus is used to compensate for the chromatic refraction of the atmosphere. The meniscus center of curvature coincides with the center of the system's pupil image. In this case the compensation is equal at all points of the field of view.d)The possibility of measuring the normal attitude of the flat mirror relative to the main instrumental plane during each observation is discussed. For these purposes there needs to be a holographic grid on the mirror surface, an artificial zenith or nadir horizon and an autocollimated source of light in the focal plane.e)Laboratory investigations of a working model of a CCD camera are being carried out and the basic software is being developed. This work is planned to be finished in the autumn of 1994. The CCD matrix ISD 011A (NPO “Electron”, St. Petersburg, Russia) 512 × 512 pixels, with pixel size of 16 × 16 mkm is used. The noise is 5 electrons/pixel/sec when the temperature is −40° C (thermoelectric cooler).

Abstract Myeloid cell commitment is regulated by factors interacting with chromatin in a progenitor cell entering differentiation. PU.1 is an ETS family transcription factor that has been well characterized in inducing myelopoiesis and blocking erythroid differentiation. Conditionally activated PU.1-Estrogen Receptor transgene in mouse PU.1 knockout-derived hematopoietic progenitors is known to induce macrophage differentiation. We observed that manipulation of PU.1 activity by using different levels of PU.1-ER activator, tamoxifen, was capable of producing major populations of myeloid progeny including macrophages, granulocytes, lymphocytes and mastocytes. Gradual activation of PU.1 produce distinct committed populations that require presence of PU.1 activator. Withdrawal of tamoxifen from macrophages restored cycling and pluripotent potential resembling parental cell line. Furthermore, very low PU.1 activity induced frequent chromosomal aberations such as polyploidy. To determine that PU.1 directly caused these phenotypic changes we determined its occupancy at genes representing PU.1 targets by quantitative chromatin immunoprecipitation (ChIP). We demonstrate that PU.1 is enriched at regulatory regions of lineage restricted cell populations including promoters of CD11b, CD14, IL7R, PAX5, RAG1, Gelatinase, Lysozyme, and MCP. Different PU.1 levels promote distinct cell populations that have different localizations of PU.1 in chromatin. We also tested mRNA levels of PU.1 target genes in progenitor cell line induced by PU.1. Macrophage-specific target genes such as CD11b and CD14 are induced at high PU.1 levels and granulocytic PU.1 targets such as Gelatinase and Lysozyme are induced at intermediate and high PU.1 levels. B-cell specific mRNAs such as IL7-R, PAX5, and RAG1 were induced at low and high PU.1 levels. Our data suggest that PU.1 is recruited to and transactivates its lineage-restricted target genes differently at distinct PU.1 levels and such mechanism may be involved in myeloid cell commitment and leukemogenesis.

Abstract Acute myeloid leukemia (AML) is a heterogenous disease according to morphology, immunophenotype, and genetics. The retained capacity of differentiation is the basis for the phenotypic classification of the bulk population of leukemic blasts and the identification of distinct subpopulations. Within the hierarchy of hematopoietic development and differentiation it is still unknown at which stage the malignant transformation occurs. It was our aim to analyze the potential involvement of cells with the immunophenotype of pluripotent stem cells in the leukemic process by the use of cytogenetic and cell sorting techniques. Cytogenetic analyses of bone marrow aspirates were performed in 13 patients with AML (11 de novo and 2 secondary) and showed karyotype abnormalities in 10 cases [2q+, +4, 6p, t(6:9), 7, +8 in 1 patient each and inv(16) in 4 patients each]. Aliquots of the samples were fractionated by fluorescence-activated cell sorting of CD34+ cells. Two subpopulations, CD34+/CD38-(early hematopoietic stem cells) and CD34+/CD38+ (more mature progenitor cells), were screened for karyotype aberations as a marker for leukemic cells. Clonal abnormalities and evaluable metaphases were found in 8 highly purified CD34+/CD38-populations and in 9 of the CD34+/CD38-specimens, respectively. In the majority of cases (CD34+/CD38-, 6 of 8 informative samples; CD34+/CD38+, 5 of 9 informative samples), the highly purified CD34+ specimens also contained cytogenetically normal cells. Secondary, progression-associated chromosomal changes (+8, 12) were identified in the CD34+/CD38-cells of 2 patients. We conclude that clonal karyotypic abnormalities are frequently found in the stem cell-like (CD34+/CD38-) and more mature (CD34+/CD38+) populations of patients with AML, irrespective of the phenotype of the bulk population of leukemic blasts and of the primary or secondary character of the leukemia. Our data suggest that, in AML, malignant transformation as well as disease progression may occur at the level of CD34+/CD38-cells with multilineage potential.

In this paper, an attempt at measure formulation of aberation in the set of results of soil analyses observed on chernozems within Soil science is presented. The statistical evaluation was realized based on the report results from separate districts. The chernozem (regardless of subtype) were chosen as suitable for statistical evaluation. The analysis of one character was calculated for the set of results in topsoil (0–30 cm) and subsoil (30–60 cm). The comparison of average content in topsoil and subsoil was made by t-test. The significant difference is shown in content of humus (topsoil 2.61% and subsoil 1.86%), phosphorus (topsoil 8.31 mg/100 g, subsoil 4.75 mg/100 g) and kalium (topsoil 16.28 mg/100 g and subsoil 9.96 mg/100 g). There is no significant difference in other characters such as content of particles under 0,01 mm (topsoil 40.16% and subsoil 42.67%), exchange reaction (topsoil 7.03 and subsoil 7.00) and CEC – cation exchange kapacity (topsoil 23.74 mmol/100 g and subsoil 23.50 mmol/100 g). With respect to chernozems character it can be deduced, that in most of not anthropically influenced samples from mollic horizon the similar features were observed in both monitored depths. This presumption is supported also by observed differencies: humus content in topsoil was distinctly increased by addition of organic substances in organic fertilisers and by cultivation of perennial forage crops. The aberation was not observed in soil reaction. This fact could be interpreted by suppressor soil ability. Similarly, no aberation was documented in CEC. In view of quality/health of soils it is positive detection because both: exchange reaction and CEC are classified in basic soil characters. The maximal aberation was observed in phosphorous content in topsoil (with log-normal (extreme) distribution of results) and, somewhat less, in kalium content. Both characters are classified in most anthropically affected due to the addition of both components in fertilisers. Similar situation can be monitored also in the influence of humus content.

The aim of research: Studing cariotype conducted cows of Ukrainian Black-and-White dairy breeds and installed corelation conection between cariotype instability and reproduction ability. Methods of research: Сytogenetic – analysis of metaphase plates, peripheral blood cells cows of Ukrainian Black-and-White dairy breeds, micronucleus test-reaction of the organism on mutagenic factors of different nature, zootechnical – analysis of reproductive properties of animals Ukrainian Black-and-White dairy breeds, statistical. Research result: Cariotype analis of Ukrainian Black-and-White dairy breeds (30 heads) farm husbandry “Progress +” showed the following results: control of spontaneous mytagenesis in cows wich was reserch showing quantitative and structural chromosomal aberation. Quantative chromosomal aberationwas find as aneuploidy. Aneuploidy was equal to 7,4% which does not exceed the limits of spontaneous chromosomal variability(1,5–8,3%), what is normal for animals of milk production breeds. Cellular frequency with asynchronous difference centroid chromosomal areas i population of cows at husbandry “Progress +” was 1,05% and did not exceed the spontaneous cytogenetic variability characteristic for the species Bostaurus. The share of metaphase plates with structural chromosomal abnormalities (chromosomal breaks) 2,6% equal to the level of spontaneous chromosomal variability for cows (0,17–11,1%). Cellular cell with micro nucleus and dual core lymphocytes was within the spontaneous level, characteristic of mammals, which were kept in the absence of direct genotoxic effects and amounted to 3.7‰ and 2.3‰, respectively. To install associate conection between chromosomal abnormalities and the reproductive ability of the studied cows was carried out a correlation analysis. Received coefficient between dual-core lymphocytes and age 1 calving (r = -0,9585) indicates a negative correlation with the strength of communication according to the Student's criteria isreliable at P > 0.99. Conclusions: Quantitative chromosomal aberrations (aneuploidy) were 7.4% which does not exceed the limit of spontaneous chromosomal variability, characteristic for animals breeds of milk production. The frequency of cells with asynchronous difference is established centroid chromosomal areas – 1,05% and a fraction of metaphase plates – 2,6% which corresponds to the level of spontaneous chromosomal variability in cattle. Detected сell frequency with micro nucleus and dual core lymphocytes which were within the spontaneous level, characteristic of mammals which are kept in the absence of direct genotoxic effects and amounted to 3.7‰ and 2.3‰. Was detected a negative correlation relationship between dual-core lymphocytes and age of the 1 calving (r = -0.9585, P > 0.99).

In vitro experiments were designed to investigate the role of aqueous extract of Rosmari nusofficinalisin the inhibition of genotoxic effect of methotrexate (MTX) in human lymphocyte. Mitotic index MI, chromosomal Aberation CA used us cytogenetic tests to detect the inhibition of genotoxic effect of Methotrexate using two concentration. The results showed that the aqueous extract of Rosmari nusofficinalis inhibited the genotoxic effect of Methotrexate in human lymphocyte. As well as the exposure of cells to aqueous extract before expose to Methotrexate T1 had a high inhibitory efficiency comparing with the combined exposureto the extract & Methotrexate together T2 and the exposure to the Methotrexate before extract. According to our obtained results it could be the aqueous extract of Rosmari nusofficinalis considereds as a antimutagene agent (Desmutagen) in first place and as a antimutagene agent (Bioantimutagene) in second place.

Results of the analyses of the crystal structure and crystal defects of high-Tc Bi2Sr2CanCun+1Oz ceramics are presented. Specimen were prepared in a conventional mixed oxide process. In series of electron diffraction patterns of the (n=1) phase(Tc≈ 80 K) reflections appear which are not consistent with the Bbmb space group symmetry suggested in the litterature. Extinction conditions obtained from electron diffraction patterns are compared to extinction conditions of space groups (e.g. Pbmb and Pbnb) which have a primitive Bravais lattice and are “klassengleiche” subgroups of Bbmb.Planar crystal defects on (001) planes were observed in Bi2Sr2CanCun+1Oz phases. These defects are (i) intrinsic and extrinsic (001) stacking faults forming lamellae of {n=0,1,2,3,4) phases and (ii) (001) twin boundaries. An extrinsic (001) stacking fault in the {n=1) phase is shown in fig.1. The image was obtained with a 400 kV electron beam in a microscope equippped with a pole piece with small spherical aberation (Cs= 1mm).

Creating optical illusions or invisible objects by using cloaking devices is still being a concern for many researchers in the world. The cloaking devices are working in visible spectrum regime, large dimention, and replicable. Addition with the full-field of light, no new materials, simple infrastructure, and scale easily may be considered an ‘ideal’ cloaking device. Based on four lenses cloaking system by Choi’s work shows an idea for solution of blindspot large vehicle’s problem. The set up for this optical system is regular covex lenses non-aberation based on educational laboratory framework. The experimental confirmation that cloaking area with 4f convex lenses is limited to certain areas only, not 100 % of the object to be observed can disappear from the view. Vibration disturbance generated by rotor module (wheels disturbance analogy on vehicles) applied on the system demonstrate that below on 2000 rotation per minutes (rpm) equal 33.34 hertz, optical illusion is still consistent. The results support the information to create optical cloaking prototype for optimizing large vehicle driver’s visibility as alternative solution blindspot’s problem.

In Biology high resolution SEM has become of special interest to study the surface representations of isolated macromolecular complexes as well as the organisation of such complexes and organelles in their natural context.Apart from the probe size, surface imaging in the nm-scale is limited by the interaction volume of the primary electrons, which directly increases with increasing primary energy. On one hand at low voltages (200-1000eV) the interaction volume can be kept minimal, but a probe size of 1-2 nm can only be obtained by a correction of the spherical and chromatic aberrations (Zach, 1990; Haider et al., 1994). On the other hand, the probe size at high voltages is mainly limited by diffraction and spherical aberation but nevertheless a probe sizes <1 nm at 30keV can be routinely achieved (Nagatani et al., 1987). To overcome the resolution limitation by the large interaction volume in bulk samples, biological specimens are coated with a very thin (1-2 nm) metal film to localize the SE-signal at the surface of the specimen (Joy, 1990).

Abstract Introduction: Gemtuzumab Ozogamicin (Mylotarg®) is a humanized anti CD33 antibody linked to the cytotoxic drug calicheamicin, which has shown considerable antileukaemic effect in the treatment of relapsed AML. With special regard to its toxicity, we present our preliminary results of Mylotarg treatment when administered on compassionate basis as consolidating monotherapy, or combined with intensive chemotherapy. Patients and methods: A total of 86 doses, corresponding to 65 courses were administered to 49 patients. Indications for Mylotarg treatment ( mono ~ monotherapy; comb ~ combination therapy. No.of courses are shown in parentheses): Primary AML: Reinduction ( mono:1; comb:3). Cytogenetic reinduction ( mono:3; comb:1). Consolidation ( mono:24; comb:2). Relapsed AML: Induction ( mono:4; comb:11). Reinduction ( mono:1; comb:2). Cytogenetic reinduction ( mono:1). Consolidation ( mono:10; comb:2). Results: Up-front consolidation with Mylotarg as monotherapy ( 30 doses; 24 patients; mean Mylotarg dose 5.7 mg/m2). Fever: 51% of doses. Focal infections: 17%. Bacteremia 7%. Biochemical findings: Platelet nadir at day 10 ( < 20 bill./l: 25% doses). Neutrophil nadir at day 11 ( mean count = 0.12 bill./l). Hepatic toxicity ( grade 1; % of doses): Alkaline phosphatase 7%; ALAT: 13%; Bilirubin 0. A similar pattern was seen in the 10 relapsed patients consolidated with Mylotarg as monotherapy. One patient receiving an allogeneic transplant 3 months after Mylotarg treatment developed fatal VOD. Three other patients who undervent allogeneic stem cell transplantation 2, 2 and 3 months, respectively, after the administration of Mylotarg did not experience severe hepatic complications. We compared twelve courses of Mylotarg combined with DaunoXome and high dose cytarabine with ( n=10) or without ( n=2) added fludarabine and etoposide, administered to10 patients in 1st relapse with 8 similar courses but without Mylotarg, administered to 7 patients with similar characteristics regarding age, CR1 duration and cytogenetics ( mean Mylotarg dose 4.4 mg/m2). More cases of grade 3 diarrhea ( 33% vs 13%) and bacteremia ( 67% vs 38%) were seen in the Mylotarg group, in which three patients suffered hypoplastic deaths. One of these patients developed intestinal perforation following postremission treatment. The two other patients, with an early refractory relapse, and a late relapse with high risk chromosomal aberation, respectively, died from infectious complications following 20 and 36 days of profound neutropenia. These 3 patients recieved 5.9, 8.3 and 5.0 mg/m2 respectively, compared to the average Mylotarg dose of 4.4 mg/m2. Conclusion: Mylotarg seems to increase toxicity when added to intensive chemotherapy in relapsed AML. The feasibility of this approach must await further studies evaluating toxicity, appropriate dosing and response pattern. When administered as consolidating monotherapy toxicity is modest.
