Background: The number of people suffering from lifestyle diseases is increasing over the world, causing significant morbidity and mortality, particularly in developing countries. Changes in behaviour and the adoption of healthy lifestyle practises can help prevent or reduce the progression of lifestyle diseases. Many patients, however, lacked the knowledge, attitude, and practise of healthy lifestyles.Methods: A cross-sectional study was conducted among the 329 participants in the urban field practice area of the department of community medicine, JSS medical college, Mysore, with study duration of 6 months. A house-to-house survey was performed to recruit study participants using convenient sampling. participants will be interviewed by using the pre-designed, pre-tested and structured questionnaire which includes socio-demographic profile, dietary patterns, physical activity, type of personality, history of stress, history of tobacco and alcohol consumption, family history of lifestyle diseases like HT, DM etc.Results: Among 329 study participants 306 (93%) have practice of doing physical exercise, 328 (99.7%) decreased salt/oil intake, 329 (100%) stopped smoking/alcohol consumption, 329 (100%) check BP regular, 327 (99.4%) check blood sugar regularly, 328 (99.7%) stopped eating junk food/cool drinks, 329 (100%) take medicationsConclusions: Healthy practices need to be subsidized with the aid of using the dependent periodic medical exam. There need to be an emphasis on incorporating bodily workout within side the day by day routine, consumption of healthful diet, and decrease/cessation of alcohol and tobacco consumption.

In H-2d mice, the immunodominant determinant of the HIV-1-IIIB gp160 envelope glycoprotein recognized by CD8+ CTL is represented by a 15-residue synthetic peptide (315-329: RIQRGPGRAFVTIGK). This peptide is seen in association with the Dd class I MHC molecule expressed on H-2k L cell fibroblast targets. We explored the structural requirements for CTL recognition of this peptide at the levels of both the peptide molecule and the class I MHC molecule. Using several transfectants expressing recombinant Dd/Ld molecules, we found that presentation of this epitope required both the alpha 1 and alpha 2 domains of the Dd molecule, in contrast to certain instances of allorecognition for which alpha 1 of Dd was sufficient in association with alpha 2 of Ld. Because this peptide derives from a hypervariable segment of the HIV envelope, substituted peptides could be used to define not only the structures affecting interaction of peptide with class I MHC molecule and with the TCR, but also the structural basis for the effect of naturally occurring viral variation on CTL recognition. The CTL-LINE specific for this HIV-1-IIIB-derived sequence could not recognize the HIV-1-RF variant-derived sequence from exactly the same site (315-329:--HIGPGRVIYATGQ). Peptides with single amino acid substitutions from the HIV-1-IIIB sequence toward the HIV-1-RF sequence were made to test the effect of each residue significantly affected recognition, and only one, 324(F), was obligatory. Moreover, both 322(R) and 324(F) substituted peptides failed to inhibit the binding of the wild type peptide to the MHC molecule. Therefore, the amino-acids 322(R) and 324(F) seem to be involved in regulating peptide interaction with the Dd class I MHC molecule. In contrast, 325(V) appeared to affect interaction with the TCR. We suggest that sequence variations among known HIV-1 isolates that affect peptide binding to MHC such as those described here, if occurring during the course of infection of an individual, could result in failure of the MHC molecules of that individual to present the peptide. If the number of dominant HIV CTL epitopes is indeed very limited, such a blind spot could allow the virus to escape immune control, proliferate rapidly, and cause AIDS.

Abstract BACKGROUND The Gas6/AXL signaling pathway plays a substantial role in cancer cell survival, proliferation, immunosuppression, and metastasis. Elevated AXL expression is associated with advanced cancer stages and decreased overall survival rates, underscoring its potential as a therapeutic target for metastatic cancers. We investigated this possibility by utilizing AB-329, a selective small molecule AXL kinase inhibitor, to treat triple-negative breast cancer (TNBC). TNBC, typified by the lack of ER, PR, and HER2 expression, frequently displays mesenchymal characteristics and carries a significant risk of metastasis and an immunosuppressive tumor microenvironment. METHODS We assessed AXL protein expression in 28 human and 11 mouse breast cancer cell lines using Western blotting. The Microarray database was utilized to examine AXL gene expression in 40 human breast cancer cells. The Sulforhodamine B proliferation and soft agar assays measured the antiproliferative effect of AB-329 in TNBC cell lines. The in vitro anti-metastatic impact of AB-329 was evaluated through migration and invasion assays. We appraised the anti-tumor effects of AB-329 in combination with paclitaxel using 4T1 and E0771-LMB mouse TNBC xenograft models. We used FACS and CIBERSORTx to perform immune cell type profiling in E0771-LMB xenograft tumor tissues. To understand AB-329’s immunomodulatory effects in tumors, single-cell RNA sequencing was conducted in a humanized mouse model with SUM149 human TNBC xenografts. RESULTS Microarray data revealed higher AXL expression in TNBC cells compared to non-TNBC cells. Western blotting supported this, showing higher AXL protein expression, particularly in basal and mesenchymal subtypes of TNBC cells, compared to luminal breast cancer cells. AB-329 presented moderate antiproliferative effects as a monotherapy in the TNBC cell lines tested, with half-maximal inhibitory concentrations exceeding 5 µM. Nevertheless, combining AB-329 with paclitaxel resulted in significant growth inhibition in all tested TNBC cell lines (p < 0.05). Importantly, AB-329 alone could inhibit TNBC migration and invasion at a concentration of 1 µM (p < 0.01) and showed enhanced inhibition rates when paired with paclitaxel (p < 0.05). The combination of AB-329 and paclitaxel considerably reduced tumor growth in the 4T1 and E0771-LMB xenograft models compared to paclitaxel or AB-329. Total RNA sequencing and CIBERSORTx analysis indicated that the AB-329 and paclitaxel combination increased Natural Killer (NK) cells’ infiltration into E0771-LMB mouse TNBC xenograft tumors compared to single agent-treated tumors. A nearly 2.5-fold increase in NKT cell infiltration was also detected with AB-329 treatment compared to Control in SUM149 human TNBC xenografts. CONCLUSIONS Our preclinical data suggest that AB-329, an AXL inhibitor, could synergize effectively with paclitaxel in TNBC treatment, notably inhibiting tumor cell proliferation and migration while enhancing NK cell infiltration. Intriguingly, we observed an increased infiltration of NKT cells into tumors following AB-329 treatment, which may promote an anti-tumorigenic immuno-microenvironment. These results underscore the necessity for further investigation into the role of AB-329 in modulating the tumor immuno-microenvironment through AXL pathway inhibition and accentuate the potential for combining AB-329 with immunotherapy. The desire to procreate in patients living with HIV (PLHIV) has been seldom investigated in Africa, particularly in Gabon. The aim of this transversal and descriptive study was to analyze the socio-demographic and behavioral factors associated with a desire to have children in a cohort of PLHIV. The study included 442 patients, predominantly females [79.9% (337/422)], and those with a secondary school education [64.2% 271/422)]. The highest prevalence of HIV was found in patients aged 30-39 years old (44.3%), of which 59% (249/422) were unemployed. The desire to have children was noted in 78% (329/422) of patients, of which 82.4% (271/329) were treated with antiretroviral drugs; this was significantly higher in subjects under 40 years versus those over 40 years old [81% (268/329) versus 19% (61/329), p<0.001]. Sero-discordant couples represented 33.4% (110/329) of patients. The frequency of patients with the desire to have a child was significantly higher when patients wanted to hold the status of parent of a child [77% (255/329) versus 23% (74/329), p<0.001]; this was influenced by the partner's desire [60% 197/329 versus 40% (132/329), p< 0.001], as well as by the absence of weight loss [56% (185/329) versus 44% (144/329), p<0.001]. The average number of children was significantly lower in patients with the desire to procreate compared to those with no desire to have children [1.7 versus 3.2, p<0.001]. These first observations in Gabon highlight the importance of the desire to have children in PLHIV and sero-discordant couples, and they show the level of interest in developing assistance methods for procreation and family planning programs to help this population, as well as to reduce the risk of mother-to-child HIV transmission.

Background: Bone cancer pain (BCP) is a common symptom occurring among patients with cancer and has a detrimental effect on their quality of life. Growing evidence has implicated microRNA-329 (miR-329) in the progression of bone diseases. In the present study, we aimed to elucidate the potential effects of miR-329 on BCP in a BCP mouse model via binding to lysophosphatidic acid receptor 1 (LPAR1) through the LPAR1/extracellular signal-regulated kinase (ERK) signaling pathway. Methods: Initially, a BCP mouse model was established via injection of 4 × 104 murine breast tumor (4T1 cell) cells (4 μl). The interaction between miR-329 and LPAR1 was identified using a bioinformatics website and dual luciferase reporter gene assay. The modeled mice were subsequently treated with miR-329 mimic, LPAR1 shRNA, or both, in order to examine the effect of miR-329 on the paw withdrawal threshold (PWT) and paw withdrawal latency (PWL) of mice, the expression of LPAR1/ERK signaling pathway-related genes. Results: The positive expression rate of LPAR1 protein and extent of ERK1/2 phosphorylation were increased in BCP mouse models. LPAR1 is a target gene of miR-329, which can inhibit the expression of LPAR1. In response to miR-329 overexpression and LPAR1 silencing, BCP mice showed increased PWT and PWL, along with decreased LPAR1 expression and ratio of p-ERK/ERK. Conclusions: Altogether, the results obtained indicated that miR-329 can potentially alleviate BCP in mice via the inhibition of LPAR1 and blockade of the LPAR1/ERK signaling pathway, highlighting that upregulation of miR-329 could serve as a therapeutic target for BCP treatment.

This article investigated the microstructure and damping property of TPU composites with different contents of ultraviolet absorbents. It can be found that the ultraviolet absorbents formed fiber-shaped precipitations in the TPU matrix. The UV-328 was randomly distributed in the matrix and exhibited a weak interfacial bonding with the matrix. In comparison, the UV-329 was well embedded in the matrix and formed a relatively better interfacial bonding and compatibility with the TPU matrix. The damping factor tanδ of both 328-composites and 329-composites had been reduced gradually with increasing content of ultraviolet absorbents at the glass transition temperature range due to the fact that the ultraviolet absorbents were in the crystalline state which decreased the volume content of the viscoelastic TPU matrix. But the tanδ increased at the temperature range of higher than the glass transition temperature, which should be related to the dominance of interfacial frictions between the ultraviolet absorbents and the matrix on the energy absorption.

Most of the dermatological research in men who have sex with men (MSM) has been in the HIV-infected subpopulation. A retrospective case-control study was performed of dermatologic diagnoses in HIV-negative MSM (n = 329) compared with male heterosexual controls (n = 600). HIV-negative MSM had a higher frequency of folliculitis (19/329 vs 12/600; P = 0.004), cutaneous herpes simplex infection (19/329 vs 14/600; P = 0.009), non-melanoma skin cancer (16/329 vs 10/600; P = 0.006) and syphilis (8/329 vs 2/600; P = 0.005) than the control group. The findings suggest that HIV-negative MSM may have a higher burden of certain skin conditions and this appears to be independent of immunosuppression or sexual behaviours.

To investigate the relative participation of peripheral (CCK-A) and central (CCK-B) cholecystokinin (CCK) receptors in satiety induced by endogenous CCK, we examined the effect of the CCK-A antagonist MK-329 (10-315 micrograms/kg) and the CCK-B antagonist L 365260 (0.1-315 micrograms/kg) on intake of a 20% sucrose solution in mildly food-deprived mice. Intraperitoneal injection of MK-329 elicited a dose-related increase in sucrose consumption with a minimal effective dose of 31.5 micrograms/kg. This dose increased sucrose intake 23% and the highest dose tested, 315 micrograms/kg, increased sucrose intake 63% above baseline. In contrast to MK-329, intraperitoneal administration of L 365260 had no effect on sucrose intake at doses up to 315 micrograms/kg. To examine the contribution of these two CCK receptor subtypes in satiety induced by exogenous CCK, CCK-8 (8 micrograms/kg) was administered alone and in combination with MK-329 and L 365260. MK-329 (10 micrograms/kg) significantly attenuated the satiety effect of CCK-8, and L 365260 (100 micrograms/kg) was without effect. These results suggest that the peripheral CCK receptor subtype mediates satiety induced by endogenous and exogenous CCK in the mouse.

Abstract (sommario):
In order to help State to ensure food security of the populations, technologies for substituting wheat flour with flour from local food resources such as rice are used in bread making. The aim of this study was to determine the rheological behavior of pastes obtained by incorporating parboiled and non-parboiled rice flour in the bread-making process. The results obtained revealed that parboiling improves the tenacity of the mixed wheat (85%) and rice (15%) paste. Indeed, the parboiled Sahel 328 and Sahel 329 rice varieties would give a tougher mixed paste (P/L = 1.24±0.030; 1.28±0.020 respectively) and closer to that of 100% wheat (P/L= 1.13±0.050). The incorporation of parboiled Sahel 329 rice flour resulted in a paste with a swelling index closest to that of the control (WS). The results also indicated that all the parboiled rice studied composite flours had satisfactory baking strength. Of all studied varieties, Sahel 329 when parboiled, gave the closest rheological properties to the control (100% wheat). In addition, we can consider from this study, the alternative use of rice in bread-making supports the idea that rice would be a better vehicle than the soft wheat flour used for iron and folic acid fortification in Senegal.

The breeding work on the competitive creation has appeared to continue on the basis of the Institute of Horticulture (IH) NAAS and its research network. According to the results of the breeding the guelder-rose ordinary increases thanks to the fruitful work activity of the specialists of the M.M. Gryshko National Botanical Garden (NBG) of the NAS of Ukraine (the cultivars Berehynia, Nasoloda), IH NAAS (Horianka, Kyivska Sadova №1, Uliana, Ania, Yaroslavna, Harmoniia, etc.) and its scientific network, in particular, the L.P. Symyrenko Research Station of Pomology of the Institute of Horticulture (NAAS) (Rubinova, Koralova, Bahriаna, Velykoplidna, Ukrainochka, etc.). Taking into consideration the present day requirements of the fruit and ornamental horticulture under the global climate changes of the above mentioned crop which would high environmental adaptability, meet the requirements of the mechanized small picking fruits as well as the processing and food industries as to the consumption quality for making products of the functional and preventive action, that requires the search, selection and creation of new relevant corresponding initial material. As a result of new guelder-rose ordinary genotypes have been created, which are characterized by a complex of economy traits valuable – these are forms F 211-35-17, F 301-19-17, F 302-22-17, F 303-16-17, F 304-23 -17, F 305-3-17, F 306-4-17, F 308-9-17, F 309-8-17, F 312-14-17, F 316-02-17, F 322-28- 17, F 328-50-17, F 329-10-17, F 332-40-17, F 335-77-17, F 347-49-17, F 350-75-17, F 360-6-17. For the purpose of the further breeding on productivity and quality the comparative evaluation of new V. opulus genotypes was carried out on the phenology of the plants development, morphological traits and fruits biochemical indices. The breeding forms F 301-19-17, F 332-40-17, F 329-10-17, F 360-6-17 have proved high productive and are characterized as valuable breeding material with a complex of economic traits that have proven themselves at a level of conditional standard, and on some signs with the significant excess too. The berries of the variety Koralova and as well as of F 308-9-17, F 305-3-17, F 350-75-17 and F 304-23-17 have a high content of total sugars (> 10.0%), that is a sugar-acid index at a level of 6.5; 6.7; 7.4 and 7.5, respectively, and indicates their dessert and the possibility of their consumption as fresh while the increased content of pectin in the berries of the varieties Yaroslavna, Koralova Podilska, Ania and F 360-6-17, F 312-14-17, F 328-50-17 and F 322-28-17 testifies their considerable jelly ability which enables to use them for manufacturing: marmalades , pastilles, jams. It is the fruits of varieties Yaroslavna, Koralova and Koralova Podilska as well as of F 322-28-17, F 301-19-17, F 322-28-17, F 211-35-17 that have the high antioxidant capacity. The article breeders should utilize the following strains of Velikoplidna, Ukrainochka, Koralova, Ania, Yaroslavna, Uliana, Elina, Koralova Podilska, Tsukrova as an source material the breeding on the fruits productivity and quality as well as F 301-19-17, F 332-40, F 329-10-17, F 360-6-17, etc.

Fish species can bioaccumulate different pollutants present in the marine environments and incorporate them into the trophic chain. In this work, the occurrence of organic ultraviolet (UV) stabilizers and filters in different species of fishes of high consumption has been studied. A multiresidue method based on microwave-assisted extraction and ultra-high performance liquid chromatography with mass spectrometry detection was developed and then it was applied to nine fish species from markets in the Canary Islands and Catalonia (Spain). Three UV filters (BP-3, OC and BM-DBM) and two stabilizers (UV-328 and UV-329) were found in some of the studied species, in concentrations ranging between 0.067 and 0.683 µg g−1 dry weight (dw). BP-3 (UV filter) was the most frequently detected compound, followed by UV-329 (UV stabilizer). Thunnus thynnus was the most heavily polluted species, with a concentration of 1.201 µg g−1 dw as the sum of all measured compounds.

Abstract Immunotherapies targeting B-lineage-specific surface marker CD19 had demonstrated promising clinical results. Two CD19 CAR-T therapies (Kymriah® and Yescarta®) have been approved by FDA to treat patients with B cell malignancies, however, the complicated manufacturing process and low throughput limit its accessibility to more patients, especially in developing countries. The first CD3-activating bi-specific antibody targeting CD19, Blincyto, or CD19 BiTE, was approved to treat relapsed and refractory acute lymphoblastic lymphoma (r/r ALL). The relatively short half-life of Blincyto requires continuous IV infusion for weeks to maintain a steady levels of drug exposure, not to mention the high risk of developing severe cytokine release syndrome in patients. We had established a bispecific antibody platform ITabTM (immunotherapy antibody) for the generation of CD3-activating bi-specific antibodies that could potentially overcome the shortcomings of BiTEs. A CH1 domain was introduced into the ITabTM construct design with the intent to increase the molecular weight thus led to extend the serum half-life of the bispecific antibody. A novel CD3-activating and monkey cross-reactive antibody was generated with a less degree of T cell activation and cytokine release compared to BiTEs. A bi-valent binding to tumor associated antigen (TAA) format was established to target tumor cells and/or stem cells expressing very low levels of TAA. We report here the biological properties of the mono-valent/bi-valent binding of CD19 bi-specific antibody with CD3-activating activity (A-319/A-329). A series of studies were conducted to evaluate the bioactivities of A-319/A-329 in vitro and in vivo including binding to CD3 and CD19 antigens, T-cell and B-cell binding activities, T cell activation and proliferation and B cell killing activities in vitro as well as in vivo efficacy using human PBMC engrafted mouse xenograft models. The in vitro data showed that the mono-valent and bi-valent CD19 binding had little effect on the CD3-associated activities including CD3 antigen binding affinity, T cell binding and T cell activation. In contrast, the bi-valent binding format A-329 showed better potency compared to the mono-valent format A-319 in CD19 binding (KD 0.89 nM vs 19.4 nM); B cell binding (EC50 at 2.3 pM vs 462 pM); in vitro human B cell killing (EC50 0.2 pM vs 3.4 pM). Both A-319 and A-329 were capable of mediating tumor cell lysis with EC50 at 0.03~4 pM. A-329 demonstrated a greater killing activity on Pfeiffer, a human diffuse large B-cell lymphoma (DLBCL) cell line with a low expression of CD19 antigen. In human PBMC engrafted NOG mouse xenograft model, a dose-dependent tumor growth inhibition was observed at 0.5~100 µg/kg in both A-319 and A-329. In monkey studies, when A-319 and A-329 was dosed at 3, 10, 30 µg/kg, twice or three times weekly via IV infusion for A-329 or A-319. Dose-dependent elimination of peripheral blood B cells were observed with both ITabTM. The CD19 bi-valent format of A-329 revealed more complete B cell killing in monkeys. No significant difference of cytokine induction or liver injuries were observed between A-319 and A-329. These results demonstrated that both A-319 and A-329 may benefit patients with B cell malignancies with less dosing frequency and lower cytokine inductions especially, A-329 may have the potential to targeting the low CD19 expressing tumor stem cells. AbstractIn Saccharomyces cerevisiae, the Rad52 protein plays a role in both RAD51-dependent and RAD51-independent recombination pathways. We characterized a rad52 mutant, rad52-329, which lacks the C-terminal Rad51-interacting domain, and studied its role in RAD51-independent recombination. The rad52-329 mutant is completely defective in mating-type switching, but partially proficient in recombination between inverted repeats. We also analyzed the effect of the rad52-329 mutant on telomere recombination. Yeast cells lacking telomerase maintain telomere length by recombination. The rad52-329 mutant is deficient in RAD51-dependent telomere recombination, but is proficient in RAD51-independent telomere recombination. In addition, we examined the roles of other recombination genes in the telomere recombination. The RAD51-independent recombination in the rad52-329 mutant is promoted by a paralogue of Rad52, Rad59. All components of the Rad50-Mre11-Xrs2 complex are also important, but not essential, for RAD51-independent telomere recombination. Interestingly, RAD51 inhibits the RAD51-independent, RAD52-dependent telomere recombination. These findings indicate that Rad52 itself, and more precisely its N-terminal DNA-binding domain, promote an essential reaction in recombination in the absence of RAD51.

Background Patient attitudes and behavior are critical to understand owing to the increasing role of patient choice. There is a paucity of investigation into the perceived credibility of online information and whether such information impacts how patients choose their surgeons. Objective The purpose of this study was to explore the attitudes and behavior of patients regarding online information and orthopedic surgeon selection. Secondary purposes included gaining insight into the relative importance of provider selection factors, and their association with patient age and education level. Methods This was a cross-sectional study involving five multispecialty orthopedic surgery groups. A total of 329 patients who sought treatment by six different orthopedic surgeons were asked to anonymously answer a questionnaire consisting of 25 questions. Four questions regarded demographic information, 10 questions asked patients to rate the importance of specific criteria regarding the selection of their orthopedic surgeon (on a 4-point Likert scale), and 6 questions were designed to determine patient attitude and behaviors related to online information. Results Patient-reported referral sources included the emergency room (29/329, 8.8%), friend (42/329, 12.8%), insurance company (47/329, 14.3%), internet search/website (28/329, 8.5%), primary care physician (148/329, 45.0%), and other (34/329, 10.3%). Among the 329 patients, 130 (39.5%) reported that they searched the internet for information before their first visit. There was a trend of increased belief in online information to be accurate and complete in younger age groups (P=.02). There was an increased relative frequency in younger groups to perceive physician rating websites to be unbiased (P=.003), provide sufficient patient satisfaction information (P=.01), and information about physician education and training (P=.03). There was a significant trend for patients that found a surgeon’s website to be useful (P<.001), with the relative frequency increased in younger age groups. Conclusions This study shows that insurance network, physician referrals, appointment availability, and office location are important to patients, whereas advertising and internet reviews by other patients were considered to be not as helpful in choosing an orthopedic surgeon. Future studies may seek to identify obstacles to patients in integrating online resources for decision-making and strategies to improve health-seeking behaviors.

Nowadays, there is a need for consensus on diagnostic criteria, measures to improve the reliability of researches and the development of effective and efficient treatment methods, despite the significant amount of researches conducted on digital addiction (both Internet and gadget addiction). At the present stage, it is advisable and possible to find a common approach that is relevant for providing preventive and medical care to children and adolescents with digital addiction. This review of recent scientific articles systematizes modern treatment tactics for the prevention, diagnosis and therapy of these conditions, primarily in children and adolescents, which can be summarized as follows: digital addiction is a behavioral addiction (1), the diagnosis of which requires a multimodal/integrated approach (2), which consists of preventive measures in childhood and adolescence ("digital hygiene") (3), psychotherapy of behavioral addiction, behavior disorders, social maladaptation and neurotic disorders (4), and psychopharmacotherapy of the existing depressive and neurotic spectrum (in particular, selective serotonin reuptake inhibitors, clomipramine) and attention deficit hyperactivity disorder (methylphenidate, atomoxetine) (5). Inhibition of the 14q32 microRNAs, miR-329-3p and miR-495-3p, improves post-ischemic neovascularization. Cold-inducible RNA-binding protein (CIRBP) facilitates maturation of these microRNAs. We hypothesized that CIRBP deficiency improves post-ischemic angiogenesis via downregulation of 14q32 microRNA expression. We investigated these regulatory mechanisms both in vitro and in vivo. We induced hindlimb ischemia in Cirp−/− and C57Bl/6-J mice, monitored blood flow recovery with laser Doppler perfusion imaging, and assessed neovascularization via immunohistochemistry. Post-ischemic angiogenesis was enhanced in Cirp−/− mice by 34.3% with no effects on arteriogenesis. In vivo at day 7, miR-329-3p and miR-495-3p expression were downregulated in Cirp−/− mice by 40.6% and 36.2%. In HUVECs, CIRBP expression was upregulated under hypothermia, while miR-329-3p and miR-495-3p expression remained unaffected. siRNA-mediated CIRBP knockdown led to the downregulation of CIRBP-splice-variant-1 (CIRBP-SV1), CIRBP antisense long noncoding RNA (lncRNA-CIRBP-AS1), and miR-495-3p with no effects on the expression of CIRBP-SV2-4 or miR-329-3p. siRNA-mediated CIRBP knockdown improved HUVEC migration and tube formation. SiRNA-mediated lncRNA-CIRBP-AS1 knockdown had similar long-term effects. After short incubation times, however, only CIRBP knockdown affected angiogenesis, indicating that the effects of lncRNA-CIRBP-AS1 knockdown were secondary to CIRBP-SV1 downregulation. CIRBP is a negative regulator of angiogenesis in vitro and in vivo and acts, at least in part, through the regulation of miR-329-3p and miR-495-3p.

Flor yeast strains represent a specialized group of Saccharomyces cerevisiae yeasts used for the production of sherry-like wines by biological wine aging. We sequenced the genome of the industrial flor yeast strain I-329 from a collection of microorganisms for winemaking “Magarach” and the metagenomes of two flor velums based on this strain and continuously maintained for several decades. The winery uses two processes for the production of sherry-like wine: batch aging and a continuous process similar to the criaderas–solera system. The 18S rRNA gene profiling and sequencing of metagenomes of flor velums revealed the presence of the yeasts Pichia membranifaciens and Malassezia restricta in minor amounts along with the dominant S. cerevisiae I-329 flor yeast. Bacteria Oenococcus oeni and Lentilactobacillus hilgardii together accounted for approximately 20% of the velum microbiota in the case of a batch process, but less than 1% in the velum used in the continuous process. Collection strain I-329 was triploid for all chromosomes except diploid chromosomes I and III, while the copy numbers of all chromosomes were equal in industrial velums. A comparative analysis of the genome of strain I-329 maintained in the collection and metagenomes of industrial velums revealed only several dozens of single nucleotide polymorphisms, which indicates a long-term genetic stability of this flor yeast strain under the harsh conditions of biological wine aging.

SummaryThe effect of selective activation of platelet prostaglandin (PG) E2 receptor subtype EP2 or EP4 on platelet aggregation remains to be determined. In platelets prepared from wild-type mice (WT platelets), high concentrations of PGE2 inhibited platelet aggregation induced by U-46619, a thromboxane receptor agonist. However, there was no significant change in the inhibitory effect of PGE2 on platelets lacking EP2 (EP2 –/– platelets) and EP4 (EP4 –/– platelets) compared with the inhibitory effect on WT platelets. On the other hand, AE1–259 and AE1–329, agonists for EP2 and EP4, respectively, potently inhibited U-46619 -induced aggregation with respective IC50 values of 590 ± 14 and 100 ± 4.9 nM in WT platelets, while the inhibition was significantly blunted in EP2 –/– and EP4 –/– platelets. In human platelets, AE1–259 and AE1–329 inhibited U-46619-induced aggregation with respective IC50 values of 640 ± 16 and 2.3 ± 0.3 nM. Notably, the inhibitory potency of AE1–329 in human platelets was much higher than that in murine platelets, while such a difference was not observed in the inhibitory potency of AE1–259. AE1–329 also inhibited adenosine diphosphate-induced platelet aggregation, and the inhibition was almost completely blocked by AE3–208, an EP4 antagonist. In addition, AE1–329 increased intracellular cAMP concentrations in a concentration- and EP4-dependent manner in human platelets. These results indicate that selective activation of EP2 or EP4 can inhibit platelet aggregation and that EP4 agonists are particularly promising as novel anti-platelet agents.

A problem-based learning (PBL) curriculum was introduced at McMaster University more than three decades ago. Not many schools have adopted the system despite its distinct advantages. The present paper examines the challenges of teaching physiology in a PBL curriculum and gleans through the literature supporting PBL. It appears that one of the reasons why PBL is not becoming readily acceptable is the lack of concrete reports evaluating the curricular outcomes. The suggestion (R.E. Thomas. Med Educ. 31:320-329, 1997) to standardize and internationalize all components of validated PBL curricula is quite valid. A database needs to be generated that can be easily accessed by traditional institutions to see the rationality and easy implementation of the PBL curriculum.

We examined, by using a specific PGE receptor subtype EP4 agonist and antagonist, the involvement of EP4 receptors in duodenal HCO3− secretion induced by PGE2 and mucosal acidification in rats. Mucosal acidification was achieved by exposing a duodenal loop to 10 mM HCl for 10 min, and various EP agonists were given intravenously 10 min before the acidification. Secretion of HCO3− was dose-dependently stimulated by AE1-329 (EP4 agonist), the maximal response being equivalent to that induced by sulprostone (EP1/EP3 agonist) or PGE2. The stimulatory action of AE1-329 and PGE2 but not sulprostone was attenuated by AE3-208, a specific EP4 antagonist. This antagonist also significantly mitigated the acid-induced HCO3− secretion. Coadministration of sulprostone and AE1-329 caused a greater secretory response than either agent alone. IBMX potentiated the stimulatory action of both sulprostone and AE1-329, whereas verapamil mitigated the effect of sulprostone but not AE1-329. Chemical ablation of capsaicin-sensitive afferent neurons did not affect the response to any of the EP agonists used. We conclude that EP4 receptors are involved in the duodenal HCO3− response induced by PGE2 or acidification in addition to EP3 receptors. The process by which HCO3− is secreted through these receptors differs regarding second-messenger coupling. Stimulation through EP4 receptors is mediated by cAMP, whereas that through EP3 receptors is regulated by both cAMP and Ca2+; yet there is cooperation between the actions mediated by these two receptors. The neuronal reflex pathway is not involved in stimulatory actions of these prostanoids.

Abstract Triple-negative breast cancer (TNBC) represents 20% of breast cancer (BC) patients (51,000 cases/year in the US). It is associated with a highly aggressive course, early metastasis and relapses, and drug-resistant phenotype. About 40% of the patients die within 5 years despite available therapies. If the disease is metastatic, patients survive only 13 months. Since TNBC does not express targetable receptors such as ER, PR, and HER2, and it is a highly heterogeneous cancer with six genetically defined sub-types, currently, there are no common actionable molecular targets and no effective targeted therapies for TNBC. microRNAs (miRNAs) are 18-20 nt long non-coding RNAs and are often dysregulated in cancer cells. Tumor-suppressive miRNAs specifically silence their protooncogenic target genes (mRNAs) by binding specifically to their 3’-UTRs located in their target mRNAs, leading to inhibition of oncogenes that play critical roles in cell proliferation, cell cycle, migration, invasion, angiogenesis, drug resistance, tumor growth, and progression. We have previously demonstrated that Eukaryotic Elongation factor-2 kinase (eEF2K) and AXL are oncogenic kinases, and their higher expression is associated with poor survival in patients with TNBC. However, currently, there is no FDA-approved eEF2K and AXL-targeted therapeutics. To effectively co-target these kinases, we extensively analyzed miRNA databases using miRNA target prediction algorithms and recently, we discovered that miR-329-3p has a specific binding site at both 3’-UTRs of eEF2K and AXL mRNAs. Analyzing the TCGA TNBC database, we found that low expression of miR-329-3p is correlated with shorter overall survival in TNBC patients, and the expression of miR-329-3p is commonly reduced or lost in TNBC patient tumors. Since the successful clinical application of miRNA-based therapeutics requires safe and effective nano-delivery systems, we have developed magnetic iron-oxide-based nanoparticles (MNPs) for the delivery of miR-329-3p. We showed that miR-329-3p significantly suppresses cell proliferation, invasion, and migration by targeting both eEF2K and AXL and their downstream mediators such as SRC/FAK and Cyclin D1 in TNBC cell lines. In vivo systemic treatment with MNPs-miR-329 completely blocks tumor growth in orthotopic MDA-MB-231 and MDA-MB-436 TNBC xenograft models in mice. Analysis of tumors shows remarkable inhibition in eEF2K and AXL expression, and clinically significant downstream targets including SRC, FAK, and Cyclin D1. We also demonstrated that miR-329 nanotherapy induces apoptosis and inhibits intratumoral proliferation in TNBC tumors in mice. Toxicity markers in mouse blood samples show that miR-329 nanotherapy is safe and not toxic to mice. Taken together, our study suggests that miR-329-3p nanotherapy may be used as a novel potential therapeutic approach in TNBC patients. The hypothesis that peripherally administered cholecystokinin C-terminal octapeptide (CCK-8) and endogenous CCK act by the same abdominal vagal mechanism to produce satiety was tested by injecting rats with CCK-8 or the type A CCK receptor antagonist MK-329 after they had received bilateral subdiaphragmatic vagotomies. CCK-8 (8 nmol/kg ip) inhibited 1-h food intake by 60%; vagotomy and MK-329 (0.5 mg/kg sc) each completely blocked this effect. In contrast, vagotomy did not alter the stimulatory effect of MK-329 (0.5 mg/kg sc) on feeding; 3-h cumulative intake in control and vagotomized animals was increased by 25 and 34%, respectively. These results suggest that satiety is mediated in part by an endogenous CCK action that is independent of abdominal vagal innervation.

Bovine oviduct epithelial cells (BOEC) are difficult to culture without dedifferentiation. Frequently observed changes in morphology during culture suggest that gene expression patterns are affected as well. We explored a novel short-term culture system for BOEC – suitable for co-culture experiments with embryos – and evaluated the cells with respect to morphological criteria, candidate gene expression, and hormone responsiveness. Simmental heifers were slaughtered on Day 3.5 after standing heat and BOEC were obtained by squeezing along the ampulla with forceps. The cell sheets were separated mechanically by repeated passages through syringes and pipetting, and recovered by sedimentation. Cells from the ipsi- and contralateral oviduct were cultured separately at a density of 106 cells per well in 24-well plates with 800 μL TCM-199 supplemented with 2% OCS (estrous cow serum, as used in embryo culture) or CS 3.5 (cow serum, Day 3.5 after standing heat, adequate to the cycle stage in which cells were obtained) and 0.25 mg/mL gentamicin. For cell counting, an aliquot was further disaggregated by passing 15 times through a 30-gauge needle to achieve a single-cell suspension. Culture took place at 38°C in a humidified atmosphere of 5% CO2 in air. Cells were examined by light microscopy at seeding and after 6, 12, 24, and 48 h. Cell aggregates showed a worm-like structure, displaying numerous vigorously beating cilia on their surfaces throughout the culture period. Trypan blue staining indicated that cells contained in aggregates were viable while single cells stained predominantly positive (non-viable). The purity of the epithelial cell culture was >95%, as determined by immunohistochemistry using antibodies against vimentin and cytokeratin. For electron microscopic investigations, BOEC were sampled at seeding and after 24 h in culture. Cultured BOEC showed a morphology highly similar to that of BOEC in vivo. Both secretory cells with numerous secretory granules and mitochondria, and ciliated cells with long, well developed and actively moving kinocilia were visible. RT-PCR data for candidate genes (ERα, ERβ, HMGCR, PHGPx, PR) obtained from BOEC samples at seeding and after 6, 12, 24, and 48 h in culture showed that gene expression was stable for the majority of transcripts after 6 h in culture. There was no significant difference between cells cultured with OCS or CS 3.5 and no difference between cells obtained from the ipsi- or contralateral oviduct. Estradiol-17β (E2, 10 pg/mL) or progesterone (P4, 10 ng/mL) stimulation showed that the cultured BOEC are able to respond to hormonal signals in a manner similar to their reaction in vivo (Ulbrich et al. 2003 J. Steroid. Biochem. Mol. Biol. 84, 279–89). Progesterone receptor mRNA was up-regulated by E2 and estrogen receptor β mRNA was up-regulated by P4. The culture system for bovine oviduct epithelial cells thus provides an adequate tool to investigate mechanisms of the embryo-maternal communication in cattle.
