Chemotherapy often results in cognitive impairment, and no neuroprotective drug is now available. This study aimed to understand underlying neurotoxicological mechanisms of anticancer drugs and to evaluate neuroprotective effects of PAN-811. Primary neurons in different concentrations of antioxidants (AOs) were insulted for 3 days with methotrexate (MTX), 5-fluorouracil (5-FU), or cisplatin (CDDP) in the absence or presence of PAN-811·Cl·H2O. The effect of PAN-811 on the anticancer activity of tested drugs was also examined using mouse and human cancer cells (BNLT3 and H460) to assess any negative interference. Cell membrane integrity, survival, and death and intramitochondrial reactive oxygen species (ROS) were measured. All tested anticancer drugs elicited neurotoxicity only under low levels of AO and elicited a ROS increase. These results suggested that ROS mediates neurotoxicity of tested anticancer drugs. PAN-811 dose-dependently suppressed increased ROS and blocked the neurotoxicity when neurons were insulted with a tested anticancer drug. PAN-811 did not interfere with anticancer activity of anticancer drugs against BNLT3 cells. PAN-811 did not inhibit MTX-induced death of H460 cells but, interestingly, demonstrated a synergistic effect with 5-FU or CDDP in reducing cancer cell viability. Thus, PAN-811 can be a potent drug candidate for chemotherapy-induced cognitive impairment.

Introduction: Telephone Triage Services (TTS) manage phone calls from the public regarding general medical problems and provide telephone advice. This telephone based care can overlap with care provided by Poison Centres. Our objective was to examine the impact of a provincial 811 TTS on the IWK Regional Poison Centre (RPC). Methods: This is a retrospective descriptive study using interrupted time series methodology. We compared monthly IWK RPC call volume in the pre-811 era (January 2007-July 2009) and the post-811 era (September 2009-December 2017). We summarized the characteristics of callers who accessed the IWK RPC in terms of client age, sex, intentionality, time of day, call disposition and outcome. Caller characteristics were compared between the pre- and post-811 eras using chi-square test for categorical variables. We used segmented regression analysis to evaluate changes in slope of call volume in the pre- and post 811 eras. The Durbin-Watson statistic was performed to test for serial correlation and the Dickey-Fuller test to investigate seasonality. Results: The dataset included 82683 calls to the IWK RPC – 27028 pre-811 and 55655 post-811. Overall, 55% of calls were for female clients and the largest age group was children aged 0-5 years (37%). Most calls originated from home (47%), followed by a health care facility (23%). Most calls were managed at home (65%). Less than 3% of calls resulted in major effect or death. The Durbin Watson statistic was not statistically significant (p = 0.94). The Dickey-Fuller test indicated series stationarity (p = 0.001). There was no statistically significant change in call volume to the IWK RPC due to the introduction of 811 (p = 0.39). There was no significant variation by time of day, day of week or month, with most calls occurring in the evening. There were significantly more calls regarding intentional ingestions in the post-811 era (23% vs. 19% pre-811, p < .001). Outcomes in the pre and post 811 eras were as follows: minor/no effect/non-toxic/minimal 80% vs. 78%; moderate 7% vs. 10%; and, major/death 1.7% vs. 2.0%. Conclusion: The introduction of a TTS did not change call volumes at our RPC. The increase in the percentage of calls about intentional ingestions may reflect an increase in call acuity as the 811-TTS likely manages calls about minor/non-toxic ingestions without consulting with the RPC. Our future research will examine the nature of poison related calls to the 811-TTS.

Background: Complications and adverse events after anterior cruciate ligament (ACL) reconstruction are well known, but they have been underestimated in previous studies. Purpose: To describe the complications and adverse events after ACL reconstruction within a 2-year follow-up and analyze them in relation to the type of graft. Study Design: Cohort study; Level of evidence, 3. Methods: From 2000 to 2012, 958 patients with an isolated ACL injury underwent surgery by a single knee surgeon. ACL reconstruction was performed with the medial portal technique for the femoral tunnel and the use of bone–patellar tendon–bone (BPTB) or hamstring tendon graft. Patients were reviewed at 6 weeks and 3, 6, 12, and 24 months after surgery with the International Knee Documentation Committee score, plain radiographs, and the KT-1000 arthrometer. Results: Of 958 patients enrolled, 147 (15%) were lost at last follow-up. The 2 groups (bone–patellar tendon–bone [n = 257] and hamstring [n = 554]) were similar regarding the mean age at the time of surgery and preoperative anterior laxity. The main complications were as follows: anterior knee pain (n = 130 of 811, 16%), stiffness (n = 72, 8.8%), secondary meniscal lesions (n = 59, 7.2%), pain attributed to fixation (n = 79, 9.7%), ACL rerupture (n = 47, 5.7%), contralateral ACL ruptures (n = 24, 3%), patellar fractures (n = 3, 0.3%), infections (n = 9, 1%), and thromboembolic complications (n = 5, 0.6%). There was no significant difference between the grafts with respect to the frequency of joint stiffness, secondary meniscal lesions, or anterior knee pain. During the first 2 postoperative years, the percentage of patients with anterior knee pain was higher in the patellar tendon group (23.3% vs 12.6%, P < .001); however, this difference was not significant after the 2-year interval (3.1% vs 2.5%, P = .63). The percentage of patients with a rerupture of the graft was significantly lower in the patellar tendon group than in the hamstring group (25 of 811 [3.1%] vs 57 of 811 [7%], P = .023). Similar results were recorded regarding the pain related to the hardware material (7 of 811 [0.8%] in the BPTB group vs 113 of 811 [13.9%] in the hamstring group, P = .001). The percentage of ACL ruptures contralateral to the repair was higher in the patellar tendon group (41 of 811 [5%] vs 17 of 811 [2%], P = .016). Conclusion: The total rate of complications after an ACL reconstruction was 39%, and the surgical revision rate for any reason was 28%. Problems with the hardware material were more frequent in the hamstring group, leading to an increased rate of surgical revision. Anterior knee pain was initially higher in the patellar tendon group, but there was no significant difference in a 2-year interval. The rerupture rate was statistically higher in the hamstring group.

За статистикою у 2021 р. в Україні діяло 1 880 858 (96%) мікропідприємств, 56 969 (3%) малих підприємств, 17 811 (0,9%) середніх підприємств та 610 (0,1%) великих підприємств. Великі підприємства займали 33,7 % у структурі загальної виробленої продукції, середні – 34,4 %, малі – 31,9%, з них мікропідприємства – 18,2%. [3].

Transferrin (Tf) is an iron transport protein expressed in MCF-7 human breast cancer cells. In nuclear run-on assays, 17beta-estradiol (E2) increased the rate of Tf gene expression approximately 3-fold within 1 h after treatment and reporter gene activity was also induced in MCF-7 cells transfected with a construct containing a -3600 to +39 Tf gene promoter insert. Deletion and mutation analysis identified an E2-responsive promoter region between -811 and -762, which was GC-rich (80%) and contained two nonconsensus estrogen response elements (EREs). E2-responsiveness of this region was associated with a GGACA(N)(3)TGGCC motif (-803 to -791) which bound human estrogen receptor alpha (hERalpha) in gel mobility shift assays. In Drosophila Schneider SL-2 cells, the -811 to -752 was E2-responsive after cotransfection with hERalpha expression plasmid plus E2, whereas Sp1 protein did not induce transactivation. These studies confirm that E2 induces Tf gene expression through a nonconsensus distal ERE.

Abstract (sommario):
The molecular mechanism for colorectal cancer to develop remains unelucidated. To find biomarkers related to colorectal cancer development, we analyzed the gene expression profile of 380 colorectal cancer patients and 51 healthy controls by R software. Finally, 1579 upregulated differential expression genes (DEGs) and 3218 downregulated DEGs were identified. Then, the top 20 upregulated DEGs were compared with 181 upregulated DEGs that we reported previously, and 11 overlapped DEGs were found. NFE2L3 (nuclear factor, erythroid 2-like 3) was among those overlapped DEGs and was rarely reported in colorectal cancer. Real-time polymerase chain reaction (PCR) results showed that higher NFE2L3 expression levels were identified in paired tumor samples than in paratumor samples (48 paired samples). Flow cytometry analysis revealed that the cell cycle was arrested at the G0/G1 phase after inhibition of NFE2L3 in both HCT116 and SW480 cell lines. Western blot detection showed that CCND1 and phosphorylated Rb transcriptional corepressor 1 at ser-807/811 (pRb1-ser807/811) expression levels were downregulated when NFE2L3 was inhibited in those two cell lines. A significant positive correlation was observed between NFE2L3 and CCND1 expression levels in colorectal tissue samples. These evidences indicate that downregulation of NFE2L3 induces cell cycle arrest at the G0/G1 phase through downregulation of CCND1 and pRb1-ser807/811.

s Looking Backward. The major concern is to question the validity of title &lsquo
socialist&rsquo
for these two texts. The reference points for such an analysis are: modernity, Marxism of the late 19th century and the practice of discipline. In this context, the intention is to find out ruptures and continuities with respect to the central ideas of socialism and basic premises of modernity. The study explorates that there are serious points of rupture in these two texts with respect to the major premises of modernity, because in Morris&rsquo
s utopia there is a romantic search for restoring communism of the 14th century, in Bellamy&rsquo
s text there are typical reactionary modernist suggestions concerning the nature of typical socialist societies. In that sense, due to the disassociation between socialism and modernity in these two texts, it is very problematic to classify these utopias as socialist. The study also questions whether the sources of such disassociation are embedded in Marxism itself. In response to such question, the study argues that this is the case to a great extent.

AbstractWhile first-in-family women’s experience of attending university has been examined in a growing body of literature, there has been little attention paid to the experiences of first-in-family males. This chapter presents an account of the motivations, transitions and participations of first-in-family male students using a narrative gender framework. The analysis especially privileges the idea of situated and relational masculinities (Hopkins & Noble, Masculinities in place: Situated identities, relations and intersectionality. Social and Cultural Geography, 10(8), 811–819, 2009). Age was found to be the chief organising category of their experiences structuring their embodied life course. Three main age and relational masculine performances emerged from these men’s stories, namely those of the Fathers, the Self-Starters and the Sons. Working to achieve or enact the breadwinner model of masculinity was found to be the dominant motivator behind their gender performances.

The author offers new possibilities for interpretation of Frankish, domestic and Byzantine sources regarding the Bulgarian political control over territories of Avar Khaganate, destroyed by Charlemagne. The main focus is placed on the certificate of embassy led by Princeрs Avarum and Canizauci in Aachen in November 811. Coordinating it with Bulgarian and Byzantine sources, leads to the following conclusions. 1. It is likely that the diplomatic mission to Charlemagne in 811, involving representatives of the Avar com­munity, led by its Tudun and Slavic tribal princes, was led by the Bulgarian prince – Omurtag, the younger brother of the ruler Krum (802? – 814), in his capacity as prince (princeps) and ombritag. i. e. Avars hegem­on, in the northwestern borders after 803 and „Khan’s beloved younger brother” (khani sev`ingi or khani sev(inč) ingi). In Aachen, he introduced himself as a cani zautzi, that is, with his post of „Khan’s envoy”. 2. The khanas uvigi Omurtag (814–831) missions to Emperor Louis in 824 and 825–826 appear to have also been led by a member of the ruling family in Plisk oba (Pliska), maybe from his second son – Zvinitsa/Zvinichis. They also appear to have had a representative/s of settlers between 813 and 837 in Trans­Danubian Bulgaria (probably in the Lower Tisza region) of Bulgarian captives of Eastern Thrace of Armenian origin. One of their leaders in 837 was named Tzantzès, and his son, Stilian, and his descendants gained fame in Byzantium under the surname Ζαούτζης, Ζαούτζας. It coincides exactly with the pro­Bulgarian official title (position) zautzi (tzautci), (=chaush) and probably derived from it. On this basis, we conclude that Τζάντζην (Öан¤·þ воеводэ) was performing the carrier of messages or emissary functions of the Bulgarian state before 837. 3. The considered evidence, facts and circumstances surrounding the Bulgarian diplomatic missions of 811, 824 and 825/6 provide new testifies for the Bulgarian state’s control over the south-eastern parts of the Avar Khaganate after its collapse in the period 791–803. They have a contribution to clarify important aspects of the Bulgarian state’s relations with the East Frankish Kingdom, as well as with the local population of Avars, Bulgarians and Slavs there.

Most work on erbium-doped fiber amplifiers (EDFA) over the past few years has focussed on amplifiers pumped at 980 nm and at 1480 nm.1-3 Recently, however, there has been renewed interest for file 800-nm pump band.4 In this paper we present a comparative study of an EDFA pumped by a Ti:sapphire laser tuned to both 800 and 980 nm in order to illustrate the effects of excited-state absorption.5 The erbium-doped fiber used in the experiment has a 4-μm-diameter SiO2-Al2O3-GeO2 core with a relative index difference of 0.035 and a cutoff wavelength of ~815 nm. The erbium concentration is 200 ppm. Gain and saturation measurements were per formed on different lengths of fiber, pumped at 980 nm and at some arbitrary wavelength in the 810-nm region, the optimum wavelength having been found to increase with length. Figure 1 is a plot of normalized gain for λs = 1533 nm as a function of pump wavelength. The 3-dB bandwidth is 14 nm. The peak gain for this 12-m piece is obtained for λ p = 811 nm and was 25 dB for λ s = 1533 nm and 20 dB for λ s = 1553 nm. The maximum absorption, however, was found to be at 797 nm.

Abstract. Microstructured sago (Metroxylon sagu) possesses an excellent potential to be used for numerous applications. In this paper, sago starch microparticles (SMPs) were fabricated using an ultra-fine grinder without any chemical treatments or purification steps thus making it a “green” and an eco-friendly procedure. The effects on increasing the numbers of passages through the grinder on the resulting SMPs and as suspension films were investigated. We observed that after 7 cycles the average diameter was reduced to a microscale region of 0.811 ± 0.754 µm (811 nm). The highest tensile strength was shown to be the highest after 3 cycles of 2.308 ± 0.210 MPa and repeated cycles beyond 7-cycled SMPs process had resulted in damaged microparticles. Furthermore, water vapor permeability was observed in SMPs with higher number of grinding cycles, with 7-cycled SMPs being 15.522 ± 0.184 g mm/m2 h Pa and 10-cycled SMPs, 19.763± 0.233 g mm/m2 h Pa. Our results demonstrated that the final particle size, water vapor permeability and tensile strength of the SMPs suspensions films were affected by the number of passages through the mechanical grinder.

ЦЕЛЬ: оценить состояние углеводного обмена у долгожителей Москвы и близлежащих регионов. МАТЕРИАЛЫ И МЕТОДЫ: в это когортное исследование включались лица в возрасте 90 лет и старше, давшие письменное согласие на участие. В домашних условиях проводилась комплексная гериатрическая оценка, включавшая сбор анамнеза, применение стандартных гериатрических шкал и опросников, выполнение антропометрических измерений. Всем участникам оценивались следующие лабораторные показатели: глюкоза, инсулин, индекс инсулинорезистентности HOMA-IR, гликированный гемоглобин (HbA1c). Интерпретация состояния углеводного обмена проводилась согласно актуальным на 2022 г. клиническим рекомендациям. Статистический анализ данных проводился с помощью языка программирования R версии 4.1.3. Количественные переменные представлены в виде Me (Q1-Q3). Для сравнения непараметрических значений применялся критерий Краскела-Уоллиса, номинальных переменных – χ2 Пирсона или точный критерий Фишера. Исследование было одобрено локальным этическим комитетом. РЕЗУЛЬТАТЫ: за период с 2019 по 2022 г. в исследование было включено 3 811 человек (75,4% женщины) в возрасте 92 (91–94) года. Средний индекс массы тела составил 25,4 (23-28,2) кг/м2, доля участников с ожирением составила 15,2%, при этом у 72,9% пациентов были значения окружности талии соответствовали абдоминальному ожирению. Средние значения глюкозы составили 5 (4,5 – 5,7) ммоль/л, HbA1c 5,6 (5,4 – 5,9)%, HOMA-IR – 1,5 (0,9 – 2,6). В группу СД были отнесены 20,5% (780); предиабета – 16,1% (613). Все случаи СД были интерпретированы как 2 тип. В группе с СД большая часть пациентов 71,7% имела диагноз на основании данных анамнеза; у 28,3% пациентов были впервые выявлены значения параметров углеводного обмена в диабетическом диапазоне. С клинической точки зрения, значения показателей углеводного обмена в группе пациентов с СД были близки к значениям в группе предиабета: глюкоза - 6,3 (5,1 - 7,9) и 5,9 (5 - 6,4) ммоль/л, HbA1c 6,2 (5,6 - 7) и 6,1 (6 - 6,2)%, HOMA-IR 2,3 (1,3 - 6,1) и 2,2 (1,4 - 4,5), соответственно. При этом лишь 4,8% (38) участников с СД получали сахароснижающую терапию (ССТ): 3,8% (30) монтерапию инсулином, 0,6% (5) – терапию пероральными сахароснижающими препаратами, 0,4% (3) – комбинированную терапию. Несмотря на то, что в группе с впервые выявленным СД значения показателей углеводного обмена были статистически значимо выше, чем в группе с ранее выявленным СД (глюкоза 7,4 и 5,9 ммоль/л; HbA1c 6,8 и 6,1%, соответственно), они всё равно были ниже предполагаемых целевых значений для этой возрастной категории. Дополнительно пациенты были разделены на возрастные подгруппы: 90-94 лет (3091), 95-99 лет (674), >100 лет (46); достоверных различий в значениях показателей глюкозы и HbA1c, частоте СД и предиабета между группами получено не было. ВЫВОДЫ: доля пациентов с СД в исследованной когорте долгожителей была практически сопоставима с данными о распространённости СД в группе пожилых людей, при этом показатели углеводного обмена указывали на компенсацию углеводного обмена даже при низкой доле пациентов, получающих ССТ.

Распространённость аденом гипофиза в популяции высокая, агрессивное течение заболевания преимущественно наблюдается при семейных формах. Поиск новых генов, вовлечённых в процесс образования аденом гипофиза является важным фактором ранней диагностики моногенных заболеваний. В ГНЦ ФГБУ «НМИЦ эндокринологии» Минздрава России внедрены технологии высокопроизводительного параллельного секвенирования (Next generation sequencing, NGS) и разработана панель генов-кандидатов, включающая следующие 15 генов MEN1, CDKN1B, PRKAR1A, GNAS, AIP, SDHA, SDHB, SDHC, SDHD, PRKCA, CDKN2C, CDKN2A, POU1F1, PTTG2, DICER1. Задачей настоящего исследования стала оценка частоты мутаций в генах среди 36 пациентов с семейными формами аденом гипофиза. По результатам генетического тестирования методом NGS среди 36 представителей семей у 7 отмечены следующие изменения нуклеотидной последовательности: мутация в гене AIP:NM_003977.2:exon 5:c.783 C>G:p.Tyr261 у пациента с семейной соматотропиномой; мутация в гене AIP:NM_003977.2:exon 6: c.811 C>T:p. R271W у двух больных в семье с соматотропиномами; мутация в гене AIP:NM_003977.4:exon :c.649 С>Т:р. A411GfsX47 у двух лиц с семейными соматотропиномами/пролактиномами в одной семье; у одного пациента с семейной пролактиномой идентифицирована мутация в гене DICER1:NM_177438:exon 16:c.2553 G>A:p. Q851Q – синонимичная мутация – замена с недоказанным патологическим значением; у одного больного с семейной пролактиномой выявлено нарушение нуклеотидной последовательности в гене DICER1:NM_ 177438.2:exon 16: c.2613 C>A:p.D871E – несинонимичная замена с недоказанным патологическим значением; у одной пациентки с семейной тиреосоматотропиномой/соматотропиномой обнаружено изменение в гене DICER1:NM_177438:exon 2:c.20 A>G:p.Q7R – несинонимичная мутация и в гене SDHA:NM_004168:exon 8:c.1002 G>A:p.A334A – синонимичная мутация с недоказанным патологическим значением; мутация в гене GNAS: NM_080425:exon 1: c.314 A>G:p.E105G подтверждена у одного пробанда с синдромом МАS и семейной соматотропиномой. Частота мутаций в исследуемых 15 генах-кандидатов у пациентов с семейными формами аденом гипофиза составила 20% случаев, в 8% из которых идентифицированы изменения нуклеотидной последовательности в генах AIP и DICER1, в 3% – в генах SDHA и GNAS. Таким образом, применение метода высокопроизводительного параллельного секвенирования NGS, позволило предположить наличие крупных делеций в генах, сократило временные и материальные затраты на диагностику, что является предпочтительным для установления генетической природы заболевания лицам с семейными формами аденом гипофиза по сравнению с прямым секвенированием каждого гена в отдельности.

Abstract The carbonate reservoir S is a giant limestone reservoir in H Oilfield, Iraq. Although the reserves account for 25%, the production contribution is only 0.4% to the total oilfield production due to poor petrophysical properties. Accordingly, the first proppant fracturing on vertical well was successfully executed in December 2016, which has already achieved a steady production period over than 3 years. In order to further improve the productivity, the first multi-stage proppant fracturing(MSPF) on horizontal well(SH01X) was successfully applied in November 2019, a technique which is rarely reported for porous limestone reservoir in the Middle East. Proppant fracturing in carbonate reservoirs is a technique difficulty worldwide, especially this is a lack of experiences in the Middle East. To ensure the success of this campaign, a holistic technical study including geology evaluation, reservoir performance analysis, drilling trajectory design, completion and fracturing technique design have been carried out based on principle of "geology-engineering integration". This paper will present a comprehensive illustration including treatment design (main completion-fracturing technique, total scale, fracturing fluid, proppant), job execution (mini-frac, main-frac) and post-frac production performance for this successful campaign. True vertical depth (TVD) of Well SH01X is 2720 m and the horizontal section length is 811 m. Based on the main technique of multi-stage proppant fracturing with open hole packers and sliding sleeves, totally 3784.3 m3 fracturing fluid and 452 m3 proppant were pumped in 8 stages. The test production was 3214 BOPD (choke size: 40/64", wellhead pressure: 970 psi). A historical breakthrough in the productivity of S reservoir has been achieved by the campaign. The post-frac evaluation shows that the treatment parameters are consistent with the design. The connectivity between artificial fractures and formation is greatly improved, and the stimulation effect is significant. Currently the "production under controlled pressure" mode has been executed and the stable production under stimulation target rate has been maintained. The systematic "geology-engineering integration" workflow is of significance to the success of the treatment as well as the stimulation effect. MSPF is planned to be a game-changing technique to develop the huge reserves of S reservoir. The experience gained from this case could provide theoretical as well as practical references for similar reservoirs in the Middle East.
