In the study, the authors conducted a multidimensional data analysis on the decrease in the number of passengers transported by air in Poland and Germany caused by the COVID-19 pandemic between 2020-2022. Forecasts of the number of passengers transported by air in Poland and Germany obtained after applying the Holt-Winters exponential smoothing method from January 2020 to December 2022 were compared with actual data. The research showed that in the case of Germany, in 2020 there was a decrease of 174 628 873 people, and in 2022, it was 90 620 354 people. In Poland, the decrease in 2020 amounted to 40 257 520 people; in 2022, it was already 21 083 066 passengers.

Abstract (sommario):
Background To assess the prognostic value of early echocardiographic indices of right ventricular function and vasoactive peptides for prediction of bronchopulmonary dysplasia (BPD) or death in very preterm infants. Methods Prospective study involving 294 very preterm infants (median [IQR] gestational age 28.4 [26.4–30.4] weeks, birth weight 1065 [800–1380] g), of whom 57 developed BPD (oxygen supplementation at 36 weeks postmenstrual age) and 10 died. Tricuspid annular plane systolic excursion (TAPSE), right ventricular index of myocardial performance (RIMP), plasma concentrations of mid-regional pro-atrial natriuretic peptide (MR-proANP) and C-terminal pro-endothelin-1 (CT-proET1) were measured on day 7 of life. Results RIMP was significantly increased (median [IQR] 0.3 [0.23–0.38] vs 0.22 [0.15–0.29]), TAPSE decreased (median [IQR] 5.0 [5.0–6.0] vs 6.0 [5.4–7.0] mm), MR-proANP increased (median [IQR] 784 [540–936] vs 353 [247–625] pmol/L), and CT-proET1 increased (median [IQR] 249 [190–345] vs 199 [158–284] pmol/L) in infants who developed BPD or died, as compared to controls. All variables showed significant but weak correlations with each other (rS -0.182 to 0.359) and predicted BPD/death with similar accuracy (areas under receiver operator characteristic curves 0.62 to 0.77). Multiple regression revealed only RIMP and birth weight as independent predictors of BPD or death. Conclusions Vasoactive peptide concentrations and echocardiographic assessment employing standardized measures, notably RIMP, on day 7 of life are useful to identify preterm infants at increased risk for BPD or death.

Abstract Background. OM is a common toxicity of conditioning regimens for HSCT that has been associated with adverse clinical and economic outcomes. Methods. A retrospective chart review study of ~400 consecutive HSCT recipients with hematologic malignancies was undertaken at a single academic center. OM severity was assessed across eight oropharyngeal sites using a validated scale which was scored as follows: no erythema/ulceration = 0; erythema only = I; ulceration one site = II; two sites = III; three sites= IV; and four or more sites = V. OM assessments began on the day of conditioning and continued bi-weekly until hospital discharge. Outcomes of interest included number of days of fever, total parenteral nutrition (TPN), and parenteral narcotic therapy, incidence of Grade III or IV (Common Toxicity Criteria) infection, and inpatient days and charges. We report data here for the 281 AL HSCT recipients in the study cohort. Results. Mean age was 41 yrs. Most common diagnoses were acute (33%) and chronic (19%) myeloid leukemia. Most patients (96%) received total body irradiation. Fifty-one percent of grafts were from unrelated donors. Seventy-six percent of patients experienced OM ≥ grade II. The relationship between worst recorded grade of OM and each of the outcomes of interest is reported below. Conclusions. OM is associated with worse clinical and economic outcomes in patients with hematologic malignancies undergoing AL HSCT. Hami, Xinjiang, is located in an extremely arid region and jujube is the main economic crop. It is important to adopt dwarf and close-planting technology under drip irrigation and to optimize water and fertilizer management to improve jujube yield and quality. Local 12-year-old jujube trees were treated using two factors of irrigation and fertilization. Three irrigation amounts (520, 700, and 880 mm) and three fertilization levels (248, 318, and 388 kg/hm2) were set up in plot experiments. The root system, yield, and quality of jujube were monitored under different irrigation and nitrogen application combinations. The effects on water use efficiency (WUE), nitrogen partial factor productivity (PNP), and economic benefit were analyzed. The water and fertilizer coupling effect of dwarf closely planted jujube was studied by combining multiple regression and spatial analyses. The yield, quality, economic benefits, WUE, and PNP of jujube were significantly affected by irrigation and fertilization. Multiple regression and spatial analyses showed that the highest yield was for irrigation of 700 mm and a nitrogen rate of 340 kg/hm2. For the maximum net benefit, irrigation was 806 mm and nitrogen was 388 kg/hm2; correspondingly, for the highest total sugar, values were 688 mm and 362 kg/hm2; for vitamin C, they were 622 mm and 376 kg/hm2; for PNP, they were 880 mm and 256.57 kg/hm2; and for WUE, they were 520 mm and 388 kg/hm2. A comprehensive evaluation of each index showed that the acceptable irrigation amount for yield, quality, WUE, and net benefit of ≥85% of the maximum value was 600–628 mm, and the nitrogen application rate was 360–372 kg/hm2. This study provides guidance for the scientific management of water and fertilizer in the drip irrigation and fertilization process of local jujube trees.

Squeeze-cast 20vol%SiCw/2024 composite was processed by rotary-die equal-channel angular pressing (RD-ECAP) and tensile-tested at elevated temperatures. RD-ECAP does not require billet removal and reinsertion between ECAP passes and can produce large cumulative deformation of type A in the billet at a controlled extrusion temperature. Tensile specimens were machined from the RD-ECAP processed billets along the longitudinal direction. 16 RD-ECAP passes at 623 K produced an average matrix grain size of about 0.8 μm in the composite. The ECAP processed 20vol%SiCw/2024 composite exhibited 330% elongation to failure when tested at 783 K and the high strain rate of 1.17×10-1 s-1. The values of strain rate sensitivity exponent of the SiCw/2024 composite determined over the strain rates between 10-2 and 1.0 s-1, was 0.39, 0.47 and 0.53 at 763, 773 and 783 K, respectively. The grain sizes of the aluminum alloy matrix in the composite after superplastic deformation increased from 0.8 μm to be about 1.5 μm. There was no cavity found within the uniformly deformed area. The apparent activation energy for the superplastic deformation of the 20vol%SiCw/2024 composite was determined to be 284 kJ·mol-1.

Abstract (sommario):
Two extra virgin olive oil (EVOO) phenolic extracts (BUO and OMN) modulate DPP-IV activity. The in vitro DPP-IV activity assay was performed at the concentrations of 1, 10, 100, 500, and 1000 μg/mL, showing a dose-dependent inhibition by 6.8 ± 1.9, 17.4 ± 6.1, 37.9 ± 2.4, 57.8 ± 2.9, and 81 ± 1.4% for BUO and by 5.4 ± 1.7, 8.9 ± 0.4, 28.4 ± 7.2, 52 ± 1.3, and 77.5 ± 3.5% for OMN. Moreover, both BUO and OMN reduced the DPP-IV activity expressed by Caco-2 cells by 2.9 ± 0.7, 44.4 ± 0.7, 61.2 ± 1.8, and 85 ± 4.2% and by 3 ± 1.9, 35 ± 9.4, 60 ± 7.2, and 82 ± 2.8%, respectively, at the same doses. The concentration of the most abundant and representative secoiridoids within both extracts was analyzed by nuclear magnetic resonance (1H-NMR). Oleuropein, oleacein, oleocanthal, hydroxytyrosol, and tyrosol, tested alone, reduced the DPP-IV activity, with IC50 of 472.3 ± 21.7, 187 ± 11.4, 354.5 ± 12.7, 741.6 ± 35.7, and 1112 ± 55.6 µM, respectively. Finally, in silico molecular docking simulations permitted the study of the binding mode of these compounds.

Abstract Background: Bone marrow (BM) has been utilized as a source of stem cells for transplantation for many years. Although the use of BM has decreased with the advent of mobilized stem cells, utilization is increasing once again due to the lower rate of chronic GVHD associated with BM as a stem cell source. There is no generally accepted technique for harvesting BM. Protocols vary both in relation to the volume of each aspirate, the number of aspirates performed at each puncture site and the total volume of harvests. Method: BM was collected from the posterior iliac crests (PIC) in 2 separate bags: 10ml aspirates from the left and 20 ml aspirates from the right. Samples taken at the start and after 100, 150, 200, 250, & 500 ml were analyzed for TNC, CD34+ and CD3+ cell counts. By the time 500 ml has been aspirated, there is no difference in the total number of CD34+ cells obtained from a 10 ml versus a 20 ml aspirate of bone marrow. Conclusion: CD34+ cell yields fall rapidly when BM is harvested along the PIC. Using additional areas such as the anterior iliac crests may be preferable to a large volume PIC harvest for optimizing CD34+ stem cell collection. After 500 ml of BM has been harvested, 20 ml BM aspirates do not increase CD34+ cell numbers and 10 ml aspirates should be taken to minimize unnecessary blood loss and reduce T cell contamination.

O dendezeiro (Elaeis guineensis) é a maior fonte de óleo vegetal no mundo. Porém, a doença do amarelecimento fatal está dizimando plantações no estado do Pará, Brasil. Para superar esse problema, híbridos interespecíficos de Elaeis oleifera x Elaeis guineensis são uma alternativa viável. Contudo alguns protocolos para o resgate in vitro desses embriões já foram estabelecidos, porém é necessário otimizá-los. O objetivo dessa pesquisa foi otimizar o protocolo in vitro para o cultivo de embriões interespecíficos de (Elaeis oleifera x Elaeis guineensis). A desinfestação das sementes foi realizada com 1 ou 2% de NaOCl por 20 ou 40 minutos. Embriões foram germinados in vitro na presença de 0; 0,1; 0,2 e 0,25% de carvão ativado e embriões foram cultivados na presença do ácido 2,4-diclorofenoxiacético (2,4–D) nas concentrações de 375, 500 e 625 μM para a indução de calos. A adição de 2,5% de carvão ativado é necessária para reduzir a oxidação fenólica dos embriões. A desinfestação das sementes pode ser realizada com 1% de NaOCl por 20 minutos. Diferentes variedades possuem diferentes concentrações ideais de 2,4–D para a indução de calos. As melhores concentrações de 2,4–D foram 375 e 625 μM para as variedades SJ-167 e SJ-165, respectivamente.

The average composition is SiO2: 30.9, Al2O3: 58.6, FeO: 0.44, MgO: 0.47, CaO: 0.02, F: 0.15. Dumortierite crystallized from supercritical magmatic fluids at T =520-620°C, P = 100 ± 30 MPa. Synthetic fluid inclusions yielded T = 510 ± 10 MPa, P = 42 ± 3 MPa at 2.9 km depth in well Venelle 2. Al data suggest present day T = 450-550°C, P = 40-80 MPa. The supercritical magmatic fluids were hypersaline brines with ~ 30 wt % LiCl and up to 2.4 wt % F, extreme contents that can be found only in pegmatites, aplites and leucogranites. Larderello granites derived from partial melting of a lower crust made up of interlayered metasediments and amphibolites. Extensive melting took place in the lower crust during the last 10 Ma owing to extensional tectonics, lithosphere thinning, roll back or break down of the west merging Adria plate and swelling of the asthenosphere below the western side of the Apennines.

Abstract (sommario):
This study assessed epidemiologic data and clinical outcomes, including venous thromboembolism (VTE) recurrence and bleeding events, in patients with cancer-associated VTE, and assessed factors associated with clinical outcomes. Data were extracted from retrospective medical-chart review of adult patients diagnosed with cancer-associated deep vein thrombosis or pulmonary embolism who received anticoagulation treatment for ≥3 months. Patients were classified by: low-molecular-weight heparin (LMWH), direct oral anticoagulants (DOACs), and other anticoagulants. First VTE recurrence and bleeding events, and factors associated with their occurrence, were assessed during the initial 6 months of treatment. Overall, 623 patients (age: 63.7 ± 11.3 years, 49.3% male) were included (119, 132, and 372 patients in LMWH, DOACs and other anticoagulants groups, respectively). The cumulative 6-month incidence of VTE recurrence was 16.6% (total), 8.3% (LMWH), 16.7% (DOACs), and 20.7% (other); respective bleeding events were 22.5%, 11.0%, 12.3%, and 30.7%). VTE recurrence and bleeding rates differed only between LMWH and other anticoagulants (HR 2.4, 95% CI: 1.2-5.0 and 3.6, 1.9-6.8, respectively). These results highlight the importance of initial VTE treatment choice for preventing VTE recurrence and bleeding events. LMWH or DOACs for ≥3 months can be considered for effective VTE management in cancer patients.

Abstract The objective of this study was to evaluate the intestinal health and growth performance of nursery pigs affected by dietary supplementation of increasing levels of xylanase and b-glucanase combinations. Pigs [n = 40; initial body weight (BW) = 6.5 ± 0.4 kg] were assigned to 5 dietary treatments and fed for 35 d in 3 phases (11, 9, and 15 d, respectively). Basal diets mainly included corn, soybean meal, and distiller’s dried grains with solubles with phytase (750 FTU/kg) supplemented with 5 levels of xylanase and b-glucanase combinations (NSPase) at 1) 0, 2) 280 TXU/kg xylanase and 125 TGU/kg b-glucanase, 3) 560 and 250, 4) 840 and 375, or 5) 1,120 and 500, respectively. Growth performance and fecal score were measured for each phase. On d 35, all pigs were euthanized and collected jejunal mucosa, jejunal digesta, jejunal tissues, and ileal digesta to determine the effects of increasing NSPase levels (%) and NSPase intake on jejunal mucosa-associated microbiota, jejunal digesta viscosity, immune responses, intestinal morphology, feed digestibility, and growth performance. Increasing NSPase level quadratically decreased (P &lt; 0.05) Bacteroidetes (min: 0.7% at 636 TXU/kg and 284 TGU/kg), Prevotellaceae (min: 0.6% at 630 TXU/kg and 281 TGU/kg) in the jejunal mucosa. Increasing NSPase intake quadratically increased (P &lt; 0.05) Firmicutes (max: 72.7% at 604 TXU/kg and 270 TGU/kg), Lactobacillaceae (max: 40.3% at 608 TXU/kg and 271 TGU/kg), and Lactobacillus (max: 40.3% at 608 TXU/kg and 271 TGU/kg) in the jejunal mucosa. Increasing NSPase intake also quadratically decreased (P &lt; 0.05) Proteobacteria (min: 3.8% at 562 TXU/kg and 251 TGU/kg), Helicobacteraceae (min: 1.6% at 560 TXU/kg and 250 TGU/kg), and Helicobacter (min: 1.6% at 560 TXU/kg and 250 TGU/kg) in the jejunal mucosa. Increasing NSPase intake tended to quadratically decrease (P = 0.059) viscosity of jejunal digesta reaching the minimum (1.74 mPa·s) at 751 TXU/kg and 335 TGU/kg. Increasing NSPase level tended to linearly decrease (P = 0.073) jejunal IgG and tended to quadratically increase (P = 0.085) jejunal villus height to crypt depth ratio reaching the maximum (2.62) at 560 TXU/kg and 250 TGU/kg. Increasing NSPase intake tended to linearly increase the apparent ileal digestibility of dry matter (P = 0.087) and ether extract (P = 0.065). Increasing NSPase intake linearly increased (P &lt; 0.05) average daily gain (ADG). In conclusion, increasing the combinational use of xylanase and β-glucanase positively modulated jejunal mucosa-associated microbiota, reduced jejunal digesta viscosity, reduced intestinal humoral immune response, enhanced villus morphology, increased ileal digestibility of nutrients, and increased growth of nursery pigs. The beneficial effects were maximized when xylanase and β-glucanase were supplemented at a range of 550 to 800 TXU/kg and 246 to 357 TGU/kg, respectively.

The hemicellulose content of cellulosic material from poplar chemi-thermomechanical pulp (CTMP) was controlled by a microwave-assisted formic acid (MFA) extraction process. The environmentally friendly, self-bonded, and cellulose-rich fiber sheets (around 0.3 mm thick and 280 g/m2 grammage) were fabricated via a molding and compression process. The results showed that the MFA treatment effectively controlled the hemicellulose content in the range from 6.8% to 20.4%. Mechanical testing results showed a good linear relationship between the tensile strength and the hemicellulose content, where a higher hemicellulose content obtained a stronger tensile strength. Meanwhile, as hemicellulose content decreased, the onset temperature, representing the thermal stability, increased from 318.1 °C to 335.7 °C and the elastic modulus, representing the mechanical stiffness, increased from 0.047 MPa to 0.068 MPa. Additionally, as the hemicellulose content decreased, the water-resistance of the cellulose-rich fiber sheets improved. During the hemicellulose removal process, the strength of single fibers was found to have a dominant role in the mechanical strength of the cellulose-rich fiber sheets, rather than the inter-fiber bonding strength. This was contrary to research on the delignification process, where the inter-fiber bonding strength was found to be more important than the strength of single fibers in deciding the material strength.

<p>La formación y documentación del hato para la conservación del banco de germoplasma de la raza bovina criolla Romosinuano se inició en 1936; actualmente se cuenta con ocho familias y 502 ejemplares que permanecen en custodia en el Centro de Investigación Turipaná de Corpoica, Cereté, Colombia. Los datos registrados han sido digitados, ordenados, clasificados y analizados; lo cual permitió determinar el origen de los 270 animales que conformaron el hato inicial, procedentes de 11 hatos de las fincas: El Torno (43%), La Granja (16%), El Recreo (14%), Los Pinos (10%) y el 17% restante de La Vorágine, Colombia, Marta Magdalena y El Chorrillo. Aplicando el método de los cuadrados mínimos y el procedimiento de modelos lineales generalizados (generalized linear model GLM) (SAS,1995), se obtuvo la caracterización fenotípica de los pesos de animales en diferentes edades y etapas de producción: edad de la vaca al primer parto e intervalo entre partos; peso del ternero al nacer, al destete, a los 16 meses ajustados; peso de la vaca al primer parto, al parto y al destete del ternero; obtenido de 12673, 6543, 4363, 620, 7178 y 2847 registros, con valores de: 29,5±3,5, 163±35, 230±45, 393±51, 444±64 y 447±60 kg, respectivamente; la edad ajustada al destete y al primer parto, obtenidas a partir de 6572 y 2064 registros fueron 256±23 y 1133±200 días, respectivamente; el intervalo entre partos a partir de 9435 registros fue de 422±131 días. La eficiencia reproductiva de 86,3% confirma la adaptación de la raza en el valle del Sinú, Colombia.</p><p> </p><p> </p>

Esta pesquisa foi realizada com o objetivo de avaliar os aspectos produtivos e econômicos de poedeiras comerciais (Gallus gallus) submetidas a diferentes densidades de alojamento. Foram alojadas 264 poedeiras em gaiolas com dimensões de 100 cm × 50 cm × 40 cm em delineamento inteiramente casualizado, com quatro grupos distribuídos em esquema fatorial 4 × 4, com 8, 10, 12 e 14 aves por gaiola, correspondendo a 625, 500, 416,6 e 357,14 cm²/ave. As características avaliadas foram: produção de ovos, peso médio dos ovos, consumo de ração, massa dos ovos, conversão alimentar por dúzia e ovos imprestáveis para consumo humano. A densidade na gaiola teve efeito apenas sobre os ovos tipos extra e segunda e sobre a produção diária dos ovos tipo segunda, mas não influenciou as demais variáveis estudadas. Para poedeiras comerciais da linhagem Dekalb White, a utilização de gaiolas com a densidade de alojamento de 625, 500, 416,6 e 357,14 cm²/ave na fase de produção não prejudica os parâmetros de qualidade da produção, sob os aspectos zootécnico e econômico.

Abstract (sommario):
LED healthy light source by using a blue LED chip to excite phosphor was prepared successfully. Four kinds of LED commercial phosphors (cyan phosphor with a peak wavelength of 498 nm, green phosphor with 520 nm, yellow-green phosphor with 546 nm, and red phosphor with 628 nm) were selected that could be excited by blue light. A series of experimental studies were carried out on the different amounts of glue, different powder-glue ratio, different combinations of phosphors, and the influence of different driving currents. Trough the optimization of the experimental scheme, we obtained a LED healthy light source spectrum with luminous efficiency higher than 124 lm/w, color rendering index higher than 80, color temperature around 4000 k. These spectra are in good agreement with the full spectrum of sunlight, and the spectral continuity and uniformity are better, which accord with the requirements of bright (dark) vision, and the proportion of the blue spectrum is less than 2.4%, which meets the high quality and healthy lighting requirements of safety and comfort.

Background: Nerve conduction studies (NCSs) help in delineating the extent distribution of neural lesion, and the diagnosis of peripheral nerve disorders. Because normative nerve conduction parameters were not yet established in Sudan EMG laboratories, this study aims towards having our own reference values, as we are using the American and British parameters. This will allow avoiding the discrepancies that might be induced by many factors. Methods: NCSs were performed in 200 Median nerves of 100 adult healthy Sudanese subjects using standardized techniques. Results: The median SNAP (sensory nerve action potential) values were as follows: distal latency, 2.6±3 ms with a range of (2.3-2.9); peak latency, 3.5±0.5 ms (3.0-4.0); amplitude, 47.7±18.0μV (29.7-65.7); conduction velocity, 53.0±7.8 m/s (45.2-60.8). The following values were obtained for the Median nerve CMAP (compound muscle action potential) at wrist stimulation: distal latency, 3.5±0.5 ms with a range of (3.0-4.0); peak latency, 9.4± 1.0 ms (8.4-10.4); duration, 5.9±0.9 ms (5.0-6.8); amplitude, 12.3±2.5 mV (9.8-14.8); area, 43.0±10.4 mVms (32.6-53.4); conduction velocity, 63.6±6.2 m/s (57.4-69.8). The F wave was 28.4±1.8 ms (26.6-30.2). Conclusion: The overall mean sensory and motor nerve conduction parameters for the tested nerve compared favorably with the existing literature with some discrepancies that were justified.

CONTEXT AND OBJECTIVE: Computed tomography (CT) guided biopsy is widely accepted as effective and safe for diagnosis in many settings. Accuracy depends on target organ and needle type. Cutting needles present advantages over fine needles. This study presents experience from CT guided biopsies performed at an oncology center. DESIGN AND SETTING: Retrospective study at Hospital do Câncer A. C. Camargo, São Paulo. METHODS: 1,300 consecutive CT guided biopsies performed between July 1994 and February 2000 were analyzed. Nodules or masses were suspected as primary malignancy in 845 cases (65%) or metastatic lesion in 455 (35%). 628 lesions were thoracic, 281 abdominal, 208 retroperitoneal, 134 musculoskeletal and 49 head/neck. All biopsies were performed by one radiologist or under his supervision: 765 (59%) with 22-gauge fine-needle/aspiration technique and 535 (41%) with automated 16 or 18-gauge cutting-needle biopsy. RESULTS: Adequate samples were obtained in 70-92% of fine-needle and 93-100% of cutting-needle biopsies. The specific diagnosis rates were 54-67% for fine-needle and 82-100% for cutting-needle biopsies, according to biopsy site. For any site, sample adequacy and specific diagnosis rate were always better for cutting-needle biopsy. Among 530 lung biopsies, there were 84 pneumothorax (16%) and two hemothorax (0.3%) cases, with thoracic drainage in 24 (4.9%). Among abdominal and retroperitoneal biopsies, there were two cases of major bleeding and one of peritonitis. CONCLUSION: Both types of needle showed satisfactory results, but cutting-needle biopsy should be used when specific diagnosis is desired without greater incidence of complications.

510 Background: In treatment of rectal adenocarcinoma, an increased time delay (TD) of 6-12 weeks from the end of radiation therapy to surgery may increase the rate of complete pathologic response (pCR), but the optimal TD with respect to survival has not been established. This study evaluates the impact of TD on overall mortality. Methods: The NCDB was queried for patients with adenocarcinoma of the rectum and no evidence of metastasis at diagnosis, who underwent preoperative chemoradiation followed by radical surgical resection. Standard statistical methods were employed for descriptive statistics and Cox model development. Results: The study included 6805 patients, predominantly Caucasian (87.2%) and males (63.9%) who generally were treated with low anterior resection (57.3%), colonanal reanastomosis (8.4%), or abdominoperineal resection (28.4%), and had median survival of 66.6 months. The effects of age, surgical margins (-/+), comorbidity index, time to discharge after surgery, TMN pathologic staging, surgical volume, and patient income significantly impacted mortality after radiation and surgery (p<0.05 for all values). There was a significant relationship between TD and pCR (p=.0002). At TD less than 30 days, 4.0% of patients achieved pCR, while 9.3% of patients have achieved pCR by 75 days. In TD of greater than 75 days, the rate of pCR decreased. Overall, 6.8% of patients (n=461) achieved pCR. Using a refined cox model, a TD of more than 60 days was associated with 20% greater risk of mortality (95% CI 1.068 – 1.367). This effect became more pronounced with increasing TD; a TD of greater than 75 days was associated with 28% (95% CI 1.06-1.55) increased risk of mortality, while patients with TD less than 60 days saw a survival benefit. Conclusions: Though an interval up to 75 days between radiation and surgery may achieve higher rates of complete pathologic response, delay of more than 60 days from radiation to surgical resection and subsequent systemic chemotherapy decreases overall survival in patients with rectal cancer.

The material for the research were publications of scientists, reporting documentation of ZHRSLSPS, RSLSPS, SLVSE of Zhytomyr and Zhytomyr region, food samples. Radiological studies were performed on the device USC Gamma-plus № 0502 BG. The content of radionuclides in mushrooms was determined on RUG-91. In 2020, 210183 samples were tested, including 90 in excess. As in previous years, the excess was found in 19 samples of milk from the Narodytsya district (103–180 Bq/kg). The gifts of the forest remain the most polluted. The specific activity of 3 samples of fresh mushrooms from Novohrad-Volynskyi district was 610 Bq/kg, 642 Bq/kg, and 819 Bq/kg (with DR 500 Bq/kg), 4 samples from Yemilchyn district – 531–761 Bq/kg, 2 samples from Luhyny district – 760 Bq/kg and 827 Bq/kg. The most significant number of exceedances in the level of contamination of fresh mushrooms was found in Narodytsky, Ovruch, Malyn, and Olevsk districts, slightly less – in Luhyny. In particular, of the 25 samples of fresh mushrooms from Narodytsya district, 23 (621–2000 Bq/kg, DR exceeded 1.2–4 times) were detected, 28 % of fresh mushroom samples in Ovruch district were exceeded at 530–916 Bq/kg, of the 48 samples from Olevsk district exceedances were in 3 samples and, accordingly, their specific activity was at the level of 983 Bq/kg, 1994 Bq/kg and 2125 Bq/kg, i.e., 1.1, 3.9 and 4 times higher for DR-2006. In Malyn district, 50 % of samples of fresh mushrooms showed an excess of 631–1450 Bq/kg (1.3–2.9 times). In the Luhyny district, the excess was found in 18 % of samples (760 Bq/kg – 827 Bq/kg). The specific activity of samples of dried mushrooms from Novohrad-Volynskyi district was 2725 and 3018 Bq/kg, respectively, Yemilchynsky – 2608–3080 Bq/kg, Ovruch – 2820 Bq/kg and 2850 Bq/kg, Narodytsky – 3425 Bq/kg. The specific activity of 2 samples of wild animal meat from Narodytskyi district was 800 and 1400 Bq/kg (exceeding 2–3.5 times), 1 sample from Ovruch – 433 Bq/kg. Of the 109 samples of beekeeping products studied during this period, exceedances were detected in 1.8 %, the specific activity was 264 Bq/kg (Narodytskyi district) and 1920 Bq/kg (Ovruch district), respectively. In 2020, no excesses were found in samples of other products. Given that the consumption of food, even with low specific activity, leads to an increase in internal exposure, the research results indicate a threat to the health of present and future generations and the need for decision-making at the state level.

Menurut penelitian terdahulu, kolom dengan sengkang spiral memiliki kinerja terbaik dalam menahan beban dibandingkan kolom dengan sengkang persegi. Namun kenyataannya dilapangan kolom sengkang persegi adalah jenis kolom yang banyak digunakan karena pelaksanaanya mudah dilakukan. Penelitian ini berfokus pada ketiga jenis penampang kolom, yaitu penampang kolom bujur sangkar, lingkaran, dan persegi panjang dan struktur penampang kolom dianalisis berdasar SNI 2847-2019 sebagai Sistem Rangka Pemikul Momen Khusus (SRPMK). Dimensi kolom bujur sangkar 500 x 500 mm, kolom lingkaran diameter 564 mm dan kolom persegi dimensi 625 x 400 mm. Jumlah tulangan utama dan sengkang semua jenis penampang sama 24D19 dan sengkang D10-100. Abstract The spatial distribution, Galactic model parameters, and luminosity function of cataclysmic variables (CVs) are established using reestimated trigonometric parallaxes from Gaia DR3. The data sample of 1587 CVs in this study is claimed to be suitable for Galactic model parameter estimation as the distances are based on trigonometric parallaxes, and the Gaia DR3 photometric completeness limits were taken into account when the sample was created. According to the analysis, the scale height of all CVs increases from 248 ± 2 to 430 ± 4 pc toward shorter periods near the lower limit of the period gap and suddenly drops to 300 ± 2 pc for the shortest orbital period CVs. The exponential scale heights of all CVs and the magnetic systems are found to be 375 ± 2 and 281 ± 3 pc, respectively, considerably larger than those suggested in previous observational studies. The local spatial density of all CVs and the magnetic systems in the sample are 6.8 − 1.1 + 1.3 × 10−6 and 2.1 − 0.4 + 0.5 × 10 − 6 pc−3, respectively. Our measurements strengthen the 1–2 order of magnitude discrepancy between the CV spatial densities predicted by population synthesis models and observations. It is likely that this discrepancy is due to objects undetected by CV surveys, such as systems with very low M ̇ and ones in the period gap. A comparison of the luminosity function of white dwarfs with the luminosity function of all CVs in this study show that 500 times the luminosity function of CVs fits very well to the luminosity function of white dwarfs. We conclude that the estimations and data sample in this study can be confidently used for further analyses of CVs.

Background:Four clinical phenotypes of IgG4-Related Disease (IgG4-RD) have been recently identified by Latent Class Analysis (LCA) - Pancreato/biliary (Group 1); Retroperitoneum/Aortitis (Group 2); Head-and-neck limited (Group 3); Mickulicz/Systemic (Group 4) - but the relevance of this classification for patient management remains unknown (1,2).Objectives:We aimed to assess whether clinical judgment can replicate LCA classification and to evaluate potential differences in epidemiological features, serological findings, and disease outcomes between disease phenotypes.Methods:The study included 179 patients. Four IgG4-RD experts were asked to classify a validation cohort of 40 patients according to published LCA derived phenotypes based on clinical judgment. Agreement between LCA and clinical clustering was calculated. To assess differences among disease phenotypes, the following variables were recorded on additional 139 patients: serum IgG4 and IgE; inflammatory markers; eosinophils; plasmablasts; IgG4-RD Responder Index (RI); history of atopy, diabetes, osteoporosis, relapses, and tumors; cumulative dose of glucocorticoids and use of rituximab.Results:Clinical judgment recapitulated LCA classification with strong agreement between IgG4-RD experts (κ= 0.841, p < 0.0005). Group 1 showed the highest levels of serum IgG4 and IgE. Group 2 and 4 had the lowest and highest IgG4-RD RI, respectively (Table 1). Increased cumulative doses of glucocorticoids and higher relapse rate were observed in Group 3 (Fig 1). Four immunologically distinct subunits were characterized in glutathione (GSH) S-transferases of human liver. Five cationic enzymes (pI 8.9, 8.5, 8.3, 8.2 and 8.0) have an apparently similar subunit composition, and are dimers of 26 500-Mr (A) and 24 500-Mr (B) subunits. A neutral enzyme, pI 6.8, is a dimer of B-type subunits. One of the anionic enzymes, pI 5.5, is also a dimer of 26 500-Mr subunits. However, the 26 500-Mr subunits of this anionic enzyme form are immunologically distinct from the A subunits of the cationic enzymes, and have been designated as A'. Immunoabsorption studies with the neutral enzyme, BB, and the antibodies raised against the cationic enzymes (AB) indicate that A and B subunits are immunologically distinct. Hybridization in vitro of the A and B subunits of the cationic enzymes (AB) results in the expected binary combinations of AA, AB and BB. Studies with the hybridized enzyme forms indicate that only the A subunits express GSH peroxidase activity. A' subunits have maximum affinity for p-nitrobenzyl chloride and p-nitrophenyl acetate, and the B subunits have highest activity towards 1-chloro-2,4-dinitrobenzene. The other anionic form, pI 4.5, present in liver is a heterodimer of 22 500-Mr (C) and B subunits. The C subunits of this enzyme are probably the same as the 22 500-Mr subunits present in human lung and placental GSH transferases. The distinct immunological nature of B and C subunits was also demonstrated by immunoaffinity and subunit-hybridization studies. The results of two-dimensional polyacrylamide-gel-electrophoretic analyses indicate that in human liver GSH transferases, three charge isomers of Mr 26 500 (A type), two charge isomers of Mr 24 500 (B type) and two charge isomers of Mr 22 500 (C type) subunits are present.

Optical Coherence Imaging (OCI) including Optical Coherence Tomography (OCT) and Optical Coherence Microscopy (OCM) uses interferometric detection to generate high-resolution volumetric images of the sample at high speeds. Such capabilities are significant for in vivo imaging, including ophthalmology, brain, intravascular imaging, as well as endoscopic examination. Instrumentation and software development allowed to create many clinical instruments. Nevertheless, most of OCI setups scan the incident light laterally. Hence, OCI can be further extended by wide-field illumination and detection. This approach, however, is very susceptible to the so-called crosstalk-generated noise. Here, we describe our novel approach to overcome this issue with spatio-temporal optical coherence manipulation (STOC), which employs spatial phase modulation of the incident light. Abstract Graft copolymerization of acrylonitrile and acryloyl chloride on to chitosan was prepared by γ-rays. Optimization of the grafting (%) was studied. The grafting (%) was observed to increase with increase in the irradiation dose and monomer concentration. The grafting percentages were about 52% and 36% from polyacrylonitrile and poly(acryloyl chloride), respectively. Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM) were used to characterize the specimens. The modified chitosan was loaded with vitamin B12, demonstrated nearly 5.0±2.3% and 50.1±4.5% release in the media of pH 1.2 and 6.8, respectively, for amidoximated chitosan-grafted polyacrylonitrile and 3.6±1.1% and 36±2.4% in pH 1.2 and 6.8, respectively, for chitosan-grafted poly(acryloyl chloride), as determined by a traditional dissolution model. The modified chitosan specimens that uploaded with vitamin B12 displayed a more decremental release in the acidic medium than the neutral one. However, in order to incorporate in vivo gastrointestinal conditions, such as acidic pH and high water content in the stomach, low water content, and the presence of semi-solid mass in the large intestine, a new model, called flow through diffusion cell, was also used to study the drug release. The results of the two approaches produced different release profiles at the same pH values.

Objective: Current studies focus on the prevalence rate of lung cancer in idiopathic pulmonary fibrosis and connective tissue disease-associated interstitial lung disease (CTD-LID). Our aim was to investigate the prevalence of malignancies in patients with various subtypes of ILD. Methods: A total of 5350 patients diagnosed with ILD between January 2015 and December 2021 were retrospectively included. The prevalence of different malignancies and different ILDs was assessed using complete follow-up data. Results: A total of 248 patients (139 males; 65—IQR, 57 to 72—years) out of 5350 patients with ILD were confirmed with malignancies. A total of 69% of patients with ILD and malignances were older than 60 years old. The prevalence of malignancies in ILD patients was 4.6%, and lung cancer had the most common incidence of 1.9%, followed by malignancies in the digestive system of 0.9%. Among the different ILD subtypes, the prevalence of malignancies such as organizing pneumonia (OP), idiopathic pulmonary fibrosis (IPF), anti-neutrophil cytoplasmic antibodies-associated vasculitis-related ILD(AAV-ILD), nonspecific interstitial pneumonia (NSIP), CTD-ILD, hypersensitivity pneumonitis (HP), sarcoidosis, and other types of ILD was 6.8%, 5.0%, 4.7%, 4.3%, 2.5%, 2.2%, 1.2%, and 6.9%, respectively. The incidence of lung cancer as the most common tumor in IPF was 3.9%, with adenocarcinoma predominating (1.7%). The highest rate of malignancy occurring in RA of CTD-ILD was 2.4%. Conclusion: Older patients with ILD (≥60 years) including OP, IPF, AAV-ILD, NSIP, CTD-ILD, and HP, were associated with a higher incidence of malignancy, especially males aged from 60 to 69 years. These epidemiological results indicate that it is essential for physicians to pay more attention to the screening of and management strategies for different malignancies, according to the specific ILD subtypes.

ABSTRACTBackgroundThere is a paucity of data about water soluble vitamin status in low birthweight infants. Therefore, the authors' objective was to assess current feeding protocols.MethodsThe authors measured serum concentrations for riboflavin, pyridoxine, and vitamin C and functional assays for thiamine and riboflavin longitudinally in 16 premature infants (birthweight, 1,336 ± 351 g; gestational age, 30 ± 2.5 weeks) before receiving nutrition (time 1, 2 ± 1 days), during supplemental or parenteral nutrition (time 2, 16 ± 10 days) and while receiving full oral feedings (time 3, 32 ± 15 days). In plasma, vitamin C was measured colorimetrically, and riboflavin and pyridoxine were measured using high‐performance liquid chromatography. The erythrocyte transketolase test as a functional evaluation of thiamine and the erythrocyte glutathione reductase test for riboflavin were measured colorimetrically.ResultsAt time 1, nutrient intake of vitamins were negligible because infants were receiving intravenous glucose and electrolytes only. Intakes differed between time 2 and time 3 for thiamine (510 ± 280 and 254 ± 115 μg · kg −1 · d −1 , respectively), riboflavin (624 ± 305 and 371 ± 193 μg · kg −1 · d −1 , respectively), and pyridoxine (394 ± 243 and 173 ± 85 μg/100 kcal, respectively), but not for vitamin C (32 ± 17 and 28 ± 12 mg · kg −1 · d −1 , respectively). Blood levels at times 1, 2, and 3 were for thiamine (4.9 ± 2.7%, 3.3 ± 6.6%, and 4.1 ± 9% erythrocyte transketolase test, respectively), riboflavin (0.91 ± 0.31, 0.7 ± 0.3, 0.91 ± 0.18 erythrocyte glutathione reductase test, respectively), riboflavin (19.5 ± 17, 23.3 ± 8.6, 17.6 ± 10 ng/mL, respectively), pyridoxine (32 ± 25, 40 ± 16, 37 ± 26 ng/mL, respectively), and vitamin C (5.2 ± 3, 5 ± 2.2, 10 ± 5 μg/mL, respectively) and did not differ at those times.ConclusionsCurrent intakes of these vitamins, except for possibly vitamin C, during parenteral and enteral nutrition seem to result in adequate plasma concentrations and normal functional indices.

Background: At diagnosis of chronic myeloid leukemia (CML) patients (pats) may present with elevated platelets (PLT) counts. Generally, high PLT counts may result in thrombosis and/or bleeding complications, the latter being due to binding of von Willebrand factor (VWF) multimers to platelets. Pediatric CML is very rare (Hijiya N, Suttorp M 2019; Blood 33:2374-2384) and no systematic investigation on clinical complications of elevated PLT counts has so far been reported. Methods: Data on PLT counts and associated thrombo-hemorrhagic complications were retrospectively analyzed in newly diagnosed pediatric pats (0 - 18 yrs old) with CML in chronic phase. Data were pooled from a German pediatric trial (CML-paed II, N= 146 pats; registered as NCT00445822, [Suttorp M, et al. 2018; Leukemia 32:1657-1669], data collection closed Dec 2016) and the International registry for CML in children and adolescents (I-CML- Ped Study, N= 475 pats, registered as NCT01281735, [Millot F, et al. 2017; Haematologica 102:1704-1708], data collection closed June 2019). Results: A total of 621 pediatric pats with CML in chronic phase could be analyzed. At diagnosis 343/621 (55.2%) pats (median age 13.2 yrs, range 1.3 - 18 yrs) presented with thrombocytosis (>500 000 PLT/µl). PLT ranged from 500 000 - 1 000 000 PLT/µl in 239/621 (38.5%) pats while extremely high PLT counts (>1 000 000 PLT /µl) were present in 94/621 (11.2%) pats. Thrombosis was observed in 2/621 (0.3%) pats including priapism occurring at 655 000 PLT/µl and lower leg venous thrombosis at 1 820 000 PLT/µl. Bleeding signs (bruises, mucosal hemorrhage) being of mild nature (grade 1, grade 2, National Cancer Institute Common Terminology Criteria v3.0) only were described in 38/343 (11.1%) pats with thrombocytosis (>500 000 PLT/µl). Bleeding occurred without correlation to elevated PLT counts but was associated in selected cases investigated with a reduced plasma concentration of large VWF multimers pointing to the diagnosis of acquired von Willebrand syndrome (AVWS). AVWS resolved after initiation of CML treatment with imatinib. Discussion: In newly diagnosed pediatric pats with CML, pathologically high PLT counts (>500 000 PLT/µL) were detected in >50% of the cohort and extremely high platelet counts in >10%. However, associated thrombo-hemorrhagic complications were rare events affecting less than 10% (40/621= 6.4%) of the pats. Especially thrombosis was a rare (0.3%) complication whereas mild bleeding symptoms formed the majority of hemostaseological complications observed. Compared to recently published findings in adults (Sora F, et al. 2018; Br J Haematol 181:267-270, N= 1591 pats; Sandal R, et al. 2019; J Clin Oncol (Suppl) abstract #e18546, N= 2350 pats; Liu Z, et al. 2017; Onco Target Ther 10:3515-3520, N= 87 pats) the proportions of adult pats with extremely high platelet counts (Sora F: 5,5%, Sandal R: not listed; Liu Z: 25.3%) is in the same range (5% - 25%) as in pediatric patients (11.2%). Surprisingly, thrombo-hemorrhagic complications were observed more frequently in children (40/621= 6.4%) than in adults (Sora F: 9/1591= 0.5%, Liu Z: 1/85= 0.1%; Sandal R: 66/2350= 2.8%). As a word of caution this comparative analysis is weakened by its retrospective nature and a non-standardized reporting of bleeding signs. Mild bleeding signs in pats with high PLT counts can be caused by an acquired low plasma concentration of large VWF multimers as could be demonstrated in selected pediatric pats from this cohort. Systematic investigations on AVWS in children with CML have not been published so far and AVWS may be underdiagnosed in pediatric CML. Disclosures No relevant conflicts of interest to declare.

622 Background: Non-Hispanic Blacks (NHB) with renal cell carcinoma (RCC) are more likely to have papillary RCC (pRCC) than Non-Hispanic Whites (NHW). Little is known about the histology of RCC in Hispanics (H). The Montefiore-Einstein Cancer Center (MECC) serves a diverse population in the Bronx, NY. We sought to investigate histological subtype-specific associations with established RCC risk factors in an ethnically diverse patient population to elucidate prospective relationships between those factors and racial differences in RCC histology. Methods: : The MECC tumor registry was used to identify patients ≥18 years of age treated with partial or radical nephrectomy between January 2000 and December 2015. An institutional software program and individual chart review were used to obtain demographic data (including self-reported race, age, and sex), pathology data, and RCC risk factors (hypertension, diabetes, smoking status, renal function, weight). Data were modeled by multinomial logistic regression to estimate odds ratios (ORs) and 95% CIs. Results: 1010 RCC cases were identified (232 NHW, 383 NHB, 181 H, and 214 other). Histology was 530 clear cell (ccRCC), 257 papillary (pRCC), 100 chromophobe (cRCC), and 123 other. Individuals with pRCC compared to ccRCC were more likely to be NHB than NHW (OR 4.41; 95%CI 2.81,6.93) and more likely to have a higher Fuhrman grade (OR 1.5; 95%CI 1.03,2.07), but were less likely to be female (OR 0.50; 95%CI 0.35,0.72) or H than NHW (OR 0.518, 95%CI 0.271,0.991). Individuals with cRCC compared to ccRCC were also more likely to be NHB than NHW (OR 2.23, 95%CI 1.06,4.67) and to have higher Fuhrman grade (OR 1.82, 95%CI 1.06,3.14). Conclusions: In the MECC dataset, histology of RCC varies by race, confirming earlier reports that non-ccRCC is more common in NHB than NHW. We also report for the first time that pRCC is less common in H than NHW. These associations will be evaluated in additional larger national data sets and may help elucidate reasons for disparate RCC outcomes among various races and ethnicities.

Postweaning liveweights and carcass traits of Hereford, 1/4 Simmental-3/4 Hereford crossbreed, first-cross Simmental-Hereford, highgrade Simmental, and Belmont Red steers were compared from 3 cohorts bred at Brigalow Research Station, Theodore, Queensland. One half of each breed was grown and fattened at Brigalow Research Station, and the other half at a different site in southern Queensland, for each of 3 cohorts. When the mean liveweight of the Hereford steers reached 450 kg, a random selection of all breed groups was slaughtered (light group), while the remainder was slaughtered when the Hereford steers reached 550 kg liveweight (heavy group). Highgrade Simmental steers had higher (P<0.05) liveweights than 114 Simmentals and Herefords at each site in each year at all ages. While highgrade Simmentals were usually heavier than first-cross Simmental-Herefords and Belmont Reds, differences were not always significant. First-cross Simmental- Hereford and Belmont Red steers were usually heavier (P<0.05) than Herefords. Response to higher nutritional levels from provision of forage crop varied according to breed, with highgrade Simmentals showing the greatest liveweight response (268 kg by 27 months) and Belmont Reds the least (164 kg) over contemporaries grazing pasture only. Highgrade Sirnmental steers had the heaviest (P<0.05) carcasses in both slaughter groups (274 and 340 kg, respectively), and there were no significant (P>0.05) differences between Belmont Red and first-cross Simmental-Hereford steers (light group, 263 and 258 kg; heavy group, 318 and 327 kg). The 114 Simmental steers had heavier (P<0.05) carcasses than Hereford steers in both slaughter groups (light, 244 v. 235 kg; heavy, 307 v. 295 kg). Belmont Red carcasses had the greatest rib fat depth for both slaughter groups (6.6 and 8.4 mm), and highgrade Simmental carcasses were leanest (3.3 and 4.1 mm; P<0.05). Carcasses from 114 Simmental and from Hereford steers had similar rib fat depth in both slaughter groups (light, 5.0 and 5.1 mm; heavy, 6.8 and 6.8 mm), while the fat depths on first-cross Simmental-Hereford carcasses were 4.3 and 6.0 mm. Although highgrade Simmental steers yielded the heaviest carcasses, the lack of subcutaneous fat cover may reduce the value of these carcasses, on a unit weight basis. By contrast, Belmont Red carcasses were more likely to meet the range of market specifications for grass-fed carcasses.

Abstract. We derive a global climatology of tropospheric NO2 profiles from OMI cloudy observations for the year 2006 using the cloud slicing method on six pressure levels centered about 280, 380, 500, 620, 720 and 820 hPa. A comparison between OMI and the TM4 model tropospheric NO2 profiles reveals striking overall similarities, which confer great confidence to the cloud-slicing approach, along with localized discrepancies that seem to probe into particular model processes. Anomalies detected at the lowest levels can be traced to deficiencies in the model surface emission inventory, at mid tropospheric levels to convective transport and horizontal advective diffusion, and at the upper tropospheric levels to model lightning NOx production and the placement of deeply transported NO2 plumes such as from the Asian summer monsoon. The vertical information contained in the OMI cloud-sliced NO2 profiles provides a global observational constraint that can be used to evaluate chemistry transport models (CTMs) and guide the development of key parameterization schemes.

<p>Salah satu upaya yang dapat dilakukan untuk meningkatkan pendapatan petani adalah peningkatan produktivitas dan peningkatan mutu hasil beras. Penelitian ini bertujuan untuk (i) mengetahui varietas unggul baru padi yang memiliki tingkat hasil dan mutu beras tinggi dan (ii) menganalisis pengaruh pemupukan terhadap hasil gabah dan mutu beras. Penelitian dilakukan di lahan sawah irigasi Kabupaten Gorontalo mulai Januari 2012 sampai Januari 2013. Penelitian dilakukan dalam dua tahap. Tahap pertama menggunakan Rancangan Acak Kelompok (RAK) untuk menguji enam varietas (Inpari 3, Inpari 4, Inpari 9, Inpari 10, Inpari 13, dan Mira) dan setiap varietas diulang sebanyak empat kali di lahan petani. Sedangkan tahap kedua menggunakan Rancangan Acak Kelompok dengan empat perlakuan (Rekomendasi pemupukan PUTS + 500 kg/ha jerami, rekomendasi pemupukan cara petani + 500 kg/ha jerami, rekomendasi pemupukan PUTS, dan rekomendasi pemupukan cara petani) yang diulang empat kali. Setiap varietas ditanam dengan jarak 25 cm x 25 cm dengan cara tanam pindah dari bibit semai yang berumur 21 hari. Hasil penelitian menunjukkan bahwa varietas Inpari 4, 10 dan Mira memiliki tingkat produktivitas yang cukup tinggi masing-masing 4,21 t/ha; 4,35 t/ha dan 4,46 t/ha GKG serta memiliki mutu beras yang lebih baik. One of the efforts to increase farmer’s income is by increasing yield and improving the quality of rice. The objectives were (i) to obtain new varieties of rice with high yield and quality and (ii) to analyze the effect offertilization to the yield and quality of rice. The research was carried out in Gorontalo district from January 2012 to January 2013. The research was conducted in two steps. The first step used a Randomized Block Design (RBD) to test six varieties (Inpari 3, Inpari 4, Inpari 9, Inpari 10, Inpari 13 and Mira) and it was repeated four times in the farmer’s fields. While the second step used a RBD with four treatments fertilizer recommendation of Paddy Soil Test Kit (PSTK) + 500 kg/ha of straw, existing fertilizer + 500 kg/ha of straw, fertilizer recommendation of PSTK and existing<br />fertilizer and it was repeated four times. Each variety was planted with measurement of 25 cm x 25 cm and transplanted in 21 days old. The results showed that the productivities of Inpari 4, Inpari 10 and Mira were quite high (4.21 t/ha; 4.35 t/ha and 4.46 t/ha of dry grain, respectively) and they had a good quality of rice. Fertilization using PSTK + 500 kg/ha of straw showed the highest yield (6.28 t/ha) and it produced a good quality of rice.<br /><br /></p><p> </p>

Abstract Abstract 2518 Introduction: Mutations in IDH1 and IHD2 at arginines 132 and 140 or 172, respectively, have recently been shown to play an important role in AML. Also the IDH1105GGT minor allele of the IDH1G105 (SNPrs11554137) polymorphism that is localized in the same exon as the IDH1R132 mutation has recently been reported to be an adverse prognostic factor in AML (JCO: 14, 2356–2364, 2010). Aim: We aimed at further delineating the frequency and impact of the IDH1105GGT minor allele in AML and also analyzed a healthy control cohort. Methods:IDH1G105 (SNPrs11554137) was analyzed in 961 AML patients by a LightCycler-based melting curve assay. Female/male ratio was 433/528 and age ranged from 13.1–100.4 years (median, 66.7 years). The results were compared to a healthy control cohort from the KORA (Cooperative Health Research in the Region of Augsburg) survey S4, which consists of 475 cases who where matched to the leukemia samples with respect to sex (193f/282m) and age (median: 67, range 32–81 years). Informed consent for participation in anonymized genetic studies was obtained from all individuals. IDH1G105 in the healthy cohort was analyzed by Sanger sequencing. Further mutation analyses were available in subsets of the AML patients, respectively, as follows: (IDH1R132 n=625, IDH2R140 n=587, IDH2R172 n=590, FLT3-ITD n=629, FLT3-TKD n=503, NPM1 n=628, CEBPA n=587, RUNX1 n=231, MLL-PTD n=629, NRAS n=273, KRAS n=133, ASXL1 n=470) and were analyzed as described previously. A subcohort of 634 AML with intermediate risk karyotype was analyzed for survival. Female/male ratio of this subcohort was 280/354 and age ranged from 15.7–86.6 years (median, 66.9 years). The adverse impact of IDH1R132, IDH2R140 and IDH2R172 on the NPM1+/FLT3-ITD- group has been shown previously for this group (Blood 2010 116: Abstract 102). Results: The IDH1105GGT minor allele was detected in 11.2% (108/961) in AML and in 8.8% (42/475) of the KORA control. This slight difference does not reach statistical significance (p=0.17) and thus there is no indication that this variance is a predisposing factor for leukemia. Also the frequency of a homozygous IDH1105GGT minor allele was not different between the AML cohort (3/108, 2.8%) and the KORA cohort (2/42, 4.8%, n.s.). In the AML cohort there was no association of the IDH1105GGT minor allele with age, WBC, platelet count or any of the above mentioned molecular mutations. In contrast, some differences in survival were observed: patients with the IDH1105GGT minor allele had a longer event free survival (EFS) than those with the IDH1105GGC major allele (median: 30.1 vs. 21.6 months, p=0.052) in the intermediate risk cohort. This prognostically favourable effect of the IDH1105GGT minor allele was most prominent in the NPM1+/FLT3- group with a median EFS of 45.1 vs 23.5 months as compared to those with the IDH1105GGC major allele (p=0.015). Conclusions: 1) The polymorphic IDH1105GGT minor allele was not found to be a marker predisposing for AML. 2) No association of the IDH1105GGT minor allele to any mutation or other biological parameters was detected. 3) We were not able to reproduce the previously published adverse impact of the IDH1105GGT minor allele on survival in AML. In contrast, in our cohort of 475 patients with intermediate risk AML the EFS was even better in patients carrying the IDH1105GGT minor allele, especially in the subcohort with NPM1+/FLT3−ITD−. Thus, we would suppose that the IDH1105GGT minor allele is a favorable molecular marker in intermediate risk AML. However, as these findings are in contrast to previously published data, further confirmation in additional studies is necessary to draw firm conclusions on the utility of the IDH1105 polymorphism as a marker for diagnostics and prognosis in AML. BackgroundPrevious guidelines (2006) recommended that tumour necrosis factor inhibitors (TNFi) be discontinued during pregnancy. Despite new guidelines (2016 & 2020) now recommending against this, the choice to stop TNFi pre-conception is patient- and provider-dependent. Observational studies have evaluated use of disease-modifying anti-rheumatic drugs (DMARDs) during pregnancy, but few have specifically assessed TNFi discontinuation pre-conception. Understanding trends and predictors of TNFi discontinuation pre-conception may help inform initiatives to improve care and optimize maternal and fetal outcomes.ObjectivesWe examined trends in TNFi discontinuation pre-conception over time, and evaluated the characteristics of pregnant women with chronic inflammatory diseases who stopped using TNFi pre-conception (without resuming in pregnancy) compared with those who used TNFi at any time during pregnancy.MethodsWe created a cohort of pregnant women with rheumatoid arthritis (RA), ankylosing spondylitis, psoriatic arthritis (PsA), psoriasis (PsO), and/or inflammatory bowel diseases (IBD) who delivered between 2011 and 2019, using the MarketScan commercial database. TNFi use was defined as at least 1 filled prescription or infusion procedure claim, categorized according to the timing of the filled prescription or infusion procedure date in relation to the gestational period: 1) TNFi pre-conception only (i.e. at least 1 prescription filled or infusion procedure claim in the 12 weeks preceding the gestational period but none within the gestational period) or 2) TNFi use at any time during pregnancy (i.e. any prescription filled or infusion procedure claim during the gestational period, including restarts, new starts, and those continuing from pre-conception).ResultsWe identified 3,372 pregnancies; 13% discontinued TNFi in the 12 weeks before conception and did not restart, and 86% were exposed to TNFi during pregnancy. Pregnancies in IBD patients accounted for 47% of all pregnancies. Nearly all pregnancies with IBD were exposed to TNFi during pregnancy (95%). Compared to those with IBD, more patients with RA (difference of 18%, 95% CI 15-21%) and PsA/PsO (20%, 95% CI 16-24%) discontinued their TNFi. Corticosteroid use was similar in both TNFi exposure groups, while patients who took TNFi during pregnancy were more likely to use non-biologic DMARDs concomitantly. Over time, a lower proportion of patients stopped TNFi pre-conception (2011-2013 19% vs 2014-2016 13% vs 2017-2019 10%; p-value for trend <0.0001).ConclusionIn our sample, 13% discontinued TNFi in the 12 weeks before conception and did not restart. The proportion of patients stopping TNFi pre-conception between 2017-2019 decreased compared to earlier years, possibly reflecting updated guidelines. Objective: While guidelines propose a single elevated prolactin measurement drawn without excess venipuncture stress as sufficient for diagnosing hyperprolactinemia, this may lead to unnecessary evaluation in the setting of stress-induced hyperprolactinemia. In this study, we aimed to define the role of the cannulated prolactin test in confirming hyperprolactinemia. Methods: We conducted a retrospective review of 757 patients with unexplained hyperprolactinemia who performed a cannulated prolactin test in a community-based referral endocrine clinic between 2000–2015. The prolactin test consisted of “test-baseline” levels taken at rest (T0), and cannulated measurements at 60 and 90 minutes (T60 and T90) without repeated venipuncture. The most recent prolactin level performed prior to the test (referral-prolactin) was collected. Results: Referral-prolactin was available for 621 (82%) patients, of whom 324 (52.2%) normalized at T0. The probability of normoprolactinemia at T0 was 50% if referral-prolactin was 2.0-fold the upper-limit-of-normal (ULN), yet only 5% if referral-prolactin was 5.0-fold the ULN. Of the 359 patients with hyperprolactinemia at T0, prolactin normalized at T60 and/or T90 in 99 (27.6%) patients. The probability of normoprolactinemia was low (<5%) in those with T0 prolactin levels >2.4-fold ULN. Overall, of 757 prolactin tests performed, only 260 (34.3%) patients had persistent hyperprolactinemia. Conclusion: Patients with referral-prolactin levels >5.0-fold the ULN, or a rested-prolactin (T0) >2.4-fold the ULN are unlikely to normalize during the cannulated test and consideration should be made to proceed directly with pituitary imaging. In patients with prolactin levels below these thresholds, the cannulated prolactin test may considerably reduce unnecessary investigations, treatment, and cost. ABSTRACT The objective of this study was to evaluate the concentration of cations in the leachate of soil cultivated with sugarcane irrigated with brackish water. An experiment was carried out in drainage lysimeters at the Federal Rural University of Pernambuco, at the Recife campus. The experimental design was completely randomized in a 5 x 2 factorial scheme, with four replicates, corresponding to five levels of electrical conductivity of the irrigation water (ECw) (0.5, 2.0, 3.5, 5.0 and 6.5 dS m-1) and two leaching fractions (LF1 = 0 and LF2 = 0.17) corresponding to water depths equivalent to 100 and 120% of crop evapotranspiration (ETc), by which the irrigations were managed. Electrical conductivity of the drained water was daily monitored and Na, K, Ca and Mg concentrations in the drainage water were determined at 129, 214, 286 and 324 days after planting (DAP). There were higher salt concentrations in the leachates when leaching fraction LF1 = 0 was used. The use of the 0.17 leaching fraction led to greater dissolution of the salts and may have caused a higher uptake of the essential cations by the crop. Sodium was the most quantitatively leached cation at all collection dates.

The objective of this study was to test the potential for using a single-entry procedure to revegetate Russian knapweed– and green rabbitbrush–infested rangeland. I hypothesized that simultaneously applying an herbicide and seeding in the fall would produce the highest establishment and growth of desired species. For Russian knapweed, treatments included three seeding rates (zero, low, and high) and two herbicides (with and without clopyralid plus 2,4-D) applied in either the spring (2003) or fall (2004). The seed mixture included pubescent wheatgrass, Siberian wheatgrass, and alfalfa. Grasses were seeded on October 21, 2003 (fall dormant), and April 5, 2004 (spring), using a no-till rangeland drill. The seeding rates consisted of 3.4, 6.8, and 9.3 (low) or 5.0, 9.0, and 12.3 (high) kg ha−1of alfalfa, Siberian wheatgrass, and pubescent wheatgrass, respectively. For green rabbitbrush, treatments included two seeding levels (13.5 kg ha−1of Siberian wheatgrass and not seeded) and three herbicides (clopyralid, dicamba, and 2,4-D) and a control applied in July. Clopyralid plus 2,4-D (0.21 plus 1.12 kg ha−1) and dicamba at 2.1 kg ha−1gave 61 and 66% control of Russian knapweed and green rabbitbrush, respectively. Herbicides interacted with seeding to provide the highest density of seeded species on the Russian knapweed site. These data support the hypothesis that simultaneously applying an herbicide and seeding in the fall would produce the highest establishment and growth of desired species. Conversely, only seeding affected Siberian wheatgrass establishment on the rabbitbrush sites. It may be reasonable to seed Siberian wheatgrass without controlling rabbitbrush if forage production is the primary objective.

2521 Background: The dose of carboplatin is usually calculated using the Calvert formula, with various bedside estimates utilized rather than directly measured GFR. The 4-v MDRD equation (Levey AS, et al. Ann Intern Med 2006; 145: 247–254) is now advocated as the routine method of estimating renal function from serum creatinine in all patients. Adoption in clinical practice is occurring despite lack of validation in oncology patients. The aim of this study was to compare carboplatin doses derived from the Calvert formula using measured GFR and the estimates of the 4-v MDRD equation and other established estimates. Methods: GFR was determined using technetium-99m diethyl triamine penta-acetic acid (Tc99mDTPA) clearance. Serum creatinine (Jaffe method) was measured and GFR estimates calculated using 4-v MDRD, Cockcroft and Gault formula (CGF), Wright, Martin, and Jelliffe (JF) formulae. Carboplatin doses were calculated using the Calvert formula, targeting an area under the curve of 7mg.ml- 1.min-1. Results: GFR was measured in 510 adult oncology patients (323 male, 187 female, mean age 63 years, range 17–87 years, mean GFR 84 mL/min, range 16–205 mL/min). The mean (range) carboplatin dose was 765 mg (287–1,610 mg), 681 mg (237–1,306 mg), 674 mg (249–2,044 mg), 721 mg (261–1,536mg), 741 mg (261–2,128mg), 620 mg (244- 1,329 mg) for measured GFR, 4-v MDRD, CGF, Wright, Martin, and JF formulas respectively. The accuracy (% within 20% of ‘true’ dose) was 58%, 63%, 73%, 72% and 49% for 4-v MDRD, CGF, Wright, Martin, and JF formulas respectively. Carboplatin doses derived using the 4-v MDRD estimate of GFR become increasingly less accurate with increasing GFR (see table ). All other formulas performed similarly. Conclusions: The 4-v MDRD equation resulted in an imprecise estimation of carboplatin doses with the degree of variability dependant on the level of renal function. Abstract Background: Anemia in the elderly is associated with significant medical consequences even with mild reductions in hemoglobin (Hb). These include worsening physical disability and heart failure. CKD is a common treatable cause of anemia in the elderly. Given the limited published data on epoetin alfa (EPO) therapy in octogenarians, we evaluated whether responses to QW EPO therapy in patients (pts) with anemia due to CKD were influenced by age. Objective: A subset analysis comparing 2 groups of elderly pts (Group 1: 65–79 years (y), Group 2: >/=80y) vs non-elderly (<65y) pts with anemia of CKD not on dialysis was performed on the dataset of a recently completed multicenter, prospective, open-label study examining response to QW EPO therapy. Efficacy was assessed by changes in Hb and transfusion requirements. Methods: All pts had entry Hb </=10g/dL and received EPO at a starting dosage of 10,000U SC QW for 16 weeks (w) titrated to 20,000U SC QW after 4w if Hb increased by <1g/dL. Results: Of 1338 pts evaluable for efficacy, 531 (40%) were 65–79y (mean age 72.5±4.2y; 55% women), 186 (14%) were >/=80y (mean age 84.8±3.6y; 58% women) and 621 (46%) <65y (mean age 52.0±10.5y; 62% women). Baseline (BL) characteristics between groups revealed those <65y, both 65–79y and >/=80y pts with CKD had significant reductions in calculated CrCL, lower SCr, and lower body weight despite comparable mean BL Hb (table). Mean Hb change from BL to final assessment was similar in all three groups (2.4±1.5g/dL for both <65y and 65–79y and 2.4±1.4g/dL for >/=80y). Responses to EPO were seen by weeks 4 and 8 (65–79y, 47.8% and 80.2%, >/=80y, 49.5% and 79.0% and <65y, 52.3% and 83.3%). The overall number of pts responding to EPO (Hb increase >/=2g/dL at any point from BL) was 471 (88.7%) in 65–79y; 166 (89.3%) in >/=80y and 564 (90.8%) in <65y. The percent of pts transfused at BL (65–79y, 10.0% >/=80y, 14.0% vs. <65, 11.3%; p=0.4794 and p=.3173 respectively) and during study was similar for all age groups assessed. Though mean weekly EPO doses were similar for all groups (65–79y, 11,331±3364U/w >/=80y, 11,251±3401U/w, <65, 11,514±3538 U/w), mean body weight dosing was significantly higher in both the 65–79y and >/=80y groups (150.0±50.7U/kg/w, 172±66.3U/kg/w vs <65, 140.0±56.9U/kg/w, p=.0018 and p<.0001 respectively). QW EPO was well tolerated in all three age groups and adverse events were those as previously reported. Conclusions: Though octogenarians with anemia due to pre-dialysis CKD have worse renal function, more comorbidities and presumably less marrow reserve, their response to doses of QW EPO is similar to younger adults. These data suggest that the “oldest old” have comparable Hb responses to EPO demonstrating that age is not a limiting factor in the administration of EPO therapy for CKD anemia. This response may in part be due to higher body weight dosing of EPO. Abstract Replicated grade samples were taken from runner, Spanish, and virginia-type farmers stock peanut lots. Each sample was graded according to the procedures of the Federal State Inspection Service. The variability of % foreign material (%FM) and % loose shelled kernels (%LSK) associated with a 1800-g sample was measured. The variability of % sound mature kernels (%SMK), % sound splits (%SS), % other kernels (%OK), % damage (%DAM), and % extra large kernels (%ELK) associated with a 500-g sample was also measured. The variance was shown to be a function of the magnitude of the grade determination and was described by a relationship derived from binomial theory. From the measured grade factors, the support price per gross ton was calculated for each grade sample using the 1988 USDA loan schedule. The variance of the price per gross ton was also estimated and appeared to be independent of the price per gross ton. The coefficients of variation averaged across all lots tested were 21.1, 18.7, 2.6, 21.2, 14.0, 55.3, 7.8, and 2.4% for %FM, %LSK, %SMK, %SS, %OK, %DAM, %ELK, and price per gross ton, respectively. The computed price per gross ton of a farmers lot that has a true value of $600 was estimated to vary from $573 to $627 95% of the time when using the 500 and 1800 g grade sample to measure each grade factor.

Bi4Ti3O12 (BTO) powders were synthesized using soft combustion techniques, specifically known as Solution Combustion Synthesis (SCS) and Low-temperature Combustion Synthesis (LCS). XRD results showed that the single phase BTO was obtained following calcinations at 800?C for the sample prepared using SCS; compared to after combustion, for the sample prepared using LCS. The TG/DTA showed a combustion temperature of around 252 - 280?C, which became stable with a single phase BTO at 500?C and above. Sintering was carried out at 1,100?C for 3 h, to determine the microstructures, grain orientation, relative density, dielectric, and ferroelectric properties, of the bulk ceramics. It was found that the bulk ceramics prepared using SCS exhibited elongated-like grains, strong (117)-preferred grain orientation, a relative density of 93.3%, ?r = 320 - 360, tan ? = 0.03 - 0.1, Pr = 6.8 ?C/cm2, and Ec = 20 kV/cm. Meanwhile, the bulk ceramics prepared using LCS showed plate-like grains, strong c-preferred grain orientation, a relative density of 83%, ?r = 143 - 195, tan ? = 0.01 - 0.38, Pr = 5.9 ?C/cm2, and Ec = 30 kV/cm.

Abstract. We derive annual and seasonal global climatologies of tropospheric NO2 profiles from OMI cloudy observations for the year 2006 using the cloud-slicing method on six pressure levels centered at about 280, 380, 500, 620, 720 and 820 hPa. A comparison between OMI and the TM4 model tropospheric NO2 profiles reveals striking overall similarities, which confer great confidence to the cloud-slicing approach to provide details that pertain to annual as well as seasonal means, along with localized discrepancies that seem to probe into particular model processes. Anomalies detected at the lowest levels can be traced to deficiencies in the model surface emission inventory, at mid-tropospheric levels to convective transport and horizontal advective diffusion, and at the upper tropospheric levels to model lightning NOx production and the placement of deeply transported NO2 plumes such as from the Asian summer monsoon. The vertical information contained in the OMI cloud-sliced NO2 profiles provides a global observational constraint that can be used to evaluate chemistry transport models (CTMs) and guide the development of key parameterization schemes.

SummaryStudies were conducted on two calves and a pregnant female captured from the forest to assess the feed utilization, growth pattern and milk composition in Mithun (Bos frontalis), a rare bovine species of north-eastern India. The DMI, kg/100 kg body weight was 0.05 with DM and OM digestibility of 56.95 ± 3.68 and 58.46 ± 3.51 percents, respectively, in calves. The growth was more during the May to October months, however, the average growth of 181.26 ± 23.51 g day-1 was obtained during the year on grazing. The Mithun was not a milk yielder, but became accustomed to milking. Fat content in milk varied from 6.8 to 7.8%. The total solids, protein, lactose, ash and solid not fat (SNF) were estimated at 18.0, 4.9, 5.0, 0.8 and 10.7% respectively with COB negative at 4°C. The birth weight of the male calf was estimated at 22.0 kg, and an average growth of 325.14 g day-1 was determined in 214 days with maximum weight gain during the first two months of age.

Cattle are essential for the growth and maintenance of a country's economy, but different blood parasites (Haemoparasites) can reduce their production, which causes both direct and indirect financial losses for the livestock industry. The incidence of hemoparasites and risk factors related to them were investigated in cattle butchered at the principal abattoir in Katsina. At the point of slaughter, 500 blood samples were aseptically taken, processed, and checked for hemoparasites by identifying parasites on thin blood smears stained with Giemsa. A prevalence of 8.4% was recorded overall. Anaplasma, Babesia, Microfilaria, and Trypanosoma, four genera of bovine hemoparasites, had prevalence rates of 4.0%, 2.4%, 2.4%, and 0.8%, respectively. The prevalence of hemoparasites was found to be greater in females (8.8%) than in males (7.7%), however there was no statistically significant difference (P 0.05). All breeds evaluated during the study tested positive for haemoparasites; the highest frequency, 16.1%, was found in Adamawa Gudali, followed by Sokoto Gudali (10.3%), White Fulani (7.2), and Red Bororo (6.8%). According to the study's findings, there is no statistically significant difference between breeds in the prevalence of haemoparasites (P 0.05). Young people were more likely to have high prevalence (9.1%) than adults (6.4%) or those who were older (8.7%). The current study has identified the haemoparasite status of cattle put to death at the principal abattoir in Katsina. Consequently, to increase livestock productivity, appropriate preventive and control measures are required.

677 Background: Napabucasin a first-in-class cancer stemness inhibitor in clinical development, suppresses cancer stemness by targeting Stat3-driven gene transcription. Preclinically, potent and broad-spectrum anti-cancer activity was observed in vitro and in vivo, alone and in combination with other agents. In a phase 1 study, napabucasin monotherapy was well tolerated with encouraging signs of anti-tumor activity at the RP2D of 500 mg BID. Methods: The current open-label, multi-center study includes phase II expansion in pts with refractory, K- Raswt mCRC to confirm safety and anti-tumor activity of napabucasin administered orally at 480mg BID in combination with panitumumab (6mg/kg bi-weekly). Results: 72 pts were enrolled, 48 pts were evaluable by RECIST of which 7 (15%) and 41 (85%) had 2 or >3 prior treatment lines, respectively. Of the 48 evaluable pts, 64.6% (31/48) were previously treated with an anti-EGFR agent. No new adverse events (AEs) were observed and most common AEs included grade 1/2 diarrhea, nausea, abdominal cramps, and vomiting. Among 48 pts enrolled who received RECIST evaluation, Disease Control Rate (DCR) was observed in 25 pts (52.1%) of which 3 pts achieved PR (6%) and 22 pts achieved SD (45%). Among 31 pts previously treated with anti-EGFR therapy, DCR was observed in 15 pts (48%) compared with DCR of 59% observed in 10 out of 17 anti-EGFR naïve pts receiving a scan. Conclusions: This phase II study confirmed that napabucasin can be safely combined with panitumumab at full dose and shows encouraging anti-tumor activity in pts with K- Ras wt mCRC, regardless of prior anti-EGFR exposure, suggesting that napabucasin may sensitize pts to repeat anti-EGFR therapy. In the present study, a simple, stability-indicating and cost-effective reversed-phase high performance liquid chromatography (HPLC) method was developed and validated for the determination of piroxicam in commercial and masterful formulation capsules as an alternative to existing methods. The improved HPLC method was carried out on a C18 column (150 mm x 4.6 mm i.d., 5 μm), maintained at 30 °C. The mobile-phase consisted of a solution of triethylamine 0.3% pH 5.0 and acetonitrile (70:30; v/v), run at a flow rate of 1.0 mL/min and using photodiode array (PDA) detection at 248 nm. The chromatographic separation is obtained with retention time of 6.8 min, presenting adequate system suitability parameters for HPLC analysis. Validation parameters such as the specificity, linearity, matrix effect, precision, accuracy and robustness were evaluated in accordance with the ICH Q2(R1) and Brazil RDC 166/17 requirements, giving satisfactory results within the acceptable range. The proposed method was successfully validated and applied for piroxicam analysis, contributing to improve the quality control, and can be used as easy, accurate, low time-consuming alternative to pharmacopeial quantification methodology of drug.

A suspension culture of Eucalyptus tereticornis was initiated from callus and grown for 7 months under indirect light in a Murashige and Skoog (1962) basal medium containing 3% glucose and 1 mg/l 2,4-D. Glucose was used, instead of sucrose, as it reduced production of phenolic-like compounds. The inoculum size for maximum cell yield was determined. Cells (0.1, 0.2, 0.5, 1.0, 2.5 and 5.0 g fresh weight) were cultured in basal medium for 14 days. Maximum fresh weight (mean 11.8 g) was attained from samples inoculated with at least 1.0 g of cells. Largest dry weight (mean 608 mg) occurred following, inoculation with at least 0.5 g fresh weight of cells. Inoculation with 0.5 g of cells resulted in the most rapid fresh weight doubling time (3.4 days).After 17 months of culture, cells were grown in basal medium or m basal medium supplemented with 1 mg/l kinetin, under continuous, direct light. Growth, based on fresh and dry weight increases, was measured over the 2-week subculture period. Growth of cells was similar in both media. The cells' chlorophyll content remained low. Fresh weight doubling time averaged 3.8 days.

BackgroundPsoriatic arthritis (PsA) is a chronic, immune-mediated inflammatory disease with heterogenous musculoskeletal manifestation (arthritis, spondylitis, enthesitis, dactylitis) and extra-musculoskeletal manifestation (skin and nail psoriasis). In addition, PsA is commonly associated with comorbidities such as metabolic syndrome and cardiovascular diseases where IL-17 is a key driver of this disease.Izokibep is a unique IL-17A inhibitor with extraordinary potency and small molecular size designed to overcome the limitations of monoclonal antibodies such as poor tissue distribution.Here, we report 16-week phase 2 results in patients with active PsA.ObjectivesTo assess efficacy, safety, pharmacokinetics and immunogenicity of izokibep versus placebo.MethodsThis is a prospective, multicenter, randomized, double-blind, placebo-controlled, parallel-groups, dose-finding trial studying bi-weekly 80 mg or 40 mg izokibep administered subcutaneously versus placebo until Week 16 (Period 1) and dose-controlled treatment until Week 46 (Period 2). PsA patients had to have ≥3 swollen and ≥3 tender joints of the 66/68 joint count, and an inadequate response to previous NSAIDs, csDMARDs or TNF inhibitor therapy. The primary endpoint was to evaluate ACR50 responses of 80 mg bi-weekly versus placebo at Week 16. Key secondary endpoints were ACR20/70, MDA, DAS28, DAPSA, SPARCC, LDI, PASI as well as tolerability and safety. Efficacy outcome measures were assessed in all patients who received at least one dose of study treatment. This trial is registered with ClinicalTrials.gov, NCT04713072.Results135 patients were randomized and treated between June 2020 and July 2021 in 22 European sites located in Austria, Belgium, Czech Republic, Germany, Hungary, Poland and Spain.At baseline, patients had a mean age of 48.5 (SD 12.0) years, a mean BMI of 29.0 (SD 4.8) kg/m2, a mean swollen joint count (SJC) of 9.9 (SD 6.6), and a mean tender joint count (TJC) of 16.7 (SD 10.4). The mean PsA disease duration was 7.1 (SD 7.8) years. 13% failed previous TNF inhibitor treatment and 80% received a concomitant csDMARD.At Week 16, the confirmatory primary endpoint ACR50 response rate was met (p=0.0003). ACR50 response rate was 52% in the 80 mg group, 48% in the 40 mg and 13% in the placebo group. Abstract (sommario):
Satafood Kehittämisyhdistys ry:llä on Manner-Suomen maaseudun kehittämisrahoituksella oleva Alituotantokasvien tuotannon kehittäminen –hanke, jossa kehitetään mm palkokasvien viljelyä. Hankkeessa on toteutettu uusien palkokasvien mm. sini-, valkolupiinin ja soijan koeviljelyä viljelijöiden pelloilla. Palkokasvit ovat typpiomavaraisia proteiinirikkaita kasveja, joilla pystytään katkaisemaan viljanviljelykierto. Viljelyksiltä on kerätty viljely- ja satotietoja sekä satonäytteitä. Näytteitä saatiin Karita, Hulda ja Stok herneestä, Kontu ja Fuego härkäpavusta, Haags Blaue ja Sonet sinilupiinista, Dieta valkolupiinista ja Elena soijapavusta. Herneestä ja härkäpavusta määritettiin proteiinipitoisuus. Sini- ja valkolupiinin sekä soijan näytteistä analysoitiin rehuanalyysi. Jokaisesta kasvilajista määritettiin aminohappoanalyysi ja kaikista vuoden 2010 näytteistä määritettiin fytiinihappopitoisuus. Stok-herneessä oli matalampi proteiinipitoisuus (214 g/kg) Karitaan (234 g/kg) ja Huldaan (223 ja 239 g/kg) verrattuna. Kontu härkäpavun proteiinipitoisuus oli jonkin verran korkeampi kuin Fuegon. Haags Blaue sinilupiinin proteiinipitoisuus oli vuonna 2010 keskimäärin 229 g/kg ja vuonna 2011 317 g/kg. Sinilupiini Sonetin proteiinipitoisuus oli 308 g/kg. Sinilupiininäytteiden rasvapitoisuus vaihteli 30-49 g/kg. Soijassa ja valkolupiinissa oli korkeimmat proteiini- ja rasvapitoisuudet. Elena soijapavun proteiinipitoisuus oli 379 g/kg ja Dieta valkolupiinin 382 g/kg. Elenan rasvapitoisuus oli 100 g/kg ja Dieta valkolupiinin 101 g/kg. Sinilupiinin kuitupitoisuus oli korkeampi kuin valkolupiinin ja soijan. Soijapapu ja valkolupiini olivat rehuanalyysituloksiltaan lähes samanarvoiset. Herne, härkäpapu, sini- ja valkolupiini sekä soija poikkesivat toisistaan aminohappokoostumuksen suhteen. Herneen (7,8 g/100 g) ja härkäpavun (7,1 g/100 g) proteiinissa oli enemmän lysiiniä kuin soijan (6,8 g/100 g) tai sini- (5,8 g/100 g) ja valkolupiinin (5.0 g/100 g) proteiinissa. Korkein rikkipitoisten aminohappojen metioniinin ja kystiinin määrä oli soijassa (3,3 g/100 g). Myös sinilupiinissa rikkipitoisten aminohappojen määrä oli melko korkea (2,9 g/100 g) proteiinia. Tutkittujen palkoviljojen fytiinihappopitoisuudet olivat erilaiset. Rehussa fytiinihappo alentaa 1-mahaisten eläinten kasvua. Matalin fytiinihappopitoisuus oli valkolupiinissa (6,2 mg/g ka) ja korkein soijapapu Elenassa (22,6 mg/g ka). Fuego härkäpavun fytiinihappopitoisuus (11,7 mg/g ka) oli alempi kuin Kontu härkäpavun pitoisuus (16,5 mg/g ka). Haags Blaue sinilupiinin fytiinihappopitoisuus (13,6 mg/g ka) oli matalampi kuin Kontu härkäpavun, mutta kuitenkin korkeampi kuin hernelajikkeiden vastaavat arvot. Hernelajikkeilla Karitalla (9,6 mg/g ka) ja Huldalla (11,1 mg/g ka) oli toiseksi matalimmat fytiinihappopitoisuudet valkolupiinin jälkeen. Vähän viljellyistä palkokasveista sini- ja valkolupiini ovat mielenkiintoiset valkuaiskasvit Suomen peltoviljelyyn.
