Abstract (sommario):
Methods borrowed from nonequilibrium thermodynamics and statistical physics have been employed in the quantitative analysis of spatiotemporal chaos in a 2D coupled map lattice (CML). Emphasis is made on entropy, entropy variation and entropy production. These quantities manifest peculiar changes in a region where spiral waves emerge. The spiral waves observed in the 2D CML are found to be dissipative objects with an elevated entropy production.

Abstract (sommario):
The present study examined the effectiveness of three types of virtual desktop displays with varying degrees of perceptual cues: two-dimensional (2D) display, three-dimensional (3D) display without motion, and 3D display with motion parallax. It was expected that performance would improve as the number of perceptual cues increased. Participants were asked to search for documents either with an organizational scheme they created themselves or with a preconstructed arrangement. As expected, those who organized their own desktops performed better than those using a preconstructed arrangement; however, these effects were moderated by cue condition. Differences among the cues had no effect on subjective organization. However, searching a preconstructed arrangement benefited from a 2D representation. These findings suggest that a 3D representation may be ill suited to tasks in which semantic material is stored in a virtual information space.

Fernandez, Hugo. "structure et auto-organisation de polymères conducteurs en films ultraminces". Electronic Thesis or Diss., CY Cergy Paris Université, 2024. http://www.theses.fr/2024CYUN1322.

Testo completo
Abstract (sommario):
Poly(3-alkylthiophenes) (P3ATs) are electronically conducting polymers used as active layers for example in photovoltaic cells or field-effect transistors typically elaborated as thick films (≈ 20-200 nm-thick) by spin-coating. Conductive properties of P3ATs are strongly dependant on their organization. Hence, this thesis work focuses on the Langmuir film technique to develop ultrathin organized films and study the relationship between structure and conductive properties. We have characterized Langmuir film structure of poly(3 hexylthiophene) (P3HT), poly(3-butylthiophene) (P3BT) and poly(3-decylthiophène) (P3DT) having different side chain length, and poly[3-(potassium-7-heptanoate)thiophene-2,5-diyl] (P3PHT) differing from P3HT by the presence of a hydrophilic COOK side chain end group. For this, several X-ray scattering techniques have been used and in certain cases, atomistic simulations of diffraction spectra allowed the precise positioning of polymer chains within the structure. P3HT and P3BT form highly organized bilayers 3 to 4 nm-thick with an edge-on conformation on water. P3HT structure has similitudes with the form I observed in P3ATs thick films, while that of P3BT is intermediate between forms I and II. Morphology of these films corresponds to a nanofiber carpet. P3DT film shows coexistence of two polymorphs close to form I and II in thick films and its morphology is also different. The presence of a hydrophilic group with P3PHT causes a drastic modification of the structure consisting in a poorly organized compressible monolayer. We also studied doping of P3ATs films with F4TCNQ using two methods, either co-deposition of dopant and polymer or sequential doping. In both cases, F4TCNQ insertion into the P3HT or P3BT bilayer induces a thickening of the films accompanied by a contraction of the 2D lattice in the π-stacking direction due to the formation of polarons, as evidenced by UV-visible spectroscopy. Upon doping of P3DT, only form I is changed, indicating partial doping of the film. The electronic conductivity of the P3DT film is significantly lower than that of the P3HT or P3BT film. For the P3HT film, conductivity reaches a maximum of 0.1 S/cm when the doping level corresponds to one F4TCNQ molecule for two bithiophene units. It can be increased up to 5 S/cm with only five stacked layers

Abstract (sommario):
The field of entrepreneurship is rapidly advancing and matures as a discipline that receives substantial amount of attention. One popular area of research in the discipline of entrepreneurship is to investigate one’s intent to start a business, which is entrepreneurial intention. This is an important construct that warrants ongoing research because entrepreneurial intention is not only a great predictor of entrepreneurial behavior but also an important step in the process of becoming an entrepreneur. The present study, based on a sample of 321 subjects along with 264 observers, makes five contributions to the entrepreneurship literature. First, I examined the psychometric property of entrepreneurial take-over intention and found that it is a construct different from entrepreneurial start-up intention. Second, the results demonstrated that risk propensity and proactive personality are positive predictors of entrepreneurial start-up and take-over intentions, whereas cognitive ability is a negative predictor of entrepreneurial start-up and take-over intentions. Rebelliousness is a positive predictor of entrepreneurial take-over intention and also has an inverted U-shaped relationship with entrepreneurial take-over intention. Third, entrepreneurial self-efficacy mediates the relationship between three individual traits (i.e., emotional intelligence, risk propensity, and proactive personality) and entrepreneurial start-up and take-over intentions. Need for cognition mediates the relationship between two individual traits (i.e., cognitive ability and proactive personality) and entrepreneurial start-up intention. Fourth, 2D:4D ratio (a proxy measure for prenatal testosterone exposure level) negatively predicts risk propensity. There also exist two two-step mediations from 2D:4D ratio to both entrepreneurial start-up and take-over intentions through risk propensity and entrepreneurial self-efficacy. Fifth, the results suggest that observer ratings of individual traits only contribute modest incremental validity above and beyond self-reported ratings of them in predicting entrepreneurial start-up and take-over intentions. I discuss implications, limitations, and future directions informed by the present study.

Abstract (sommario):
Mitochondria are the power plant of most non-green eukaryotic cells. In order to understand mitochondrial functions and their regulation, knowledge of the spatiotemporal organization of their proteins is important. Mitochondrial membrane proteins can diffuse within membranes. They are involved in diverse functions e.g. protein import, cell respiration, metabolism, metabolite transport, fusion, fission or formation of the mitochondrial architecture. Furthermore, mitochondria compose of different subcompartments with different tasks. Especially, the inner mitochondrial membrane (IM), where the oxidative phosphorylation (OXPHOS) takes places, has a complex architecture with cristae extending into the matrix. The present work revealed the restricted localization of some mitochondrial proteins to specific membrane sections and linked it to their function or bioenergetic circumstances in the living cell.
Single particle tracking (SPT) techniques like tracking and localization microscopy (TALM) allow to localize proteins with a precision below 20 nm. Additionally, tracking single proteins provides information about their mobility, dynamic and their spatiotemporal organization. TALM uses proteins, which were genetically tagged either with the HaloTag® (HaloTag) or the fSnapTag® (fSnapTag). These tags can be orthogonally and posttranslationally stained with specific and self-marking dyes. If the dyes are conjugated to the respective substrate of the tag. Single molecule labeling of mitochondrial proteins was performed substoichiometrically using membrane permeable rhodamine dyes, either tetramethylrhodamine (TMR) or silicon rhodamine (SiR). TALM allowed to localize proteins in different mitochondrial subcompartments. The gained trajectories and trajectory maps of mitochondrial proteins revealed their spatiotemporal organization. In the case of IM proteins like F1FO ATP synthase (Complex V - CV) a restricted diffusion in the CM, which is part of the continuous IM, was determined. The unimpeded diffusion of mitochondrial proteins in the outer mitochondrial membrane (OM) was compared with the mobility of IM proteins. The diffusion of mitochondrial IM proteins was restricted by the IM architecture and their diffusion coefficients were lower. Furthermore, significant differences of different mitochondrial IM proteins were compared, showing different localizations in the IM often coupled to their function, accompanied by different spatiotemporal organization and diffusion coefficients. Furthermore, a distinction was made between diffusion of proteins in the inner boundary membrane (IBM) and proteins that preferentially diffuse in the cristae membrane (CM). Evaluating trajectory maps, the different subcompartments in the IM were revealed by trajectories and the trajectory directionality, allowing the identification of mitochondrial proteins, which mark these subcompartments.
The morphology of mitochondria / mitochondrial networks and their bioenergetic parameters are linked to the metabolic states of the cell. In this work, the connection of the spatiotemporal protein organization of CV and the IM architecture was uncovered on the micro- and nanoscopic level and linked to the metabolic state of the cell. It was determined that the spatiotemporal organization of the CV was altered, when CV was inhibited. In addition, the bioenergetic influence of cells on the spatiotemporal behavior of CV and the reorganization of the IM architecture was investigated by TALM and compared with results of electron microscopy images. It was shown that starvation of cells led to a loss of cristae and thus to an increased mobility and spatiotemporal reorganization of CV. Taken together, the results presented in this work showed that a correctly functioning and active CV helps to maintain the IM architecture and both, the spatiotemporal organization of CV and the IM architecture were coupled to the metabolic state..
In order to investigate putative protein-protein interactions by colocalization and co-locomotion studies on single molecule level, dual color SPT is needed. Therefore, posttranslational and substoichimetric labeling as performed in TALM was tested for its potential of protein-protein interaction studies of mitochondrial membrane proteins. Here, a genetically double tagged translocase of the outer membrane subunit-20 (Tom20) (Tom20:HaloTag:fSnapTag) acted as a positive control. It turned out that substoichimetric, posttranslational labeling of mitochondrial proteins was not suitable for protein-protein interaction studies on mitochondrial proteins, because it was restricted by the low labeling degrees needed for TALM. However, dual-color TALM still allowed to study effects of proteins influencing the IM architecture and to study their influence on the spatiotemporal organization of CV. The co-transfection of Mic10, as the central protein of the mitochondrial inner membrane organizing system / mitochondrial contact site complex / mitochondrial organizing structure (MINOS / MICOS / MitOS (MINOS/MICOS)), altered the regular and aligned organization of the cristae. This was measured by a changed spatiotemporal organization of the CV, such as the loss of the perpendicular oriented of CV subunit-γ (CV-SUγ) cristae trajectories. In contrast to this, co-transfection of CV subunit-e (CV-SUe), important for dimerization of CV, increased the number of cristae trajectories.
Mitochondria are three-dimensional (3D) cell organelles. Consequently, subcompartments like the IBM and CM are a 3D space in which CV is localized and diffuses. Thus, the diffusion of mitochondrial proteins is underestimated by two-dimensional SPT e.g. lateral confined diffusion can result from mitochondrial proteins diffusing along the z-axis of the microscope. In order to reveal the 3D spatiotemporal organization of CV, the potential of TALM to be extended to a 3D-SPT technique was investigated. Therto a cylindrical lens was installed in the emission path of a total internal reflection fluorescence (TIRF) microscope. This leads to an astigmatically distorted point spread function (PSF) of the fluorescent single molecule signals. This distortion allowed the reconstruction of single molecule localizations of CV to a superresolved image of the IM, in living cells. In addition, 3D-TALM enabled to display the 3D architecture of the IM by 3D trajectories of CV. 3D-TALM was able to detect whether CV diffuses in the IBM or in the CM, and extended the information about its mobility in the CM that it takes place in a disc-like manner. In this way it could be shown that CV is mobile within the cristae in all directions. Finally, 3D-TALM revealed an altered IM architecture caused by the metabolic state of the cell. As performed in two-dimensional TALM, the cells were kept under starving conditions. Here the now tubular IM architecture was revealed by 3D-TALM. The reversed metabolic state under improved respiratory conditions unexpectedly led to a more diverse IM architecture. These ultrastructural changes were also revealed by electron microscopy. Consequently, 3D-TALM enabled the study of IM architecture by tracking CV under different metabolic conditions, allowing an ultrastructural analysis of mitochondria in living cells. In addition, 3D TALM provided the spatiotemporal organization of CV under different metabolic conditions, so that the diffusion coefficients of CV could be related to changes in IM architecture caused by the metabolic condition.

Abstract (sommario):
AbstractSpatial organization on the atomic scale is one of the key objectives of nanotechnology. The development of DNA nanotechnology is a hallmark of material programmability in 2D and 3D, in which the large variety of available DNA modifications allows it to be interfaced with a number of inorganic and organic materials. Nature’s solution to spatiotemporal control has been the evolution of self-organizing protein systems capable of pattern formation through energy dissipation. Here, we show that combining DNA origami with a minimal micron-scale pattern-forming system vastly expands the applicability of DNA nanotechnology, whether for the development of biocompatible materials or as an essential step toward building synthetic cells from the bottom up. We first describe the interaction of DNA origami nanostructures with model lipid membranes and introduce the self-organizing MinDE protein system from Escherichia coli. We then outline how we used DNA origami to elucidate diffusiophoresis on membranes through MinDE protein pattern formation. We describe how this novel biological transport mechanism can, in turn, be harnessed to pattern DNA origami nanostructures on the micron scale on lipid membranes. Finally, we discuss how our approach could be used to create the next generation of hybrid materials, through cargo delivery and multiscale molecular patterning capabilities.

Abstract (sommario):
AbstractHumanities and scientific collections institutions and cultural memory organizations around the world are incorporating advanced digital representations of their materials into their conservation practice, service to the public, and materials preservation strategy. Understanding the reliability of digital assets is key to the success of these activities. Imaging tools and methods are now available that produce 2D and 3D digital representations which carry a metadata record describing the process of their creation. The open-source Digital Lab Notebook software simplifies the creation of such a metadata record and assists in its long-term archiving and preservation. This transparent record allows users, both within and outside of collection organizations, to evaluate the quality of the representation, understand its precision, and determine its utility for their own purposes.

Abstract (sommario):
Abstract One of the central tenets of Gestalt psychology is that configurations of stimulus elements are perceptually organized to have the lowest possible structural complexity—what is often referred to as the ‘‘minimum principle.’’ Consider, for example, the image of a Necker cube presented in the left panel of Figure 31.1. It is important to keep in mind that the actual physical structure of this figure is a 2D configuration of lines, but that is not how it is perceived. It appears instead as a 3D cube, presumably because that structure is simpler than the alternative 2D interpretation.

Abstract (sommario):
The Canada in 3D (C3D) project (https://canada3d.geosciences.ca/), formally initiated in the spring of 2020 by the National Geological Surveys Committee (NGSC) is required to provide a working group update to all its provincial and territorial partners. There have been several informal C3D working meetings with the partners prior to the creation of the C3D Charter and there has been a hiatus in communication through the Covid-19 pandemic. To re-engage the C3D community, a video tele-conference was held on June 6th, 2022 with approximately 44 participants. There was representation and presentations of all provinces and territories with various managers, technical and scientific observers. The purpose of this compilation of presentations and discussions from this 2022 C3D-NGSC reconnection meeting is to provide activity information to all participants, and their respective organizations, highlighting current geoscience compilation and modelling efforts in 2D and 3D. The aim is to help identify opportunities for collaboration on data standards, methods, applications and best practices but with the overall goal of working toward the C3D vision, outlined in the C3D charter of an updated 2D and 3D geological map/model of Canada.
