This type of research is a qualitative descriptive type by conducting observations and interviews directly to CV Percetakan Karya. The result of this study shows that the accurate program can make easier to compile CV Percetakan Karya financial statements, the resulting report in the form of income statement and balance sheet report. The accurate program also makes easier fo CV Percetakan Karya to entry transaction data into the accurate program. The accurate program can provide detailed and complete financial statements, so companies can easily analyze the financial statements.

Abstract Triple-negative human breast cancers (TNBC) lack three proteins commonly targeted by chemotherapy; estrogen and progesterone receptors, and her-2-neu. Most current protocols used to treat TNBC are largely ineffective and aggressive tumors frequently re-emerge, leading to metastasis and patient death. Thus, new therapies for TNBC are needed. Recent studies show that around 80% of TNBC express mutant p53 (mtp53), a functionally defective form of the p53 tumor suppressor protein. If mtp53 is converted into the active wild-type protein (wtp53), tumor suppressor functions are recovered. Most p53 mutations occur in the DNA-binding domain, causing normal regulation of p53 target genes involved in apoptosis, cell-cycle arrest, and angiogenesis to be blocked. This promotes metastasis and renders tumors resistant to chemotherapy. APR-246 is a small-molecule drug that re-activates mtp53 by covalent modification of the DNA-binding core domain of the mutant protein through alkylation of thiol groups and has been shown to reactivate mtp53 and restore p53 function. We examined whether APR-246 could inhibit TNBC growth, both in vitro and in vivo. Cell viability assays and FACS were used to measure in vitro TNBC cell growth and apoptosis respectively, in MDA-MB-231 and MDA-MB-468 cells, with MCF-7 cells (which express wtp53) as controls. Analysis of TNBC growth in vivo was assessed in a mouse model of MDA-MB-231 derived xenografts. Nuclear extracts of APR-246-treated TNBC cells exhibited significantly increased p53 DNA binding compared with untreated cells, indicating that APR-246 converts mtp53 to wtp53 in these cells. APR-246 significantly reduced TNBC cell viability in vitro, but had no effect on normal mammary cells or wtp53-expressing MCF-7 cells. Pro-apoptotic proteins, Bax, p21 and caspase-3 were elevated in APR-246 treated cells, while the cell survival protein Bcl-2 was suppressed. In the xenograft model, animals were given an intravenous (iv) tail vein injection of APR-246 alone (100 mg/kg/day) once tumors reached 100 mm3. A second group received an intraperitoneal (ip) injection of 2aG4 antibody (100 µg/mouse/day), which targets phosphatidylserine and disrupts tumor blood vessel formation. A third group was given both APR-246 and 2aG4 using the same doses above. A control group received antibody C44 (100 µg/mouse/day, ip) and/or PBS (0.1 mL/day, iv). A total of 18 treatments were used. Administration of APR-246 alone or in combination with 2aG4, significantly reduced TNBC tumor growth, as well as two markers of angiogenesis (vascular endothelial growth factor expression and blood-vessel density). APR-246 in combination with 2aG4 completely eradicated almost 20% of the TNBC tumors. We conclude that TNBC is inhibited by APR-246 and 2aG4. Such treatment could represent an effective and innovative means of combating these particularly aggressive and deadly types of cancer.

ABSTRACT ST-246 was evaluated for activity against cowpox virus (CV), vaccinia virus (VV), and ectromelia virus (ECTV) and had an in vitro 50% effective concentration (EC50) of 0.48 μM against CV, 0.05 μM against VV, and 0.07 μM against ECTV. The selectivity indices were >208 and >2,000 for CV and VV, respectively. The in vitro antiviral activity of ST-246 was significantly greater than that of cidofovir, which had an EC50 of 41.1 μM against CV and 29.2 μM against VV, with selectivity indices of >7 and >10, respectively. ST-246 administered once daily by oral gavage to mice infected intranasally with CV beginning 4 h or delayed until 72 h postinoculation was highly effective when given for a 14-day duration using 100, 30, or 10 mg/kg of body weight. When 100 mg/kg of ST-246 was administered to VV-infected mice, a duration of 5 days was sufficient to significantly reduce mortality even when treatment was delayed 24 h postinoculation. Viral replication in liver, spleen, and kidney, but not lung, of CV- or VV-infected mice was reduced by ST-246 compared to levels for vehicle-treated mice. When 100 mg/kg of ST-246 was given once daily to mice infected by the intranasal route with ECTV, treatment for 10 days prevented mortality even when treatment was delayed up to 72 h after viral inoculation. Viral replication in target organs of ECTV-infected mice was also reduced.

Abstract In murine schistosomiasis, H-2k CBA and C3H mice develop severe CD4 T cell(CD4) mediated liver pathology(LP) and a high Th1/17 response very focused on a single epitope of the parasite egg antigen Sm-p40(p234-246). Low pathology C57BL/6 mice do not recognize this antigen. A possible reason for this result is that p234-246 exacerbated dominance(ED) polarizes the anti-egg response causing severe LP. To test this hypothesis we: a)analyze the LP of SJL mice whose H-2 haplotype also exhibit ED of a single Sm-p40 epitope and b) analyze the LP of CBA mice whose p234-246 ED was neutralized by an epitope cocktail. Methodology:H-2s Sm-p40 restriction pattern was determined with the rankpep software. SJL LP was documented by comparative liver morphometric analysis(LMA) of infected CBA, C57BL/6 and SJL mice euthanized 7 weeks-post-infection(wpi). The effect of p234-246 ED neutralization was studied by LMA of 3 groups of infected CBA mice euthanized at 7wpi. The 1st group was treated with an emulsion of adjuvant and 4 synthetic epitopes designed to compete for I-Ak with an affinity equal or higher than p234-246, the 2nd was treated with adjuvant alone and the 3rd was not treated. Results:LMA shows that SJL mice develop as severe a LP as CBA mice and that p234-246 ED neutralization prevents development of severe LP. Conclusion:These results show that ED may be pathogenic in CD4 mediated diseases and that its neutralization may be an effective therapeutic strategy that deserves exploration.

Copper(II) complexes with 2,3,4-trimethoxybenzoic acid (H234-tmbz) and 2,4,6-trimethoxybenzoic acid (H246-tmbz), [Cu2(234-tmbz)4(H2O)2] (6) and [Cu(246-tmbz)2(µ-H2O)2(H2O)2]n (7), were synthesized and characterized by elemental analysis, infrared and UV-vis spectra and temperature dependence of magnetic susceptibilities (1.9–300 K). The X-ray crystal structures revealed that the former 6 is a dinuclear cluster having syn-syn-bridged Cu2(µ-234-tmbz)4 core with Cu···Cu separation of 2.6009(7) Å, while the latter 7 is a µ-aqua-bridged chain molecule consisting of Cu(246-tmb)2(µ-H2O)2(H2O)2 units with Cu···Cu separation of 4.1420(5) Å. Temperature dependence of magnetic susceptibilities showed that an antiferromagnetic interaction with 2J = −272 cm−1 for 6 and a weak antiferromagnetic interaction with J = −0.21 cm−1 for 7, between the two copper(II) ions. The adsorption isotherm of 6 showed Types I behavior having a 125.4 m2g−1 of specific surface area.

Purpose APR-246 (PRIMA-1MET) is a novel drug that restores transcriptional activity of unfolded wild-type or mutant p53. The main aims of this first-in-human trial were to determine maximum-tolerated dose (MTD), safety, dose-limiting toxicities (DLTs), and pharmacokinetics (PK) of APR-246. Patients and Methods APR-246 was administered as a 2-hour intravenous infusion once per day for 4 consecutive days in 22 patients with hematologic malignancies and prostate cancer. Acute myeloid leukemia (AML; n = 7) and prostate cancer (n = 7) were the most frequent diagnoses. Starting dose was 2 mg/kg with dose escalations up to 90 mg/kg. Results MTD was defined as 60 mg/kg. The drug was well tolerated, and the most common adverse effects were fatigue, dizziness, headache, and confusion. DLTs were increased ALT/AST (n = 1), dizziness, confusion, and sensory disturbances (n = 2). PK showed little interindividual variation and were neither dose nor time dependent; terminal half-life was 4 to 5 hours. Tumor cells showed cell cycle arrest, increased apoptosis, and upregulation of p53 target genes in several patients. Global gene expression analysis revealed changes in genes regulating proliferation and cell death. One patient with AML who had a p53 core domain mutation showed a reduction of blast percentage from 46% to 26% in the bone marrow, and one patient with non-Hodgkin's lymphoma with a p53 splice site mutation showed a minor response. Conclusion We conclude that APR-246 is safe at predicted therapeutic plasma levels, shows a favorable pharmacokinetic profile, and can induce p53-dependent biologic effects in tumor cells in vivo.

Abstract The objective of this study was to evaluate different nutritional management strategies (drylot vs. supplementation on pasture) to optimize growth and reproductive performance of Nellore heifers submitted to the fixed-time artificial insemination (FTAI) at 14–15 mo. On d0, 126 weaned Nellore heifers (152±22 kg of BW; 240±28 days of age) were stratified by BW and age, and randomly assigned to receive: (1) high-concentrate-based diet (62.2% TDN, 12.0% CP of DM of diet) ad libitum in feedlot from d 0 to 246 (FDL; 6 pens and 7 heifers/pen); (2) fixed supplementation at 1% of BW on Brachiaria pastures from d0 to 246 (PAS10; 6 pastures; 7 heifers/pastures and 4 ha/pasture); and (3) increasing supplementation on Brachiaria pastures at 0.8% of BW from d 0 to 80 and 1.2% of BW from d 81 to 246 (PAS812; 6 pastures; 7 heifers/pastures and 4 ha/pasture). Supplement offered to heifers on pasture consisted of corn, soybean meal, urea, and minerals (78% TDN, 22% CP of DM at dry season and 79% TDN, 20% CP of DM at rainy season). Final pregnancy rate was determined on d238. Heifer BW on d160 was greater (P < 0.01) for FDL vs. PAS10 and PAS812 (264 vs. 251 and 253±10.1 kg, respectively). Overall ADG from d0 to the start of breeding season (d160) and BW on d246 were greater (P < 0.01) for FDL vs. PAS10 and PAS812 (0.72 vs. 0.62 and 0.64±0.02 kg/d, and 325 vs. 310 and 316 ±10.3 kg, respectively). Heifer BW on d160 and 246 and ADG from d0 to 160 did not differ between PAS10 and PAS812 (P ≥ 0.33). Final pregnancy rates did not differ (P = 0.75) among treatments. Therefore, growth performance, but not pregnancy rates following FTAI, was enhanced for Nellore heifers developed in drylot vs. pasture systems and submitted to FTAI at 14–15 mo.

Objectives: The study aimed at determining the frequency and severity of depression in primary caregivers of psychotic patients. Study Design: Cross sectional study. Setting: Out-patient as well as in-patient setting of Department of Psychiatry, Military Hospital Rawalpindi. Period: Six months, from 20th April 2017 to 19th October 2017. Material and Methods: Purposive, non-probable sampling technique was used. A total of 246 caregivers of the same number of psychotic patients were studied after consent, using the General Health Questionnaire 28 (GHQ-28) and Beck’s Depression Inventory (BDI). The mean total BDI scores were calculated and correlated with the socio-demographic variables. Results: The mean total GHQ-28 score of 246 caregivers was 7.32 (cut off score is >7) which indicate definite psychopathology among them. Out of 246 caregivers, 175 (71.1%) showed scores greater than 7 (GHQ positive) while 71 (28.9%) showed scores less than 7 (GHQ negative). The mean total BDI score of 175 caregivers (who were GHQ positive) was 17.83 (cut off score for depression is >9). Out of 175 GHQ positive caregivers, 72 (41.14%) showed subclinical or no depression, 49 (28%) showed mild depression, 40 (22.86%) showed moderate depression and 14 (8%) showed severe depression on BDI. The results show that there is considerable caregiver burden in relatives of psychotic patients. Conclusion: Caregivers of psychotic patients suffer clinical depression with obvious negative outcomes for the patient. Younger age group, female gender and illiteracy typically carry a greater risk of experiencing higher burden in terms of depression.

Abstract Abstract 3773 Poster Board III-709 Introduction The tumor suppressor protein p53 induces cell cycle arrest and/or apoptosis in response to various forms of cellular stress, through transcriptional regulation of a large number of down stream target genes. p53 is frequently mutated in cancer, and cancer cells carrying defects in the p53 protein are often more resistant to conventional chemotherapy. Thus, restoration of the wild type function to mutant p53 appears to be a new attractive strategy for cancer therapy. APR-246 is a novel small molecule quinuclidinone compound that has been shown to reactivate non-functional p53 and induce apoptosis. Although the exact molecular mechanism remains to be determined, recent results suggest that an active metabolite of APR-246 alkylates thiol groups in the core domain of p53, which promotes correct folding of p53 and induces apoptosis (Lambert et al., Cancer Cell 15, 2009). Currently, APR-246 is in Phase I/IIa clinical trials for hematological malignancies and prostate cancer. In the present abstract results from in vitro, ex vivo and in vivo preclinical studies with APR-246 are presented. Results The lead compound of APR-246, PRIMA-1 (p53 reactivation and induction of massive apoptosis), was originally identified by a cellular screening of the NCI library for low molecular weight compounds (Bykov et al., Nat. Med., 8, 2002). Further development and optimization of PRIMA-1 led to the discovery of the structural analog APR-246 (PRIMA-1MET), with improved drug like and preclinical characteristics. In in vitro experiments APR-246 reduced cell viability (WST-1 assay) in a large number of human cancer cell lines with various p53 status, including several leukemia (CCRF-CEM, CEM/VM-1, KBM3), lymphoma (U-937 GTP, U-937-vcr), and myeloma (RPMI 8226/S, 8226/dox40, 8226/LR5) cell lines, as well as many solid cancer cell lines, including osteosarcoma (SaOS-2, SaOS-2-His273,U-2OS), prostate (PC3, PC3-His175, 22Rv1), breast (BT474, MCF-7, MDA-MB-231), lung (H1299, H1299-His175) and colon cancer (HT-29). In human osteosarcoma cell lines APR-246 reduced cell viability and induced apoptosis (FLICA caspase assay) in a concentration dependent manner being more potent in the p53 mutant (SaOS-2-His273) than in the parental p53 null (SaOS-2) cells. The IC50 values (WST-1 assay) were 14 ± 3 and 27 ± 5 μM, respectively (n=35). In in vivo subcutaneous xenograft studies in SCID (severe combined immunodeficiency) mice APR-246 reduced growth of p53 mutant SaOS-2-His273 cells in a dose-dependent manner, when injected i.v. twice daily with 20 -100 mg/kg (64 – 76% inhibition). An in vivo anticancer effect of APR-246 was also observed in hollow-fiber test with NMRI mice using the acute myeloid leukemia (AML) cell line MV-4-11. An ex vivo cytotoxic effect of APR-246 and/or its lead compound PRIMA-1 has also been shown in primary cells from AML and CLL (chronic lymphocytic leukemia) patients, harbouring both hemizygously deleted p53 as well as normal karyotype (Nahi et al., Br. J. Haematol., 127, 2004; Nahi et al., Br. J. Haematol., 132, 2005; Jonsson-Videsater et al., abstract at this meeting). APR-246 was also tested in a FMCA (fluorometric microculture assay) test using normal healthy lymphocytes (PBMC) and cancer lymphocytes (CLL). It was 4-8 fold more potent in killing cancer cells than normal cells, indicating a favorable therapeutic index. This is in contrast to conventional cytostatics that often show negative ratio in this test. Furthermore, when tested in a well-defined panel of 10 human cancer cell lines consisting of both hematological and solid cancer cell lines, the cytotoxicity profile/activity pattern of APR-246 differed from common chemotherapeutic drugs (correlation coefficient less than 0.4), suggesting a different mechanism of action. Conclusion In relevant in vitro, in vivo and ex vivo cancer models, APR-246 showed unique pharmacological properties in comparison with conventional cytostatics, by being effective also in cancer cells with p53 mutations and by demonstrating tumor specificity. Moreover, in experimental safety/toxicology models required to start clinical trials, APR-246 was non toxic at the predicted therapeutic plasma concentrations. Thus, APR-246 appears to be a promising novel anticancer compound that may specifically target cancer cells in patients with genetic abnormality associated with poor prognosis. Objective: To determine frequency of hyperuricemia among the patients having metabolic syndrome. Study Design: Cross Sectional study. Setting: Indus Medical College, Tando Muhammad Khan. Period: 28-06-2021 till 28-12-2021. Material & Methods: After informed consent, 246 cases of metabolic syndrome who met the inclusion criteria were included. Inclusion criteria were the known case of metabolic syndrome for more than six months, of age group 20 to 60 years and of either gender. Exclusion criteria were the non-consenting patients as well as the patients with history of hyperthyroidism and hypothyroidism, gout, congestive cardiac failure, chronic renal failure, Post myocardial infarction, chronic liver disease, stroke, COPD & Pregnancy. Data analyzed on IBM SPSS 22.0 and p value< 0.05 taken as statistically significant. Results: Out of 246 patients mean age in our study was 40.80 years with the standard deviation of ±7. Mean FBS, triglycerides, HDL, SBP, DBP and waist circumference was 115.64±10.16 mg/dl, 144.7±10.43 mg/dl, 41.61±4.03 mg/dl, 168.5±4.16, 98.2±2.78 and 101±8.16 cm respectively. 93 (37.8%) were male and 153 (62.2%) were female. Out of 246 patients, 81 (32.9%) had hyperuricemia and 165 (67.1%) did not have hyperuricemia in patients with metabolic syndrome. Conclusion: This study concluded that hyperuricemia is 32.9% common in metabolic syndrome patients.

Abstract Introduction: TP53 mutant (mTP53) MDS and AML, accounting for 5-10% of de novo MDS and 25-30% of therapy related MDS (t-MDS), represent a distinct molecular cohort with inferior outcomes. Hypomethylating agents (HMA) are preferred treatments for patients (pts) with these mutations, although with CR rates of only 20-30% and median OS of 6-12 months. APR-246 is a novel, first-in-class small molecule that selectively induces apoptosis in mTP53 cancer cells through mutant p53 protein re-activation by restoring the wild-type conformation, with single agent activity in mTP53 AML. We report the planned, completed Phase 1b results of APR-246+ azacitidine (AZA) in mTP53 MDS/AML. Methods: This is a multicenter Phase 1b/2 trial of APR-246+AZA in HMA naïve mTP53 MDS and oligoblastic AML (≤ 30% blasts) pts ≥ 18 years of age. Pts received APR-246 in a 3+3 dose escalation design (50, 75, 100 mg/kg lean body weight (equivalent to 4500mg fixed dose based on PK studies)) IV daily over 4 days in a lead-in phase (days -14 to -10) followed by the same dose of APR-246 (days 1-4) + AZA 75 mg/m2 SC/IV over 7 days (days 4-10 or 4-5 and 8-12) in 28 day cycles. The primary objective was to define safety and the recommended Phase 2 dose (RP2D), with AEs graded by CTCAE v4.03 and DLT assessment over 6 weeks. Secondary objectives included response by IWG 2006 criteria as well as serial next generation sequencing (NGS) and p53 IHC for evaluation of clonal suppression and remission depth as predictors of outcomes. For minimal residual disease (MRD) analysis, a custom target-capture NGS assay was developed using unique molecular Identifiers for error correction with a limit of detection of 0.1% with results validated by pt specific digital droplet PCR (ddPCR). Nanostring nCounter RNA expression analysis was conducted on a panel of 770 genes after the lead-in phase to assess transcriptional effects induced by APR-246. Results: As of July 30, 2018, 12 pts (42% male; median age 66 years (39-73)) were enrolled. Three pts had AML-MRC and 9 had MDS; all pts had poor risk cytogenetics (17% poor, 83% very poor) and higher risk disease by IPSS-R (25% high, 75% very high). T-MDS occurred in 5 pts (42%) and 7 pts (58%) were transfusion-dependent at baseline. Median BM blasts were 9% (4-30). Eleven of 12 pts (92%) had a TP53 missense mutation in the DNA binding domain with multiple mutations in 4/12 pts (33%). For 9/12 pts (75%), TP53 was the sole mutation. Median time on study is 176 days (41-298) with 7 pts ongoing. Treatment (Tx) related AEs during the lead-in phase (all G1) included nausea (n=5), neuropathy (n=5), decreased appetite (n=2), and dizziness (n=2) which were all transient. Tx related AEs occurring in > 1 pt in the combination phase included nausea/vomiting (n=6), dizziness (n=3), headache (n=3), neuropathy (n=3), fall (n=2), pruritus (n=2), thrombocytopenia (n=6), neutropenia (n=5), and leukopenia (n=4); all G1/G2 except cytopenias (G3/G4). No DLTs have occurred to date. Eleven of twelve pts were response evaluable with 1 pt discontinuing tx prior to 1st disease assessment (Fig 1A). ORR by IWG was 100% (11/11) with 9 CR (82%) and 2 marrow CR (mCR; 18%). Median time to first response was 70 days (4-91) and one CR patient achieved mCR and partial cytogenetic response after APR-246 lead-in prior to combination therapy. All CR pts had high p53 positivity by IHC at baseline (25-80%) which normalized on serial assessment with the 2mCR pts having <5% p53+ at baseline. Serial NGS with a variant allele frequency (VAF) cutoff of 5% was negative in 73% of patients (8/11). In NGS negative pts, MRD analysis, validated by ddPCR, was performed with a median VAF of 0.3% (0.1%-3.1%) at best molecular response. Enriched pathway analysis via Reactome following APR-246 lead-in phase showed transcriptional activation of p53 targets (FDR = 9.16E-09), including pathways involved in cell cycle arrest, apoptosis, DNA repair, and regulation of TP53 activity. At a median follow up of 7 months, the median OS or PFS has not been reached. In comparison to a internal historical cohort of 51 mTP53 MDS/AML treated with AZA alone, APR-246+AZA had a trend for improved OS (NR vs 7.6months; HR 0.30, P=0.07; Fig 1B). Conclusions: APR-246+AZA combination is well tolerated in mTP53 MDS/AML. Responses have been achieved in all evaluable pts (82% CR) accompanied by deep molecular and durable remissions. - In this paper we conduct a dynamic panel data analysis using 246 municipalities of the Marche region in order to investigate the presence of electoral budget cycle in local tax setting. Results show that local governments significantly decrease property tax rate during the election year. Although incumbent politicians can choose the tax rate in a range from 4% to 7%, results highlight that the range is not a hindrance to engaging electoral competition at the local government level.

Background: Prescribing patterns provides prescribing behaviors of prescriber. Hypertension is most common cardiac complication among middle and old age population. Thus, study about prescribing trends helps to select appropriate drug for treatment of hypertension. The study aim was to analyze the patterns of antihypertensive drug prescribed in patients diagnosed with hypertension.Methods: The study was cross sectional and observational study. A questionnaire was specifically designed factoring patients’ demographical profile, diagnosis of disease, drug regimen.Results: The totals of 100 patients were analyzed for the prescribing patterns of antihypertensive drug maximum patients belonged to the age group of 61-80. The proportion of male (62%) patients was more as compared to female patients (38%). Total drug prescribed was 246 in 100 prescriptions. Average drug per prescription was 2.46. Among 246 drugs, 97% were antihypertensive prescribed and 3% were other concomitant drugs. 65% single drug prescribed, 25% two drug prescribed and 10% three drug prescribed. Most commonly single antihypertensive drug prescribed was telmisartan. Most commonly two antihypertensive drug prescribed was telmisartan + hydrochlorothiazide. Most commonly three antihypertensive drug prescribed was telmisartan + amlodipine + hydrochlorothiazide. Among total 246 drug prescribed, angiotensin receptor blocker was prescribed (ARB) 28%, calcium channel blocker (CCB) 17%, angiotensin converting enzyme inhibitor (ACEI) 12%, beta blocker 5% followed by ARB + diuretics 13%, ACEI + diuretics 8%, beta blocker + CCB 4%, ARB + CCB + diuretics 7%, ARB + beta blocker + diuretics 3% and 3% other drug prescribed. There was no fixed dose combination prescribed and no injectable prescribed in present study.Conclusions: The most common drug prescribed was ARB as single drug therapy. The most common drug which was used for combination therapy was diuretics.

Topramezone is a newly introduced herbicide for use in field corn (Zea mays L.) that may have potential for weed management in sweet corn. Tolerance of eight sweet corn hybrids to topramezone applied postemergence (POST) at 0, 50, 75, 100, 150, and 300 g a.i. ha− 1 were studied at one Ontario location in 2000 and two locations in 2001 and 2002. Topramezone applied POST at 50, 75, 100, and 150 g·ha− 1 did not cause any visual injury in Calico Belle, CNS 710, Delmonte 2038, FTF 222, FTF 246, GH 2684, Reveille, and Rival sweet corn hybrids at 7 days after treatment (DAT) and caused minimal injury (less than 5%) at 300 g·ha− 1 in all hybrids. The initial sensitivity observed in these hybrids was minimal and transient with no effect on visual injury at 14 and 28 DAT. Topramezone applied POST did not reduce plant height, cob size, or marketable yield of the sweet corn hybrids included in this study. Based on these results, topramezone applied POST at the rates evaluated can be safely applied to Calico Belle, CNS 710, Delmonte 2038, FTF 222, FTF 246, GH 2684, Reveille, and Rival sweet corn.

Background: The objective was to study the hypotheses that an advanced zone II flexor tendon rehabilitation protocol would avoid rupture, achieve a high range of excursion, and minimize interphalangeal contracture during both the early phases and at the conclusion of healing. We also proposed the null hypothesis of no difference between any two of the zone II subdivisions. Methods: Fifty-one consecutive adult patients with zone II flexor tendon repairs of a single finger were retrospectively evaluated on an active contraction rehabilitation protocol with no splint, no tenodesis protection, and immediate full composite extension. There were 38 males and 13 females with a mean age of 39 years (range 18–69) involving 15 index, 7 long, 6 ring, and 23 small fingers. Repairs were located in flexor subzone IIA for 8 fingers; subzone IIB, 14; subzone IIC, 19; and subzone IID, 10. Differences in outcome between any two subzones were compared by T-test with p < 0.05. Results: Mean active arcs of motion in degrees at 3 weeks post repair were PIP 1-93; DIP 0-44; and total active motion (TAM) 221. At 6 weeks PIP 2–98; DIP 1–51; and TAM 236. At 10–12 weeks PIP 1–101; DIP 1–56; and TAM 246. Final TAM by flexor subzone IIA was 243; IIB, 251; IIC, 246; and IID, 246. There were no significant differences between any two subzones. Mean final DASH score was 5. There were no ruptures. Conclusions: The results support the hypotheses. Outcomes of the therapy protocol demonstrated the lack of interphalangeal joint flexion contractures, high range of total active motion achieved early and sustained, and no ruptures. No differences were identified between and two of the flexor subzones.

Introduction: Generalized pustular psoriasis (GPP) is a rare, neutrophilic, chronic skin disease in which an overactivation of inflammatory pathways causes widespread erythema and eruption of sterile pustules that may coalesce into ‘lakes of pus’. GPP flares are heterogeneous, and there is no standard clinical definition. GPP flares are painful; severe cases may require emergency treatment and can lead to life-threatening complications such as sepsis. The objective of this study was to characterize the frequency, number, and duration of flares in patients with GPP. Methods: We conducted a retrospective chart review including patients of all ages with a confirmed diagnosis of GPP after 2011 receiving care at 27 sites in France, Malaysia, and Tunisia. Results: Data from 175 patients were included (France: 58.9%, Malaysia: 31.4%, Tunisia: 9.7%). Mean (SD) follow-up was 5.0 (3.1) years. Flare-related care was the second most common reason for patient visits, accounting for 200 of 1378 follow-up visits after GPP diagnosis, surpassed only by routine follow-up. The mean (SD) annual number of flare episodes was 2.3 (2.0); the total number of flare episodes since the initial flare leading to diagnosis ranged from 0 to 9. Eighty-three (47.4%) patients experienced ≥1 flare episode after diagnosis, though individual country data suggested a higher flare burden in Tunisia (70.6%) and Malaysia (54.5%) versus France (39.8%). Twenty-eight patients (16.0%) had ≥2 flare episodes (60 visits); median (min, max) time between episodes was 11.6 (0.1, 52) months. The mean (SD) duration of flares was 34.3 (41.3) days during follow-up. Of 254 hospitalizations, 128 (50.4%) were flare-related inpatient admissions. Overall, 37.5% of GPP flares required inpatient care. The mean (SD) duration of flare-related admissions was almost twice as long as non-flare admissions at 19.2 (23.1) days versus 10.5 (8.1) days. Mean (SD) time between flare-related hospitalizations among 66 patients (128 hospitalizations) was 14.1 (17.1) months, with a median of 9.5 months indicating a skew distribution. Conclusion: Flare episodes are a heavy burden for patients with GPP, with flares recurring as often as once a year and lasting up to 2 weeks or more. Flares account for >50% of hospitalizations for patients with GPP and are an important cause of follow-up care visits. The frequency of flares varied between patients, with some individuals experiencing up to 10 episodes over the study period. Heterogeneity of flares and rarity of GPP make diagnosis and clinical study challenging, while the severity of acute episodes and the burden of flare reoccurrence demonstrate the need for more effective treatment options.

A Prática como Componente Curricular (PCC) com base na legislação dos últimos anos constituía-se uma carga horária específica na formação inicial de professores, que junto ao estágio supervisionado, representava um espaço e tempo para a aproximação e apropriação do fazer docente. Nessa perspectiva, esse estudo objetivou analisar as percepções sobre a PCC na licenciatura em Física. Para isso, fez-se uso da revisão integrativa, como um procedimento metodológico realizado em seis etapas sucessivas, tendo a partir da quarta etapa, respostas à pergunta norteadora. Os resultados trazem evidências importantes, com destaque ao fato de que a implementação da PCC sem fundamentação e discussão com os professores corroborou para a uma concepção e interpretação equivocada da legislação, ocasionando a falta de preparao dos professores para desenvolver a PCC. Conclui-se com o apontamento das lacunas de estudos, voltadas à: diferença entre PCC e estágio supervisionado; a PCC nas disciplinas de conteúdo da Física e a PCC na Resolução do Conselho Nacional de Educação/Conselho Pleno (CNE/CP) nº 2/2019, pelas quais, tem-se interesse na construção de uma discussão que possa corroborar à prática de ensino na referida licenciatura, de modo a reconhecer, conforme nos lembra Oliveira (2023) o caráter dialético das relações entre teoria e prática pedagógicas, o qual pressupõe a autonomia e a dependência simultâneas e recíprocas entre elas.

Abstract. Improved quantification techniques of natural sources are needed to explain variations in atmospheric methane. In polar regions, high uncertainties in current estimates of methane release from the seabed remain. We present unique 10- and 3-month time series of bottom water measurements of physical and chemical parameters from two autonomous ocean observatories deployed at separate intense seabed methane seep sites (91 and 246 m depth) offshore western Svalbard from 2015 to 2016. Results show high short-term (100–1000 nmol L−1 within hours) and seasonal variation, as well as higher (2–7 times) methane concentrations compared to previous measurements. Rapid variability is explained by uneven distribution of seepage and changing ocean current directions. No overt influence of tidal hydrostatic pressure or water temperature variations on methane concentration was observed, but an observed negative correlation with temperature at the 246 m site fits with hypothesized seasonal blocking of lateral methane pathways in the sediments. Negative correlation between bottom water methane concentration (and variability) and wind forcing, concomitant with signs of weaker water column stratification, indicates increased potential for methane release to the atmosphere in fall and winter. We present new information about short- and long-term methane variability and provide a preliminary constraint on the uncertainties that arise in methane inventory estimates from this variability.

e22224 Background: BC is the most common malignancy and the 1st cause of death by cancer in women in Tucuman, Argentina.The expression of hormone receptors and Her 2 neu in tumor tissue with other microscopic variables have shown to be indicators of prognosis and is also useful in therapeutic decisions.The molecular basis for treatment selection is actually the best way for a treatment “target” identification.The objective of this trial was to determine the patients profile in the population with BC under program (all pts studied under methodologic norms of a BC Detection Program) from Tucumán Argentina, defined by risk categories (high, intermediate or low) and endocrine response (high, incomplete or no response)according St Gallen 2007. Methods: The population included in the study were 246 pts with BC from public and private institutions in the province of Tucumán.HE routine biopsies were used for the assessment of histological type and subtype, tumor grade (according to Elston et al. and Nottinghan breast cancer grading method), axillary nodes status, tumor size, and the IHC staining for estrogen, progesterone receptors and Her2 neu.The results were studied grouping and evaluating the data for risk and endocrine responsiveness.The statistic applied was the exploratory analysis.IHC methods were validated with external and internal controls, in 7 cases Her2 was determined also by Fish. Results: 217/246 pts completed all parameters and were classified on risk categories: high (60 patients- 27,6%-); intermediate (156 patients-71,9%-) and low (1 patient-0,5%).No significant differences were found between the ages of groups with different levels of risk.The ER of the pts(n:242/246) showed highly endocrine responsive in 109/242 (45%), incompletely endocrine responsive in 49/242 (20,2%), endocrine no responsive 84/242 (34,7%). and association between endocrine levels of response and age range and tumor size (p <0001 chi-square test and p<0,017). Conclusions: These results are comparable with international data and allows us to know the patients profiles in our population under BC program. No significant financial relationships to disclose.

Abstract Abstract 1344 Poster Board I-366 Male recipients of female hematopoietic cell grafts, when compared with all other donor/recipient gender combinations, have an increased risk for both acute and chronic GVHD, but also have a significantly decreased risk of posttransplant relapse. F→M HCT is also characterized at the cellular level by donor (female) T cell responses against male-specific minor histocompatibility (H-Y) antigens, which can contribute to both graft-versus-host disease (GVHD) and graft-versus-leukemia (GVL) activity. SMCY is a Y-chromosome gene that has previously been shown to encode at least two distinct MHC class I-restricted H-Y antigens presented by HLA-A*0201 and HLA-B*0702, respectively. Also, association between CD8+ T cell responses specific for the SMCY311-319 FIDSYICQV epitope and GVHD or GVL has been reported. A CD8+ FIDSYICQV-specific T cell clone was also reported to induce histological signs of GVHD reaction in an in vitro skin-explant assay. To date, however, only two MHC class I-restricted, and no MHC class II-restricted, H-Y antigens encoded by SMCY have been characterized. Given the large size of the SMCY and the homologous SMCX proteins and the fact that they are only 85% identical at the amino acid sequence level, we hypothesized that SMCY encodes other MHC class I- and class II-restricted H-Y antigens, and that T cell responses against these epitopes may likewise contribute to GVHD and GVL activity after F→M HCT. Arrays of pentadecapeptides with eleven-residue overlap were designed to tile regions of the SMCY protein that are non-identical to the corresponding regions of its X chromosome-encoded homologue SMCX, and then used to generate SMCY-specific T cell lines recognizing novel SMCY-encoded MHC class I- and class II-restricted H-Y antigens. Peripheral blood mononuclear cells (PBMC) were obtained on posttransplant day +126 from a 46 year-old male patient with monosomy 7 AML who had received a hematopoietic cell graft from his MHC-identical sister, and were stimulated in vitro with dendritic cells derived from his pretransplant PBMC that had been pulsed with the SMCY pentadecapeptides. After three stimulations, a SMCY peptide-specific CD4+ T cell line as well as a SMCY311-319 (FIDSYICQV)-specific CD8+ T cell line were obtained. After cloning by limiting dilution, we further characterized the SMCY-specific CD4+ T cell clone, 13H3. The 13H3 T cell clone recognizes the SMCY232-246 15-mer peptide, ELKKLQIYGPGPKMM, presented by HLA-DRB1*1501, and has a CD3+, CD4+, CD8−, CD45RA−, CD45RO+ surface phenotype. The cytokine release profile of this clone when assessed with SMCY232-246-loaded donor-derived EBV-LCL, as measured by the Luminex assay, is characterized mainly by Th1 cytokines (IFN-g and IL-2), but the clone also produced low to moderate levels of the Th2 cytokines IL-4, IL-10, and TGF-β. A minigene encoding SMCY232-246 was recognized by the 13H3 clone in a HLA-DRB1*1501-dependent fashion when transfected into COS-7 cells, but a minigene encoding the homologous SMCX-derived ELKKLQIYGAGPKMM peptide was not recognized, demonstrating that the clone is SMCY-specific. The 13H3 clone recognized 3 of 5 HLA-DRB1*1501+ male primary leukemia cells, but did not recognize either of 2 HLA-DRB1*1501− male or either of 2 HLA-DRB1*1501+ female primary leukemia cells. These results suggest that CD4+ T cell responses against the SMCY232-246 epitope could potentially contribute to GVL activity after F→M HCT. A SMCY232-246/HLA-DRB1*1501 tetramer has been constructed which specifically marks the 13H3 T cell clone, and future studies will use this reagent to determine whether CD4+ T cells specific for this epitope can be detected directly ex vivo in posttransplant blood samples from HLA-DRB1*1501+ F→M HCT recipients. Introduction : TP53 mutated (TP53m) MDS and AML have very poor outcome irrespective of the treatment received, including 40% responses (20% CR) with azacitidine (AZA) with short response duration and a median overall survival (OS) of about 8 months (Bejar, Blood 2014). APR-246 is a prodrug spontaneously converted to methylene quinuclidinone (MQ), a Michael acceptor that binds covalently to cysteines in mutant p53, leading to protein reconformation that reactivates its pro apoptotic and cell cycle arrest functions. The combination of AZA and APR 246 showed promising results in a phase Ib study in TP53m MDS (Sallman, ASH 2018). We report interim results of a GFM phase 2 study of AZA+ APR-246 in TP53m MDS and AML, conducted in parallel with a similar US MDS consortium study. Patients and Methods : The study included hypomethylating agent (HMA) naïve and not previously allografted intermediate, high or very high IPSS-R TP53m MDS and AML adult patients. Patients received APR-246 4500 mg IV /d (6 hour infusions) (days 1-4) followed by AZA 75 mg/m²/d (days 4-10) in 28 day cycles. Response (primary endpoint, assessed by IWG 2006 for MDS and ELN criteria for AML) was evaluated after 3 and 6 cycles in the intent to treat (ITT) population, ie all patients who had received any protocol treatment, and in patients who had at least a blood and bone marrow evaluation after cycle 3 (evaluable population). Allo-SCT, when possible, was proposed after 3 to 6 cycles, and treatment with reduced APR 246 and AZA doses could be continued after allo-SCT. Results : 53 patients were enrolled between Sept 2018 and July 2019 in 7 GFM centers, with a median age of 73 years (range 44-87), and M/F: 28/25. 34 patients had MDS (including 74% very high IPSS-R) and 19 had AML. IPSS-R cytogenetic risk was very poor in 30/34 MDS, and unfavorable in 18/19 AML, complex in 89% of the patients. Median baseline mutated TP53 VAF was 21% (range 3-76). Nineteen of the 53 patients had been included at least 7 months before date of analysis (25 July 2019), had received protocol treatment and were thus potentially evaluable for response after 6 treatment cycles (ITT population). One of them died after only one cycle from an unrelated cause (cerebral ischemic stroke), and 2 during the third cycle (from bleeding and sepsis, respectively). In the remaining 16 patients (evaluable population per protocol), the response rate was 75% including 9 (56%) CR, 3 (19%) marrow CR or stable disease with hematological improvement (HI), and 4 treatment resistance. In the ITT population, the response rate was 63%, including 47% CR, and 16% stable or marrow CR+ HI. Among CR patients, complete cytogenetic CR and negative NGS for TP53 mutation (VAF cutoff of 2%) were achieved in 7/9 (78%) and 8/8 (100%), respectively. So far, 1 patient has undergone allo-SCT. All 53 patients had received at least one treatment cycle, and no increased myelosuppression, compared with AZA alone, was apparent. Treatment related AEs observed in ≥ 20% of patients were febrile neutropenia in 19 (36%) and neurological AEs in 21 (40%) of the patients. The latter, reviewed with a neurological team, were mainly grade 1 or 2 and consisted of ataxia (n=13), sometimes associated with cognitive impairment (n=4), suggesting a cerebellar origin. Other patients experienced acute confusion (n=4), isolated dizziness (n=3) and facial paresthesia (n=1). Neurological AEs reached grade III in 3 cases (1 acute confusion, 2 ataxia). Occurrence of neurological AEs was correlated with lower glomerular filtration rate at treatment onset (p&lt;0.01) and higher age (p=0.05). Neurological symptoms spontaneously regressed within 5 days of drug discontinuation (after a median of 1 day). They did not recur in the following cycles after per protocol APR 246 dose reductions. Conclusion : In this very high-risk elderly population of TP53m MDS and AML, generally with complex karyotype, a promising 56% CR rate at 6 cycles was reached in the evaluable population with AZA+ APR 246 combination, with deep molecular remission in all CR patients. We observed manageable neurologic AEs, mainly in elderly patients with reduced renal function, who therefore require close monitoring and dose reduction if necessary. An update regarding safety and efficacy in the 53 patients, including survival data, will be available at the meeting. Wrist actigraphy measures sleep activity and circadian rhythm. This study examined nighttime variability in Actiwatch parameters in a sample of breast cancer survivors (BCSs) to determine a minimum number of nights needed to obtain an accurate picture of objective sleep. A descriptive, quantitative, and repeated measures design was used. Consenting participants wore an actigraph and completed a sleep diary across 7 nights. There were no significant differences in wake after sleep onset (WASO), total sleep time (TST), sleep latency, or sleep disturbances across nights of week (Monday to Sunday) or monitoring nights (1st to 7th). Sleep efficiency was significantly better at Night 6 compared with Night 7. The coefficients of variation (CVs) for WASO ranged from 46% to 86%, TST 23%–34%, sleep latency 154%–246%, sleep efficiency 12%–22%, and sleep disturbances 33%–41%. Although the CVs indicated high variability across women, there was little internight variability in WASO or TST during across 7 nights of sleep. This suggests that in BCSs, Actiwatch data could be collected and evaluated from any single night for an accurate measure of usual sleep.

Tujuan penelitian ini yaitu untuk menemukan hubungan minat belajar dan motivasi belajar terhadap hasil belajar pada pendidikan jasmani di smk negeri 7 Pontianak. Metode dalam penelitian ini adalah korelasional, pengambilan data berupa kuesioner, dengan meteode penelitian kuantitatif. Populasi dalam penelitian ini adalah siswa SMK Negeri 7 Pontianak yang berjumlah 1138 siswa dengan sampel berjumlah 246 siswa kelas X. Teknik pengumpulan data yang digunakan yaitu angket dan nilai siswa. Hasil perhitungan data motivasi belajar pendidikan jasmani di SMK Negeri 7 Pontianak yang telah dihitung presentase mencapai 85,45% dengan kategori “Sangat Baik”, Data minat belajar pendidikan jasmani di SMK Negeri 7 Pontianak mencapai 87,03% dengan kategori “Sangat Baik”, dan data hasil belajar pendidikan jasmani mencapai 88,41% dengan kategori “Sangat Baik”. Dibuktikan dari hasil perhitungan menggunakan SPSS versi 25 dapat diketahui bahwa koefesien korelasi yang diperoleh sebesar 0,410. Sesuai dengan interprestasi koefesien korelasi nilai r, maka 0,410 berada pada skala 0,41 samapai 0.599 dengan tingkat hubungan “sedang”. Dibuktikan dari hasil perhitungan menggunakan SPSS versi 25 dapat diketahui bahwa koefesien korelasi yang diperoleh sebesar 0,420. Bovine CIDR were used for 11 d to synchronize estrus in 18 farmed muskoxen. Nine muskoxen went into harem for 1 wk (BRED) following CIDR removal and nine animals served as treated, non-mated controls (CTL). Progestin treatment prevented short estrous cycles (100% BRED; 88.9% CTL). The interval from CIDR removal to CL formation was 7.2 d (BRED) and 11.6 d (CTL) and fertility rate at the estrus following treatment was 88.9% (BRED). The use of bovine CIDR to synchronize estrus reduced the harem period to 1 wk and resulted in synchronized calving (7 d) following a mean gestation of 246 d. Zusammenfassung: Bei einer Stichprobe von 246 Personen, deren Anwerbung stratifiziert nach Geschlecht und Alter erfolgte, wurde der SVF78, eine Kurzvariante des SVF120 von Janke und Erdmann (1997) , hinsichtlich seiner Reliabilität und internen Validität überprüft. Die inneren Konsistenzen nach Cronbach und die Halbierungsreliabilitäten sind ebenso wie die von Janke und Erdmann (2002) in der Handanweisung zum SVF120 angegebenen gut bis sehr gut. Faktorenanalysen ergaben eine Faktorenstruktur, die der vergleichbar ist, die aus den Daten der Standardisierungstichprobe bei Verwendung des SVF120 errechnet wurde. Ebenso bildeten sich Geschlechts- und Altersunterschiede entsprechend denen der Standardversion ab. Die Untersuchungsergebnisse legen nahe, dass der SVF78 als zuverlässige Kurzvariante des SVF120 angesehen werden kann, die bei Außerachtlassung von 7 Stressverarbeitungsweisen die wesentlichen Stressverarbeitungsweisen des SVF120 erfasst.

To characterize autonomic influences on the Purkinje system in vivo, we measured Purkinje relative refractory period (PRRP) in response to sympathetic (SNS) and vagal nerve stimulation (VNS). Effects of SNS on PRRP were primarily mediated via beta-adrenergic mechanisms because shortening of PRRP during SNS [from 215 +/- 7 (SE) to 202 +/- 8 ms, P less than 0.01] was entirely blocked by metoprolol (1 mg/kg). Vagal influences in the ambient state did not prolong PRRP, even when effective refractory period of adjacent muscle did prolong. When VNS was augmented with physostigmine, PRRP prolonged from 205 +/- 12 to 212 +/- 13 ms, P less than 0.05. Similar provocation of parasympathetic effects on PRRP occurred when VNS was performed during SNS; PRRP prolonged from 188 +/- 9 to 193 +/- 9 ms, P less than 0.05. Also, when alpha-adrenergic stimulation was produced by phenylephrine infusion (25 micrograms.kg-1.min-1) in the presence of metoprolol (1 mg/kg), which prolonged PRRP from 242 +/- 8 to 246 +/- 9 ms, P less than 0.05, the addition of VNS further prolonged PRRP from 246 +/- 9 to 253 +/- 9 ms, P less than 0.05. Thus some refractory period responses in the Purkinje system were similar to adjacent muscle, because beta-adrenergic activation shortened refractory period and vagal stimulation antagonized the shortening. Findings unique to Purkinje tissue were refractory period prolongation by vagal stimulation when facilitated by concurrent prolongation of refractory period during alpha-adrenergic stimulation.

Bambara groundnut is a neglected legume crop that contributes to improve global food and face food insecurity. In Burkina Faso, research on Bambara groundnut cropping system were essentially focused on mounding times. Less attention has been paid on the application of fertilizers. Experiments was carried out on the site of Tenkodogo University Centre (11°48’37’’N, 0°22’19’’W), Burkina Faso to study the response of two Bambara groundnut varieties to different NPK application periods. The experiments consisted of a factorial combination of two factors, including the varieties (KVS 235 and KVS 246) and the different NPK application dates (At basal, 7, 15 and 21 days after sowing) in completely randomised blocks with three replications. Characters were collected, including plant height and spread, total biomass at flowering, stalks weight, yield-components and yield. The results indicated that most of characters not varied significantly for both varieties whatever NPK application period except for stalks weight (P = 0.013). The interactions between varieties and NPK application dates had significant effects for only total biomass at flowering (P = 0.048). In addition, pod length has significant effect (P = 0.032) between varieties. The significant effect on the characters mentioned above in both varieties implies that application of NPK fertilizer have influenced the characteristics differently, depending on the period of bringing. NPK application at basal lead to best performance of the results regarding pod and seed dimension and 100-seeds weight and yield. In general, KVS235 outperformed than KVS 246.
