Abstract Accumulating evidence suggest that miRNAs play important roles in immune system. At present study, we first found that miRNA expression profiles were significantly different among vary immunocytes by microarray. Here, We focused on miR-128-2 which was highly expressed in DP T cells, but lower in other detected cells. In order to further study the function of miR-128-2 in lymphocytes development, we established the miR-128-2 chimeric and transgenic mice, confirmed by FACS analysis and PCR. We found B cell development appeared profoundly defects in both mice models, however, the development of T cells had no significant changes after over-expressing mir-128-2. Further analysis suggest that CLP was significantly higher in mir-128-2 TG mice compared to WT mice, but HSC and MMP cells were comparable in mir-128-2 TG mice and WT mice. Brdu and annexinV staining showed that over-expressing mir-128-2 did not affect the proliferation and apoptosis of CLP. These suggested that miR-128-2 inhibited the CLP cells to develop into B cell. Finally, we try to seek for the targets of miR-128-2 through software analysis and molecular biology methods. We found that miR-128-2 might down-regulate a group of genes including BCL11a, MALT1, A2b et. al. to inhibit B cell development confirmed by luciferase assay, real time PCR and western blot. Thus, our present study suggests that miR-128-2 mainly blocks CLP to differentiate into PreProB cells by down-regulating the BCL11a, MALT, A2b et al. genes.

Abstract Cefoperazone/sulbactam (CSL) and piperacillin/tazobactam (TZP) are commonly used in clinical practice in China because of their excellent antimicrobial activity. CSL and TZP-nonsusceptible Enterobacteriaceae are typically resistant to extended-spectrum cephalosporins such as ceftriaxone (CRO). However, 11 nonrepetitive Enterobacteriaceae strains, which were resistant to CSL and TZP yet susceptible to CRO, were collected from January to December 2020. Antibiotic susceptibility tests and whole-genome sequencing were conducted to elucidate the mechanism for this rare phenotype. Antibiotic susceptibility tests showed that all isolates were amoxicillin/clavulanic-acid resistant and sensitive to ceftazidime, cefepime, cefepime/tazobactam, cefepime/zidebactam, ceftazidime/avibactam, and ceftolozane/tazobactam. Whole-genome sequencing revealed three of seven Klebsiella pneumoniae strains harbored bla SHV-1 only, and four harbored bla SHV-1 and bla TEM-1B. Two Escherichia coli strains carried bla TEM-1B only, while two Klebsiella oxytoca isolates harbored bla OXY-1-3 and bla OXY-1-1, respectively. No mutation in the β-lactamase gene and promoter sequence was found. Outer membrane protein (Omp) gene detection revealed that numerous missense mutations of OmpK36 and OmpK37 were found in all strains of K. pneumoniae. Numerous missense mutations of OmpK36 and OmpK35 and OmpK37 deficiency were found in one K. oxytoca strain, and no OmpK gene was found in the other. No Omp mutations were found in E. coli isolates. These results indicated that narrow spectrum β-lactamases, TEM-1, SHV-1, and OXY-1, alone or in combination with Omp mutation, contributed to the resistance to CSL and TZP in CRO-susceptible Enterobacteriaceae. Abstract Kiasu-BC and Joltik-BC are internal tweakable block ciphers of authenticated encryption algorithms Kiasu and Joltik submitted to the CAESAR competition. Kiasu-BC is a 128-bit block cipher, of which tweak and key sizes are 64 and 128 bits, respectively. Joltik-BC-128 is a 64-bit lightweight block cipher supporting 128 bits tweakey. Its designers recommended the key and tweak sizes are both 64 bits. In this paper, we propose improved meet-in-the-middle attacks on 8-round Kiasu-BC, 9-round and 10-round Joltik-BC-128 by exploiting properties of their structures and using precomputation tables and the differential enumeration. For Kiasu-BC, we build a 5-round distinguisher to attack 8-round Kiasu-BC with $2^{109}$ plaintext–tweaks, $2^{112.8}$ encrytions and $2^{92.91}$ blocks. Compared with previously best known cryptanalytic results on 8-round Kiasu-BC under chosen plaintext attacks, the data and time complexities are reduced by $2^{7}$ and $2^{3.2}$ times, respectively. For the recommended version of Joltik-BC-128, we construct a 6-round distinguisher to attack 9-round Joltik-BC-128 with $2^{53}$ plaintext–tweaks, $2^{56.6}$ encryptions and $2^{52.91}$ blocks, respectively. Compared with previously best known results, the data and time complexities are reduced by $2^7$ and $2^{5.1}$ times, respectively. In addition, we present a 6.5-round distinguisher to attack 10-round Joltik-BC-128 with $2^{53}$ plaintext–tweaks, $2^{101.4}$ encryptions and $2^{76.91}$ blocks.

Lactiplantibacillus plantarum could regulate certain physiological functions through the AI-2/LuxS-mediated quorum sensing (QS) system. To explore the regulation mechanism on the growth characteristics and bacteriostatic ability of L. plantarum SS-128, a luxS mutant was constructed by a two-step homologous recombination. Compared with ΔluxS/SS-128, the metabolites of SS-128 had stronger bacteriostatic ability. The combined analysis of transcriptomics and metabolomics data showed that SS-128 exhibited higher pyruvate metabolic efficiency and energy input, followed by higher LDH level and metabolite overflow compared to ΔluxS/SS-128, resulting in stronger bacteriostatic ability. The absence of luxS induces a regulatory pathway that burdens the cysteine cycle by quantitatively drawing off central metabolic intermediaries. To accommodate this mutations, ΔluxS/SS-128 exhibited lower metabolite overflow and abnormal proliferation. These results demonstrate that the growth characteristic and metabolism of L. plantarum SS-128 are mediated by the AI-2/LuxS QS system, which is a positive regulator involved in food safety. It would be helpful to investigate more bio-preservation control potential of L. plantarum, especially when applied in food industrial biotechnology.

Retarding the protein deterioration of shrimp during storage is important for maintaining its quality. Lactobacillus plantarum SS-128 (L. plantarum SS-128) is a biocontrol bacterium that can effectively maintain the fresh quality of food. This research establishes a myofibril simulation system and refrigerated control system to explore the impact of L. plantarum SS-128 on the quality and shelf life of refrigerated shrimp (Litopenaeus vannamei). Through the bacterial growth assay and AI-2 signal molecule measurement, the effect of the AI-2/LuxS quorum sensing (QS) system of L. plantarum SS-128 and shrimp spoilage bacteria was established. In the myofibril simulation system, a study on protein degradation (dimer tyrosine content, protein solubility, sulfhydryl content, and carbonyl content) showed that adding L. plantarum SS-128 effectively slowed protein degradation by inhibiting the growth of food pathogens. The application to refrigerated shrimp indicated that the total volatile basic nitrogen (TVB-N) value increased more slowly in the group with added L. plantarum SS-128, representing better quality. The total viable count (TVC) and pH results exhibited similar trends. This study provides theoretical support for the application of L. plantarum SS-128 in storing aquatic products.

Paracoccidioides brasiliensis is a thermo-dimorphic human pathogenic fungus that lives at 23”C in the mycelium phase (infecting phase) and at 37”C in the yeast phase (parasite phase). In attempt to survive, the cell wall of fungi can change its composition and/or structure in response to environmental stress by compensatory mechanisms. The molecules involved in these mechanisms are possible target for the development of effective antifungal agents. In P. brasiliensis, the main components of the cell wall are glucans and chitin polymers. These polymers make a primary barrier that is responsible for the structural integrity and form of the cell wall. In this work the behavior of P. brasiliensis was evaluated against stress conditions with the aim of study, for the first time, the mechanisms used by this fungus in the maintenance of the cell wall integrity. Our results shown that P. brasiliensis yeast cells are sensitive to cell wall stressors calcofluor white (CFW), congo red (CR), SDS, KCl, NaCl and sorbitol. There was an increase in the PbFKS1 transcripts expression and in the content of cell wall β- 1,3-glicana after treatment with all stressor agents. After treatment with SDS and KCl the PbGFA1 transcripts expression and the cell wall GlcNAc residues also increased. The transcript expression of PbGEL3 was also evaluated being increased after treatment with CFW, NaCl and sorbitol. Thus we showed that these molecules are involved in the maintenance of the cell wall against stress conditions. Apart from these analyses we obtained the active recombinant protein PbFks1pc. Through the anti-PbFks1pc antibody we performed immunocitolocalization assays. These experiments revealed the localization of PbFks1p in regions of apical growth in the mycelium phase and in the cell surface in yeast phase.

In this thesis we will discuss the privacy issues in the RFID system. In the past any RFID reader can read from and write to the tags within the valid range in the neighborhood. When the articles are embedded with tags, they can be traced and monitored. Therefore it demonstrates the security holes as regards to information safety used. It shows that all information of articles after being attached with tags can be obtained without needing the permission from tag users. In general there two kinds of methods that provide the data security of RFID tags. One of the methods is to encrypt the data of tags. In the processes the data can be protected by substituting the plain-edited text with the cipher-edited text. Any data obtained by unauthorized readers will be not readable unless the data obtained can be decrypted. The other method is to set up confirmation between tags and readers. It means that if the data has already been confirmed before, the readers and tags will be mutually ascertained the right party. Therefore the readers can be allowed to obtain the data of tags and continue to carry out operations within the authorized ranges. The research in this thesis mainly uses HMAC inlayed with MD5 hash functions to prevent the unauthorized readers from invading the data information in the tags. The hatched method is applied to the passive RFID of EPC-128 Class Therefore it will not take too long read time that EPC-128 tags are encrypted.

Spatial light modulators (SLMs) are the key component for real time electro-optical signal processing, computing, holographic optical memory storage, free space optical interconnect, information processing, and data conversion systems. We have used our SLM technology to develop several optical processing and data routing demonstration units including beam steering units and optical correlators. The later have been created with maximum speed of 600,000 128 x 128 2-D correlations per second.

The Ar2 excimer band at 128 nm has generally been thought to arise from excimers in the closely spaced A 3 Σ u + ( 1 u ) and B 1 Σ u + ( 0 u + ) states. We have investigated this designation spectroscopically by irradiating the dc discharge plasma of Ar2 after supersonic expansion with the beam of a tunable titanium–sapphire laser to probe the 3p54s manifold of atomic Ar and simultaneously monitoring the effects on atomic transitions as well as on the 128 nm band of Ar2. Intensity increases and decreases of up to 5% were measured in the 128 nm band, depending on the wavelength of the incident radiation. The analysis of these results indicates that under the present discharge conditions, the 128 nm band is primarily due to excimers in the A 3 Σ u + ( 1 u ) state, formed from atoms in the 4s[3/2]2 level. It is also shown that the observed FWHM bandwidth of 8.7 nm can be explained by significant vibrational cooling of the excimer after expansion, with ≈80% population in the v = 0 level. Thus, when the laser was tuned to probe an absorption band of Ar2 at 985 nm, and a decrease in intensity of the 128 nm band was recorded having the shape and bandwidth of the 128 nm band, this was proof that the 985 nm band originates from the A 3 Σ u + ( 1 u ) ( v = 0 ) state.

Liquid Crystal on Silicon (LCOS) spatial light modulators (SLM’s) and microdisplays are made by placing a thin layer of liquid crystal directly on top of a silicon chip. We have constructed a high speed 256 by 256 binary SLM which uses a ferroelectric liquid crystal as the light modulating layer [1]. We have also recently constructed a high speed distorted helix effect analog 128 by 128 SLM [2] and a high voltage 128 by 128 analog SLM using the electroclinic effect in BDH764E [3]. This work has been motivated by the performance requirements of coherent applications which typically include high frame rate, small pixel pitch and good diffraction efficiency into a single (zeroth) diffracted order. These devices have been sucessfully used in optical correlators and holographic interconnect systems [4].

A numerical, two-dimensional hydrodynamic model of the Mississippi River, from Thebes, IL, to Tiptonville, TN (128 miles/206 km), was developed using the Adaptive Hydraulics model. The study objective assessed current patterns and flow distributions and their possible impacts on navigation due to Birds Point New Madrid Floodway (BPNMF) operations and the Len Small (LS) levee break. The model was calibrated to stage, discharge, and velocity data for the 2011, 2015–2016, and 2017 floods. The calibrated model was used to run four scenarios, with the BPNMF and the LS breach alternately active/open and inactive/closed. Effects from the LS breach being open are increased river velocities upstream of the breach, decreased velocities from the breach to Thompson Landing, no effects on velocity below the confluence, and cross-current velocities greater than 3.28 ft/s (1.0 m/s) within 1186.8 ft (60 m) of the bankline revetment. Effects from BPNMF operation are increased river velocities above the confluence, decreased velocities from the BPNMF upper inflow crevasse (Upper Fuseplug) to New Madrid, cross-current velocities greater than 1.5 ft/s (0.5 m/s) only near the right bank where flow re-enters the river from the BPNMF lower inflow/outflow crevasse Number 2 (Lower Fuseplug) and St. Johns Bayou.

Rocky Flats National Wildlife Refuge (“the Refuge”) in Colorado near Denver, Colorado, has a history (1952-1 989) of producing components for nuclear weapons. The current goal for the area is “to restore and preserve the native prairie ecosystems, provide habitat for migratory and resident wildlife, conserve and protect habitat for Preble’s meadow jumping mouse, and provide research and education opportunities”. The grasslands of the Refuge are surrounded by busy roads to the west (Hwy 93, 18,000 AADT), north (Hwy 128, 4,200 AADT) and east (Indiana St. 7,000 AADT), and there are houses and associated roads on its southern boundary. Other open space with non-motorized trails and protected areas with predominantly grassland are to the west, north and east. Large ungulates, including mule deer, elk, and moose cross the roads. This results in large ungulate -vehicle collisions and the roads also represent a barrier to the movements of animals. Creek crossings under the roads are a concern as they are likely a barrier for species dependent on riparian habitat, including the Preble’s meadow jumping mouse. The objectives of the current project were to 1. Formulate measures that reduce collisions with large wild mammals, and 2. Formulate measures that improve connectivity across roads for large wild mammal species and one small mammal species in specific, the Preble’s meadow jumping mouse. We suggest large open span bridges at creek crossings (for deer, moose, black bear, mountain lion, and Preble’s meadow jumping mouse) and designated wildlife overpasses for elk and also f or mule deer. The crossing structures may be combined with human co-use to connect the trails on the refuge with the trail system in the surrounding areas.

Researchers conducted eight large-scale laboratory tests to assess the combined impact of hot-mix asphalt (HMA) overlay mix and thickness on its performance to control reflective cracking. Bonding efficiency, flexibility, and stiffness of the HMA mix as well as overlay thickness significantly affect an overlay’s performance against reflective cracking. Researchers developed a generalized 3D finite-element model to predict an overlay’s reflective cracking potential and generated a database of 128 cases. They also developed a data-driven surrogate model to predict reflective cracking potential that engineers can easily use. Life-cycle cost analysis of overlay alternatives was performed using Illinois Department of Transportation’s unit prices from contracts between 2018 and 2019. The researchers identified optimal overlay configurations to control reflective cracking. An overlay composed of a 1.5 in (38.1 mm) SMA-9.5 or 1.25 in (31.8 mm) IL-9.5FG surface course and a 0.75 in (19.1 mm) IL-4.75 binder course had the lowest annual cost per mile among non-interstate projects. For interstate projects, an overlay composed of a 2 in (50.8 mm) SMA-12.5 surface course and a 2.25 in (57.2 mm) IL-19.0 binder course was the most cost-effective. The study concluded that to control reflective cracking and to reduce life-cycle cost, an overlay composed of an SMA-9.5 surface course and an IL-4.75 binder course is recommended for non-interstate projects. An IL-9.5FG surface course and an IL-4.75 binder course are suggested for low-volume and low-speed roads. For interstate projects, an overlay comprised of an SMA-12.5 surface course and an IL-19.0 binder course is recommended. A data-driven surrogate model may be used to design overlay thicknesses.
