The structure of butorphanol hydrogen tartrate {(9R,13S,14S)-(-)-17-(cyclobutylmethyl)morphinan-3,14-diol (2S,3S)-(-)-hydrogen tartrate} (C21H29NO2 .C4H6O6) was solved by direct methods and refined anisotropically to the R value of 0.029 for 2 069 observed reflections. The title morphine analogue crystallizes in the triclinic space group P1 with lattice parameters a = 7.620(1), b = 9.140(1), c = 9.591(1) Å, α = 105.48(1), β = 112.91(1), γ = 84.29(1)°, Z = 1. The butorphanol B ring possesses the 3E envelope conformation with small 3H2 distortion, C and D rings have a regular chair conformation. The intramolecular N17-H17...O2 hydrogen bond is observed in crystal structure of the title compound. The butorphanol and hydrogen tartrate molecules are joined together by means of O2-H(O2)...O7, O1-H(O1)...O7' and O3-H(O3)...O8" hydrogen bonds to form networks.

ABSTRACT In fresh-cut produce production, antimicrobials may be used during washing to control the risk of cross-contamination by microbial hazards. Surrogate microorganisms have long been used to validate processes, but none have been identified for validating the efficacy of antimicrobial washing of fresh-cut produce. The objective of this study was to develop procedures by which surrogates may be identified for use in validating the control of cross-contamination for fresh-cut lettuce operations. Four microbial characteristics, which may be important factors in cross-contamination events, were quantitatively evaluated in potential surrogate microorganisms for comparison to a reasonably foreseeable hazard, Escherichia coli O157:H7: sensitivity to chlorine in solution, sensitivity to chlorine on lettuce leaf surfaces, shedding from contaminated lettuce leaves into the water during washing, and cross-contamination from inoculated to uninoculated lettuce leaves during chorine washing. A procedure of practical quantitative experiments for comparing the characteristics reduced the original pool of 80 potential strains, which consisted of lactic acid bacteria, probiotics, and isolates obtained from lettuce enrichment cultures, to five strains: Lactobacillus plantarum, Pediococcus pentosaceus, probiotic 22C, and two lettuce enrichment isolates. These strains may be evaluated in additional studies involving comparisons to other reasonably foreseeable hazards and including other potential process variables that should be understood and controlled to prevent cross-contamination in fresh-cut lettuce operations.

It has been discussed previously that older adults’ living arrangements are associated with mortality. This study investigated the relationships between older adults’ living arrangements and sleep-related outcomes in China. The nationally representative sample included 4,731 participants who participated on two different occasions, with a total of 9,462 observations (2012 and 2014 waves). Panel logistic regression and panel ordinary least squares regression models were estimated with outcomes of sleep quality and average hours of sleep daily, respectively. Approximately 62% of individuals reported good quality of sleep. We observed that older adults who lived with family members had 17% greater odds of reporting good quality of sleep (adjusted odds ratio = 1.17, 95% confidence interval [1.03, 1.34], p < .05) and reported longer sleep duration daily (β = .334, standard error = .069, p < .01), compared with those who lived alone. Social support is needed to strengthen the residential relationship, especially with family members.

ABSTRACTObjective: identify the major stressors related to psychoactive substances shown in nurses from units opened and closed at a university hospital in the city of Recife-PE. Method: the study population was composed of 44 nurses who work in open and closed units of the hospital. The analysis of data was composed of two parts: characterization of the population with 17 questions and sources of stress factors and with 49 questions grouped into five categories (conflict of duties, on the workload, interpersonal relationships, management of personnel and critical situations). This project was approved by the Ethics Committee on Human Research of Universidade Federal de Pernambuco, Protocol No. 069/2008. Results: the open sector showed the highest score of stress. With the data we trace the socio-demographic profiles and occupational sample. The stress on the activity of nurses is related to several variables and the individual. Conclusion: according to these findings there is a need to discuss the working conditions of nurses, regardless of their area of operation because the successful delivery of patient care and family is with professionals who are ready globally, ie in sense of knowledge, emotions and actions with structure adequada. AbstractObjectiveThe study evaluated the association between consumption frequencies of the major food categories and the risk of new depression four years later in older Taiwanese.DesignA prospective cohort study with multistage random sampling. Logistic regression analysis evaluated the significance of the longitudinal associations of intake frequencies of the major food categories with future (4 years later) risk of new depression, controlled for possible confounding factors with or without adjustment for cognitive status.SettingPopulation-based free-living elderly.SubjectsMen and women (n 1609) ≥65 years of age.ResultsIn a regression model that controlled for demographic, socio-economic, lifestyle and disease/health-related variables but not cognitive status, both fruits (OR = 0·66, 95 % CI 0·45, 0·98, P = 0·038) and vegetables (OR = 0·38, 95 % CI 0·17, 0·86, P = 0·021) were protective against depressive symptoms 4 years later. However, when the same regression model was also adjusted for cognitive status, only vegetables (OR = 0·40, 95 % CI 0·17, 0·95, P = 0·039) were protective against depressive symptoms. Higher consumption of eggs was close to being significant in both regression models (P = 0·087 and 0·069, respectively). Other food categories including meat/poultry, fish, seafood, dairy, legumes, grains and tea showed no significant associations.ConclusionsResults suggest that although confounding factors cannot be totally ruled out, more frequent consumption of vegetables seems to be protective against depressive symptoms in the elderly. Further studies are needed to elucidate the causal role and the mechanism of the association.

The purpose of this study was to investigate the personality characteristics in adolescents with SLE. The research design is a case-control study by means of the Rorschach Method and the Wechsler Intelligence Scale. Study group: 30 female adolescents with lupus, 12–17 years of age. The SLE Disease Activity Index was administered during the period of psychological evaluation. Control group: 32 nonpatient adolescents were matched for age, sex, and socioeconomic level. In the Wechsler Intelligence Scale the mean IQ of the experimental group was significantly lower than that of the control group (77 and 98, respectively, p < .001). In the Rorschach, the lupus patients showed greater difficulty in interpersonal interactions, although they displayed the resources to process affect and to cope with stressful situations. A positive moderate correlation (p = .069) between the activity index of the disease and the affect constriction proportion of the Rorschach was observed: the higher the SLEDAI score, the lower the capacity to process affect. There is a negative correlation between the activity index of the disease and the IQ (p = .001): with a higher activity index of the disease, less intellectual resources are available.

AbstractMesaite (IMA2015-069), ideally (V2O7)3·12H2O, is a new mineral from the Packrat mine, Gateway district, Mesa County, Colorado, USA. Crystals of mesaite occur as orangish red blades up to 0.1 mm long and ∼10 μm thick. The streak is light pinkish orange and the lustre is vitreous, transparent. Mesaite has a brittle tenacity, {010} perfect cleavage; fracture is irregular, and no parting was observed. The mineral has a Mohs hardness ≈ 2. The measured density of mesaite is 2.74(1) g cm–3. Mesaite is biaxial (–), α = 1.760(calc), β = 1.780(5), γ = 1.795(5) in white light; the measured 2V value = 81(2)°. Dispersion is strong, r < v, and pleochroism is present in shades of brownish orange. Mesaite is monoclinic, P2/n, with a = 9.146(2), b = 10.424(3), c = 15.532(4) Å, β = 102.653(7)° and V = 1444.7(6) Å3. The strongest four diffraction lines in the powder diffraction pattern are [(dobs in Å, (Iobs), (hkl)]: 10.47 (100) (010), 2.881 (25) (132, 3̄12, 033, 310), 3.568 (24) (1̄14, 1̄23, 2̄13), 3.067 (17) (1̄24, 1̄32, 2̄23). The composition of mesaite was determined by electron microprobe, and yielded an empirical formula of Mn5.32Ca0.56Zn0.31V5.96As0.04O33H23.61 on the basis of 33 O atoms per formula unit (apfu).The atomic arrangement of mesaite was solved and refined to R1 = 0.0600. The structure is formed of zigzag octahedral chains of edge-sharing Mn2+O6 octahedra. Oxygen atoms of the octahedra are shared with V2O7 groups, which link with adjacent octahedral chains to form {010} heteropolyhedral layers. The interlayer region contains Ca atoms and H2O groups. Each Ca bonds to two O6 atoms in the heteropolyhedral layer and to two fully occupied and six partially occupied O (H2O) sites in the interlayer, resulting in an effective Ca coordination of approximately seven. Similar zigzag chains of edge-sharing MnO6 octahedra decorated with V2O7 groups are also found in the mineral fianelite. Mesaite has beenapproved by the Commission on New Minerals, Nomenclature and Classification of the International Mineralogical Association (IMA2015-069). The name mesaite is conferred for Mesa County, Colorado, USA.

Abstract. The aim of the study was to characterize the population of the German Sport Horse, a sub-population of the German Riding Horse, in terms of population structure and genetic parameters. Data from 26 490 horses with information from foal inspections (FI; n=17 881), broodmare inspections (BI; n=12 069) and mare performance tests (MPT; n=2 756) from the years 1990-2006 were used for analysis. A total of 21 traits as scored in the three types of performance tests were analysed. Heritability estimates for traits from FIs ranged from 0.24 ± 0.01 (conformation) to 0.50 ± 0.01 (type). For BIs estimates ranged between 0.15 ± 0.02 (forelegs) and 0.50 ± 0.02 (head). Traits evaluated in the MPT showed heritabilities between 0.18 ± 0.02 (rideability) and 0.46 ± 0.02 (trot). Genetic correlations within and across the three tests were all positive. An analysis of the structure of genetic relationships revealed that the population is influenced by different German Riding Horse sub-populations as well as by thor ough breds. The genetic connectedness within the population is of a complex structure and appeared to be sufficient for genetic analyses. Differences between the genetic pa rame ters estimated in this study for the GSH and those used in the national breeding value esti ma tion system are predominantly insignificant. These results confirm that the GSH is both adequately represented in the national system and that the population itself is a rep re sen tative part of the entire German Riding Horse population.

As there are not enough measurement instruments to assess emotions during epistemic activities, this research aimed to evaluate the psychometric properties of the epistemic emotions scale among speakers of the Persian language. The scale has previously been used and studied in different cultural contexts. The present study sought to expand the application of this scale in the Iranian context. A total of 1153 students (58.6% women) within the age range of 17 to 29 years participated in this study. The mean and standard deviation of the age of the participants were 21.63 and 2.24, respectively. The data collection tools in this study were the Epistemic Emotions Scale, the Achievement Emotions Questionnaire, the Academic Buoyancy Scale, and the Academic Satisfaction Scale. CFA was performed to test the factorial structure (?2 = 1895.96, df = 394, GFI = .86, CFI = .97, IFI = .97, NFI = .97, NNFI = .96, SRMR = .069, and RMSEA = .086) and its validity and reliability were also evaluated. The seven-factor model of the Epistemic Emotions Scale was the same as that of the original scale version, and the Epistemic Emotions Scale showed good internal consistency with Cronbach's alpha for this scale ranging from .85 to .88.. The results showed that the EES scores also correlated in expected directions with measures of related constructs included in this study thus also supporting the external validity of this version of the scale. We can conclude that the Epistemic Emotions Scale is a valid and reliable tool for measuring students' epistemic emotions, and as a result, researchers can use this tool for research purposes.

ObjectivesThis study examined predictors of lung cancer mortality, beyond age and smoking, among construction workers employed at US Department of Energy (DOE) sites to better define eligibility for low-dose CT (LDCT) lung cancer screening.MethodsPredictive models were based on 17 069 workers and 352 lung cancer deaths. Risk factors included age, gender, race/ethnicity, cigarette smoking, years of trade or DOE work, body mass index (BMI), chest X-ray results, spirometry results, respiratory symptoms, beryllium sensitisation and personal history of cancer. Competing risk Cox models were used to obtain HRs and to predict 5-year risks.ResultsFactors beyond age and smoking included in the final predictive model were chest X-ray changes, abnormal lung function, chronic obstructive pulmonary disease (COPD), respiratory symptoms, BMI, personal history of cancer and having worked 5 or more years at a DOE site or in construction. Risk-based LDCT eligibility demonstrated improved sensitivity, specificity and positive predictive value compared with current US Preventive Services Task Force guidelines. The risk of lung cancer death from 5 years of work in the construction industry or at a DOE site was comparable with the risk from a personal cancer history, a family history of cancer or a diagnosis of COPD. LDCT eligibility criteria used for DOE construction workers, which includes factors beyond age and smoking, identified 86% of participants who eventually would die from lung cancer compared with 51% based on age and smoking alone.ConclusionsResults support inclusion of risk from occupational exposures and non-malignant respiratory clinical findings in LDCT clinical guidelines.

Abstract Abstract 5042 Bortezomib is a proteasome inhibitor with potent antimyeloma activity in relapsed/refractory multiple myeloma (MM) patients. We evaluated the type and factors affecting the onset of infectious complications and mortality owing to infection in MM patients treated with bortezomib-based regimens. We reviewed 139 patients with MM treated with regimens containing bortezomib in order to assess the type and factors affecting the development of severe infections. Infections occurred in 56 (40. 3%) of 139 patients and 83 (7. 8%) cases of the 1, 069 evaluable cycles. Severe infections developed in 43 (30. 9%) patients and ten patients (7. 1%) died during bortezomib-based treatment. Multivariate analysis determined lymphocytopenia grade 3–4 (OR 3. 17, 95% CI 1. 38–7. 31, p = 0. 007) and number of cycle ° Â 8 cycles (OR 3. 91, 95% CI 1. 39–11. 02, p =0. 010) as risk factors associated with increased severe infection. This study showed that MM patients who received bortezomib-based regimens are at a higher risk of severe infections within eight cycles of treatment during especially severe lymphocytopenic period. AbstractObjectiveConsuming a variety of fruit and vegetables provides many different micronutrients and bioactive compounds. Whether this contributes to the beneficial association between fruit and vegetables and incident CHD and stroke is unknown.DesignProspective population-based cohort study.SettingThe Netherlands.SubjectsMen and women (n 20 069) aged 20–65 years. Participants completed a validated 178-item FFQ, including nine fruit and thirteen vegetable items. Variety in fruit and vegetables was defined as the sum of different items consumed at least once per 2 weeks over the previous year. Hazard ratios (HR) for variety in relation to incident CHD and stroke were calculated using multivariable Cox proportional hazards models additionally adjusted for quantity of fruit and vegetables.ResultsVariety and quantity in fruit and vegetables were highly correlated (r = 0·81). Variety was not associated with total energy intake (r = −0·01) and positively associated with nutrient intakes, particularly vitamin C (r = 0·70). During 10 years of follow-up, 245 cases of CHD and 233 cases of stroke occurred. Variety in vegetables (HR per 2 items = 1·05; 95 % CI 0·94, 1·17) and in fruit (HR per 2 items = 1·00; 95 % CI 0·87, 1·15) were not related to incident CHD. Variety in vegetables (HR per 2 items = 0·93; 95 % CI 0·83, 1·04) and in fruit (HR per 2 items = 1·03; 95 % CI 0·89, 1·18) were also not related to incident stroke.ConclusionsMore variety in fruit and vegetable consumption was associated with higher intakes of fruit and vegetables and micronutrients. Independently of quantity, variety in fruit and vegetables was related neither to incident CHD nor to incident stroke.

We aimed to identify psychopathological, anthropometric and sociodemographic factors that may influence the risk of low MD adherence.DesignLongitudinal study in three phases. MD adherence was assessed using the Krece Plus food questionnaire and psychopathological symptoms using the Screen for Childhood Anxiety Related Emotional Disorders, Children’s Depression Inventory, Youth’s Inventory-4 and Eating Disorder Inventory-2. Anthropometric data were collected in the first and third phases.SettingsAll five representative areas in Reus, Spain.SubjectsAdolescents (n241).ResultsRegardless of past and current BMI, socio-economic status was a protective factor for low MD adherence (OR=0·805,P=0·003) and a risk factor for high BMI (OR=0·718,P=0·002; OR=0·707,P=0·001). Regardless of socio-economic status, depression was involved with risk of low adherence (OR=1·069,P=0·021). Girls with lower MD adherence presented significantly higher scores for eating disorders measured using the Eating Disorder Inventory-2 (low adherence, mean 18·9 (sd13·5); high adherence, mean 8·9 (sd9·0),P=0·020) and the Youth Inventory-4 (low adherence, mean 5·2 (sd4·3); medium adherence, mean 3·6 (sd3·2),P=0·044). They also presented higher depression symptoms (low adherence, mean 17·7 (sd9·6); medium adherence, mean 12·3 (sd7·2),P=0·01) than girls with high adherence.ConclusionsThe results highlight the influence of psychosocial factors on levels of MD adherence. These factors need to be taken into account when developing prevention and health promotion initiatives.

BackgroundTreatment-free survival (TFS) characterizes disease control after discontinuation of immune checkpoint inhibitors (ICIs) until subsequent therapy or death. We previously evaluated TFS in a pooled analysis of the CheckMate 067 and CheckMate 069 trials of the ICIs nivolumab and ipilimumab, alone or in combination, in patients with advanced melanoma after minimum follow-up of 36 months. This analysis investigated TFS differences between treatments in CheckMate 067 after a minimum follow-up of 60 months, and their relation to overall survival (OS) differences.MethodsData were from 937 patients who initiated treatment (nivolumab plus ipilimumab, nivolumab, or ipilimumab) in CheckMate 067 (NCT01844505). TFS was defined as the area between the Kaplan-Meier curves for time to protocol therapy cessation and time to subsequent systemic therapy initiation or death, each measured from randomization. TFS was partitioned as time with and without toxicity. Toxicity included persistent and late-onset grade ≥2 select treatment-related adverse events (ie, those of potential immunologic etiology). The area between Kaplan-Meier curves was estimated by the difference in 60-month restricted-mean times of the endpoints. Between-group differences were estimated with bootstrapped 95% CIs.ResultsAt 60 months from randomization, 39%, 24%, and 11% of patients assigned to treatment with nivolumab plus ipilimumab, nivolumab, and ipilimumab, respectively, had survived and were treatment-free. The 60-month mean TFS was approximately twice as long with the combination (19.7 months) than with nivolumab (9.9 months; absolute difference, 9.8 (95% CI 6.7 to 12.8)) or ipilimumab (11.9 months; absolute difference, 7.8 (95% CI 4.6 to 11.0)). In the respective groups, mean TFS represented 33% (8% with and 25% without toxicity), 17% (2% and 14%), and 20% (3% and 17%) of the 60-month period. Compared with 36-month estimates, mean TFS over the 60-month period represented slightly greater percentages of time in the nivolumab-containing regimen groups and a lesser percentage in the ipilimumab group. TFS differences between the combination and either monotherapy increased with longer follow-up.ConclusionsAlong with improved long-term OS with the nivolumab-containing regimens versus ipilimumab, TFS without toxicity was sustained with nivolumab plus ipilimumab versus either monotherapy, demonstrating larger between-group differences with extended follow-up.

Worldwide, it is estimated that about 1.3 million new gynecological cancer cases are diagnosed each year. For 2018, the predicted annual totals were cervix uteri 569 847, corpus uteri 382 069, ovary 295 414, vulva 44 235, and va​gina 17 600. Treatments include hysterectomy with or without bilateral salpingo-oophorectomy, radiotherapy, and chemotherapy. These can result in loss of ovarian function and, in women under the age of 45 years, early menopause. The aim of this position statement is to set out an individualized approach to the management, with or without menopausal hormone therapy, of menopausal symptoms and the prevention and treatment of osteoporosis in women with gynecological cancer. Our methods comprised a literature review and consensus of expert opinion. The limited data suggest that women with low-grade, early-stage endometrial cancer may consider systemic or topical estrogens. However, menopausal hormone therapy may stimulate tumor growth in patients with more advanced disease, and non-hormonal approaches are recommended. Uterine sarcomas may be hormone dependent, and therefore estrogen and progesterone receptor testing should be undertaken to guide decisions as to whether menopausal hormone therapy or non-hormonal strategies should be used. The limited evidence available suggests that menopausal hormone therapy, either systemic or topical, does not appear to be associated with harm and does not decrease overall or disease-free survival in women with non-serous epithelial ovarian cancer and germ cell tumors. Caution is required with both systemic and topical menopausal hormone therapy in women with serous and granulosa cell tumors because of their hormone dependence, and non-hormonal options are recommended as initial therapy. There is no evidence to contraindicate the use of systemic or topical menopausal hormone therapy by women with cervical, vaginal, or vulvar cancer, as these tumors are not considered to be hormone dependent.

6546 Background: Anti-PD-1 mAbs have changed the landscape of R/M HNSCC treatment, but physical, immunologic, and metabolic barriers present in the tumor microenvironment are likely drivers of low response rates. Hypoxia is a well-established feature of the tumor microenvironment and may act as a barrier to T cell infiltration and function. We evaluated the effect of hypoxia on the efficacy of anti-PD-1 mAb treatment in R/M HNSCC patients. Methods: We conducted a retrospective analysis of R/M patients treated with anti-PD-1 mAb that had consented to the UPMC Hillman tissue banking protocol (HCC 99-069). Pre-treatment archival FFPE samples were analyzed via immunofluorescent imaging for number of CD8+ T cells (CD8), Tregs, and the percent area (% CAIX) and mean intensity (Int) of carbonic anhydrase IX, a well-described marker of hypoxia. Tissue sections stained with PanCK, CAIX, CD8, Foxp3, and DAPI were imaged with an Olympus IX 83 microscope. ImageJ software and custom software plugins were used to determine %CAIX, Int, CD8, and Treg. PD-L1 by IHC was reported as a combined positive score (CPS) defining positive as CPS > 1. We compared non-responders (NR) i.e. PD to responders (R) i.e. PR or SD, and analyzed OS, PFS. All data were analyzed using GraphPad Prism software. Two-tailed unpaired t test was used when comparing 2 groups, 1-way ANOVA was used for multiple comparisons, and log-rank test was used for survival analysis. Results: The 36 patients included were 69% male, median age 59, 58% smokers. 61% were platinum failure. Primary site included 39% OC, 22% OPC (38% HPV positive), 17% Larynx, 17% other, 5% hypopharynx. Low %CAIX/Int, high CD8, and high CD8/Treg were all significantly associated with R. Patients with low %CAIX/Int (12 month OS Low: 75% vs. Mid: 17% vs. High:8%, p = 0.02) and high CD8/Treg had a significant increase in OS. Only high CD8 was associated with significantly higher PFS. Low %CAIX alone showed a non-significant trend towards increased R and no difference in PFS/OS. There was no difference in CD8, CD8/Treg, PD-L1 and Treg between %CAIX/Int groups. Conclusions: To our knowledge this is the first evaluation of tumor hypoxia as a predictive biomarker in anti-PD-1 mAb treated R/M HNSCC patients. Lower hypoxia by %CAIX/Int was associated with significantly increased response and OS. While further analysis in a larger dataset is needed to confirm, the lack of significant difference in CD8, Treg, PD-L1, and CD8/Treg between %CAIX/Int groups (Low, Mid, High) suggests that hypoxia may be an independent predictive marker.

AbstractA group of subarboreal tropical beetles, the Xystosomi of subtribe Xystosomina new subtribe, is revised and reclassified based on a reevaluation of structural characters. Xystosomi are found in tropical Australia (Queensland) and tropical/subtropical America (Guerrero, México, to Aguas Blancas, Argentina). The largest concentration of species occurs near the equator in the Amazon Basin, but a significant radiation of flightless forms was recently discovered in the northern Andes of Colombia, Ecuador, and Venezuela. Xystosomina also includes the Mioptachyi, which at present is composed of the genera Mioptachys and Inpa.Seventy-six species of Neotropical and Australian Xystosomi are described or re-described, illustrated, or keyed. This assemblage includes 12 classic species, 24 species described in the last 3 decades, and 40 new species, a 6-fold increase since the time of Henry Walter Bates, the last 19th-century entomologist to study this remarkable lineage of carabid beetles. The main purpose of the present study was to examine factors that affect bone mineral density (BMD) in female ballet dancers. Training history, Ca intake, body composition, total body BMD (TBMD) and site-specific BMD, and bone mineral content were described in twenty-four female ballet dancers (mean age 22·6 (SD 4·5) years). Training history was determined by questionnaires, Ca intake by 7 d dietary record, BMD and bone mineral content by dual-energy X-ray absorptiometry (DXA), total body water by 2H dilution, extracellular water by bromide dilution, body fat by underwater weighing (UWW; two-component model), DXA, and the four-component (4C) model. Dancers had a significantly lower body mass index (BMI 18·9 (SD 1·0) kg/m2) than controls (21·3 (SD 19) kg/m2), with significantly lower percentage body fat (17·4 (SD 3·9) v. 24·4 (SD 5·1)) but comparable fat-free mass. Mean TBMD (1·147 (SD 0 ·069) g/cm2) was significantly higher (6 %) compared with that of a reference population. These high values could be attributed to the high BMD of legs and pelvis, the weight-bearing sites of the dancer's body. No relationship was found between age, start of ballet classes, period (years) of dancing, Ca intake, and BMD (total and site-specific). However, TBMD was positively related to BMI, and negatively related to the age of menarche. BMD of the legs was significantly related to daily period (h) of training. Depending on the method used the percentage body fat ranged from 16·4 (by DXA) to 18·3 by the 4C model. These differences were significantly related to the TBMD. Percentage body fat by the different methods was not significantly different, except for DXA and 4C model. The present study showed that, despite the factors that have a negative effect on BMD, such as low body mass and late menarche, BMD in female ballet dancers was relatively high. These high values were probably caused by high levels of weight-bearing physical activity.

4002 Background: Cetuximab prolongs survival in pretreated advanced colorectal cancer (CRC) patients (pts). For these pts, the effect of palliative treatment on quality of life (QOL) is also an important outcome. Methods: CO.17 was a randomized controlled trial (n=572) of cetuximab combined with best supportive care (BSC) vs BSC alone in pts with pretreated epidermal growth factor receptor-positive advanced CRC. Primary endpoint was survival. QOL was a secondary endpoint using the EORTC QLQ-C30 at baseline, 4, 8, 16, and 24 weeks (wks). Primary endpoint for QOL compared physical function (PF) and global health status (Global) mean changes from baseline to 8 and 16 wks using Wilcoxon test. Negative change scores denote worsening QOL. The proportion of pts in either arm with at least a 10 unit deterioration at 8 and 16 wks from baseline was compared using Fischer’s exact test. Kaplan-Meier estimates and logrank tests on median time to definitive QOL deterioration, defined as at least a 10 unit deterioration in PF or Global, were evaluated. Exploratory analyses of all other scale and symptom change scores at 8 and 16 wks were also performed. Results: Compliance rates for QOL questionnaires was high at baseline, 94% each arm, but did drop off over time: 73% vs 60% at week 8 and 60% vs 39% at week 16 for cetuximab vs BSC, respectively. PF change scores at 8 wks were -3.9 for cetuximab vs -8.6 for BSC (p=.046); at 16 wks -5.9 vs -12.5 (p=.027). Global change scores were -0.5 vs -7.1 (p=.008) and -3.6 vs -15.2 (p=.008) at 8 and 16 wks respectively. The proportion of pts with deteriorating PF was 25% for cetuximab vs 35% for BSC at 8 wks (p=.051) and 30 vs 43% at 16 wks (p=.069). Deteriorating Global was seen in 23 vs 38% at 8 wks (p=.004) and 31 vs 49% at 16 wks (p=.0011). Time to PF deterioration was 5.4 months [95% CI 3.8–5.7] for cetuximab vs 3.7 months [2.0–3.9] for BSC (p=.022). Time to Global deterioration was 5.4 [3.9–5.7] vs 3.7 months [2.1–3.9] (p=.062). Exploratory analyses of all other scale and symptom change scores at 8 and 16 wks showed all differences of p<.05 also favored cetuximab. Conclusions: Cetuximab provides palliation in pretreated pts with advanced CRC, delaying deterioration in QOL as well as improving survival. [Table: see text]

ABSTRACT Insulin-like growth factors (IGFs) and IGF-binding proteins (IGFBPs) may be important factors in the control of neonatal growth. We have examined the production, in vitro, of IGFBPs and IGFs by hindlimb skeletal muscle from normal and small-for-gestational age (SGA) neonatal rats. Conditioned medium was collected from muscle strips after incubation at 37 °C for 2 h in Ham's F-12 medium. The conditioned medium was subjected to acid-gel permeation chromatography to separate IGFBPs from IGFs. The binding of 125I-labelled IGF-I to IGFBPs from both control and SGA muscle was displaced equipotently by IGF-I and IGF-II and not at all by insulin. IGFBPs from control and SGA muscles bound IGF-I with comparable affinities (Kd = 0·071 and 0·069 nmol/l respectively). When IGF-II was used as tracer, neither IGF-I nor insulin competed for binding. Western ligand blots of IGFBPs in conditioned media from both control and SGA muscles showed three bands of radioactivity at molecular masses equivalent to 24, 30 and 40 kDa. When the release of IGFBPs by muscle tissue in vitro was quantified by measuring the number of IGF-I binding sites in acid-fractionated medium it was apparent that the muscles from SGA pups secreted significantly more IGFBPs (39·3±7·5 fmol/mg muscle protein per 2 h) than the muscles from control pups (17·8±2·7 fmol/mg protein per 2 h; P < 0·05). In contrast to the IGFBPs, more IGF activity was secreted by the muscles from the control pups (61·1±15·6 fmol/mg muscle protein per 2 h) than the muscles from the SGA pups (12·6±5·8 fmol/mg muscle protein per 2 h; P < 0·05). Analysis of the IGF activity with assays specific for IGF-I and IGF-II showed that both SGA and control muscles secreted predominantly IGF-II with approximately 10% of the total IGF activity measurable as IGF-I. Background. The assessment of medical technologies based on digital information is prompt and accurate, but it is currently not widely used, and analysis methods for this have not been developed, which became the basis for this study. Aim. To economically evaluate the technologies of reperfusion therapy and dispensary observation in coronary heart disease using electronic data on medical care. Material and methods. 586 patients with myocardial infarction treated in 2016 in two vascular centers of the Orenburg region were identified according to the registers-accounts of the compulsory health insurance fund. Among them were 349 patients with reperfusion therapy (main group) and 237 without it (control group). In the groups, the cost-effectiveness indicator, the cost of inpatient treatment of myocardial infarction and treatment in the postinfarction period were determined. Also, according to the registers-accounts for 2016, 17 225 patients with angina pectoris were identified. Among them, 13 208 patients, who applied to polyclinics for dispensary purposes during 20162017, and 4017 patients who did not apply, were registered the main and control groups. Between groups, the total cost of outpatient, emergency, emergency hospital care and the cost-effectiveness ratio were compared. When processing the data, linear statistics methods and parametric comparison methods (Student's t-test and z-test) were used. Results. The cost-effectiveness indicator in the group with reperfusion therapy was 425 879, without it 643 069 rubles/unit, 1.5 times more in the control group. In the postinfarction period, the cost of treatment in the main group was lower than in the control group, 1223.6619.04 and 1321.1121.15 rubles, respectively, for outpatient care (p=0.001), 35 138.91299.85 and 51 189.64903.15 rubles for inpatient care (p=0.002), 4093.058.98 and 5428.652.95 rubles for an ambulance (p=0.001). The cost of angina treatment in the main group was 2.1 times less than in the control group, 6541.385.58 and 13 671.492.12 rubles (p=0.001). The cost-effectiveness indicator in the main group was 6867, in the control group it was 15 293 rubles/unit, 2.2 times more in the control group. Conclusion. The use of reperfusion therapy and dispensary observation in coronary heart disease is economically feasible.

440 Background: Up to one-third of patients with renal cell carcinoma present with metastatic disease (mRCC). Cytoreductive nephrectomy remains the standard of care for appropriately selected patients. However, cytoreductive nephrectomy is not always practical. We sought to identify the indications and outcomes for partial nephrectomy (PN) in our cohort of patients with mRCC, with particular attention to different PN subgroups. Methods: Using our institutional database, 30 patients with mRCC who underwent PN between 1996 and 2011 were identified. Demographic, clinical, and pathologic variables were collected. Non-parametric statistics and log-rank tests were used. Cancer specific survival (CSS) was estimated using Kaplan-Meier method according to presentation, tumor size, and presence of metastatic disease, from the time of PN to last follow-up or death. Results: The median age at PN was 57 years (range 32-84). 8 patients presented with bilateral synchronous renal masses; 17 presented with a metachronous contralateral renal mass; and 5 presented with a unilateral renal mass (including 3 in a solitary kidney). Median follow-up after PN was 32 months (range 1-184). Overall, 23 patients (77%) died of disease at a median of 27 months (range 7-86) after PN. Patients who underwent PN for a metachronous contralateral renal mass had a median CSS of 61 months compared to those with bilateral synchronous or unilateral renal masses (CSS 26.5 months, HR 2.98, p =.012 and CSS 31, HR 2.12, p =.069, respectively). Patients who underwent PN for a renal mass ≤4cm and >4cm had a median CSS of 42 and 26.5 months, respectively (HR 2.49, p =.037). Median CSS for patients with and without metastatic disease at original diagnosis was 27 and 61 months, respectively (HR 2.85, p =.013). In this study, patients who became M0 after metastasectomy did not have improved CSS compared to patients who did not (42 and 32 months, p =0.152). Conclusions: Our findings suggest that the burden of disease at initial diagnosis, timing of presentation of the PN index lesion, and the size of the renal mass at PN play an important role in survival. These factors should be taken into consideration when determining which patients would benefit from PN in the setting of mRCC.

Abstract Thalidomide-Dexamethasone (TD) is an active regimen both in patients (pts) with relapsing/refractory and in untreated pts with multiple myeloma (MM). Here we compare TD with standard Melphalan-Prednisone (MP) in previously untreated elderly pts with MM. 274 pts have been enrolled (median age: 72 yrs, stage I: 9 (3%), stage II: 84 (31%), stage III: 179 (65%). Pts were randomized to T 200mg/day and D 40mg, days 1–4 and 15–18 (on odd cycles) and days 1–4 (on even cycles) or M 0.25mg/kg days 1–4 and P2mg/kg days 1–4, q 4–6 weeks. T should be dosed up to 400mg/day, if feasible. Pts achieving response or SD were randomized to maintenance therapy either with T (≤200mg/day)-Interferon-a2b (IFN, 3Mega U, TIW) or IFN (3Mega U/TIW). Zoledronic acid (4mg) was administered monthly to all pts during the entire treatment period. Response is defined according Blade’s criteria, plus nCR defined as IF positive or >90%↓ in PP and VGPR defined as >75% ↓reduction in PP. 231 pts are evaluable per protocol. Best response to TD: CR (14%) nCR 17%, VGPR 17%, PR 21%, yielding an ORR (CR-PR) of 68%. Best response to MP: CR 7%, nCR 8%, VGPR 14%, PR 22%; ORR 51% (ORR in TD vs. MP p=.0044). Time to response and time to best response were shorter in the TD (6, 16 weeks, respectively) compared to the MP group (16, 25 weeks, respectively; p<.001, p<.002, respectively).There was a tendency for more early treatment discontinuations in the TD arm (15 vs. 11, p=.416) and a higher mortality during the first treatment year (31 vs. 17, p=.026). Pts treated with MP showed a tendency for longer EFS (median: 43 vs. 25mos; p<.07) and had sign. longer OS compared to those on TD (median: 58 vs. 45mos;p=.029). A retrospective comparison of pts ≤ aged 72 showed no difference in OS (median: 57.9 vs. 50 mos, p<.186) but OS was longer in pts >72 yrs treated with MP (median: 46.5.9 vs. 25.4, p<.0625). Maintenance treatment in the 108 pts randomized to either T-IFN, or IFN only, did not result in a sign. difference in OS (from start of maintenance: not reached vs. 41.1 mos, p<.95). Pts on MP had a higher incidence of G 3–4 leucopenia (14% vs. 3%; p< .0001), while for thrombocytopenia only a tendency for a higher frequency was noted. Pts on TD had more G 2–3 neuropathy (28% vs. 10%, p<.001), constipation (30% vs. 10%, p<.001), psychological toxicity (18% vs. 6%, p<.0006), and a tendency for a higher rate of skin toxicity (9% vs. 5%, p=.069), and infections (23% vs. 16%, p<.12) compared to those on MP. The cumulative incidence of DVTs was 15% in pts on TD and 6% in those on MP (p=.089). In conclusion, TD treatment resulted in sign. higher rate of CR/nCR and ORR, a sign. shorter time to response and to best response. Pts on TD had similar EFS but sign. shorter OS. This was particularly true in pts aged >72 years but not in those aged ≤72 who showed similar OS with TD and MP. Pts on TD had more neuropathy, DVT, psychological toxicity, and a higher rate of early treatment discontinuations, while haematological toxicity was higher in pts treated with MP.

440 Background: Fibrolamellar hepatocellular carcinoma (FL-HCC), encompassing 1%-5% of all liver cancers, is a rare, malignant cancer that typically affects patients less than 40 years old. FL-HCC usually affects individuals with healthy livers and initially presents asymptomatically. Prior studies describe the importance of surgery on survival, with a 5-year survival of about 44%-68% with surgery versus 2%-17% without surgery. Disparities in outcomes regarding surgical factors and survival between private insurance, Medicare, Medicaid, and no insurance remain unknown. This study aims to determine differential surgical factors and survival of patients with FL-HCC based on insurance status. Methods: The National Cancer Database was used to identify patients diagnosed with FL-HCC from 2004-2019 using the histology code 8171 as assigned by the Commission on Cancer Accreditation program. Kaplan-Meier, Chi-Square, ANOVA, and Cox Proportional Hazards tests were performed. Data was analyzed using SPSS version 29 and statistical significance was set at α=0.05. Results: Of the 436 patients included, 241 (55%) had private insurance and 96 (22%) had Medicare. Patients with private insurance had a longer median survival (49 months) than patients with Medicare (18 months; p<.001) and Medicaid (26 months; p<.004);no significant survival difference existed between Medicare, Medicaid, and no insurance (39 months) ( p’s≥.063). After controlling for age, income, tumor size, adjuvant therapy, receiving surgery, and surgical margins of primary site, private insurance was associated with an independent decrease in hazard (HR=.899; p=.004).Patients with private insurance received more surgery (68%) than those with Medicaid (50%), no insurance (48%), and Medicare (32%) ( p’s<.001); grade did not differ amongst insurance types ( p=.069).Furthermore, patients with private insurance had more surgical margins of the primary tumor site without residual tumor (57%) compared to those with Medicaid (40%), no insurance (38%), and Medicare (23%) ( p’s<.001).While patients with Medicare presented with greater co-morbidities, age at diagnosis (67), tumor size (476 mm), positive regional lymph nodes (90), and days between diagnosis and treatment (45), patients with private insurance presented at a younger age (31) with fewer co-morbidities, smaller sized tumors (366 mm), and fewer days between diagnosis and treatment (26) ( p’s<.018). Conclusions: Since patients with private insurance received the most surgery, best surgical margins, and displayed the highest overall survival, while patients with Medicare were diagnosed at the highest age with the largest tumor size and most positive regional lymph nodes, future research should explore ways to alleviate the disparity in factors affecting surgical resections and early diagnoses among insurance statuses.

Abstract Previous studies have shown that specific chromosomal abnormalities are of major prognostic significance in patients with multiple myeloma (MM). It has been recently suggested that a t(11;14)(q13;q32) may be an indicator of favorable outcome in MM. In this investigation, we analyzed 163 patients with newly diagnosed MM (53% treated by high-dose therapy) to address the question whether or not the simultaneous occurrence of a t(11;14) and a deletion 13q [del(13q)], an established negative prognostic factor in MM, has any impact on prognosis. DNA-specific probes for IgH (14q32) and cyclin-D1 (11q13) were used for interphase FISH analysis of clonal plasma cells (cytoplasmic Ig positive). A t(11;14) by FISH was shown in 27 of the 163 MM patients (16.6%); the abnormality was present in the majority (median, 89%) of clonal plasma cells. Immunohistochemical analysis of CYCLIN-D1 expression was carried out in 72 patients, of whom 11 had a t(11;14) by FISH; all 11 patients had evidence for CYCLIN-D1 protein expression. Presence of a t(11;14) did not show significant correlations with standard laboratory and clinical MM features including type of the paraprotein, hemoglobin, creatinine, LDH, albumine, calcium, CRP, and beta-2-microglobulin (b2M). In contrast to a recent report, there was also no association with CD20 expression by MM cells. With respect to survival, presence of any 14q-translocation (52% of patients) was associated with similar overall survival times (OS) compared to patients lacking a t(14q), whereas patients with a t(11;14) experienced prolonged OS (median, 70+ months vs. 59.8 months among patients without t(11;14); P = .071). This survival advantage was even greater among the 16 patients with t(11;14) who were also normal for 13q (P = .02); however, occurrence of a del(13q) concomitantly with a t(11;14) was indicative for shortened progression-free survial (17.7 months vs. 31.6 months; P = .17) and OS (P = .07). A survival benefit of MM patients with a t(11;14) was particularly evident for the population receiving standard-dose chemotherapy (median OS not yet reached; P = .02). By multivariate Cox regression analysis, low serum b2M at diagnosis (P = .001), absence of a del(13q) (P = .004), high-dose therapy (P = .034), and presence of t(11;14)/no del(13q) (P = .069) emerged as independent favorable parameters for OS. Thus, according to the cytogenetic pattern, three prognostic groups of patients could be discriminated (P < .001): patients with good [t(11;14), no del(13q)], intermediate [no t(11;14), no del(13q)], and poor prognosis [no t(11;14), del(13q)]. We conclude that MM with t(11;14) represents a heterogenous entity, and only the cytogenetic pattern t(11;14)/no del(13q) characterizes the most favorable prognostic group of MM, with sensitive disease to multiple lines of anti-MM therapy.

8504 Background: Neoadjuvant immune checkpoint inhibitors (ICIs) induce major pathologic response (MPR) rates of 20 to 45% in resected NSCLCs. We report the results of NEOSTAR - a phase 2 trial of neoadjuvant N or NI for NSCLCs. Methods: Pts with stage I-IIIA (single N2) resectable NSCLC (AJCC 7th), PS 0-1, were randomized to N (3 mg/kg IV, D1, 15, 29) or N plus I (1 mg/kg IV, D1) followed by surgery (n = 44). Primary endpoint: MPR (≤10% viable tumor), hypothesized to be higher than MPR to induction chemotherapy historical controls. Tumor immune infiltrates and pre- & post-ICI tumor PD-L1 % were assessed by flow cytometry & IHC. Wilcoxon ranked sum test & Fisher’s exact test were used for comparisons. Results: 44 pts were randomized, 23 N, 21 NI: mean age 66, 64% males, 18% never smokers, 59% adenocarcinomas, stages: IA 8 (18%), IB 15 (34%), IIA 7 (16%) IIB 5 (11%); IIIA 9 (20%). Only 3 pts received < 3 doses due to TRAEs (7%). 34 pts had surgery post ICIs (7 not resected [7/41], 17%, [2 N, 5 NI], 3 pending). There were 10 MPRs in 41 pts overall (24%, 4 N, 6 NI), of which 6 were path CRs (15%, 2 N [9%], 4 NI [21%]). Among 34 resected pts, MPR rate was 29% (N 20%, NI 43%). Median % of viable tumor was lower post NI vs N (20% vs 65%, p = .097). ORR (RECIST v1.1) was 22% (8 PRs [5 N, 3 NI], 1 CR [NI]); 15% of pts had PD (3 N, 3 NI). The proportion of CR+PR in MPR+ was higher than in MPR- (6 [60%] vs 2 [7%], p < .001). Surgical complications included 2 bronchopleural fistulas (BPFs) in N & 8 air leaks (5 N, 3 NI). G3-G5 TRAEs included a death due to BPF post steroid-treated pneumonitis (G5, N); G3 pneumonia, hypoxia, hypermagnesemia (1 each, all N), G3 diarrhea (1 NI). CD3+ & CD103+ tissue resident memory CD8+ TILs were higher in NI- vs N-treated tumors (CD3+ 81.2% vs 54.4%, p = .028; CD8+ 56.2% vs 38.3%, p = .069). Median pre-treatment tumor PD-L1 was higher in responders (MPR+, CR+PR) vs non-responders (80% vs 1%, p = .024), and the % of viable tumor was lower in tumors with PD-L1 > 1% vs PD-L1 ≤1% (median 20% vs 80%, p = .046). Conclusions: Overall a 24% MPR rate to neoadjuvant ICIs was observed. NI induced a higher % of non-viable tumor and of tissue resident memory TILs vs N. 184 Background: SETD2, a key methyltransferase modulating histone 3 gene transcription, has been shown to act as a tumor suppressor gene by reducing oxidative stress, colonic inflammation and tumorigenesis in animal models. SETD2 gene expression has been reported to be significantly downregulated in CRC and linked with poorer patient survival. Additionally, SETD2 plays an important role in DNA repair and loss of function mutations have been associated with increased tumor mutational burden (TMB), mismatch repair (MMR) deficiency, and benefit from immunotherapy. Hence, we aimed to characterize the molecular features associated with SETD2 gene expression in CRC. Methods: 15,425 CRC tumors tested at Caris Life Sciences (Phoenix, AZ) with NextGen Sequencing on DNA (592 genes or whole exome sequencing), RNA (whole transcriptome sequencing), and IHC were analyzed. SETD2-high and -low expression were defined as ≥ top and < bottom quartile of SETD2 transcripts per million (TPM), respectively. Cell infiltration in the tumor microenvironment (TME) was estimated by QuantiSEQ. Mann-Whitney U and X2/Fisher-Exact tests were applied where appropriate, with P-values adjusted for multiple comparisons ( q < .05). Gene expression profiles were analyzed for transcriptional signatures predictive of response to immunotherapy (T cell-inflamed) and MAPK pathway activation (MPAS). Real-world overall survival information was obtained from insurance claims data and Kaplan-Meier estimates were calculated for molecularly defined patients. Results: SETD2 expression was significantly increased in left-sided/rectal compared to right-sided tumors (median TPM [mTPM] 55.8 vs 51.1, q < .001). SETD2 mutations were associated with reduced SETD2 expression in pMMR/MSS CRC (mTPM 37.9 vs 54.3 in WT, q < .001), although not statistically significant in dMMR/MSI-H CRC (mTPM 43.6 vs 51.7 in WT, P = .17). Compared to SETD2-high tumors, SETD2-low was associated with increased rates of TMB-high (10.4% vs 8.2%, P = .009), dMMR/MSI-H (7.3% vs 5.6%, P = .02), and PD-L1 IHC levels (4.4% vs 2.5%, q < .002). Only minor differences in gene mutation and copy number rates were observed between SETD2-high and -low tumors , whereas SETD2-high TME was associated with increased immune cell infiltration, including neutrophils, B cells, NK cells, M2 macrophages, and dendritic cells ( q < .05). SETD2-high was also associated with increased T cell-inflamed and MPAS signatures (152% and 159% median increase, respectively, q < .001), regardless of tumor MMR status. Among dMMR/MSI-H CRC, SETD2-high tumors receiving immune checkpoint inhibitors had longer time-on-treatment (HR: 0.39, 95% CI: 0.2-0.76, P = .005) and a trend towards longer OS (HR: 0.29, 95% CI: 0.07-1.2, P = .069). Conclusions: Our data show distinct immune biomarkers and TME cell infiltration associated with SETD2 gene expression in CRC. These findings support a key role for SETD2 in modulating anti-tumor immunity and TME.

Long-term preservation of seed germplasm is a high agricultural priority. It assures that genetic diversity will be available for future generations for continued plant improvement. This experiment reports on the affect that storage temperature had on the viability of 65 selections of lettuce seed stored for 30 years. The average seed moisture content was 5.5% ± 0.5% (fresh weight basis). Fresh seed samples were placed at 5 °C storage in 1969. In 1975 they were then transferred to -18 °C storage. Viability remained at 98% ± 5% for the first 14 years of 5 /-18 °C storage, then viability declined. At 17 years storage, the average viability had dropped to 75% and continued to drop at about 4%/year. At the 17-year mark, individual samples were split, one-half remained at -18 °C the other half was placed under liquid nitrogen vapor (lnv) conditions (about -150 to -190 °C). The -18 °C stored samples continued to deteriorate to 14% viability at the 30 year test period (1999). The samples placed in lnv did not decrease further in viability and remained at 75% viability at the 30-year mark. Seed vigor was reduced in the -18 °C stored seeds that were still viable. The lnv-preserved samples were significantly more vigorous. It is clear from this experiment that lnv preservation was significantly superior to -18 °C storage and, in fact, stopped or significantly reduced the rate of viability loss in samples that are rapidly deteriorating.

e14705 Background: Increased use of immune checkpoint inhibitors (ICI) has resulted in a rise in immune related adverse events (irAEs), which many non-oncology clinicians may underrecognize. Controversy exists surrounding whether irAE presence has an impact on survival. Our primary goal was to evaluate the timing between detection to diagnosis of irAE, and determine the department responsible for making the irAE diagnosis. We also assess the incidence of single vs multisystem irAEs, impact of irAE on survival, and assess whether standard irAEs (s-irAEs) those diagnosed within 90 days of initial ICI administration vs delayed irAE (d-irAEs) those diagnosed >90 days impacts survival. Methods: We performed an IRB approved retrospective study of patients 18 years and older who received ICIs for a malignancy at Gundersen Health System between January 1, 2011 and December 31, 2021. Statistical analysis included Chi-square, Fisher’s Exact, and Wilcoxon rank sum tests and Kaplan-Meier survival analysis. All analysis was completed in R version 4.2.3 with a p-value <0.05 considered significant. Results: Our study included 664 patients who were 99.6% white, 60% male, and an average age of 67 years. Median Charlson Comorbidity Index was 10. There were 245 (36.8%) patients with at least 1 irAE, and 62 (9.3%) with a multisystem irAE. While 28 (11%) irAEs were initially detected by non-oncology providers, only 10 (4.1%) irAEs were diagnosed by a non-oncology department. Median time (days) from symptoms to diagnosis was 3 for those diagnosed in non-oncology departments and 0 for those diagnosed in oncology (p<0.01). A total of 142 (58.0%) s-irAEs were detected with grades 1-2-3-4-5 accounting for 21.1%-54.9%-19.7%-1.4%-2.8%. There were 103 (42.0%) d-irAEs with grades 1-2-3-4-5 accounting for 23.3%-53.4%-17.4%-5.8%-0%. Of the d-irAEs, 17 (16.5%) occurred >365 days from initiation of ICI. The 3 most common irAE in this patient population included thyroiditis (34%), dermatitis (21%), and pneumonitis (12%). Our data shows a significant survival difference in patients with an irAE compared to those without (p<.01). No significant survival difference was noted between patients who had s-irAE vs d-irAE (p=.069). Conclusions: Our results show that irAEs may be underrecognized and often not formally diagnosed by non-oncology providers. However, median time to diagnosis is short within this context. This may be due to non-oncology providers feeling uncomfortable making the final decision, which emphasizes the importance for oncologists to finalize irAE diagnoses. We did see a positive impact on survival with irAE presence. While oncologists often clinically expect irAEs to surface within the first 6-10 weeks of treatment, our study demonstrates that 42% of irAEs occur after 12 weeks from ICI initiation. There is no difference in survival between s-irAEs vs d-irAEs, and overall grade distribution between these two groups appears similar.

The circumambulatory rearrangements of some 8,8-dimethylhomotropylium cations and their corresponding iron tricarbonyl complexes have been examined. 8,8-Dimethylhomotropylium cation, 4, has been prepared by protonation of 8,8-dimethyl-bicyclo[5.1.0]octa-3,5-dien-2-ol in FSO3H/SO2ClF. Evidence for homoaromatic delocalization in 4 was found in its characteristic 1H nmr spectrum, particularly when this was compared to that of the non-cyclically delocalized 8,8-dimethyl-bicyclo[5.1.0]octadienyliron tricarbonyl complex 7. Cation 4 was found to be stable at low temperatures but to isomerize to isopropyltropylium ion, 8, at −50 °C, [Formula: see text] The specifically deuterated cation 4D was prepared and a slow circumambulation of C8 around the basal 7-membered ring of this cation was found to occur at −90 °C. The free energy of activation for this circumambulation is 14.5 kcal/mol. Homotropylium cation 4 was also prepared by protonation of 8,8-dimethylbicyclo[3.2.1]octa-3,6-dien-2-ol, 14, at −120 °C. No evidence for the presence of the corresponding bicyclo[3.2.1]octadienyl ion 12 was obtained. The 2,8,8-trimethylhomotropylium cation, 17, was prepared and shown to undergo a circumambulatory rearrangement to give the 4,8,8-trimethyl substituted cation 20 as the major product. In this trimethyl series it also proved possible to generate the 2,8,8-trimethylbicyclo[3.2.1]octadienyl cation, 18, and observe its 1H nmr spectrum at very low temperatures (−116 °C). The proton chemical shifts of 18 suggest that the charge is predominantly located on the 3-carbon bridge. Cation 18 isomerized to 17, 20, and other products at temperatures above −100 °C. The 2,8,8-trimethylbicyclo[5.1.0]octadienyliron tricarbonyl complex 22 was also shown to undergo a circumambulatory rearrangement in which both C8 and the Fe(CO)3 grouping migrate.

We previously implicated 3-hydroxy oxylipins and ascospore structure in ascospore release from enclosed asci. Using confocal laser scanning microscopy on cells stained with fluorescein-coupled, 3-hydroxy oxylipin-specific antibodies, we found that oxylipins are specifically associated with ascospores and not the vegetative cells or ascus wall of Ascoidea corymbosa. Using gas chromatography – mass spectrometry the oxylipin 3-hydroxy 17:0 could be identified. Here, we visualize for the first time the forced release of oxylipin-coated, hat-shaped ascospores from terminally torn asci, probably through turgor pressure. We suggest that oxylipin-coated, razor-sharp, hat-shaped ascospore brims may play a role in rupturing the ascus to affect release.Key words: Ascoidea corymbosa, ascospore release, confocal laser scanning microscopy, gas chromatography – mass spectrometry, hat-shaped ascospores, 3-hydroxy oxylipins.

Growth of Pinus taeda L., with and without control of competing herbaceous vegetation, was evaluated over 15 years at planting densities ranging from 747 to 2990 trees/ha. A height response to vegetation control of 0.9 m at age 5 decreased to 0.3 m at age 15. Convergence in the cumulative height between treated and untreated stands did not translate into converging basal area and volume yield. For 1500 and 2200 trees/ha, volume response to early vegetation control increased to age 15. For 800 trees/ha volume gains increased to age 12 and were stable between ages 12 and 15. Higher densities apparently captured more of the site resources made available through vegetation control. Age-15 volume gains for 2200, 1500, and 800 trees/ha were 71 (23%), 48 (17%), and 19 m3·ha-1 (8%), respectively. A separate adjustment term in height and basal-area models captured the short-term positive effect of vegetation control on height growth and longer term basal-area responses. Previously published height and basal-area models were modified to account for planting-density effects.

We examined the methanotrophs in the Trail Road Landfill soils, Ottawa, Ontario, through cultivation-independent molecular assay and the culturing approach. Denaturing gradient gel electrophoresis (DGGE) analysis of amplified methanotroph-specific 16S rDNA gene fragments revealed a more diverse type I (RuMP pathway) methanotrophic community than type II (serine pathway) in 17 soil samples taken along a 50 m transect. The type II methanotrophic community was less diverse, with the dominance of Methylocystis in almost all samples, and clustering with high similarity (85%–88%). Also, the results showed that the C/N ratio of soil organic matter could significantly affect the methanotrophic community structures. The DGGE results were supported by sequence analysis of cloned pmoA. Members of the genera Methylobacter (type I), Methylocaldum (type X), and Methylocystis (type II) appeared to be the dominant methanotrophs. From methanotrophic enrichments, we isolated type I Methylobacter sp., and 3 type II Methylocystis spp.,which appeared to be one of the dominant bacteria species in the soil sample from which isolates were obtained.

Background:Attention is drawn to the frequent combination of osteoarthritis (OA) with cardiovascular disease. Non-specific inflammation plays a significant role in the pathogenesis of OA and atherosclerosis. Limiting the physical activity of patients with OA is an additional important factor aggravating the course of cardiovascular disease (CVD). Chronic pain syndrome, causing a neuroendocrine response, is often the cause of the development of complications of atherosclerotic disease. Dyslipidemia is the main cause of atherosclerosis and vascular thrombosis.Objectives:To study variants of lipid metabolism disorders in female and male patients of different age groups with osteoarthritis.Methods:Case histories of 90 patients with OA were analyzed. The average age of patients was 63.27 ± 11.31 years. The average body mass index (BMI) is 39.8 ± 3.2. All patients underwent questionnaires, general clinical and biochemical blood tests with lipid profile determination, anthropometry, bioimpedansometry, and the main metabolic rate assessment using indirect calorimetry in dynamics (at the beginning of the study and after 3 months).Results:Burdened heredity for obesity, arterial hypertension (AH), diabetes mellitus (DM) was revealed. AH was diagnosed in 76 patients (84.4%), type II diabetes in 17 (18.9%), dyslipidemia and hypercholesterolemia in 56 (62.2%). Statins were taken by 43 patients (47.8%) - group I patients, which is associated with low adherence to therapy, group II included patients who did not initially take statins or stopped taking them at least 6 months before inclusion in the study.Against the background of diet therapy and physiotherapy exercises, BMI (R0.99; p <0.05), fat mass (R0.95; p <0.05) significantly decreased, lipid profile normalization was noted: total cholesterol (R0.66; p <0 .05), LDL (R0.69; p <0.05), HDL (R0.95; p <0.05), TG (R0.57; p <0.05), AST decreased (R0.64; p <0.05) and ALT (R0.76; p <0.05) in both groups of patients, regardless of lipid-lowering therapy. A decrease in fat mass correlated with TG levels (R0.51; p <0.05), an increase in skeletal muscle mass (R0.60; p <0.05), lean mass (R0.72; p <0.05), and active cell mass (R0.59; p <0.05). The lipid profile in the I group of patients was significantly better before and at the end of the study. Long-term effects have not been investigated due to the short duration of the study.Conclusion:In patients with OA, a high frequency of concomitant diseases of the cardiovascular system, lipid metabolism disorders was found. Non-drug therapy has a positive effect on the lipid profile and the level of transaminases. The decrease in body weight due to loss of fat mass reliably correlates with the level of TG. Timely use of statins contributes to the normalization of the lipid profile, reduces the risk of cardiovascular disease in patients with OA. It is necessary to study lipid profile disorders in patients with OA with recommendations for lifestyle modification (diet, physical activity), and if necessary, prescribe lipid-correcting therapy.References:[1]E. It is composed of 17 species: T. sentus Pritchard and Baker, T. neosentus van der Merwe, T. acanthus van der Merwe, T. sennarensis El Badry, T. parva (Matthysse and Denmark) comb, nov., T. kuzini (Wainstein), T. relenta (Matthysse and Denmark) comb, nov., T. ecclesiasticus De Leon, T. scytinus Chazeau, T. prunusus van der Merwe, T. yokogawae Ehara and Hamaoka, T. operantis (Chaudhri) comb, nov.,T. loricatus (Wainstein) comb, nov., T. bregetovae (Wainstein and Beglyarov) comb, nov., T. neosoleiger Gupta, T. ignavus (Chaudhri) comb, nov., and T. niloticus El Badry. Typhlodromus vitreus (Chaudhri) comb. nov. is designated as a junior synonym of T. bregetovae. Two species, T. ignavus and T. niloticus, are designated as "species inquirendae" because of insufficient information on their characteristics. Various generic concepts relating to the species in this group are reviewed. The group is described and a key to adult females and descriptions of each species are provided.

The sources of rare diamonds reported in northwestern British Columbia, southwestern Yukon, and parts of Alaska are enigmatic. We carried out a heavy-mineral survey of 17 streams draining bedrock in the Atlin–Nakina region in northwestern British Columbia to determine if high-pressure igneous and metamorphic rocks exhumed in the area could be potential sources for the diamond. Heavy-mineral fractions returned flakes of gold but no diamond. The ferromagnetic fractions were examined optically and by electron microprobe analysis of key indicator minerals, namely olivine, orthopyroxene, clinopyroxene, garnet, spinel, and titanite. Detritus from a horizon of coarse pebble conglomerate in the Jurassic Laberge Group south of Atlin is the most likely source of the anomalous diamond. Garnets and pyroxenes in the latter sedimentary unit were derived by rapid erosion of peridotite and eclogite massif bodies exhumed from depths approaching the diamond stability field during collision and Pliensbachian uplift in the northern Cordillera. Evidence is shown for glacial transport of detritus from both the Laberge Group sediments and Neogene volcanics, the latter of which evidently covered a much wider area before the last glaciation.

El Ejército de Tierra (ET) participa en la Campaña Antártica Española desde el año 1988, que es la participación militar en el extranjero más antigua actualmente en vigor. Esta misión militar [1] es, sin embargo, única tanto por la lejanía y las dificultades climatológicas/físicas que entraña como por la importancia de la colaboración del Ejército con la investigación científica. Todo ello convierte a la Antártida en uno de los laboratorios científicos más importantes del planeta y, por tanto, en un lugar único para desarrollar proyectos y trabajos, bajo la supervisión del Tratado Antártico que implica la total protección del medio ambiente. Este laboratorio científico ha despertado el interés de la XXXI Campaña Antártica (17-18) en volar DRONES/RPAS por primera vez en dicho continente antártico por el Ejército de Tierra, con distintos fines y misiones. La estricta legislación, dentro del marco del Tratado Antártico y en España, dirigida por el CPE, está regulada por el “Protocolo del Comité Polar Español (CPE) para la operación de aeronaves pilotadas por control remoto (RPAS) y automatizadas (UAV) en la Antártida”. Fundamentalmente basada en las regulaciones de Agencia Estatal de Seguridad Aérea (AESA), es temporal e indica el procedimiento para poder realizar actividades aéreas en la Antártida y aplicada a aeronaves inferiores a 25 kg de peso.

Nineteen mature Double Muscled (DM) cattle consisting of 11 cows and eight bulls were slaughtered between 470 and 710 kg to determine the influence of sex on carcass composition and muscle, bone and fat in DM cattle. Expressed as a percentage of the total side weight, DM bull carcasses had 15% more muscle and 55% less total fat. When sides were compared DM bulls showed a 17% increase in the muscle:bone ratio compared with DM cows. In bull carcasses, there were shifts in muscle weight distribution towards the forequarter. The ratio of hindquarter muscle:forequarter muscle was greater in cows than in bulls. Expressed as a percentage of the total side muscle, significant differences between sexes were found in 48 of the 95 muscles. The most striking sexual dimorphism was found in the neck region, particularly among the muscles responsible for secondary sexual features and those which act to elevate and extend the head. Sexual dimorphism was less pronounced in the distal parts of the both limbs. The cervical vertebrae and scapula made up a greater proportion of total side bone in DM bulls than in DM cows. There was a consistent but nonsignificant trend for the cows to have more of their bone caudally and less anteriorly compared with the bulls. Abstract Introduction Confronted with a constant increase of occupational diseases in the retail industry over the past quarter-century, the governmental organizations (CARSAT et DREETS Grand EST) together with the occupational health and safety services (OHSS) and in partnership with the French National Education have developed the first virtual project for the prevention of occupational health in retail. Methods The methodology involves the creation of a virtual model of a supermarket that serves as the backdrop for scenarios simulating risky situations. The technology provider HOLO3 designed the “Optimus Prev” application, furnished the necessary equipment to partners and handled the maintenance. The working group deployed the system within the medical-technical team of OHSS and the CARSAT addressed the national and regional management of retail chains, the trade unions, the employer associations, vocational schools and higher education institutions for immersive experimentation. The multidisciplinary teams of OHS analyzed workplace situations in super and hypermarkets, employing standardized measurements based on harmonized recommendations. Results A working group comprising 17 specialists from different institutions, driven by common goals, achieved a prevention system through 29 meetings that closely addressed the 600 hyper and supermarkets in Alsace and Moselle (France). “Optimus Prev” was integrated into 14 portable stations in France, primarily in the Grand Est region. Training deployment began in Alsace and Moselle with 160 workers from 22 supermarkets. Additionally, 37 educational institutions, including engineering students from INSA, used the tool to raise awareness of occupational risks. Discussion and conclusion This multi-institutional collaboration introduced innovative virtual prevention simulations to the retail sector, unifying prevention stakeholders in a shared message.

Soil organic constituents may strongly affect the kinetics of soil chemical processes, including K exchange reactions. We investigated the influence of organic matter on the rate of K adsorption by selected soils (Ferric Ultisol, Orthic Ultisol and Vertisol) using a H2O2 treatment and a K ion-selective electrode technique. In the reaction period of 0–30 s, in which the adsorption was too fast for one to determine rate coefficients of K adsorption, the amount of K adsorbed by the untreated soils was 158–363 mg kg–1, compared with 0.5–47 mg kg–1 for the treated soils. In the reaction period of 30–120 s, K adsorption data based on the first-order kinetics show that rate coefficients of K adsorption by the untreated soils were 47 × 10–5 s–1 (Ferric Ultisol), 59 × 10–5 s–1 (Orthic Ultisol) and 61 × 10–5 s–1 (Vertisol); by contrast, after H2O2 treatment, the rate coefficients were 23 × 10–5 s–1 (Ferric Ultisol), 17 × 10–5 s–1 (Orthic Ultisol) and 42 × 10–5 s–1 (Vertisol). Similar treatment effects were observed for the reaction period of 120–600 s, though the difference in the rate coefficients between the treatments was not as great as that for the reaction period of 30–120 s. These results indicate that organic matter considerably promotes the initial fast rate of K adsorption and has more easily accessible adsorption sites for K compared with mineral constituents of the soils. To research effects of the artificial selection of Gallus gallus on G. domesticus' nucleotide diversity of immune genes, sequence polymorphisms of G. domesticus (23 genes), G. gallus (23 genes), G. lafayetti (17 genes), and G. sonneratii (17 genes) were obtained from GenBank. The data set included 819 polymorphisms. Immune gene polymorphism and selection efficiency in the data from those four species of Gallus were calculated. By calculating the qw (Watterson's estimator) of each site, an average qw for each species and the minimum number of re-combinations in each species and by estimating the selection efficiency for G. domesticus and G. gallus, neither significant nucleotide diversity nor genetic-diversity-qw- difference was found between G. domesticus and G. gallus. The results indicated that the patterns of genetic diversity in G. domesticus were strongly influenced by recombination and, because Tajima's D has a negative value, recombination was the main mechanism responsible for the immune gene evolution of G. gallus.

Abstract (sommario):
The responses and tolerances of early-weaned piglets to transport are not well documented. It is believed that the additive stress incurred by transporting early-weaned pigs predisposes them to increased disease risk and compromised performance. The objective of this work was to investigate the relationship between two primary transportation stressors, duration and temperature, and their effects on piglet performance. Prior to housing on flatdecks at 30°C, transport was simulated by placing 17-d (± 1 d) old weaned piglets into wooden boxes of dimensions 1.2 × 1.2 × 1 m (space allowance range 0.18 to 0.36 m2 pig–1) with straw bedding. Two trials were conducted. In each trial, 96 piglets were randomly mixed in groups of four and assigned to the following simulated transport durations: no transport (control), 6 h, 12 h (Trial 2 only) and 24 h. Piglets undergoing transport simulation were kept at one of the following temperatures 20, 25 (Trial 1 only), 30 and 35°C. There was a significant interactive effect between transport duration and temperature upon liveweight change in the first 24 h in both trials (P < 0.01). The animals that incurred the greatest liveweight deficit after weaning relative to untransported control groups were predominantly either those that had been transported for 24 h at high transport temperatures (35 and 30°C) or those transported for 6 h at 20°C in Trial 1 and 35°C in Trial 2. The effect of simulated transport was measureable for up to 5 d post-transport. However, by 14 d post-transport there was no detectable influence of transport treatment on feed consumption or weight gain (P > 0.05). The study found piglets are able to recover and perform adequately in the early post-weaning period if extremes of transport duration and temperature are avoided.
