We explore various aspects of electrochemistry and photoelectrochemistry using in situ spectroscopy of electrode (metal) and photoelectrode (semiconductor) interfaces under electrochemical working conditions. Using sum frequency generation (SFG) spectroscopy, we measure the voltage dependence of the orientation of D2O molecules at a graphene electrode surface, which enables us to extract the free energy orienting potential of interfacial water.1 In particular, we measure the “free OD” feature in the spectra, which corresponds to the topmost water molecule that is rotated up out of the bulk water solution and is, therefore, not hydrogen bonded. Using transient absorption spectroscopy (TAS), we measure the lifetime of hot electrons photoexcited in plasmon resonant nanostructures.5 In a similar study, we use transient reflectance spectroscopy (TRS) to measure the photoexcited carrier dynamics in a GaP/TiO2 photoelectrode, as well as the electrostatic field dynamics at this semiconductor-liquid interface in situ under various electrochemical potentials.2 Here, the electrostatic fields at the surface of the semiconductor are measured via Franz−Keldysh oscillations (FKO). These spectra reveal that the nanoscale TiO2 protection layer enhances the built-in field and charge separation performance of GaP photoelectrodes. Using surface enhanced Raman scattering (SERS) spectroscopy, we monitor local electric fields via Stark-shifts of nitrile-functionalized silicon photoelectrodes.6 By monitoring Stark shifts in graphene-enhanced Raman spectroscopy (GERS), we measure local electric fields and local charge densities at monolayer graphene electrode surfaces.3 Lastly, we measure the stacking dependence of monolayer WSe2/MoSe2 heterostructures and observe resonant excitation of interlayer excitons for photocatalytic energy conversion.4 Montenegro, A., C. In this forum, three leading experts on Sino-Soviet relations and Mao Zedong's policy toward the Soviet Union offer their appraisals of Sergey Radchenko's Two Suns in the Heavens, The Sino-Soviet Struggle for Supremacy, 1962–1967, published by the Woodrow Wilson Center Press. The commentators praise many aspects of Radchenko's book, but Michael Sheng and to a lesser extent Qiang Zhai and Deborah Kaple wonder whether Radchenko has gone too far in downplaying the role of ideology in Mao's foreign policy. Unlike Lorenz Lüthi, who gives decisive weight to ideology in his own book about the Sino-Soviet split, Radchenko argues that a classical realist approach is the best framework for understanding Chinese foreign policy and the rift between China and the Soviet Union. Sheng and Zhai also raise questions about some of the sources used by Radchenko. Replying to the commentaries, Radchenko defends his conception of Mao's foreign policy, arguing that it is a more nuanced view than Sheng and Zhai imply. Radchenko also stresses the inherent shortcomings of the source base scholars are forced to use when analyzing Chinese foreign policy.

Abstract Background Camrelizumab and nab-paclitaxel demonstrated promising anti-tumour activity in refractory metastatic immunomodulatory triple-negative breast cancer (TNBC) in FUTURE trial. Anti-angiogenic agents have been reported to facilitate immune infiltration. Famitinib is a tyrosine kinase inhibitor targeting VEGFR-2, PDGFR and c-kit. The FUTURE-C-PLUS trial (NCT04129996) which added famitinib to camrelizumab and nab-paclitaxel is a single-arm, phase 2 trial evaluating this novel triplet combinatorial strategy in patients with advanced immunomodulatory TNBC. Study design and the primary endpoint ORR has been reported previously (Zhi-ming Shao, et al. ASCO 2021, Abstract 1007). Here, we reported the updated results of this trial. Method Briefly, this study enrolled women aged 18-70 years, with previously untreated, histologically confirmed, unresectable, locally advanced, recurrent or metastatic immunomodulatory TNBC. Immunomodulatory TNBC was defined as CD8 expression on at least 10% of cells using immunohistochemistry analysis. Eligible patients received the triple therapy. Study design has been reported previously in ASCO 2021. Results Between Oct 2019 and Oct 2020, 48 patients were enrolled and treated. 39 (81.3%, 95% CI 70.2-92.3) patients had a confirmed objective response which has been reported in ASCO 2021. At this updating data cutoff (June 30, 2021), the median progression-free survival was 11.9 months (95% CI, 7.3-16.5) with the median follow-up was 14.0 months. While overall survival data were not mature yet, a promising overall survival rate was observed at 12 months (84•2%, 95% CI 73.4-95.0) and 18 months (73.6%, 95% CI 52.0-95.2). In the 39 responders, median duration of response was also not mature. The disease control rate was 95.8% (46/48). The most common treatment-related grade 3 or 4 adverse events were neutropenia (16 [33.3%]), anemia (5 [10.4%]), febrile neutropenia (5 [10.4%]), and thrombocytopenia (4 [8.3%]). No treatment-related deaths were reported. Conclusions These data, combined with those from our previous reports, provide further evidence for the triplet combination of famitinib, camrelizumab and nab-paclitaxel as an active therapy in advanced Immunomodulatory TNBC. To our knowledge, this is the best objective response rate reached in first-line treatment of advanced TNBC. A randomized controlled FUTURE-Super trial (NCT04395989) is keeping recruiting patients to further validate those findings. Abstract Background: Garsorasib (D-1553), a potent KRAS G12C inhibitor, has previously demonstrated promising anti-tumor activity in KRAS G12C-mutated non-small-cell lung cancer (NSCLC) in a phase 1 study. Here we present the efficacy and safety of garsorasib in a pivotal phase 2 study. Methods: In this open-label, multicenter, single-arm phase 2 study, patients received garsorasib 600 mg twice daily in 21-day cycles. Eligible criteria included patients with locally advanced or metastatic NSCLC harboring KRAS G12C mutation, who had disease progression after prior anti-PD-(L)1 therapy and platinum-based chemotherapy or intolerance to the above regimens due to toxicity, and had measurable tumor lesions according to the RECIST, v1.1. The primary end point was objective response rate (ORR) evaluated by independent review committee (IRC) per RECIST v1.1. The secondary end points included duration of response (DOR), disease control rate (DCR), time to response (TTR), progression-free survival (PFS), overall survival (OS), and safety. Results: As of November 17, 2023, 123 patients (108 [87.8%] male, median age 64 [range: 33-80], and 11.4%/88.6% ECOG PS 0/1) were enrolled and treated, with a median follow-up of 7.92 months (range, 0.7, 16.5) and a median duration of treatment of 6.24 months (range, 0.7, 15.9). At data cutoff date, 82 patients (66.7%) discontinued treatment, with disease progression (46 patients [37.4%]) being the most common reason. Among 123 patients, IRC confirmed-ORR was 49.6% (61/123, 95% CI, 40.5 to 58.8) and DCR was 88.6% (109/123, 95% CI, 81.6 to 93.6). The median TTR was 1.38 months (range, 1.12 to 5.55). The median DOR was 12.78 months (95% CI, 6.21 to NE). The median PFS was 7.56 months (95% CI, 5.55 to 9.69), and the median OS was not reached. A total of 117 (95.1%) patients reported treatment-related adverse events (TRAEs) with 61 (49.6%) patients experiencing grade 3 or higher events. The most commonly reported (≥25%) TRAEs (any grade) were increased AST, ALT and γ-GGT, anemia, increased blood bilirubin, and increased blood ALP. No new safety signals were identified, and most of the adverse events were well managed. No TRAE resulted in permanent discontinuation of garsorasib. Conclusions: Garsorasib has shown a high tumor response rate and long response duration in patients with KRAS G12C mutated NSCLC that have progressed after prior anti-PD-(L)1 therapy and platinum-based chemotherapy. Its safety profile was also shown to be well-tolerated and manageable. Garsorasib could be considered as a promising treatment option for KRAS G12C-mutant NSCLC patients with a high unmet medical need. Abstract Background: Intratumor heterogeneity (ITH) refers to variations observed among cancer cells within a single tumor, posing significant challenges in clinical practice due to its contribution to treatment resistance and unfavorable outcome. However, the inconvenience of multi-region biopsy and limitations of genomic sequencing based on limited tissue impede the practical detection of ITH. Consequently, there is an urgent need for a non-invasive method that comprehensively captures ITH from a holistic perspective of the entire tumor. Methods: We utilized a large multicenter dataset comprising dynamic contrast-enhanced magnetic resonance images from breast cancer patients (n = 1423) along with matched multiomics data (n = 468) to develop a non-invasive machine learning approach for measuring ITH. We extracted quantitative radiomic features from both the entire tumor and peritumor regions, with a specific focus on features associated with imaging heterogeneity. Using these radiomic features, we established an imaging ITH (IITH) model. The robustness of IITH evaluation was determined by assessing its correlation with genomic ITH (employing the mutant-allele tumor heterogeneity [MATH] algorithm) and pathological cellular ITH (analyzing variation in quantitative nuclear features extracted through digital pathology). Prognostic power was evaluated using Kaplan-Meier and multivariate Cox proportional hazards analyses. Integrated multiomics analyses were performed to investigate the molecular basis of different IITH subgroups. Results: We developed a non-invasive radiomic signature to quantify imaging ITH (IITH) in the FUSCC cohort. Breast cancer patients were retrospectively categorized into high and low IITH groups. Multivariate Cox analysis identified high IITH as an independent predictor of poor prognosis in breast cancer patients, even after adjusting for clinical risk factors such as tumor size, positive lymph nodes, clinical subtype and lymphovascular invasion status. These findings were further validated in the DUKE cohort, confirming the prognostic value of IITH. Moreover, we substantiated the robustness of IITH by demonstrating its association with genomic and pathological ITH. Multiomics analysis revealed the activation of oncogenic pathways and metabolic dysregulation in high-IITH tumors. Intriguingly, our investigation also highlighted ferroptosis as a vulnerability and potential therapeutic target in high-IITH tumors, which was further supported by evidence from the TCGA cohort. Conclusion: Radiomic-based assessment of ITH provides a non-invasive approach to comprehensively capture ITH and predict the prognosis of breast cancer patients. Targeting ferroptosis may hold promise as a treatment strategy for patients with high IITH. On the basis of understanding the indexes of different tributaries of Bai Ta Pu river and according to the contribution rate of each principal component, the comprehensive score of different tributaries was calculated through principal component analysis. Each tributary was ranked according to the score, and the evaluation results were analyzed. It is pointed out that the tributary with the highest comprehensive index of Bai Ta Pu river is in Shi Jia Zhai bridge and the tributary with the lowest comprehensive index is in De Sheng Tun bridge.

In 2022, the insect inventories organized by Prof. Zhi-Sheng Zhang of Southwest University were constructed in Yintiaoling Nature Reserve, Chongqing Municipality, China. The neopseustid moth from Chongqing based on three specimens of Neopseustis archiphenax by light trapping were reported in this paper. The photos of the male and female adults, genitalia, and abdominal special structures are presented with the female described firstly. Also, the DNA barcoding sequence data is provided, and the key to the Neopseustis species is given.

The Old World genus Mesocomys Cameron (1905) (Hymenoptera: Eupelmidae: Eupelminae) is revised. Eleven species, including two newly described species, are recognized and keyed in two previously established species groups, the albitarsis and the pulchriceps species groups sensu Gibson (1995), but with additional features provided to distinguish members of the two groups. Five species are recognized in the pulchriceps group—Mesocomys anelliformis n. sp., M. longiscapus n. sp., M. orientalis Ferrière, 1935, M. pauliani Ferrière, 1951, and M. pulchriceps Cameron, 1905. Seven species are assigned to the albitarsis group, but one, M. aegeriae Sheng, 1996 is treated as a nomen dubium; the six recognized and keyed species in the albitarsis group are M. albitarsis (Ashmead, 1904), M. breviscapis Yao, Yang & Zhao, 2009, M. menzeli (Ferrière, 1930b), M. obscurus (Ferrière, 1930b) revised stat., M. superansi Yao, Yang & Zhao, 2009, and M. trabalae Yao, Yang & Zhao, 2009. Within the albitarsis group, the species are further discussed relative to two newly established species subgroups, the albitarsis subgroup for M. albitarsis, M. menzeli and M. obscurus, and the aegeriae subgroup for M. aegeriae, M. breviscapis, M. superansi and M. trabalae. Females of the albitarsis subgroup possess a finely sculptured mesoscutal medial lobe in combination with partly infuscate fore wings and/or at least partly pale flagellum, whereas females of the aegeriae subgroup possess a much more coarsely sculptured mesoscutal medial lobe and hyaline fore wings in combination with a dark flagellum. Members of the albitarsis species group are restricted to the Oriental and eastern Palaearctic regions except for a single female of the aegeriae subgroup seen from Algeria that is provisionally identified as M. breviscapis; members of the pulchriceps group are restricted to the Afrotropical region except for M. orientalis from the Oriental region. Newly placed in synonymy are M. aegeriae Sheng, 1998 under M. aegeriae Sheng, 1996 n. syn., M. sinensis Yao, Yang & Zhao, 2009 under M. breviscapis Yao, Yang & Zhao, 2009 n. syn., M. atulyus Narendran, 1995 under M. orientalis Ferrière, 1935 n. syn., M. vuilleti (Crawford, 1912) under M. pulchriceps Cameron, 1905 n. syn., and Semianastatus orientalis Kalina, 1984 and Mesocomys kalinai Özdikmen, 2011 under M. albitarsis (Ashmead, 1904) n. syns. Lectotypes are newly designated for M. menzeli, M. obscurus, M. orientalis, M. pauliani, M. pulchriceps and M. vuilleti. Morphological features characteristic of the genus and of the highly dimorphic sexes are described and illustrated, and the species are keyed, described, and illustrated through macrophotography. Phylogenetics are discussed for the genus, the two species groups, and species within the pulchriceps group. Distribution and host records are also summarized for each species.

Abstract Background: Anti-HER2 targeted therapy has achieved a series of breakthroughs. However, the current treatment strategy regarding HER2-positive breast cancer remains indiscriminate and lacks specificity, which limits the further improvement of overall treatment response and may lead to overtreatment and extra cost for some patients. Our study aims to reveal the molecular heterogeneity of HER2-positive breast cancer to guide a more precise treatment. Patients and methods: We selected HER2-positive breast cancer patients treated at Fudan University Shanghai Cancer Center between 2013 and 2014 and conducted genomic, transcriptomic, proteomic and metabolomic profiling. We then applied a non-negative matrix factorization algorithm on transcriptomic data to obtain an unsupervised classification. And we further studied the correlation between subtypes and corresponding treatment strategies in multiple cohorts of adjuvant and neoadjuvant therapy. For clinical accessibility, we developed convolutional neural network models through deep learning algorithm based on digital pathology to identify different subtypes. Additionally, we explored novel treatment strategies using the patient-derived organoids (PDOs) models. Results: We established a novel multiomics cohort of 180 HER2- breast cancer patients and classified them into four clinically significant molecular subtypes: (1) A classical HER2-enriched (HER2-CLA, N=51) subtype characterized by strong ERBB2 signaling and remarkable sensitivity to anti-HER2-targeted therapy (pathologic complete response with dual-targeted therapy: 93%). (2) an immunomodulatory (HER2-IM, N=36) subtype characterized by an immune-activated microenvironment and excellent prognosis with current treatment (no relapse in 97% of patients with a median follow-up of 86 months). Tumors of this subtype were therefore candidates for de-escalatory treatment. (3) A luminal-like (HER2-LUM, N=55) subtype distinguished by activated estrogen receptor signaling and (4) a basal/mesenchymal-like (HER2-BM, N=38) subtype enriched in activated receptor tyrosine kinase pathways. HER2-LUM and HER2- BM showed limited benefit from anti-HER2 therapy, and thus, add-on therapies might be needed. The overall area under the curve (AUC) of the convolutional neural network model based on digital pathology for identifying different subtypes is 0.77. In the exploration of novel treatment strategies, we found in the PDO model that the HER2-LUM subtype is more sensitive to a treatment regimen combining standard (chemotherapy and targeted therapy) with subsequent endocrine therapy and CDK4/6 inhibitors compared to other subtypes. Additionally, the HER2-BM subtype demonstrated greater sensitivity to treatment with a combination of EGFR inhibitors, PDGFR inhibitors or VEGFR inhibitors. Conclusion: We uncovered a high degree of molecular heterogeneity in HER2-positive breast cancer and illustrated its impact on treatment response. More precise treatment can be given according to the characteristics of different subtypes, which may achieve good efficacy and simultaneously reduce overtreatment and extra cost. The comprehensive profiling of HER2-positive breast cancers could also serve as an important resource for further exploration. This paper studies new solitary waves for the Klein–Gordon–Zakharov equations. The obtained results are diverse and some specific ones emerge as dark, bright and bell-shape. The two integration schemes lead to obtaining waves which propagate without deformation as illustrated in graphical representations. The present work is a continuation of two previously published articles in which the biography of Zhao Rugua 趙汝适 (1170-1231), the author of Zhufan zhi 諸蕃志 (Records of Foreign Peoples; 1225) an important historical geographical source about the foreign lands during the Song dynasty (960-1279), was studied through the usage of the text of his funeral epitaph as well as other historical sources. They also included the translation of the text of inscription into Russian and English [1; 2]. The goal of this article is to deepen the study of Zhao Ruguas funeral epitaph by researching the material features of the source, analyzing the structure of the text and comparing it with other similar sources of Medieval China. In contrast with the previous works the inscription in this article is analyzed both as a textual source and as an object of material culture which enables to understand the context of the creation of the source as well as the extent of detalization of the deceaseds biography.

This study aimed to investigate the therapeutic effect of a traditional Chinese medicine on the treatment of painful leg ulcer. Twelve cases of painful leg ulcer were recruited and their characteristics (etiology, size, Visual Analog Scale score, and duration) were recorded. Then, an herbal medicine–oriented treatment protocol combining Zhi Tong Sheng Ji powder (ZTSJ) and SJXP (Sheng Ji Xiang Pi plaster) was applied to these patients. The results suggested that this therapeutic protocol contributed to pain relief and promoted wound healing simultaneously. Specifically, 2 days after applying ZTSJ on wounds, the intensity of pain reduced substantially, and the difference between pre- and post-treatment Visual Analog Scale scores (46 mm vs 23 mm, P < .001) was statistically significant. Epithelialization varied according to ulcer depth. If the wound was deep to the fascia, islands of epithelia did not form within wounds, but rather formed at the edge of wounds, generating island of epithelial cells (IECs) or newly formed epithelial tissue. If the wound was superficial and the dermis remained, scattered IECs appeared within ulcers and merged eventually. Furthermore, as wound pain decreased, IECs appeared, expanded, and merged, or epithelial proliferation and migration occurred in wound margin, eventually healing wounds. No infection spread, topical rashes, itching, or other allergies occurred in these 12 cases, and the time for recovery ranged from 16 to 163 days. ZTSJ is effective for the treatment of painful leg ulcers, but the specific mechanism is not clear.

Clara Law (Law Cheuk-yiu) has amassed a substantial body of work as a writer and/or director, with upward of twenty feature credits between 1985 and 2015. Starting with her early Hong Kong–based films addressing sexuality, The Reincarnation of Golden Lotus (Pan Jin Lian zhi qian shi jin sheng, 1989) and Temptation of a Monk (1993), Law embraced transnational cinema through invitations and awards from major international film festivals, as well as participating in Erotique (1994), a compilation film with multinational contributors. Her work insistently interrogates the interconnections among concepts of Asia, transcultural migrations, and filmmaking practice through narratives that exceed “Chineseness” both in terms of culture and ethnicity, and in terms of expectations for Asian women directors. Reconfigurations of sexual and cultural identity, and geopolitical correspondences and differentiations, consistently recur in her filmic narratives, as characters encounter new personal, cultural, racial and ethnic formations in the wake of geopolitical transmigrations.

Much attention has been focused on the recrudescence of Chinese nationalism in the 1990s. The PRC government generally denies, but occasionally defends, the existence of nationalism within China (Xi 1996). Many Chinese scholars acknowledge the phenomenon and debate its impact, with most evincing a positive attitude toward nationalism (Zhang Xudong 1997; Xiao 1997; Sheng 1996; Wen 1996; Li 1995). Outside the PRC, Chinese nationalism is increasingly scrutinized (Zhao 1997; Zhu 1997, Gries 1999). Some hold that it is rational and manageable (Nathan and Ross 1997; Metzger and Myers 1998; Zheng 1999), while others speak in more premonitory tones (Chang 1998; Su 1997; Mosher 2000). All conclude that it is statesponsored, popular, and fills an “ideological vacuum” left by the waning of socialism (see, for example, Oksenberg 1997; Christensen 1996).

Abstract This article presents a historical exploration of the concept of qili qiji 契理契机 (according with the [core] principles [of Buddhism] and the circumstances or capacities [of sentient beings]) in the context of Buddhist thought from the Eastern Jin era (317–420 CE) to contemporary times, examining its origins, development, and dissemination, and the progressive expansion of its definition. The study explores the significance of qili qiji in the writings and discussions of influential figures and organizations in the domain of “Buddhism for the Human Realm” (renjian fojiao 人间佛教), such as Taixu 太虚, Yin Shun 印顺, Sheng-yen 圣严, Hsing Yun 星云, Zhao Puchu 赵朴初, and the Buddhist Association of China. Lastly, the study investigates the adoption of this term beyond the Buddhist community, specifically its incorporation into the official discourse of governmental religious affairs departments and its usage within other religions such as Daoism.

Background.Traditional Chinese medicine (TCM), when given as a therapy for symptom relief, has gained widespread popularity among diabetic patients. The aim of this study is to analyze the utilization of TCM among type 2 diabetic patients in Taiwan.Methods.The use of TCM for type 2 diabetic patients were evaluated using a randomly sampled cohort of 1,000,000 beneficiaries recruited from the National Health Insurance Research Database.Results.Overall, 77.9%(n=31,289)of type 2 diabetic patients utilized TCM and 13.9%(n=4,351)of them used TCM for the treatment of type 2 diabetes. Among the top ten most frequently prescribed herbal formulae, four remedies,Zhi-Bo-Di-Huang-Wan, Qi-Ju-Di-Huang-Wan, Ji-Sheng-Shen-Qi-WanandBa-Wei-Di-Huang-Wanare derivative formulae ofLiu-Wei-Di-Huang-Wan.In other words,Liu-Wei-Di-Huang-Wanand its derivatives were found to be the most common herbal formulae prescribed by TCM doctors for the treatment of diabetes in Taiwan.Conclusion.Although some evidence does support the use TCM to treat diabetes, the results from the current study may have been confounded by placebo effect, which emphasize the need for well conducted, double-blind, randomized, placebo-controlled studies in order to further evaluate the efficacy ofLiu-Wei-Di-Huang-Wanon patients with type 2 diabetes.

Abstract The overexpression of ATP-binding cassette (ABC) transporters has known to be one of the most important mechanisms responsible for the development of multidrug resistance (MDR). OTS964 is a potent T-LAK cell-originated protein kinase (TOPK) inhibitor. Herein, we investigated the interaction of OTS964 and MDR-associated ATP-binding cassette sub-family G member 2 (ABCG2, breast cancer resistance protein/BCRP). The cell viability assay indicated that the effectiveness of OTS964 is limited in drug-resistant and gene-transfected cells overexpressing ABCG2. We found that the known ABCG2 inhibitor is able to sensitize ABCG2-overexpressing cells to OTS964. In mechanism-based studies, OTS964 shows inhibitory effect on the efflux function mediated by ABCG2, and in turn, affects the pharmacokinetic profile of other ABCG2 substrate-drugs. Furthermore, OTS964 upregulates ABCG2 protein and mRNA expression levels, resulting in enhanced resistance to ABCG2 substrate-drugs. The ATPase assay demonstrated that OTS964 stimulates ATPase activity of ABCG2 in a concentration-dependent manner, and that this stimulation can be antagonized by a verified ABCG2 ATPase inhibitor. Additionally, the computational molecular docking analysis combined with results from ATPase assay suggested that OTS964 interacts with drug-binding pocket of ABCG2 protein and has substrate-like behaviors. Thus, OTS964 is an MDR-susceptible agent due to its interactions with ABCG2, and overexpression of ABCG2 transporter may attenuate its therapeutic effect in cancer cells. Abstract Resistance to chemotherapies such as temozolomide presents a major hurdle to effectively treat glioblastoma. This challenge arises possibly from the activation of phosphatidylinositol 3-kinase (PI3K) signaling, which makes PI3K an appealing therapeutic target. However, non-selectively blocking the four highly homologous PI3K catalytic kinases p110α, β, δ, and γ has yielded undesired clinical outcomes. It is therefore imperative to investigate individual PI3K kinases in glioblastoma’s chemosensitivity. Here we report that PI3K kinases differentially regulate chemosensitivity. We found that the four kinases were unequally expressed in grade II/III glioma and glioblastoma with levels of p110β being the highest. Concomitantly, glioblastoma patients deficient of O6-methylguanine-DNA-mehtyltransferase and expressing high levels of p110β and low levels of other kinases (p110β-high) were more likely resistant to temozolomide, thereby experiencing poor prognosis. Perturbation of p110β, but not other kinases, sensitized p110β-high glioblastoma cells or mouse xenograft tumors to temozolomide. Moreover, p110β-selective inhibitors and temozolomide synergistically mitigated the growth of resilient glioblastoma stem cells. Our results have demonstrated the essential role of p110β in regulating chemosensitivity, suggesting that selectively blocking p110β is an effective chemosensitizer for therapy-resistant glioblastoma. Abstract Background: Unlike patients with advanced colorectal cancer (CRC) with deficient DNA mismatch repair or microsatellite instability-high (dMMR/MSI-H) tumors, most patients with advanced CRC with proficient DNA mismatch repair or microsatellite-stability (pMMR/MSS) tumors are not sensitive to immune checkpoint inhibitor (ICI) therapy. The current challenge is to find out innovative therapeutic combinations with ICIs for such patients. Case Presentation: In this study, we report a case of heavily pretreated refractory colon cancer with MSS but high-tumor mutation burden (TMB-H) tumors. After experiencing five lines of therapy, including chemotherapy and targeted therapy with anti-angiogenic inhibitor as well as anti-EGFR antibody, she received sixth-line therapy with combination of Sintilimab, Bevacizumab and chemotherapy, and obtained a durable clinical benefit. The patient’s progression-free survival (PFS) of sixth line therapy was about 9 months and overall survival (OS) has been over 4.6 years. Conclusion: To the best of our knowledge, this study represented the longest PFS of response to combination of ICI with other agents with different mechanisms of action in a heavily pretreated refractory colon cancer patient with MSS. Abstract Polycomb repressive complex 2 (PRC2) catalyze the writing of H3K27me3 epigenetic marker and suppresses the expression of genes including tumor suppressors. Activation mutation of EZH2, the catalytic subunit of the PRC2 complex, drives the progression of tumors such as DLBCL. The embryonic ectoderm development (EED) protein acts as the scaffolding and H3K27me3-binding subunit of PRC2 and is essential for its activity. We have developed potent, selective, and orally bioavailable EED inhibitor BR1733. BR1733 blocks the interaction between EED and H3K27me3, and inhibits catalytic activity of PRC2. BR1733 showed superior PK profile across species and exhibited tumor growth inhibition against several B cell lymphoma CDX models. Indication expansion studies demonstrated that BR1733 could inhibit solid tumors with SWI/SNF loss-of-function mutation only when both CDKN2A and TET1 are expressed. BR1733 also inhibited tumor angiogenesis and showed tumor growth inhibition in several CDX models of highly vascularized solid tumors such as HCC and RCC. BR1733 is currently in Phase I clinical trials. Abstract Pancreatic cancer (PCa) is one of the most lethal malignancies in human cancers. The enhanced infiltration of stromal tissue of PCa tumor microenvironment limits the identification of key tumor-specific transcription factors and epigenomic abnormalities in malignant epithelial cells. Integrated transcriptome and epigenetic multi-omics analysis of paired PCa organoids indicated that the basic helix-loop-helix transcription factor 40 (BHLHE40) was significantly upregulated in tumor samples. Increased chromatin accessibility at promoter region and enhanced mTOR pathway activity contribute to the elevated expression of BHLHE40. Integrated analysis of chromatin immunoprecipitation (ChIP)-seq, RNA-seq and high-through chromosome conformation capture (Hi-C) data together with chromosome conformation capture (3C) assay indicate that BHLHE40 not only regulates the sterol regulatory element-binding factor 1 (SREBF1) transcription as a classic transcription factor, but also links the enhancer and promoter regions of SREBF1 via forming liquid-liquid phase-separated droplets. Then, we find the BHLHE40-SREBF1-stearoyl-CoA desaturase (SCD1) axis protects PCa cells from ferroptosis, leading to the reduced accumulation of lipid peroxidation in cells. Moreover, SREBF1 inhibitor, Fatostatin, significantly suppresses the cell growth of PCa tumors with higher expression of BHLHE40. This study highlights the important roles of BHLHE40 mediated lipid peroxidation in inducing ferroptosis of PCa cells and provides a novel mechanism underlying the SREBF1 overexpression in PCa. Background. Ischemic heart disease (IHD) related to cardiovascular or cerebrovascular disease is the leading cause of mortality and an important issue of public health worldwide. The cost of long-term healthcare for IHD patients may result in a huge financial burden. Objectives. To analyze the medical expenditure incurred for and survival of IHD patients treated with Chinese herbal medicine (CHM) and Western medicine. Methods. Subjects were randomly selected from the National Health Insurance Research Database in Taiwan. The Cox proportional hazards regression model, Kaplan–Meier estimator, logrank test, chi-square test, and analysis of variance were applied. Landmark analysis was used to assess the cumulative incidence of death in IHD patients. Results. We identified 11,527 users of CHM combined with Western medicine and 11,527 non-CHM users. CHM users incurred a higher medical expenditure for outpatient care within 1 (24,529 NTD versus 18,464 NTD, P value <0.0001) and 5 years (95,345 NTD versus 60,367 NTD, P value <0.0001). However, CHM users had shorter hospitalizations and lower inpatient medical expenditure (7 days/43,394 NTD in 1 year; 11 days/83,141 NTD in 5 years) than non-CHM users (11 days/72,939 NTD in 1 year; 14 days/107,436 NTD in 5 years). The CHM group’s adjusted hazard ratio for mortality was 0.41 lower than that of the non-CHM group by Cox proportional hazard models with time-dependent exposure covariates. Danshen, Huang qi, Niu xi, Da huang, and Fu zi were the most commonly prescribed Chinese single herbs; Zhi-Gan-Cao-Tang, Xue-Fu-Zhu-Yu-Tang, Tian-Wang-Bu-Xin-Dan, Sheng-Mai-San, and Yang-Xin-Tang were the five most frequently prescribed herbal formulas in Taiwan. Conclusions. Combining Chinese and Western medicine can reduce hospital expenditure and improve survival for IHD patients.

Abstract Tumor-infiltrating lymphocytes (TILs) has demonstrated significant clinical benefits for patients with solid tumor. A robust expansion process is needed in order to produce a large number of cells for clinical usage. We modulated the PI3K-AKT signaling pathway in TILs isolated from cervical and ovarian cancer patients and found the inhibition of either PI3K or AKT can promote TIL proliferation during the REP stage. Inhibiting PI3K-AKT led to an increase of the effector CD8+T cell population with higher CD25+ and CD28+ in the CD8+ population, more TCF-1+ and CD39-CD69- T cells in both CD4+ and CD8+ T cell population, and downregulated exhaustion markers CD39+ and TIM-3+ in CD8+ T cell population. This inhibition significantly increased cytotoxicity when cocultured with tumor cell lines and patient-derived tumor samples. Moreover, dual knockout of ATK1 and ATK2 promoted TIL proliferation, long-term survival and decreased apoptosis of TIL, which were further confirmed by the RNA-seq results. Thus, inhibition of PI3K-AKT signaling represents a promising strategy to generate large numbers of TIL with enhanced stemness, activation and cytotoxicity, and could be used to support adoptive cell therapy to treat solid tumors. Abstract F-box/WD repeat-containing protein 7 (FBXW7) is the substrate recognition component of SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins. FBXW7 recognizes and binds phosphor-degrons within target proteins and capture them to SCF complex for ubiquitination. Know FBXW7 substrates include cyclin-E, DISC1, JUN, MYC, NFE2L1, NOTCH2, MCL1, RICTOR, and NOTCH1 released notch intracellular domain (NICD). FBXW7 is frequently mutated in cancer, results in loss of substrate recognition and binding. R465C is a one of the major FBXW7 mutations and is found in over 70,000 cancer patients worldwide. From a high-throughput screen, we discovered compounds that selectively bind to FBXW7 R465C mutant. PROTACs against various protein targets have been made with these mutant specific ligands. Some of these PROTACs could induce targeted protein degradation in cells carrying FBXW7 R465C mutant but not wild-type FBXW7. Targeted protein degradation with these PROTACs could represent a novel therapeutic approach against FBXW7 R465C mutant cancer. Abstract Photoacoustic (PA) imaging can map the physiological conditions of tissues and track the biodistribution of contrast agents. Ultrasound localization microscopy (ULM) with microbubbles provides deep-tissue super-resolution blood vessel images and blood velocity maps. The integration of these techniques offers a potential tool for oncological applications, such as visualizing tumor hypoxia and vasculature. Additionally, because PA imaging and ULM can share the same ultrasound detector and data-acquisition system, PA images can seamlessly fuse with ULM images without requiring post co-registration processing. However, the acquisition time of ULM is considerably longer (several minutes per frame) compared to PA imaging (10 milliseconds to 100 milliseconds per frame), limiting the simultaneous acquisition of the two imaging modalities. To address this issue, we developed a deep learning model based on U-Net to accelerate the frame rate of ULM using information from sparse microbubble locations and power Doppler images. By enabling fast ULM acquisition, we developed an interleaved PA/fast ULM imaging technique to simultaneously capture photoacoustic and ULM images. Our experiments demonstrated up to a 16 times improvement in acquisition speed with U-Net, utilizing in vivo mouse brain and skin tumor data. The hybrid imaging sequence achieved super-resolution vascular and tissue oxygenation imaging with less than 2 seconds of acquisition time per frame in vivo. The implementation of this rapid dual imaging scheme enables the longitudinal monitoring of 3D tumor microvasculature and hypoxia conditions using a common linear ultrasound transducer. We monitored tumor growth of epidermoid carcinoma (A431) in a mouse model from Day 7 to Day 17 of tumor implantation, and the results reveal a significant decrease in tumor vasculature density and oxygen saturation as the tumor grew larger over time. The results indicate a positive relationship between oxygen saturation and vessel density. Our findings showcase the potential of this innovative technique for advancing deep tissue, high-resolution, multi-biological parameters imaging in oncological research. Abstract Though docetaxel plus cisplatin and 5-fluorouracil (TPF) induction chemotherapy becomes the standard care for locoregionally advanced nasopharyngeal carcinoma (NPC), some patients could not benefit from this therapy. The underlying mechanisms remain unclear. We found that DCAF7 was highly expressed in TPF resistant NPC patients, and promoted the cisplatin resistance and metastasis of NPC cells. Mechanistically, DCAF7 facilitates the interaction between USP10 and G3BP1, resulting in the removal of K48-linked ubiquitylation of G3BP1 at Lys76 mediated by USP10, thus preventing the degradation of G3BP1 through ubiquitin-proteasome pathway, and facilitates the stress granule (SG)-like structures formation. Moreover, knockdown of G3BP1 successfully reversed the SG-like structures formation and oncogenic effects exerted by DCAF7. Importantly, NPC patients with elevated DCAF7 expression exhibited high risks of metastasis, and is associated with a poor prognosis. This study identifies DCAF7 as a pivotal cisplatin resistance gene, and sheds light on the underlying mechanism of TPF resistance in NPC patients. The DCAF7-USP10-G3BP1 axis provides potential therapeutic targets and biomarker for NPC treatment. Abstract Werner helicase (WRN) is a RecQ DNA helicase involved in genome maintenance and DNA damage repair. It has recently been shown to be essential to cancer cells with high microsatellite instability (MSI-H). In MSI-H cancer cells, WRN depletion was shown to induce DNA damage and disrupt mitoses, resulting in a distinct phenotype of enlarged and fragmented nuclei. Here, we present a high-throughput phenotypic screen to identify compounds that selectively induce enlarged nuclei in MSI-H cancer cells. In HCT116, an MSI-H colorectal cancer cell line, approximately 40% of cells exhibited enlarged nuclei when WRN was depleted using siRNA. In contrast, fewer than 10% of control cells displayed this phenotype. In HT29, a microsatellite stable (MSS) cancer cell line, no such difference in nuclear morphology was observed, suggesting that the phenotype is specific to MSI-H cells. We used high content imaging to screen our in-house diversity library of approximately 175,000 compounds in 384-well format. 4,260 compounds induced enlarged nuclei in HCT116 cells, a 2.4% hit rate. After re-screening hits in HCT116 along with two additional MSI-H cancer cell lines, SW48 and SNU-407, and counter-screening in HT29 cells, 144 hits were confirmed to specifically enlarge nuclei in MSI-H but not MSS cells. Using the ADP-Glo Kinase Assay, two of these compounds were confirmed to inhibit WRN ATPase activity, suggesting WRN inhibition as a potential mechanism for their specific effects in MSI-H cells. This screen will also identify compounds which selectively kill MSI-H cells through mechanisms beyond direct inhibition of WRN, including interaction with WRN complex members or other members of DNA damage repair pathways. Validation studies including the assessment of cell viability and DNA damage in a panel of MSI-H and MSS cell lines will be performed. Our study proposes a phenotypic screening strategy for identifying small molecules with the potential to target WRN and uncover additional novel mechanisms for treating MSI-H cancers. Samchulkunbi-tang (SCT, Shen Zhu Jian pi tang in Chinese) is said to have been first recorded by Zheng Zhi Zhun Sheng during the Ming Dynasty in China. Records of SCT in Korea are known to have been cited in Donguibogam (Dong Yi Bao Jian in Chinese), Uibang Hwaltu (Yi Fang Huo Tao in Chinese), and Bang Yak Hapyeon (Fang Yao He Bian in China). Although SCT is widely used in treating chronic gastritis and gastric ulcers, the beneficial effect on renal vascular function is unknown. Hypertension is a risk factor for cardiovascular disease and endothelial dysfunction in humans and experimental animal models of arterial hypertension. In addition, kidney dysfunction is characterized by hypertension diseases. This study was conducted to evaluate the effect of SCT on the vascular function in vitro (human umbilical cord endothelial cells, HUVECs) and in vivo (NG‐nitro‐L‐arginine methyl ester, L-NAME-induced hypertensive rats). The phosphorylation of protein kinase B (Akt) and endothelial nitric oxide synthase (eNOS) is closely related to nitric oxide (NO) production in HUVECs, and SCT in this study significantly increased these. For three weeks, hypertensive rat models were induced by L-NAME administration (40 mg/kg/day) with portable water. It was followed by oral administration with 100 and 200 mg/kg/day for two weeks to confirm the effectiveness of SCT. As a result, systolic blood pressure decreased in the SCT-treated groups, compared with that in the L-NAME-induced hypertensive group. SCT treatment restored vasorelaxation by stimulating acetylcholine and cGMP production in the thoracic aorta. In addition, SCT treatment decreased intima-media thickness, attenuated the reduction of eNOS expression, and increased endothelin-1 expression. It also increased p-Akt and p-eNOS expression in hypertensive rat aorta. Furthermore, regarding renal function parameters, SCT ameliorated urine osmolality, urine albumin level, serum creatinine, and blood urea nitrogen levels. These results demonstrate that the oriental medicine SCT exerts potent vascular and renal protective effects on nitric oxide-deficient hypertensive rats and HUVECs

Traditional Chinese medicine (TCM) divides fracture treatment into three stages. Many TCM herbs and formulas have been used to treat fractures for thousands of years. However, research regarding the Chinese herbal products (CHPs) that should be used at different periods of treatment is still lacking. This study aims to identify the CHPs that should be used at different periods of treatment as well as confirm the TCM theory of fracture periods medicine. We used prescriptions of TCM outpatients with fracture diagnoses analyzed using the Chang Gung Research Database (CGRD) from 2000 to 2015. According to the number of days between the date of the fracture and the clinic visit date, all patients were assigned to one of three groups. Patients with a date gap of 0-13 days were assigned to the early period group; those with a date gap of 14-82 days were assigned to the middle period group; and those with a date gap of 83-182 days were assigned to the late period group. We observed the average number of herbal formulas prescribed by the TCM doctor at each visit was 2.78, and the average number of single herbs prescribed was 6.47. The top three prescriptions in the early fracture period were Zheng-gu-zi-jin-dang, Shu-jing-huo-xue-tang, and Wu-ling-san. In the middle fracture period, the top three formulas were Zheng-gu-zi-jin-dang, Shu-jing-huo-xue-tang, and Zhi-bai-di-huang-wan. In the late fracture period, the top three formulas were Shu-jing-huo-xue-tang, Gui-lu-er-xian-jiao, and Du-huo-ji-sheng-tang. The main single herbs used in the early fracture period were Yan-hu-suo, Gu-sui-bu, and Dan-shen. From the middle to the late period, the most prescribed single herbs were Xu-duan, Gu-sui-bu, and Yan-hu-suo. We concluded that the results showed that the CGRD utilization pattern roughly meets the TCM theory at different fracture periods.

Abstract DDX3X is an ATP-dependent RNA helicase involved in diverse cellular processes. Somatic mutations in the DDX3X gene exhibit oncogenic or tumor suppressive activities depending on cancer types. However, helicase modulating roles of specific DDX3X mutations remain elusive mainly due to lack of an appropriate cell culture model system. Here, we report our development of novel transgenic cell lines capable of interrogating DDX3X helicase activity. To evaluate the helicase activity of the DDX3X mutations in question, we designed a reporter plasmid that measures the luciferase signal corresponding to the helicase activity. Using 8 different point mutations in the DDX3X that we identified earlier in relapsed/refractory patients with diffuse large B cell lymphoma (DLBCL), we show that these DDX3X mutants displayed defective helicase activity. DDX3X mutant knock-in DLBCL cells also displayed abolished helicase unwinding capabilities, validating our reporter cell models. We were able to extend the application of our reporter cells to interrogate Y chromosome-linked male paralog DDX3Y. In male lymphoma patients, ectopic DDX3Y expression can compensate for DDX3X loss to restore global protein synthesis. We narrowed down DDX3Y helicase and ATPase domains to be critical for cellular translation, ruling out DDX3Y involvement in ribosomal elongation. Unexpectedly, DDX3Y helicase is less efficient in RNA unwinding as compared to DDX3X revealing subtle differences despite having a high level of homology. The developed DDX3X reporter cells can be applied widely to evaluate the effects of DDX3X mutations in diseases as well as study the biology of DDX3X and DDX3Y. Background:Treat-to-target (T2T) approach is recommended as a standard management strategy in rheumatoid arthritis (RA) treatment by Chinese guideline for diagnosis and treatment of RA[1]. However, there is little known about its current implementation in China.Objectives:This study aimed to evaluate the implementation and achievement of T2T approach and explore their associated factors in Chinese RA cohort.Methods:A comprehensive cross-sectional survey of rheumatologists and their RA patients was conducted in China. Data were collected during May-Aug 2019 via physician-completed patient record forms. 60 rheumatologists provided data on demographic, clinical characteristics, treatments, and T2T approach implementation for 600 RA patients. Two logistic regressions were used to evaluate factors associated with T2T approach implementation and T2T goal achievement, respectively. Patients with missing data were not included in the models.Results:600 patients were included in this study (48.8±11.7 years, 70.3% female). 39.0% (N=234) of 600 patients were being treated with T2T approach, and 64.9% (N=366) of 564 patients had achieved T2T goal. Patients with longer disease duration (>2 years diagnosis) (odds ratio (OR) [95%CI]=1.61 [1.05, 2.49], vs. diagnosis ≤2 years), higher pain score (OR [95%CI]=1.26 [1.04, 1.51]), or receiving advanced therapy (OR [95%CI]=6.91 [3.64, 13.13]) were more likely to use T2T. Patients with BMI >23.9kg/m2 (OR [95%CI]=2.83 [1.59, 5.04], vs. BMI≤23.9kg/m2), or who worked full-time (OR [95%CI]=2.12 [1.26, 3.57]) were more likely to achieve T2T goal, while patients with more pain (OR [95%CI]=0.77 [0.64, 0.92]) were less likely to achieve T2T goal.Conclusion:Low implementation of T2T approach is observed in Chinese RA treatment. Longer disease duration, more pain, and receiving advanced therapy are associated with higher probability of T2T use, while higher BMI, full-time work and less pain are associated with higher probability of T2T goal achievement. Background: Acute ischemic stroke (AIS) is characterized by high morbidity, disability, mortality, recurrence, and economic burden. Clinical trials have demonstrated that the clinical efficacy of combining oral Chinese patent medicines (CPMs) with chemical drugs (CDs) is better than that of CDs alone. In this study, we performed a network meta-analysis (NMA) of RCTs to assess the efficacy of different CPMs in combination with CDs in the treatment of AIS. Methods: Search 6 databases from the beginning to January 10, 2023. The Cochrane Risk of Bias tool assessed the methodological quality of the included studies. The NMA was then performed using the STATA 13.0 program. The surface under the cumulative ranking curve (SUCRA) probability values were applied to rank the studied treatments, and cluster analysis was used to compare the effects of CPMs between 2 different outcomes. Results: A total of 94 eligible RCTs, involving 9581 participants, were included in this analysis. Nine CPMs, including Nao-mai-li granule (NML), Nao-mai-tai granule (NMT), Qi-long granule (QL), Long-sheng-zhi capsule (LSZ), Nao-xin-tong capsule (NXT), Nao-xue-shu oral liquid (NXS), Tong-xin-luo capsule (TXL), Xiao-shuan-chang-rong capsule (XSCR), and Xue-shuan-xin-mai-ning capsule (XSXMN) were included. Regarding the clinical effective rate, all types of CPMs + CDs treatments were significantly better than CDs treatments alone, with significant differences among the 9 selected CPMs. Compared with CDs, results showed that NXS + CDs performed best in improving clinical effective rate [OR = 4.73; 95% CI: 1.26–17.78; (SUCRA: 76.1%)]. TXL + CDs showed the most effective effect in alleviating National Institutes of Health Stroke Scale (NIHSS) [MD = −3.84; 95% CI: −5.23, −2.45; (SUCRA: 81.6%)]; NXT + CDs were most effective in improving Barthel index [MD = 13.05; 95% CI: 3.98–22.12; (SUCRA: 63.5%)]. Combined with other outcome indicators and the results of cluster analysis, NXS + CDs may assist in the potential optimal treatment regimen for AIS. Conclusion: In conclusion, CPMs were found to be beneficial as adjuvant therapy in patients with AIS. Taking into account the clinical effective rate and other outcomes, NXS + CDs may be the most effective option to improve the condition of AIS patients.

Thermal runaway (TR) reactions of lithium ion cells pose a particular risk to the safety of battery systems. Beside the energy remaining in the cell body, particles and gases exiting the cell during the venting process cause major thermal load for adjacent cells and further system components [1]. In addition, high energy particles can act as an ignition source in the presence of ignitable venting gases and oxygen [2]. Therefore, measurements minimizing the risk of thermal propagation (TP) caused by venting ejecta material and the ignition of combustible gases are introduced on system level. For the dimensioning of the measures, the energy transported by venting particles and gases is identified as a crucial parameter. Depending on the amount of transported energy, the gas cooling distance as well as the complexity of the particle separation structure differ. Simulation methods are used to evaluate the effectiveness of such system measures. More information is needed regarding energy release to accurately predict the system behavior in simulations and to use them as a tool for design. Introducing a novel approach for TR tests enables the determination of the energy remaining in the cell body, separated from the energy transported by venting ejecta. The test setup combines a calorimeter combined with a cyclone separator placed inside an autoclave reactor. The TR energy release of 156 Ah prismatic lithium ion cells have been analyzed using the setup. Venting particles and gases transported a main part of the released energy out of the cell during the TR. Using an analytical approach, the energy transferred to the calorimeter is calculated.
