MADS-box is a key transcription factor regulating the transition to flowering and flower development. Lagerstroemia indica ‘Xiang Yun’ is a new cultivar of crape myrtle characterized by its non-fruiting nature. To study the molecular mechanism underlying the non-fruiting characteristics of ‘Xiang Yun’, 82 MADS-box genes were identified from the genome of L. indica. The physicochemical properties of these genes were examined using bioinformatics methods, and their expression as well as endogenous hormone levels at various stages of flower development were analyzed. The results showed that LiMADS genes were primarily classified into two types: type I and type II, with the majority being type II that contained an abundance of cis-acting elements in their promoters. By screening nine core proteins by predicted protein interactions and performing qRT-PCR analysis as well as in combination with transcriptome data, we found that the expression levels of most MADS genes involved in flower development were significantly lower in ‘Xiang Yun’ than in the wild type ‘Hong Ye’. Hormonal analysis indicated that ‘Xiang Yun’ had higher levels of iP, IPR, TZR, and zeatin during its early stages of flower development than ‘Hong Ye’, whereas the MeJA content was substantially lower at the late stage of flower development of ‘Hong Ye’. Finally, correlation analysis showed that JA, IAA, SA, and TZR were positively correlated with the expression levels of most type II genes. Based on these analyses, a working model for the non-fruiting ‘Xiang Yun’ was proposed. During the course of flower development, plant hormone response pathways may affect the expression of MADS genes, resulting in their low expression in flower development, which led to the abnormal development of the stamen and embryo sac and ultimately affected the fruiting process of ‘Xiang Yun’.

Abstract Chemoradiation (CRT) is widely used to treat locally advanced non-small cell lung cancer (NSCLC) patients, but only around 30% of patients achieve pathological complete response. Here, we utilized an in vitro high-content clonogenic survival screening method to identify CRT sensitizers from the NCI Cancer Therapy Evaluation Program (CTEP) portfolio. Talazoparib (PARPi) was identified among the top candidates to show potent CRT sensitization effects in both KRAS mutant and TP53 mutant NCSLC cell lines. We validated the CRT sensitization effect of PARPi on lung tumors in vivo with cell line xenografts mouse models. To further investigate alterations in the tumor microenvironment (TME) induced by PARPi, we utilized syngeneic mouse models and showed that the combination of radiotherapy, PARPi, and PD-L1 blockade achieved superior tumor control effects. By applying single cell RNA sequencing technologies to the mouse tumors, we found that PARPi combined with CRT or immunotherapy significantly reprogrammed the tumor cells and TME, resulting in the increase of tumor-infiltrating T cells which elicit potent tumor control effects. Our data showed that PARPi sensitizes NSCLC tumors to CRT and immunotherapy through a combination of tumor-intrinsic and extrinsic phenomenon. Abstract Hepatocellular carcinoma (HCC) remains one of the most challenging tumors to treat. Cancer stem-like cells (CSCs) are believed to be important in HCC development, immune evasion, and treatment resistance. Dysadherin is a cell membrane glycoprotein linked to cancer progression by endowing HCC with CSC features. However, the underlying mechanism of increasing CSC characteristics in refractory advanced HCC is not entirely understood. A comprehensive bioinformatics study revealed that dysadherin-expressing HCC cells were associated with cancer stemness and the YAP signaling activation. Dysadherin knockdown or overexpression in liver cancer cell lines attenuated or increased cancer stemness and aggressiveness via YAP signaling, respectively. Correspondingly, dysadherin deficiency lowered cancer stemness and aggressiveness in the HCC mouse model by blocking the YAP pathway. Mechanistically, dysadherin promoted YAP nuclear localization, which activated stem cell transcription factors such as OCT4, KLF4, and MYC. Of note, our comprehensive gain and loss of function studies in CSCs demonstrated that targeting the dysadherin/YAP axis could boost sorafenib’s therapeutic effect and suppress PD-L1 expression, which helps tumor cells escape from immune surveillance. Thus, our results highlight that the dysadherin/YAP axis drives the aggressive phenotype of HCC, providing a potential therapeutic strategy for HCC. Lithium-metal electrodes are of particular interest for next-generation rechargeable batteries because of their high specific capacity and low redox potential. Therefore, Li-metal-based batteries may afford higher energy densities than commercially available Li-ion batteries with graphite anodes. However, various bottlenecks have hampered the commercial development of these batteries, including uncontrolled Li dendrite formation and huge volume changes during cycling. Consequently, Li-metal batteries suffer from low Coulombic efficiency and poor cycling stability. In recent years, there has been extensive research on the design and construction of three-dimensional (3D) porous electrodes that can host metallic Li. However, the low pore utilization and uneven Li plating remain crucial issues. This can be understood in terms of electronic and ionic transport through the framework electrode. The carbon frameworks exhibit high electronic conductance; however, large resistance to Li+ migration in the electrolytes of internal and interparticle pores inhibits the penetration of Li+ deep into the electrode. In this work, we demonstrate that a strong interaction between Li and a lithiophilic nanolayer on a substrate plays a critical role in enhancing pore utilization in carbon framework electrodes. As a model architecture, we examine a Li storage process in a framework electrode consisting of porous carbon derived from metal-organic frameworks (MOFs) and a galvanically displaced Ag layer on a Cu substrate (Cu@Ag). The electrochemical experiments combined with operando XRD measurements and microstructural characterizations suggest that a lithiophilic Ag on the Cu substrate preferentially reacts with Li+ to form Li x Ag during the initial stage of Li plating. This Li x Ag phase acts as a seed that can regulate the subsequent Li plating, promoting confined Li storage in the carbon framework electrode while suppressing top plating. Because of these advantages, the MOF-C framework electrode on Cu@Ag exhibits better cycling stability (>250 cycles) than the MOF-C framework electrode on Cu (140cycles). However, when the thickness of the MOF-C framework is increased to 90 μm, the diffraction peak for Ag remains dominant throughout Li plating-stripping, and the formation of Li x Ag alloys is not clearly detectable in the diffraction patterns, suggesting that only a limited amount of Ag is involved in the alloying reaction with Li+. Based on the computational studies, the efficacy of lithiophilic layers toward improving pore utilization is discussed in terms of the kinetic competition between Li+ transport through porous channels and the interfacial reaction of Li+ with the substrate. This study conveys an important message that the Li-substrate interaction plays a vital role in promoting the confined Li storage; hence, it should be considered a key design factor for porous carbon frameworks with high capacity and long cycle lifetime. Abstract Background: Treatment (tx) for LA SCCHN includes a combination of surgery, radiation and/or chemotherapy followed by monitoring for local recurrence/distant metastases as standard of care. Given poor outcomes, there remains a clear unmet need in this pt population. IMvoke010 (NCT03452137) evaluated the efficacy and safety of atezo in pts with LA SCCHN who are at high-risk for disease progression following multi-modal definitive tx. Methods: Eligible pts with LA SCCHN (Stage IVa or IVb involving the oral cavity, larynx, hypopharynx, HPV negative oropharynx or Stage III HPV positive oropharynx [per AJCC 8th edition]) with no disease progression after multi-modal definitive tx were randomized (1:1) to receive atezo 1200 mg or placebo. Tx was given every 3 weeks for 1 year or until disease progression, unacceptable toxicity or consent withdrawal. Primary endpoint: investigator-assessed event-free survival (INV-EFS). Other key endpoints: overall survival (OS, secondary for efficacy) and safety. Results: A total of 406 pts were randomized to receive either atezo (n=203) or placebo (n=203). 156 (38.4%) pts underwent surgery as part of definitive tx. At clinical cutoff (27 September 2023), median follow-up was 46.5 months (mos). Median INV-EFS was 59.5 mos with atezo vs 52.7 mos with placebo (HR, 0.94; 95% CI, 0.70-1.26). INV-EFS results were generally consistent across all subgroups. OS showed no difference between arms for atezo vs placebo. Safety data are reported (Table). Conclusion: This study did not meet the primary endpoint of INV-EFS, a numerical improvement in INV-EFS was observed for atezo in pts with LA SCCHN, but this was not statistically significant. Atezo was generally well tolerated, and no new safety signals were identified. The genus Haplotropis Saussure, 1888 belongs to family Pamphagidae, superfamily Acridioidea. It contains five species: Haplotropis brunneriana Saussure, 1888; Haplotropis xiai Ye, Shi & Li 2016; Haplotropis zhuoluensis Ye, Shi & Li 2016; Haplotropis aqiensis Zhang, Lin et Yin, 2018 and Haplotropis shandongensis Zhang, Yin et Liu, 2019 distributed mainly in Amur River region of Asia and China only (Saussure, 1888; Bey-Bienko et Mishchenko, 1951; Otte, 1994; Yin, Shi et Yin, 1996; Storozhenko & Paik, 2011; Yin et al, 2014; Dong et al, 2015; Ye et al, 2016; Zhang et al, 2017; Zhang et al, 2018; Zhang et al, 2019; Cigliano et al, 2019). A new species Haplotropis heiheensis sp. nov. is described in this paper. The new species is similar to Haplotropis aqiensis Zhang, Lin et Yin, 2018, but differs from latter by Krauss’ organ of male without ramified keel; tegmine narrower, length 2.4 times width; cercus of male gently tapering at apical half part; minimum width 1.4 times length in mesosternum and epiphallus with 36 spikes. Type specimens are deposited in the College of Plant Protection, Shandong Agricultural University, Taian, Shandong, China.

It contains three species: Haplotropis brunneriana Saussure, 1888; Haplotropis xiai Ye, Shi & Li 2016 and Haplotropis zhuoluensis Ye, Shi & Li 2016 distributed mainly in Amur River region of Asia and China only (Saussure, 1888; Bey-Bienko et Mishchenko, 1951; Otte, 1994; Yin, Shi et Yin, 1996; Storozhenko & Paik, 2011; Yin et al, 2014, Dong et al, 2015; Ye et al, 2016; Zhang et al, 2017; Cigliano et al, 2018). A new species Haplotropis aqiensis sp. nov. is described in this paper. The new species is similar to H. xiai Ye et al., 2016, but differs from latter by tegmina of male extending over the hind margin of first abdominal tergite slightly, length of tegmina 1.7 times width, cover 2/3 tympanum; frontal ridge of male not widened at median ocellus; length interspace of mesosternum in male equal narrowest width; apical half part of cercus suddenly tapering in male and lower margin of epiphallus without projection in the middle. Type specimen is deposited in the College of Plant Protection, Shandong Agricultural University, Taian, Shandong, China.

Objective: We aimed to show whether the antioxidant properties of pomegranate juice (PJ) show a protective effect against renal damage associated with oxidative stress caused by Extracorporeal Shock Wave Lithotripsy (ESWL) in patients who have undergone ESWL for kidney stones. Material and Methods: Ninety patients who were to undergo ESWL for kidney stones were included in the study and were divided into 3 groups with 30 patients each ESWL. Group 1 (control group) patients received only ESWL treatment. Group 2 (study group) patients were recommended to take 30 ml/kg/day pomegranate juice mixture orally for 2 days before ESWL. Group 3 (placebo group) patients were recommended to take 30 mg/kg/day water orally for 2 days before ESWL. All ESWL-treated patients received 2000 to 2500 shock waves at 18 to 24 in order to measure lipid peroxidation markers showing the DNA lesion associated with oxidative stress and to show tubular damage after ESWL, Results: There was a statistically significant mean increase of 52.39± 10.49 in 8-Hydroxy deoxyguanosine (8-OHdG) levels after treatment compared to pre-treatment levels in Group 1 (p<0.01). There was a statistically significant mean increase of 13.16±7.71 and 33.24±12.82 in Groups 2 and 3, respectively (p<0.01; p<0.01, respectively) There was a statistically significant mean increase of 19.20±5.21 in N-acetyl-beta-D-glucosaminidase (NAG) levels after treatment compared to pre-treatment levels in Group 1 (p<0.01). There was a statistically significant mean increase of 3.44±5.21 and 13.07±5.50 in Groups 2 and 3, respectively (p<0.01; p<0.01, respectively). Conclusion: Our study concluded that pomegranate juice (PJ) has protective effects against ESWL-related renal damage. Keywords: Antioxidant, kidney stones, lithotripsy, pomegranate juice.

Abstract Background: Colorectal cancer (CRC) remains one of the most commonly diagnosed cancers and a leading cause of cancer-related deaths worldwide. Between 3 and 6% of all CRC cases are attributed to hereditary colorectal cancer (HCRC). It is important to distinguish CRCs that arise hereditarily from those that arise sporadically, because patients with HCRC and their relatives can benefit from intensive clinical management and surveillance. The detection of germline variants using tumor tissue and paired non-cancerous tissue/whole blood cells (WBC) (conventional method) is considered as the gold standard for germline confirmation, but which could introduce high cost and long turnaround time. In this work, we aimed to investigate the feasibility of diagnosing HCRC using only tumor next-generation sequencing data without normal tissue/WBC data. Methods: One-hundred patients with suspected HCRC were prospectively enrolled according to the clinical diagnosis. Both tumor tissue and paired WBC were collected from each patient. Capture-based targeted sequencing using a panel consisting of 41 cancer-related genes was performed on both tumor tissue and WBC sample. The germline variants were identified using conventional and ColonCore method, respectively. The sensitivity and positive predictive value (PPV) of ColonCore method in identifying germline alterations were calculated, when conventional method was used as a reference. Results: A total of 636 alterations were identified using ColonCore method, including 606 somatic alterations and 30 germline alterations. Among the somatic alterations detected from this panel, 599 single-nucleotide variants and 7 copy number variants were observed. The most frequently mutated genes were APC, TP53, and KRAS, occurring in 76%, 66%, and 46% of patients, respectively. For ColonCore analysis, 17, 9, 2, and 2 patients harbored germline pathogenic variants in DNA mismatch repair (MMR) genes, APC, MUTYH, and ATM, respectively. Compared to the conventional method, the sensitivity of ColonCore method in the diagnosis of HCRC achieved 88.5% with a PPV of 82.1%. Next, the diagnostic performance of ColonCore method in Lynch syndrome (LS) and familial adenomatous polyposis (FAP) was subsequently investigated. We found its sensitivity in the diagnosis of LS achieved 100% (with a PPV of 98.8%), which was obviously higher than that in the diagnosis of FAP (70.0% with a PPV of 63.6%). Twenty-three patients identified as having a microsatellite instability (MSI)-high tumor using ColonCore method were also identified as having an MMR-deficient (dMMR) tumor using immunohistochemistry staining analysis. Conclusions: Our study suggests that ColorCore method is a one-stop approach for detecting germline/somatic alterations and MSI/dMMR status for suspected HCRC. This thesis seeks to examine the extent of the role that the Industrial Technology Research Institute (ITRI) played in Taiwan's high-technological development and whether developing countries of today can promote such development by creating similar institutional arrangements. Literature on innovation systems was reviewed, particularly national innovation systems and the role of R&D institutions within these. Taiwan's recent economic success, deemed attributable to economic and institutional reforms in recent decades, was also studied. In depth analysis was carried out of its leading high-technological research institute, ITRI, which bridges the gap between industry and academia. Although the case of Taiwan is sometimes presented as a unique example of industrial success of an SME-based state, this thesis argues that this success was possible because the research and development process had a large institute at its core. One way of creating such a research scale is by merging existing institutes, a process that would result in more efficient use of capital and human resources. The case of high-technological development in Pakistan is briefly assessed in order to gauge how its existing institutions structure could be amended to allow such changes to be made. The study concludes with the following three main points: (i) scale is an important factor: Taiwan's SME-based industry was able to succeed because of a large research institute at its core; (ii) in developing countries, governments decide which form of high-technology to pursue and when; thus, timing and choice of sector are important; and (iii) political leadership was seen to be important in the case of Taiwan's development in high-technology, and can play a key role in developing countries of today.

This thesis is a study of Yuan ye, a Chinese treatise on garden design written by Ji Cheng and published in or after 1635. Although not prominent in Chinese discourse on gardens during the 17th to the 19th centuries, Yuan ye became an indispensable classic for many scholars of Chinese gardens in the 20th century. This thesis develops a path of thinking that enhance our understanding of Yuan ye in three ways: first, by articulating an interdisciplinary field of discussion that shows how recent developments in the fields of architecture, landscape architecture and comparative philosophy have opened up significant possibilities for re-reading Yuan ye. Second, by offering nuanced readings of selected passages from Yuan ye that go beyond conventional readings of this treatise that take the form of content summary. And, third, by considering Yuan ye in relation to a range of Chinese texts from the 13th to the 20th century that have been selected with two concerns in mind: movement and stillness in the experience of landscape design, and the formation of the cultural memory of gardens.

The purpose of the research is to outline the structure of the main methodological ideas within the frames of interpretive thinking in the essay of the famous Vistnyk’s writer, critic and essayist Yevhen Malaniuk. Considering the purpose and tasks of the studio, an interdisciplinary methodological base, related to the author’s “national approach”, has been worked out. The epistemological potential of national philosophy as a philosophy of national existence, national science as a theory of nation, hermeneutics as a theory and practice of interpretation and post-colonialism as interpretation of cultural phenomena from the standpoint of anti- and post-imperial consciousness are used in the work. The scientific novelty is that on the basis of the previous hermeneutic generalization and definition of national-existential methodology, a propaedeutic outlining of the structure of national-philosophical concepts within the frames of the essayistic interpretation of reality in Ye. Malaniuk is proposed. In the methodological sense, the writer’s essayism is structured by such concepts as nation-centrism, idealism, voluntarism, heroism, and can be considered as one of the variants (close by the experiences of D. Dontsov, Yu. Lypa, M. Mukhyn, etc.) of the Vistnyk’s national-philosophical (national-existential, nationalistic or nation-centric) hermeneutics, that is, the way of understanding, which the author by himself outlined as a “national approach”. The support of Ye. Malaniuk as a culture-philosopher and exegete on the eternal nation-centric values and criteria in his essayistic studies makes his reflections not only historically interesting, but also theoretically productive, classically important for the development of modern Ukrainian hermeneutics and humanities in general.
