Résumé :
The Hippo pathway is a master regulator of cell proliferation and organ size, namely through regulation of transcriptional co-activators YAP and TAZ which bind TEAD1-4 transcription factors. The Hippo effector YAP is dysregulated in many human solid tumours including rhabdomyosarcoma and oesophageal cancer. Additionally, persistent hyperactivity of YAP in activated but not quiescent satellite cells can give rise to embryonal rhabdomyosarcoma. However, the question of exactly how YAP acts as an oncogene and actively gives rise to tumour progression in these cancers remains unknown. In this thesis I characterised the mechanisms which determine the functional role of YAP in driving instability in the genome. Secondly, lentiviral mediated knockdown of YAP is performed to determine and investigate its effect on tumorigenesis. Thirdly, gene sets from constitutive YAP S127A induced mouse ERMS tumours subjected to array-CGH were further analysed. Finally, I cloned chicken Yap1, Tead1 and Fstl5 to identify its role during chick embryonic development, by the retroviral mediated loss of function approach. The results demonstrated that constitutive YAP S127A expression in-vitro as well as in-vivo induces chromosomal instability by increasing the rate of mitotic chromosome segregation errors and copy number alterations of oncogenes and other cancer related genes. Recurrent copy number gains of the p53 inhibitor Mdm2 were observed in YAP S127A-driven ERMS tumours. Moreover, lentiviral mediated YAP knockdown showed significant reduction in proliferation, migration and invasion as well as transformation potential in human cultured cancer cells. Moreover, retroviral YAP S127A expression during early stages of chick embryo development did not lead to an overt phenotype and showed poor survival. Additionally, I have cloned RCAS-RNAi vectors to study the loss of function effect on Hippo targets and Fstl5 during chicken embryo development. Collectively, my data provides insight into the mechanisms with which YAP could drive tumorigenesis and that YAP knockdown can be considered a potential therapeutic target to reduce cancer progression.

A capacidade dos organismos em alterar os seus padrões de expressão de genes em resposta a perturbações do meio ambiente é essencial para a sua viabilidade. Neste trabalho, utilizando Saccharomyces cerevisiae como organismo eucariota modelo, foi estudada a relevância do factor de transcrição da família Yap, o Yap1, na desintoxicação do excesso de cobalto. Os resultados obtidos neste trabalho demonstram que, após a incubação das células com cobalto, o factor de transcrição Yap1 é acumulado no núcleo, através de um mecanismo independente de Orp1/Gpx3. Verificámos que o cobalto tem a capacidade de ligar-se directamente ao Yap1, sugerindo que, à semelhança do que acontece com outros metais, seja este o mecanismo de retenção nuclear após o stress. Confirmámos que uma vez no núcleo, o Yap1 medeia a indução de genes envolvidos na regulação do stress oxidativo em levedura (tais como GPX2, SOD1, TRR1 e TRX2) e constatámos que a presença de cobalto aumenta os níveis de carbonilação das proteínas, sendo esse aumento mais acentuado na ausência de Yap1. Verificámos ainda que os níveis de cobalto estão levemente diminuídos no mutante yap1 ao contrário do ferro que está em concentrações mais elevadas neste mutante relativamente à estirpe selvagem, quer na presença como na ausência de cobalto. Este facto leva-nos a propor que o cobalto altere a homeostase do ferro e que em consequência, conjuntamente com a diminuição das defesas anti-oxidantes, gere stress oxidativo. Foi descrito que a glutationa (GSH) é importante na homeostase do ferro e o excesso de cobalto leva ao aumento dos níveis de ferro intracelulares. Verificámos não só que a expressão de GSH1 está diminuída aos 30 minutos de tratamento do mutante yap1 com cobalto mas também que a adição de GSH faz recuperar os fenótipos de crescimento na presença deste metal. Finalmente apresentámos um modelo de acção do factor Yap1 no stress provocado pelo cobalto.
The ability of organisms to reprogram gene expression in response to changes in the environment is essential for their viability. In this work, using Saccharomyces cerevisiae as a eukaryotic model organism, we studied the relevance of the transcription factor, Yap1, in the detoxification of cobalt excess. The results here reported show that after incubation of cells with cobalt, Yap1 accumulates in the nucleus, through a mechanism independent of the peroxidase Orp1/Gpx3. We found that cobalt has the ability to directly bind to Yap1, suggesting that this is the mechanism for its nuclear retention after cobalt treatment. We confirmed that, once in the nucleus, Yap1 mediates the induction of genes involved in the regulation of oxidative stress in yeast (such as GPX2, SOD1, TRX2 and TRR1). Moreover, we found that the presence of cobalt increases the levels of protein carbonylation, being the damage more pronounced in the absence of Yap1. It was also found that cobalt levels are slightly decreased in the yap1 mutant. In contrast, the iron concentration is higher in this mutant than in the wild type, in the presence and absence of cobalt. This led us to propose that the cobalt alters iron homeostasis and iin consequence generate oxidative stress. It was reported that glutathione (GSH) is important in the homeostasis of iron. As the excess of cobalt leads to increased levels of intracellular iron, we found not only that in the yap1 mutant GSH1 expression is decreased after 30 minutes of treatment with cobalt but also, the addition of GSH can recover the growth phenotypes in the presence of this metal. . Finally we present a working model of the transcription factor Yap1 in stress caused by cobalt.

Oxidative stress is a harmful condition in a cell, tissue, or organ, caused by an imbalance between reactive oxygen species or other oxidants and the capacity of antioxidant defense systems to remove them. These oxidants cause wide-ranging damage to macromolecules, including proteins, lipids, DNA and carbohydrates. Oxidative stress is an important pathophysiologic component of a number of diseases, such as Alzheimerâ s disease, diabetes and certain cancers. Cells contain effective defense mechanisms to respond to oxidative stress. Despite much accumulated knowledge about these responses, their kinetics, especially the kinetics of early responses is still not clearly understood. The Yap1 transcription factor is crucial for the normal response to a variety of stress conditions including oxidative stress. Previous studies on Yap1 regulation started to measure gene expression profile at least 20 minutes after the induction of oxidative stress. Genes and pathways regulated by Yap1 in early oxidative stress response (within 20 minutes) were not identified in these studies. Here we study the kinetics of early oxidative stress response induced by the cumene hydroperoxide (CHP) in Saccharomyces cerevisiae wild type and yap1 mutant. Gene expression profiles after exposure to CHP were obtained in controlled conditions using Affymetrix Yeast Genome S98 arrays. The oxidative stress response was measured at 8 time points along 120 minutes after the addition of CHP, with the earliest time point at 3 minute after the exposure. Statistical analysis methods, including ANOVA, k-means clustering analysis, and pathway analysis were used to analyze the data. The results from this study provide a dynamic resolution of the oxidative stress responses in S. cerevisiae, and contribute to a richer understanding of the antioxidant defense systems. It also provides a global view of the roles that Yap1 plays under normal and oxidative stress conditions.

Le cancer gastrique (CG) est une maladie multifactorielle, fréquemment associée à l’infection chronique par des souches CagA+ d’Helicobacter pylori. La transition épithélio-mésenchymateuse (EMT) est un processus réversible dans lequel une cellule épithéliale polarisée acquiert un phénotype mésenchymateux. L’EMT est à l’émergence de cellules souches cancéreuses (CSC) qui expriment CD44 et présentent une activité ALDH élevée. L’infection des cellules épithéliales gastriques humaines (CEGs) par CagA+ H. pylori induit des cellules CD44+ avec des propriétés des CSCs via une EMT. La voie Hippo est composée par les kinases MST et LATS, et leurs cibles, les YAP1 et TAZ. Suite à la phosphorylation, YAP1 et TAZ sont inhibés. YAP1 et TAZ activés lient les facteurs TEAD pour promouvoir la croissance cellulaire et l’inhibition de l’apoptose.Notre premier objectif était de rechercher si H. pylori change l’état d’activation de la voie Hippo et l'effet sur l’EMT et les CSC in vitro et in vivo. Le deuxième but est la caractérisation du rôle de YAP1/TEAD dans les propriétés de CSCs gastriques in vitro et les conséquences de son inhibition dans la croissance tumorale in vivo.Pour étudier la régulation de la voie Hippo pendant l’infection par H. pylori, LATS2, YAP1 et CD44 ont été évalués dans la muqueuse gastrique de sujets non-infectés et infectés par H. pylori, qui ont été augmentés avec l’infection et leur surexpression a été associée avec la gastrite et la métaplasie intestinale. Dans les CEGs l’expression de gènes de la voie Hippo a été altérée par l’infection. La régulation de la voie Hippo par H. pylori a une cinétique diphasique et dépendante de CagA. Dans l’infection précoce, H. pylori déclenche l’activité transcriptionelle de YAP1. Cette période d’inactivité de la voie Hippo est suivi de son activation progressive, soutenue par l’accumulation de LATS2 et la phosphorylation inhibitrice de YAP1. La répression de LATS2 avec siRNAs a accéléré l’acquisition du phénotype mésenchymateux après l’infection, l’augmentation de marqueurs de l’EMT (Zeb1 et Snail1), et la diminution des miR-200 épithéliaux. Les CSC induites par H. pylori ont été potentialisées par l’inhibition de LATS2, ce qui suggère que LATS2 limite l’EMT et le phénotype de CSC acquis pendant l’infection. L’inhibition de LATS2 ou YAP1 diminue l’expression de ces deux protéines, révélant ainsi une boucle de régulation positive. Dans des coupes de tissu de CG, l’expression de LATS2 et YAP1 est hétérogène et positivement corrélée, fait qui a été confirmé dans 38 CEGs de la CCLE. L’expression LATS2 est fortement corrélée à celle de CTGF et CYR61, ce qui suggère que LATS2 peut aussi être un gène cible de YAP1/TEAD.La verteporfine (VP) est capable d’interrompre l’interaction YAP1/TEAD, et donc d’inhiber son activité transcriptionelle. In vitro, utilisant CEGs et des cellules de tumeurs de patients amplifiées chez la souris (patient-derived xenograft PDX), le traitement à la VP a diminué la croissance cellulaire, l’expression de gènes cible de YAP1/TAZ/TEAD, l’activité du rapporteur TEAD-luciférase et la capacité de formation de sphères. L’activité de la VP a été testée in vivo par injection péri-tumorale dans un modèle de greffe sous-cutanés des CEGs MKN45 et MKN74 et le PDX GC10 chez la souris NSG. La croissance tumorale a été diminuée. Le poids des tumeurs, l’analyse par IHC (CD44, ALDH, Ki67) et la capacité de formation de sphères des CSCs résiduelles ont été diminuées. Ces résultats montrent une activité inhibitrice de la VP sur les CSCs gastriques in vitro et in vivo.Ce travail montre pour la première fois que l’axe LATS2/YAP1/TEAD est précocement activé pendant l’infection chronique avec H. pylori et que celui-ci contrôle l’EMT et les propriétés de CSC. Le ciblage de la voie Hippo a été montré comme étant efficace dans la prévention de la croissance tumorale, mettant en évidence le potentiel de son inhibition dans le traitement du cancer gastrique
Gastric cancer (GC) is a multifactorial disease, most frequently associated to chronic infection with CagA-positive Helicobacter pylori strains. Epithelial-to-mesenchymal transition (EMT) is reversible process in which polarized epithelial cells acquire a mesenchymal phenotype. EMT is at the origin of cancer stem cells (CSC). In GC, CSCs express CD44 and high aldehyde-dehydrogenase (ALDH) activity. Infection with H. pylori of human gastric cancer cell lines (hGECs) in vitro induces the emergence of a population of CD44+ cells with CSC-properties through an EMT process in a CagA-dependent manner. The Hippo pathway is composed by the kinases MST and LATS, and their phosphorylation targets,YAP1 and TAZ. Upon phosphorylation by LATS, YAP1 and TAZ are inhibited. Active YAP1 and TAZ bind to TEAD transcription factors to promote the expression of genes that regulate cell growth and apoptosis.The first aim of this work was to investigate whether H. pylori affects the activation state of the Hippo pathway, and its effect on the EMT process and the CSCs. Second, we intended to characterize the role of YAP1/TEAD in gastric CSC properties in vitro and the consequences of its pharmacological inhibition on tumor growth in vivo.To study the Hippo pathway regulation during infection, LATS2, YAP1 and CD44 were evaluated in gastric mucosae of non-infected or H. pylori-infected patients. They were upregulated in infected mucosae and were associated to pathology. Hippo pathway regulation by H. pylori infection has biphasic kinetics and is CagA-dependent. Early in infection, H. pylori transiently triggered YAP1 expression and co-transcriptional activity, along with LATS2. This period of Hippo pathway inactivity is followed by a progressive activation, sustained by LATS2 accumulation and inhibitory YAP1Ser127-phosphorylation. LATS2 siRNA-mediated repression accelerated the acquisition of the EMT-phenotype upon infection, the up-regulation of EMT-markers ZEB1 and Snail1, and the decrease of the epithelial miR-200. H. pylori-induced CD44 upregulation, invasion and sphere-forming capacity were further enhanced upon LATS2 knockdown, suggesting that LATS2 restricts the EMT and CSC-like phenotype in hGECs upon H. pylori infection. Inhibition of either LATS2 or YAP1 reduced the expression of both proteins, revealing a positive feedback loop. In tissue sections of GC, LATS2 and YAP1 were heterogeneous and co-expressed. The positive correlation between LATS2 and YAP1 was confirmed in the 38 hGECs of the CCLE. The expression of CTGF and CYR61 was also strongly correlated to LATS2, suggesting that LATS2 could also be a YAP1/TEAD target gene.hGECs of the CCLE. The expression of CTGF and CYR61 was also strongly correlated to LATS2, suggesting that LATS2 could also be a YAP1/TEAD target gene.Verteporfin (VP) disrupts the YAP1/TEAD interaction inhibiting its transcriptional activity. In vitro, using hGECs and cells from patient derived primary tumor xenogratfs (PDXs), we showed that treatment with VP decreased cell growth, expression of YAP1/TAZ/TEAD target genes, TEAD-luciferase reporter activity and sphere-forming capacity. The activity of VP was tested in vivo, by peritumoral injection in a model of subcutaneous graft of hGECs (MKN45 and MKN74) and PDX (GC10) in NGS mice. Tumor growth was followed and a decrease was observed. Tumor weight measurement, IHC analysis (CD44, ALDH and Ki67), and CSCs were decreased in treated tumors. These results show the CSC-inhibitory activity of VP both in vitro and in vivo.We showed for the first time that the LATS2/YAP1/TEAD axis is early activated during the carcinogenesis process induced by chronic H. pylori infection and controls the subsequent EMT and CSC-like features. Targeting the Hippo pathway efficiently prevented tumor growth in a PDX model, highlighting the potential of its inhibition to be implemented in gastric cancer therapy

O trabalho aqui desenvolvido teve como objeto o estudo estrutural e funcional de várias proteínas do fitopatógeno Xanthomonas axonopodis pv citri (Xac), dentre as quais se destacam as proteínas hipotéticas conservadas YaeQ e SufE, as proteínas RpfC, RpfF e RpfG envolvidas em quorum sensing e proteínas PilZ, FimX e PilB envolvidas na biogênese do pilus tipo IV. Para o desenvolvimento deste trabalho foram utilizadas diferentes técnicas incluindo: clonagem, expressão, purificação, desnaturação térmica, cristalografia, difração de raios-X, RMN, ensaios de 2-híbrido, produção de nocautes, mutação sítio dirigida, Western- e Far- Western, entre outras. Dentre os resultados mais importantes obtidos temos a determinação estrutural das proteínas YaeQ e PilZ pela técnica MAD. Em ambos os casos, as estruturas representaram topologias inéditas. Com base nos dados estruturais, mostramos que YaeQ pertence à família PD-(D/E)XK presente em endonucleases dependentes de magnésio, e a partir de ensaios funcionais obtivemos evidências que sugerem que YaeQ está envolvida em alguma via de reparo de DNA em Xac. A estrutura tridimensional de PilZ revelou uma inesperada variedade estrutural dentro da família PilZ e mostrou de forma clara porque ortólogos não interagem com o segundo mensageiro bacteriano, c-diGMP. A cadeia principal de PilZ foi assinalada por RMN e a estrutura secundária de PilZ em solução é consistente com aquela determinada por cristalografia. Duas proteínas que interagem com PilZ foram identificadas: PilB e FimX. Como PilZ, ambos exercem papéis na biogênese do pilus tipo IV (T4P). Mostramos que PilZ interage especificamente com o domínio EAL de FimX e que resíduos conservados na região do C-terminal de PilZ estão envolvidos na interação com PilB, mas não com FimX. Ensaios de mutação sítio dirigida mostraram que a Y22 de PilZ pode estar envolvida na regulação da interação de PilZ com FimX e com PilB. Apesar de PilZ não interagir com c-diGMP seu parceiro, FimX, interage. PilZ consegue interagir com PilB ao mesmo tempo em que interage com FimX, formando um complexo ternário que é independente da interação de FimX com c-diGMP. Com base em todos estes resultados propusemos possíveis mecanismos de ação de PilZ e FimX no controle da biogênese do T4P. Além dos resultados acima descritos, determinamos a estrutura de SufE e mostramos que esta aumenta a atividade cisteína dessulfarase de seu parceiro, SufS, em torno de 10 vezes, como ocorre com SufE-SufS de E.coli. Clonamos, expressamos, purificamos e fizemos ensaios de cristalização de algumas proteínas envolvidas no controle de quorum sensing em Xac. Tivemos êxito na cristalização do domínio HPT (histidina fosfotransferase) da proteína chave deste sistema, RpfC
The aim of the project was to perform structural and functional studies of different Xanthomonas axonopodis pv citri (Xac) proteins including the hypothetical proteins YaeQ and SufE; RpfC, RpfF and RpfG involved in the quorum sensing and PilZ, FimX and PilB that play roles in type IV pilus (T4P) biogenesis. Several experimental techniques were employed including cloning, expression and purification of recombinant proteins, thermal denaturation, protein crystallography, X-ray diffraction, NMR, two-hybrid assays, Western- and Far-Western Blotting assays, site direct mutagenesis, and the production of Xac knockouts strains. The most important results include the determination of the three-dimensional crystal structures of PilZ and YaeQ using the MAD technique. In both cases, the structures reveled new protein topologies. The comparison of the YaeQ structure with others deposited in public databases revealed that YaeQ proteins represent a new variation within the PD-(D/E)XK magnesium dependent endonucleases superfamily. Functional assays suggest that YaeQ may be envolved in DNA repair in Xac. The PilZ three-dimensional structure revealed an unexpected structural variation within the PilZ domain superfamily and showed why PilZ orthologs are not able to bind the important bacterial second messenger, c-diGMP. We assigned the PilZ main chain by NMR and used this information to demonstrate that the PilZ secondary structure in solution is consistent with the PilZ crystal structure. We identified two proteins that interact with PilZ: PilB and FimX. As with PilZ, both PilB and FimX are involved in T4P biogenesis. PilZ binds specifically to the EAL domain of FimX and the conserved residues located in the PilZ unstructured C-terminal region contribute to binding with PilB but not with FimX. Site direct mutagenesis studies showed that PilZ residue Y22 is necessary for its capability to interact with both PilB and FimX. Although PilZ does not bind c-diGMP, her partner, FimX, does. We present evidence that PilZ can bind simultaneously to FimX and PilB, forming a ternary complex that is independent of c-diGMP. These results allow us to propose possible mechanisms by which PilZ and FimX control T4P biogenesis. Other results obtained during this period include the resolution of the crystal structure of the SufE protein from Xac using the molecular replacement technique. We show that SufE induces a 10-fold increase in the cysteine desulfurase activity of SufS, similar to that observed for the SufE-SufS complex from E. coli. Several proteins involved in quorum sensing and c-di-GMP signaling were cloned, expressed and submitted to crystallization trials. Quando submetidas a condições de estresse as células respondem a partir da indução de genes e ativação de proteínas em mecanismos regulados por cascatas de eventos metabólicos. Na levedura Saccharomyces cerevisiae muitos desses sinais externos são recebidos e amplificados por vias das chamadas MAP quinases. Em estudos anteriores mostramos a eficácia do biocida catiônico polihexametileno biguanida (PHMB) no combate a leveduras que contaminam o processo de fermentação alcoólica industrial. Entretanto, foi observado que certas linhagens de S. cerevisiae são sensíveis a esse biocida. Portanto, para que se possa propor o uso deste composto no controle de contaminações industriais é necessário que conheçam os mecanismos moleculares envolvidos na resposta à ação biológica do PHMB e dos mecanismos de resistência celular a este composto. Trabalhos da literatura mostraram que células de E. coli respondem a este composto a partir da expressão de genes envolvidos na manutenção da integridade da parede celular (CWI). A partir da identificação desses homólogos em Saccharomyces cerevisiae realizamos uma série de experimentos de análise da expressão gênica e da avaliação de linhagens mutantes dessa levedura. Os experimentos mostraram que a cascata regulatória da via da proteína quinase C (PKC) regula a expressão dos genes do mecanismo CWI que respondem às lesões na parede celular induzidas pelo PHMB, causando um estresse osmótico que é sentido e restaurado pelos genes da via HOG. O efeito protetor da trealose, a geração de desequilíbrio redox e a natureza do efeito tóxico sugerem que PHMB dispara o mesmo sinal molecular que é sentido pelas células quando submetidas a estresse etanólico. Além disso, a proteína Yap1, principal regulador da transcrição dos genes de resposta a estresse oxidativo, parece ser crucial para a resistência ao PHMB, embora neste caso a função desse fator de transcrição estaria relacionada com um efeito protetor sobre a parede celular. Com isso, estamos propondo uma nova função biológica para a proteína Yap1 na resposta a estresse industrial em Saccharomyces cerevisiae

Les gliomes malins sont les tumeurs les plus fréquentes du système nerveux central. Les glioblastomes représentant plus de 50% des gliomes, constituent la forme la plus agressive et sont particulièrement résistants à la radiochimiothérapie. Au sein de ces tumeurs réside une sous-population de cellules souches tumorales (CSG) qui pourrait être responsable de leurs initiation, progression et résistance aux traitements. Ces processus sont gouvernés par des voies de signalisation, pour la plupart activées de manière constitutive et dont l’étude est nécessaire afin de mieux comprendre les mécanismes impliqués dans la gliomagenèse. L’objectif de ces travaux de thèse consistait en l’exploration des voies de signalisation STAT3 et Hippo dans les gliomes dans le but d’identifier de nouveaux marqueurs pronostiques et de nouvelles cibles thérapeutiques potentielles. La première partie de ces travaux a montré que la phosphorylation S727 de STAT3 jouait un rôle important dans la radioresistance des CSG et que son inhibition pharmacologique induisait leur radiosensibilisation. Dans un second temps, ces travaux ont montré que deux effecteurs de la signalisation Hippo, YAP1 et TEAD3, sont associés à un mauvais pronostic et qu’ils seraient impliqués dans la prolifération cellulaire et le phénotype des CSG notamment par inhibition de la signature proneurale. Ainsi, ces travaux visent à proposer de nouvelles pistes thérapeutiques, d’une part l’inhibition de la pS727-STAT3 afin de potentialiser les effets de la radiothérapie et d’autre part, les effecteurs de la signalisation Hippo comme biomarqueurs pronostiques et potentielles cibles thérapeutiques
Malignant gliomas are the most common tumors of central nervous system. Glioblastomas represent more than 50% of all glioma and constitute the most aggressive form of the tumor which is particularly resistant to radiotherapy. The presence of the subpopulation of glioblastoma stem cells (GSC) could be involved in tumor initiation, progression and therapeutic resistance. Hence, these processes are governed by signaling pathways which are mostly constitutively activated and their study is necessary for a better understanding of gliomagenesis. The aim of this PhD thesis was to assess STAT3 and Hippo signaling pathways in glioma to identify new prognostic markers and potential therapeutic targets. The first part on this work showed that pS727 phosphorylation of STAT3 could be involved in radioresistance and its inhibition induced GCS radiosensitization. Additionally, this work showed that YAP1 and TEAD3, two effectors of Hippo signaling, are associated with poor patient survival and could be involved in GSC proliferation and phenotype maintenance by inhibiting proneural gene signature. Thereby, this work aims to offer new therapeutic avenues, on the one hand the inhibition of pS727-STAT3 for radiotherapy potentiation and on the other hand the effectors of Hippo signaling as prognostic biomarkers and potential therapeutic targets

Molecular dynamics (MD) is a powerful tool that can be applied to protein folding and protein structure. MD allows for the calculation of movement, and final position, of atoms in a biomolecule. These movements can be used to investigate the pathways that allow proteins to fold into energetically favorable structures. While MD is very useful, it still has its limitations. Most notable, computing power and time are of constant concern. Protein structure is inherently important due to the direct link between the structure of a protein and its function. One of the four levels of protein structure, the secondary structure, is the first level to accommodate for the three-dimensional shape of a protein. The main driving force behind secondary structure is hydrogen bonding, which occurs between the carboxyl oxygen and the amine hydrogen of the backbone of a peptide. Determining a greater link between hydrogen bond patterns and types of secondary structure can provide more insight on how proteins fold. Because molecular dynamics allows for an atomic level view of the dynamics behind protein folding/unfolding, it becomes very useful in observing the effects of particular hydrogen bond patterns on the folding pathway and final structure formed of a protein. Using molecular dynamic simulations, a series of experiments in an attempt to alter structure, hydrogen bonding, and folding patterns, can be performed. This information can be used to better understand the driving force of secondary structure, and use the knowledge gained to manipulate these simulations to force folding events, and with that, desired secondary structure features.

Cardiovascular disease is a leading cause of death in the United States. Toll-like receptor (TLR)-mediated pathways have been demonstrated to play a role in myocardial ischemia/reperfusion (I/R) injury. We and others have shown that PI3K/Akt signaling is involved in regulating cellular survival and protecting the myocardium from I/R induced injury. In this dissertation, we provide compelling evidence that miR-125b serves to “fine tune” TLR mediated NF-kB responses by repressing TNF-a and TRAF6 expression. We constructed lentiviral expressing miR-125b, delivered it into the myocardium. The data showed that delivery of lentivirus expressing miR-125b significantly reduces myocardial infarct size and improves cardiac function in I/R hearts. Mechanistic studies demonstrated that miR-125b negatively regulates TLR mediated NF-kB activation pathway by repressing TNF-a and TRAF6 expression in the myocardium. We also observed that transfection of the myocardium with lentivirus expressing miR-214 markedly attenuates I/R induced myocardial infarct size and cardiac dysfunction. We demonstrated that miR-214 activates PI3K/Akt signaling by targeting PTEN expression in the myocardium. We also investigated the role of TLR3 in neonatal heart repair and regeneration following myocardial infarction (MI). Wild type (WT) neonatal mice showed fully cardiac functional recovery and small infarct size, while TLR3 deficient mice exhibited impaired cardiac functional recovery and large infarct area after MI. Poly (I:C), a TLR3 ligand, administration significantly enhances glycolysis, YAP1 activation and the proliferation of WT neonatal cardiomyocytes. 2-deoxyglucose (2-DG), a glycolysis inhibitor treatment abolished cardiac functional recovery and YAP1 activation in neonatal mice after MI. In vitro either inhibition of glycolysis by 2-DG or inhibition of YAP1 activation prevents Poly (I:C) induced YAP1 activation and neonatal cardiomyocyte proliferation. Importantly, YAP1 activation increases miR-152 expression, leading to cardiomyocyte proliferation through suppression P27kip1 and DNMT1 expression. We conclude that microRNAs play an important role in TLR modulation induced protection against myocardial I/R injury by increasing the activation of PI3K/Akt signaling pathway, decreasing TLR/NF-kB mediated inflammatory response, and suppressing activation of apoptotic signaling following myocardial I/R injury. In addition, TLR3 is an essential for neonatal heart repair and regeneration after myocardial infarction. TLR3 modulation could be a novel strategy for heart regeneration and repair.

Lung cancer is the leading cause of cancer-related death in both men and women, nationally and internationally and kills more people each year than breast, prostate, and colon cancers combined. Non-small cell lung carcinoma (NSCLC) is the most common histological subtype of lung cancer, and accounts for 85% of all cases. Cigarette smoking is the single greatest risk factor for lung cancer, and is correlated with 80-90% of all lung cancer deaths. Nicotine, the addictive component of tobacco smoke, is not a carcinogen and cannot initiate tumors itself; however, it is known to act as a tumor promoter, by enhancing the proliferation, migration, and invasion of cells in vitro, thus accelerating tumor growth and metastasis in vivo. Nicotine exerts is tumor promoting effects primarily by binding to, and activation of, nicotinic acetylcholine receptors (nAChRs), specifically the α7 subunit of nAChRs. While α7 nAChR is expressed in a wide array of cells, how its expression is regulated is not fully understood. Here we sought to elucidate the transcriptional regulation of α7 nAChR in NSCLC cells. We report that α7 nAChR expression is induced by nicotine in an autoregulatory feedforward loop, and that the α7 gene promoter is differentially regulated by E2F1 and STAT1 transcription factors at an overlapping binding site suggesting a competitive interplay. Depletion of E2F1 resulted in a reduced ability of nicotine to induce α7 nAChR, while depletion of STAT1 resulted in enhanced induction, suggesting that nicotine might use these two transcription factors to modulate the expression of α7 nAChR in a very precise fashion. More recently, nicotine has been implicated in promoting self-renewal of stem-like side-population cells from lung cancers. Cancer stem-like cells have been implicated in tumor initiation as well as the maintenance, drug resistance, dormancy, recurrence, and metastasis of various tumor types. We had previously shown that the embryonic stem cell transcription factor, Sox2, is indispensable for self-renewal of stem-like cells from lung adenocarcinoma cell lines; hence we sought to determine whether nicotine enhances stemness of lung cancer stem-like cells through Sox2. We find that nicotine can induce the expression of Sox2 at the transcriptional level and this occurs through a nAChR-Src-Yap1-E2F1 signaling axis. Over recent years, electronic cigarettes (e-cigarettes) have emerged as healthy alternatives to traditional cigarette smoking as they do not contain tobacco; however, they do still contain nicotine. Our studies show that e-cigarette components can enhance tumor promoting properties of NSCLC cells similar to that observed with nicotine alone, and find that they can induce expression of Sox2 and mesenchymal markers as well as enhance migration and stemness of NSCLC cells. Taken together, these studies reveal novel molecular mechanisms by which exposure to nicotine, via cigarette smoke or e-cigarettes, could alter the oncogenic potential of NSCLC cells.

Inspired by the contrapuntal and relational critiques of Edward Said and Ella Shohat, this thesis conducts a comparative analysis of the literature and film of Arab Jews in order to deconstruct discourses on Jewish identity that privilege the dichotomies of Israel-diaspora and Arab-Jew. Sami Michael’s novel Refuge, Naim Kattan’s memoir Farewell, Babylon, Karin Albou’s film Little Jerusalem and b.h. Yael’s video documentary Fresh Blood: a Consideration of Belonging reveal the complexities and interconnections of Sephardic, Mizrahi and Arab Jewish experiences across multiple geographies that are often silenced under dominant Eurocentric, Ashkenazi or Zionist interpretations of Jewish history. Drawing from these texts, Jewish identity is explored through four philosophical themes: Jewish beginnings vs. origins, boundaries between Arab and Jew, the construction of Jewish identities in place and space, and, the concept of diaspora and the importance Jewish difference. As a double-diaspora, with the two poles of their identities seen as enemies in the ongoing conflict between Israel-Palestine, Arab Jews challenge the conception of a single Jewish nation, ethnicity, identity or culture. Jewishness can better be understood as a rhizome, a system without a centre and made of heterogeneous component, that is able to create, recreate and move through multiple territories, rather than ever settling in, or being confined to a single form that seeks to dominate over others. This dissertation contributes a unique theoretical reading of Jewish cultures in the plural, and includes an examination of lesser known Arab Jewish writing and experimental documentary in Canada in relation to Iraq, France and Israel.

The present work brought the Digital Information and Communication Technologies (DICT) inclusion in the modality of Youth and Adult Education (YAE) for the Chemistry teaching and learning process, in order to identify the constituent elements of the teaching and learning process, from the perspective of the YAE, which denote conceptual and social meaning to the teaching of chemistry through DICT. There were moments of dialogue sessions to expose the substance content that preceded the use of the educational applications "MERCK PTE" and "molculator". The research developed in the action research methodology, had the lessons recorded and filmed for later transcription, reflection and analysis. In the data analysis, from three categories, "Rejuvenation of the public of YAE; Didactic perspectives in the use of DICT and (In) dependence on the applications use ", it was observed the importance of the dialogue between teacher and students, based on the life experiences and the practice of using the DTIC, among the older students and the younger ones. This exchange of experiences was decisive for the effective learning process of the chemical contents mediated by digital technologies. It’s concluded from the analysis that it is possible to approach chemical contents through the DTIC in classrooms of YAE, since it is a profitable public to exchange experiences and dialogue and, because of being rejuvenated, absorb study habits that involve the technologies as a new school configuration element.
O presente trabalho trouxe a inclusão de Tecnologias Digitais da Informação e Comunicação (TDIC) na modalidade de Educação de Jovens e Adultos (EJA) para o processo de ensino e aprendizagem de Química, tendo como objetivo identificar elementos constitutivos do processo de ensino e aprendizagem, na perspectiva da EJA, que denotem significado conceitual e social ao ensino de química por meio das TDIC. Foram utilizados aplicativos educativos, como “MERCK PTE” e “molculator”, voltados ao ensino dos conteúdos de química na EJA, buscando assim, estabelecer relações entre conceitos e contexto a partir da interação entre diferentes faixas etárias e o uso das TDIC. Houve momentos de aulas dialogadas para exposição do conteúdo de quantidade de matéria que antecederam o uso dos aplicativos educativos “MERCK PTE” e “molculator”. A pesquisa, desenvolvida na metodologia de pesquisa-ação, teve as aulas gravadas e filmadas para posterior transcrição, reflexão e análise. Na análise dos dados, a partir de três categorias, “Rejuvenescimento do público de EJA; Perspectivas didáticas no uso das TDIC e (In)dependência no uso dos aplicativos”, observou-se a importância do diálogo entre docente e alunos, pautado nas experiências de vida e da prática de uso das TDIC, entre os estudantes mais velhos e os mais novos. Essa troca de experiências foi determinante para o efetivo processo de aprendizagem dos conteúdos químicos mediados pelas tecnologias digitais. Conclui-se a partir da análise, que é possível abordar conteúdos químicos por meio das TDIC em salas de aulas de EJA, visto ser um público profícuo à troca de experiências e diálogo e, por estar rejuvenescido, absorvem hábitos de estudos que envolvem as tecnologias como novo elemento da configuração escolar.

La presente tesis realiza un estudio sobre la reescritura del mito griego en el teatro contemporáneo. El centro de la atención es la reescritura de la Orestíada, la tragedia esquilea en la que se representa el paso de una sociedad arcaica y violenta a una democrática, donde, sin embargo, el matricida queda impune. A lo largo de la historia cultural europea la Orestíada ha representado un importante modelo teatral así como un núcleo para reflexionar sobre el nexo entre tragedia, política y rito. Reescribir el mito significa deconstruir el imaginario y trabajar sobre las representaciones presentes en el canon occidental que ha desarrollado una función normativa. En la primera parte de esta tesis, se propone un recorrido teórico y propedéutico alrededor de tres núcleos: una aproximación a la Orestíada a lo largo de la historia cultural; un análisis detallado de los conceptos de intertextualidad y reescritura a partir de las teorías literarias y teatrales; una descripción panorámica de las distintas tendencias estéticas y culturales presentes en el teatro contemporáneo (las tendencias postmodernas y posdramáticas y las conexiones con lo intercultural y postcolonial y el género). En la segunda parte se analizan cuatro reescrituras contemporáneas de diferentes países: Orestea. Una Commedia Organica? de Romeo Castellucci y la Socìetas Raffaello Sanzio, (Italia, 1995); Orestea ex machina de Mapa Teatro (Colombia,1995); Agamenón. Volví del supermercado y le di una paliza a mis hijos de Rodrigo García y la Carnicería Teatro (España, 2003); Molora de Yael Farber y la Farber Foundry (Sudáfrica, 2003). El objetivo es analizar la reescritura de la Orestíada dentro y fuera de Europa y observar cómo el contexto histórico, político y cultural influye en la reescritura. Por una parte, el análisis de las obras desarrolla los vínculos entre los sistemas escénicos de representación y los contextos histórico-culturales específicos. Por otra, se apoya en la reconstrucción de la estética de cada compañía y de las estrategias de re-apropiación del mito clásico. La investigación toma como eje vertebrador el teatro actual como espacio cultural y artístico propicio para replantear una reflexión sobre la relación entre el mito griego, lo político y lo ritual en la contemporaneidad. La perspectiva teórica es interdisciplinar y entrecruza varios discursos presentes en la contemporaneidad: artes escénicas, teoría del teatro y de la literatura, semiótica, estudios de género y postcoloniales. Dentro de este marco, la reescritura constituye un complejo laboratorio para trabajar sobre la dialéctica pasado/presente y, también, nosotros/los otros.
This thesis presents a study on the rewriting of Greek myth in contemporary theatre and, in particular, analyzes the rewriting of the Oresteia in different countries. The Aeschylus’ trilogy is concerned with the transition from a violent society to a democratic one. Nevertheless, the resulting democracy is not a fair one because it is based on impunity: Orestes is absolved. Within the European cultural history, the Oresteia represents an important theatrical model as well as an interesting topic to re-interpret the relationship between tragedy, politics and ritual. Re-writing the myth entails deconstructing the stereotypes and working through the western canon, which involves a normative connotation. In the first part of my dissertation, I examine three clusters: the myth of the Oresteia; the topics of “intertextuality” and “rewriting” from a literary and theatrical perspective; the description of the different trends in contemporary theatre (postmodern and post-dramatic movements and the connections with intercultural, postcolonial and gender studies). In the second part of the dissertation, I examine four rewritings of the Oresteia: Orestea. Una Commedia Organica? by Romeo Castellucci and the Socìetas Raffaello Sanzio (Italy, 1995); Orestea ex machina by Mapa Teatro 1995 (Colombia, 1995); Agamenón. Volví del supermercado y le di una paliza a mis hijos by Rodrigo García and the Carnicería teatro (Spain, 2003); Molora by Yael Farber and the Farber Foundry (South Africa, 2003). The main purpose is to approach the “rewriting” of Greek myth in contemporary theatre from an interdisciplinary and theoretical perspective as well as to analyze the connections between the historical and political contexts and the texts. On the one hand, the thesis analyzes the relationship between theatrical strategies and specific historic-cultural contexts. On the other hand, the purpose is to reconstruct the aesthetics of the performances and the different models of reappropriation of the Greek myth. In my thesis, I consider theatre as a cultural tool which contributes to the survival of Greek culture intertwining political and ritual aspects. In this respect, the “re-writing” is like a huge laboratory, where it is possible to explore the borders between classic and contemporary representations of us/the others.

Le contrôle de la concentration du peroxyde d'hydrogène [H2O2] est un processus essentiel à la survie des cellules. Chez S. Cerevisiae, le facteur de transcription Yap1 est le régulateur majeur de ce contrôle : il est activé par oxydation par l'H2O2, de façon indirecte, via Orp1, détecteur de l'H2O2 et transducteur de ce signal à Yap1. Nous avons, dans cette étude, exploré des déterminants de la régulation redox de Yap1. Nous avons identifié deux paramètres cruciaux dans cette régulation : la concentration d'H2O2 résiduelle qui, au-dessus d'un certain seuil, est responsable de la réoxydation itérative du facteur, et la réduction catalytique de Yap1, probablement via les thiorédoxines (Trx). Les Trx interviennent aussi sur le premier paramètre, puisqu'elles sont les réductases des peroxyrédoxines (Prx), assurant la dégradation de l'H2O2. Trx et Prx étant contrôlées par Yap1, il y a autorégulation de Yap1 par le biais des deux paramètres identifiés. Ces résultats établissent que Yap1 est un moniteur hautement fiable de [H2O2]. Dans une deuxième partie, nous avons identifié la forme oxydée de Yap1 in vivo, deux ponts disulfures intramoléculaires et proposé un modèle pour leur formation, faisant intervenir l'association séquentielle de deux molécules d'Orp1 oxydé avec Yap1. Notre étude montre que la configuration de la forme oxydée de Yap1 assure la stabilité de sa forme active. Finalement, nous avons tiré parti de la fonction de moniteur redox de Yap1 pour étudier la contribution des Prx, Trx et catalases dans le contrôle de [H2O2], ainsi que celle des régulateurs Msn2/4 et conclure à l'existence d'une régulation fine et permanente de l'H2O2, de type homéostatique
Respiring organisms produce H2O2 and must regulate its concentration [H2O2] to maintain viability. In the yeast S. Cerevisiae, the Yap1 transcription factor is the major regulator of this homeostatic control. Yap1 is activated by oxidation by H2O2. This oxidation is indirect, involving Orp1, a Gpx-like enzyme acting as the H2O2 sensor and the transducer of this signal to Yap1. We have explored here several determinants of Yap1 redox regulation. We identified two essential parameters in this regulation : the residual H2O2 that re-oxidizes iteratively Yap1 when above a threshold-concentration, and Yap1 catalytic reduction, operated by thioredoxins (Trx) probably. Trx act also on the first parameter by being the electron donor for peroxiredoxins, the major H2O2 scavenging enzymes. Prx and Trx being under transcriptional control of Yap1, the all system is auto-regulated. Our data establish Yap1 as a faithful monitor of [H2O2]. In a second part, we have identified the nature of the oxidized form of Yap1 in vivo, containing two intramolecular disulfide bonds, and proposed a model for their formation, involving the sequential association of two oxidized Orp1 molecules with Yap1. Our data demonstrate that the configuration of the oxidized form of Yap1 provides the stability of the active form of the regulator, a crucial parameter for its function. In the last part, we took advantage of Yap1 as a monitor of [H2O2] to measure the pharmacokinetics of this oxidant and to test the respective contributions of Prx, Trx and catalases in the control of [H2O2] as well as that of the stress regulators Msn2/4. Les plantes de la famille des légumineuses sont capables d'établir une interaction symbiotique avec des bactéries du sol, collectivement appelées Rhizobia. Cette interaction aboutit à la formation d'un nouvel organe appelé nodule et dans lequel les bactéries différenciées fixent l'azote atmosphérique au bénéfice de la plante hôte. Les facteurs Nods (NFs) rhizobiens sont des molécules clés responsables de la spécificité d'hôte, et sont capables à eux seuls d'induire de nombreuses réponses développementales racinaires. MtNF-YA1 est un gène dont l'expression est fortement induite au cours du processus de nodulation chez Medicago truncatula. Ce gène code pour une sous-unité du complexe NF-Y, également appelé " CCAAT-Binding Factor " (CBF). Le facteur de transcription NF-Y est composé de trois sous-unités différentes appelées NF-YA, NF-YB, et NF-YC. Alors que chez les animaux, chaque sous-unité n'est généralement codée que par un seul gène, une diversification structurelle et fonctionnelle s'est produite chez les plantes, menant à l'apparition de familles multigéniques. Des analyses de plantes RNAi ou de mutants nf-ya1 ont précédemment montré que MtNF-YA1 est impliqué dans le maintien de l'activité méristématique nodulaire. Au cours de ma thèse, nous avons apporté l'évidence d'un rôle clé additionnel de MtNF-YA1 pendant l'infection rhizobienne. Nous avons de plus montré que MtNF-YA1 et son homologue MtNF-YA2 sont impliqués au cours de la voie de signalisation de réponse aux NFs menant à l'activation de l'expression du gène marqueur MtENOD11. En parallèle, l'étude de complexes trimériques NF-Y dans lesquels agissent NF-YA1 pour contrôler les étapes précoces de la nodulation a conduit à l'identification de partenaires potentiels NF-YB et NF-YC
Plants belonging to the legume family are able to establish a nitrogen fixation symbiosis with soil bacteria called Rhizobia, resulting in the formation of new organs called root nodules, in which differentiated bacteria fix atmospheric nitrogen to the benefit of the host plant. The bacterial Nod Factors (NFs) are known as key players responsible for the host specificity, and are able, in the absence of bacteria, to trigger many early plant responses. MtNF-YA1, in Medicago truncatula, is a gene whose expression is strongly up-regulated during the nodulation process. It encodes a subunit of the CCAAT-Binding Factor (CBF) also called NF-Y complex. The NF-Y transcription factor is composed of three different subunits (NF-YA, NF-YB and NF-YC). While in animals each NF-Y subunit is encoded by a single gene, a structural and functional diversification has occurred in plants, leading to the emergence of gene families. Using RNAi and mutant analyses, we previously showed that MtNF-YA1 is involved in the maintenance of the nodule meristematic activity. During my PhD, we provided evidences for the involvement of MtNF-YA1 during earlier stages of symbiotic root infection. We have shown that MtNF-YA1, together with the closely-related MtNF-YA2, is involved in the early NF signaling pathway leading to the activation of the MtENOD11 marker-gene. In parallel, the study of NF-Y trimeric complexes in which MtNF-YA1 acts to control early steps of nodulation led to the identification of potential NF-YB and NF-YC partners

Para tanto, descortinou-se neste ensaio a âteceturaâ entre as concepÃÃes histÃricas da avaliaÃÃo no cenÃrio da educaÃÃo de jovens e adultos, da subjetividade individual e social que dÃo sustentaÃÃo à formaÃÃo da cultura escolar, por meio de suas falas, dialogicidade, relaÃÃes sociais e da avaliaÃÃo enquanto prÃtica pedagÃgica adotada na EJA. O ato de revolver e vasculhar a temÃtica da avaliaÃÃo enquanto prÃtica pedagÃgica na EducaÃÃo de Jovens e Adultos possibilita que o problema ocupe um espaÃo epistemolÃgico significativo, principalmente quando referenciadas Ãs inÃmeras variÃveis que entrecortam o trabalho do professor na construÃÃo de uma prÃtica avaliativa humanizada. The present research aims to supply theoretical subsidies for reflection in the field of the educational evaluation facing the practices adopted in the education of youth and adults in the initial series of basic education and if these benefit or not the exercise of the education right, focused on the permanence aspect, access and scholar inclusion of the pupil. So, the âweavingâ was disclosed in this essay among the historical conceptions of the evaluation in the scene of youth and adults education, of the individual and social subjectivity that supports the schooling culture formation, through its speech, dialog aspect, social relations and evaluation concerning the adopted pedagogic practice in YAE. The topic might be translated into the following question: the evaluative practice adopted in the scene of the and Adults and Youth Education in Public Schools of Fortaleza suggests the permanence process of the pupil in the school, corroborates for its inclusion in the heart of society and then, in the exercise of education right? The work is based on bibliographical, documental and realistic experiments research. Through the experiments, we tried to adopt, as instrumentals, the commonly applied survey to ten percent of the MUNICIPAL public school teachers and the focused group, applying with 06 students from Regional VI schools, where we tried to point the âmodus operandiâ of the evaluative practice concerning the possibility of compliance of the social role of including the pupil in the school and in the society.Th e act of digging and searching the thematic of the evaluation as a pedagogical practice in Youth and Adult Education allows the problem to occupy a significant epistemologic space, mainly when we refer to the several variable aspects that cross the work developed by the teacher in the construction of a humanized evaluative practice. After an excursion on the designated conceptions, we can announce it as one of the main final considerations, which the act of evaluating is tied to the formation of the internalized âhabitusâ culturally.

Les peroxydases à thiol jouent un rôle central dans la physiologie du peroxyde d’hydrogène (H2O2), un oxydant possédant une fonction de messager cellulaire. Ces enzymes catalysent la réduction de H2O2 par réaction avec une Cys catalytique, ce qui leur confère la capacité de jouer le rôle de détecteur et relais du message redox. Chez Saccharomyces cerevisiae, l’activation H2O2-dépendante du facteur de transcription Yap1, un régulateur clef de la réponse au stress oxydant, dépend de la formation de ponts disulfure intramoléculaires catalysés par la peroxydase à thiol Orp1, via la réaction de l’intermédiaire acide sulfénique avec une Cys de Yap1 pour former un complexe disulfure mixte. L’étude des mécanismes à l’origine de la spécificité de la réaction entre Orp1 et Yap1, et du rôle de la protéine Ybp1 comme partenaire essentiel de l’activation de Yap1, constitue la question centrale de ce travail. Nos résultats montrent que Ybp1 permet de recruter Yap1 et Orp1 au sein d’un complexe ternaire au sein duquel (i) la cinétique de la réaction entre les deux partenaires est fortement activée, et (ii) la compétition avec la formation d’un pont disulfure intramoléculaire dans Orp1 est inhibée. La spécificité de l’activation de Yap1 par H2O2 repose donc sur des mécanismes qui combinent à la fois la réactivité chimique intrinsèque de l’acide sulfénique, et la reconnaissance moléculaire entre Yap1, Orp1 et Ybp1 qui jouerait un rôle de protéine « de charpente ». Ces principes, assurant une activation rapide et spécifique des défenses antioxydantes de la levure Saccharomyces cerevisiae, pourraient s’appliquer à d’autres voies de signalisation dépendantes des peroxydases à thiol
Thiol-peroxidases play a central role in the physiology of hydrogen peroxide, an oxidant which can act as a cellular messenger. They catalyze H2O2 reduction by the very efficient reaction of a catalytic Cys residue, responsible for their ability to act as an H2O2 sensor and relay. In Saccharomyces cerevisiae, the H2O2-activation of the transcription factor Yap1, a key regulator of oxidative stress response, depends on the formation of intramolecular disulfide bonds catalyzed by the thiol peroxidase Orp1, through the reaction of the sulfenic acid intermediate with a Cys of Yap1 to form a mixed disulfide complex. Due to the high reactivity of the sulfenic acid species, several reactions can compete with Yap1. The study of the mechanisms underlying the specificity of the reaction between Orp1 and Yap1, and of the role of the Ybp1 protein as an essential partner of Yap1 activation, is the central question of this work. Our results show that Ybp1 can recruit Yap1 and Orp1 within a ternary complex that allows (i) strong activation of the reaction between the two partners and (ii) inhibition of the competition raised by the formation of an intramolecular disulfide bond within Orp1. The specificity of the activation of Yap1 by H2O2 therefore relies on mechanisms that combine intrinsic chemical reactivity of the sulfenic acid species and molecular recognition between Yap1, Orp1 and Ybp1, which would act as a scaffold. These principles, which afford rapid and specific activation of antioxidant defenses in Saccharomyces cerevisiae, could apply to other redox signaling pathways dependent on thiol peroxidase as a H2O2 sensor

Les salmonelles, responsables de fièvres typhoïdes et gastro-entérites, sont un problème majeur de santé publique. Les systèmes de sécrétion de type III (TTSS) sont des facteurs de virulence cruciaux de Salmonella. Nos travaux ont montré que délétion du cadre ouvert de lecture yfgL conduit à une faible transcription des gènes codant les 3 TTSS conduisant à une baisse importante de la virulence de Salmonella Enteritidis. Or, la délétion de ce gène, dont la protéine chez E.coli est en complexe avec les protéines YaeT, YfiO, NlpB et SmpA, conduit à une altération de la membrane externe. Chez S. Enteritidis, nous avons montré que le rôle du complexe « YaeT » dans l’assemblage des protéines de membrane externe est conservé. Or, seule l’inactivation d’yfiO, conduit une diminution de l’expression des TTSS. Nos travaux ont toutefois suggéré que le défaut d’assemblage des protéines de membrane externe n’est pas la cause de ce défaut d’expression des TTSS observés chez les mutants yfgL et yfiO
Salmonella, responsible for typhoid fever and gastroenteritis, is a worldwide health problem. Type three secretion system (TTSS) are crucial virulence factors in Salmonella. Our work showed that deletion of the open reading frame yfgL led to a transcriptional down-regulation of the genes encoding the proteins involved in the biosynthesis of the 3 TTSS in Salmonella Enteritidis. It was shown that inactivation of yfgL whose encoded protein is in complex with YaeT, YfiO, NlpB and SmpA in E. coli, causes an outer membrane alteration. In S. Enteritidis, we observed that the role of the “YaeT” complex in outer membrane protein assembly is conserved in S. Enteritidis. In addition, only yfiO inactivation resulted in a down-expression of the TTSS. However, we presented elements suggesting that the outer membrane protein targeting defect was not responsible for the TTSS down-expression observed in the yfgL and yfiO mutants

The Youngster and Adult Education (YAE) faces major challenges related to contemporary social and economic needs. If this system once has had the stigma that was enough to teach adults and adolescents who dropped out or never went to school to read and write his name, today YAE has the challenge of providing a broad and inclusive education that enables its students to not only be part of the labor market, but also to worthy and effective a social inclusion. Due to these new goals sought by YAE, the pedagogical and management planning of this teaching modality need to be aligned with contemporary social, cultural and economic conditions. Therefore, the large-scale assessments done to diagnose the skills and competencies of students and offer portraits of the macro institutional activity, may appear as valuable tools for the development of actions and interventional projects aimed at the adequacy of management and pedagogical actions to improve the quality of education for the students and the performance of the educational systems. In this scenario, this study is meant to perform the analysis of an evaluation project on Youngster and Adult Education program as a way to demonstrate the importance of conducting an external assessment also to this type of teaching modality
A Educação de Jovens e Adultos (EJA) enfrenta grandes desafios relacionados às necessidades sociais e econômicas contemporâneas. Se algum dia essa modalidade já teve o estigma de que bastava ensinar adultos e adolescentes, que abandonaram ou nunca frequentaram a escola, a ler e a escrever seu nome, hoje a EJA tem o desafio de oferecer um ensino amplo e inclusivo que possibilite aos estudantes não só uma inclusão no mercado de trabalho, mas também uma inclusão social digna e efetiva. Devido aos novos objetivos almejados pela EJA, os planejamentos pedagógicos e de gestão dessa modalidade de ensino necessitam estar alinhados às necessidades sociais, econômicas e culturais da atualidade. Para tanto, as avaliações em larga escala que diagnosticam competências e habilidades dos estudantes e oferecem retratos macros da atuação institucional podem figurar como valiosas ferramentas para a elaboração de ações e projetos de intervenção que visem à adequação da gestão dos programas e das ações pedagógicas para a melhoria da qualidade da educação, da formação dos estudantes e da atuação de redes e sistemas educacionais. Nesse cenário, o estudo aqui apresentado tem o objetivo de analisar um projeto de avaliação na Educação de Jovens e Adultos a fim de demonstrar a importância da realização de uma avaliação externa aplicada em larga escala também para essa modalidade de ensino

A dissertação trata de um estudo de natureza didática para uma reflexão sobre a leitura na Educação de Jovens e Adultos (EJA). Enfocamos a importância da formação de alunos com inúmeras pretensões e necessidades comunicativas, carentes da formação fundamental, que buscam a escola como oportunidade para que sejam reconhecidos e para validarem conhecimentos e competências resultantes de experiências construídas ao longo da vida, fora dos bancos escolares. O estímulo à prática da leitura revelou-se um dos caminhos para melhorar o letramento e contribuir para a inclusão social. Nas relações entre linguagem, sociedade e escola, é indispensável uma prática de ensino em que se ofereça a oportunidade de o homem continuar a educação durante o tempo necessário, numa concepção democrática do conhecimento. O segmento EJA tem uma Proposta Curricular, fundamentada nos Parâmetros Curriculares Nacionais, mas considerando as especificidades e características dessa modalidade de ensino. O trabalho com diferentes gêneros e tipos de textos facilita tanto a interpretação como a produção escrita. A proposta-desafio que ora apresentamos mostrou-nos que o exercício constante, lendo e escrevendo, é fundamental para que os estudantes participem das variadas práticas sociais da língua, ao abrir um leque de conhecimentos e novos significados, levando-os a apreciar e a valer-se deste poderoso instrumento, a LEITURA. Ao refletirmos sobre aspectos pedagógicos, voltamo-nos sobre a nossa prática, como forma de avaliar o desempenho face ao processo ensino-aprendizagem que buscamos aperfeiçoar quanto aos procedimentos adotados com alunos da EJA.
This dissertation deals with a didactic study concerning a consideration about reading in the Youth and Adult Education (YAE). We focus the importance of students education with various aspirations and communicative needs, deprived of basic education, who seek the school as an opportunity to be acknowledged and to confirm knowledge and abilities acquired through experiences throughout life, outside school. Motivation for the reading practice turned out to be one of the ways to improve literacy and to contribute to social inclusion. In the relations between language, society and school, it is essential a teaching practice in which the student is offered the opportunity to carry on his education during the required period, in a democratic conception of knowledge. The segment YAE has a Curricular Proposal, based on the National Curricular Parameters, but taking into account the specifities and particulars of this type of teaching. The work with different sorts and types os texts helps interpretation as well as the written output. The challenge-proposal we now present showed us that the constant exercise, reading and writing, is essential for students to participate in the various social practices of the language, by opening a series of knowledges and new meanings, leading them to enjoy and to take advantage of this powerful instrument, READING. As we think over the pedagogic aspects, we make use of our practice as a means to evaluate the performance in view of the teaching-learning process which we try to improve as for the procedures adopted with YAE students.

This Master s Degree Dissertation sought to analyze with educators at permanent formation, the challenges to integrate the world reading of youths and adults in works with thematic project, making the relation between you - I more lively and participatory. The research thematic came to meet educators group of the researched school because it caused the reflection about real conditions of learners and their specificities since they are not children anymore (over 15 years old), excluded from school, and belonged to a disadvantaged social group. It brings the need of a knowledge approach related to everyday life and favors the schooling with collective construction of works with Projects. The investigation was accomplished at Permanent Formation of Educators of a state school of Santa Maria focused on the challenge of working with projects from educators life knowledge. The methodology used for the meetings was Investigative-formative Dialogued Circles inspired on Paulo Freire s conception in which each educator was invited to say his/her word , without embarrassment, obligation of speech, slowly loosening up and interacting with the group from the moment that feels comfortable. The present study brought as a result the need of research co authors subject self(trans)formation who see in the permanent formation of teachers possibilities of getting their pedagogical praxis better, demonstrating that educators have to search alternatives in group for permanent self(trans)formation through dialogue and opinions sharing which facilitates the appearance of new what doings mainly when integrating meaningful contents at Thematic Projects from learners real interests, and according to this world reading, being that a possibility for more lively and participatory you and I relations.
Esta Dissertação de Mestrado buscou analisar com os educadores, na formação permanente, os desafios de integrar as leituras de mundo dos jovens e adultos nos trabalhos com projetos temáticos, tornando as relações eu-tu mais alegres e participativas. A temática da pesquisa veio ao encontro do grupo de educadores da escola pesquisada, pois provocou a reflexão sobre as condições reais do educando e as suas especificidades, uma vez que estes se apresentam não mais como crianças (acima de 15 anos), tendo sido excluídos da escola e, ainda, pertencem a grupos sociais menos favorecidos. Traz a necessidade de uma abordagem de conhecimentos relacionados ao cotidiano e que favoreça a escolarização com a construção coletiva dos trabalhos com Projetos. A investigação foi realizada na Formação Permanente de Educadores de uma escola estadual de Santa Maria focada no desafio de trabalhar com projetos a partir dos saberes de vida dos educandos. A metodologia utilizada para os encontros foi a dos Círculos Dialógicos Investigativo-formativos, inspirados na concepção freireana, em que cada educador e educadora foi convidado a dizer a sua palavra , sem constrangimento, sem obrigação nenhuma de fala, podendo ir aos poucos se soltando e interagindo com o grupo a partir do momento que se sentiu a vontade para isso. O presente estudo trouxe como resultado a necessidade de uma auto(trans)formação dos sujeitos coautores da pesquisa que veem na formação permanente de professores possibilidades de melhorar a sua práxis pedagógica. Apontando que educadores tem que buscar alternativas em conjunto pela auto(trans)formação permanente, por meio do diálogo e do compartilhamento de opiniões, o que vai facilitar o surgimento de novos quefazeres, principalmente ao integrar nos Projetos Temáticos conteúdos significativos que partam dos reais interesses dos educando e de acordo com sua leitura de mundo, sendo estes uma possibilidade para que as relações eu-tu sejam mais alegres e participativas.

La rhizosphère, définie comme l'espace du sol qui entoure les racines, héberge de nombreux microorganismes à l'origine d'une grande variété d'interactions avec la plante. Au cours des dix dernières années, un grand nombre de gènes végétaux impliqués dans le dialogue moléculaire qui s'établit dans le cadre des différentes symbioses ou lors de la réponse de la plante aux parasites ont été identifiés. Les résultats de ces travaux ont notamment révélé l'existence de fortes similitudes entre les signaux et les récepteurs impliqués dans les deux types d'interactions. Toutefois, l'identification de gènes essentiels à la fois aux réponses immunitaires et mutualistes n'a pas encore été décrite, en raison notamment de l'incapacité de la plante modèle Arabidopsis thaliana à établir des interactions symbiotiques. Pour étudier l'existence d'interconnexions moléculaires entre symbioses et résistances aux parasites, la légumineuse modèle Medicago truncatula a été utlisée. En exploitant des mutants symbiotiques et deux pathosystèmes développés avec cette plante, les résultats décrits dans ce travail relatent la découverte de l'implication de plusieurs gènes avec un rôle clé dans la mise en place de la symbiose et l'immunité de la plante. L'implication du récepteur putatif aux facteurs NOD NFP et celle du facteur de transcription NF-YA1, marqueur de la symbiose fixatrice d'azote ont été particulièrement étudiées après inoculation d'Aphanomyces euteiches (Ae), un oomycète racinaire. Des approches de phénotypage complémentaires (notations des symptômes, cytologie, détection moléculaire du parasite) ont montré que NFP est impliqué dans la résistance de la plante tandis que NF-YA1 est associé à la sensibilité de celle-ci. Le comportement des mutants nfp vis-à-vis de Colletotrichum trifolii, un champignon parasite, s'est révélé similaire à celui observé vis-à-vis d'Ae, permettant de généraliser son rôle face aux organismes filamenteux pathogènes. En ce qui concerne NF-YA1, il est apparu que sa mutation implique aussi un meilleur développement des racines latérales. Son expression a d'ailleurs pu être détectée dans les primordia de racines, suggérant qu'il possède un rôle régulateur de ces organes. Des analyses transcriptomiques comparant les mutants de ces deux gènes et la plante sauvage inoculée par Ae ont révélé des modifications non seulement sur l'expression de gènes dans la signalisation et la mise en place de réponses immunitaires mais aussi dans des fonctions associées à des processus de dynamique cellulaire. Ainsi l'analyse détaillée du rôle de NFP et NF-YA1, ainsi que l'initiation de plusieurs autres travaux sur d'autres mutants symbiotiques suggèrent un nombre d'interconnexions moléculaires beaucoup plus important qu'imaginé initialement entre symbiose et immunité
Rhizophere can be defined as the soil area surrounding the roots. It contains important amount of microbes that initiate a wide variety of interactions with plants. During the last decades, numerous plant genes involved in the molecular dialog leading either to symbiotic or immune responses has been discovered. These studies unravelled strong homologies between signals and their cognate receptors involved in both types of interactions. Nonetheless, whether some genes can act in both kind of interaction is not an addressed question yet, notably beacause the model plant Arabidopsis thaliana cannot establish any kind of symbiosis. To investigate potential molecular cross-talks between symbiosis and disease resistance, the model legume plant Medicago truncatula has been used. Taking advantage of symbiotic mutants and two pathosystems developped with this plant, the results described in this work highlight roles in immune responses of several symbiotic actors. Implication of the putative Nod factor receptor NFP and of the transcription factor NF-YA1, marker of nodule development maintenance, has been particularly studied upon inoculation of the root oomycete Aphanomyces euteiches (Ae). Complementary phenotyping approaches (symptoms notations, cytology, molecular detection of parasite) showed the involvement of NFP in plant immunity whereas NF-YA1 appears as a susceptibility gene. Nfp mutants are also more susceptible to Colletotrichum trifolii, a parasitic fungus. This result indicates that the role in resistance of this gene can be generalised to filamentous microbes. In parallel, we showed that NF-YA1 disruption lead also to a better root development upon oomycete invasion and its expression was localised in lateral root primordia through histological observations, suggesting a regulation role for this gene in root architecture. Transcriptomic analyses comparing mutants and wild type plants inoculated with Ae were generated and revealed expression alterations of genes involved in signaling and immune responses but also associated to cell dynamic processes. Hence, detailed analyses of NFP and NF-YA1 roles along with others studies with symbiotic genes and Ae initiated in this work suggest an unexpected number of molecular crosstalks between symbiosis and immunity than previously thought

This work is in search line of Reading, Writing and Teaching of the Portuguese language and has as theme the study of the expansion of the degree of literacy of a person that is already literate in the Student Elementary School II of the Youth and Adults Education (YAE), through the teaching of meaningful reading, developed under school. We have as a central objective to contribute to the studies of the literacy associating it to dialogical studies and enunciates of Mikhail Bakhtin and Paulo Freire, who understand the interaction between the person s discourse and the world. We have as specific objectives to contribute to the teaching of meaningful reading, developed in the Portuguese Language classes, taught for the students (YAE), and contribute to the teaching of meaningful reading in State School, an institution in which we work and where we believe that we can develop a work dedicated to the teaching of that reading. In discussing the importance of an education geared to the meaningful reading in the classroom, we propose an analysis on the statement I love you, based on the context of interaction, the dialogism and the multiplicity of ways. We suggest, also, two activities of reading that, we believe, can be worked in the (YAE), and they consider the social dimension and discursive of language, as well as the literate practices of the involved people in any situation of speech communication. The research enabled us propose a dialogue between the (YAE), studies of literacy and the theoretical principles of Bakhtin and Freire. We, therefore, that proficient reading goes beyond the linguistic code, pure and simple, and it is effective through factors such as the context of interaction, dialogical resources and the multiplicity of ways
Este trabalho insere-se na linha de pesquisa de Leitura, Escrita e Ensino de Língua Portuguesa e tem como tema o estudo da expansão do grau de letramento de um sujeito já alfabetizado e estudante do Ensino Fundamental II da Educação de Jovens e Adultos (EJA), por meio do ensino de uma leitura significativa, desenvolvido no âmbito escolar. Temos como objetivo central contribuir com os estudos do letramento ao associá-lo aos estudos dialógicos e enunciativos de Mikhail Bakhtin e de Paulo Freire, que compreendem a interação discursiva entre o(s) sujeito(s) e o mundo. Temos como objetivos específicos contribuir para o ensino da leitura significativa, desenvolvida nas aulas de Língua Portuguesa, ministradas para os(as) alunos(as) da EJA, e, contribuir para o ensino da leitura significativa na escola pública, instituição em que trabalhamos e onde acreditamos que seja possível desenvolver um trabalho voltado para o ensino dessa leitura. Ao discutirmos a importância de um ensino voltado à leitura significativa em sala de aula. Assim propomos uma análise em torno do enunciado eu te amo, tendo como base o contexto de interação, o dialogismo e a multiplicidade de sentidos. Sugerimos, ainda, duas atividades de leitura que, acreditamos, possam ser trabalhadas na EJA, e que consideram a dimensão social e discursiva da linguagem, bem como as práticas letradas dos sujeitos envolvidos em qualquer situação discursiva de comunicação. A pesquisa realizada nos possibilitou propor um diálogo entre a EJA, os estudos do letramento e os princípios teóricos de Bakhtin e de Freire. Defendemos que a leitura proficiente vai além do código lingüístico, puro e simples, e que ela se efetiva por meio de fatores como o contexto de interação, os recursos dialógicos e a multiplicidade de sentidos

This work intended to talk about the posture of the Portuguese teacher while developing activities in the writing classes, in particular, in classrooms of high schools of the Youth and Adult Education (YAE). Today, it is said very in the knowledge that the student, in activities of writing, must possess to constitute its text of direction. In this scene, its important to comprehend the paper and the work of the professor to develop activities where the writing become one significant practice. Thus, the objetive was to demonstrate that the teacher, knowing the writing class as social event and the text produced by the student like a moment of this event, can establish with bigger security the ways for the teaching and learning of the written production. In order to develop this investigation, the studies of the Critical Discourse Analysis will provide to one better agreement of the dynamics of relations between speech and society, reproduction of power and social inaquality, inherent to the school, the lessons of writing and its especificidades. The CDA, considering the linguistic in the social interior, where the understanding of the social and psychological construction of the individuals is basic, demonstrate that the practice of writing in the school are not practices without historical and social context, but yes inserted in discourse and social practice of accomplishment. With the analyses was possible to understand the form how a student/author acts, represent the world and identify oneself inside text; this way of seeing the written production is what will allow to a teacher a clear and solid work of the writing competences and skills
Este trabalho discorre sobre a postura do professor de Língua Portuguesa ao desenvolver atividades de produção textual escrita nas aulas de redação, em particular, salas do Ensino Médio da Educação de Jovens e Adultos (EJA). Hoje, fala-se muito nos conhecimentos que o aluno, em atividades de redação, deve possuir para constituir seu texto de sentido. Nesse cenário, coloca-se em questão o papel e o trabalho do professor para desenvolver atividades onde a escrita seja uma prática significativa. A partir disso, o objetivo foi demonstrar que o professor, ao entender a aula de redação como um evento social e o texto produzido pelo aluno como um momento deste evento, pode estabelecer com maior segurança os caminhos para o ensino e aprendizagem da produção escrita. Para tal, uma abordagem a partir dos estudos da Análise Crítica do Discurso proporcionou um melhor entendimento da dinâmica de relações entre discurso e sociedade, reprodução de poder e desigualdade social, inerentes à escola, as aulas de redação e suas especificidades. A ACD, considerando o lingüístico no interior social, onde é fundamental a compreensão da construção social e psicológica dos indivíduos, mostrou que as práticas de redação realizadas na escola não são práticas textuais descontextualizadas histórica e socialmente, mas sim inseridas em práticas discursivo-sociais de realização. As análises realizadas possibilitaram compreender a forma como um aluno/autor age, representa o mundo e se identifica dentro de um texto; essa maneira de ver a produção escrita é o que permitirá ao professor um trabalho claro e consistente das competências e habilidades escritoras

Human cytosolic monothiolglutaredoxin-3 (GLRX3) is a protein essential for the maturation of cytosolic [4Fe−4S] proteins. We show here that dimeric cluster-bridged GLRX3 transfers its [2Fe−2S]2+ clusters to the human P-loop NTPase NUBP1, an essential early component of the cytosolic iron−sulfur assembly (CIA) machinery. Specifically, we observed that [2Fe−2S]2+ clusters are transferred from GLRX3 to monomeric apo NUBP1 and reductively coupled to form [4Fe−4S]2+ clusters on both N-terminal CX13CX2CX5C and C-terminal CPXC motifs of NUBP1 in the presence of glutathione that acts as a reductant. In this process, cluster binding to the C-terminal motif of NUBP1 promotes protein dimerization, while cluster binding to the N-terminal motif does not affect the quaternary structure of NUBP1. The cluster transfer/assembly process is not complete on both N- and C-terminal motifs and indeed requires a reductant stronger than GSH to increase its efficiency. We also showed that the [4Fe−4S]2+ cluster formed at the N-terminal motif of NUBP1 is tightly bound, while the [4Fe−4S]2+ cluster bound at the C-terminal motif is labile. Our findings provide the first evidence for GLRX3 acting as a [2Fe−2S] cluster chaperone in the early stage of the CIA machinery. Iron-sulfur (Fe-S) clusters are among the most versatile cofactors in biology. Although Fe-S clusters formation can be achieved spontaneously in vitro with inorganic iron and sulfur sources, the in vivo behaviour is more complex and requires the so-called Fe-S biogenesis machineries. In the cytosol, the biogenesis of Fe-S proteins is assisted by the cytosolic Fe-S protein assembly machinery, which comprises at least thirteen known proteins, among which there are human ORAOV1 and YAE1. A hetero-complex formed by the two latter proteins facilitates Fe-S cluster insertion in the human ABC protein ABCE1 within a chain of binding events that are still not well understood. In the present work, ORAOV1 and the YAE1-ORAOV1 complex were produced and their structural and cluster binding properties spectroscopically investigated. It resulted that both ORAOV1 and the YAE1-ORAOV1 complex are characterized by well-structured alpha-helical regions and by unstructured, flexible regions, and are both able to bind a [4Fe-4S]2+ cluster. Bioinformatics and site-directed mutagenesis studies indicated that ORAOV1, and not YAE1, is the protein involved in [4Fe-4S]2+ cluster binding in the hetero-complex. ORAOV1 has indeed a conserved cluster-binding motif able to coordinate a [4Fe-4S] cluster. Overall, our data suggested that the YAE1-ORAOV1 complex might actively participate in the Fe-S cluster insertion into ABCE1 thanks to the [4Fe-4S]2+ cluster binding properties of ORAOV1.

Oxidative stress mechanism in yeast presents an innovative pathway to understand in creating the next generation of antifungal drugs. Rgc1 and Rgc2 are paralogous proteins that regulate the Fps1 glycerol channel in hyperosmotic stress. Hyperosmotic conditions lead Hog1 MAP kinase to phosphorylate Rgc2 and cause its dissociation from Fps1, allowing the channel to close and protect the cell from damage. Rgc2 contains pleckstrin homology (PH) domains broken up by long insertions and more phosphorylation sites than targeted by Hog1 in response to hyperosmotic stress. Since none of the other MAP kinases in yeast were seen to phosphorylate Rgc2 during oxidative stress, it is thought that Rgc2 may bind to other proteins. In this study, the sensitivity of a strain deleted for both RGC1 and RGC2 was compared to strains with single deletions in either gene in response to oxidative stress. Having deletions in both RGC1 and RGC2 caused increased sensitivity to hydrogen peroxide whereas strains with deletions in either gene seemed unaffected, correlating with the fact that Rgc1 and Rgc2 are paralogous proteins, able to recover each other's functions. A second analysis compared mutated Fps1 (fps1∆-FKSV) and a strain with deletions for both RGC1 and RGC2 (rgc1/2∆). The fps1∆-FKSV strain has four amino acid substitutions in the C-terminal region where Rgc2 binds to Fps1. While both strains grew less than wild-type in hydrogen peroxide, the rgc1/2∆ strain was more sensitive suggesting that Rgc1/2 has an additional role in oxidative stress. To identify the oxidative stress function of Rgc1/2, a genomic overexpression library was transformed into the rgc1/2∆ strain and used for a suppressor screen in the presence of hydrogen peroxide. Although the screen revealed a manageable amount of 49 candidates, only four produced sequences that spanned a protein-encoding region. The candidate plasmids were transformed back into the rgc1/2∆ strain for preparation of a sensitivity assay which showed that the colonies did not survive any better than the starting rgc1/2∆ strain. Without a plausible plasmid candidate, we decided to look into the effect of YAP1 on the rgc1/2∆ strain. Yap1 is a transcription factor known to activate many genes in oxidative stress. Two forms of YAP1 were transformed into rgc1/2∆: wild-type YAP1 and YAP1-A627E which contains a mutation in the nuclear export signal. Compared to the controls, YAP1-A627E allowed the rgc1/2∆ strain to grow at 1.5mM H2O2 while wild-type YAP1 did not. This result showed that a constitutively nuclear Yap1 can overcome deletions in RGC1 and RGC2. It also suggested that an increased activity in the nucleus was important in hydrogen peroxide resistance and another suppressor screen of rgc1/2∆ was performed looking for spontaneous mutations in the genomic DNA. The screened colonies were tested for their survival on hydrogen peroxide but their resistance appeared to be transient. We have shown Rgc1 and Rgc2 to be important cellular components in oxidative stress in addition to hyperosmotic stress. Further research on Rgc1/2 would provide invaluable knowledge on oxidative stress protection in yeast and a better foundation on which to build antifungal drugs.

Array comparative genomic hybridization (array-CGH) was used in this study to analyze the chromosomal aberrations in 9 gastric cancer cell lines. Our results showed good concordance with those of conventional CGH. We correlated the results from array-CGH with expression profiling and found some novel and independent target genes which deserved further confirmation.
Gastric cancer is one of the most common malignancies worldwide and is the second most frequent cause of cancer related death. A variety of genetic and epigenetic aberrations underlie development abnormality of gastric cancer.
Taken together, our findings supported YAP1 is a functional oncogene in gastric cancer. We provided the first evidence that YAP1 exerted the oncogenic function by enhancing the capacity to activate the early response gene pathway. YAP1 could be a prognostic biomarker and potential therapeutic target for gastric cancer.
The study was focused on the putative oncogene Yes-associated Protein 1 (YAP1) located in 11q22.1. Up-regulation of YAP1 was observed in 92.3% of gastric cancer by immunohistochemistry (IHe) on gastric cancer tissue microarrays. YAP1 nuclear accumulation correlated with cancer specific survival. In addition, multivariate Cox regression showed that YAP1 was an independent predictor of short disease specific survival time for patients with early stage gastric cancer (P=0.042) in addition to T stage ( P=O.038). Knockdown YAP1 in gastric cancer cell lines MKN1 and AGS resulted in a significant reduction in proliferation, anchorage-dependent colony formation, cell invasion and cell motility. Ectopic YAP1 expression in MKN45 cells promoted anchorage-independent colony formation, induced a more invasive phenotype and accelerated cell growth both in vitro and in vivo. Microarray analysis highlighted the alteration of MAPK pathway by YAP1. We confirmed a constitutive activation of RAF/MEKJERK in YAP1-expressing MKN45 cells and further demonstrated that YAP1 enhanced serum/EGF induced c-Fos expression in gastric cancer cells. Furthermore, we demonstrated that ectopic MST1 promoted phosphorylation and cytoplasmic translocation of YAP1 and subsequently quenched the oncogenic function of YAP1 in the nucleus.

Background   Obesity has now become one of the issues that need to be addressed by countries around the world. In addition to affecting the quality of life and psychological burden of individuals, obesity can even cause problems in the social and economic losses of the entire family and the country. Many studies have found that overweight and obesity are associated with chronic diseases such as cardiovascular disease, cancer, metabolic syndrome, hypertension, and hyperlipidemia. However, in China, eight of the top ten causes of death in 2017 are highly correlated with obesity. Obesity is a very important public health problem today.   The study pointed out that bad living habits and the environment are one of the important factors that cause obesity. Therefore, this study returns to the basic level and intends to explore the changes in the fat environment of Taichung City from different perspectives. Purpose   The purpose of this study is divided into: multi-faceted Taichung City, 2014-2015 workplace fattening environment changes. Method   This study conducted a secondary database analysis. The source of the data was the 2014-year and 2015-year “Workplace Fat Obesity Environmental Assessment Questionnaire” of the Taichung City Government Health Bureau. The Taichung City Government Health Bureau conducted a two-year fattening environment survey for 170 workplaces. This study divided the workplace fattening environment into 8 strategies and analyzed the changes. The statistical method uses SPSS 20.0 version for descriptive statistics and dual variable analysis. Result   The total number of samples in this study was 56, of which women were the mainstays, and the education level was higher in universities or colleges (78.6%). In healthy life surveys, higher in middle school or high school (62.5%), in the presence or absence In the case of chronic diseases, the proportion of patients without chronic diseases is higher (85.7%); on the contrary, the proportion of chronic diseases in healthy life surveys is higher (63.5%). Bivariate analysis This study found that in the 2014-year and 2015-year workplace fattening environment changes in Taichung City, the eight major-oriented aspects were discussed. Although most of them did not meet the statistical standards, the two-year fattening environment involved in workplace changes. In comparison, people still have basic cognitive attitudes, but the public still chooses higher risk factors for obesity. Conclusion and Suggestion   After the intervention of the obesity environment in the workplace, the results of this study show that the people's eating habits have an improvement trend, and it is impossible to control the incidence of obesity. If China can continue to conduct the "obesity prevention and evaluation tool" survey, accumulate long-term tracking. According to the data, it is suggested that future research can use the tracking method to explore the factors related to the fat environment in the workplace, so as to improve the improvement of the fattening environment, in order to effectively help China solve the problem of obesity related problems, and also to explore the degree of obesity of the Chinese people through this study.
