Résumé :
Abstract In the early 1950s, East German officials at the storied Maxhütte steel mill in Thuringia collected short CVs or “life stories” (Lebensläufe) written by approximately 370 blue- and white-collar workers who had recently become Stakhanovite “activists”. These documents, which all contain the same basic biographical information about the authors – from their socioeconomic background to their political activities – shed light on a group that has received little systematic scholarly attention, namely, ostensibly loyal and ordinary East Germans at the grass roots. Their early support of the SED state and its economic goals ensured the longer-term stability of a largely unloved regime. These valuable documents thus provide important clues for understanding the puzzling political and economic viability of the German Democratic Republic.

Résumé :
In this essay, I discuss specific ways in which the Odyssey ensures its reception by broader and future audiences by employing both professional and amateur storytellers and internal listeners as guarantors of the story's viability. My focus here is on the differences between Demodocus’s professional storytelling and Odysseus's nominally nonprofessional storytelling. While the former serves to please groups of an audience who expect “safe” pleasures by listening to traditional epic tales, the latter opens an affective space between listeners and the storyteller, where the listeners’ affects are mobilized by Odysseus’s unpredictable and perhaps less skillful ways of narrating stories. Odysseus’s amateurism (as I would call it) instigates the audience’s desire to listen more and becomes a successful strategy to captivate the audience’s minds. In this age and culture often defined by the “narrative turn,” many of us are interested in telling good stories that can impact broad audiences. This much-loved, ancient work of fiction can teach us various techniques of good storytelling that can move the hearts of generations of audiences.

Résumé :
Abstract Bigleaf maple is a common hardwood in western Oregon and Washington. Stored seeds from this species are generally believed to lose viability within a few weeks. In this study, two experiments were conducted with seeds collected in 1986 and 1987 in the Oregon Coast Range west of Corvallis, OR. Seeds collected in 1986 had a field moisture content (percentage of dry weight) of 49% and in 1987, 16%. The viability of the seed with the lowest moisture content declined from 75 to 60% during 1 year of storage at 1°C; the seed with the highest moisture content declined from about 90 to less than 10% viability. Storing the seeds at -10°C did not increase viability. Stratification of seeds for more than 60 days did not appear to increase germination. West. J. Appl. For. 5(2):52-55, April 1990.

Abstract A series of controlled-pollination tests with fresh pollen and pollen stored for up to 10 years demonstrates the conditions required to maintain high pollen viability and vigor. During handling, freshly extracted pollen is highly resistant to exposure to temperatures up to 50°C for as long as 24 hours. Moisture content must be reduced to less than 10% prior to storage. It can then be held in a refrigerator at 3°C for up to 1 year in a container that maintains low moisture content. For longer storage, dried pollen should be placed in tightly closed glass or plastic bottles and held at -20°C. Pollen can be stored in vacuum-sealed ampules at 3°C for at least 10 years. South. J. Résumé :
Abstract Seeds of Hippeastrum × hybridum Hort. were subjected to dehydration, freezing, and storage for 12 months at various temperature and humidity levels. Amaryllis seed viability was unaffected by dehydration that removed 97% of available moisture content at seed harvest. Seeds were tolerant of freezing, but germination was reduced gradually as freezing temperatures decreased from 0° to −80°C. Moisture content at freezing influenced loss of viability; the lower the moisture content the higher the germination percentages. Temperature and relative humidity during seed storage governed seed viability loss rate. No measurable losses in viability resulted from storing seeds at 11% or 52% RH at 5° or 15°, respectively, for 12 months, but viability was significantly decreased when seeds were stored at these humidities at 25° or 35°. Viability decreased substantially or was lost after 3 months of storage at 75% and 95% RH and 25° and 35°.

Résumé :
Purpose: The main objective of this paper was to assess whether lifestyle audit as an anti-corruption tool is viable. Methodology: The study was based on a desk review of existing studies and documented statistics. Further, legal framework and case laws were cited. A narrative analysis was performed and at this point the information was interpreted by comparing the results with the results of other empirical studies. This information was interpreted using the “stories in the stories” and linked to the existing literature. Results: The study found that lifestyle audit is a viable tool for fighting corruption. However, there are challenges in the legal framework, particularly on the implementation. The study noted that the viability of lifestyle audits depends on judicial interpretation, political will and more importantly addressing of the current software and hardware issues existing in the current legal framework. Recommendation: The study calls for political will to ensure the Lifestyle Audit Bill 2019 is not watered down by judicial interpretation. It further recommends that the political will should be accompanied by executive action to fight corruption through wealth declaration.

Résumé :
ABSTRACT: Macauba palm stands out for having favorable features to biodiesel production such as the high oil content of its fruit. Considering the great potential of the species and their applicability in the renewable energy field, it becomes indispensable to establish the right conditions for storing the seeds for propagation purpose. The aim of this research was to evaluate the effect of seed moisture content, packaging, and storage conditions such as temperature and relative humidity on the quality of seeds from Minas Gerais State, during a 12-month storage period. The research had two independent assays: (I) the seeds were stored with three moisture contents/ranges 4.0≤6.0%; 6.0≤8.0% and 8.0≤10.0% in impermeable packages, under room temperature and at 10ºC; (II) seeds with approximately 5.9% of moisture content were stored in three different types of packages: a) permeable, b) semi-permeable and c) impermeable. Three storing conditions were tested: a) room temperature and RH under laboratory conditions; b) 15ºC and 45% RH; c) 20ºC and 55% RH. Water content, germination rate and germination speed index were evaluated at 0, 4, 8 and 12 months of storing. The best germination results were obtained with the moisture range of 6.0≤8.0%, with seeds kept at room temperature; while the seeds stored at 10ºC, regardless the moisture range, did not survive. The stored seeds with 5.9% moisture content and at both 15ºC/45%RH and 20ºC/55% RH conditions, independently of the package type used, showed the best results. Thus, macaw palm seeds can be classified as intermediates seeds.

Ultracold preservation is widely used for storage of genetic stocks of Caenorhabditis elegans. Current cryopreservation protocols are vulnerable to refrigeration failures, which can result in the loss of stock viability due to damage during re-freezing. Here we present a method for preserving worms in a dehydrated and frozen form that retains viability after multiple freeze-thaw cycles. After dehydration in the presence of trehalose or glycerol, C. elegans stocks can be frozen and thawed multiple times while maintaining viability. While both dauer and non-dauer larvae survive desiccation and freezing, the dauer defective mutant daf-16 does not survive desiccation. Our technique is useful for storing stocks in a manner robust to freezer failures, and potentially for shipping strains between laboratories.

Résumé :
Using a battery on a household level has become easier after the launch of Tesla’s Powerwall. Storing electricity during daytime’s PV overproduction or charging the battery during night with an attractive tariff is the most prominent applications. This paper explores the economic impact of the usage of residential battery storage combined with solar photovoltaics (PV) based on real load data from Northern California, USA. A data-driven, deterministic model to benchmark electricity cost savings for single households is presented and the financial viability of such systems is scrutinized for California. Our results indicate that under current capacity and price points, battery systems have limited financial viability and have a payback period exceeding 20 years in most cases. We deepen our analysis and compare the results of our deterministic model to that of a stochastic model to demonstrate that for an hourly time resolution the deterministic model provides an adequate benchmark for estimating cost (within 3%) savings with a short (1/60th) computation time.

Résumé :
ABSTRACT The medium for storing biological tissues is of great importance for their optimal use in surgery. Glycerin has been proven efficient for storing diverse tissues for prolonged time, but the preservation of the bovine amniotic membrane in glycerin 99% at room temperature has never been evaluated to be used safely in surgical procedures. This study evaluated the preservation of 80 bovine amniotic membrane samples stored in glycerin 99% at room temperature. The samples were randomly divided evenly into four groups. Samples were microbiologically tested after 1, 6, 12 and 48 months of storage. The presence of bacteria and fungi in the samples was evaluated by inoculation on blood agar and incubation at 37 ºC for 48 hours and on Sabouraud agar at 25 ºC for 5 to 10 days. No fungal or bacterial growth was detected in any of the samples. It was concluded that glycerin is an efficient medium, regarding microbiology, for preserving pre-prepared bovine amniotic membrane, keeping the tissue free of microorganisms that grow in the media up to 48 months at room temperature.

This paper present the results of (experiments and models) biosynthesized prodigiosin (PG) released from an implantable biomedical device on the viability of cancer cells. The implantable biomedical devices were obtained from poly-di-methyl-siloxane (PDMS) packages with well-controlled micro-channels and drug storage compartments, along with a drug storing polymer core (which contains thermosensitive Poly (N-isopropylacrylamide)(PNIPA)-based gels). The results were compared with drugs elution from devices loaded with paclitaxelTM. The effects of localized release of PG and paclitaxel (PTx) on cell viability were elucidated via clonogenic assay testing on MDA-MB-231 breast cancer cell line. The effects of PG and PTx released were also tested over a range of temperatures (37-45 ̊C) in which localized hyperthermia is applicable. The trends in the results were analysed using statistical models before discussion their implications for localized treatment of breast cancer.

Résumé :
<p>Soybean seed viability declines during seed storage. Soybean seed deteriorates rapidly, affected by its high protein content and often high humidity in the tropical environment. This research was aimed to develop dynamic model of soybean seed viability in an open storage. The study was conducted in three stages, namely: 1) desk study, 2) seed storage experiment, 3) development of seed storage model, simulation and verification of the model. The second stage of the experiment consisted of soybean seed storing and germination testing using completely randomized design. Treatments were three initial moisture content (7-8%, 9-10% and 11-12%) and four varieties of soybean (Anjasmoro, Wilis, Detam-1 and Detam-2). The results showed that the seed behaviour during storage period were affected by initial seed moisture content, initial viability, varieties and environmental condition. Therefore, seed moisture content, initial viability and varieties may be used as input model. Moisture content, integreting seed respiration, electric conductivity and seed storability vigor (VDSDB) were as model output. Simulation of Seed Storability Vigor Prediction Model with Model Construction Layer-Stella (MCLS) using relative humidity (RH), temperature, seed permeability, initial moisture content and initial viability as input model could logically predict the seed moisture content and seed storability vigor (VDSDB).</p>

Seed development of tea was studied to identify the maturity index and the optimal time of seed collection. After harvest, the moisture content (28–30%fresh weight basis) of mature seeds, which germinated 100%, declined progressively (19% moisture content) after shedding, with a decrease in seed germination and viability. However, this viability loss could be prevented to some extent by storing seeds within intact fruits. The maximum rate of seed dry matter accumulation coincided with the accumulation of starch in the embryos and seeds at stage 8, the embryo maturation phase. Although the embryo abscisic acid (ABA) content was highest at stage 8, free ABA declined in the tea embryos throughout the remainder of the seed maturation cycle.

Abstract The microbiological profile illustrates the existence and growth of microflora in fermentation products, such as concentrate yogurt. The study aimed to identify and evaluate microflora in yogurt paste observed from the total bacteria, total lactic acid bacteria (LAB), total yeast, LAB viability, and level of acidity. Concentrate yogurt was made of full-fat milk and low-fat milk that was fermented and hung for 24h and stored at a low temperature for observation on day 0,15, 30, 45, and 60. The data were subjected to a Generalized Linear Model (GLM) followed by the Duncan test. The result showed significant (P<0.05) changes in the total bacteria, total LAB, total yeast, LAB viability dan acidity (pH) of the fermentation products; however, no significant difference (P>0.05) was identified regarding the use of raw material. Storage made the fermented products suffer from the decreased total bacteria, LAB dan its viability, but there was an increase in the total yeast and pH. Conclusively, storing concentrate yogurt in a cool place for 60 would decrease the microflora profile (total bacteria, LAB dan LAB viability) but increase the total yeast and pH value.

Résumé :
Cytotoxicity, measured as inhibition of cell growth of cultured cell lines, is a widely used method for testing the safety of biomaterials and chemicals. One major technical disadvantage with this method is the continuous routine maintenance of the cell lines. We decided to investigate the possibility of storing stock cultures of fibroblasts (L-929) in an ordinary refrigerator as a means of reducing the routine workload. Stock cultures of the mouse fibroblast cell line L-929 were prepared in plastic vials with Eagle's minimum essential medium. The vials were stored in a refrigerator at 4–10°C for periods of 7–31 days. The condition of the cells after storage was determined as cell viability, cell growth and the toxic response to acrylamide, measured as cell growth inhibition. We found that the L-929 cell line can be stored for 2–3, weeks with a viabilty > 90% and a cell growth of about 95%, compared to L-929 cells grown and subcultured in the normal manner. The results also show that the toxic response to acrylamide, using refrigerator stored L-929 cells, corresponds to that of control L-929 cells. We concluded that it is possible to store L-929 cells in a refrigerator for periods of up to 3 weeks and still use the cells for in vitro cytotoxic assays.

Résumé :
This research proposes sociological, linguistic, folklore (psychoanalytic and structuralsemiotic) approaches to the analysis of the contemporary anecdote about adultery. Academic interpretations of satirical texts are quite different, but they allow us to explain the popularity and cultural reproduction of anecdotes about married life, gender stereotypes, the reasons for the women demonizing and men derision, as well as the modern specifics of a joke and recreation of its laughing effect. However, the viability of such stories about female infidelity is connected not only with the communicative situation. The article also analyzes the unified structure of the plot, developing around a trick, and the connection between medieval fabulous stories about “women’s tricks” and modern jokes with a typical beginning “the husband returns from a business trip”. Such anecdotes often use the same plots with a trick, motifs and formulaic beginning, as well as anecdotal stories about women’s cheating. In a contemporary joke, there is no explicitly pronounced didactics and morals, but it has some similarities with fabulous stories – exposure of its fictional status, formulaic beginning, plotting caused by tricks and savvy of characters, comic denouement and laughing effect.

Abstract: Seeds storing under appropriate conditions reduces the speed of the seeds deterioration process. The aim of this study was to evaluate pre-packing cooling and types of packages in the maintenance of the physiological quality of soybean seeds during storage. The completely randomized experimental design was used, with four replications, considering a factorial arrangement involving or not the pre-packing cooling of the seed mass, three types of packages (multi-layer paper bag, big bag, and polyethylene container), and five storing periods (0, 2, 4, 6 and 8 months). The physiological quality was evaluated by the tests of first germination count, germination, accelerated aging, electrical conductivity, emergence speed index and emergence. The use of packages with larger amount of seeds (big bag and polyethylene container) did not favor the maintenance of the pre-packing cooling temperature. The types of packages did not show significant differences in the seeds viability during storage, up to eight months, regardless the pre-packing cooling. The cultivars seeds showed different tolerance to storing, i.e. seeds of the cultivar TMG 1176 had lower storing potential comparing to the seeds from the cultivar SYN 9074.

In recent decades, probiotic products have been increasingly used to prevent certain gastro-intestinal and urogenital disorders, to improve the general condition of the body and as a supplement to pharmacological therapy. They are most often registered as dietary supplements, and less often as drugs in the form of capsules, powders, and solutions. Optimization of technological processes of production and packaging of these products aims to maintain probiotic characteristics while adhering to all criteria during production that ensure quality, bioavailability and optimal therapeutic effects. In this paper, the importance of choosing primary and secondary packaging materials was explored with the aim of preserving the viability of probiotic cells in capsules for two years, i.e. during the shelf life. By comparing the applications of polyvinyl chloride (PVC) and PVC/polyvinylidene chloride/polyethylene blister foils, better protection of probiotic cells was observed by applying the multilayer foil. In addition, in this research, further improvements of probiotic cell protection were achieved by applying a secondary packing-flow pack bag with inert gas for storing multilayered blisters.

Résumé :
Abstract Artificial insemination supports the prevention of infectious dispersion in organisms which share the same environment. Meanwhile, spermatozoa usually experience a decrease in quality during processing due to the peroxidation process. This study aims to identify the effect of vitamin E antioxidant addition in goat milk diluent on the quality of thin-tailed sheep semen after storing at 5°C for five days. A total of 4 male thin-tailed sheep were used and the treatments consisted of controls, namely goat milk sperm diluent without vitamin E antioxidant (P0), and others with the addition of vitamin E at a dose of 2 gL-1 diluent (P1); 3 gL-1diluent (P2); and 4 gL-1 (P3). Moreover, the variables observed include individual motility, viability, and intact plasma membrane of spermatozoa after storing at 5°C for five days. The data were analyzed statistically using analysis of variance (ANOVA) with a completely randomized design in one-way. The results showed that the addition of vitamin E antioxidant to goat milk diluent during sperm dilution had a significant effect (P < 0.05) on the motility, viability, and intact plasma membrane. Spermatozoa motility at P0, P1, P2, and P3 were 44.0%, 48.2%, 50.4%, and 53.2%, while the viability was 48.2%, 51.4%, 54.2%, and 56.0%, respectively. Additionally, the intact plasma membranes at P0, P1, P2, and P3 were 41.0%, 46.6%, 48.2%, and 51.4%, respectively. The addition of antioxidant vitamins in goat milk diluent with a concentration of 4 gL-1 produced the best results on sperm quality after cooling. Based on the results, it was concluded that the addition of vitamin E antioxidant to the sperm diluent can improve the quality of thin-tailed sheep chilled semen.

Abstract A comparative study of the effect of different storage regimes of seeds (at + 5 ° С, -20 ° С and under deep freezing conditions at -196 ° С) of a valuable bast fiber culture - kenaf - Hibiscus cannabinus L. on their viability and the development of plants from them in field conditions. Freezing increases the laboratory and field germination of kenaf seeds, and also increases the energy of their germination. It did not cause the appearance of non-viable, ugly seedlings and plants, as well as their death. Plants went through a full development cycle and set full-fledged seeds. Biometric parameters of plants obtained from thawed seeds and control ones are very close. Résumé :
Following insemination, ram spermatozoa are transported to the isthmus region of the oviduct where they bind to the oviductal epithelial cells (OEC), remaining viable for several hours. The aim of the present study was to begin to decipher which component(s) of the ewe oviduct actively participates in maintaining the viability of ram spermatozoa. A series of experiments was conducted to investigate whether: (1) soluble OEC apical plasma membrane proteins (sAPM) isolated from ewes prolong survival of ram spermatozoa over an extended (48 h) coincubation period at 39°C; (2) a recombinant form of one of these oviductal proteins, namely heat shock 70 kDa protein 8 (HSPA8), prolongs survival of ram spermatozoa; and (3) pretreatment with HSPA8 antibody compromises the ability of sAPM to prolong the survival of ram spermatozoa. Both sAPM and recombinant HSPA8 had a beneficial effect on the viability of ram spermatozoa during coincubation, although both these effects were dose dependent. In contrast, pretreatment with HSPA8 antibody significantly negated the ability of sAPM to maintain the viability of ram spermatozoa. These findings suggest that HSPA8 is an active component of the ewe oviduct that participates in maintaining the viability of ram spermatozoa. This is a potentially valuable observation given that there is a great deal of room for improving existing diluents for storing fresh ram semen.

Résumé :
This paper describes a research investigation on a project led by two libraries, Hamilton Public Library and McMaster University Library, in Hamilton, Canada, concerning the use of proximity-based technologies to share digital stories about a city’s culture. Proximity-based technology systems, such as iBeacons, allow users to receive information automatically when they are close to a physical spot. The project involved the setup of iBeacons that disseminated digital stories pertaining to Gore Park – a prominent historical park in the heart of downtown Hamilton. To test the viability of using iBeacon technologies to raise interest in a city and promote appreciation for a city’s cultural heritage, a pilot study was conducted. The study included one-on-one interviews and a short survey with 50 participants from the general public immediately after these participants used an iBeacon app to experience digital stories about Gore Park. Findings suggest iBeacons are viable tools to share city cultural heritage stories that yield improved perceptions of a city and greater appreciation for a city’s culture and history. Participants were appreciative of the digital stories and the iBeacon app. All participants mentioned that they learned something new about the city and that the app was very informative. Findings indicate that individual differences are important and can affect not only the acceptance and use of an iBeacon digital storytelling app, but also the extent to which the app can promote interest in a city and appreciation for a city’s cultural heritage.

Résumé :
Abstract Do the media cover men and women politicians and candidates differently? This article performs a systematic analysis of 90 studies covering over 25,000 politicians in over 750,000 media stories, and presents the accumulated knowledge in a comprehensive theoretical framework. The paper shows that there is a gender bias in the amount of coverage of politicians in proportional electoral systems, where women politicians lag behind men in media attention, but that, surprisingly, this gender bias is absent in majoritarian electoral systems. In addition, we systematically review gender differences in the content of media reports on political candidates, such as differences in attention to private life and family, viability and horse-race coverage, issue coverage, and gender stereotypes. Overall, women politicians receive more attention to their appearance and personal life, more negative viability coverage, and, to some extent, stereotypical issue and trait coverage. We conclude by pointing out promising avenues for future research.

Résumé :
Fifteen male New Zealand rabbits were used in this study, with the aim of storing their platelet-rich plasma (PRP) for 30 days at 4-6 °C to investigate its conservation and viability during this period. Thirty samples of PRP were prepared and sorted into three equal groups (G1, G2, and G3), and every three days a sample was taken out for evaluationof the number of platelets, mean platelet volume (MPV), pH of the plasma, aggregation post addition of calcium thromboplastin, and for the presence of bacterial and fungal contamination. Results suggested that, for the number of platelets, there was no linear relationship over time. However, when comparing the number of platelets pre-storage to that post-storage, a statistical difference was observed. The hemogram MPV variables, pre and post-storage, also did not relate with time however, there was a statistical difference between the MPV of hemogram and MPV pre-storage, and between MPV pre-storage and MPV post-storage. From the pH evaluation, no influence of time on the variables was found, but statistical differences were found in the samples after storage between 30 and 6 days, 30 and 24 days, and 30 and 27 days. Platelet aggregation occurred within twenty seconds in all samples, independent of storage time. There was no growth of bacteria or yeast in any sample; however, mold growth occurred in the samples stored for 21 days from G1 and G3. It can be concluded that the PRP of rabbits can be stored in 4-6 °C refrigeration for up to18 days and still maintain the number of platelets, with no significant pH alteration or bacterial or fungal contamination.

We evaluated the fate of Listeria monocytogenes on commercial pork scrapple, a regionally popular, ready-to-eat (RTE) meat. We also conducted an informal survey to address consumer practices for storing and reheating scrapple. Of the 129 consumers who responded to at least one of the eight questions posed in the survey, about half (46.4%; 52 of 112) considered scrapple RTE, the majority (69.7%; 76 of 109) stored it in the refrigerator, and all (100%; 112 of 112) preferred to reheat it prior to consumption. Most respondents (83.9%; 94 of 112) reheated the scrapple by pan frying for 1 to 10 min at medium to high temperature. To study pathogen behavior, slices of pork scrapple were surface inoculated with a five-strain cocktail of L. monocytogenes (ca. 2.0 log CFU/g), vacuum sealed, and stored for up to 60 days. Pathogen levels increased to 8.9, 9.5, and 9.9 log CFU/g after 44 (4°C), 21 (10°C), and 5 (21°C) days, respectively. When slices 1.3 cm (ca. 55 g) and 1.9 cm (ca. 85 g) thick were surface inoculated with L. monocytogenes (ca. 7.0 log CFU/g) and then reheated in a skillet (191°C) for 0.5 to 4 min per side or to target instantaneous internal temperatures of 48.9 to 71.1°C, it was possible to achieve pathogen reductions ranging from ca.

To determine the long-term effects of a lactic acid rinse on viability and recovery of pathogens, Salmonella Hadar was isolated from poultry and bioluminescent constructs obtained by transformation with the lux (CDABE) gene cassette from Photobacterium phosphoreum. Results indicated that the transformed Salmonella Hadar lux was otherwise phenotypically similar to the wild-type strain. Viability studies were performed by measuring luminescence following lactic acid treatment of turkey breast and subsequent storage at −12, 0, 5, and 10°C. The ability of the S. Hadar lux strain to recover was determined by monitoring light output after incubation at 22°C for 10 h. The results showed that metabolic activity was significantly (P &lt; 0.05) affected by lactic acid and by storage temperatures of −12, 0, and 5°C. The lowest recovery rate was observed after rinsing with lactic acid and storing at 5°C. The study demonstrated that bacterial bioluminescence is an effective way of monitoring in “real time” the ability of bacteria to recover from stress.

ABSTRACT The Multicenter AIDS Cohort Study (MACS), an ongoing prospective study of the natural history of human immunodeficiency virus (HIV), has stored biologic specimens, including peripheral blood mononuclear cells (PBMC), from 5,622 participants for up to 12 years. The purpose of the present analysis was to evaluate the quality of the PBMC in the MACS repository in order to test the validity and feasibility of nested retrospective studies and to guide the planning of future repositories. PBMC were collected from MACS participants at four centers at 6-month intervals from 1984 to 1995, cryopreserved, and transported to a central repository for storage. A total of 596 of these specimens were subsequently tested for viability and used to evaluate cell function, to conduct immunophenotype analysis, or to isolate HIV. Simple linear regression models were applied to evaluate trends in recovery and viability over time and by center. Results indicated that from a nominal 107 cells cryopreserved per vial at all four centers, the median number of viable cells recovered was at least 5 × 106 (50% of the number stored) and the median viability was at least 90%. Results suggested that cryopreserved cells can be stored for at least 12 years with no general tendency toward cell loss over time. Furthermore, there were no statistically significant changes in the percent cell viability according to the length of time frozen, regardless of HIV serostatus or the level of CD4+ lymphocytes. Storing 107 PBMC per vial yields sufficient viable cells for phenotypic and/or functional analysis. Results from the MACS provide the basis for the planning of future repositories for use by investigators with similar research goals.

Résumé :
Abstract Hematopoietic progenitor cell (HPC) transplant product quality indicators after prolonged cryopreservation are variable. A recent study in Europe demonstrated wide variability of cryopreservation times, from 1 to >20 years, with most centers storing products for 5 or 10 years. Thawed HPC viability using various ex vivo means and in vivo by studying neutrophil and platelet engraftment has generally supported preservation of CD34+ viability with prolonged storage. However, sample size, methods, and timepoints varied, and clear guidelines currently do not exist regarding how long HPCs can be stored. This study’s objectives include an evaluation of HPC quality indicators over time and if HPC product age and/or viability correlates with HPC engraftment. Retrospective record review was conducted on 837 HPC products thawed between January 1, 2011, and December 31, 2018. HPC products were sampled and characterized at thawing by flow cytometry for total nucleated cell (TNC) recovery, CD34+ recovery, and CD34+ viability (by 7-AAD). Preestablished acceptability criteria for TNC recovery, CD34+ recovery, and CD34+ viability were >80%, >40%, and >60%, respectively. If available, products with ANC engraftment data were further analyzed to correlate data with time to HPC engraftment. HPC products were stored between 3 and 5,943 days; 816 products were infused, ranging from 3 to 5,273 days in storage. Most (81%) HPC transplants occurred within the first year, 47 infusions occurred after storage >5 years, and 4 infusions occurred after storage >10 years. CD34 viability and percent recovery of TNC and CD34 were not significantly different as time in storage increased. Given sample paucity, 19 samples stored for >10 years were analyzed in individual years and grouped. In total, 96.5% samples stored for <1 year and 68% in samples stored >10 years met acceptability criteria for >60% CD34+ recovery; 98.9% samples stored <1 year met the >60% CD34+ viability acceptability threshold versus 90% of samples stored for >10 years. The oldest sample infused after storage of 5,273 days (14.4 years) showed 42% TNC recovery, 19% CD34+ recovery, and 84% CD34 viability; ANC engraftment data were not available. Sixty-five samples containing ANC engraftment data showed that time to ANC engraftment ranged from 9 to 15 days. No clear trend was present for viability or CD34 recovery when compared with days to engraftment based on limited data. Although rare, infusion of HPC products of extended duration occurs. This study suggests that HPC product quality may decrease after extended cryopreservation based on preestablished acceptability criteria. The oldest sample infused had preserved CD34+ viability despite significantly less TNC and CD34+ recovery, suggesting thawing might select for more robust HPCs. More studies with older samples are needed to determine the clinical impact of cryopreservation and if modification of current quality indicators could be useful clinically.

Résumé :
The purpose of this descriptive study was to co-create oral histories of Midwestern farm widows. Rural widows constitute a vulnerable population due to issues of bereavement and depression compounded by emotional and geographical isolation. A farm widow is often forced to maintain viability of the farm for the family’s livelihood. Oral history interviews with nine Midwest farm widows were conducted and analyzed. Three overarching themes emerged: competence, industriousness, and inner strength. Women shared stories of overcoming insurmountable obstacles. This study contributes to the literature on grief and expanding inner strength among rural widows. Further research could inform theory related to inner strength following a challenging life event.

Résumé :
This piece offers an autoethnographic examination of the unraveling of epistemic certainty. The reader is offered three stories of falling, which explain the author's loss of knowing as a result of education, trauma, or apostacy. The aporias and assumptions in each story are teased out as we explore what they profess, where they fall silent, and how they understand uncertainty. Rather than determining which story is true, the text asks ethical and practical questions about where each story takes us. The irreconcilability of these three narratives raises questions about the limits of pluralism, how we respond to ideological difference, and the viability of our need to know.

Résumé :
<em>During the storage period, cocoa seeds undergo various processes that will affect the content of their food reserves and will have an impact on germination capacity and seeds vigor. The objective of this study is to investigate the changes of biochemical and vigor of cocoa seeds after storage for a maximum of 4 weeks with an interval of 1 week. The experiment was conducted at the Integrated Laboratory and Greenhouse of Balittri, Sukabumi, West Java, from October to December 2019. The completely randomized design with five treatments of seeds storage duration: (a) seeds unstored, (b) seeds stored for 1 week, (c) seeds stored for 2 weeks, (d) seeds stored for 3 weeks, and (e) seeds stored for 4 weeks, and five replications were used in this study. Parameters observed during seed storage were the biochemistry change of seeds and the percentage of germinated seeds, while during seed germination is an indicator of seed vigor. The results showed that storing cocoa seeds for 1-4 weeks decreased the fat content of the seeds, while the free fatty acids, protein, and percentage of germinated seeds increased. Until the 21st day after sowing, seed storage increases electrical conductivity but decreases growth rate, maximum growth potential, viability, and hypocotyl length. Seeds stored for one week still good in their vigor (viability of 80%), while seeds stored for 2-4 weeks have decreased their vigor (maximum viability of 54.4%).</em>

Résumé :
This article reviews the germinability and viability of seeds of threatened species collections in the New South Wales (NSW) Seedbank with the manifold aims of: ensuring that existing storage treatments and conditions provide effective ex situ storage of threatened species seed; providing baseline viability and seed storage life data on threatened species; and, identifying research gaps in seed germination and storage protocols for threatened species and communities. The germinability and viability of a range of seed accessions, of various ages and stored under different (although mainly identifiable) conditions in the NSW Seedbank, was determined through germination and cut-tests. The results indicated that many of the Fabaceae, Myrtaceae and Proteaceae species tested are orthodox and can be stored at 5 to 10% moisture content at 5C for up to ten years without significant loss of viability (short- to medium-term storage). The best results were obtained in the lower seed moisture content range (2 to 9%), which appeared to be especially critical for long-term storage of many Proteaceae accessions, reinforcing the need to attain the correct seed moisture content for long-term storage. Around 10% of accessions exhibited some degree of dormancy even after long storage periods. Storage of the widest range of species, for periods greater than ten years for long-term conservation purposes, is generally best conducted by storing at sub-zero temperatures. Freezing at -18C had little effect on the germinability of a range of seeds tested and is recommended over storage at 5C. Collection and seed banking procedures for the NSW Seedbank will be regularly reviewed and procedures modified in order to identify the best long-term storage conditions for species within this and other seedbanks. Seed collection strategies to maximize diversity and uses of seedbanks in conservation are discussed.

Résumé :
ABSTRACT Objective: Green tea extract (GTE) was shown to be effective in preserving periodontal ligament fibroblasts (PDLFs) of avulsed teeth. This study aimed at determining the potential of GTE in preserving the viability of PDLFs comparing with different storage media. Materials and Methods: Periodontal ligament cells were obtained from freshly extracted healthy impacted third molars and cultured in Dulbecco's Modified Eagle Medium (DMEM). Cell viability was determined by storing the cells in seven media; DMEM, tap water, Hank's balanced salt solution (HBSS), whole milk, hypotonic sucrose solution, GTE, and GTE + sucrose for 1, 2, 4, and 24 h at 37°C using tetrazolium salt-based colorimetric (3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyl tetrazolium bromide) assay. Statistical analysis was performed by one-way analysis of variance and post hoc tests. Results: GTE showed significantly higher protective effect than HBSS at 2, 4, and 24 h (P = 0.009, P = 0.02, P = 0.016), DMED at 2 h (P = 0.003), and milk at 4 h (P = 0.039). Conclusion: Although with undesirable osmolality and pH, GTE had a good ability in preserving the PDLFs comparing with other studied media.

Résumé :
Storage Potentials and Tolerance of High Protein Maize (HPM) and Quality Protein Maize (QPM) to Seed Storage Pests in Controlled EnvironmentStudies on seed storage were conducted at seed processing and storage section of the Institute of Agricultural Research and Training, Obafemi Awolowo University, Moor Plantation, Ibadan Nigeria in 2007 and 2008, to evaluate High protein maize (HPM) seed for resistance/tolerance to storage insect pest, and assess the resultant effect of seed treatment chemicals on its germination potential. Seeds of HPM variety (ART-98-SW1) and Quality protein maize (QPM) variety (ILE-1-OB) and two varieties of field corn (SUWAN-1-SR and TZPB-SRW) were treated with chemicals, namely Fitscophos™, Actellic 25 EC™, Apron Star™ and combination of Actellic 25 EC and Fitscophos before storing them under controlled environment for six months. The results show that HPM, QPM and field corn require storage chemicals for effective storage, but both QPM and HPM varieties were significantly damaged by storage insect pests compared to field corn varieties regardless of the chemical used. Apron Star effectively reduced infestation by insect pests, but it may bring about significant reduction in seed viability if used for long term storage. Combination of Actellic 25 EC and Fitscophos successfully reduced infestation by insect pests without significant injurious effect on seed viability. Breeding programmes for resistance to storage pests is recommended while long term effects of Apron Star on seed viability needs to be investigated before recommending it for use in HPM/QPM maize varieties.

Résumé :
Trauma causes spinal cord injury, and the devastating consequences of the injury are due to the failure of the damaged central nervous system (CNS) axons to regenerate. Previous studies have shown that olfactory ensheathing cells (OECs) are a unique type of glial cell and they can promote regeneration of CNS axons to aid recovery after spinal cord injury. Transplantation of OECs, in particular from the olfactory bulb (OB), is considered one of the most promising therapeutic strategies for the repair of CNS injuries, including spinal cord injury. Transplantation of OECs can be autologous or allogenic. Here we focused on the less invasive and more error-proof allograft approach which needs a collection of donor OB tissue for OEC production. In this study, we investigated the effects on the yield and proportions of OECs and olfactory nerve fibroblasts (ONFs) from storing OB tissue in various media for periods of 24 and 48 hours. The OEC yield contributes to the viability of a successful cell transplant. We concluded that storing OB tissue for a period longer than 24 hours negatively impacted the total cell number and subsequently the OEC population. This study provides useful information for future clinical applications.

Résumé :
The continuity of Chekov’s text adheres to the existence of his protagonists everywhere and then. The presence of Chekhov's heroes in every place and time is the key to the text's long-term viability. He described various characters, upon whom feelings were shown in different situations, specifically feeling scared among his protagonists. The character of (Belekov) in “The introversive man” who fears everything is different from the protagonist in “Khemech” by the contemporary writer Yuri Buyda. The protagonist’s wife made an unusual reaction against Chekov after her husband's death. She screamed: “I hate your writer Chekov! I hate him! I hate him!”. This reaction was a protest and a denial of the cover idea which had become a literary mode in literary works and daily life as was presented by Chekov in “The Introversive man”. The modern writer V. Bitsokh was able in “The cabinet” to show fear in people’s behavior in the Soviet Union, and so he did show how permanent fear of the protagonists’ environment was a common factor with Chekov’s. As for the female writer G. Shcherbakova in her “The introversive man”, she presented a character of a school headmistress named (Vania) who was dedicated and ambitious. However, she neglected an important aspect of her life, making a family. This was a struggle for her cover in Chekov. Shcherbakova named her story after Chekov’s “The introversive man”, but her protagonist differed from Chekov’s (Varinka) who did not recognize others’ feelings. Here, Chekov’s text gained its controversy and dialogue in a contemporary and modernist way.

Résumé :
The relevance of the study to establish methods of storing corn grain is related to the problem that exists in the technology of storage of crops of different crops. Methods and material and technical base with preservation do not fully take into account the characteristics of individual crops as objects of storage, which leads to loss of products and deterioration of its quality. There is also no technical and technological assessment of new methods of storage and granaries, including metal type. The article presents the results of research, which revealed the peculiarities of storage of corn grain in metal granaries. The temperature regime, which is formed in different places of the embankment and grain quality indicators - humidity, purity, acidity, viability (germination) depending on the thermal protection of the storage are established. Layers of grain embankment were found in the warehouse, where unfavorable conditions for preservation and stable condition of products are created. The technical and technological assessment was carried out and the efficiency of different granaries was determined depending on the storage and the direction of its use. As a result of the conducted researches the technical and technologicalcharacteristic of various ways applied to storage of grain is made, their advantages and lacks are established. For the purpose of safe storage in metal granaries it is recommended to carry out their thermal protection by drawing heat-insulating material on external walls of storage. For long-term storage of corn grain stocks, its humidity should not exceed 12% with the removal of the total mass of the unstable fraction. In the long run, it is necessary to reduce the storage of grain in a controlled gaseous environment, as one that increases a high degree of preservation of products and their quality.
