Acceptor splice site recognition (3′ splice site: 3′ss) is a fundamental step in precursor messenger RNA (pre-mRNA) splicing. Generally, the U2 small nuclear ribonucleoprotein (snRNP) auxiliary factor (U2AF) heterodimer recognizes the 3′ss, of which U2AF35 has a dual function: (i) It binds to the intron–exon border of some 3′ss and (ii) mediates enhancer-binding splicing activators’ interactions with the spliceosome. Alternative mechanisms for 3′ss recognition have been suggested, yet they are still not thoroughly understood. Here, we analyzed 3′ss recognition where the intron–exon border is bound by a ubiquitous splicing regulator SRSF1. Using the minigene analysis of two model exons and their mutants, BRCA2 exon 12 and VARS2 exon 17, we showed that the exon inclusion correlated much better with the predicted SRSF1 affinity than 3′ss quality, which were assessed using the Catalog of Inferred Sequence Binding Preferences of RNA binding proteins (CISBP-RNA) database and maximum entropy algorithm (MaxEnt) predictor and the U2AF35 consensus matrix, respectively. RNA affinity purification proved SRSF1 binding to the model 3′ss. On the other hand, knockdown experiments revealed that U2AF35 also plays a role in these exons’ inclusion. Most probably, both factors stochastically bind the 3′ss, supporting exon recognition, more apparently in VARS2 exon 17. Identifying splicing activators as 3′ss recognition factors is crucial for both a basic understanding of splicing regulation and human genetic diagnostics when assessing variants’ effects on splicing.

My project has been focused on the identification and validation of new mitochondrial gene variants and new mitochondria-related genes. The first report is about a woman presenting with a stroke-like episode and an history of severe hearing loss, frequent migraines, exercise intolerance, myalgias and limb-girdle muscle weakness indicating a slowly progressive myopathy and secondary amenorrhea with low gonadotropin levels. A muscle biopsy showed ragged-red, cytochrome c oxidase-negative fibers, and an isolated defect of cytochrome c oxidase activity in muscle mitochondria and sequence analysis of muscle mtDNA revealed a new heteroplasmic m.6597C>A transversion in the MTCOI gene, encoding subunit I of cytochrome c oxidase. Analysis on transmitochondrial cybrids demonstrated that the mutation is indeed associated with COX deficiency, i.e. pathogenic. The second report is about a new phenotype associated to mutations in the AARS2 gene encoding for the mitochondrial aminoacyl tRNA synthetase, identified in six patients presenting with primary ovarian failure, cerebellar and pyramidal signs and cognitive or behavioural disturbances. Two patients underlined a muscle biopsy which showed a severe complex IV defect at histochemical and biochemical analyses. The third report is about the clinical and biochemical phenotypes associated with mutations in two new mitochondrial aminoacyl tRNA synthetases (ARSs2) genes. In the first patient, an 8 years old child presenting with psychomotor delay, seizures, facial dysmorphisms and hyperlactacidemia and a brain MRI showing hyperintense lesions in the insula and fronto-temporal right cortex, whole exome sequencing (WES) identified a homozygous missense mutation in VARS2, encoding the mitochondrial valyl tRNA-synthetase. In two siblings presenting with a phenotype characterized by hypotonia and psychomotor retardation, high plasma and liquor lactate, both died at few months of age WES revealed two variants in TARS2, encoding the mitochondrial threonyl tRNA-synthetase: a missense and a splice site mutation. VARS2 and TARS2 mutations segregate within patients families. Patients’ clinical- biochemical phenotype and in silico and in vitro analyses of VARS2 and TARS2 mutations clearly indicate these genes as disease-causative. Expression of the corresponding wild-type enzymes led to recovery of the biochemical impairment of mitochondrial respiration in immortalized mutant fibroblasts; yeast modelling of the VARS2 mutation confirmed its pathogenic role.

Variability (the ability of a software system or software artifact to be adapted for use in a specific context) is reflected in and facilitated through the software architecture. The Second International Workshop on Variability in Software Architecture (VARSA) was held in conjunction with the Joint 10th Working IEEE/IFIP Conference on Software Architecture & 6th European Conference on Software Architecture 2012 in Helsinki, Finland. The workshop aimed at exploring current and emerging methods, languages, notations, technologies and tools to model, implement, and manage variability in the software architecture. It featured one industrial talk, five research paper presentations, and three working group discussions. Working groups discussed topics that emerged during the workshop. This report summarizes the themes of the workshop and presents the results of the working group discussions.

Los argumentos de la tesis son los modelos VAR estructurales y los modelos dinámicos de equilibrio general ambos aplicados a la macroeconomía.
El primer capítulo analiza, por medio de modelos VAR, los efectos de la políticas monetaria y fiscal sobre el producto interior bruto (PIB) y
el nivel de los precios en la economía norteamericana a partir de los años sesenta. Ambas políticas producen efectos pequeños. El capítulo demuestra que si en un modelo VAR para el análisis solo de la política monetaria se incluyen variables fiscales, sus efectos se educen de la mitad.
El segundo capítulo analiza los efectos de aumentos de los tipos de interés a corto plazo sobre los índices de bolsas en los países que forman el G-7 y en España. Los efectos, en general negativos y transitorios, son diferentes en termino de reducción de los índices entre los países analizados. Variaciones exógenas de los tipos de interés no parecen ser responsables de los principales movimientos en los índices de bolsa.
El tercer capítulo presenta un modelo de equilibrio económico general en el cual las familias consumidoras pueden invertir en acciones y en depósitos bancarios. El modelo, calibrado sobre los datos de la economía norteamericana es capaz de reproducir, desde un punto de vista cualitativo, los efectos de la política monetaria sobre el índice de la bolsa.
El ultimo capítulo confronta tres modelos de equilibrio económico general alternativos del ciclo económico. En el primero las fricciones financieras determinan endógenamente costes para variar el nivel de capital. En los otros dos estos costes son exógenos. Los modelos son estimados mediante el método de la máxima verosimilitud utilizando datos sobre la economía norteamericana de 1966 hasta el 2001. El resultado principal es que el primer modelo no parece explicar mejor que los modelos alternativos las dinámicas de las principales variables del modelo.
Chapter 1 investigates if and how the standard results of the VAR literature on the macroeconomic effects of monetary policy, which typically overlooks fiscal policy, are affected when monetary and fiscal policy are jointly considered. To this end, structural VAR models are set up using U.S. post-war data. It is found that the magnitude of the responses of output and price to a monetary policy shock are halved once fiscal policy is considered. Both monetary and fiscal policy shocks have small effects on output and the prices.
Chapter 2 evaluates the effects of monetary policy shocks on stock market indices in the G-7 countries and Spain using structural VARs. A contractionary shock has a negative and transitory effect on stock market indices.
In Chapter 3 a limited participation model with households trading in stocks is set up and validated by means of impulse responses using U.S. data.
The model is able to account for the empirical response of stock prices to monetary policy shocks.
Chapter 4 compares three alternative models of the business cycle that rely on sticky prices and real rigidities in the form of adjustment costs for investment. In the first model these costs arise endogenously as the result of asymmetric information and agency costs. In the second model the costs for adjusting the level of investment are exogenously imposed while in the last model these costs are imposed on the changes in investment. The models are estimated with maximum likelihood using U.S. post-war data. The model with exogenous adjustment costs on the level of investment seems to provide the best representation of the U.S. business cycle and the responses to technology and monetary policy shocks.

This paper proposes a large Bayesian Vector Autoregressive (BVAR) model with common stochastic volatility to forecast global equity indices. Using a dataset consisting of monthly data on global stock indices the BVAR model inherently incorporates co-movements in the stock markets. The time-varying specification of the covariance structure moreover accounts for sudden shifts in the level of volatility. In an out-of-sample forecasting application we show that the BVAR model with stochastic volatility significantly outperforms the random walk both in terms of root mean squared errors as well as Bayesian log predictive scores. The BVAR model without stochastic volatility, on the other hand, underperforms relative to the random walk. In a portfolio allocation exercise we moreover show that it is possible to use the forecasts obtained from our BVAR model with common stochastic volatility to set up simple investment strategies. Our results indicate that these simple investment schemes outperform a naive buy-and-hold strategy. (authors' abstract)

Buried esker systems are a major source of potable water in Eastern Ontario. Ever since the Clean Water Act was passed in 2006, efforts have been placed on understanding aquifers and characterizing watersheds in order to protect source water. This thesis focuses on one esker, extending from Vars to Winchester in the South Nation River watershed, which provides municipal water for seven villages -- an estimated population of 17,000. The esker complex is conceptualized as a 50-km long deposit of glaciofluvial outwash high-permeability material that is generally surrounded by fine, low permeability materials, except in a number of locations where the esker outcrops at ground surface. The complex recharges at its northern- and southern-most areas, which correspond to topographic highs; it discharges in a topographically low area at its center near the village of Embrun. Regional stratigraphy was simplified for modeling purposes and brought down to six hydrostratigraphic units, two of which represent the esker: a coarser core, composed of a mixture of coarse sand and gravel, and sandy fan deposits. A three-dimensional hydrogeological model was developed using a finite element groundwater flow system, FEFLOW. Recharge and hydraulic conductivities were adjusted during calibration. Calibration was assessed using the mean error of residuals, standard deviation of residuals and normalized Root Mean Square calculations; correlation was estimated using the determination coefficient and Pearson's coefficient. Results favour the conceptualization of the esker as a continuous, heterogeneous deposit comprising a gravelly core and sandy fans (ME=-4.33m, RMS=5.67m, n=18 wells in close proximity of esker); however, mean residuals are not significantly different when testing the discontinuous and homogeneous esker hypotheses. Sensitivity analyses testing the model's response to recharge and pumping showed that the system is relatively resilient; however, our better judgement tells us that this is an artefact due to boundary conditions and not an adequate prediction of what may happen. Nevertheless, we hope this model will be a facilitating tool for understanding, managing and protecting the aquifer's resource.

Målet är att religionslärare ska kunna tillägna sig några av dessa förslag, för att ge deras elever en mer givande och inspirerande undervisning.   Religionsundervisningen ska ses som en kunskapsresa, där det huvudsakliga , målet är att eleverna ska lära för livet. Att skapa en mer levande undervisning som stimulerar elevernas lärande, kräver att läraren utgår ifrån elevernas referensramar liksom möter dem utifrån deras egna villkor.

Syftet med denna litteraturstudie var att beskriva vilka copingstrategier föräldrar använder sig av då deras barn har cancer. Data samlades in via databaserna MedLine (PubMed) och CINAHL. Femton artiklar valdes ut och de valda artiklarna kvalitetsgranskades och analyserades. Utifrån valda artiklar framkom sju kategorier. I studiens resultat framkom det att föräldrar till barn med cancer använde sig av olika copingstrategier. Copingstrategierna var socialt stöd, andligt stöd, att få visa känslor som förälder, jämförande tillvägagångssätt, positivt tänkande, undvikande coping och att söka information. Författarnas slutsats av denna litteraturstudie är att det finns många olika copingstrategier som föräldrar kan använda sig av och att anhöriga, i det här fallet föräldrar, måste få en större plats inom sjukvården. Sjuksköterskor behöver också ha kunskap om copingstrategier hos föräldrar till barn med cancer för att i sin tur kunna undervisa föräldrarna i det, så att de skall kunna handskas med situationen.

Abstract Muscle disorders and induced muscle atrophy impose critical risks to the well-being of an individual, limiting normal activities of daily living. Several resistance training methods exist that have effectively reversed the progression of muscle atrophy. Weightlifting and hydrotherapy are the two widely practiced schemes for resistance training; however, there is the potential risk of excessive loads exerted on the muscles during weightlifting, and limited accessibility and cost are barriers to hydrotherapy. An alternative is using a resistance band. Some limitations include engaging multiple muscles/joints while only unidirectional resistance is feasible. To address these limitations, a VAriable Resistance Suit (VARS) is designed to provide speed-dependent, bi-directional, and variable resistance at a single joint. As a proof of concept, an elbow module of VARS is developed and validated experimentally. A pilot study shows the changes in flexor and extensor muscle activations in response to eight different levels of resistance modulated by VARS. The clinical implications of VARS remain as future work, but the functionalities implemented in the presented VARS prototype and preliminary results suggest that VARS could be a viable solution and complementary to existing tools and techniques utilized in resistance training.

The article presents the tests results of narrow-leafed lupin for some morphological and biological characters: height and initial growth of plants, their productivity (seeds amount and weight), seed amount per a pod, microdistribution coefficient etc. Vars. Beloro-zovy 114, BL 78-07 and Hybrid 1215 stood out in plant height (68.6–81.3 sm); Uzkolistny 53-02, Belozerny 110 and Smena — in seeds amount per a pod (4.44–4.08 seeds). Low alkaloid content (0.038%) had the vars. Belorozovy 144 and Smena.

The preliminary findings of an exploratory study conducted on a novel Oxyfuel Combustion Turbine Cycle (OCTC) using ASPEN PLUS for a range of 40% to 90% CO2 capture are presented. Starting from a GE Energy IGCC, the OCTC retains the Gasifier with a Radiant Cooler-only section and the Warm-Gas-Clean Up (WGCU) section with the desulfurization process eliminated for a combined carbon and sulfur co-sequestration approach. The conventional gas turbine combined cycle is also removed. With no integration between the Air Separation Unit (ASU) and the modified oxyfuel combustion turbine, the High Pressure (HP) ASU is replaced by a Low pressure (LP) ASU. The added attributes of this novel coal-based power system configuration are (1) the Vapor Absorption Refrigeration System (VARS), (2) the associated water production in the VARS evaporator, (3) the recirculation of combustor flue gas for lower NOx emissions and additional power production, as well as (4) an original concept of carbon dioxide compression as proposed by SouthWest Research Institute (SwRI) and Dresser Rand (D-R). Assuming a reasonable Coefficient of Performance (COP) for the VARS, the overall process efficiency results (about 35% HHV) were equivalent or even better when compared with studies of current simulated IGCC systems with CO2 capture. Once an optimum scheme has been finalized in future activities, an economic analysis would be conducted. The combined performance and economic results could then be compared with alternate advanced coal based power systems.

A novel cooling and power cycle has been proposed that combines a semi-closed cycle gas turbine called the High Pressure Regenerative Turbine Engine (HPRTE) with a vapor absorption refrigeration system (VARS). The refrigeration cycle (VARS) interacts with the power cycle (HPRTE) solely through heat transfer in the generator and the evaporator. Waste heat from the recirculated combustion gas of the HPRTE is used to power the absorption refrigeration unit, which cools the high-pressure compressor inlet of the HPRTE to below ambient conditions and also produces excess refrigeration, in an amount which depends on ambient conditions. Water produced as a product of combustion is intentionally condensed in the evaporator of the VARS, which is designed to provide sufficient cooling for three purposes: chilling the inlet air to the high pressure compressor, water extraction, and for an external cooling load. In a previous study, the combined cycle was modeled using zero-dimensional steady-state thermodynamics, with the specified values of efficiencies and pressure drops for the turbo-machinery and heat exchangers. The model predicts that the combined cycle with steam blade cooling for a medium-sized engine will have a thermal efficiency of 49%, in addition to the external refrigeration load generated in the cycle which is 13% of the net work output. It also produces about 1.4 kg of water for each kg of fuel (propane) consumed. A small experimental unit demonstrating the HPRTE/VARS combined cycle has been constructed and is currently being tested in the Energy & Gas-dynamic Systems Laboratory at the University of Florida. A 45 HP Rover 1S-60 engine is integrated with a NH3/H2O vapor absorption refrigeration unit having a capacity of 19 Ton Refrigeration. The engine flow-path has been significantly modified to include partial recirculation of exhaust products, turbocharging, and recuperation, thus implementing the HPRTE concept. In addition, a significant modeling effort has been undertaken to simulate the combined cycle operation under design and off-design conditions. Initial experimental results show good agreement with the model predictions, including overall efficiency and water extraction rates.

We propose a method to adjust for data outliers in Bayesian Vector Autoregressions (BVARs), which allows for different outlier magnitudes across variables and rescales the reduced form error terms. We use the method to document several facts about the effect of outliers on estimation and out-of-sample forecasting results using euro area macroeconomic data. First, the COVID-19 pandemic led to large swings in macroeconomic data that distort the BVAR estimation results. Second, these swings can be addressed by rescaling the shocks’ variance. Third, taking into account outliers before 2020 leads to mild improvements in the point forecasts of BVARs for some variables and horizons. However, the density forecast performance considerably deteriorates. Therefore, we recommend taking into account outliers only on pre-specified dates around the onset of the COVID-19 pandemic.
