We previously confirmed that transducer of ERBB2, 1 (TOB1) gene, can induce autophagy in gastric cancer cells. Studies have shown the biogenesis of exosomes overlaps with different autophagy processes, which helps to maintain the self-renewal and homeostasis of body cells. This study is aimed at verifying whether overexpressing TOB1 induces autophagy by secreting exosomes in gastric cancer cells and its underlying mechanisms. Differential ultracentrifugation was used to extracted the exosomes from the culture medium of gastric cancer cell line AGS-TOB1 ectopically overexpressing TOB1 (exo-AGS-TOB1, experimental group) and AGS-empty-vector cell line with low expression of endogenous TOB1 (exo-AGS-Vector, control group). Exosomal markers CD9 and TSG101 were determined in both the cell supernatants of exo-AGS-TOB1 and exo-AGS-Vector by Western blot. Under the transmission electron microscope (TEM), the exosomes were round and saucer-like vesicles with double-layer membrane structure, and the vesicles showed different translucency due to different contents. The peak size of exosomes detected by nanoparticle tracking analysis (NTA) was about 100 nm. When the exosomes of exo-AGS-TOB1 and exo-AGS-Vector were cocultured with TOB1 knockdown gastric cancer cell line HGC-27-TOB1-6E12 for 48 hours, the conversion of autophagy-related protein LC3-I to LC3-II in HGC-27-TOB1-6E12 gastric cancer cells cocultured with exo-AGS-TOB1 was significantly higher than that in the control group, and the ratio of LC3-II/LC3-I was statistically different ( P < 0.05 ). More autophagosomes in HGC-27-TOB1-6E12 cells cocultured with exo-AGS-TOB1 for 48 hours were observed under TEM, while fewer autophagosomes were found in the control group. Lastly, miRNAs were differentially expressed by cell supernatant-exosomal whole transcriptome sequencing. Thus, our results provide new insights into TOB1-induced autophagy in gastric cancer.

Background/Aims: This study investigated the effect of silencing TOB1 (Transducer of ERBB2, 1) expression in bone marrow-derived mesenchymal stem cells (MSCs) on MSC-facilitated tendon-bone healing in a rat supraspinatus repair model. Methods: Rat MSCs were transduced with a recombinant lentivirus encoding short hairpin RNA (shRNA) against TOB1. MSC cell proliferation was analyzed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays. The effect of MSCs with TOB1 deficiency on tendon-bone healing in a rat rotator cuff repair model was evaluated by biomechanical testing, histological analysis and collagen type I and II gene expression. An upstream regulator (miR-218) of TOB1 was determined in MSCs. Results: We found that knockdown of TOB1 significantly increased the proliferative activity of rat MSCs in vitro. When MSCs with TOB1 deficiency were injected into injured rat supraspinatus tendon-bone junctions, the effect on tendon-bone healing was enhanced compared to treatment with control MSCs with normal TOB1 expression, as evidenced by elevated levels of ultimate load to failure and stiffness, increased amount of fibrocartilage and augmented expression of collagen type I and type II genes. In addition, we found that the TOB1 3′ untranslated region is a direct target of miR-218. Similar to the effect of TOB1 deficiency, overexpression of miR-218 effectively promoted tendon-bone healing in rat. Conclusion: These results suggest that TOB1 may play a negative role in the effect of MSCs on tendon-bone healing, and imply that expression of TOB1 may be regulated by miR-218.

How proliferative and inhibitory signals integrate to control liver regeneration remains poorly understood. A screen for antiproliferative factors repressed after liver injury identified transducer of ErbB2.1 (Tob1), a member of the PC3/BTG1 family of mito-inhibitory molecules as a target for further evaluation. Tob1 protein decreases after 2/3 hepatectomy in mice secondary to posttranscriptional mechanisms. Deletion of Tob1 increases hepatocyte proliferation and accelerates restoration of liver mass after hepatectomy. Down-regulation of Tob1 is required for normal liver regeneration, and Tob1 controls hepatocyte proliferation in a dose-dependent fashion. Tob1 associates directly with both Caf1 and cyclin-dependent kinase (Cdk) 1 and modulates Cdk1 kinase activity. In addition, Tob1 has significant effects on the transcription of critical cell cycle components, including E2F target genes and genes involved in p53 signaling. We provide direct evidence that levels of an inhibitory factor control the rate of liver regeneration, and we identify Tob1 as a crucial check point molecule that modulates the expression and activity of cell cycle proteins.

Reliable biomarkers corresponding to disease progression or therapeutic responsiveness in multiple sclerosis (MS) have not been yet identified. We previously reported that low expression of the antiproliferative gene TOB1 in CD4+ T cells of individuals presenting with an initial central nervous system (CNS) demyelinating event (a clinically isolated syndrome), correlated with high risk for progression to MS. We report that experimental autoimmune encephalomyelitis (EAE) in Tob1−/− mice was associated with augmented CNS inflammation, increased infiltrating CD4+ and CD8+ T cell counts, and increased myelin-reactive Th1 and Th17 cells, with reduced numbers of regulatory T cells. Reconstitution of Rag1−/− mice with Tob1−/− CD4+ T cells recapitulated the aggressive EAE phenotype observed in Tob1−/− mice. Furthermore, severe spontaneous EAE was observed when Tob1−/− mice were crossed to myelin oligodendrocyte glycoprotein–specific T cell receptor transgenic (2D2) mice. Collectively, our results reveal a critical role for Tob1 in adaptive T cell immune responses that drive development of EAE, thus providing support for the development of Tob1 as a biomarker for demyelinating disease activity.

Background We previously identified the tumor suppressor gene TOB1 as related to gastric cancer. The purpose of this study was to explore whether TOB1 induces autophagy through the AKT/mTOR signaling pathway in gastric cancer. Methods Western blotting was used to detect the protein levels of TOB1, LC3, AKT, mTOR, phosphorylated (p) AKT, and p-mTOR. A double fluorescent GFP-RFP-LC3 fusion protein was used to trace autophagy by laser confocal microscopy. Autophagosomes were observed by transmission electron microscopy. Results The conversion of LC3-I to LC3-II and the LC3-II/LC3-I ratio were significantly increased in AGS cells overexpressing TOB1 compared with control cells. Fluorescence imaging showed LC3 puncta at 48 h, and these puncta increased significantly at 72 h after TOB1 transfection compared with control tumor cells. The presence of autophagosomes in AGS cells was observed at 72 h after TOB1 transfection by transmission electron microscopy, and no autophagosomes were found in the control cells. Moreover, the levels of p-AKT and p -mTOR were lower in AGS cells than in control cancer cells. Conclusion Our results provide novel insight that TOB1 might suppress gastric cancer by inducing autophagy, possibly through decreasing phosphorylation and the subsequent activation of the AKT/mTOR signaling pathway.

Abstract Abstract 5125 The TOB1 gene is a transcription factor responsible for the transduction of the gene ERBB2. It is a member of a family of cell suppressor proliferation proteins called TOB/BTG1 family; also, this gene operates on the inhibition of neoplastic transformation. The TOB1 gene presents a decreased expression in several types of cancer such as lung, breast, thyroid and stomach cancer. However, the function of this gene in chronic myeloid leukemia (CML) remains unknown. Aiming to evaluate the inhibition of gene TOB1 into BCR-ABL positive cells and trying to elucidate the molecular mechanisms associated with the inhibition of this gene in the CML we proceed to a more detailed study of this gene. The inhibition of this gene in K562 cells was performed using specific lentivirus. The effect of silencing TOB1 in the proliferation of K562 cells was assessed by the MTT assay after 48 hours of culture; in shTOB1 the proliferation was increased in comparison with shControl cells. To evaluate the synergistic effect between the inhibition of kinase tyrosine activity of BCR-ABL and the inhibition of TOB1 we performed a treatment with different concentrations of imatinib (0. 1, 0. 5 and 1μM), but we observed the decrease in cell proliferation of shTOB1 cells to similar levels of shControl cells only at the 1μM concentration. Therefore, the TOB1 silencing increased the proliferation of K562 cells without an additional effect of a treatment with Imatinib. To analyze the clonogenicity, we performed a formation of colonies assay, in methylcellulose, to determine whether silencing TOB1 could cause a change in the clonal growth of positive BCR-ABL cells. There was no significant change in the number of colonies that grew in cell culture shTOB1 compared to shControl cells. These results suggest that silencing TOB1 in K562 cells may not change the clonogenicity. In the assessment of cell cycle, the flow cytometry analysis revealed a significant accumulation of K562 cells in S phase, with consequent reduction of cells in the G2 phase of the cell cycle in cells shTOB1 compared to cells shControl. The TOB1 gene silencing in K562 cells kept the cells in the S phase and prevented the entry of cells in the G2 phase showing that the inhibition of gene TOB1 induced an increase in proliferation of K562 BCR-ABL cells. The level of apoptosis was assessed by flow cytometry after labeling the cells with anexin-V/PI. The Imatinib treatment presented dose-response in the induction of apoptosis as expected. However, a cumulative effect with TOB1 silencing was not observed. Furthermore, the apoptosis was also assessed by assays of caspases 3, 8 and 9, which showed an increase of the caspase activity of shControl cells in relation of the shTOB1 cells, showing that inhibition of this gene also changes the level of apoptosis. These results corroborate the literature data that report the relationship of this tumour suppressor gene in signalling pathways related to angiogenesis, carcinogenesis, apoptosis and metastasis. When we relate the results obtained with the LMC, we can consider the possibility of TOB1 regulation changes be related to modification of important signalling pathways such as AKT, PI3K, STAT3 and STAT5, among others. Furthermore, the inhibition of TOB1 may be related with an increase on the number of BCR-ABL positive cells and subsequent disease progression. In conclusion, this study confirmed literature data showing that TOB1 gene works as a tumour suppressor protein in cells of many types of cancer. From this work we can infer that in CML the expression of this gene is transformed, resulting in changing of the capacity of induction of apoptosis, decrease tumour necrosis and increase cell proliferation. Abstract Gastric cancer (GC) is the most common tumors and the molecular mechanism underlying tumor metastasis is still largely unclear. Here, we show that miR-25 was overexpressed in plasma and primary tumor tissues of GC patients with TNM stage (III or IV) or lymph node metastasis. MiR-25 inhibition significantly decreased the metastasis, invasion and proliferation potential of GC cells in vitro, and reduced their capacity to develop distal pulmonary metastases and peritoneal dissemination in vivo. Furthermore, miR-25 repressed transducer of ERBB2, 1 (TOB1) expression by directly binding to the 3’-UTR, and the inverse correlation was observed between the expressions of miR-25 and TOB1 mRNA in primary GC tissues. Moreover, the loss of TOB1 increased the metastasis, invasion and proliferation of GC cells, and the restoration of TOB1 led to the metastasis, invasion and proliferation suppression. The receiver operating characteristics (ROC) analysis yielded an area under the curve (AUC) value of 0.7325 in distinguishing the GC patients with death from those with living. The analysis of optimal cutoff value revealed significant difference of patients’ survival between GC patients with high plasma concentrations of miR-25 (&gt; 0.2333 amol/μL) and those with low (&lt; 0.2333 amol/μL). Taken together, miR-25 promotes GC progress by directly down-regulating TOB1 expression, and the high concentrations of miR-25 in plasma of GC patients predict poor patient survival.

Adaptor proteins, like Tollip and Tom1, facilitate cellular cargo sorting through their ubiquitin-binding domains. Tollip and Tom1 bind to each other through their TBD and GAT domains, respectively, whereas Tollip interacts with phosphatidylinositol-3-phosphate (PtdIns(3)P)-containing endosomal membranes. Tom1 and Tollip interaction and association with endosomes is proposed to be involved in the lysosomal degradation of polyubiquitinated cargo. Through cellular, biochemical, and biophysical techniques, we have further characterized the association of Tom1 with Tollip. Mutations in the binding interface of the Tom1 GAT and Tollip TBD complex leads to a subcellular mis-localization of both proteins, indicating that Tom1 may serve to direct Tollip to specific cellular pathways. It was determined that Tom1 inhibits the binding of Tollip to PtdIns(3)P and inhibition was reversed when mutations in the binding interface of the Tom1 GAT and Tollip TBD were present. Furthermore, it was established that, upon the binding of Tollip TBD to Tom1 GAT, ubiquitin is inhibited from binding to Tom1 GAT. It was also demonstrated that Tom1 GAT, but not Tollip TBD, can weakly bind to PtdIns(3)P. Consequently, we propose that association of Tom1 may serve to direct Tollip for involvement in specific cell signaling pathways. Gaining insight into the function of Tom1 and Tollip may lead to their use as therapeutic targets for increasing the efficiency of cargo trafficking and also for patients recovering from various cardiac injuries.

The ~ 73 ka ‘super-eruption’ of the Toba caldera in Sumatra is the largest known eruption of the Quaternary. The products of this eruption, the Youngest Toba Tuff (YTT), have been implicated in global and regional climate deterioration with widespread ecological effects. In this thesis I study the YTT co-ignimbrite ashfall, in particular the mechanisms of transport, sedimentation and preservation of ash deposits. I use distal marine and terrestrial ash sediments: a) to estimate the volume of YTT ash fallout; b) to quantify variability in the geochemistry of YTT ash; c) to assess the reliability of YTT ash as a chronostratigraphic marker; and d) to determine local influences on the reworking of YTT ash deposits. Following the introductory chapters, I address topics a) and b) through detailed investigations of published physical and chemical evidence. Chapter three shows that particle size and sediment thickness do not decline exponentially with distance from the eruption vent, highlighting the limitations of current methods of volume estimation for co-ignimbrite super-eruptions. Chapter four analyses geochemical variation in 72 YTT samples, and reveals the signatures of magma chamber zonation and post-depositional alteration. I address topics c) and d) through fieldwork in six locations, and detailed analysis of ash samples from a wide variety of local depositional environments. Chapter five uses high-resolution stratigraphic analysis of the YTT layer in the Son Valley, India, to show that variable deposition and sediment reworking may compromise the reliability of the ash layer as an isochronous marker for interpreting archaeological sequences. Chapter six combines a new understanding of the mechanisms of reworking, using new data on microscopic characteristics of reworked ash at four sites in Malaysia to demonstrate the necessity of accounting for reworking in palaeoenvironmental reconstructions. I conclude that accurate analyses of distal ash deposits can reliably determine the chemical properties of the YTT eruption, and that a detailed understanding of deposition and reworking processes is essential to inference of the environmental impacts of super-eruptions.

Pacto poético alude, obviamente, al concepto de “pacto autobiográfico” acuñado por Philippe Lejeune. Internet, al medio por el cual se ha difundido esta experiencia poético-autobiográfica centrada en poemas y entrevistas concedidos por un tal Cristóbal “Tobi” Kanashiro. Y, por último, propiamente el caso de este sujeto o agente inventado, entre alumnos y profesor, en el marco de un curso denominado “Literatura”, para la Facultad de Arte de la PUC del Perú (semestre académico I, marzo-julio, 2010). El presente ensayo, más que ahondar o debatir los problemas teóricos inherentes a la autobiografía -al que este tipo de experiencia invita, sobre todo, en cuanto aquello de la identidad real del autor-, trata más bien de establecer o poner en paralelo los requisitos del pacto autobiográfico con -y esta es la hipótesis que intentaremos demostrar- las exigencias propias, asimismo, a una recepción productiva o eficaz en el campo de la poesía. Relación entre productor y lector, esta última, a la que vamos denominando “acto poético”. Productores de Kanashiro que dialécticamente fuimos, además, lectores de primera mano. El presente trabajo, por lo tanto, utilizará como fuente fundamental de análisis los testimonios de los alumnos involucrados en dicha experiencia.

Q'eqchi' is one of many Mayan languages spoken in Guatemala, C.A. This study provides the first Tone Break and Indices (ToBI) transcription system (Silverman et al., 1992) labeling of Q'eqchi' within the Autosegmental-Metrical (AM) model of intonation (Liberman, 1975; Pierrehumbert, 1980; Ladd, 1996). As an exploratory study into the basic intonation patterns of the language, observations were made on a variety of phenomenon relating to the intonational structure and contour pattern of the language. Three native male speakers of Q'eqchi' each provided 75 spoken sentences designed to best observe the basic patterns of intonation in the language. Each spoken utterance was analyzed through the labeling of pitch accents, phrase accents, and boundary tones in accordance with ToBI transcription guidelines (Beckman & Hirschberg, 1994; Beckman & Elam, 1997). The study reinforces previous observation on the stress pattern in the language, identifies the pitch accents and boundary tones which best describe the behavior of the intonational contour of the Q'eqchi' speakers, and proves the existence of prosodic phrases which dictate the intonational patterns of speech. In addition, the different patterns observed in declarative, imperative, and interrogative sentences are exemplified and discussed along with other phenomenon observed in the spoken data.

A TOBI (tangential on board injection) or preswirl system is a critical component of a high pressure turbine cooling delivery system. Its efficient performance and characterization are critical because the blade and disk life depend on the accuracy of delivering the required flow at the correct temperature and pressure. This paper presents a TOBI flow discharge coefficient validation process applied to a low radius radial configuration starting from a 1D network flow analysis to a 3D frozen rotor CFD analysis of the rotor cooling air delivery system. The analysis domain commences in the combustor plenum stationary reference frame, includes the TOBI, transitions to the rotating reference frame as the flow travels through the rotating cover plate orifice, continues up the turbine disk into the slot bottom blade feed cavity, and terminates in the turbine blade. The present effort includes matching a 1D network model with 3D CFD results using simultaneous goal-matching of pressure predictions throughout the circuit, defining test rig pressure measurements at critical “non-disturbing” locations for quanification of pressure ratio across the TOBI, and finally comparing the TOBI flow coefficient resulting from stationary cold flow tests with what was obtained from the 3D CFD results. Analysis of the results indicates that the discharge coefficient varies with pressure ratio and that the traditional method of using a constant discharge coefficient extracted from a cold flow test run under choked conditions leads to over-predicting turbine cooling flows. TOBI flow coefficient prediction for the present study compares well with the stationary data published by author researchers for the configuration under investigation, and the process described in this paper is general for any TOBI configuration.

This work summarizes the results of the CFD analyses to investigate the effect of the geometrical parameters for a typical coverplate-disk cavity and blade broach system also known as the blade cooling flow supply system. A turbofan high pressure turbine was used as the test vehicle for this investigation. The main objective was to explore potential improvements in engine SFC (aerodynamic performance) by reducing the parasitic work while minimizing the impact on the factors that affect the durability of the turbine blades; feed pressure, temperature, and mass flow. Various tangential on-board injection (TOBI) blade cooling flow supply systems were considered: i) Phase 1 compared the radial TOBI and axial TOBI, ii) Phase 2 compared coverplate-disk cavity shapes, and iii) Phase 3 compared blade broach shapes. The in-house CFD code NS3D was used for the analyses. Compared to the radial TOBI, the axial TOBI has a positive impact on the parasitic work (lower) and blade feed temperature (lower) while it has a negative impact on the blade feed pressure (lower). Further, the coverplate-disk cavity shapes investigated had no significant impact on the parasitic work, blade feed pressure, and blade feed temperature. The CFD solutions show that the major portion of the parasitic work is due to flow turning at the broach entrance. Finally, reducing the blade broach cross-section by sloping up the lower wall has no significant impact on the parasitic work and blade feed temperature but a negative impact on the blade feed pressure and mass flow. Modifying the broach pressure side wall shape is preferred among the blade broach geometries investigated. Future work to improve the CFD analysis consists of performing unsteady analyses to better capture the vortex flow in the blade broach, and including upstream stationary components with either iterative boundary condition modeling or an unsteady multi-stage approach.

Unfolding processes of timbre and memory in improvisational piano performance This exposition is an introduction to my research and practice as a pianist, in which I unfold processes of timbre and memory in improvised music from a performer’s perspective. Timbre is often understood as a purely sonic perceptual phenomenon. However, this is not in accordance with a site-specific improvisational practice with changing spatial circumstances impacting the listening experience, nor does it take into account the agency of the instrument and objects used or the performer’s movements and gestures. In my practice, I have found a concept as part of the creating process in improvised music which has compelling potential: Timbre orchestration. My research takes the many and complex aspects of a performance environment into account and offers an extended understanding of timbre, which embraces spatial, material and bodily aspects of sound in improvised music performance. The investigative projects described in this exposition offer a methodology to explore timbral improvisational processes integrated into my practice, which is further extended through collaborations with sound engineers, an instrument builder and a choreographer: -experiments in amplification and recording, resulting in Memory piece, a series of works for amplified piano and multichannel playback - Piano mapping, a performance approach, with a custom-built device for live spatialization as means to expand and deepen spatio-timbral relationships; - Accretion, a project with choreographer Toby Kassell for three grand pianos and a pianist, where gestural approaches are used to activate and compose timbre in space. Together, the projects explore memory as a structural, reflective and performative tool and the creation of performing and listening modes as integrated parts of timbre orchestration. Orchestration and choreography of timbre turn into an open and hybrid compositional approach, which can be applied to various contexts, engaging with dynamic relationships and re-configuring them.
