Filarioid nematodes (Nematoda: Filarioidea) parasitic in Australasian monotremes, marsupials and murid rodents are described from three genera, Breinlia (Breinlia), Pelecitus and Sprattia. New host and geographic records are presented for 24 previously described species from these genera, a second subgenus, Breinlia (Johnstonema) and an additional three genera, Cercopithifilaria, Monanema and Pelecitus. Nine species are described as new, B. (B.) beveridgei sp. nov. from the peritoneal cavity of Macropus parryi Bennett, B. (B.) bigenera sp. nov. from the peritoneal and pleural cavities of Aepyprymnus rufescens (Gray), Onychogalea fraenata (Gould), O. unguifera (Gould) and possibly Antechinus swainsonii (Waterhouse), B. (B.) dorcopsis sp. nov. from the peritoneal cavity of Dorcopsis hageni Heller and D. luctuosa (D’Albertis), B. (B.) melomyos sp. nov. from the peritoneal cavity of Melomys cf. burtoni, B. (B.) oweni sp. nov. from the peritoneal cavity of Dactylopsila trivirgata Gray, B. (B.) presidentei sp. nov. from the peritoneal cavity of Mesembriomys gouldii (Gray), M. macrurus (Peters) and the pleural and peritoneal cavities of Conilurus penicillatus (Gould), B. The female of B. (J.) woerlei from the pulmonary artery, right ventricle and lung cysts of Petrogale brachyotis (Gould) is described for the first time. Breinlia (J.) andersoni Spratt & Varughese, 1975 from the subcutaneous connective tissues of Macropus rufus (Desmarest), M. giganteus Shaw, M. robustus erubescens Sclater and Wallabia bicolor (Desmarest) is placed as a synonym of B. (J.) annulipapillata (Johnston & Mawson, 1938) from the subcutaneous connective tissues of M. dorsalis (Gray), O. fraenata and W. bicolor. In general, filarioids predominate in macropodoid hosts and in the north of Australia.

Abstract The plant fossil genera Umkomasia Thomas 1933 and Pteruchus Thomas 1933 emend. Townrow 1962 are known chiefly from the Middle and Upper Triassic of Gondwana. The structure of these fructifications has been conjectural, some being identified as pinnate, others as helically arranged. Specimens from the Ladinian-lower Norian of Queensland (northeastern Australia) show that the female and male fructifications - U. geminata (Shirley 1898) Rigby in Playford et al. 1982 emend. nov. and P. dubius Thomas 1933 emend. Townrow 1962, respectively - have a bipinnate structure. Those fructifications and the bipinnate leaf, Dicroidium feistmantelii (Johnston 1894) Gothan 1912, probably all belonged to the same parent plant. It was first suggested by John Townrow in 1962 that the sporangial heads of P. dubius have a pinnate structure; this character is confirmed herein. Pteruchus is recorded for the first time from the Carnian Tarong Basin, Queensland.The holotype of Stachyopitys simmondsii Shirley 1898 is shown to be ovuliferous. The species is recombined as Umkomasia simmondsii (Shirley 1898) comb. et emend. nov. Genuine male fructifications, previously identified as S. simmondsii, and later as Pteruchus simmondsii (Shirley 1898) Jones and de Jersey 1947, are assigned to P. minor Thomas 1933. That species comprises the smallest fructifications of the genus. Townrovia polaris Bomfleur et al. 2011 and Stachyopitys lacrisporangia Anderson & Anderson 2003 are identified as junior synonyms to P. minor. The diagnosis of Townrovia Retallack 1981 is inaccurate and the genus is insufficiently distinguished from Pteruchus.

RESUMO: O presente artigo tem por objetivo explorar, dentro da perspectiva da semiótica discursiva, alguns conceitos de desenho animado, utilizados há longa data na produção de filmes de animação dos estúdios da Walt Disney, explicitados em Frank Thomas e Ollie Johnston (1981). Queremos compreender em que medida a composição das imagens em movimento – que é, em essência, a própria forma do movimento – produz sentido, configurando-se como signo, com significante e significado próprios. À luz do percurso gerativo de sentido, focando-se na dimensão semio-narrativa (GREIMAS; COURTÉS, 2013) e na semiótica tensiva (ZILBERBERG, 2006a, 2006b), com a conceitualização de valências e missividade, podemos notar como cada sequência do movimento animatorial organiza-se em um sintagma animatorial próprio. Por meio dessa ordenação sintática, que possui um fundo tensivo, formula-se um programa narrativo e revelam-se nitidamente seus valores, actantes e fazeres. Afirma-se, então, como a forma da expressão dos movimentos não é mero suporte a um plano de conteúdo, desfazendo-se com a eficácia da comunicação, mas sim o centro de que se origina e que norteia o sentido do conteúdo, com níveis de profundidade tanto no plano do conteúdo (níveis tensivo, missivo e narrativo) como no plano da expressão (níveis rítmico e motor).PALAVRAS-CHAVE: animação; tensividade; narratividade.ABSTRACT:In this article, we propose to explore some cartoon’s concepts used by the Walt Disney Animation Studios for a long time, which we find in Frank Thomas and Ollie Johnston’s The Illusion of Life: Disney Animation (1981). By the perspective of Discoursive Semiotics, we can comprehend how the motion pictures’ composition, the form of motion, has its meaning, becoming a sign itself, with a signifier and a signified. Using traditional Greimasian Semiotics’ generative course (GREIMAS; COURTÉS, 2013), focusing on the semio-narrative level, and Zilberberg’s Tensive Semiotics and its valencies and missivity (ZILBERBERG, 2006a 2006b), we note that every motion sequence in a film organizes itself in a syntagm. Within this syntactic order with tensive base, narrative programs appear along with its values, actants and doings. Hence, we see how expression forms are not only support to content forms which disappear after effective communication, but that expression forms are, in this animatorial articulation, the originating center of meaning, with its own generative levels in the expression plane (rhythm and motor levels) and content plane (tensive, missive and narrative levels). It also opens a door to formulate a grammar for expression forms in SemioticsKEYWORDS: animation; tensivity; narrativity.

A number of casts of complete or portions of plesiosaurs from the Lower Jurassic of England are stored or displayed in the Geological Museum of Trinity College, Dublin. The historical significance of these has only relatively recently been realised. They include specimens from the collections of Thomas Hawkins, William Willoughby Cole the Earl of Enniskillen, and the Bristol City Museum. They came into the possession of Trinity College, Dublin either by donation from the Geological Society of Dublin in 1848 or from William Johnston Sollas in the late 1800s. These casts include the holotype of Thalassiodracon hawkinsi (Owen, 1838); a �sternum� and �scapula� illustrated by Thomas Hawkins and now referable to Eurycleidus arcuatus (Owen, 1840); a complete skeleton of Plesiosaurus macrocephalus Owen, 1838; the right and left side of a skull of Eurypterygius communis(Conybeare, 1822) the original of which was at the Birmingham Philosophical Institution; the skull and the right front flipper of Rhomaleosaurus megacephalus(Stutchbury, 1846); and a badly damaged cast of Attenborosaurus conybeari (Sollas, 1881). The latter two examples are important because the originals once in Bristol were destroyed in 1940 during the Second World War.

O objetivo deste artigo é apresentar uma possibilidade de análise para a videoanimação Menina bonita do laço de fita (2014), tendo em vista as linguagens verbal, visual e sonora que compõem esse texto multimodal. Para tanto, adotamos como procedimento metodológico um levantamento bibliográfico e webliográfico, que apoia a análise da videoanimação nos pressupostos da Gramática do Design Visual (KRESS; VAN LEEUWEN, 2006), dos 12 Princípios da Animação da Disney (THOMAS; JOHNSTON, 1995) e em concepções sobre as vozes, as músicas e os efeitos sonoros (FURNISS, 2014). Dessa forma, chegamos a considerações que demonstram como essa abordagem pode não só contribuir para o ensino de língua portuguesa, mas também tratar de temas atuais, como o combate ao racismo e a valorização da identidade negra.

Dalam perancangan sebuah film animasi, penerapan prinsip dasar animasi merupakan tahapan penting untuk membantu memperkuat kualitas visual film. Duabelas prinsip dasar animasi diperkenalkan oleh dua animator Disney, Ollie Johnston dan Frank Thomas, untuk membuat film animasi terlihat lebih realistis. Film AAA Walk Cycle adalah film animasi pendek yang dirancang menggunakan prinsip dasar animasi. Film ini memperlihatkan seorang tokoh melakukan walk cycle (siklus berjalan kaki) secara berulang (loop). Perancangan film ini dilakukan dalam beberapa tahapan, antara lain proses penggambaran keypose, inbetween, dan penambahan gerakan penunjang menggunakan beberapa perangkat lunak seperti Adobe Photoshop dan After Effects. Terlepas dari terbatasnya jenis gerakan yang ditampilkan, penerapan beberapa prinsip dasar animasi menjadi poin penting dalam perancangan animasi walk cycle yang dibutuhkan dalam film. Lebih lanjut, penerapan prinsip dasar animasi dapat memperkuat kualitas visual film AAA Walk Cycle.

The nematode genus Labiobulura Skrjabin & Schikhobalova, presently known from bandicoots (Isoodon Desmarest and Perameles Geoffroy), and bilbies (Macrotis Reid) from Australia and rodents (Leptomys Thomas) from Papua New Guinea is revised. Diagnoses of Labiobulura, Labiobulura (Archeobulura) Quentin and Labiobulura (Labiobulura) Quentin and a key to all species of the genus are given. Five species are redescribed: L. (A.) leptomyidis Smales from L. paulus Musser, Helgen & Lunde, L. (A.) peragale Johnston & Mawson from M. leucura (Thomas), L. (L.) baylisi Mawson from I. macrourus (Gould) and P. nasuta Geoffroy, L. (L.) inglisi Mawson from I. obesulus (Shaw), P. bougainville Quoy & Gaimard and P. gunnii Gray, L. (L.) peramelis Baylis from I. macrourus and two are described as new: L. (A.) perditus from P. bougainville, L. (L.) quentini from I. obesulus and the identification of the hosts determined. The significance of the relationship between the placement of the amphids and cephalic papillae and the labial lobes is discussed and the denticles surrounding the mouth opening in the sub genus Labiobulura are described, both for the first time. There is evidence for host specificity in the Archeobulura with each parasite species limited to a single host species but less so for the Labiobulura with three of five species found in more than one host species.

The text on these pages is a brief introductory note by the guest-editor on the contents of Part 1 of the present issue. Of the three parts of this special number of Ekistics, the first, entitled "Reflections on Gottmann's thought," contains 13 articles in three groups. In the first group are those that focus on the thought of Jean Gottmann, such as the papers of Robert Harper, John Agnew, Luca Muscarà, Nicolas Prevelakis, Jean-Paul Hubert as well as Pavios Tzermias whose essay concentrates more on the relationships between history and geography. The papers in the next group - contributed by Christian Lagarde, Jean Laponce, Ron Johnston, Yasuo Miyakawa and the present guest editor - refer indirectly to the thinking of Gottmann. Lastly are essays by Paul Claval who highlights some relationships with Gottmann's thought, and Thomas Pierre Fournel who provides an in-depth analysis of new geographical realities.

This article reviews the analysis of the functions and roles of 12 basic principles of animation in the animation produced in Indonesia “Adit & Sopo Jarwo”. This serial produced by MD Animation was released on Januari, 27th, 2014 and broadcasted from a TV station in Indonesia. This film is about the friendships and daily adventures developed in the plots between the main characters, Adit, Dennis, and Sopo Jarwo. This study is based on the initial assumption, that is, one of the indicators of a good animation is how the impression of every movements is created by the animators. To achieve such a good and natural impression of movements, the animators should apply the 12 basic principles of animation, as formulated by Frank Thomas and Ollie Johnston (1981), two former animators of Disney’s. The analytical process itself was using descriptive qualitative method. The purpose of this article is to show the intensity of the using of 12 basic principles of animation in “Adit & Sopo Jarwo”.

To investigate chromosome segregation in Saccharomyces cerevisiae, we examined a collection of temperature-sensitive mutants that arrest as large-budded cells at restrictive temperatures (L. H. Johnston and A. P. Thomas, Mol. Gen. Genet. 186:439-444, 1982). We characterized dbf8, a mutation that causes cells to arrest with a 2c DNA content and a short spindle. DBF8 maps to chromosome IX near the centromere, and it encodes a 36-kDa protein that is essential for viability at all temperatures. Mutational analysis reveals that three dbf8 alleles are nonsense mutations affecting the carboxy-terminal third of the encoded protein. Since all of these mutations confer temperature sensitivity, it appears that the carboxyl-terminal third of the protein is essential only at a restrictive temperature. In support of this conclusion, an insertion of URA3 at the same position also confers a temperature-sensitive phenotype. Although they show no evidence of DNA damage, dbf8 mutants exhibit increased rates of chromosome loss and nondisjunction even at a permissive temperature. Taken together, our data suggest that Dbf8p plays an essential role in chromosome segregation.

This study documents the helminth assemblages of grassland melomys: Melomys burtoni (Ramsay, 1887), Melomys sp. cf. burtoni and Melomys lutillus (Thomas, 1913). In total, 22 helminth species comprising one cestode and 21 nematodes from 112 hosts were found. All the specimens of Physaloptera spp. examined proved to be Physaloptera banfieldi Johnston & Mawson, 1941 with P. troughtoni Johnston & Mawson, 1941 the junior synonym. The dominant helminth group was the trichostrongyloid nematodes including Odilia melomyos (Mawson, 1961) and O. mackerrasae (Mawson, 1961). The most prevalent, O. melomyos, occurred in each of the host species across all areas sampled (New South Wales, Northern Territory, Papua New Guinea, Queensland and Western Australia). The helminth assemblage of M. burtoni from hosts from New South Wales and Queensland was the most diverse. The helminths of M. burtoni from the Northern Territory and of M. lutillus from Papua New Guinea were subsets of that assemblage. That of M. cf. burtoni from Western Australia, with only six helminth species, was not dominated by trichostrongyloids, three of the six species were not found in other localities, and, with Sorensen’s Indices of 18.2% when compared with the helminths from Papua New Guinea and the Northern Territory and 24% when compared with Queensland, was the least similar. No substantial differences were found between the helminth assemblages of the grassland melomys group, excluding M. cf. burtoni, and Melomys cervinipes (Gould, 1852), the fawn footed melomys. This was reflected in a Sorensen’s Index of 67.9%. The time between the arrival of Melomys into Australia during the Pleistocene and the present day suggests that the trichostrongyloids O. melomyos, O. mackerrasae and O. mawsonae (Durette-Desset, 1969) may have travelled with their rodent hosts from New Guinea to Australia and other helminths in the assemblage may have been acquired in Australia.

Multimedia is an amalgamation of several components, namely text, audio, animation and video with digital processes managed by computers. Animation is one of the components contained in multimedia, to convey information and as a means of entertainment. In the animation and design of cartoon characters, animators have often turned to the study of biological theories and observation of human actors and animals to capture lifelike movements and emotions more successfully. Charles Darwin’s principle of antithesis, as one of the principles he considered to be responsible for the expression of emotions in animals, would seem to be of distinctive importance in the development of animation. By revisiting Darwin’s original idea in the context of the principles of animation formulated by Thomas and Johnston, we are able to assess its application and relevance in the expressions of emotions in cartoon animal characters. The article concentrates on the emotive function of animal social communication as outlined in zoosemiotics, while taking into account that the expressions of animal characters are directed at the viewer. The principle of antithesis, as a descriptive tool, aids us in considering the diversity of modalities used simultaneously in affective communication, and serves to explicate human interpretations of the anthropomorphic and zoomorphic projections onto the behaviour of cartoon animal characters. This paper offers insight into the potential expansion and re-evaluation of unattested principles in animation, which can be utilized by animators in the creation of more dynamic and expressive animated characters.

The Geological Survey of Canada, the oldest scientific organization in the nation, was founded in 1842, at a time when major changes were taking place in the sciences of geology, chemistry and biology. Geochemistry was not yet recognized as a science in North America, although the word had been coined and the field outlined in 1838 by C. F. Schönbein, the Swiss chemist. Thomas Sterry Hunt (1826-1892) was appointed chemist and mineralogist to the Geological Survey in 1847 by Sir William Logan, first director of the Survey. For the next 25 years Hunt carried out geochemical research on most aspects of the science and added materially to the establishment of various geological principles. Sterry Hunt has been called the "Father of Chemical Geology, or Geochemistry, in North America." Following Hunt, many able men, including B. J. Harrington, F. D. Adams, G. C. Hoffman, and R. A. A. Johnston occupied the position of chemist and mineralogist to the Survey during the period 1872-1922. They were followed by E. Poitevin and H. V. Ellsworth who served from 1922 until 1952. H. V. Ellsworth laid the foundations of 20th Century geochemistry in the Geological Survey. His extensive studies of the distribution of rare elements (e.g., Li, Zr, Nb, U, Th, etc.) in Canadian minerals and rocks, and his studies of geochronology employing uranium-lead methods, had universal significance in geochemistry.

According to the two editors, it has been a long time since the entire history of Welsh literature was treated in one volume, so the new effort by Geraint Evans and Helen Fulton must be certainly welcomed. But for a little housekeeping, so to speak, they only refer to the volume Hanes Llenyddiaeth Gymraeg hyd 1900, published by Thomas Parry in 1953, translated into English in 1955. A simple search in any online catalog, however, unearths other valuable studies, such as Bobi Jones’s The Dragon’s Pen: A Brief History of Welsh Literature (1986), Mathias Roland’s Anglo-Welsh Literature: An Illustrated History (1986), The Oxford Companion to the Literature of Wales, ed. Meic Stephens (1986), and Dafydd Johnston, The Literature of Wales (1994), none of which are included in the final cumulative bibliography. Of course, this does not mean at all that new efforts in that regard could be dismissed, on the contrary. In fact, as Evans and Fulton correctly emphasize, both with respect to the use of English and the use of Welsh, the time has come to approach the entire corpus of literary texts as produced in Wales from the early Middle Ages until today in a holistic fashion, although this work was here divvied up among a larger number of scholars responsible for individual literary-historical periods. It would have been helpful, however, if the editors had reviewed critically the previous efforts to write a literary history of Wales in order to highlight better the new approaches and methodologies, which are explained subsequently, but not clearly enough in contrast to previous publications.

Formation of a novel structure, the aggresome, has been proposed to represent a general cellular response to the presence of misfolded proteins (Johnston, J.A., C.L. Ward, and R.R. Kopito. 1998. J. Cell Biol. 143:1883–1898; Wigley, W.C., R.P. Fabunmi, M.G. Lee, C.R. Marino, S. Muallem, G.N. DeMartino, and P.J. Thomas. 1999. J. Cell Biol. 145:481–490). To test the generality of this finding and characterize aspects of aggresome composition and its formation, we investigated the effects of overexpressing a cytosolic protein chimera (GFP-250) in cells. Overexpression of GFP-250 caused formation of aggresomes and was paralleled by the redistribution of the intermediate filament protein vimentin as well as by the recruitment of the proteasome, and the Hsp70 and the chaperonin systems of chaperones. Interestingly, GFP-250 within the aggresome appeared not to be ubiquitinated. In vivo time-lapse analysis of aggresome dynamics showed that small aggregates form within the periphery of the cell and travel on microtubules to the MTOC region where they remain as distinct but closely apposed particulate structures. Overexpression of p50/dynamitin, which causes the dissociation of the dynactin complex, significantly inhibited the formation of aggresomes, suggesting that the minus-end–directed motor activities of cytoplasmic dynein are required for aggresome formation. Perinuclear aggresomes interfered with correct Golgi localization and disrupted the normal astral distribution of microtubules. However, ER-to-Golgi protein transport occurred normally in aggresome containing cells. Our results suggest that aggresomes can be formed by soluble, nonubiquitinated proteins as well as by integral transmembrane ubiquitinated ones, supporting the hypothesis that aggresome formation might be a general cellular response to the presence of misfolded proteins.

The paper has not been peer reviewed. Hydraulic fractures created during completions are assumed to produce back to the original well. While multiwell-pad completions increase efficiencies, they complicate fracture connectivity between wells. The proximity of newly completed wells to a pre-existing producing well suggests a depleted zone that can “steal” fracture surface area. The complete paper presents a real-time method to estimate the stage-to-stage interference and well-to-well interference and their implications on completions efficiency. Introduction The primary challenge for the industry with regard to unconventional resource development is to optimize well spacing to improve well net present value. Shale heterogeneity and completion design influence fracture orientation and geometry. The industry still lacks accurate methods in characterizing these. Fig. 1 shows three well-spacing types that the authors write could be classified as “good,” “bad,” or “ugly”—the A+ spacing type (good), the B spacing type (bad), and the C spacing type (ugly). The A+ spacing type has no interference between wells that perfectly drain the reservoir; however, it is hard to achieve. In the B spacing type, there may be 10–15% interference between either hydraulic or natural fracture systems. B spacing can be achieved with proper integration of reservoir and completion engineering. This spacing type usually can provide more predictable well-level estimated ultimate recovery (EUR). C spacing type is characterized by closer-than-needed spacing, resulting in shared stimulated reservoir volume (SRV) between wells. Some operators use this spacing when oil and gas prices are high in order to accelerate production.

ISSN 0804–4643 Short-term trials of growth hormone (GH) substitution in hypopituitary adults have shown beneficial effects on body composition. To evaluate the long-term effects on body composition, we followed thirteen GH-deficient adults (GH < 6 mU/l following standard provocative tests) for 4 years of GH replacement. At yearly intervals, serum insulin-like growth factor I (IGF-I), body weight, body mass index (BMI), waist, waist-to-hip circumference ratio (WHR) and resting systolic (SBP) and diastolic blood pressure (DBP) were determined, and body composition was assessed using three independent methods: total body potassium (TBK), bioelectrical impedance analysis (BIA) and dual-energy X-ray absorptiometry (DXA). Compared to baseline, IGF-I levels increased significantly at 1 (p = 0.0001), 2 (p = 0.0004), 3 (p = 0.006) and 4 years (p = 0.002). Body weight and BMI changed minimally at 1, 2 and 3 years and increased significantly only at the fourth year (p = 0.012 and p = 0.0009, respectively) of GH therapy. Waist and WHR decreased significantly at 1, 2 and 4 years (waist: p = 0.0009, p = 0.0004, p = 0.049; WHR: p = 0.0025, p = 0.012, p = 0.047, respectively). Neither resting SBP nor DBP changed significantly. Fat-free mass (FFM) derived from TBK and BIA increased significantly at 1 (p = 0.004; p = 0.004), 2 (p = 0.003; p = 0.05), 3 (p = 0.005; p = 0.04) and 4 years (p = 0.02; p = 0.002). Using DXA, the increase in FFM was significant at 1 (p = 0.007) and 2 years (p = 0.008) but not at 3 and 4 years. Percentage body fat measured by TBK, BIA and DXA decreased significantly at 1 (p = 0.008; p = 0.003; p = 0.03), 2 (p = 0.018; p = 0.06; p = 0.049) and 4 years (p = 0.03; p = 0.002; p = 0.04). A rise in total body water, calculated from BIA, was observed at 1 year (p = 0.004) and was maintained throughout the treatment period. These data demonstrate that 4 years of GH treatment in hypopituitary adults is associated with sustained improvement in body composition. IntroductionSocial care teams are increasingly leading the delivery of care for elderly clients and their families who are living with a terminal illness. Marie Curie, in partnership with the City of Edinburgh, East Lothian, Midlothian and West Lothian Council’s identified the need for a bespoke education programme. The LCADP encompassed a single study day, followed by an online education programme with mentorship in the participants’ work setting.AimThe overall aim of the project was to increase the knowledge and confidence of frontline staff employed by each of the four Lothian councils to care for clients living with a terminal illness.MethodsA mixed methods approach was used. A survey was completed at four time points, including an impact survey up to six months post completion of the programme. Focus groups and interviews of a sample of social care workers, mentors and managers were also completed.Results514 participants completed the single day programme. 71 social care workers continued on to the online module and mentorship programme. Both quantitative and qualitative data will be presented, demonstrating longer term impacts such as team approaches to planning and delivering care. Increased knowledge and confidence by the social care staff has resulted in a more person centred approach to care, prompt recognition of uncontrolled symptoms, and policy change.ConclusionsThrough partnership working, the programme team were able to reach more people and influence the care that is being delivered to people living with a terminal illness in their own homes and in care homes in Lothian.References. Abstract Introduction/Background: Endometrial cancer (EC) is the most common gynecological cancer in the developing world. While significant progress has been made through molecular sub-classification, the treatment of EC for many women remains contentious and better biomarkers are required to guide therapy selection. Circulating tumor DNA (ctDNA) a ‘liquid biopsy’, involves collection of tumor specific DNA via a blood test and can quantify cancer burden and detect tumor specific genomic information. The CODEC study is a prospective pilot study to assess the utility of ctDNA as a prognostic biomarker in EC with the hope that it may help predict relapse and guide selection of adjuvant therapy. Methodology: Women with Type 2 EC planned for surgery and consenting to the collection of biospecimens were enrolled prior to surgery and followed prospectively. Blood was collected perioperatively and then every three months for up to two years and at relapse. Broad sequencing of participants’ tumors was performed, the results of which guided the design of personalized droplet digital PCR (ddPCR) assays for ctDNA analysis. ctDNA was extracted from 5ml of plasma and tested in quadruplicate. PET defined metabolic tumor volume was compared to ctDNA and analysis of ctDNA and relapse free survival (RFS) performed using a Cox proportional hazards regression model. Results: Somatic mutations to guide ddPCR analysis were detected in 45 enrolled women stages 1- 4 including pathogenic variants in POLE. ctDNA results are currently available for 30 participants (remaining results awaited). 12/30 participants had detectable ctDNA preoperatively and 5/25 at Time Point 3 (2-6 weeks post-surgery). ctDNA levels varied significantly across the cohort but results to date indicate that perioperative ctDNA positivity is associated with relapse. Interestingly in those that had detectable ctDNA preoperatively, the majority had a rise in ctDNA intraoperatively. Conclusion: This proof-of-concept analysis showed that ctDNA can successfully be detected in the perioperative setting and is the first study in EC to compare ctDNA with PET findings. Although testing is still underway, the early results indicate that ctDNA maybe a useful biomarker to predict relapse and guide the selection of adjuvant therapy. Abstract Background: Arginine deprivation with ADI-PEG20 (pegargiminase) alone or combined with chemotherapy displayed antitumor activity in early phase clinical trials of argininosuccinate synthase 1 (ASS1)-deficient cancers, including pleural mesothelioma (PM). The non-epithelioid subtype of mesothelioma is particularly aggressive and arginine-auxotrophic due to frequent ASS1 loss. ATOMIC-meso is a pivotal phase 2-3 trial comparing standard of care (SOC) chemotherapy plus pegargiminase or placebo for patients (pts) with non-epithelioid PM. Methods: In this double-blind, placebo-controlled, phase 2-3 trial, we randomly assigned ECOG PS 0-1 pts with chemonaïve non-epithelioid PM in an intention-to-treat 1:1 ratio to receive q3 week SOC chemotherapy with pemetrexed (500mg/m2 i.v.) and cisplatin (75mg/m2 i.v.) with q1 week pegargiminase (36 mg/m2 i.m.) or placebo i.m. up to 6 cycles. Carboplatin (AUC5) was permitted in the phase 3 portion instead of cisplatin. Maintenance therapy with pegargiminase or placebo was continued until progression, toxicity or 24 months. The primary end point was median OS (mOS) and secondary end points included median PFS (mPFS), safety, pharmacodynamics and immunogenicity. ORR was assessed by blinded independent central review (BICR) for the phase 2 portion only using modified RECIST or RECIST v1.1. Results: 249 pts with non-epithelioid PM (median age 71, range 28-86; male 82.7%; 48.2% biphasic and 51.8% sarcomatoid) from 5 countries were randomized between Aug 2017 and Aug 2021: 125 pts were assigned to the pegargiminase-chemotherapy group and 124 pts to the placebo-chemotherapy group. The experimental arm showed a superior mOS respect to SOC: 9.3 months (95% confidence interval [CI], 7.9-11.8) vs. 7.7 months (95% CI, 6.1-9.5) (hazard ratio [HR], 0.71; 95% CI, 0.55-0.93; p=0.023). Also, the mPFS was higher in the experimental vs. control arm: 6.2 months (95% CI, 5.8-7.4) vs. 5.6 months (95% CI, 4.14-5.91) ([HR], 0.65; 95% CI, 0.46-0.90; p=0.019). The ORR by BICR was 13.8% in the pegargiminase-chemotherapy group vs. 13.5% in the placebo-chemotherapy group (p=0.95) with more stable disease in the former (71.3% vs. 62.9%). Plasma arginine declined with a reciprocal increase in citrulline. Anti-ADI-PEG20 antibodies were detected in 97.4% of patients by week 25 on pegargiminase. Grade ≥ 3 treatment-related adverse events to pegargiminase occurred in 28.8% and to placebo in 16.9%; grade ≥ 3 drug hypersensitivity and skin reactions occurred in 3.2% and 1.6% in the experimental arm, respectively, and none on placebo. Post-study drug treatment was comparable in both arms (45.6% post-pegargiminase vs. 46.8% post-placebo). Abstract Background: Dying cells play an important role in CD8+ T cell-mediated immunity. While apoptosis is mainly immunologically silent, necroptosis is immunogenic and can more efficiently initiate an adaptive immune response that could be augmented by immune checkpoint inhibition (ICI). Inhibitor of Apoptosis (IAP) proteins are key gatekeepers of how a cell dies, and frequently overexpressed in cancer. ASTX660 (tolinapant) is a triple blocker of cIAP1/2 and XIAP, sensitizing cancer cells to death ligands through ripoptosome formation, activating ‘danger’ co-stimulatory signals and dendritic cell (DC) maturation required to prime a potent antitumor response. Methods: We assessed the safety, pharmacodynamic (PD), and clinical activity of 7 days of oral ASTX660 given prior to pembrolizumab 200 mg (Q3W), in patients (pts) refractory to standard therapy. TITE-CRM was used to explore ASTX660 doses 90-180 mg. Serial paired blood and tumor samples were analyzed to interrogate the effect of ASTX660 on cell death, tumor microenvironment and host immune system. Results: 22 pts have been enrolled; median age 55 years (range 38-81); male:female 5:17; all ECOG PS ≤1; median 4.5 prior therapies. Tumor types included breast, head and neck (HN), hepatocellular, cervical, pancreatic and cholangiocarcinoma. Combination ASTX660-pembrolizumab is currently recruiting at highest dose 180 mg without any dose limiting toxicities (DLT) seen within the 6-week DLT observation period. Most common adverse events (AE) were G1-2 rash, nausea, and asymptomatic increases in amylase, lipase and transaminases; all reversible and easily manageable. Four iRECIST confirmed partial responses were seen in 10 efficacy evaluable pts treated at biologically active dose levels 120 and 150 mg (Overall response rate 40%), including 2 pts with ER+ HER2- breast cancer, 1 cervical adenocarcinoma pt (exceptional response following initial pseudoprogression) and a HN cancer pt refractory to ICI monotherapy. 3/4 responders are ongoing. Efficacy data from pts treated at highest dose level 180 mg is pending. FACS of paired blood samples from responders shows a significant rise in %CD57+/CD3+ cytotoxic effector T cells [mean 11.90% (pre-) vs 14.25% (post-ASTX660); p=.0176]. These changes were not seen in the non-responders group. Full PD from paired blood and tumour sampling will be shared at the meeting. Conclusion: ASTX660-pembrolizumab is is well-tolerated with durable antitumor responses in pts with immunologically cold tumors, providing the first clinical proof-of-concept of immunogenic cell death induction leading to effective adaptive immune response when combined with ICI. Recommended Phase 2 dose optimization and expansion cohorts in immunologically cold cancers are ongoing. Jones, Stanley C., SPE, Marathon Oil Co. Abstract Displacements were conducted in Berea cores to gain insight into the mechanism of tertiary oil displacement and propagation by a micellar slug. Contrary to expectation, propagation by a micellar slug. Contrary to expectation, the first oil mobilized by micellar fluid was among the first oil (instead of the last oil) to be produced, giving the appearance of either viscous fingering or of unusually large dispersion. To eliminate the possibility of unfavorable mobility ratios caused by oil/water/surfactant interaction, we conducted several runs in which an injected hydrocarbon displaced another hydrocarbon, initially at residual saturation. In other experiments, water (the wetting phase) at irreducible saturation was displaced by a distinguishable injected aqueous phase. Injected hydrocarbon appeared in the produced fluids immediately after oil breakthrough, yielding behavior similar to the micellar-slug experiments. Even with a favorable viscosity ratio of less than 0.01, the apparent dispersion was huge. However, mixing zones in the wetting-phase displacements were quite normal and similar to those observed for single-phase flow. Nonwetting-phase fronts (injected hydrocarbon displacing resident hydrocarbon) are smeared much more than wetting-phase fronts because the entrance of hydrocarbon into smaller water-filled pore throats is delayed until the capillary entrance pressure is overcome by differences in the flowing oil and water pressure gradients. Oil might not be displaced from the smaller pores until long after oil breakthrough. Nonwetting-phase dispersion, which occurs in many EOR processes, can be expected to be one or two orders of magnitude greater than dispersion measured in single-phase-flow experiments. Entrance of the wetting phase, however, is not delayed; hence, wetting-phase Mixing zones are short. Introduction Experiments for this study were inspired by the question: How is residual oil, which has been mobilized by a micellar slug, transported? More specifically, does the first oil mobilized by a slug (near the injection end of a core) contact and mobilize oil downstream from it, which displaces more oil even farther downstream? If this were the case, the first oil to be produced would be the most-downstream oil (i.e., oil nearest the outlet). The last oil produced would be the first oil mobilized from the produced would be the first oil mobilized from the injection end of the core. This scheme is somewhat analogous to pushing a broom across a floor covered with a heavy layer of dust. The first dust encountered by the broom stays next to the broom. As the accumulated layer of dust in front of the broom becomes adequately compacted, it pushes dust ahead of it to from an ever-widening band or "dust bank" ahead of the broom. The dust farthest ahead of the broom is the first to be pushed into the dustpan, and the dust first encountered by the broom is the last to be pushed in. Or is this concept all wrong? Another model postulates that the oil first contacted by a micellar slug is mobilized and quickly travels away from the slug so that the downstream oil is contacted and mobilized by the slug, not by the first-mobilized oil. If this process were to proceed to its logical conclusion, the first-produced oil would proceed to its logical conclusion, the first-produced oil would be from the inlet end of the core, and the last-produced from the outlet end. Either of these two extremes would be modified by dispersion, which smears sharp fronts by mixing displaced and displacing fluids. Dispersion in porous media has been investigated extensively. Perkins and Johnston have reviewed several studies, mostly involving single-phase flow. The simultaneous injection of the water with light hydrocarbon solvents is a technique used to reduce solvent mobility and viscous fingering. Raimondi et al. performed steady-state experiments in which flowing performed steady-state experiments in which flowing water and oil were miscibly displaced by the simultaneous injection of water and solvent. They found that the longitudinal mixing coefficient for the hydrocarbon phase increased sharply with increasing water above the irreducible saturation. The displacement of the wetting phase was not greatly affected by the presence of the nonwetting phase. However, a large amount of oil that initially phase. Character education can equip young children to become Indonesia's golden generation who are superior and skilled in noble work. This research aims to produce an early childhood character learning model based on local Indonesian wisdom through the Playing Center title which will be implemented at Early Childhood Education Institutions (ECE) in Indonesia. This research method uses the steps of the Borg and Gall research and development model. The subjects in this research were 30 teachers from five ECE institutions in the DKI Jakarta area. The data used as the basis for building a framework for early childhood character learning models based on local intelligence were collected using observation and interview techniques. The findings show that the teacher prepares a learning plan program at the beginning of each semester which is listed in the weekly program and daily program. However, the teacher has not fully prepared a simulation program that leads directly to the introduction of early childhood character. Identification of character values ​​in early childhood is done by providing examples of behavior and speech at school. The media that has been used to identify children's characters is through posters and non-digital story books. The learning environment needs to be improved because teachers have not been optimal in organizing the play environment in providing stimulation to children in class. The results of this research will enrich the character education model following the potential of the institution and the cultural background of early childhood as Indonesia's golden generation. The prevalence of ADHD children increases every year. Some researchers have shown that psychosocial behavior therapy (antecedents-consequences modification) was effective to decrease hyperactivity and increase attention to ADHD children. This study aims to find out the effectiveness of antecedents-consequences modification by parents and teachers to decrease hyperactivity and increase attention to a 6 years old boy with ADHD. The study was a single case experimental design. Psychosocial behavior therapy has been used with antecedents-consequences modification. The antecedents-consequences modification was applied by teacher at school and parents at home. Data were analyzed using Wilcoxon Signed Rank Test. Results showed that there’s a significant decrease of hyperactivity behavior and significant increase of doing his assignment both at school and also at home. Not only about the content of behavior therapy itself, but how to give the therapy is important. Parents and teacher should do the therapy consistently, immediately, specifically and saliency to reach the target of intervention.
