Résumé :

Abstract We recently incorporated the use of the automated Sepax Cell Processing System (Biosafe SA, Switzerland) for red cell and volume reduction of cord blood units (CBU) before cryopreservation. Now that we have been routinely using this new technique in the laboratory for about six months, we decided to compare the results of this method with our standard manual processing method (Rubinstein, et al, PNAS1995,; 92: 10119–22). For both methods, hespan is added to the cells at final concentration of 20% (v/v). With the Sepax system, after addition of hespan, the cell bag is connected to the Sepax tubing set with the final freeze bag pre-attached to the set. After completion of the automated procedure, buffy coat is collected in the final freeze bag. Cryoprotectant solution is then added directly to the freeze bag. In the manual method, buffy coat and white cell rich plasma layer is collected after first centrifugation step and the white cells are separated from the plasma after the second centrifugation step. Cryoprotectant solution is then added to the cells before transfer to the final freeze bag for cryopreservation. Since the Sepax device is an automated procedure, issues could arise (i.e. short term loss of electrical power) that would require us to reprocess the CBU before cryopreservation. The Sepax system allows for recovery and reprocessing of the cell using the ‘Purge mode’. We used 5 CBU units to evaluate TNC recovery and viability after purging the cells once and reprocessing. These data demonstrates that both methods are equivalent with respect to cell recovery. However, the Sepax System substantially reduces processing time and hands-on operator intervention. Additionally system provides, closed-system processing, bar code reading capability and run data print-out suitable for GMP manufacturing settings.

Abstract Fetal stem cells have several therapeutical advantages when compared to their adult counterparts. Cell populations residing in “waste” tissues (cord blood, umbilical cord and placenta) may be collected without any medical or ethical contraindications concerning mother or newborn baby. Cord blood hematopoietic stem cells are routinely used for clinical transplantations, and the methods for their collection, isolation and freezing are now well standardized. We have tested the efficiency of cell separator (Sepax, Biosafe) in volume reduction of cord blood units, and in washing-out DMSO after cell thawing. Results of the volume-reduction procedure revealed that the average MNC recovery rate was 91±5%, cell viability was ≥90%, and CD34+ cell recovery rate was 92±4%. Volume reduction process eliminated 88±4.5% of red cells. In vitro clonogenic potential of cells isolated with Sepax did not differ from this of cells isolated according to the standard HES sedimentation protocol. Protocol of Sepax-based washing of thawed cells resulted of 9±4% cell loss and 10±6% decrease in cell viability, when compared to thawed non-processed cells. We conclude, that Sepax separator may be used both for cord blood volume reduction, and DMSO elimination from thawed samples. The other cord blood stem cell population, identified by us (J Cell Sci 2002, 115:2131–2138), was plastic-adherent, nonhematopoietic stem cell differentiating at least into neuropoietic progeny. Attempted optimisation of cell isolation/culture methods did not allow for successful isolation of these cells from more than approximately 20% cord blood samples - we postulate, that this cell population, when detected in mature newborn blood, is rather a remnant of cells circulating in earlier stages of fetus development. The population of cells residing in Wharton jelly of umbilical cord (CD34−, CD45−, CD29+, CD44+, CD51+, CD105+, SH-2+, SH-3+, plastic-adherent) is capable, according to published data, to differentiate into several tissues, resembling the adult mesenchymal stem cells. We were able to isolate these cells from 85% of cord collections, and to expand them for 5–10 passages in in vitro cultures in IMDM + 10% FCS media without growth factors added. We have also developed the protocol, allowing for storage of frozen umbilical cord tissue without preliminary cell isolation and expansion, allowing for labor-economical and cost-economical banking of large numbers of umbilical cord cell samples. Conclusions: Sepax application for cord blood volume reduction, or DMSO elimination from thawed cell samples, is an improvement in cord blood bank routine protocols. The low frequency of non-hematopoietic plastic-adherent stem cells in cord blood samples, collected during mature deliveries, is rather a biological phenomenon than the inadequacy of laboratory techniques, and may reduce the utility of these cells for clinical purposes. Stem cells from Wharton’s jelly of umbilical cords, if confirmed their “stemness”, can be an easy to obtain cell source for tissue engineering. Our technique of freezing-storage of the umbilical cord tissue may be useful for low-cost, time-saving banking of large numbers of fetal stem cells.

Résumé :

OBJETIVO: Analisar a segurança e a eficácia da infusão percutânea no foco da pseudoartrose do aspirado de medula óssea total e da infusão de células mononucleares purificadas e concentradas. MÉTODOS: Foram tratados seis pacientes portadores de pseudoartrose de tíbia ou de fêmur. Quatro receberam infusão do aspirado de medula óssea autóloga total obtida da crista ilíaca e dois receberam o aspirado de medula óssea autóloga processada com Sepax®. Não foi alterado o método de fixação primária ou exposto o foco da pseudoartrose. Realizou-se exame físico e avaliação radiográfica 2, 4 e 6 meses após o procedimento. Após a consolidação, foi avaliado o grau de satisfação do paciente. RESULTADOS: Não verificamos qualquer complicação decorrente dos procedimentos. No primeiro grupo (aspirado total) a consolidação foi obtida entre três e vinte e quatro semanas e, no segundo grupo (Sepax®), entre 12 e 16 semanas. O grau de satisfação dos pacientes teve a média aumentada de 2,75 para 9 no grupo I e de 1 para 8,5 no grupo II (p=0,0156). CONCLUSÃO: Concluímos que o tratamento das pseudoartroses com os métodos avaliados mostraram-se eficazes e seguros independente do método de estabilização utilizado. Nível de Evidência II, Estudo Prospectivo Comparativo.

Abstract Abstract 3219 Poster Board III-156 Introduction Dimethylsulfoxide (DMSO) containing cryoprotective solutions are routinely used for the storage of hematopoietic progenitors (HP). At room temperature, DMSO is toxic for the cells and may produce severe adverse reactions during their infusion, especially in the pediatric patients. These problems can be avoided by washing the cells prior to the infusion. Our objective was to test if an automatic washing method (Sepax S-100, Biosafe) allowed us to preserve the CD34+ cell numbers with an adequate viability and engraftment potential. Material and Methods Forty five peripheral blood HP apheresis that have been cryopreserved using autologous plasma plus 9% DMSO were studied. After rapid thawing in a water bath at 37° C, an automatic wash with the Sepax S-100 (2 washes cycle) was performed. Nucleated cell levels determined by an hematology analyzer, flow cytometry CD34+ cell counts and Trypan Blue cell viability test were performed on aliquots collected prior to and after the washing technique. The paired Student's t-test and the Pearson's correlation coefficient were used for the statistical analysis. Results The mean total nucleated cell (TNC) and CD34+ cell recovery was 75,47% ± 3, and 94,66% ± 4,62 respectively. In spite of the TNC significant loss (p<0,001), there were no significant differences between the pre and post-washing CD34+ cell numbers (p=0,08) or viability (p=0,19). The TNC loss had no correlation with the pre or post-washing number of platelets (p=0,18 and p=0,16 respectively), neither with the loss of platelets during the procedure (p=0,31). In contrast with the 40% of untoward reactions recorded in our historical data of 226 DMSO containing cell infusions, we observed just one adverse effect (2,22%) with the washed cells. One patient reported a transient mild abdominal pain with nausea and one vomit. Median time to neutrophil engraftment (>500 cells /mL) and platelet engraftment (>50.000 cells/mL) were 11 ± 0,2 and 20 ± 1,4 days respectively. Conclusions The Sepax S-100 automatic wash protocol of DMSO containing peripheral blood progenitor cells determines a good CD34+ cell recovery and preserves their viability and engraftment potential. This method avoids the DMSO infusion related adverse events and it constitutes a closed and easy to do procedure. Disclosures No relevant conflicts of interest to declare.

Abstract Background Cord blood transplantation (CBT) is an increasingly used alternative to bone marrow or peripheral blood stem cell transplantation, but is associated with slower neutrophil and platelet recovery. One strategy to overcome the delayed hematopoietic recovery of CB is to correct the decreased fucosylation of CB cell surface molecules which is thought to impair homing to the bone marrow of the limited numbers of stem and progenitor cells in the CB graft. Our pre-clinical murine xenograft models have demonstrated that human CB-derived progenitor cells treated with recombinant human fucosyltransferase-VI (ASC-101:America Stem Cell Inc.) prior to infusion resulted in more rapid and higher levels of human engraftment as compared to untreated CB progenitors (Robinson et al Exp Hematol, 2012). We therefore sought to study this novel strategy in a clinical trial where recipients with hematologic malignancies receive a double CBT where one CB unit is fucosylated prior to infusion. Objective In an effort to improve engraftment following cord transplant, we tested the ability of a 30-minute ex vivo fucosylation treatment to shorten time to neutrophil and platelet recovery. Methods Cell Processing: The unmanipulated CB unit with the highest total nucleated cell (TNC) dose was thawed, washed on the Sepax device (Biosafe) and infused first. The second CB unit which had the smaller TNC dose was thawed and washed using 10% Dextran-40/5% human serum albumin and the cells treated at 106cells/ml for 30 minutes at room temperature with recombinant human fucosyltransferase VI (ASC-101) and substrate GDP-fucose (America Stem Cell Inc). The fucosylated cells were then washed on the Sepax and infused. After the procedure fucosylated (CD34+ CLA+) CD34+ cells increased from a median of 33% to 99%. Clinical 10 patients with AML (5), MDS (2), NHL (2), or HL (1) have been enrolled to date. Three patients are too early to evaluate engraftment; therefore, first 7 evaluable patients are reported here. Median age was 55 (range 26 -62) years and median weight 87 (range 61-97) kg. All patients were conditioned with fludarabine 160mg/m2, melphalan 140mg/m2, and ATG 3mg/kg. Tacrolimus and MMF were used for GVHD prophylaxis. Results Median time to absolute neutrophil count ≥ 0.5 X 109 /L was 14 (range 12-28) days. Median time to platelet count ≥ 20 X 109/L was 33 (range 18-100) days. One patient had secondary graft failure and was rescued with backup autologous stem cells. Four patients had engraftment of the fucosylated unit and 2 of the unfucosylated unit. Two patients developed grade 2 acute graft versus host disease. No infusion related toxicities were seen. Conclusion Ex vivo fucosylation appears to be a safe, simple and rapid approach to enhancing neutrophil and platelet engraftment in the setting of double cord transplantation. Résumé :

Abstract Generation and characterization of unrestricted somatic stem cells (USSC) from cord blood (CB) was described by our group and has been well established under laboratory conditions [Koegler et al 2004, 2005 and 2006; Sensken et al 2007]. Due to their proliferative and differentiation capacity, USSCs are an interesting candidate for the future development of cellular therapy for tissue repair and tissue regeneration as well as a supportive cell layer to support hematopoietic reconstitution. Since generation and expansion under GMP-grade conditions is mandatory for use in clinical application, the automated cell processing system Sepax (BIOSAFE) with the CS900 separation kit was used for mononuclear cell separation and the subsequent generation of the USSC colonies in the presence of 30% GMP-grade fetal calf serum (Perbio), low-glucose DMEM-medium/10-7M dexamethasone. Expansion of USSC was performed in a closed system (Macopharma) applying cell stacks (Costar Corning). Results achieved so far indicate that the generation frequency and quality of generated USSC under GMP conditions are equal or even superior (45%) to manual generation under laboratory conditions (43%). 20 cord-blood units (mean volume 88,5 +− 15,8 ml; mean number of mononuclear cells 3,1 +−0,6 *108 MNC) have been processed, resulting in 9 USSC-colony formations and lines within 14–28 days. Growth kinetics is equal to the previously established USSC-lines (∼36–48 h / population doubling). Analysis of the immunophenotype as well as the differentiation potential towards the mesenchymal, neural and endodermal lineages also showed no difference to those lines generated manually using Ficoll-separation and normal cell culture flasks (Costar Corning T225). The closed system applied here is perfectly suitable to ensure safe and easy handling of the USSC, including seeding, trypsination and harvesting. In combination with the cell stack system (1, 2, 5 and 10 layers), cell amounts of more than 1.0×109 USSC can be achieved within 4 passages. These USSC products were temperature controlled cryopreserved in the presence of 10% DMSO, HSA and dextran. USSC can be thawed and further expanded in clinical grade quality. On the basis of their pluripotency and expansion under GMP-conditions into large quantities, these USSC from cord blood, when pretested for infectious agents and matched for the major transplantation antigens, may serve as a universal allogeneic stem cell source for tissue repair and tissue regeneration.

Abstract Introduction Barcelona Cord Blood Bank (BCBB) program started for CB units (CBU) donations in 1991. In 2002, Automated Volume-Reduced method was introduced applying the closed System (Sepax®, Cytiva). In order to ensure the quality of cryopreserved units after long-term storage, CBB programs evaluate cell recovery, progenitor cells functionality such as viability and Colony forming unit (CFU), and myeloid recovery after cord blood transplant (CBT). Objectives This retrospective study investigated quality parameters of CBUs stored for 1-20 years processed by automated volume-reduced and cryopreserved and stored in Bioarchive® system, (Thermogenesis) showing the effect of cryopreservation on variables measured both pre- and post-thaw. Additionally, we analyzed engraftment after CBT transplantation according to CBU time storage. Methods CBUs used for transplantation between2002 to 2022 were included in the study. To evaluate the influence of long-term storage of cryopreserved CBUs, Kruskal-Wallis correlation test between quality variables was performed. We studied correlation between pre-cryopreservation and post-thaw values (recovery) of TNC and CD34+ counts, CD45+ and CD34+ viability, CFU and CFU-FM counts. Additionally, myeloid engraftment was evaluated categorizing whole population in 3 groups according CBU storage duration: short-term (≤ 2 years), medium-term (> 2 and ≤ 5 years) and long-term (> 5 years). Results A total of 1577 CBUs with a median of 3.9 (range 1.3-20 years) of cryopreservation storage were evaluated. The median of TNC and CD34+ recovery, CD45+ and CD34+ recovery, CFU and CFU-GM recovery were 81.7%, 79.9%, 54.4%, 73.3%, 58.2% and 26.5%, respectively. Analysis demonstrated a non-significance decreased in CD34+ counts recovery, infused CD34 + 7AAD-viability and CFU-GM counts (P = 0.12, P = 0.28 and P = 0.96, Figure 1), respectively. Neutrophil engraftment at 42 days after CBT was found significantly faster for short-term stored CBUs (98%; CI95%, 96-99%, P = 0.015, Figure 1). No significance difference was found in platelet engraftment at 60 days between CBUs storage time groups (P = 0.5). Discussion Most quality parameters results evaluating CBUs demonstrated no significance decrease up to 20 years of cryopreservation meaning likely successful clinical transplantation of CBUs following long-term storage. Long-term cryopreserved CBUs continue to fulfil release FACT-Netcord requirements.

Résumé :

L'optimisation de la purification des cellules d'îlots est essentielle pour faire progresser les thérapies cellulaires ciblant le diabète de type 1, nécessitant l'intégration de technologies innovantes, conformes aux normes GMP, afin de renforcer l'efficacité des processus, l'automatisation et l'évolutivité. Cette thèse analyse deux technologies majeures employées dans la purification des îlots. La première étude examine l'impact d'un nouveau système de refroidissement sur le processus de purification des cellules d'îlots. Ce système permet un contrôle précis de la température durant la purification par gradient de densité grâce à un refroidissement sous pression, stabilisant efficacement la température tout en préservant la stérilité de l'environnement de salle blanche GMP. Comme solution rentable et peu invasive, ce système de refroidissement présente également des applications potentielles pour d'autres équipements de thérapie cellulaire, dont la majorité est dépourvue d'option de refroidissement. Le principal axe de cette thèse porte sur l’adaptation du système Sepax C Pro - Sefia, une technologie en circuit fermé initialement conçue pour le traitement des cellules souches hématopoïétiques, à la purification des cellules d'îlots humains. Avec le retrait progressif du système Cobe 2991 en Europe d'ici 2025 et mondialement d'ici 2031, la plateforme Sepax C Pro - Sefia constitue une alternative automatisée et conforme aux normes GMP. Elle intègre l’automatisation des étapes critiques du processus de purification des îlots, réduit la manipulation manuelle et améliore la reproductibilité des processus, en s'imposant comme une solution innovante pour les applications cliniques et de recherche en transplantation d'îlots. À la suite de ces avancées, un brevet intitulé « Systèmes et Méthodes pour le Traitement des Tissus » a été déposé pour protéger la méthode innovante développée pour le système reconverti Sepax C Pro - Sefia, assurant ainsi la sécurisation de la propriété intellectuelle et facilitant son application future en milieu clinique. Grâce à l'intégration de ces innovations technologiques, y compris une méthode brevetée pour le traitement des tissus, cette thèse propose un cadre exhaustif pour le remplacement du système Cobe 2991, garantissant la continuité de l'isolement clinique des îlots et contribuant à des thérapies plus efficaces pour les patients atteints de diabète de type 1 (allogreffes), ainsi que pour ceux souffrant de pathologies pancréatiques (autogreffes)
The optimization of islet cell purification is crucial for advancing cell-based therapies for type 1 diabetes, requiring innovative, GMP-compliant technologies to improve process efficiency, automation, and scalability. This thesis evaluates two key technologies used in islet purification. The first study evaluates the impact of a novel cooling system on the islet cell purification process. This system ensures precise temperature control during density gradient purification by providing pressurized cooling, effectively stabilizing the temperature while maintains the sterility of the GMP cleanroom environment. As a cost-effective and minimally invasive solution, this cooling system holds promises to cool another cell therapy equipment the majority of which offer no cooling option.The core focus of My thesis is the repurposing of the Sepax 2C Pro- Sefia system, a closed-system technology originally developed for hematopoietic stem cell processing, for human islet cell purification. With the Cobe 2991 system being phased out in Europe by 2025 and globally by 2031, the Sepax 2 - Sefia platform offers a fully automated, GMP-compliant alternative. It automates key steps in the islet purification process, reduces manual handling, and improves process reproducibility, making it a ground-breaking solution for both clinical and research applications in islet transplantation. Building on the findings a patent: “Systems and Methods for Tissue Processing,” was filed to protect the novel approach developed for the repurposed Sepax 2 - Sefia system ensuring the intellectual property is secured and facilitating the future application of this system in clinical settings. Through the integration of these technological advancements, including a patented method for tissue processing, this thesis provides a comprehensive framework to replace the Cobe 2991 system, ensuring the continuity of clinical islet isolation and contributing to more effective therapies for patients with type1 diabetes (allografts) but also patients with pancreatic pathologies (autografts)

Aquest treball analitza la conceptualització i representació de la sepat dels dos ceptres-uas en el context de la Geografia sagrada d'Egipte des d'una perspectiva diacrònica a partir de la documentació papirològica i epigràfica relativa a aquesta regió, des del Regne Antic fins a època grecoromana. Durant més de 2500 anys, aquest territori de l'Alt Egipte es representà als temples del país en un intent de reactualització constant de la seva hipòstasi i dels textos que l'acompanyen. Les fonts analitzades són el reflex d'una geografia simbòlica i selectiva, validada a través de mites i arquetips que, en la mentalitat egípcia antiga, contituïa ritualment la més eficaç. Un dels principals eixos del treball el constitueix l'estudi textual, iconogràfic i espacial de les representacions de la sepat i dels seus components topogràfics en cada una de les processons geogràfiques dels temples que han pogut ser constrastades in situ. La caracterització i anàlisi de l'evolució dels cultes locals en aquest territori d'orígen sethià ha esta l'altre pol de la recerca. El mètode d'anàlisi emprat ha tingut en compte les regles de sintaxi naològica i la "Gramàtica del temple" que regeixen el programa "decoratiu" de cada temple en particular.
The principal aim of this research is to characterise the conceptualisation and representation of the double was-scepter sepat and its place in Egypt's ritual topography. A diachronic approach has been adopted, based on epigraphic sources of this region dating from the Old Kingdom to the Graeco-Roman period. Through more than 2500 years this territory in Upper Egypt was depicted in temples in order to update its hypostasis and its accompanying texts. The sources articulate a sacred topography expressed in myths and archetypes considered by the ancient Egyptians as the most valid and efficacious. One of the main research axis is the study of textual, iconographical and spatial analysis of the representations of this territory and its topographical components in geographical processions laid out in temple contexts that had been verified in situ. This is complemented by the characterisation and the analysis of local cults dynamics in this sethian region. The methodology involves the study of the rules of the naological syntax and the "Grammar of the temple" that govern a temple's decoration, revealing that the representation of Egypt's sacred landscape varies from temple to temple.

El confinamiento domiciliario derivado de la expansión del virus SARS-CoV-2 supuso realizar la Formación Universitaria de forma no presencial. Aunque para cierto porcentaje de la población universitaria supuso enfrentarse a una nueva situación, en realidad la Formación Universitaria a Distancia en España tiene más de 50 años de antigüedad. La educación a distancia es una modalidad educativa que permite que las dificultades para realizar la educación presencial (distancia, incompatibilidad de horarios, falta de espacios) no condicionen el proceso de enseñanza-aprendizaje. En este sentido, los avances tecnológicos de los medios de información y comunicación son fundamentales. En la educación a distancia, además de fomentar la interactividad del alumno, los profesores deben formarse en las técnicas y medios específicos para transmitir de la forma más adecuada los conocimientos y aprendizajes conforme a las características de cada materia. El incremento de la oferta formativa universitaria online en castellano permite ubicar a las universidades en posiciones competitivas en los países hispanoparlantes al vencer las restricciones de movilidad internacional. Así también permite incrementar la internacionalización de las universidades mediante el desarrollo de proyectos formativos en colaboración con otras universidades, tanto para la conformación del profesorado del curso, como para la captación de alumnado internacional. Esta apertura de la Universidad a la formación internacional requiere que los docentes conozcan los medios y sepan utilizar las herramientas y mecanismos que tienen a su disposición para seleccionar los más adecuados para conseguir sus objetivos formativos. Diferentes instituciones educativas han publicado documentos-guía de docencia en línea, centradas principalmente en criterios docentes. Sin embargo, esta guía aporta información práctica sobre la utilización de las herramientas de docencia en línea, enfocada principalmente al personal docente. Saber cómo utilizar las herramientas disponibles aumentará la calidad de la enseñanza y el control del docente sobre el funcionamiento de las herramientas, aportándole seguridad pedagógica.

Abstract Just as in classical mechanics, the study of single particle systems in one dimen- sion provides invaluable experience in the use of quantum mechanical concepts and techniques. To extend these ideas to more realistic applications (to be able to enter the Quantum World of Part II) we also need to develop the formalisms necessary to handle multiparticle systems. In this chapter we will discuss separ- able systems (in Section 14.2), the important special case of two-body systems (Section 14.3), notation for spin-1/2 wavefunctions (Section 14.4), and lastly, in Section 14.5, the important constraints placed on quantum wavefunctions of multiparticle systems due to indistinguishability.

El presente capítulo de investigación busca realizar un análisis sobre el proceso contable que llevan a cabo los pequeños negocios de la comuna 2 del municipio de Soacha, Cundinamarca. Sin duda alguna, la gestión contable dentro de cualquier tipo de organización, sin importar su tamaño, toma una gran relevancia para quienes los administran, pues es gracias a esta que se pueden tomar decisiones adecuadas y a tiempo, así como conocer su salud financiera, poder llevar a cabo el control de su operatividad y dar cumplimiento a sus obligaciones tributarias. Ninguna organización puede inventar cualquier tipo de proceso contable, ya que este no permitirá detectar alguna pro-blemática administrativa que pueda poner en riesgo la continuidad y prevalencia de la organización. Es por lo anterior que quienes ejerzan alguna función contable deben ser personas idóneas, que sepan organizar la información contable por medio de las herramientas tecnológicas existentes.Teniendo como referente la información primaria consultada, se conoce cómo debe ser manejada la gestión contable en los pequeños y medianos negocios. Posteriormente, se aplica un instrumento encuesta a especialistas conocedores de la contabilidad para pequeñas empresas, con el fin de conocer su percepción en este tema y con ello plantear unos aspectos mínimos contables que deben tener presente las organizaciones estudiadas.

New discoveries play a major part in archaeological research, but coincidence can also have a role. When four copper-alloy scabbard-studs with Style II ornament were excavated in the smith’s grave at Tattershall Thorpe in 1981 (Fig. 2.18), they were the first of their kind to have been found in England, despite being well known on the continent, where they are dated to between 640 and 670. Within a couple of years, however, another set turned up, on a scabbard in a cemetery in Buttermarket, Ipswich, Suffolk. Then, in 1999, yet another set was found, in a grave at the new football stadium in Southampton, Hampshire (Fig. 3.1). These studs adorned scabbards that were not for double-edged swords, but for the single-edged long seax, not a very practical weapon, but one that was probably used in hunting and was therefore redolent of aristocratic practice. At Tattershall Thorpe the studs were not attached to anything, and were presumably going to be shown to a prospective patron with a view to reuse. At Ipswich and Southampton both sets were in cemeteries at what were about to become major trading-places, Gippeswic and Hamwic. These wic sites had continental counterparts and suggest new ways of organizing and systematizing exchanges of goods; others in England were London, Lundenwic, and York, Eoforwic, both former Roman towns, with the wics outside the walls but episcopal churches inside. Neither Ipswich nor Southampton had a major church, so there was no reason for important burials at either unless they were of people involved in the places’ emergence as commercial centres. One explanation is that some of the graves were for kings’ ‘reeves’ and their families, royal agents placed to oversee merchants and to ensure that tolls were paid, who were buried slightly away from where the commerce was to take place. The Southampton cemetery had other signs of an elite presence, such as a woman’s grave that contained a gold pendant with garnets and Style II animals in filigree gold wire on it (Col. pl. C.2), which seems likely to be mid- to later seventh-century.

Para popularizar a computação de maneira lúdica em feiras científicas, criamos um estande para desmistificar a computação física e, ao mesmo tempo, motivar e estimular o interesse pela computação. Foram projetados dois robôs programáveis interativos, em forma de minion (um deles com mais de um metro de altura). Com ambos, pode-se mostrar como é fácil e interessante criar programas em poucos minutos utilizando uma linguagem visual como Scratch 2 ou Snap!. O hardware consiste em uma placa microcontrolada Arduino Nano e um conjunto de sensores e atuadores. A experiência na SEPEX 2015, com mais de 4 mil visitantes, mostra que a oferta de tais estandes ao público em geral pode ser uma alternativa envolvente para popularizar a computação com um custo baixo.

Abstract Over the years, multilateral well technology has been one of the most rapidly evolving and widely utilized production technologies for new and maturing reservoirs. Multilateral wells have the potential for reservoir productivity improvement. The characteristics used to evaluate multilateral well completion are connectivity, isolation, and accessibility. All these focus on the completion design of the main bore, lateral bores, and junctions that connect the lateral and main bores. Hence, one of the factors to consider in designing multilateral wells is the junction type, which depends on the required degree of mechanical integrity and pressure integrity at each lateral. Previous studies establish that the lateral junctions are a critical element of multilateral completions and can fail under formation stresses, temperature-induced forces, and differential pressures during production. Thus, the reliability of a multilateral completion design is the ability to construct and complete the multilateral junction successfully. The Technology Advancement of Multilaterals (TAML) has categorized the distinct types of multilateral junctions based on support and hydraulic integrity provided at the junction. The objectives of this paper are: (1) to provide a detailed discussion on each classification level and the conditions in which they are applicable, (2) to present a conceptually digitized application of a multilateral well on a stacked reservoir XXXX in a Niger Delta field using SEPAL software. To achieve the latter goal, after a preliminary and detailed casing design, we applied the SEPAL software to design and digitize the proposed multilateral well schematics for the stacked reservoir. From the analysis, a multilateral level 5 junction was selected to overcome specific problems (e.g., wellbore collapse) due to the unconsolidated sands of the reservoir in the field of interest.
