Texte intégralNella prima parte del dottorato è stato effettuato lo studio e il miglioramento delle condizioni del differenziamento in vitro di cellule staminali pluripotenti presenti nel fegato, le HLSC, in senso epatico. Dopo aver valutato che la semina in condizioni statiche di tali cellule in spugne di collagene non permetteva una buona distribuzione in tutto lo scaffold e queste rimanevano per lo più indifferenziate, sono state effettuate delle colture dinamiche utilizzando un bioreattore per garantire una corretta perfusione del terreno di coltura. Inoltre sono state effettuate sia delle co-colture di HLSC con le cellule Ito del fegato sia semina in un terreno normalmente utilizzato per la coltura di epatociti e in un medium per cellule staminali, per valutare come la variazione di un terreno di coltura potesse influire sulla staminalità delle cellule. Sono state effettuate analisi morfologiche, istologiche, di proliferazione cellulare e di espressione genica delle colture. Queste hanno dimostrato come l’utilizzo di un bioreattore e di un terreno per cellule staminali possa permettere l’organizzazione delle HLSC in clusters tipici del fegato fino a strati profondi della spugna e l’aumento del differenziamento funzionale in una popolazione di epatociti maturi. Un’altra parte del dottorato è stata improntata nello studio del differenziamento in vitro di cellule staminali in Isole di Langerhans all’interno di biomateriali. Durante una prima fase di studio si sono utilizzate cellule Beta del pancreas per capire in quale forma di scaffold a base dell’estere benzilico dell’acido ialuronico potessero crescere meglio. La situazione migliore riguardava l’uso di spugne. Avendo però le cellule una scarsa capacità proliferativa, si è deciso di utilizzare come alternativa le Cellule Staminali Mesenchimali (MSC) e sono state effettuate prove di co-colture con cellule Beta utilizzando terreni diversi per valutare lo stato differenziativo delle MSC. Tali prove sono ancora in corso.

Texte intégralIt is known that melanoma develops as a consequence of multifactorial alterations. To date several studies indicate the effective implication of p16 as a tumor suppressor gene with a major role in either the development or progression of human melanoma. Deregulation of melanoma cell growth has been widely associated with mutations in the p16-cyclin D/cdk4-pRb pathway. Recently anticancer therapies are focused on restoration of p16 CDK inhibitory function and other proteins unregulated in melanoma cell cycle pathway (e.g., c-myc, p27). A combined strategy for restoration of normal homeostasis in the melanoma skin with targeted delivery of apoptosis-inducing agents does not seems to be far obtain. New class of antitumoral agents are emerging: histone deacetylase (HDAC) inhibitors have attracted much interest because of their ability to arrest cell growth, induce cell differentiation, and in some cases, induce apoptosis of cancer cells. Recently, attention has been focused on the ability of HDAC inhibitors to induce perturbation in cell cycle regulatory protein (e.g., p21CIP1) and down-regulation of survival signalling pathway. In the present study, we have examined the effect of valproic acid (VPA) on M14 and JR8 human melanoma cell lines. Here we observed that VPA induces cell cycle arrest and apoptosis sensitising melanoma cells to cis-Platin and etoposide treatment. VPA was able to induce G1 arrest (up to 75 %) in association with up-regulation of p16, p21 and cyclin-D, inducing apoptosis (50 %) in M14 and JR8 (55%) cells. To evaluate the possibility of VPA to be used as co-adjuvant in the chemotherapy protocol for melanoma, two different sequences of in vivo administration of VPA and cis-Platin were designed. The concomitant treatment of tumour (M14) in vivo, with VPA+cis-Platin induced a growth inhibition of 48% compared to control with an anti- metastatic effect of about 45%. The treatment sequence with VPAcis-Platin combination inhibited tumour growth in M14 of about 61% and induced an anti-metastatic effect of about 70%. Moreover p16 gene expression was found up-regulated 2,5-fold in tumor treated with the sequence VPAcis-Platin compared to control and cis-Platin single treated tumor. The ability of valproic acid to re-established the G1 pathway in melanoma both in vitro and in vivo suggests a potential application of VPA in melanoma therapeutic protocols.

Texte intégralIn questo lavoro sono stati ricostruiti in vitro 'soft and hard tissue' seminando Cellule Staminali Adulte su supporti tridimensionali in presenza di opportuni fattori differenziativi. In particolare, sono stati preparati i seguenti costrutti: (a) tessuto nervoso a partire da cellule staminali di pelle (SKP) e tessuto adiposo (ADSC) seminate su un supporto a base di acido ialuronico o fibrina; (b) tessuto osseo vascolarizzato utilizzando ADSC seminate su scaffold a base di idrossiapatite e (c) tessuto simil-dentale associando le cellule staminali della polpa dentale (DPSC) a un biomateriale a base di acido ialuronico. Lo studio ha previsto una fase preliminare in monostrato, in cui il differenziamento delle cellule staminali è stato valutato mediante analisi di immunofluorescenza. Stabilita la plasticità delle cellule staminali ai fattori differenziativi, sono stati effettuati gli esperimenti nelle tre dimensioni. A diversi tempi, i costrutti sono stati sottoposti a test di proliferazione, ad analisi morfologica mediante microscopia elettronica a scansione e ad analisi dell'espressione genica mediante Real Time PCR. Inoltre sono state condotte analisi di citogenetica mediante cariotipo e CGH array per verificare la stabilità genetica delle cellule durante il differenziamento. I dati raccolti hanno mostrato: (a) il differenziamento neuronale e gliale delle SKP e delle ADSC su supporti a base di acido ialuronico e fibrina mediante un processo che non altera lâassetto cromosomico delle cellule differenziate. (b) Il co-differenziamento osteogenico ed endoteliale delle ADSC su supporti a base di idrossiapatite e la generazione in vitro di un sostituto osseo bioingegnerizzato privo di alterazioni geniche e dalla promettente osteointegrazione. (c) Un possibile modello per la rigenerazione della polpa dentale che associa le DPSC a un supporto tridimensionale a base di acido ialuronico sottoforma di tessuto non tessuto, dove il co-differenziamento gliale, neuronale ed endoteliale delle DPSC favorisce la rigenerazione della componente nervosa e vascolare del tessuto pulpare.
