This article consists in establishing a geotechnical stability study of the -4m/zh quay belonging to the Ksar Sghir Port. First of all, using the TALREN software for verification against large slips. the study presents a verification of the stability for the rear slope slip circles and for the front slope slip circles in the static case. Secondly, stability was determined for the back slope slip circles and for the front slope slip circles in the seismic case. Moreover, PLAXIS software was used to model the displacements and constraints in the structure. Following the results of the safety coefficients, we notice that the stability of the structure gives acceptable values. In addition, the soil does not reach breaking stress. The total mean stress and total displacement mostly impacted the surface area that is related to external impact.

The Moroccan road network is susceptible to multiple landslides annually, particularly in the northern regions due to high rainfall and specific geology. These events result in significant economic and social negative consequences, highlighting the need for sustainable and cost-effective solutions for network maintenance. This study outlines the methodology employed in addressing the issues within the RR410 regional road (Rifain region of Morocco), which entailed a thorough examination of the malfunctions, specific surveys, laboratory testing, and problem modeling. By incorporating long-term test-derived shear strength parameters, the model indicated that the road platform was stable, and back analysis using TALREN 4 software allows for model calibration. At kilometric point 23, using earthwork-based solutions (e.g., purging and replacing the base layer, employing granular water-insensitive substitution material) was found to provide a sustainable alternative to the expensive reinforced concrete-based solutions commonly used. Furthermore, these solutions contributed to the use of environmentally friendly and locally sourced materials. Road alignment rectification to anchor the platform in suitable soil was also an effective solution, as demonstrated at kilometric point 48. Additionally, enhancing the drainage and sanitation infrastructure, such as installing draining trenches, spurs, and reinforcing existing water structures, is a crucial aspect of addressing most landslides in the region.

Transcription activator-like effector (TALE) nucleases (TALENs) mediated gene editing methods are becoming popular and have revealed the staggering complexity of genome control during development. Here, we present a simple and efficient gene knockout using TALENs in kawakawa, Euthynnus affinis, using slc24a5. We examined slc24a5 gene expression and functional differences between two TALENs that hold the TALE scaffolds, +153/+47 and +136/+63 and target slc24a5. Developmental changes in slc24a5 transcripts were seen in early-stage embryos by real-time PCR; slc24a5 expression was first detected 48 h post fertilization (hpf), which increased dramatically at 72 hpf. Four TALENs, 47- and 63-type of two different target loci (A and B), respectively, were constructed using Platinum TALEN and evaluated in vitro by a single-strand annealing (SSA) assay. TALEN activities were further evaluated in vivo by injecting TALEN mRNAs in the two-cell stage of the zygote. Most of the TALEN-induced mutants showed mosaic patterns in the retinal pigment epithelium (RPE) and fewer melanin pigments on the body at 72 hpf and later when compared to the control, implying the gene’s association with melanin pigment formation. A heteroduplex mobility assay (HMA) and the genome sequence further confirmed the TALEN-induced mutations of substitution, insertion, and deletion at an endogenous locus.

Abstract Abstract 2403 Background: Pediatric Burkitt Lymphoma (BL) is the most common histological subtype of Non Hodgkin Lymphoma (NHL) in children and adolescents (Cairo et al., Blood, 2007; Miles/Cairo, BJHaem., 2012). We previously identified in a subset analysis that children with BL and a 13q deletion, particularly 13q14.3, had significantly poorer outcome and inferior overall survival (OS) despite aggressive short, intensive multiagent chemotherapy (Poirel/Cairo et al., Leukemia, 2009; Nelson/Cairo/Sanger et al., BJHaem., 2009). Deleted in Lymphocytic Leukemia 1 (DLEU1) is a long non codign RNA gene with the BL classifier genes as reported by Dave et al (NEJM. 2006) on chromosome 13q14.3 region and the expression levels of RASSF1, TUBB2C and UBR1 were significantly higher in BL than in diffuse large B-cell lymphoma (DLBCL) (Day/Cairo et al., AACR, 2008). Sequence-specific Zinc Finger Nucleases (ZFN) and Transcription Activator-Like Effector Nucleases (TALENs) technologies have been developed for precision targeted genome editing in in vitro experiments with high efficiency and TALEN technology has been applied in a variety of organisms and stem cells for new experimental and therapeutic tools (Carlson et al., Mol Ther., 2012). Objectives: We hypothesize that 1) TALEN technology is suitable for the modification of and silencing of DLEU1 locus and 2) DLEU1 may have function as a tumor suppressor gene and therefore examined whether the loss of DLEU1 by TALEN-mediated DLEU1 knockdown Raji BL cells result in change in proliferation and programmed cell death. Methods: TALENs were constructed based on REAL (restriction enzyme and ligation) assembly methods (Sander et al., Nat. Biotech, 2011) for DLEU1 gene modification. DLEU1 TALEN pairs or combinations were transfected into 293T and Raji cells using lipofectamin reagent (Invitrogen) and Amaxa nucleofection kit, respectively. Genomic DNA and total RNA were extracted using genomic DNA extraction kit (Promega) and Trizol reagent (Invitrogen). Surveyor mutation detection assay (Transgenomic) was applied for TALENs validities and confirmed by sequencing analysis. Quantitative RT-PCR was performed by CFX96 Real-time system (Bio-rad) using qScript™ cDNA Synthesis Kit (Quantas) and SsoFast™ EvaGreen® Supermix (Bio-rad). Transfected cells were re-plated (1×104) into 48 well plates at 24hours post transfection and then counted every 24hrs for cell proliferation assay. For spontaneous apoptosis assay, DLEU1 TALENs transfected Raji cells were re-plated into 96 well plates at 24hours post-transfection and measured caspase 3/7 activities (Promega) using Clarity Luminescence microplate reader (Biotek) and statistical significance was determined by a one or two-tailed Student t-test. Results: Three pairs of functional DLEU1 TALENs (T1, T2, and T3) targeting endogenous DLEU1 gene generated and confirmed their validities with gene modification caused by non-homologous end joining (NHEJ) in TALENs mediated 293T [Figure 1]. Forty eight hours after transiently transfection in Raji cells, combined DLEU1 TALENs, T1/T2 (T12), T1/T3 (T13) and T2/T3 (T23) mediated Raji cells were dramatically reduced expression of DLEU1 mRNA (about 82–93% reduction) compared to untransfected Raji cells [92% reduction (p<0.002), 93% (p<0.0006), and 92% (p<0.0003)], respectively, and compared to empty vector as a control [82% reduction (p<0.05), 87% (p<0.0001), and 82% (p<0.001, respectively)]. T12 and T13-mediated DLEU1 knockdown Raji cells showed significantly an increase of cell proliferation [1.2-fold (p<0.02) and 1.5-fold (p<0.04) at day 5, respectively]. In spontaneous apoptotic assays, T13 mediated Raji showed significantly 20% (p<0.05) of reduction of caspase 3/7 activities. In comparison of mRNA expression of DLEU1 network genes in T13 mediated Raji cells, there were no significant differences in mRNA expression of c-myc compared to control, however, there was significantly reduced expression of RASSF1 and UBR1 mRNA (<7.7-fold, p<0.005 and <11.1-fold, p<0.0005), respectively, and significantly increased expression of TUBB2C mRNA (>7.5-fold, p<0.005). Conclusions: We demonstrate that 1) DLEU1-TALENs are useful tools for the modification of endogenous DLEU1 gene expression, 2) DLEU1 may have as a tumor suppressor gene in BL, and 3) DLEU1 TALEN mediated gene knockdown resulted in inhibition of BL apoptosis and increase in cell proliferation. Transcription activator-like effector (TALE) nucleases (TALENs) efficiently recognize and cleave DNA in a sequence-dependent manner. However, current TALE custom synthesis methods are either complicated or expensive. Here we report a simple and low-cost method for TALE construct assembly. This method utilizes the denaturation/reannealing nature of double-stranded DNA to create a unique single-stranded DNA overhang for proper ordering of TALE monomers in an engineered multimer. We successfully synthesized two TALEN pairs targeting the endogenous TET1 locus in human embryonic kidney cells and demonstrated their editing efficiency. Our method provides an alternative simple, low-cost method for effective TALEN assembly, which may improve the application of TALE-based technology.

Abstract Diamond-Blackfan Anemia (DBA) is a congenital bone marrow failure syndrome that manifests as a profound macrocytic anemia and classically presents within the first year of life. Heterozygous mutations in, or genomic loss of one of several Ribosomal Protein (RP) genes have been identified in over 50% of DBA patients, most commonly RPS19, accounting for 25% of all cases. DBA shares a similar erythroid phenotype to the 5q- subtype of myelodysplastic syndrome in which anemia is thought to arise from heterozygous loss of RPS14. Anemia in these conditions is at least partially due to p53-mediated apoptosis and cell cycle arrest of erythroid progenitors. To further study the role of p53 in the pathogenesis of DBA and 5q- syndrome, we employed genome editing tools to generate stable Rps14 and Rps19 knockout zebrafish lines. We generated Transcription Activator-Like Effector Nucleases (TALENs) targeting exon 1 of rps19 and exon 1 of rps14 as well as Clustered, Regularly Interspaced, Short Palindromic Repeats (CRISPR) single guide RNAs (sgRNA) targeting exon 2 of rps19. TALENs or CRISPRs were injected into p53m214k/m214k zebrafish embryos at the single-cell stage. This zebrafish line carries a mutated p53 that is insensitive to DNA damage and hence prone to tumor formation. rps19 CRISPR sgRNAs were injected with mRNAs encoding Cas9, Cas9D10A nickase, and a ssDNA guide with a human DBA mutation. For each cohort of embryos injected, genomic DNA analysis from 20 phenotypically normal embryos from each clutch was screened to determine the efficacy of cleavage by TALEN and CRISPR using MiSeq. Mutations were identified in 30% (rps14 TALEN) 29% (rps19 TALEN), 27% (rps19 Crispr Cas9) and 12% (rps19 Crispr Cas9D10A) of reads. None of the rps19Crispr Cas9D10A carried the ssDNA guide mutation, rather single nucleotide variants and indels similar to those observed with Cas9. The remaining embryos from each F0 clutch were raised in order to generate stable mutant lines in the F1 generation; however, early, overt tumor growth was noted in all RP injected lines. Tumors were observed from 4 months post fertilization compared with 9 months for uninjected controls. F0 RP mosaic fish continued to develop tumors earlier than uninjected counterparts. At 10 months of age tumor development was statistically significantly higher in rps19 and rps14 TALEN and rps19 Cas9D10A and trended towards significance in rps19 Cas9 injected fish. Overall survival was significantly reduced in each of the cohorts compared to p53m214k/m214k uninjected controls (p<0.0001). Preliminary histology of grown tumors has shown melanomas and malignant peripheral nerve sheath tumors. Zebrafish injected with an unrelated TALEN targeting a zinc transporter (SLC30A10), into p53m214k mutant embryos do not show any increase in tumor formation compared to uninjected controls. Notably several RP’s have been shown to be haploinsufficient tumor suppressor genes in their own right in zebrafish and drosophila models. To determine if the early tumor development in p53m214k zebrafish was simply additive to a potential tumor suppressor effect of Rps14 or Rps19 alone, we injected WT embryos with the rps14 and rps19 TALENS. High mortality in rps14 and rps19 TALEN injected WT embryos impeded this analysis; however recent published reports on stable Rps19 mutant zebrafish do not report an increase in tumor incidence. Interestingly, embryo survival was not affected when TALENs were injected into p53m214/+. Analysis of these zebrafish is ongoing. Our results show that loss of Rps14 or Rps19 accelerates the development of tumors in the p53m214k/m214k mutant line. This effect is independent of the RP or the method of mutation (TALEN vs CRISPR), indicating that off target effects are unlikely to be responsible for this observation. As these are mosaic F0 fish, it is possible that tumors may arise from cells with homozygous RP mutations. Further molecular analysis will reveal this. We have now identified 2 stable Rps19 mutant lines, and tumor analysis of F1 fish from these lines is ongoing. In conclusion, we have shown that loss of Rps14 or Rps19 cooperates with a loss of function p53 mutation to accelerate tumor formation and death. Our results highlight the importance of caution in using p53 suppressors as a therapeutic option in RP deficient patients. Disclosures No relevant conflicts of interest to declare.

Abstract Transcription activator-like effector nucleases (TALEN) are engineered proteins used for precise genome editing by generating specific DNA double strand that are repaired by homologous recombination and by non-homologous end joining. TALENs can be used to study gene regulation by deleting putative regulatory elements in the context of the native chromosome and measuring mRNA synthesis. We designed TALENs to delete individual DNAse I-hypersensitive sites (HS) of the β-globin locus control region (LCR) followed by an assessment of globin gene expression and assessment of epigenetic effects in K562 erythroleukemia cells. The β-globin LCR is composed of five HSs and functions as a powerful regulatory element responsible for appropriate levels of the five β-like globin genes during development. Introduction of plasmid DNA encoding a pair of TALENs and targeting individually the flanking region of the HS2, HS3 and HS4 core elements along with a donor 100 base single-stranded oligonucleotide resulted in the successful deletions of each of the three core elements in K562 cells. Individual K562 cells were seeded to produce clones and the mutations were screened by PCR to identify both heterozygous and homozygous clones. The TALEN-mediated 288 bp HS2 core deletion resulted 32 heterozygous (48.5%) and 6 homozygous clones (9.1%) in a total of 66 clones screened. K562 carries three copies of chromosome 11 emphasizing the robustness of TALEN technology to target each of the alleles. In the 199 bp HS3 core deletion, from 113 clones we identified 28 heterozygous (24.8%) and 3 (2.7%) homozygous clones. Lastly, the 301 bp HS4 core deletion yielded 9 homozygous (5.9%) and 12 heterozygous (7.9%) clones from 151 clones screened. Total RNA was isolated from wild-type K562 cells, and from both the heterozygous and homozygous mutant clones and subjected to RNase Protection analysis to quantitate the levels of globin mRNA. Deletion if the HS3 core in K562 cells in a ∼30% reduction in ε-globin mRNA and 2-fold reduction in γ-globin mRNA. A more dramatic effect on globin expression is observed in the HS2 core deletion, as ε- and γ-globin expression is reduced by 2- and 5-fold, respectively. These results suggest that HS2 contributes the majority of the LCR enhancer function in K562 cells. The HS4 core deletion resulted in a modest ∼20% reduction in both ε- and γ-globin expression. TALENs were designed to knockout trans-acting factors implicated to be involved in globin gene regulation and/or globin switching. TALENs bracketing the gene promoters and the first exon of 25 genes encoding either a transcription factor or histone-modifying enzyme were synthesized and post-transfection PCR screens of the transfected pool of K562 cells resulted in the successful identification of 17 gene knockouts. The 17 target genes are PRMT5, LDB1, EIF2AK3, BCL11A, HBSIL, MYB, SOX6, NFE4, NR2F2, NR2C1, NR2C2, CHTOP, NFE2, DNMT3A, RBBP4, MTA2 and MBD2. Single cell clones have been generated by limited dilution of transfected K562 pools and thus far we have identified heterozygous and homozygous clones of 8 of 17 gene knockouts, importantly all clones were identified without selection. The frequency of identifying the knockout clones, represented by the number of clones screened/ number of heterozygous clones/ number of homozygous clones, are as follows: HBS1L (63/3/0), SOX6 (68/13/2), NFE4 (56/13/7), LBD1 (300/2/0), MBD2 (301/0/1), CHTOP (288/66/6), NFE2 (712/44/5) and NR2C1 (96/40/11). The remaining nine gene knockouts and globin gene expression data will be presented at the meetings. These studies highlight a powerful TALEN-mutagenesis platform for target deletions of both cis- and trans-elements to study globin gene switching. TALENs can be synthesized in several days and the screening of the individual clones for the desired knockouts is completed within two weeks. This highly efficient mutagenesis platform will further our understanding of the molecular basis of globin switching. Disclosures: No relevant conflicts of interest to declare.

Cytomegalovirus (CMV) infections are still a global health problem, because the latent viruses persist in humans and cause recurring diseases. Currently, there are no therapies for CMV latent infections and the therapies for active infections are limited by side effects and other problems. It is impossible to eradicate latent viruses in animals. HCMV (human CMV) is specific to human diseases; however, it is difficult to study HCMV due to its host specificity and long life cycle. Fortunately, MCMV (murine CMV) provides an excellent animal model. Here, three specific pairs of transcription activator-like effector nuclease (TALEN) plasmids (MCMV1–2, 3–4, and 5–6) were constructed to target the MCMV M80/80.5 sequence in order to test their efficacy in blocking MCMV lytic replication in NIH3T3 cell culture. The preliminary data showed that TALEN plasmids demonstrate specific targeting and cleavage in the MCMV M80/80.5 sequence and effectively inhibit MCMV growth in cell culture when the plasmid transfection is prior to the viral infection. The most specific pairs of TALEN plasmids (MCMV3–4) were further used to confirm the negative regulation of latent MCMV replication and gene expression in Balb/c mice. The injection of specific TALEN plasmids caused significant inhibition in the copy number level of immediately early gene (ie-1) DNA in five organs of mice, when compared with the controls. The result demonstrated that TALENs potentially provide an effective strategy to remove latent MCMV in animals.

This is an empirical study of how the mathematical talk of adult learners constructs/reconstructs different mathematical discourses. The study is to be regarded as an attempt to develop a discursive approach within the field of mathematics education and to complicate the status of mathematics in education and in society in general. My theoretical underpinnings consist of three possible mathematical discourses – coercive, regulative and emancipative mathematics. From a discursive psychology perspective, I let these discourses function as analytical interpretive repertoires in relation to the adult learners’ rhetorical use of mathematics and their claiming of mathematical subject-positions, named the coerced, the self-regulating and the responsible mathematician. The conclusions are concentrated around a discussion about the rhetorical turn of adult mathematics in which I focus on mathematics and its relation to the becoming of the self-regulating subject. I continue by arguing that a focus on how the participants deal with mathematics, rather than how they learn mathematics, can generate new perspectives on the teaching praxis of mathematics education.

Talin is a multifunctional cytoskeletal protein that plays a critical role in linking the actin cytoskeleton to the integrin family of transmembrane cell adhesion receptors. Two distinct integrin binding sites have been identified in talin, one present in the globular head domain (IBS1) and involved in integrin activation, and a second (IBS2), that has been delineated to a 130 residue fragment of the talin rod domain, but whose functional role is still elusive (Tremuth et al.2004). The objective of the present study was to define the minimal structure of talin IBS2 and to investigate its functional role in the integrin-cytoskeleton connection.

In the first part of this study, we used a combination of three different experimental approaches to define the minimal structure of talin IBS2: 1) an in silico bioinformatics approach to analyse sequence conservation of talin IBS2, 2) an in vivo cell biology approach to study the subcellular localization of recombinant talin fragments covering IBS2 in CHOáIIbâ3 cells, and 3) an in vitro biochemical approach consisting in protein overlay, pull down and Surface Plasmon Resonance (SPR) assays, to study the direct interaction between talin IBS2 and the integrin â3 subunit. We delineated IBS2 to a single amphipathic á-helical repeat of 23 residues within the talin rod domain. We further provided evidence that a two amino acid mutation(L2094I2095/AA) was sufficient to inactivate the IBS2 site, due to a disruption of the á helix structure, as demonstrated by infrared spectroscopy. In addition, we identified 2 lysine residues (K2085, K2089) exposed on the solvent face of á helix 50, which are directly involved in the talin IBS2-integrin interaction.

In the second part of this study, we investigated the functional role of talin IBS2 in spreading defective talin (-/-) cells and showed that in contrast to full-length wild type talin, an IBS2 LI/AA mutant talin was unable to fully rescue the spread phenotype of these cells. These results provide the first direct evidence that IBS2 in the talin rod is essential to link integrins to the actin cytoskeleton.

Syftet med denna studie är att kontrastera en utvald organisations strategier för att attrahera, behålla, utveckla och avveckla de viktigaste resurserna mot Talent Management och dess komponenter. Studien ämnar således mot att göra en kontrastering mellan traditionellt kompetensförsörjningsarbete mot det mer moderna konceptet Talent Management. Författarna har valt att undersöka detta genom att samla empiri från intervjuer med sex medarbetare kombinerat med analys av interna dokument. Resultatet visar att delar av myndighetens arbete med kompetensförsörjning kan likställas med Talent Management men att vissa komponenter är uteblivna. Baserat på resultatet framhålls i resultatdiskussionen att myndigheten möter stora utmaningar gällande enhetligt arbete med kompetensutveckling, kompetensförsörjning och ledarskap på grund utav det delegerade ansvaret från central nivå. I slutskedet av avsnittet för resultatdiskussion redovisas även en sammanfattande slutsats som grundar sig i ett framgångsrikt arbete inom blocken attrahera och avsluta samtidigt som arbete inom blocket behålla och utveckla varierar inom organisationen, dels på grund av delegerat ansvar. Slutligen presenteras förbättringsförslag inom organisationen och vidare forskning inom ämnet.
The purpose of this bachelor level thesis is to compare a Swedish public authority’s practical work with competence supply towards the components of Talent Management. The study is aiming to contrast traditional competence supply operations with the more modern concept of Talent Management. The writers have chosen to collect empirical data through interviews with six employees combined with analysis of internal documents. The result shows that parts of the public authority’s work with competence supply have similarities to Talent Management, but some of the components are left out. Based on the results, the discussions shows that this public authority is struggling with united approaches towards competence development, competence supply and leadership because of delegated responsibilities from a central level. In the end the writers are presenting an overall conclusion that is based on the organizations successful work within the blocks of attract and terminate while the work within the block of retain and develop varies within the organization, partly because of delegated responsibility. Finally suggestions for improvement within the organization and further research are presented.

During cell migration, adhesion complexes control the production of force and the adaptation to the mechanical properties of the environment. The aim of this project was to understand the molecular mechanisms by which these complexes control the force produced by actin assembly and encode mechanical information into biochemical reactions. The first study shows that the proteins talin, RIAM and VASP assemble on the surface of a membrane to stimulate actin assembly by a novel. mechanism The second part is based on the reconstitution of the mechanosensitive machinery of the adhesion complexes with pure proteins on a micropatterned surface observed in microscopy. The study reveals that the stretchable protein talin exchanges its partner RIAM for vinculin in response to the force transmitted by the actin cytoskeleton. Talent management has been a heating-up topic in recent years. It has a positive effect on the ability to create a compelling, productive, and valued enterprise for all stakeholders — employees, customers, business partners and investors. Moreover, talent management facilitated talent flexibility and enables the rapid growth of the business, ensuring rapid alignment with the requirements established by business leaders as the company evolved. Meanwhile, the importance for a company to attract and retain a high-quality workforce is moving into the boardroom agenda, more and more attention is pain on this acute topic.

In this thesis, we are doing the research on how to build up a talent factory within the company – how to recruit the most outstanding people to meet the business needs, how to maximize the potential of employees, how to put the right people in the right position and finally how to keep the best people in the company. In order to achieve it, we conducted our research from different methodologies (literatures, case studies, interviews) to find the answer our research question: is talent management a science or an art in order to build up a talent factory? At the end, we came out of our own understanding about the talent management and the accommodations of building up a talent factory.

Dessa böcker och artiklar har gemensamt att de betonar vikten av att företag adopterar konceptet och de ödesdigra följderna om de låter bli. Talent Management är enligt många ett utav det största och senaste begreppen inom personaladministration.

Under personaladministrationens historia har emellertid en mängd olika begrepp kommit och gått ur tiden såsom Personnel Management, Human Resource Management och Strategisk Human Resource Management och många forskare har hävdat att dessa begrepp inte skiljer sig nämnvärt åt, utan snarare kan karaktäriseras som ett kontinuerligt strävande efter legitimitet och status av personalansvariga. Det huvudsakliga temat i detta strävande har varit att ett företags personal utgör en viktig och betydande del av organisationen och därigenom kan utgöra skillnaden mellan framgångsrika och icke framgångsrika företag.

Syftet med den här uppsatsen var att ta reda på de bakomliggande faktorerna och motiven till varför företag implementerar Talent Management, samt att undersöka i vilken utsträckning konceptet kan sägas karaktäriseras av ny och värdefull kunskap.

Genom att jämföra teorier om HRM och personalutveckling med normativ litteratur och intervjuer kring Talent Management har författarna kommit fram till att Talent Management inte kan sägas karaktäriseras av ny och värdefull kunskap, utan snarare som ett försök att paketera om gamla idéer och tekniker under en ny etikett. Förfat-tarna till den här uppsatsen tror att konceptet kan sägas känneteckna ännu ett försök av personalansvariga att stärka sin legitimitet och status i sina respektive organisationer.


The authors of this thesis have found that, during the last years, the world has witnessed a dramatic explosion of articles and books about the concept Talent Management. These books and articles, all emphasise the urgency for companies to adopt the concept and the devastating consequences if they don’t. The concept is by many re-searchers seen to be the top issue and, the latest trend within Human Resource Management.

Nevertheless, throughout the history of the personnel profession the world has witnessed several different concepts such as Personnel Management, Human Resource Management, and Strategic Human Resource Management and several researches have claimed that these concepts describes the same thing. Some researchers have argued that the different concept instead represent a continuous rhetoric struggle by HR professionals to enhance their legitimacy and status by becoming more business oriented and demonstrate that employees indeed can make a difference in distinguish-ing successful organizations from others.

The purpose of this thesis was to investigate the underlying reasoning and logic to why companies adopt talent management and explore what the concept represents in terms of new knowledge.

By comparing traditional theories of HRM and HR planning with normative literature and interviews on Talent Management the authors have found that the concept does not represent any new and distinctive knowledge, but rather can be considered as an effort to repackage old ideas and techniques with a new label. The authors of this thesis believe that Talent Management is another illustration of the struggle by HR professionals to enhance their legitimacy and status in their organization.

El trabajo que a continuación se presenta tiene por objeto de estudio el Taller de Artes Visuales (TAV), taller de grabado y gráfica, fundado en Santiago de Chile en 1974. El propósito elemental de la investigación es reconstruir la historia del TAV, que surge en la escena artística local ligado a un contexto de suyo complejo para el país y que, no obstante aquello, pervive hasta el día de hoy. Es decir, la presente tesis ofrece al lector un estudio monográfico sobre el TAV y su devenir a través de más de tres décadas de existencia. Escribir la historia de un taller especializado en las técnicas de la gráfica –o de cualquier disciplina artística- supondría el rastreo y la catalogación de las referencias al respecto registradas en las fuentes fundamentales para la historia del arte local, es decir, estudios sobre arte chileno, catálogos y crítica especializada; como también de los.documentos oficiales de los principales centros de enseñanza artística del país, esto es, universidades, institutos, academias. Mas, este no es el caso. La presencia del TAV en el campo artístico nacional se caracteriza por su limitada referencia en las historias del arte, por su casi nula existencia en la crítica artística, y su marginalidad respecto de centros de formación institucionalizada. Por consiguiente, esta pesquisa acerca del Taller de Artes Visuales tiene una condición capital, a saber: la carencia parcial de fuentes escritas, en relación a los principales discursos historiográficos referidos al arte en Chile.

This thesis employs discourse analysis in order to understand how the concepts of “talent” and “talent management” are socially constructed within an organisational setting and the ways in which these are then brought into being and enacted. The research highlights that the meanings attributed to these often “taken for granted” concepts is not commonplace or self-explanatory, but rather that they come about via ongoing processes of negotiation within organisational settings. The thesis is a case study of talent management at a single Australian Professional Services Firm - PSF - and its multiple business units. It is one of very few empirical studies that examines talent management in an organisation and provides important empirical insights into the how, what and why of talent management which are notably absent in the literature. A discourse analytic framework which distinguished between discursive concepts, objects and subjects is employed to examine the talk about talent in the case study organisation. This approach enables the thesis to contribute to our understanding of talent management in several ways. While previous publications on talent management have offered prescriptive and normative assumptions about how organisations should or could define talent and conduct talent management, the thesis examines the discursive processes associated with attributing meanings to concepts and highlights how talent concepts adopted within organisations are themselves social constructions with multiple meanings potentially attributed to the same term. The thesis challenges the assumption that stakeholders in an organisation will maintain the same definition of talent, and instead argues that the meaning of talent can be debated and attributed a variety of meanings, not only between organisations, but also within organisations. The thesis also challenges the existing conceptual frameworks which imply that categories of talent are mutually exclusive. It illustrates that executives, business units and organisations such as PSF can simultaneously draw upon multiple conceptualisations to attribute multiple meanings to talent. The thesis makes an original contribution to the understanding of talent management by indicating that talent management concepts, objects and subjects are interconnected. This is because executives and business units within PSF were required to operationalise and bring into reality their idea of talent (concept) through material arrangements indicative of talent management (objects) which are then enacted on employees (subjects). It argues that “talent” is a not separate, incidental or intermittent aspect of talent management, but rather an integral facet of the associated practices and shows that the meaning of talent arises from ongoing processes of negotiation, with these meanings affecting the practice of talent management and the individuals who are the subject of these practices.

The article is devoted to the study of the modern North Russian fairy tale tradition. My colleagues and I have many years of field experience in the villages of the Mezen River basin led to the need to find justification for other new forms of fixation and publication of fairy tales. We understand by a fairy tale, first of all, a speech work that occupies a special position in the situation of communication between the storyteller and his listeners, which turn out to be, including folklorists. Interviews in which our villagers mention fairy tales can be divided into two types: interviews-memories of how they themselves listened to fairy tales (most often when they were children); interviews in which the interlocutors explain how they themselves told/tell fairy tales to their children or grandchildren; finally, interviews, which are accompanied by the performance of a fairy tale to us, folklorists. For a very long time, the first two types of interviews were not the focus of research interest. Folklorists were interested in fairy tales addressed to themselves or told in their presence. Refs 16.

The present work aims to address the literary genre of fairy tales on the basis of their psychologicaland educational value aimed at satisfying the psychological-emotional and cognitive-ethical aspects. The objectives will be to present the conscious and unconscious aspects of the characters and to reveal the creative abilities that fairy tales contain: what are the main characteristics and their psychological value. The fairy tale represents an important point both in the field of imagination and creativity and you know in the didactic one. It is important to give the younger generations the opportunity to develop creative thinking, to test themselves with different types of intelligence, to get dirty with the experience of the world in which one lives, to get excited, to be empathetic. The method used will be the psychological analysis of the characters; in particular we will focus on the reading and analysis of the fairy tale "The Little Prince" by Antoine de Saint- Exupery, a sublime tale, full of meanings, which through its images has fascinated entire generations and which aims to transmit the true meaning of life: contact with the other. The psychodynamic aspects of the fairy tale will be externalized through the possibility of knowing oneself thus allowing a constant development of the recipient's creativity and imagination. In conclusion, fairy tales represent the key for the child to enter the world of fantasy, creativity and objective reality, without getting confused with his affective-emotional balance and with his future possibilities of socialization and logical understanding.

: This paper is aimed at analysing the functions of names in Romanian and foreign fairy tales by highlighting similarities and differences of names in various cultures (English, French, German and Romanian). In comparison with names in real life, proper names in literature feature more functions than that of identification, as such names are varied from a semantic and structural perspective. The relationship between a name and its bearer is motivated; thus, literary names are generally more innovative and their structure is much more complicated than in the case of names in real life. One finds it impossible to talk about a set of stable functions of names in fairy-tale discourse, since it is impossible for one to separate their hierarchy from the text in which they appear. The functions of proper names in fairy tales are inferred by means of analogy based on the more general functions of verbal communication defined by Jakobson (1964: 88–94). In the framework of fairy-tale discourse, the functions are not independent, but actually depend on one another, as names in fairy tales are polyfunctional in themselves. Several functions occur: dominant (referential), as well as subjacent or latent (allusive, camouflage, localisation, didactic-educational, expressive, semantic and sociological) functions. The research framework of this paper is onomastics, but the approach is transdisciplinary. The methods belong to pragmatics, semantics and semiotics. The corpus was compiled from anthologies of folktales and modern fairy tales.

The important advantages of fairy tales as linguodidactic material are authenticity, informative richness, concentration of linguistic means, and emotional impact on the addressee. There are several stages of work on the text of a fairy tale: before text, text, after text. At each of them, it is necessary to use various tasks that contribute to the expansion of the vocabulary of students, increase the level of proficiency in a foreign language, and contribute to the solution of linguistic and cultural problems.

Este trabajo parte del testimonio de mi experiencia didáctica de un taller de artes plásticas en el Centro cultural de España en Malabo, Guinea Ecuatorial, realizado entre los meses febrero y abril de 2019. Se trata de un proyecto personal de enseñanza de técnicas gráficas en torno al retrato como hilo conductor y la maestría de conceptos y técnicas básicas para formar capacidades plásticas y expresivas de los alumnos. La especificidad del país donde transcurre el taller consiste en su situación sociopolítica y vinculación reciente a España, en la ausencia de enseñanzas artísticas regladas como tales, y la extrema dificultad al acceso a los materiales y a fuentes pictóricas y bibliográficas. Política- y socialmente nos encontramos pues en un contexto muy diferente al europeo, y el objetivo de este paper será hacer una reflexión sobre el retrato y la didáctica del arte como campos para la subjetividad en términos políticos y estéticos a partir de una experiencia concreta en un territorio donde de facto se interponen factores materiales, culturales y políticos, así como explorar el contexto artístico y educativo del país que apenas cumple 50 años de independencia respecto a España y tiene una de las rentas per cápita más elevadas del continente africano.

The feature film Aquaman (2018, directed by James Wan) is the most commercially successful superhero movie belonging to the DC Extended Universe. Produced by DC Films and Warner Bros. Pictures, the motion picture portrays a rebellious superhero with an extraordinary physical presence. The paper aims to reflect on the movie Aquaman and its ability to function as a late modern fairy tale. Aquaman’s genre structure includes elements of fantasy, science-fiction and action film. However, the authors work with the assumption that the story is, in its nature, a fairy tale involving late modern means of expression. The first part of the text is largely theoretical, outlining the movie’s importance and defining the genre of a fairy tale in the context of late modern culture. Following the given line of thought, the second part of the paper presents a narrative analysis of the film in question, which is based on Propp’s morphology of fairy tales.

The author analyzed the functional, symbolic, and semantic features of the traditional dwelling of the Bulgarians of the Republic of Moldova in fairy-tale folklore (novelistic tales and anecdotes). The types of dwellings and their constituent elements (internal and external) were considered as a part of the material; culture is reflected through subject realities as a component of ethnic culture in the human microcosm. The connection of dwelling with traditional values of the Bulgarians of the Republic of Moldova was also analyzed.

As well as to identify the level of preservation of basic ethnic mentality under conditions of new “mental environment”. Thus, the studying of regional ethnic culture is an interdisciplinary research, which allows to discover how localization in time and space affects ethnic culture in general and oral folk art in particular.
