Texte intégralThis endeavour represents a pioneering effort to design and develop a cost-effective low-field Dip-NMR or Immersible NMR system dedicated to monitoring reaction mixtures at the closest possible source of information. While we have successfully designed and optimized the first version of prototype capable of acquiring single scanned NMR spectra of pure samples, further focused study and research are highly warranted to develop and integrate prototype into an immersible probe unit capable of multi-channel acquisition to realize the proposed Dip-NMR system. The NMR spectra obtained from Dip-NMR system were notably broad and necessitated dedicated chemometric methods for quantification and kinetic data extraction. Known reactions were monitored on a high-field high-resolution NMR spectrometer, with the kinetic data from this instrument serving as benchmark data for comparison and evaluation against the data obtained from Dip-NMR system. Various chemometric methods were explored and tested on the mimicked spectra for quantification and kinetic data extraction, with the results subsequently compared with the benchmark data. In parallel with the project’s goal, hydrosilylation reactions involving organic substrates such as nitriles, cyclic amides, and esters were catalyzed using an Ir(III) catalyst and monitored on a high-field high-resolution NMR spectrometer. These investigations have successfully yielded valuable insights into the reactions, contributing to a deeper understanding of the processes involved and laid important reference kinetic data for future testing of an advanced Dip-NMR probe prototype

Texte intégralIn swimming-pools, chlorine is used as a disinfectant to minimize the risk to users from microbial contaminants. In water, chlorine reacts with nitrogen compounds generated by human activity like saliva, sweat, urine and skin, leading to the formation of toxic compounds, such as monochloramine (NH2Cl), dichloramine (NHCl2), nitrogen trichloride (NCl3), chloroform (CHCl3), etc… The detection and the quantification of these volatile compounds at ppb level (part per billion) is an important and significant challenge to be able to monitor the air quality in swimming pool. Or, there is currently no commercially available and low-cost system which can instantaneously measure at ppb concentrations.The aim of this research is to develop a cheap, sensitive and selective chemical and colorimetric sensors of monochloramine, nitrogen trichloride and chloroform. For this purpose, we developed chemical sensors based on the use of nanoporous silicate matrices doped with probe-molecules. The NCl3 sensor doped with NaI and amylose can detect NCl3 at ppb level (5 ppb – 180 ppb) in humid atmospheres (from 50% to 80% relative humidity) at ambient pool temperatures. Due to the fast change of color, visible with naked eyes, these sensors can be used to detect peaks of pollution and to monitor the air quality of indoor pools. The NH2Cl selective sensor is based on the Berthelot reaction. The nanoporous silicate matrices doped with sodium nitroprusside and phenol in an alkaline medium, turn from transparent to blue upon exposure to gaseous NH2Cl. This sensor can detect NH2Cl in the range from 60 to 250 ppb in a very humid atmosphere (≈ 80%). Used to probe the quality of pool water, this sensor can detect NH2Cl in water with a detection limit of 0,1 µmol•L-1. A preliminary study of the CHCl3 detection was also conducted to identify probe-molecules capable of reacting with chloroform to form colored products. The reagents of the Fujiwara reaction were selected. The study of the 2,2’-bipyridine reactivity in solution in the presence of a strong base allowed highlighting the simultaneous formation of two colored compounds, whose formation depends on the nature of the reaction environment

Texte intégralIn this work, we are interested in studying dirofilarial infections in dogs and vectors “Mosquitoes” especially detection, monitoring and prophylaxis. The first objective is to develop a real-time duplex PCR targeting the COI gene capable of simultaneously detecting and differentiating D. immitis and D. repens. Subsequently, we applied this tool to a canine dirofilariosis surveillance process in different endemic areas of Mediterranean basin (Corsica and Algeria). We have thus detected by this molecular tool for the first time in France, D. immitis and D. repens in Aedes albopictus mosquitoes. We reported, also, the presence of D. immitis in dogs from northern Algeria.The second aim was to assess whether the MALDI-TOF MS can detect changes in the protein profiles of Aedes aegypti infected experimentaly with filarial nematodes (D. immitis, Brugia malayi and B. pahangi). Obtained results showed the potential of MALDI-TOF MS as a reliable tool for differentiating non-infected and filariae-infected Ae. aegypti mosquitoes with a best correct classification rate obtained from the thorax-head part with 94.1 and 86.6, 71.4 and 68.7% for non-infected and D. immitis, B. malayi and B. pahangi infected mosquitoes respectively.The third aim of this work has focused on the evaluation of the anti-feeding and insecticidal efficacy of an ectoparasiticide (Vectra® 3D) containing three active ingredients: dinotefurane, pyriproxyfen and permethrin (DPP) against Ae. albopictus. Results demonstrated that the DPP combination has significant anti-feeding and insecticidal efficacy against Ae. albopictus for at least 4 weeks

Texte intégralKidney transplant is the most performant strategy for renal replacement therapy. Increasing treatment targetting T cell response has led (i) to reappraise the importance of humoral allogenic response, (ii) to underline two main complications subsequent to immunosuppressive pressure : cytomegalovirus infection and tumorigenesis. Here, we first report the pathological impact of two of these factors on kidney allograft deterioration : CMV infection prior a biopsy for cause and Donor Specific Alloantibodies (DSA) detected within the graft with single antigen flow bead assay. Then we showed that CMV-induced Vδ2neg γδ T cells are a new player and a potentially useful clinical biomarker in antibody-mediated lesions of kidney transplants. By engaging DSA on their Fcγ-receptor CD16, γδ T cells participate in allograft lesions mediated by DSA through antibody-dependent cell cytotoxicity (ADCC). In addition to this new indirect allogenic function, CMV-induced Vδ2neg γδ T cells displayed a dual anti-CMV and anti-tumor reactivity. Finally, we here identified EphA2 as a new stress-regulated antigen targetting by a non Vδ2neg γδ T cell clone, which recognition implies the hijacking of the natural EphA2-ephrin interactions to activation. These data suggest that in humans, a fortiori when immunosuppressed, γδ T cells compose a lymphoid stress-surveillance compartment, capable of recognizing the dysregulated state of infected or transformed cells and to take part to allogenic response through an ADCC mechanism
