The process by which a zygote develops from a single cell into a multicellular organism is poorly understood. Advances are hindered by detection specificity and sensitivity limitations of single-cell protein tools and by challenges in integrating multimodal data. We introduce an open microfluidic tool expressly designed for same-cell phenotypic, protein, and mRNA profiling. We examine difficult-to-study—yet critically important—murine preimplantation embryo stages. In blastomeres dissociated from less well-studied two-cell embryos, we observe no significant GADD45a protein expression heterogeneity, apparent at the four-cell stage. In oocytes, we detect differences in full-length versus truncated DICER-1 mRNA and protein, which are insignificant by the two-cell stage. Single-embryo analyses reveal intraembryonic heterogeneity, differences between embryos of the same fertilization event and between donors, and reductions in the burden of animal sacrifice. Open microfluidic design integrates with existing workflows and opens new avenues for assessing the cellular-to-molecular heterogeneity inherent to preimplantation embryo development.

Today, for almost all products, the appearance is very important. Thus, for example, the appearance of universal gear reducers was not given so much attention, but today there are many very interesting solutions of shape, colour and graphic design. It should have in mind that universal gear reducers have much complex construction comparing them with special gear reducers. In the same time, they have to provide different positions and way of mounting and different speeds with different powers (torques) at the output shaft. Particularly, it should be noticed, that within all universal gear reducers single-stage gear units are nowadays increasingly used because their gear ratio values are increased (values of gear ratio can go up to 15) so that these reducers largely cover most of the two-stage gear reducers with their gear ratio values, so thereby their exploitation is significantly increased. Single-stage gear reducers are very simple products, and today there is a large number of manufacturers of such product. However, their mounting dimensions and way of mounting are not defined by a standard, so there is a variety of units with different dimensions and way of mounting. Although, most manufacturers look up to dimensions of leading manufacturers (usually SEW) in order to ensure greater exchangeability of their gear units. It is interesting to note, that most manufacturers tend to adopt the design of housing that will be substantially different from competing solutions. The reason is probably to avoid the possibility of being accused of copying someone else’s solution. However, there are manufacturers that retain almost identical housing design as a competing solution (probably they bought the licence or the solution is copied without permission). This paper considers only the design solutions of single-stage gear reducer in order to indicate the expected directions of their further development.

This paper presents results of rig testing of a transonic, single-stage turbine at off-design conditions. Mapping of the 3.4 pressure ratio, 1.9 stage loading turbine ranged from 70 through 120 percent of design speed and 75 to 125 percent of design pressure ratio. Results show expansion efficiency dropping over 4 percent from 100 to 80 percent of design speed at design pressure ratio, while remaining within half a percent from 90 to 110 percent of design pressure ratio at design speed. Efficiency lapse rate from equivalent sea-level to cruise altitude Reynolds numbers at the design point was measured and found to be worth over 1.5 percent. Analyses of test results using a viscous three-dimensional solver showed very good agreement for the efficiency change with speed.

The design and analysis of the multistage common emitter (CE) amplifier for improved voltage gain over the single stage CE amplifier is presented in this paper. The design procedure started with the design of a single stage CE amplifier. The design specifications for the CE amplifier were specified. The design of the multistage CE amplifier was done using the designed single stage CE amplifier as the basic configuration. The designed single stage and multistage CE amplifiers were simulated in the linear technology simulation program with integrated circuit emphasis (LT SPICE). The results obtained show that using analytical method a voltage gain of 45 dB was obtained for the single stage and 54 dB was obtained for the multistage respectively, the LT SPICE simulation software presented a result of 44 dB for the single stage and 54 dB for the multistage. The proposed amplifier may find suitable applications in devices operating in the low frequency such as audio signal applications. Furthermore, judging from these results coupling of more stages of the transistor amplifier to obtain greater voltage, current and power gains is recommended.

It is well known that multiplierless finite impulse response (FIR) filters in multiple-stage cascade form can achieve lower hardware cost and lower coefficient sensitivity than that of single stage design. In this work, a novel algorithm is proposed for the design of multiplierless multiple-stage cascaded FIR filters. Unlike to the conventional algorithms in which the number of stages is fixed and usually is fixed to two, the number of stage in the proposed algorithm is automatically determined. The design examples show that the proposed algorithm significantly outperforms the best existing algorithm in terms of hardware cost and the design time is also saved.

A single-stage axial fan having a pressure ratio of 1.01 is designed in the current study. The design pressure ratio is chosen based on the power available from the existing motor (2.2 kW). The design space for the axial flow fan was generated by varying specific flow and geometrical parameters in suitable steps, using a program written in MATLAB. The varied flow parameters are mass flow rate, inlet Mach number, inlet flow angle, and rotor speed. The geometrical parameters that were varied are hub to tip ratio, aspect ratio, and blade solidity. Using these as the input variables and applying free vortex theory for 3-dimensional blade design, the aerodynamic design of the axial flow fan was carried out. Performance parameters like flow coefficient, stage loading coefficient, degree of reaction, diffusion factor, De Haller’s number, and blade angles were calculated at the blade’s hub, mean, and tip. Total design space of 92160 data points was obtained from the combination of input parameters. Several constraints were applied to optimise the design space based on the available power from the existing motor and in-house manufacturing limitations. The initial design space was reduced to 82 data points using these constraints. To further reduce the number of points in the design space, off-design performance was evaluated for each of these data points. Following this, one design point was selected based on the optimum performance range in off-design operation, while considering manufacturing limitations. Using Mellor charts, a suitable blade profile was chosen based on the inlet and exit blade angles. NACA 65-410 airfoil was selected with a stagger of 55 degrees and an incidence of 6 degrees for optimum performance.

<p class="Abstract"><span>The demand of low noise amplifier (LNA) has been rising in today’s communication system. LNA is the basic building circuit of the receiver section satellite. The design concept demonstrates the design trade off with NF, gain, power consumption. This paper reports on with analysis of wideband LNA. This paper shows the schematic of LNA by using Darlington pair amplifier. This LNA has been fabricated on 90nm CMOS process. This paper is focused on to make comparison of three stage and single stage LNA. Here, the phase mismatch between these patameters is quantitavely analyzed to study the effect on gain and noise figure (NF). In this paper, single stage LNA has shown the 23 dB measured gain, while the three stages LNA has demonstrated 29 dB measured gain. Here, LNA designed using darlington pair shows low NF of 3.3-4.8 dB, which comparable to other reported single stage LNA designs and appreciably low compared to the three stages LNA. Hence, findings from this paper suggest the use of single stage LNA designed using Darlington pair in transceiver satellite applications.</span></p>

With the global financial crisis happened, companies and investors pay more attention to development expense. Development costs mainly include design cost at the design stage. We analyze the design process and extract many design factors in order to estimate design cost at the conceptual and embodiment design stage. A number of process-based design features that impact the design cost are considered. These design factors include: 1) the number of performances and constraints, 2) sub-functions level and relationships, and 3) solutions at the conceptual design stage. At the embodiment of the design stage the design factors include: 1) the number of parts and their relationships, 2) the types of materials, 3) load resources, and 4) the number and types of tolerances. We use relative cost to estimate design cost by comparing the number of design factors among designs. To demonstrate the relative design cost estimation in practice, Kodak single-use cameras are used as example products. Use of these methods proves beneficial in estimate design cost in the design process.

Abstract This paper presents an improved power quality converter (IPQC)-based power supply design for high brightness light emitting diode (HB-LED) low power lighting. The IPQC circuit uses a Cuk buck-boost converter to operate it in a discontinuous conduction mode (DCM) using the voltage follower technique for the mitigation of harmonic contents present in the AC mains current. Subsequently, reduction in harmonic contents results in improving the power quality indices at the AC mains. Single-stage single switch converter topology is used, which has less component count, size and cost as compared to the two-stage converter topology. DCM operation has an advantage that only output voltage control loop is required as compared to three control loops required in the continuous conduction mode operation. An 18-W LED driver is designed, modeled and simulated using MATLAB/Simulink software for 220 V, 50 Hz AC mains. The performance of the proposed LED driver is observed in terms of total harmonic distortion of the input current (THDi), input power factor (PF) and crest factor (CF) taking into account the strict international standard of IEC 61000-3-2 for class C equipments.

This paper presents a new single-stage single-switched forward converter with magnetic coupled nondissipative snubber, which gives good power factor correction (PFC), low current harmonic distortion, and tight output voltage regulation. The proposed converter features low switch current and voltage stresses, essential for the design of a single-stage power factor correction converter. The prototype shows that the IEC1000-3-2 requirements are met satisfactorily with nearly unity power factor. This proposed converter with magnetic coupled nondissipative snubber is particularly suited for power supply applications with low power level.

The performance of a single-stage low-speed compressor has been measured both before and after the introduction of certain features of the multistage flow environment. The aim is to make the single-stage rig more appropriate for developing design rules for multistage compressors. End-wall blockage was generated by teeth on the hub and casing upstream of the rotor. A grid fitted upstream produced free-stream turbulence at rotor inlet typical of multistage machines and raised stage efficiency by 1.8 percent at the design point. The potential field that would be generated by blade rows downstream of an embedded stage was replicated by introducing a pressure loss screen at stage exit. This reduced the stator hub corner separation and increased the rotor pressure rise at flow rates below design, changing the shape of the pressure-rise characteristic markedly. These results highlight the importance of features of the flow environment that are often omitted from single-stage experiments and offer improved understanding of stage aerodynamics.

A single-phase, single-stage, differential boost inverter comprises two independently-controlled boost DC-DC converters, with the load connected between their outputs. The net voltage on the load is sinusoidal and has a controllable frequency and magnitude that is larger than that of the DC source. The present work first derives steady-state and small-signal models of the inverter with parasitic elements. The results obtained from the line-to-output transfer function, control-to-output transfer function, open-loop input impedance, and open-loop output impedance models are compared with that of the ones obtained from the experimental testbed. Using the new models, a voltage mode controller is designed in the synchronous reference frame. The regulator design is explored through the use of an example. The results are verified against the small-signal model, then PLECS simulations, and finally a laboratory experiment. The results indicate excellent agreement between the model and experiment during transients in voltage reference, input source voltage, and output load. A sensitivity analysis is performed based on the inverter model considering the parameter variation. Finally, loss and efficiency estimations are provided in this work.

The use of variable profiles in engineering is the most effective way to reduce metal consumption and improve performance, increase the lifetime, reliability of machines and mechanisms, and reduce costs for their production, repair and operation. The purpose of the present work is to systematize the blanks of machine-building industries obtained by methods of plastic deformation and to develop general principes of the con cept of creating single-stage rolling mills on the basis of analysis of their design and operation. Three main classes of profiles are distinguished, differing in the type of the initial workpiece: strip, shaped and tube periodic billets. All these types of blanks can be used as semi-finished products for the production of the main parts of suspension and chassis of cars. Strip billets are widely used for the manufacturing of elastic suspension elements, tubular – for manufacturing of hollow body parts with increased requirements for strength and rigidity, in particular, driving axle housing, semi-axles of cars and trailers, jet engine casings. Shaped periodic profiles of various shapes are used as blanks for hot volumetric stamping. Mills and automatic lines for the rolling of blanks of low-spring springs, under-springs, guide bearings of air suspension, rolling of the bars of the front axle beam, manufacturing of protective elements and semi-axes of agricultural machinery developed under the guidance or with the direct participation of the author are presented. General requirements for the deformation equipment of machine-building industries are formulated. Recommendations to use induction heating and high-temperature thermomechanical treatment are justified. It is not recommended to change the direction of movement of the workpiece in the process chain. As a power drive equipment it is better to take electromechani cal, rather than hydraulic systems. It is also advisable to choose separate drives for each working element of the equipment.

This paper describes a bidirectional twisted single-phase single-stage buck-boost dc-ac converter based on an output unfolding circuit. This solution is derived by the combination of an inverting buck-boost dc-dc converter and an unfolding circuit. The operation principle, component design guidelines, along with the control approach are presented. The zero-crossing distortion problem is discussed and solved by a simple approach. The simulation and experimental results confirm all theoretical statements. Loss distribution and achievable efficiency are analyzed. Finally, the pros and cons of the proposed solution, along with the most promising application field, are analyzed and discussed in the conclusion.

The work presented in this paper concerned with the analysis, design and software implementation of the Solid State Transformer as an alternative to the conventional power transformer. The proposed transformer aims to perform the same task as the conventional one with additional facilities and advantages. Three stages are considered to configure the Solid State Transformer. The first stage which is known as input stage and implemented using Vienna rectifier which converts the AC voltage of the main supply to a DC voltage. The second stage (isolation stage) step down the DC voltage to a lower level DC voltage. This stage consists of a single – phase five-level diode clamped inverter, 1 KHz step – down transformer and fully controlled bridge rectifier. The output stage (third stage) is a three-phase three-level diode clamped inverter which converts the low level DC voltage to a three-phase, 50 Hz AC voltage. Model Predictive Current Control has been employed for driving transformer’s stages. The gating signal is produced directly when the given cost function is minimized, therefore there is no need of any modulator. Behavior of the proposed structure is achieved by simulation which shows high quality power conversion with low Total Harmonic Distortion.
