Texte intégralAdequate nutritional practices are the basis of profitability and sustainability of animal production and are one of the main factors influencing animal welfare. In addition to the chemical composition, the safety quality, in terms of fermentation quality and microbial contamination, plays an important role in determining the actual palatability and safety of feed. In the current PhD thesis, we addressed, through heuristic method of data and sample collection, the study of interactions between feed quality and impact on animal performance. In particular, the interactions between silage quality and diets were evaluated. Given the complexity of these matrices in terms of microbial populations influencing and driving feed quality, new challenges in nutritional assessment for cattle must move toward multi-parameter assessments that include chemical-biological, microbiological, and safety characterizations. The collection of this information conducted without predetermined aims, has allowed to analyze with multivariate statistics and machine learning techniques the relationships between feed quality and the effects they have on herd performance, proposing new approaches to classify feed quality and nutritional strategies adopted in dairy farms.

The object of work: six different Sultana raisin producers were examined. 42 samples were collected, 28 were collected from packed raisins and 14 from unpacked. YGC (Yeast Extract Glucose Chloramphenicol) agar was used to count yeasts and molds number. For E. coli identification was used Chromocult coliform agar. In every raisin sample yeasts and molds were identified.

Texte intégralIl presente lavoro, intitolato Il razionale e l’irrazionale: la psicologia del volere in Hegel, ha alla propria base l’intento di indagare le forme limitate del volere finito e il loro possibile superamento nella dimensione della libertà. Per far ciò si è scelto di prendere in considerazione da vicino il concetto hegeliano di “volontà” per come viene presentato da Hegel nelle sezioni interne alla Psicologia enciclopedica e nell’Introduzione ai Lineamenti di filosofia del diritto. I due perni concettuali sui quali ruota la presente analisi sono quelli di “formalismo” e di “volontà libera”. Da una parte, il “formalismo” viene individuato come elemento caratterizzante tutte le forme finite e limitate dello spirito pratico. In questo senso, l’opposizione tra razionale e irrazionale esprime tutta la conflittualità interna ai soggetti finiti tra la dimensione della loro naturalità e immediatezza (impulsi, inclinazioni, passioni, ecc.) e quella della razionalità a loro immanente, ma ancora non effettivamente compresa. Tale razionalità, partendo dal tentativo di controllo arbitrario del soggetto, conduce infine al superamento dello stesso arbitrio umano. Ogni passaggio interno alle singole forme finite della volontà può essere infatti letto attraverso la chiave ermeneutica di un progressivo autodeterminarsi razionale della volontà stessa, che progressivamente diviene sempre più consapevole dei limiti irrazionali, naturali e immediati della sua condizione finita a livello psicologico. Dall’altra, la “volontà libera” rappresenta invece quel momento dello sviluppo dello spirito, nel quale la consapevolezza dei limiti psicologici del singolo volere porta alla necessità del superamento degli stessi in una nuova dimensione dello spirito. I risultati ai quali giunge una tale analisi possono essere così riassunti: a) il “formalismo” esprime una caratteristica propria del volere del singolo soggetto psicologico. Esso si mostra ai nostri occhi come l’opposizione continua tra l’irrazionale, costituito dagli elementi ancora naturali e immediati del volere umano, e il razionale, proprio della dimensione universale di autodeterminazione e autocomprensione dello spirito; b) l’analisi delle forme limitate dello spirito pratico conduce tanto alla comprensione della necessità del loro porsi psicologico nello sviluppo dello spirito, quanto al loro finale superamento in virtù di tale comprensione; c) la volontà libera rappresenta quella dimensione, nella quale il formalismo viene compreso come condizione propria del singolo soggetto finito a livello psicologico. Essa determina il superamento del formalismo, ma, allo stesso tempo, apre alla problematizzazione della nozione di “libertà”, come eccedente la stessa psicologia umana.

Texte intégralLa tesi di dottorato si propone di studiare alcuni aspetti teorici e pratici, sia su scala di campo che su scala territoriale, relativi all’uso razionale dell’ acqua con l’irrigazione. Il primo studio riguarda la quantificazione degli effetti correlati all’intercettazione fogliare dell’acqua di pioggia e irrigua in un campo investito a mais e l’effetto di questo fenomeno, a livello della fila, sulle dinamiche dell’acqua nel suolo. I risultati delle prove evidenziano che l’intercettazione fogliare si ripartisce mediamente per il 24% in throughfall (acqua che cade dalle foglie direttamente al suolo) e per il 76 % in stemflow (acqua che arriva al suolo scorrendo lungo il culmo); si è anche riscontrato che il rapporto stemflow/throughfall diminuisce in modo logaritmico all’aumentare della pluviometria. Successivamente, utilizzando la curva che descrive la ripartizione dell’acqua al variare della pluviometria e i dati di umidità rilevati durante il periodo di prova, si è simulato in termini di evapotraspirazione del mais, con il programma Hydrus 2D/3D, l’effetto dell’intercettazione vegetale, di un’ipotetica distribuzione uniforme al suolo e di un sistema di irrigazione localizzata. Dalle simulazioni appare che l’evapotraspirazione è superiore per l’ipotetica distribuzione uniforme rispetto al sistema localizzato, entrambi però superiori rispetto al flusso ripartito dalla copertura vegetale. Il secondo argomento riguarda due sperimentazioni che sono state condotte per valutare l’efficacia dell’irrigazione a goccia nel razionalizzare l’uso dell’acqua: una in un’azienda situata nella provincia di Verona su mais ed una nella provincia di Mendoza in Argentina su vite. La prima ricerca sperimentale è stata condotta presso la Cooperativa Agricola Zootecnica “LA TORRE” di Isola della Scala (VR) e ha riguardato la valutazione dell’impatto dell’uniformità di distribuzione dei due sistemi irrigui sulla resa del mais a maturazione cerosa. I sistemi utilizzati sono stati da una parte l’irrigazione per aspersione gestita con interventi di soccorso e dall’altra l’ irrigazione a goccia gestita a calendario. È stata calcolata inoltre l’efficacia d’uso dell’acqua e la convenienza economica di entrambi i sistemi irrigui. L’uniformità di distribuzione, espressa mediante i coefficienti DUlq e CU, è risultata buona ma non ottima nel sistema microirriguo (DUlq 0,73 e CU 84,74%) a causa della lunghezza dell’appezzamento che ha portato a perdite di pressione nella parte terminale delle linee gocciolanti, mentre è stata bassa per l’aspersione (DUlq 0,33 e CU 58,94%) a causa soprattutto dell’errato posizionamento degli irrigatori che non consentiva una omogenea copertura dell’area. L’efficacia d’uso dell’acqua è stata superiore per il sistema irriguo a pioggia rispetto alla goccia, indicando che la gestione razionale dei volumi irrigui ha un’influenza maggiore rispetto all’uniformità di distribuzione. Dal bilancio economico di entrambi i sistemi irrigui è risultato che l’utile sul tal quale risulta appena superiore per il rotolone rispetto alla gestione a goccia. La seconda sperimentazione ha valutato l’effetto dell’uniformità di distribuzione dell’irrigazione a goccia su alcune caratteristiche delle piante di vite e il confronto con l’irrigazione superficiale mediante indici di vegetazione a partire da immagini satellitari Landsat. Questo lavoro sperimentale, realizzato in un’azienda viticolo frutticola nella zona est di Mendoza ha avuto tre obiettivi: 1) determinare l’impatto dell’uniformità di distribuzione sulla vite; 2) osservare l’effetto della conversione dall’irrigazione per scorrimento superficiale all’irrigazione a goccia mediante l’utilizzo di indici vegetativi a partire da immagini satellitari Landsat; 3) comparare la variabilità della resa e del diametro del tronco in viti irrigate a goccia e per scorrimento superficiale. I risultati indicano che l’uniformità di distribuzione dell’irrigazione a goccia, secondo i coefficienti DUlq, CU e CV, è stata eccellente nonostante si siano osservati un importante numero di orifizi ostruiti. Inoltre, si nota una bassa correlazione tra i parametri quantitativi e qualitativi della vite e la portata degli erogatori e la percentuale di orifizi ostruiti. Gli indici di vegetazione hanno permesso di verificare il significativo miglioramento dello stato vegetativo degli appezzamenti irrigati a goccia rispetto a quelli irrigati per scorrimento superficiale. Per ultimo, il confronto in termini di tendenza della variabilità di resa e diametro del tronco, mostra che negli appezzamenti irrigati a scorrimento superficiale appare lievemente superiore rispetto a quelli irrigati a goccia. Questo può essere attribuito alla influenza dell’irregolarità topografica del suolo. Infine, è stato effettuato uno studio sui fabbisogni idrici territoriali nell’ambito del consorzio di bonifica sinistra Piave situato sulla parte nord della provincia di Treviso. In questa ricerca è stato applicato un modello supportato da un sistema GIS per valutare l’uso dell’acqua per l’irrigazione su un’area all’interno del bacino del Piave. Il modello georefernziato è stato realizzato integrando le informazioni raccolte da un campione di agricoltori che hanno compilato un questionario mirato a caratterizzare l’uso dell’acqua irrigua dell’area, dalla mappa dei suoli, dalla mappa d’uso dei suoli, dai dati medi di evapotraspirazione e pioggia, e dalla fornitura d’acqua per l’irrigazione all’interno dell’area. Grazie all’utilizzo integrato di uno strumento GIS (Geographical Information System) e del programma Management Zone Analyst (MZA), è stata effettuata un’analisi cluster per la stima del bilancio idrico e degli eccessi di fornitura idrica. Al fine di analizzare le possibili strategie che permettono di ottenere una fornitura idrica ottimale garantendo le rese delle colture sono stati analizzati quattro diversi scenari con gestioni diverse nella frequenza e durata dei turni irrigui mensili. Dai risultati delle simulazioni si può concludere che passando da 3 a 4 turni al mese, con una riduzione però delle ore per turno tale che la quantità di acqua fornita sia inferiore a quella prevista per i tre turni, visto la maggiore efficienza irrigua, potrebbe essere una buona strategia per garantire le rese delle colture il più possibile dai cambiamenti climatici che come sembra aumentano la variabilità degli eventi piovosi. Il metodo proposto è un’applicazione semplificata che può essere usata per altri fiumi.

American industrial cities, particularly in the Northeast and Midwest, have seen a steep decline in manufacturing jobs over the past so years. For those cities whose economies were not diversified and relied on one or two major companies for their employment base, the job loss has been particularly crippling. This decline, coupled with the flight of retail, service, and other jobs to the outlying suburbs as well as a loss in upper- and middle-income residents to those same areas, has left many cities decimated. Perhaps nowhere has the effect of this been more apparent than in the downtown areas of these cities. This thesis will examine the growth and decline of the downtown in American cities as well as the development of one city's downtown in particular, Schenectady, New York. A number of interventions, projects, and proposals that have been part of the struggle to reverse the decline of that city's core will be examined. These will be considered within a problem-solving process to discover where assumptions were faulty or where externalities negatively influenced them-contributing to missed objectives and goals. Where applicable, alternative means of reaching the stated goals will be explored. This thesis will also explore the idea of a community vision and explore the role it plays in determining the direction that downtown revitalization takes. After outlining the past interventions, this thesis will consider and evaluate the current programs, projects, and organizations that are involved in the latest round of redevelopment efforts in the downtown. It will look at the assets that the city should consider building on as well as the liabilities that are holding it back. This thesis will also make recommendation for future strategies and consider the questions: What makes a healthy, downtown, and how can Schenectady direct its efforts toward achieving this goal? and Are the current projects and likely to bring the city closer towards that objective, and if not, what other options should be explored.

Texte intégralThis multi-modal dissertation examines the historical hegemonic making of U.S. education, and how compulsory schooling has framed acceptable notions of culture, language/literacy, and knowledge production. Through this criticism of colonization and education, theoretical and practical alternatives are explored for the opportunities outside mainstream schooling in the US. In examining the literary work on decolonizing education, these efforts can engage in unlearning of coloniality by finding examples from a time before colonization. In contemporary society, the practice of de/unschooling can hold the possibilities for decolonizing education. To demonstrate how families of color in the U.S. engage with unschooling, interview questions serve as the sharing of knowledge and experience so as to ground the research in lived reality. A brief survey of critical education and critical pedagogy broadens those already critical of schools and/or receptive to the criticism of schools and the un/deschooling alternative then places student and family/community as the center of learning and teaching.

Texte intégralAcute myeloid leukemia (AML) has a very poor 5-year survival of ~20% in Europe. The internal tandem duplication (ITD) mutation of the Fms-like receptor tyrosine kinase 3 (FLT3) (FLT3-ITD) is the most frequent mutation (~25%) in normal karyotype AML. In recent clinical studies, few patients display prolonged remissions with receptor tyrosine kinase (RTK) inhibitors, such as FLT3 inhibitors (FLT3i) therapy, highlighting a substantial unmet need for novel effective treatment. Persistence of leukemia stem cells (LSC) drive AML leukemogenesis, responsible for drug resistance and disease relapse following conventional chemotherapy. Growing evidence recognizes that FLT3-ITD mutation leads to the constitutive activation of FLT3 kinase and its downstream pathways, including PI3K/AKT/mTOR signaling, strongly associated with LSC survival and crosstalk between LSC and stromal cells associated bone marrow (BM) tumor environment (TME). The TME provides protection of FLT3-ITD AML cells against FLT3 inhibitors. Thus, the PI3K/AKT/mTOR pathway may represent as a putative target for FLT3-ITD AML. This study aims to test the hypothesis that PI3K/AKT/mTOR inhibition could sensitize FLT3-ITD AML cells to RTKi-lead targeted therapy using human AML cell lines and primary patient blasts. First, I uncover the phenotypic profile of FLT3-ITD versus FLT3 wildtype cell lines following treatment with selected FLT3i or PI3K/AKT/mTORi that have failed treatment of AML as monotherapy in clinical studies. More specifically, I determine the drug efficacy by means of cell growth measurement and assessment of cell cycle status and apoptosis. I was able to demonstrate that BAY-806946 (pan PI3Ki) and PF-04691502 (dual PI3K/mTORi) exerted growth inhibitory activity caused by G1 cell cycle arrest and apoptosis, and this effect was irrespective of FLT3 status. Quizartinib (FLT3i) selectively inhibited cell growth in FLT3-ITD AML and this effect was mainly caused by apoptosis. The observed drug-induced apoptotic effect was however not as efficient as chemotherapy. Next, I provide proof-of-concept for the combination of quizartinib and BAY-806946 using both FLT3-ITD AML cell lines and primary patient blasts. When evaluating on primary patient blasts, I take into consideration the protective role of mesenchymal stromal cells and physiological growth factors to mimic the BM microenvironment. Hereby, I co-cultured FLT3-ITD AML blasts with stromal cell line MS-5 and added growth factors essential for AML survival and differentiation such as IL-3, TPO and G-CSF at physiological concentration. As expected, treatment with BAY-806946 enhanced both cytostatic and cytotoxic effect of quizartinib in FLT3-ITD AML cell line MOLM-13 as well as primary patient blasts in co-culture. More importantly, enhanced apoptosis was measured in the stem cell like CD34+CD38- population. Lastly, I elucidate the cytokine profile and persistent phosphoproteins as putative targets following combination treatment. Ultimately, this study demonstrates the potential of PI3K/AKT/mTORi to enhance the efficacy of RTKi quizartinib for the treatment of FLT3-ITD AML.

Texte intégralEnzymes involved in NAD(P) metabolism are attractive targets for drug discovery against human diseases such as tuberculosis. NAD kinase catalyzes a phosphorylation of NAD using ATP or inorganic polyphosphates (poly(P)) as phosphoryl donors to give NADP. No other pathway of NADP biosynthesis has been found in the procariotic or eukaryotic cells. The validation of this enzyme as drug target is based on its essentiality in Mycobacterium tuberculosis, as well as on differential structural features between mycobacterial and human enzyme. In order to find a compound able to selectively inhibit the M. tuberculosis NAD kinase, structure-based analyses of this enzyme, resulted in the design of the several modified NAD and poly(P) analogues. We found that dinucleoside disulfide mimics of NAD analogues are novel selective inhibitors of NAD kinases that bind at the NAD-binding domain of NAD kinases, but do not affect the majority of other NAD-dependent enzymes such as NAD dehydrogenase. Instead, the dinucleotide polyphosphates and poly(P) analogs showed a weak selectivity against mtNAD kinase because, while the human enzyme is strictly ATP-dependent, the mycobacterial can use polyphosphates as phosphoryl donors. The differences existing in the catalytic behaviour or in the substrate specificity between the mycobacterial and human enzyme, will be exploited for future design of new potent and selective antitubercular drugs. Inhibitors of human enzyme are also of interest as they can be used to reduce the critical supply of exogenous NADPH during oxidative stress and in cancer cells. Recently, it was suggested that reactive oxygen species (ROS) derived from NADPH oxidase plays important roles in physiological and pathological angiogenesis making this enzyme an excellent target for anticancer therapy. The suppression of NADPH oxidase by inhibition of NAD kinase may show some anticancer effects. Finally, we search for another human NAD kinase gene. C5orf33, a putative NAD kinase, has been cloned and expressed in Escherichia Coli and partially purified to check the kinase activity.

Texte intégralLinear Rational Insurance Model Abstract The aim of the first work is to provide a closed pricing formula for insurance contracts in a linear rational framework, which consists in assuming the existence of a linear drift diffusion process and a state price density which is a linear function of it. The main advantage of this process is that we can compute the conditional expectation of polyno- mials function of this diffusion: in fact a polynomial can be see as a linear combination of an enlarged set of variables with a linear drift, this can be proofed by means of the Itô’s formula. This result is very important because it allows us, under the hypothesis that the diffusion part is a martingale, to use all the results we have about linear drift diffusions for this new set of variables. As a consequence, we are able to give the price of three important life insurance contracts: the sur- vival and death benefit and the guaranteed annuity option (also called GAO). It is about the GAO that we can see the advantage of the framework we are using: actually the payoff of the GAO is not an affine or a polynomial function, so the only way to treat it is by performing a change of measure or a Monte Carlo simulation. We show that, under the assumption that the state space is compact, we are able to approximate the GAO payoff by a polynomial, which will allow us to find a closed formula for the price of this contract. The end of this work is dedicated to some numerical experiments which have the aim to point out the importance of the choice of the degree of the approximated polynomials in order to have reliable results. We show that a ten degree polynomial is able to estimate with a small error the Monte Carlo price of the GAO. This work extend the existing literature concerning polynomial models and their application in life insurance, proposing a pricing method also for liabilities which are not necessarily building blocks, but more complicated functions, like the guaranteed annuity option. Economic Scenario Generator The aim of this second work is to build an economic scenario generator with the intention of improving the portfolio allocation of Bpifrance. In order to do that, we have to pass through a different number of steps. The first thing is to study, by a principal component analysis, the present portfolio of Bpifrance, in order to find the variables which explains the most of its variability. A second step consists in selecting from the market the financial instruments that allows us to replicate the components we retained from the step before. This part is then completed by both an univariate and multivariate analysis of these assets, finding in this way the stylized facts that we need to take into account when choosing a model for the diffusion of the price of these financial factors. The third step, and last concerning our work, is to estimate the parameters of the models we retained and see if they are able to fit the empirical data and, as a consequence, if they could be used as a part of our future economic scenario generator. In order to achieve this point, we focus only on the diffusion of the equity indices, proposing also a model who takes into account the dependency on the inflation. We will see that on the basis of our data there is no evidence to link the return on equity indices on the realisations of this macroeconomic factor.

Texte intégralThe observation of the Italian Funds Market shows that the real estate closed-end funds trade at an average discount on their Net Asset Value (NAV). As regards to the Italian market, anecdotical and empirical surveys stress that the main reasons of this phenomenon are referred to the structural characteristics of the market (the scarce outstanding shares of the Italian real estate funds market, the poor liquidity of the secondary market, the low interest in dealing shown by institutional investors, etc), to the peculiarities of real estate funds (the holding discount, the low disclosure on corporate governance of management companies, etc) and to the features of the underlying real estate assets (the fairness of the independent expert evaluations, the real estate systematic risk, etc). This paper enters the wide international debate about closed-end fund puzzle and argues, following a rational approach, if the puzzle can be explained by technical characteristics and performance aspects of Italian Real Estate funds. The empirical evidence points out that the NAV discount increases as the time to maturity and the nominal size of shares increase, that it is higher for blind pool funds and lower for seeded funds, and that the NAV discount is higher for income distribution funds and lower for income accumulation funds. Furthermore, the analysis bears that the NAV discount decreases as the ROA increases while it increases as the Price/earning ratio increases. Finally, the study shows the NAV discount increases as the leverage increases.

Scant attention has been paid to the event-decisions of corrupt agents in order to explore how an agent decides to pay, how he identifies his partner, how he contacts him, how he negotiates with him, how he manages to achieve his goal, and how he protects himself against “lemons”. The aim of this thesis is to understand the corruption-commission process by using crime script analysis to delve into the event-decisions of corrupt agents. Therefore, within the Italian context, this thesis seeks to answer the following research questions: how do corrupt deals in public procurement take place? What are the tactics, dynamics and the modi operandi of corrupt agents? What are the steps of corruption-commission in the specific area of public procurement? How can these steps be explained? What are the facilitators of corruption-commission? After the relevant literature has been reviewed (Chapter 1) and the methodology has been discussed (chapter 2), the results of the research are presented in chapters 3 and 4, where the procedural aspects and the procedural requirements of the crime are presented and discussed. Some concluding remarks close the thesis.

Texte intégralProfessionalism and experimentation are the key concepts of the following research and are identified from a feeling of adherence to the work of Robaldo Morozzo della Rocca, architect born in Turin, educated in Rome and based in Genoa. Through the critical definition of the dimensions and the features of Morozzo's work (observed within his social, cultural, economic and political framework) – the work aims to decipher both the newness and the contemporary feasibility of na un-programmatic, free and disenchanted approach that nonetheless is always characterized by a cultured methodological-professional coherence. The following contribution to the critical debate around the definition of modern architecture, takes its cue from the observation of a basic condition, detectable in the work of Morozzo, which we will define post-ideological for the linguistic autonomy from any external conditioning. A sort of sui generis professionalism, made up of design itineraries that are hardly recognizable and can never be summarized in an univocal image, which make it an unreliable 'product' for historiography. Morozzo appears as an irregular figure, individualistic, syncretic, simultaneously rigorous and arbitrary, complex and contradictory, sophisticated and popular that makes Genoa a particular observation point for the transformation of the reality and an intimate laboratory of formal experimentation in the field of architecture.

Texte intégralThe present thesis analyses the reception of the thought of Origen of Alexandria (c. 184-c. 253) in Jean Le Clerc (1657-1736). Its particular focus is on Origen's conception of freedom and the theological doctrines related to it. The goal of this thesis is to uncover, through Le Clerc's use of Origen, some of the argumentative practices and the intellectual dynamics of the time, in particular in religious, especially inter-confessional, debates. This thesis is divided into three main parts. The first part has mainly an introductory character and looks at the epistemological assumptions of Le Clerc and his relationship with intellectual and religious authorities of the past. The second part reviews the various ways in which Le Clerc quoted, referred to or otherwise made use of the thought or the name of Origen in his vast production. This part provides a first result in that it frames, in general, Le Clerc's reception of Origen. This step is, at the same time, also preparatory for the material contained in part three. In the third part, only the material is considered which is strictly related to Origen's idea of freedom and the related theological doctrines of original sin, grace/predestination, and the problem of evil. The result of this analysis, as it appears form the examination of argumentative practices in the previous sections, is that Le Clerc was no simple "Origenist" but neither was he was fully uncommitted to the Origenian cause. A full commitment to Origen, despite this strong sympathy, was still hindered by Le Clerc's epistemological, scriptural and theological outlook.
