Lith., 18(1): 40-45. The genomic DNA purification method for Gonyostomum semen algae was optimized by applying different DNA purification techniques and rational modifications. This method allowed to obtain high quality DNA preparations suitable for the phylogenetic analysis and genetic variability investigation of algae. DNA isolated by this method yielded strong and reliable amplification products showing their applicability for RAPD-PCR using random decamer primers. In the present study, the RAPD protocol was optimized for the evaluation of Gonyostomum biodiversity.

The new records of genera and species of algae (Chlorodendrophyceae, Chlorophyceae, Cryptophyceae,Mesostigmatophyceae, Raphidophyceae, Xanthophyceae) for the West-Siberian Plain and the Far East, including AltaiKrai, Novosibirsk Oblast and Primorskii Krai, were revealed. The raphidophyte flagellate Chattonella Marina from South Australia that was associated with the mortality of farmed tuna in April 1996 was successfully cultured. This study investigates ecophenotypic variation in physiology, chemotaxonomy and ichthyotoxicity of Australian and Japanese C. marina. Australian C. marina had similar temperature and salinity requirements to the Japanese strain but was adapted to higher light intensities than the Japanese strain. This differentiation was reflected in high concentrations of mycosporinelike amino acids (MAA's), especially the antioxidant MAA mycosporine-glycine, in the Australian cultures. Mycosporine-glycine was absent in the Japanese strain which instead used a violaxanthin:zeaxanthin cycle to moderate inhibition by high PAR irradiance. Ecophenotypic variations in lipid profiles were also observed between Chattonella strains from different geographic locations. Fatty acid and sterol profiles allowed for a clear discrimination between the raphidophyte genera Chattonella, Heterosigma, Fibrocapsa and Olisthodiscus, but exhibited little differentiation between C. marina, C. antiqua and C. subsalsa. Sterol and fatty acid profiles do not support the separation of C. antiqua and C. marina as distinct species. Sterol signatures, which may be useful as chemotaxonomic markers, were identified. Lipid composition correlated more closely to recent molecular classification of raphidophytes than classification based upon carotenoid pigments. Previous research on ichthyotoxic principles of C. marina has focused on production of high levels of reactive oxygen species (ROS), a brevetoxin-like compound and free fatty acids. This study found that Chattonella marina produces levels of the ROS superoxide 100 times higher than most algae, which is partially controlled by electrons donated through photosynthetic electron transfer. Differing superoxide production and toxic effects on zooplankton and fish are documented between different geographic strains of C. marina and light treatments. These results suggest a synergistic effect between ROS and an ichthyotoxin, and cannot be explained on the basis of these mechanisms of toxicity on their own. Our investigations into Australian C. marina demonstrate an absence or only very low concentrations of brevetoxin-like compounds by LC-MS techniques and negative mouse bioassays. All raphidophyte species were found to have high levels of eicosapentaenoic acid (EPA), which tested positive as a potential ichthyotoxin using damselfish as a model organism. EPA produced a similar mortality and fish behavioural response to that of intact C. marina cells while superoxide alone was not sufficient to cause fish mortality. However, superoxide in combination with low concentrations of EPA accelerated fish mortality. Implications of this work for mitigating the impact of Chattonella algal blooms on finfish aquaculture are discussed.

Particularly the work was focused on: Fibrocapsa japonica atypical occurrence offshore, in a cyclonic eddy in the eastern Alboran Sea (Western Mediterranean Sea), and Heterosigma akashiwo photosynthetic parameter (alpha, beta and Pmax) co-variation during experiments under sinusoidal and continuous light regime, and nitrogen starvation, which were conducted at the Laboratoire d’Oceanographie, Observatoire Oceanologique, Villefranche-sur-mer, Unviersitè Pierre et Marie Curie, CNRS, France.

Chattonella marina algal blooms have been associated with major farmed and wild fish mortalities in tropical (Mexico), subtropical (South Australia) and temperate regions (Japan). However, the precise toxic mechanisms involved remain incompletely known and disputed. A novel sensitive in vitro assay using the rainbow trout cell line RTgill-W1 was developed to assess toxicity of lipid extracts, ruptured cells and intact cultures of 6 strains of C. marina. Chattonella was found to be less toxic than the haptophyte Prymnesium parvum, but more toxic than the raphidophytes Heterosigma akashiwo, Fibrocapsa japonica and dinoflagellate Karenia mikimotoi. Ruptured cells from Australian CMPL01 and Japanese N-118 C. marina strains were the most toxic, decreasing gill cell viability by 71 and 65%, respectively. The Mexican CMCV-1 strain was the least toxic (35%), possibly because it is larger and less fragile than other strains. Chattonella marina is unique among ichthyotoxic microalgae in its high production of superoxide anion (19 pmol cell\(^{-1}\) hr\(^{-1}\)). Sonicated cultures showed higher levels of superoxide than intact cultures (19.0 vs 9.5 pmol cell\(^{-1}\) hr\(^{-1}\), respectively). However, O\(^2\) - on its own did not appear to be the main cause of toxicity (only 14% loss of cell viability). Superoxide production was highest when grown in medium enriched with 1 μM Fe(III) compared to the 5 μM standard medium (25 vs 9.5 pmol cell\(^{-1}\) hr\(^{-1}\)), especially after a sudden change from dark adaptation to light conditions (200 μmol photons m\(^{-2}\) s-1) (up to 37.6), but lowest (4.1) at 100 nM Fe(III). The three major fatty acids in C. marina were palmitic (PA), eicosapentaenoic (EPA), and octadecatetraenoic (OTA) acids. Higher levels of EPA were found in ruptured cells (21.4 – 29.4% of total fatty acids) compared to intact cells (8.5 – 25.3%). Gill damaging effects from free fatty acid (FFA) fractions were conclusively demonstrated (LC\(_{50}\) at 1 hr of 0.44 μg mL\(^{-1}\)), and remain candidates of ichthyotoxicity when well-defined toxins such as brevetoxin or karlotoxin can be ruled out as causative factors. The aldehydes 2E,4E-decadienal and 2E,4E-heptadienal, which may be generated by FFA oxidation, also showed high impacts on gill cell viability, with LC\(_{50}\) (at 1 hr) of 0.34 and 0.36 μg mL\(^{-1}\), respectively. In conclusion, C. marina was more toxic after cell disruption and when switching from dark to light conditions, associated with higher EPA levels and a higher production of superoxide anion. Implications of this work for finfish aquaculture management and mitigation are discussed.
