Radioactive iodine-capturing materials are urgently needed for the emerging challenges in nuclear waste disposal. The various pore structures of covalent organic frameworks (COFs) render them promising candidates for efficient iodine adsorption. However, the detailed structure–property relationship of COFs in iodine adsorption remains elusive. Herein, two polymorphic COFs with significantly different crystalline structures are obtained based on the same building blocks with varied molecular ratios. The two COFs both have high crystallinity, high specific surface area, and excellent chemical and thermal stability. Compared with the [C4+C4] topology (PyT-2) with an AA stacking form, the [C4+C2] topology (PyT-1) with an AB stacking form has more twisted pore channels and complex ink-bottle pores. At ambient conditions, PyT-1 and PyT-2 both exhibit good adsorption properties for iodine capture either in a gaseous or liquid medium. Remarkably, PyT-1 presents an excellent maximum adsorption capacity (0.635 g g−1), and the adsorption limit of PyT-2 is 0.445 g g−1 in an n-hexane solution with an iodine concentration of 400 mg L−1, which is highly comparable to the state-of-the-art iodine absorption performance. This study provides a guide for the future molecular design strategy toward novel iodine adsorbents.

Objective: To corroborate the result of postlaryngectomy olfactory rehabilitation evaluating the efficacy of the Polite Yawning Technique (PYT) with rhinomanometry and odours with the Smell diskettes Olfaction test (SDOT). Materials and methods: Thirty-two laryngectomised patients were subjected to olfactory rehabilitation consisting of 15-minute training and independent exercising for two weeks. The sense of smell and nasal airflow in laryngectomised patients were examined before and after implementation of PYT. Results: The differences in SDOT results prior to and after introduction of PYT were statistically significant, most patients achieving better results after rehabilitation. A significant positive correlation was obtained in results of SDOT and rhinomanometry prior to and after introduction of PYT. The results of measurements after rehabilitation showed that 75% of patients had improved nasal breathing, 78% of patients had a better sense of smell and 40% of patients became normosmic. Conclusions: PYT has proved to be an effective method in olfaction rehabilitation following total laryngectomy. Using rhinomanometry, evaluation of the rehabilitation success is substantiated while the technique is simplified.

AbstractAn experimental pedology research project, based on a “process-system” pedogenetic model, was initiated to investigate the weathering of Phlegraean Yellow Tuff (PYT) – the yellow facies of the Campanian Ignimbrite with phillipsite > chabazite – as representative of the parent rock material as the soil-forming factor. PYT was weathered by water (W) and tannic acid solutions (TA) at concentrations ranging from 3 × 101to 3 × 104μmol l–1. The most concentrated TA solution (3 × 104μmol l–1) showed the greatest extracting efficiency. Na and K were continually solubilized by W; Al, Ca and Mg contributed to the ionic composition of all the TA supernatants. A significant dissolution of the amorphous component and an associated decrease of the zeolite content, especially phillipsite, were noted. The features of PYT weathered outcomes fitted the “process-system” model very well. PYT showed a pedogenic potential, in being prone to weathering, as well as in releasing elements of importance for plant settlement and development, such as K, Ca, Mg and Fe.

O objetivo deste trabalho foi determinar a concentração de manitol para a germinação e crescimento mínimo in vitro do coqueiro (Cocos nucifera). Os seguintes acessos foram utilizados: Anão-vermelho-do brasil-de-gramame (BRDG), Anão-amarelo-da-malásia (MYD), Gigante-do-brasil-praia-do-forte (BRA), Gigante-da-polinésia (PYT) e Gigante-de-rennel (RIT). Os embriões foram colocados em meio de cultura Y3 a diferentes concentrações de manitol (0, 0,1, 0,2, 0,3 e 0,4 mol L-1). Verificou-se inibição do crescimento da parte aérea na presença de 0,1, 0,2 e 0,3 mol L-1, para MYD e BRDG, e de 0,1 e 0,2 mol L-1, para PYT, BRA e RIT. O uso do manitol é uma estratégia promissora para a conservação por crescimento mínimo.

Introduction. Based on data on the role of blood erythrocytes in the development and implementation of the vaccine and infectious processes in plague, it was of interest to evaluate changes in erythrocyte surface architecture from the position of searching for informative criteria for the preclinical evaluation of anti-plague vaccines. Aim — using atomic force microscopy to characterize the state of the blood erythrocyte membrane of guinea pigs in response to subcutaneous administration of the vaccine strain Yersinia pestis EV NIIEG and its isogenic derivatives. Materials and methods. For immunization of animals strain Y. pestis EV NIIEG (pYT+, pYV+, pYP+) and its isogenic derivatives Y. pestis KM216 (pYT–, pYV–, pYP+), Y. pestis KM217 (pYT–, pYV+, pYP–), Y. pestis KM218 (pYT–, pYV–, pYP–) were used. Analysis of the erythrocyte membrane was carried out using a Solver P47-PRO scanning probe microscope (NT-MDT, Russia). Results. The most pronounced changes in the surface architectonics of the membrane of guinea pig erythrocytes were established during the first three days of the formation of the immune response to the Y. pestis EV strain NIIEG and its isogenic variant Y. pestis KM217, in the genome of which the pYV plasmid is preserved, administered at a dose of 5 × 108 CFU. A significant increase (p 0.05) in the proportion of transformed cell forms (43.67 ± 3.63% and 37.83 ± 7.03% versus 4.08 ± 0.86% in the control group), root mean square roughness (319 ± 8 nm and 312 ± 7 nm versus 70 ± 6 nm in the control group), Young's modulus (125.73 ± 4.48 kPa and 113.8 ± 5.41 kPa versus 53.03 ± 1.47 kPa in the control group). By the 21st day, the value of these indicators decreased by an average of 2.7, 2.0 and 1.5 times, respectively, indicating restoration of the erythrocyte membrane. Conclusion. The dependence of the changes in the erythrocyte membrane and the rate of their restoration on the plasmid composition of Y. pestis strains has been established. The data obtained contribute to the understanding of the processes of interaction of Y. pestis with the erythrocyte membrane and can be used as additional characteristics in the development of new criteria for preclinical evaluation of plague candidate vaccines.

Breeding for high yielding rice varieties with VVEM (&lt;90 days after sowing/ das) is considered important to increase cropping indexes in the fully irrigated farm lands. This research was aimed to evaluate the yield potential of 200 promising VVE and VE maturing rice lines. A total of 200 VVE and VE maturing rice lines and five check varieties (Ciherang, Dodokan, Inpari 1, Inpari 13, Silugonggo) were evaluated in Preliminary Yield Trial (PYT) at Sukamandi Experimental Farm using augmented design, during the dry season of 2012. The 17 lines selected from PYT along with three check varieties (Silugonggo, Inpari 13, Ciherang) were further evaluated in Advanced Yield Trial (AYT) using randomized complete block design at Sukamandi, Kuningan, Magelang and Klaten, during wet season of 2012. Of the 200 lines tested in PYT, three lines yielded significantly higher than the best check variety (Inpari 1) and 2 lines did equal to Inpari 1. A total of 155 lines were VVE (&lt;90 das), and 45 lines were VE maturing (91-104 das). Based on yield per day, 17 lines were selected to be evaluated in AYT in WS 2012. Based on combined analyses from four locations of AYT, seven lines produced grain yield/ha and grain yield/day higher than did the best check Silugonggo (5.51 t/ha and 51.7 kg/day); there were14 lines did better than Ciherang (5.07 t/ha; 41.9 kg/day), and 13 lines yielded better than did Inpari 13 (5.27 t/ha; 46.7 kg/day). The best seven lines and lines with higher productivity per day than that of the best check, with an average yield of 5.62 t/ha up to 6.12 t/ha, with days to maturity from 87 up to 94 das, were ready to be evaluated in Multi Locational Yield Trial to meet the requirement for the release of new variety.

O povo Parkatêjê vive no sudeste do estado do Pará e fala uma língua timbira pertencente ao agrupamento linguístico Macro-Jê. Suas narrativas têm como função principal a acumulação, o armazenamento e a transmissão de conhecimentos. Por meio delas, o discurso é expresso linguisticamente. Desse modo, o presente trabalho, com fundamentação na metodologia da linguística descritiva, na linguística textual e em estudos de textos orais, visa abordar questões sobre a estrutura formal e discursiva da narrativa selecionada.

Abstract New bismuth(III) bromine compounds of the heterocyclic thioamides were prepared and structurally characterized. The reaction of heterocyclic thioamides with bismuth(III) bromide resulted in the formation of the {[BiBr2(μ2-Br)(MMI)2]2·CH3COCH3·H2O} (1), {[BiBr2(MBZIM)4]·Br·2H2O} (2), {[BiBr2(μ2-Br)(tHPMT)2]2·CH3CN} (3), {[BiBr2(μ2-Br)(PYT)2]2·CH3CN} (4) and {[BiBr2(μ2-Br)(MBZT)2]2 2CH3OH} (5) complexes (MMI: 2-mercapto-1-methylimidazole, MBZIM: 2-mercaptobenzimidazole, tHPMT: 2-mercapto-3,4,5,6-tetrahydro-pyrimidine, PYT: 2-mercaptopyridine and MBZT: 2-mercaptobenzothiazole). The complexes 1–5 were characterized by melting point (m.p.), elemental analysis (e.a.), molar conductivity, Fourier-transform infrared (FT-IR), Fourier-transform Raman (FT-Raman), nuclear magnetic resonance (1H and 13CNMR) spectroscopy, UV-Vis spectroscopy and thermogravimetric-differential thermal analysis (TG-DTA). The molecular structures of 1–5 were determined by single-crystal X-ray diffraction. Complex 2 is a first ionic monomuclear octahedral bismuth(III) bromide, while the complexes 1, 3–5 are the first examples of dinuclear bismuth(III) bromide derivatives. Complexes 1–5 were evaluated in terms of their in vitro cytotoxic activity against human adenocarcinoma breast (MCF-7) and cervix (HeLa) cells. The toxicity on normal human fetal lung fibroblast cells (MRC-5) was also evaluated. Moreover, the complexes 1–5 and free heterocyclic thioamide ligands were studied upon the catalytic peroxidation of the linoleic acid by the enzyme lipoxygenase (LOX).

Titanatos de chumbo dopados com itérbio e ítrio, PYbT e PYT foram preparados pelo método Pechini. Os materiais obtidos foram calcinados sobre fluxo de oxigênio em diferentes temperaturas e investigados utilizando-se as técnicas de difração de raios X (DRX), microscopia eletrônica de varredura (MEV) e espectroscopia na região do infravermelho (IV). O comportamento térmico de PbTiO3 dopados com itérbio e ítrio é descrito através de curvas termogravimétricas (TG) e análise térmica diferencial (ATD). Os resultados revelaram que a fase perovskita foi obtida em uma baixa temperatura e que o aumento da dopagem favorece a estabilidade térmica dos materiais obtidos.

La Tomographie par Emission de Positrons (TEP) est une technique d’imagerie médicale utilisée largement dans le traitement du cancer et dans la recherche neurobiologique, afin d’imager l’activité biologique des organes. Il s’agit de détecter deux photons de 511 keV produits par l’annihilation d’un positron dans les tissus, ce qui permet d’en reconstruire la carte 3D. En mesurant avec une très bonne précision la différence de temps de détection des deux photons, il sera possible d’améliorer la qualité d’image (technique du temps de vol). Dans ce manuscrit, nous présentons le développement de deux détecteurs innovants, rapides et efficaces, pour la détection de la lumière Cherenkov produite par la conversion des photons de 511 keV. Le premier, destiné à un scanner clinique (cerveau) et pré-clinique à haute précision spatiale, utilise comme milieu de détection du TriMéthylBismuth. Le second, pouvant être utilisé pour construire un scanner corps entier, met en œuvre un cristal de PbF₂ comme radiateur Cherenkov. Dans les deux configurations, un photomultiplicateur à micro-canaux (MCP-PMT) est utilisé pour détecter les photons Cherenkov. Notre électronique de détection montre une résolution temporelle limitée à 5 ps (RMS). La chaîne de détection est limitée par les performances du MCP-PMT. Après étalonnage, nous avons mesuré une efficacité de 25 % (grande pour un détecteur Cherenkov), et de résolution temporelle de 200 ps (FWHM).Nous exposons les facteurs limitant la résolution temporelle des détecteurs et proposons des développements qui permettront d’en améliorer les performances
Positron Emission Tomography (PET) is a nuclear imaging technique widely used in oncology and neuroscience to observe biological activity in the body. Detection of two gamma quanta with the energy 511 keV emitted by positron annihilation in tissues allows one to reconstruct the tracer activity distribution in the body of the patient. Additional measurement of the difference in time detection between the two photons lets us to improve significantly the quality of the reconstructed image (time-of-flight method).In this manuscript, we present the development of two innovative detectors, fast and efficient, used to detect Cherenkov light produced by electrons from the photo-ionization conversions of 511 keV gamma quanta. The first one, intended for use in a brain PET scanner of a high spatial resolution, uses TriMethylBismuth for the detection medium. The second one, planned to be used to construct a whole-body PET scanner, enforces a PbF₂ crystal as Cherenkov radiator. In both configurations a micro-channel photo-multiplier (MCP-PMT) is used to detect Cherenkov photons. We commissioned an electronic detection chain with a time resolution limited to 5 ps (RMS). Using precise MCP-PMT calibration, we were able to develop simultaneously detectors with high efficiency, up to 25 %, and time resolution as good as 200 ps (FWHM).We highlight the limitations of detectors time resolution and suggest several developments in order to improve performances of Cherenkov light detectors

[ES] El crecimiento de las plantas se ve afectado por el estrés abiótico, sequía, salinidad o altas temperaturas. La transducción de señales de estrés abiótico es fundamental para generar una respuesta fisiológica adecuada, que implica la participación de diferentes hormonas vegetales, siendo el ácido abscísico (ABA) el regulador hormonal crítico en la regulación de la respuesta de la planta a situaciones de estrés por déficit hídrico. La vía de señalización de ABA y los componentes principales están bien caracterizados molecular y bioquímicamente. Los receptores de ABA "Pyrabactin Resistance 1"(PYR)/"PYR1-LIKE" (PYL)/ "Regulatory Component of ABA Receptor" (RCAR) juegan un papel importante en la regulación cuantitativa de la señalización ABA tanto en semillas como en tejidos vegetativos. Aunque la función bioquímica de los receptores PYR/PYL/RCARs de ABA, está bien caracterizada, se conoce poco sobre otros aspectos con relevancia biológica, como sus modificaciones postraduccionales o la regulación de su vida media. Uno de los avances recientes en este campo ha sido el descubrimiento de una nueva familia de E3 ligasas llamadas RSL1/RFAs ("RING-finger-ABA-related") que consta de al menos 10 miembros, reguladores clave de la estabilidad de los receptores PYR/PYL/RCAR de ABA en tejidos de raíces y hojas, regulando su degradación en diferentes ubicaciones celulares. Un estudio detallado de esta familia génica reveló que RSL1/RFA se caracterizan estructuralmente por la presencia de tres dominios RING putativos en tándem, denominados "RING1-IN BETWEEN RING-RING2" (RBR), y en consecuencia pertenecen a la familia de E3 ligasas de tipo RBR. Cinco miembros de la familia RSL1/RFA, RSL1 y RFA6-RFA9, contienen un dominio TM en el extremo C-terminal, lo que sugiere que RFA6-RFA9 también se localizan en la membrana plasmática. Sin embargo, otros miembros de las E3 ligasas como RFA1-RFA5 carecen del dominio TM C-terminal y su caracterización funcional, así como su ubicación celular, aún no se conocen. Nosotros mostramos que la E3 ligasa RFA1 se localiza en núcleo y citosol, mientras que RFA4 muestra una localización específica en el núcleo promoviendo la degradación nuclear de los receptores ABA. Por lo tanto, los miembros de la familia RSL1/RFA interactúan con los receptores ABA en la membrana plasmática, el citosol y el núcleo, dirigiéndolos a su degradación a través de la vía endosomal/vacuolar (en el caso de RSL1) o el proteosoma 26S (para RFA1 y RFA4). Proporcionamos información sobre la función fisiológica de estas E3 ligasas de tipo RBR. Realizando tanto mutagénesis como ensayos bioquímicos para identificar la cisteína 361 (Cys361) en RFA4 como la Cys del sitio activo, que es una característica distintiva de las E3 ligasas de tipo RBR. Demostramos mediante análisis de inmunotransferencia del mutante con pérdida de función de rfa1rfa4 que los niveles endógenos de los receptores de ABA PYR1 y PYL4 aumentan en comparación con las plantas de tipo silvestre. Hemos identificado una enzima E2, "Ubiquitin Conjugating Enzyme 26" (UBC26), como la enzima nuclear canónica E2 que interactúa con la E3 ligasa RFA4 y forma complejos UBC26-RFA4-Receptor, formando agregados nucleares. Generamos alelos ubc26 con pérdida de función que mostraban una mayor sensibilidad a ABA y acumulación de receptores ABA en comparación con el tipo silvestre. En definitiva, hemos revelado un sofisticado sistema de ubiquitinación de receptores ABA en diferentes ubicaciones subcelulares llevado a cabo a través de la familia de E3 ligasas RSL1/RFA de tipo RBR. Por otro lado, hemos iniciado pruebas bioquímicas para identificar la S-acilación en el dominio TM de RSL1. [CA] El creixement de les plantes es pot veure afectat per l'estrès abiòtic, sequera, salinitat o altes temperatures. La transducció de senyals d'estrès abiòtic és fonamental per a generar una resposta fisiològica adequada, que implica la participació de diferents hormones vegetals, sent l'àcid abscísic (ABA) el regulador hormonal crític en la regulació de la resposta de la planta a situacions d'estrès per dèficit hídric. La ruta de senyalització d'ABA i els components principals de la ruta estan ben caracteritzats molecularment i bioquímica. Els receptors "Pyrabactin Resistance 1"(PYR)/"PYR1-LIKE"(PYL)/"Regulatory Component of ABA Receptor" (RCAR) exerceixen un paper important en la regulació quantitativa en resposta a l'estrès tant en llavors com en planta. Encara que la funció bioquímica dels receptors PYR/PYL/RCARs d'ABA, està ben caracteritzada en els últims anys, es coneix poc sobre altres aspectes amb rellevància biològica, com les seues modificacions postraduccionals o la regulació de la seua vida mitjana. Un dels avanços recents en aquest camp ha sigut el descobriment d'una nova família d'E3 ligases anomenades RSL1/RFAs ("RING-finger-ABA-related") que consta d'almenys 10 membres, que són reguladors clau de l'estabilitat dels receptors PYR/PYL/RCAR d'ABA en teixits d'arrels i fulles, regulant la seua degradació en diferents ubicacions cel·lulars. Un estudi més detallat d'aquesta família gènica va revelar que RSL1/RFAs es caracteritzen estructuralment per la presència de tres dominis RING putatius en tàndem, denominats "RING1-IN BETWEEN RING-RING2" (RBR), i en conseqüència pertanyen a la família d'E3 ligases de tipus RBR. Cinc membres de la família RSL1/RFA, RSL1 i RFA6-RFA9, contenen un domini TM en l'extrem C-terminal, la qual cosa suggereix que RFA6-RFA9 també es localitzen en la membrana plasmàtica. No obstant això, altres membres d'aquesta família d'E3 ligases com RFA1-RFA5 manquen del domini TM C-terminal i la seua caracterització funcional, així com la seua ubicació cel·lular, encara no ha sigut investigada. Vam mostrar que l'E3 ligasa RFA1 es localitza tant en el nucli com en el citosol, mentre que RFA4 mostra una localització específica en el nucli promovent la degradació nuclear dels receptors ABA. Per tant, els membres de la família RSL1/RFA interactuen amb els receptors ABA en la membrana plasmàtica, el citosol i el nucli, dirigint-los a la seua degradació a través de la vía endosomal/vacuolar (en el cas de RSL1) o el proteosoma 26S (per a RFA1 i RFA4). Proporcionem informació sobre la funció fisiològica d'aquestes E3 ligases de tipus RBR. Realitzant tant mutagènesis com a assajos bioquímics per a identificar la cisteïna 361 (Cys361) en RFA4 com la Cys del lloc actiu, que és una característica distintiva de les E3 ligases de tipus RBR. Hem demostrat mitjançant una anàlisi d'immuno-transferència del mutant amb pèrdua de funció de rfa1rfa4 que els nivells endògens dels receptors d'ABA PYR1 i PYL4 augmenten en comparació amb les plantes de tipus silvestre. D'altra banda, hem identificat un enzim E2, "Ubiquitin Conjugating Enzyme 26" (UBC26), com l'enzim nuclear canònic E2 que interactua amb l'E3 ligasa RFA4 i forma complexos UBC26-RFA4-Receptor, formant agregats nuclears. També generem al·lels ubc26 amb pèrdua de funció que mostraven una major sensibilitat a ABA i acumulació de receptors ABA en comparació amb el tipus silvestre. [EN] Plant growth is affected by abiotic stress, drought, salinity or high temperature. Signal transduction of abiotic stress is crucial to generate an appropriated physiological response, which involves the participation of different plant hormones, being abscisic acid (ABA) the critical hormonal regulator in regulating the plant's response to situations of stress due to water deficit. The ABA signaling pathway and the major components of the pathway are well characterized molecularly and biochemically. Pyrabactin Resistance 1 (PYR)/PYR1-LIKE (PYL)/Regulatory Component of ABA Receptor (RCAR) ABA receptors play an important role in quantitative regulation of ABA signaling both in seeds and vegetative tissues. Although the biochemical function of the PYR/PYL/RCAR ABA receptors has been well established in recent years, little is known about other aspects with biological relevance, such as their post-translational modifications or the regulation of their half-life. One of the recent advances in this field has been the discovery of a new family of E3 ligases called RSL1/RFAs (RING-finger-ABA-related) that consists of at least 10 members, which are key regulators of the stability of PYR/PYL/RCARs in root and leaf tissues, and regulate the degradation of ABA receptors at different cellular locations. Further inspection of the gene family revealed that RSL1/RFAs are structurally characterized by the presence of three putative RING domains in tandem, named as RING1-IN BETWEEN RING (IBR)-RING2, and accordingly they belong to the RBR-type E3 ligase family. Five members of the RSL1/RFA family, that is, RSL1 and RFA6-RFA9, contain a TM domain at the C-terminal end of the proteins, which suggests that RFA6-RFA9 are also localized in plasma membrane. However, other members of this family of E3 ligases such as RFA1-RFA5 lack the C-terminal TM domain and their functional characterization, as well as their cellular location, has not been investigated yet. In this study we show that the E3 ligase RFA1 is localized both in the nucleus and in the cytosol, while RFA4 shows a specific localization in the nucleus promoting the nuclear degradation of ABA receptors. Therefore, we members of the RSL1/RFA family interact with ABA receptors at the plasma membrane, cytosol and nucleus, targeting them for degradation via the endosomal/vacuolar pathway (in the case of RSL1) or the 26S- proteasome (for RFA1 and RFA4). We provide information on the physiological function of these RBR-type E3 ligases, which are hardly explored in plants. Additionally, we performed mutagenesis and biochemical assays to identify Cys361 in RFA4 as the active site cysteine, which is a distinctive feature of RBR-type E3 ligases. We have shown by immunoblot analysis of the rfa1rfa4 loss-of-function mutant that endogenous levels of ABA receptors PYR1 and PYL4 are increased compared to wild-type plants. On the other hand, we have identified an E2 enzyme, Ubiquitin Conjugating Enzyme 26 (UBC26), as the canonical nuclear enzyme E2 that interacts with the E3 ligase RFA4 and forms UBC26-RFA4-Receptor complexes, forming nuclear aggregates. We also generated loss-of function ubc26 alleles that exhibited higher sensitivity to ABA and accumulation of ABA receptors compared to wild type. We have revealed a sophisticated ubiquitination system of ABA receptors in different subcellular locations carried out through the RBR-type RSL1/RFA family of E3 ligases. We have proceeded with the biochemical and genetic study of the different members of the family. We have started biochemical tests to identify the S-acylation in the TM domain of RSL1. To this end, we have generated RSL1C334S, RSL1 C5S and RSL1C6S by mutagenesis as well as RSL1ΔTM, a deletion of the TM domain. Initial studies have shown that Cys residues close to the TM domain are S-acylated. Finally, we have also generated new combined mutants: rsl1rfa1, rsl1rfa5, rfa1rfa5 and rsl1rfa1rfa5.

La sequía y la salinidad representan estreses ambientales que afectan de forma crítica el crecimiento de las plantas y limitan enormemente su potencial agrícola. La fitohormona ácido abcísico (ABA) juega un papel fundamental en la coordinación de la respuesta y adaptación de las plantas a este tipo de estreses, así como en la regulación del crecimiento y desarrollo vegetal. Elementos intermediarios de la ruta de señalización ya habían sido caracterizados, pero aún se desconocía el mecanismo de percepción y transducción de señal de la hormona. Este trabajo de tesis ha contribuido a la caracterización de una nueva familia de receptores intracelulares de la hormona ABA, formada por 14 miembros y denominada PYR/PYL (de pyrabactin resistance / PYR1-like) /RCAR (de Regulatory Component of Abscisic acid Receptor), y a su caracterización estructural y bioquímica. Estas proteínas son capaces de unir de forma específica la hormona ABA. La unión de la hormona induce en estos receptores un cambio conformacional, que les permite regular la actividad de los reguladores negativos de la ruta: fosfatasas del grupo A como ABI1, ABI2 o HAB1 ( Leung et al., 1994; Meyer et al.,1994; Saez et al., 2004). Para la caracterización de estos receptores se han llevado a cabo abordajes genéticos, bioquímicos, de calorimetría y estudios estructurales.

To establish accurate and early stage tumour diagnosis, quantitative molecular imaging modalities such as positron emission tomography (PET) are being employed. Molecular probes containing a positron emitting isotope (e.g. ¹⁸F, ⁶⁴Cu) are designed to target biomarkers or metabolic processes within the tumour. A therapeutic molecule could then be conjugated to this diagnostic probe to create a multifunctional construct (MFC). MFCs are a new concept in drug design offering the potential for both diagnosis and therapy. This work exploits the chelating properties of saturated tetraazamacrocycles known herein as macrocycles as well as their known affinity for the chemokine receptor CXCR4, a cell surface receptor involved in embryonic development and cell migration known to be overexpressed in at least twenty three different tumour types, along with the photodynamic activity of porphyrin molecules to create novel MFCs with diagnostic and therapeutic properties. A series of configurationally restricted copper(II), zinc(II) and nickel(II) mono-ring macrocycles bearing a methylamine pendant arm are presented. Monomacrocycles possess the potential for improved pharmacological properties. Biological studies assessing the affinity and potency of these macrocycles as CXCR4 antagonists demonstrated that zinc(II) monomacrocycles were the most potent in their class. A side bridged (SB) zinc(II) monocyclen compound; [Zn9]²⁺ synthesised in 64% yield, demonstrated the highest affinity for CXCR4 in its class showing a 2.5-fold higher affinity in comparison to the clinically licensed antagonist AMD3100 (IC₅₀ = 12 nM vs. 31 nM). In response to the high affinity observed for a meta-substituted biscyclen ligand (L48) a novel cross bridged (CB) meta-substituted biscyclen compound bearing a methylamine pendant arm 20 was synthesised along with its copper(II), nickel(II) and zinc(II) complexes in 77%, 88% and 90% yields, respectively. These metal complexes displayed high affinity for CXCR4 when competed with a CXCR4 specific antibody (% inhibition >85%) and required sub-micromolar concentrations to reduce the ‘normal’ signalling of CXCR4 by 50%. [Cu₂20]⁴⁺ has the potential to be a targeted PET probe through the use of ⁶⁴Cu and so a detailed biological evaluation into its binding mode was undertaken. These assays concluded that [Cu₂20]⁴⁺ exerts its antagonistic effect by locking the receptor on the surface of the cell, stopping the internalisation and recycling processes, which would result in formation of a new receptor. An analogous meta-substituted biscyclam ligand (28) and its copper(II), zinc(II) and nickel(II) complexes were synthesised in 74%, 58% and 46% yields, respectively. These metal complexes also demonstrated high affinity for CXCR4 (IC₅₀ < 0.1 μM). In fact [Cu₂28]⁴+ showed unexpectedly high affinity for CXCR4 demonstrating almost a 3-fold higher affinity than AMD3100 (IC₅₀ = 11nM vs. 31 nM), the potential of [Cu₂28]⁴⁺ as a targeted PET probe should also be considered. Various organic spacer arms were attached to bismacrocycle 20, to produce a CXCR4 targeting probe with a functional arm for further conjugation which would also act to prevent steric hindrance. An acidic terminating chain was successfully attached to bismacrocycle 20 in quantitative yield, via ring-opening of diglycolic anhydride as well as an amine terminating variant (93% yield). A biotin tag was also attached to bimacrocycle 20 and its copper(II) complex was successfully isolated in a 94% yield. Addition of a spacer arm was found to have no significantly detrimental effect on binding affinity to CXCR4. A series of known water and organic soluble metalloporphyrins were synthesised. Subsequent conjugation to macrocyclic compounds proved challenging but analytical data for the conjugation of bismacrocycle 20 and metalloporphyrin [Zn59] indicated a successful reaction had occurred. This reaction represents important steps towards the synthesis of MFCs with diagnostic and therapeutic properties.

Abstract The general goal of this thesis is the development of new generation photoactivable smart nanomaterials that can be modulated into tremendous way to achieve multimodal anticancer and antimicrobial therapeutic actions. Here the cyclodextrins act as the building block for the development of the nanocarrier in order to convey the therapeutic payload to the desired site. With this goal in mind, we have developed several kinds of nanosystems and are discussed in each chapter. The entire dissertation is divided into four sections based on the types of materials and the sections are further divided into twelve chapters based on the functional behavior of each nanosystems.

Resumo: Pesquisas no campo da tecnologia têm buscado avaliar técnicas e/ou processos econômicos e eficientes para o tratamento de efluentes que contenham metal pesado. Atualmente a bioadsorção vem se apresentando como uma boa alternativa às tecnologias existentes. Este trabalho teve como objetivo a investigação da macrófita Azolla pinnata como bioadsorvente de cinco íons metálicos: Cd2+, Cu2+, Ni2+, Pb2+ e Zn2+ em soluções sintéticas. A caracterização do biomaterial foi realizada utilizando-se técnicas como Microscopia Eletrônica de Varredura (MEV) para análise da superfície do material; análise de Espectroscopia de Energia Dispersiva de Raios-X (EDX), que mostrou que qualitativamente a diminuição de íons de competição (Ca, Na e K) após o processo de adsorção, evidenciando que a troca iônica pode ser um dos mecanismos do processo; e análise de Infravermelho por Transformada de Fourier (FTIR), em que se pôde constatar que alguns grupos funcionais como carboxilas e grupos sulfonados são fundamentais no processo. A cinética de adsorção foi ajustada pelos modelos de pseudoprimeira e pseudossegunda ordem, sendo que o melhor ajuste variou de acordo com o íon estudado. O modelo de difusão intrapartícula mostrou que muitos mecanismos podem ser limitantes na taxa com a qual ocorre o processo. Os dados de equilíbrio foi ajustado pelos modelos de isoterma de Langmuir e de Freundlich em duas temperaturas, 25 e 45 ºC. O processo foi favorecido pelo acréscimo de temperatura para todos os íons. O modelo de Langmuir se ajustou melhor aos dados experimentais de todos os íons e o valor de capacidade máxima de adsorção (qmax) a 25 ºC foi de 0,359; 0,448; 0,254; 0,472 e 0,357 mmol/g para Cd2+, Cu2+, Ni2+, Pb2+ e Zn2+, respectivamente. Os eluentes HCl, EDTA e CaCl2 foram testados no processo de dessorção em batelada e o cloreto de cálcio se mostrou eficiente além de não causar danos ao material nem diminuir a capacidade de adsorção da macrófita. Ensaios dinâmicos em coluna de leito fixo foram realiados para os íons Cu2+ e Pb2+ com soluções mono e bicomponentes. Nos ensaios em sistema binários, pode-se constatar uma grande preferência pela adsorção de íons chumbo e, portanto, uma grande queda na adsorção de cobre. Foram testados ciclos de adsorção/dessorção em coluna utilizando o CaCl2 como eluente e, em três ciclos, a macrófita manteve suas propriedades adsortivas. Com estes resultados, pode-se constatar que a realização deste trabalho contribuiu para a avaliação deste material como bioadsorvente alternativo e que, especificamente, macrófita A. pinnata mostrou-se atrativa para utilização nos processos de tratamento e/ou estabilização de metais pesados presentes em baixas concentrações em efluentes industriais
Resumo: No processo Kraft para produção de papel e celulose, os cavacos de madeira são cozidos em um digestor com uma solução aquosa denominada licor branco, constituída de hidróxido de sódio (NaOH) e sulfeto de sódio (Na2S). Durante o cozimento uma pasta fibrosa é produzida. Esta pasta é filtrada e segue para o branqueamento. O produto filtrado chama-se licor negro, devido à mudança da sua cor nesta etapa. O licor negro possui aproximadamente 15 % de sólidos em massa. Ele é bombeado para a unidade de recuperação, na qual ele será concentrado em evaporadores de múltiplosefeitos até 75 % de sólidos em massa. Em seguida, ele é bombeado para uma caldeira onde é queimado, para recuperar os sais inorgânicos e, principalmente, fornecer calor para geração de calor. Durante a evaporação do licor negro, quando a porcentagem em sólidos ultrapassa 50 %, ocorre a precipitação de sais inorgânicos. Estes sais se depositam nas superfícies de troca térmica dos evaporadores diminuindo sua eficiência e causando prejuízos à indústria. Os principais sais que causam este problema são o carbonato de sódio (Na2CO3) e o sulfato de sódio (Na2SO4). O objetivo do presente trabalho foi testar e validar um método experimental para a determinação da quantidade de Na2CO3 presente em amostras industriais de licor negro. Além disso, também foram analisadas algumas propriedades físico-químicas do licor negro, como massa específica, teor de sólidos e viscosidade. Com os resultados obtidos, tentou-se obter uma correlação entre o teor de carbonato de sódio e as demais propriedades do licor que foram determinadas. Os resultados das análises das características físico-químicas demonstraram coerência e boa reprodutibilidade, porém, os resultados das análises de teor de carbonato de sódio não foram satisfatórios e mostraram que a técnica deve ser aperfeiçoada
Abstract: Research in the technology field has been looking for evaluating efficient and economic techniques and/or process for effluents that contains heavy metal. The biodsorption process has been seemed as a good alternative to the existing technology. This work had the aim to investigate the use of Azolla pinnata macrophyte as biosorbent of five metallic ions: Cd2+, Cu2+, Ni2+, Pb2+ and Zn2+ in synthetic solutions. The biomaterial characterization was carried out by techniques as scanning electron microscopy (SEM) for the analyze of material superficies; the energy dispersive X-ray spectroscopy (EDX), which showed, qualitatively, a decreasing in the competitions ions (Ca, Na e K) after the sorption process, indicating that the ion exchange can be one o the process mechanism; and Fourier Transform Infrared Spectroscopy (FTIR), in which it was possible implied functional groups as carboxyl and sulfonate play a important role in the adsorption. The kinetics was adjusted by primer and second order models and the model which fit better depends on each ion. The intra-particle diffusion model showed that several mechanisms may be limiting to the process rate. The equilibrium data were adjusted by Langmuir and Freundlich models in two temperatures: 25 and 45 ºC. The process was favored by the increasing in the temperature for all five ions. The Langmuir model fit better the experimental data and the maximum adsorption capacity (qmax) were 0,359; 0,448; 0,254; 0,472 and 0,357 mmol/g for Cd2+, Cu2+, Ni2+, Pb2+ and Zn2+, respectively. The HCl, EDTA and CaCl2 eluents were tested in the desorption process and the calcium chloride was efficient and did not cause any damage in the biomaterial or decreasing the biosorption capacity. Dynamics runs were carried out in fixed-bed in mono and bi-components solutions. In the binary were found a great preference for lead instead of copper and as a result, a significant decrease in the removal of copper. Adsorption/desorption cycles were tested in fixed-bed system using CaCl2 as eluent. After three cycles no damage, weight lost or significant decrease in the biosorption capacity was noticed. These results indicating that the A.pinnata macrophyte is an attractive potential new biosorbent to be used in heavy metal removal especially in low concentrations effluents
Abstract: In the Kraft process for pulp and paper production, wood chips are cooked in a digester with an aqueous solution named white liquor, consisted by sodium hydroxide (NaOH) and sodium sulfide (Na2S). During this cooking process, wood chips are transformed generating a fibrous paste and a liquid solution. This paste is filtered and separated from the liquid solution, named black liquor due to its color. The paste goes to the bleaching stage to produce cellulose. Black liquor contains inorganic and organic (mainly lignin) compounds from wood. Initially this black liquor contains approximately 15 mass/% of solids and is sent to the recovery unit to have this concentration raised to approximately 75 mass/% to be burned in a recovery boiler in order to generate power and recover inorganic reactants, recycling them to the process. The concentration process of black liquor occurs in a multiple effect evaporators unit. During this process, when the concentration of solids exceeds 50%, the precipitation of inorganic salts (mainly Na2CO3 and Na2SO4) begins. These salts are deposited in the heat transfer surfaces of the evaporator tubes, reducing their efficiency, leading to a reduction in the production capacity. The scope of this work was testing and validate an experimental method to estimate the content of Na2CO3 present in industrial black liquor samples. Moreover, some black liquor's physical-chemical properties as density, solids content and viscosity were also analyzed. An attempt to develop a predictive model to evaluate sodium carbonate content from the other properties was made. The results obtained for physical-chemical properties have shown a good consistency. However, the results of sodium carbonate analyses were not satisfactory, showing that the technique must be improved

Este trabajo de investigación nace por el amor a nuestras mascotas, el querer darles una mejor calidad de vida y la necesidad que tenemos los dueños de mascotas de tener una atención inmediata y de calidad para nuestros amigos peludos cuando pase alguna emergencia. De esa necesidad es que nace el seguro para mascotas SAFE PET, que ofrece un paquete estándar que incluyan varios beneficios y coberturas para las mascotas afiliadas. La principal fuente de ingreso de SAFE PET será la venta de nuestras pólizas, nuestra inversión inicial será de s/ 124,890 que la financiaremos el 60% los 4 socios de la empresa y 40% con un préstamo a una entidad financiera. Por las investigaciones que realizamos mediante encuestas a dueños de mascotas, estos en su mayoría nos manifestaba el querer tener un seguro para su mascota, y luego gracias nuestra landing page pudimos observar que del 100% de visitas a nuestra landing el 54.6% dejó sus datos porque le interesa nuestro seguro, este porcentaje es mucho mayor al que esperábamos y eso nos alienta mucho más a ser uno de los primero en el país en dedicarnos exclusivamente en seguro integral de mascotas. Finalmente, luego del estudio financiero realizado a nuestra empresa hemos considerado en el análisis de sensibilidad COK (89.71%) y un costo promedio ponderado de capital de WACC (74.53%), para terminar después de todo nuestro plan financiero podemos concluir que SAFE PET, es una empresa viable económicamente.
This research was born for our love to pets and we want to give them a better life quality and the need that pet owners to have immediate and quality care for our furry friends when an emergency happens. From that need is born SAFE PET, that is a pet insurance, which offers a standard package that includes many benefits and coverages for affiliated pets. The main source of income for SAFE PET will be the sale of our policies, our initial investment will be S/ 124,890 which will be financed by 60% the 4 partners of the company and 40% with a loan to a financial institution. Due to the research we conducted through surveys of pet owners, most of them told us that they wanted to have insurance for their pet, and then thanks to our landing page we could observe that of 100% of visits to our landing, 54.6% left their data Because you are interested in our insurance, that percentage is much higher than we expected and that encourages us much more to be one of the first in the country to dedicate ourselves exclusively to comprehensive pet insurance. Finally, after the financial study carried out on our company, we have considered in the sensitivity analysis COK (89.71%) and a weighted average cost of capital of WACC (74.53%), to finish after all our financial plan we can conclude that SAFE PET, it is an economically viable company.

El presente trabajo plantea la puesta en marcha de un negocio innovador, para ello, se ha considerado los factores externos e internos que podrían repercutir en el desarrollo del mismo. Asimismo, se ha evaluado el contexto actual y se ha elaborado el respectivo estudio que sustenta la viabilidad del proyecto. Hoy en día las personas se preocupan más por la seguridad de sus mascotas, existe una tendencia petfriendly que ha llevado a la sociedad a incrementar el consumo de alimentos balanceados, gastos en veterinarias y spas caninos. Considerando que no existe en la actualidad un negocio similar al planteado, los alumnos autores del presente trabajo han considerado que la creación de un seguro para mascotas en el Perú sería altamente rentable. Para sustentar la idea expuesta, se ha tomado en consideración todos los aspectos concernientes a la creación de la persona jurídica, los objetivos estratégicos, políticas internas y externas, análisis financieros y la rentabilidad del proyecto. Después de analizar los aspectos antes mencionados, y de considerar todos los aspectos que puedan influir en su desarrollo, se llega a la conclusión de que el negocio es viable.
This work proposes the start-up of an innovative business, for this, the external and internal factors that could have an impact on its development have been considered. Likewise, the current context has been evaluated and the respective study that supports the viability of the project has been prepared. Nowadays people care more about the safety of their pets, there is a petfriendly trend that has led society to increase consumption of feed, veterinary expenses and canine spas. Considering that there is currently no business similar to that proposed, the student authors of the present work have considered that the creation of pet insurance in Peru would be highly profitable. To support the idea presented, all aspects concerning the creation of the legal entity, strategic objectives, internal and external policies, financial analysis and the profitability of the project have been taken into consideration. After analyzing the aforementioned aspects, and considering all aspects that may influence its development, it is concluded that the business is viable.

Abstract Ferro-electric thin film materials have been used for nonvolatile random access memories(FRAM*). PZT(Pb(Ti,Zr)O3) is one of several promising materials for use as a FRAM capacitor in combination with a platinum film as an electrode. However the ferroelectric properties of this capacitor are degraded by commonly used LSI processes, such as a passivation/ insulator(CVD-SiO2) process. Recently, mechanisms of process degradation are being investigated**,***, but are not clearly understood yet. For example, to our knowledge the existence of hydrogen in the capacitor after an annealing in hydrogenous atmosphere has not yet been measured by analytical methods. As a result in this study, we measured electrical properties of Pt/PZT/Pt capacitors after a 7.6 torr N2-H2 annealing at various hydrogen partial pressures and temperatures. Then we measured composition profiles of these capacitors by Secondary Ion Mass Spectrometry(SIMS). We found that after the N2-H2 annealing, the non-volatile polarization(Pnv)(uC/cm2) of the capacitors rapidly decreased under the presence of hydrogen at lower temperatures. At the same time, SIMS profiles showed that hydrogen concentration increased in the PZT film, and oxygen concentration decreased in the upper Pt film. We considered that the increase of hydrogen in the PZT film causes the degradation of electric properties, and the decrease of oxygen in the Pt film makes the adhesion force between the upper platinum electrode and the PZT film weaker. Moreover we found that the degradation can be eliminated by annealing in oxygen at temperatures as high as 400 °C.

Crystallographically engineered Relaxor-PT single crystals, specifically PMN-PT (Generation I) and PIN-PMN-PT/PMN-PZT (Generation II), offer much higher piezoelectric and electromechanical coupling coefficients (d33 > 1,500 pC/N, k33 > 0.9), when compared to polycrystalline PZT ceramics. Recently Ceracomp Co., Ltd. (www.ceracomp.com) has developed the solid-state single crystal growth (SSCG) technique and successfully fabricated Gen III PMN-PZT single crystals modified with acceptors or donors [Fig. 1]. The piezoelectric constants (d33) of (001) Gen III PMN-PZT single crystals were measured to be higher than 4,000 pC/N and thus about two times higher than those of PMN-PT/PZN-PT (Gen I) and PIN-PMN-PT/PMN-PZT (Gen II) single crystals. The Gen III PMN-PZT single crystals have been firstly applied to single crystal-epoxy composites, ultrasonic transducers, piezoelectric sensors, and piezoelectric actuators [Fig. 2]. In this presentation, we will introduce the development of high performance piezoelectric sensors and actuators by using the Gen III PMN-PZT single crystals for SHM (structural health monitoring) application.

Vibration-to-electricity conversion using piezoelectric transduction has been studied by several researchers over the last decade. PZT (lead zirconate titanate) - based piezoelectric ceramics such as PZT-5A and PZT-5H have been very frequently employed in design of piezoelectric energy harvester beams. Recently, the single-crystal piezoceramics PMN-PT (lead magnesium niobate – lead titanate) and PMN-PZT (lead magnesium niobate – lead zirconate titanate) have also been investigated for electrical power generation due to their large piezoelectric constants (particularly the d31 constant for the bending mode). Piezoelectric, elastic and dielectric properties of these piezoceramics differ from each other considerably. Even though the d31 constants of two piezoceramics might differ by an order of magnitude (e.g. PZT-5A and PMN-PZT), this large difference is not necessarily the case for their power outputs. It is theoretically discussed and experimentally demonstrated in this paper that the d31 piezoelectric constant alone is an insufficient parameter for selecting the best piezoelectric material to design a power generator for vibration-based energy harvesting. Elastic compliance of a piezoceramic has a strong effect on its electrical power output. In addition, since these devices are usually designed for resonance excitation, mechanical damping constitutes another parameter that might change the entire picture regarding the power generation performance. The last one is particularly critical considering the fact that it is difficult to control mechanical damping due to clamped interfaces and adhesive layers in practice. Theoretical comparisons are given for geometrically identical bimorphs with PZT-5A, PZT-5H, PMN-PT (with 30% PT), PMN-PT (with 33% PT) and PMN-PZT layers using an experimentally validated distributed-parameter electromechanical model. Two experimental demonstrations are presented. The first case compares two geometrically identical bimorphs (using PZT-5A and PZT-5H piezoceramics) and shows that the bimorph with PZT-5A can generate larger power than the one with PZT-5H in spite of the larger d31 constant of the latter. The second experimental case compares the power generation performances of a PZT-5H unimorph and a PMN-PZT unimorph. In agreement with the theory, considerably large damping identified for the PMN-PZT unimorph results in much lower power output compared to that of the PZT-5H unimorph.
