Résumé :
The spinocerebellar ataxias are dominantly-inherited neurodegenerative disorders whose major clinical feature is incoordination. Although 32 have been described to date, those characterized by (CAG)n repeat expansions resulting in elongated polyglutamine tracts in their respective host proteins (SCAs 1, 2, 3, 6, 7, 17, and in part 8) are the most common and have been subject to the most detailed investigation of their pathogenic mechanisms. All are characterized by polyglutamine tract aggregates, toxicity of which was initially thought to be their pathogenic mechanism. However, recent research has emphasised the importance of host protein context, and the disease-specific mechanisms that this implies.

Résumé :
The 3,144–amino acid huntingtin protein (HTT) folds in water into a structure consisting of compact, organized domains interspersed with intrinsically disordered protein (IDP) elements. The IDPs function as sites of post-translational modifications and proteolysis as well as in targeting, binding, and aggregation. Although the dominant structural motif of HTT is the α‎-helix–rich HEAT repeat, the expanded polyglutamine (polyQ) toxicity responsible for Huntington’s disease is most likely played out within intrinsically disordered HTT exon 1–like fragments consisting of the 16– to 17–amino acid N-terminal HTTNT segment, the polyQ segment, and a proline-rich segment. The physical behavior of HTT exon 1 fragments is dominated by interactive, polyQ repeat length–dependent structural transitions responsible for membrane and protein–protein interactions and the formation of tetramers, higher oligomers, amyloid fibrils, and inclusions. Understanding the basis of this solution behavior may be the key to disease mechanisms and molecular therapeutic strategies.

Patterson, Marc C. Congenital Disorders of Glycosylation. Oxford University Press, 2017. http://dx.doi.org/10.1093/med/9780199937837.003.0066.

Texte intégral
Résumé :
Congenital disorders of glycosylation (CDG) comprise a family of multisystem diseases in which N- and O-linked glycosylation and glypiation of a variety of proteins and lipids is deficient. The hypoglycosylation of multiple glycoconjugates impairs normal development of the brain (and other organs), and is associated with both episodic and chronic organ dysfunction. Developmental disorders; seizures; strokelike episodes (and stroke); hearing and visual loss; peripheral neuropathy; coagulopathy; and immune, liver, endocrine, cardiac, and cutaneous manifestations may occur in varying combinations. Specific therapy is available for MPI-CDG and SLC35C2-CDG. Most forms of O-linked CDG affect muscle; these include congenital muscular dystrophies and limb girdle dystrophies.

Résumé :
The aim of the study was to determine the effect of feed additives (algae, soybean, and sunflower oil) used in the rabbit feed on: growth indices and slaughter traits, pH, colour, texture, chemical composition, fatty acid profile and oxidative stability (TBARS) of the meat as well as FTO and FABP4 genes expression in the meat’s intramuscular fat (m. longissimus lumborum), depending on the age and sex. The experimental material consisted of Termond White rabbits (n = 160, 80 females and 80 males). Animals were weaned on the 35th day of life, and housed in metal cages arranged in batteries (4 rabbits of the same sex in a cage). From weaning to 12 or 18 weeks of age, the rabbits were fed pellets ad libitum. Animals in the control group (C) received non-supplemented pellets throughout the experiment. In the other groups, the pellet contained 1% algae (A), 3% sunflower oil (OS), and 3% soybean oil(SO).The experimental diets were formulated to have similar protein and energy content. Diets were balanced by lowering the proportion of other feed components. The total share of all components remained at 100%. The results indicate that 3% vegetable oils (soybean or sunflower) supplementation of diets for growing rabbits leads to an increase of body weight and improvement of some of the slaughter traits, while 1% addition of algae to the feed causes deterioration of body weight and slaughter traits. The effect of oil additive depends on the animals’ age. Supplementation of the rabbits’ diet with algae (1%) or sunflower and soybean oils (3%) led to an increase in the dressing percentage of rabbits slaughtered at 18 weeks of age (approx. 3%), but had no effect on the dressing percentage of rabbits slaughtered at 12 weeks of age. Feeding pellets with either 3% vegetable oils or 1% algae additive to the rabbits did not significantly change the chemical composition of the meat. Protein content increased and intramuscular fat content decreased with age, while ash and water content were similar. The feed additives significantly differentiated meat acidity without deteriorating meat quality. Diet modification has not affected negatively meat colour. 24 h after the slaughter, the colour of rabbit meat was similar across the studied feeding groups. Correlation between diet and rabbits’ age was found. Meat texture (hardness, springiness and chewiness) of all rabbit groups slaughtered at 12 weeks of age was similar, and the shear for cewas greater in rabbits fed pellets with algae and soybean oil. At 18 weeks of age, rabbit meat from experimental groups had lower hardness and chewiness, compared to meat of the animals from the control group. Meat shear force was higher in the control group, and from algae-supplemented group. The correlation between diet and age was also found. The use of 3% vegetable oils or 1% algae as feed additives significantly reduced meat oxidative stability. Soybean or sunflower oil (3%) usedas feed additives favourably modified the fatty acid composition of intramuscular fat. Polyunsaturated fatty acids (PUFA) content was increased, including linoleic acid, and PUFA/MUFA ratio was improved. The content of these acids decreased with age. The use of algae (1%) as a feed additive resulted in positive effect on the increase of n-3 fatty acid content (EPA and DHA) in meat intramuscular fat. Algae supplementation improved pro-health properties of meat, with low n-6/n-3 acid ratio (2.5), indicating that diet modification may affect the fatty acid composition of rabbit meat. The influence of diet and age on FTO and FABP4 gene expression in meat intramuscular fat (m. longissimus lumborum) was found. FTO and FABP4 gene expression increased with age and was the highest in the group of rabbits with 1% algae supplementation in the diet. The effect of rabbits’ gender on growth, slaughter traits, meat quality and gene expression in rabbits was not observed. In conclusion, the use of natural feed additives, such as sunflower, soybean oil or algae, can improve the nutritional value of rabbit meat, without changing its chemical or physical properties, and therefore the meat can serve as functional food, with properties beneficial to human health. The results obtained in this study also indicate that the expression of FTO and FABP4 genes in rabbit muscles is regulated by dietary factors and age, which, in addition to cognitive significance, has practical implications for improving technological and dietary quality of rabbit meat.

Résumé :
We usually associate the sounds of classic rock ‘n’ roll with youthful rebellion, by juvenile delinquents, student demonstrators, idealistic hippies, or irreverent punks. But an important strain of rock from the late 1960s onward spoke to and for a very different audience: the regular working-class fans who didn’t want to change the world as much as they wanted to protect their place in it. From Creedence Clearwater Revival to Bruce Springsteen, from Lynyrd Skynyrd to AC/DC, and from Judas Priest to Ted Nugent, the music provided the anthems of an increasingly distinct—and increasingly vulnerable—demographic, which has since become a key influence on political culture around the world.

Résumé :
This chapter examines early discourses on the relationship between television and the developing black freedom movement, with particular emphasis on optimistic hopes that television could be a progressive tool for African American advancement and racial justice. Unlike radio, early network television appeared to take seriously obligations to present African Americans in respectful ways. In the early 1950s, for example, NBC's politically progressive chief censor worked to eradicate offensive black stereotypes from programming by scrubbing references to “darkies,” images of Stepin Fetchit–style characters. This chapter first considers the National Association for the Advancement of Colored People's protest against the Amos 'n' Andy and response to the Beulah radio shows before discussing the role of entertainment television in the pre-civil rights period. It looks at the ABC program The Beulah Show. While Beulah exemplifies early television's initial foray into the arena of race relations and black representation, this chapter argues that it did not give viewers a concept of black and white on equal terms.

Résumé :
Pyromania corresponds to fire setting not done for criminal reasons, for profit or sabotage, for monetary gain, as an expression of sociopolitical ideology (an act of terrorism or protest) or anger, or for revenge. Pyromania, in the sense of arson without a separate motive, is a rare phenomenon. In the DSM-IV-TR, pyromania is classified as an impulse control disorder (ICD) not elsewhere classified. It is characterized by a failure to resist impulsive, repetitive, deliberate fire-setting urges that are unrelated to external reward. The only study of the prevalence of fire setting derived from the National Epidemiological Survey on Alcohol and Related Conditions found a lifetime prevalence of 1% for fire setting in the U.S. population. The prevalence of pyromania in adult psychiatric inpatients was 3.4% (n = 7), and the lifetime prevalence was 5.9%.Fire setting is significantly associated with a wide range of antisocial behaviors. Multivariate logistic regression analyses identified strong associations between lifetime alcohol and marijuana use disorders, conduct disorder, antisocial and obsessive-compulsive personality disorders, and a family history of antisocial behavior. Intentional illicit fire-setting behavior is associated with a broad array of antisocial behaviors and psychiatric comorbidities. The most prevalent psychiatric disorders among persons with a history of fire setting are any lifetime alcohol use disorder (71.7%), antisocial personality disorder (51.46%), marijuana use disorder (43.17%), and nicotine dependence (42.95%). A family history of antisocial behavior is also frequent (60%).
