Texte intégralThe circular economy is one of the ways of dealing with various environmental, economic and societal issues. A global vision is therefore essential to consider all stakeholders and their interactions, so that the industrialization of such an economy is possible. We therefore propose to extend the concept of regeneration to consider all circular strategies (called regenerators) that enable value to be recovered for a product at the end of its life cycle. However, the literature does not analyze all possible regeneration trajectories in their entirety, and therefore does not characterize regenerators in a generic way. Thus, our work proposes a formalization based on product health state to characterize all regenerators in an ecosystem, which will enable us to determine which regenerators are able to regenerate a product at end-of-life. To consider the context when choosing a regenerator, several indicators are identified. Some of these need to be estimated, as they are dependent on the product's health state, and on the realization of regeneration, which may be uncertain. Thus, thanks to product's health state/regenerator modeling and Monte-Carlo simulation, these indicators are estimated and the whole set is finally used in a multi-criteria decision support tool (AHP) to rank possible regenerators and help decision-makers to choose the best regeneration trajectory for the end-of-life product. The results obtained show that the various proposals contribute to the implementation of the circular economy and to increasing knowledge for its industrialization

Texte intégralPancreatic cancer is the fourth most lethal cancer and ranks as the eighth most commonly diagnosed cancer worldwide. This is due to its rapid proliferation, strong metastatic potential and its delayed detection. One major risk factor for developing pancreatic cancer is the aggressive inflammatory disease chronic pancreatitis. Chronic inflammation frequently precedes the development of certain pancreatic cancers.
Inflammation is a protective and necessary process by which the body can alert the immune system of the existence of a wound or infection and mount an immune response to remove the harmful stimuli and start wound healing. The cross-talking of cells of the immune system and infected cells happens through cytokines, soluble proteins that activate and recruit other immune cells to increase the system’s response to the pathogen. Failure to resolve the injury can result in persistent cytokine production that in turn allows a cell that is damaged or altered to survive when in normal conditions it would be killed. Inflammation is thought to create a microenvironment that facilitates the initiation and/or growth of pancreatic cancer cells.
Cytokines use two important kinases for their signaling: Janus Kinases (JAKs) and Signal Transducers and Activators of Transcription (STATs). The JAKs are activated upon the binding of cytokines to their corresponding receptors. When activated, the JAKs activate STATs through tyrosine phosphorylation. The STATs transduce signals to the nucleus of the cells to induce expression of critical genes essential in normal physiological cellular events such as differentiation, proliferation, cell survival, apoptosis and angiogenesis. STAT3 (a member of the STAT family) is constitutively activated in some pancreatic cancers, promoting cell cycle progression, cellular transformations and preventing apoptosis. Therefore, STAT3 is a promising target for cancer treatment. Novel therapies that inhibit STAT3 activity in cancers are urgently needed. Natural products are a very good resource for the discovery of new drugs against pancreatic cancer.
Covering more than 70% of the Earths surface, The Ocean is an excellent source of bioactive natural products. Harbor Branch Oceanographic Institute’s Center for Marine Biomedical and Biotechnology Research (HBOI-CMBBR) situated in Florida, aims to find new marine natural products useful in disease prevention and drug therapy. Their current focus is to look for novel treatments for preventing both the formation of new pancreatic tumors and the metastasis of existing tumors.
The hypothesis of this degree project was that novel inhibitors of STAT3 useful in the treatment of pancreatitis and/or pancreatic cancer could be found from marine-natural products. The first specific aim of this degree project was to set up an assay to identify bioactive marine natural products as inhibitors of inflammation. Furthermore the assay was validated using a commercially available inhibitor of inflammation (Cucurbitacin I). The last aim was to further validate the assay by screening pure compounds and peak library material from the HBOI marine specimen collection.
At the end of the experimentation time, the assay still was not set-up as there were difficulties in proper cell culture techniques and the cell line did not respond as advertised. While the results were not as expected, the work performed resulted in familiarization with research laboratory practices and increased laboratory skills. Moreover, the results from the assays point to future directions to accomplish this project.

Job tasks: Evaluate the solid state fermentation (thereinafter – SSF) fermente by lactic acid bacteria (thereinafter – LAB) in different Jerusalem artichoke influence: pork ready-to-cook physical- chemical parameters; beef ready-to-cook physical-chemical parameters; changes of volatile compounds in beef ready-to-cook. Evaluate the SSF of fermented in different LAB lupine influence: pork ready-to-cook physical- chemical parameters; beef ready-to-cook physical-chemical parameters; changes of volatile compounds in pork and beef ready-to-cook. Evaluate SFF ferment in different LAB Jerusalem artichoke and lupine influence to pork ready-to-cook impurity. In this experiment pork and beef ready-to-cook have been used with 5 percent product (fermented lupine and Jerusalem artichoke, for fementation were used 3 LAB: P. acidilactici KTU -05-7, P. pentosaceus KTU -05-8, L. sakei KTU -05-6). Texte intégralThe theses is about the control of production, commerce, use and publicity of tobacco by the State, in three ways of performance (state control), according to Federal Constitution of 1988, the Framework Convention on Tobacco Control and the internal Legislation. It is due to the Legislative Power, to elaborate the restritive rules of the using of tobacco; it is due to the Executive Power the planning and the execution of public politics concerning to the prevention of tobacco use; and to Judiciary, to judge the compensation procedure by the healthy damage caused by tobacco use. Because it involves the fundamental right to the health of the smoking and non-smoking citizen (second hand smoking), this work analyses the collision of fundamental rights and the acting of the tobacco industry, for misinformation from the 1920 decade in XX century. At last, it stablishes a comparison between hard cases to judge, taking into account the relationship between right and reality, specially the conclusions of science concerning to the diseases caused by cigarette smoking which can lead to death.
A tese aborda o controle pelo Estado da produção, comercialização, consumo e publicidade do tabaco, numa tríplice esfera de atuação, denominada de dirigismo estatal, de acordo com a Constituição Federal de 1988, a Convenção-Quadro de Controle do Tabaco e a legislação infraconstitucional no Brasil. Cabe ao Poder Legislativo, notadamente, a elaboração de normas restritivas à utilização do tabaco; cabe ao Executivo o planejamento e a execução de políticas públicas voltadas à prevenção do tabagismo; e, ao Judiciário, o julgamento das ações de indenização pelos danos causados à saúde pelo consumo de cigarro. Em razão de envolver direito fundamental à saúde do cidadão fumante e não fumante (fumante passivo), este trabalho analisa a colisão de direitos fundamentais e a conduta da indústria de tabaco, por defeito de informação perpetrado desde a década de 1920, do século XX. Por fim, estabelece um parâmetro de julgamento de casos difíceis, levando-se em conta a relação direito e realidade, especialmente as conclusões da ciência no tocante às doenças causadas pelo cigarro, que podem levar à morte.

Texte intégralEste trabalho propõe analisar o fenômeno da terceirização desenvolvido ao longo das duas últimas décadas em Bio-Manguinhos. Trata-se de um estudo de caso realizado na unidade de produção de vacinas e reagentes para diagnóstico da Fundação Oswaldo Cruz. Inicialmente foi realizado um estudo de cenário, analisando a política de gestão do trabalho desenvolvida pelo Estado brasileiro para as instituições públicas. A seguir, a análise passou a verificar a inserção da Fiocruz nessas políticas, desde a sua criação e no decorrer de vigência de diferentes formas de contratação, alternando da extrema rigidez para a total flexibilidade sempre com foco na gestão do trabalho, e, como essas políticas se refletiram no desempenho de Bio-Manguinhos. Baseado no contexto da conformação do Estado e na análise do desempenho da unidade, o trabalho critica a falta de planejamento e de prospecção dessas políticas, que favoreça a inovação de processos, produtos e procedimentos e o desempenho pleno de uma unidade de produção de insumos para a saúde, tão necessários ao atendimento das necessidades da população brasileira. Critica também, a ausência de um modelo de Estado consolidado que sirva de arcabouço à formulação dessas políticas. Devido às fortes críticas dos órgãos de controle acerca da extrapolação dos limites da terceirização praticada nas últimas duas décadas e à falta de definição clara desses marcos legais, o trabalho apresenta uma metodologia baseada nas atribuições dos cargos do plano de carreiras da Fiocruz e culmina com a apresentação de uma matriz de atribuições passíveis de serem realizadas por contratação indireta. Conclui pela necessidade de convivência de dois quadros de trabalhadores: um permanente formado por servidores, estáveis e de carreira e outro flexível, executado por contratação indireta, de caráter eventual, temporário ou de apoio às atividades relacionadas à missão de Bio-Manguinhos. O trabalho ainda sugere a aplicação da mesma metodologia às demais unidades da Fiocruz e que a instituição afirme, frente aos órgãos de controle, quais atividades que ela precisa manter no quadro de servidores permanente e quais ela quer delegar a terceiros sem contudo, ferir a legislação vigente. Por fim, constata-se que a metodologia apresentada ameniza, mas não resolve o problema, dessa forma, recomenda-se à Fiocruz buscar mecanismos que altere o modelo de gestão pelo qual está submetida a fim de viabilizar as duas formas de incorporação de mão-de-obra.
