Résumé :
Abstract The pathological process underlying Alzheimer disease (AD) unfolds predominantly in the cerebral cortex with the gradual appearance and regional progression of abnormal tau. Intraneuronal tau pathology progresses from the temporal transentorhinal and entorhinal regions into neocortical fields/areas of the temporal allocortex. Here, based on 95 cases staged for AD-related neurofibrillary changes, we propose an ordered progression of abnormal tau in the temporal allocortex. Initially, abnormal tau was limited to distal dendritic segments followed by tau in cell bodies of projection neurons of the transentorhinal/entorhinal layer pre-α. Next, abnormal distal dendrites accumulated in the prosubiculum and extended into the CA1 stratum oriens and lacunosum. Subsequently, altered dendrites developed in the CA2/CA3 stratum oriens and stratum lacunosum-moleculare, combined with tau-positive thorny excrescences of CA3/CA4 mossy cells. Finally, granule cells of the dentate fascia became involved. Such a progression might recapitulate a sequence of transsynaptic spreading of abnormal tau from 1 projection neuron to the next: From pre-α cells to distal dendrites in the prosubiculum and CA1; then, from CA1 or prosubicular pyramids to CA2 principal cells and CA3/CA4 mossy cells; finally, from CA4 mossy cells to dentate granule cells. The lesions are additive: Those from the previous steps persist.

Résumé :
This paper takes the parallel indexing cam mechanism as research object and analyzes the formation of the profile of cam. On the basis of Visual C++6.0 object-oriented method, we have successfully developed the CAD/CAE/CAM system of parallel indexing cam mechanisms, which provides the parameter input dialogue frame with friendly interface. We can generate accurate profile data by using it, then create the three-dimensional model of the parallel indexing cam and assemble it with other parts under the Pro/E environment. At last, we don’t build virtual prototype until we transform the model into ADAMS through special interface Mechanism/Pro. Finally, this paper has researched the theory of contact and collision, carries out the motion simulation. We have received many valuable conclusions of practical application through all of the process.

Résumé :
Calmodulin (CaM) is most recognized for its role in activating Ca2+–CaM-dependent enzymes following increased intracellular Ca2+. However, CaM's high intracellular concentration indicates CaM has the potential to play a significant role as a Ca2+ buffer. Neurogranin (Ng) is a small neuronal IQ-motif–containing protein that accelerates Ca2+ dissociation from CaM. In cells that contain high concentrations of both Ng and CaM, like CA1 pyramidal neurons, we hypothesize that the accelerated Ca2+ dissociation from CaM by Ng decreases the buffering capacity of CaM and thereby shapes the transient dynamics of intracellular free Ca2+. We examined this hypothesis using a mathematical model constructed on the known biochemistry of Ng and confirmed the simulation results with Ca2+ imaging data in the literature. In a single-compartment model that contains no Ca2+ extrusion mechanism, Ng increased the steady-state free Ca2+. However, in the presence of a Ca2+ extrusion mechanism, Ng accelerated the decay rate of free Ca2+ through its ability to increase the Ca2+ dissociation from CaM, which in turn becomes subject to Ca2+ extrusion. Interestingly, PEP-19, another neuronal IQ-motif protein that accelerates both Ca2+ association and dissociation from CaM, appears to have the opposite impact than that of Ng on free Ca2+. As such, Ng may regulate, in addition to the Ca2+–CaM-dependent process, Ca2+-sensitive enzymes by influencing the buffering capacity of CaM and subsequently free Ca2+ levels. We examined the relative impact of these Ng-induced effects in the induction of synaptic plasticity.

The automotive development process is advancing through the utilization of CAD/CAM/CAE. This advancement is being enhanced by proper utilization of CAD/CAM integration, Digital prototyping and Digital mock-up. Until recently, CAD has played the leading role for the digitalization of automotive development. However, recent trend shows that, the leading role is shifted from CAD to analysis simulation. Three-dimensional design, which utilizes digital mock-up, is becoming the main tool for product design. Here, the functional capability and predictive assessment by CAE is the core technology. In addition, this supporting role is helping to realize the concurrent engineering in production engineering field.

Résumé :
One of the major barriers to the integration of CAD, CAPP and CAM is insufficiency and incompatibility of product model data among various application systems. This paper presented an effective approach to achieve integrated process planning with CAD and CAM under a product data management environment based on 3D product models. The system architecture and the processes of an integrated process planning system (3D-IPP) were constructed using feature information and specific CAD/CAPP/CAM integration technologies based on feature. A feature-based product information model is designed as the information source for the 3D-IPP. A feature-based integrated process planning method was developed based on group technology, intelligent decision-making algorithms and parametric operation templates in numerically controlled (NC) programming. The procedures of intelligent process route planning and parametric NC operation planning were addressed. The 3D-IPP system and its implementation provide an effective solution to strive for complete information sharing among application systems of CAD, CAPP and CAM.

Résumé :
The authors' company is a medium-sized engineering company specializing in naval ship repair. The bulk of corporate work centers on the Phased Maintenance (PM) of three classes of ships. Typically, each PM contract covers three to five different ships per class scheduled for 90-day Availabilities at approximately one-year intervals over a period of five years. The type of work to be performed during each Phased Maintenance Availability (PMA) falls into one of two categories: ship alterations or ship repairs. The first group, ship alterations, is characterized by detailed, long-lead engineering and planning efforts, typically beginning 540 days prior to the vessels's arrival. The second group, ship repairs, makes up the other end of the spectrum with short lead times and compressed service details. The majority of repair items are identified 60 days prior to an Availability, while some are not determined until after the vessel has arrived in the shipyard. The engineering department that services PMA work is composed of three disciplines: structural, mechanical and electrical. The mechanical discipline is further subdivided into the areas of machinery and piping/HVAC. While the nature of PMA work within each discipline is peculiar to the application, the process is similar in each. Reference information is gathered and verified, technical analysis is provided where necessary, and detailed drawings are prepared and submitted for Navy approval prior to shipyard production. All drawings are developed using two-dimensional drafting techniques at various sites by teams of computer-aided design (CAD) input operators utilizing color graphic workstations on a multi-shift basis as required by the workload. Completed drawings are transferred to the engineering site over a network link, where additional workstations are available for engineers to check and correct them as necessary

Résumé :
In this paper, a comparison between CNC manufacturing and Rapid Prototyping technology (FDM – Fused Deposition Modeling process), applied for a cam fabrication, is presented. In the products development area, a substantial support is offered by models, as intermediate between product configuration and technology design. The CAD/CAM/CNC technology is a widely used technique for creating prototypes, as well as production parts, using a subtractive type material-removal procedure from a semi-manufactured article. The rapid prototyping (RP) technologies are additive processes, where the part is built up layer by layer until done, directly from the 3D CAD model, within the precision limits of the chosen process. Similarities and differences between these two coexisting computer driven prototyping processes, the subtractive CNC 3 – axes milled part production and the additive RP/ FDM technique, are pointed out for a disk cam manufacturing as sample part.

Résumé :
Recently, computer-aided design and computer aided manufacturing (CAD/CAM) is largely applied in universal machinery due to the accelerated development of computer technologies. Owing to the technical application of CAD/CAM tool in the manufacturing industry, enterprises have progressively changed their traditional approaches to design and manufacture of products. The CAD module provides product designers with capabilities of designing product model by feature-based design and obtaining product data. The product model is displayed on a screen in the CAD environment before transferring it downstream to the CAM module. This paper presents the application of CAD/CAM in solid modelling, design of machining process, fixture design and virtual manufacture of a flanged tube. Pro/Engineer (Pro/E) Wildfire 4.0 was used to create the three dimensional (3D) model of the flange tube based on its two-dimensional drawing (2D) blank drawing and the mould was designed and created using the same software. The mould creation was based on the clamping and positioning devices of the flange tube being manufactured. The technical schedule of the flange tube was worked out including the choice of machining method, machine tool, cutting tool and selection and computation of machining parameters based on finished drawing the flange tube. Two dedicated fixtures for holding the flange tube in the machine tool were designed and finally Master CAM 9.0 software was applied to the virtual machining of the mould. It is concluded that CAD/CAM tool is important in manufacturing technology as it automates the manufacturing process thereby saving time, energy, cost, and making production highly flexible.

Résumé :
“CyberCut™” is a testbed for an Internet-based CAD/CAM system. It was specifically designed to be a networked, automated system, with a seamless communication flow from a client-side designer to a server-side machining service. The creation of CyberCut required several new software modules. These include: a) a Web-based design tool in which Design-for-Manufacturing information and machining rules constrain the designer to manufacturable parts; b) a geometric representation called SIF-DSG, for unambiguous communication between the client-side designer and the server-side process planner; c) an automated process planning system with several sub-modules that convert an incoming design to a set of tool-paths for execution on a 3-axis CNC milling machine. Using this software-pipeline, a CyberCut service, modeled on the MOSIS service for VLSI chips, has been now been launched for limited student-use at a group of cooperating universities.

Résumé :
An integrated digital routine is applied in the near net shape manufacturing of marine propeller. Firstly, the 3D CAD file of propeller is created by parametric modeling. Secondly, the propeller casting process is simulated using CAE software, through which an optimized casting scheme is obtained. Thirdly, fabricates the mold using laser rapid prototyping and cast the metal propeller. Finally, evaluate the casting precision performance. CAD, CAE and CAM are integrated in this process. The dimensional accuracy of the final piece is controlled within 1mm and its surface roughness achieves Ra 6.3μm. The result shows that the pattern-less casting of propeller can be achieved by this method, reducing cost and performing high accuracy.

Résumé :
An approach to CAD and CAM modeling and to the design of CAD/CAM systems is presented. Models of the product and of the process are represented by logical assertions in a common logical language. CAD/CAM functions are represented by the application of logical inference rules, which correspond to the derivation of new information as well as to actions. This allows all the different kinds of model and specification used in design and manufacturing to be represented in a computer in a common form. It therefore allows the representation of constraints and rules connecting any aspects of design and manufacturing together. This approach has all the advantages of formal specifiction, namely, ease of expression, communication, standardization and abstraction. At the same time, we demonstrate its practical implementation in an efficient form, and which is industry compatible, and we report practical experience with using this approach for CAD/CAM models and for intelligent CAD/CAM functions.

Résumé :
Advances in computer-aided design (CAD) / computer-aided manufacturing (CAM) technologies and their ease of application enabled the development of novel treatment concepts for modern prosthodontics. This recent paradigm shift in fixed prosthodontics from traditional to minimally invasive treatment approaches is evidenced by the clinical long-term success of bonded CAD/CAM glass-ceramic restorations. Today, defect-oriented restorations, such as inlays, onlays, and posterior crowns, are predominately fabricated from glass-ceramics in monolithic application. The variety of CAD/CAM ceramic restorative systems is constantly evolving to meet the increased demands for highly aesthetic, biocompatible, and long-lasting restorations. Recently introduced polymer-infiltrated ceramic network CAD/CAM blocks add innovative treatment options in CAD/CAM chairside 1-visit restorations. The material-specific high-edge stability enables the CAD/CAM machinability of thin restoration margins. Full-contour zirconia restorations are constantly gaining market share at the expense of bilayered systems. Advancements in material science and bonding protocols foster the development of novel material combinations or fabrication techniques of proven high-strength zirconia ceramics. CAD/CAM applications offer a standardized manufacturing process resulting in a reliable, predictable, and economic workflow for individual and complex teeth-supported restorations. More evidence from long-term clinical studies is needed to verify the clinical performance of monolithic polymer-infiltrated ceramic network and zirconia teeth-supported minimally invasive and extensive restorations.

Résumé :
Objectives. This study aimed at comparing the methodologies for achieving an occlusal-mesial ceramic inlay using both the conventional and digital methods in terms of workflow protocol, aesthetic and functional results, working time, but also costs and equipment needed. Material and methods. A 22-year-old patient presented with a direct mesio-occlusal composite restoration with a secondary marginal decay in tooth 3.6 that was subsequently restored by a ceramic inlay manufactured using both traditional and digital methods. Outcomes. The dental technician's talent, experience, and vision guided the technical process of creating the conventional inlay. The technical process of producing the digital inlay involved fewer laboratory stages, but also contamination risks than conventional ones, removing potential human errors associated with each stage and allowing possible changes to be made more quickly and efficiently. The conventional method took more time to complete all of the laboratory steps than the digital method. Conclusions. When compared to the digital method, the traditional method allowed the dental technician to achieve a high level of individualization of the prosthetic restoration. Digital techniques are a method of the future that is rapidly growing and improving. Digital techniques for obtaining an inlay involved high-performance equipment, which is pricey to purchase and maintain.

Résumé :
Neurodegenerative diseases cause neural cells to lose both the functional and sensory abilities as a result of genetic factors, proteopathies and mitochondrial dysfunction. Neurodegeneration forms the basis of most neurodegenerative disorders for example Alzheimer’s disease, Huntington’s diseases, and Parkinson’s diseases. The mechanism that underlines the process of neurodegeneration is not well understood. Understanding the process and mechanism involved in neurodegeneration might offer a better therapeutic approach to positively manage cases of neurodegenerative diseases. Therefore, this study’s target was to create an animal model to study neurodegeneration. Sixteen adult male Wistar rats were used in the study and divided into two groups. Control (0.2 mL of normal saline (NS)), and trimethyltin-treated (TMT, 8 mg/kg stat dose only). These animals underwent perfusion with 4% paraformaldehyde, brain excision and analysis of p53 antigen, GFAP and Bielshowsky on these tissues. The results showed that animals in the control group showed presence of activated p53 antigen, reactive astrogliosis, neurofibrillary tangles, and amyloid plaques within the cytoplasm of the hippocampal cells. Cornus Ammonis (CA2) and (CA3) showed more of the trimethylrtin injury than CA1 and CA4. This study thus revealed that, intra-peritoneal administration of single dose of 8mg/kg of trimethyltin can offer an attractive disease model to study some neurodegenerative diseases. Keywords: p53 antigen, Bielshowsky, Glia fibrillary acidic protein, Trimethyltin, Hippocampus,
