Texte intégralAbstract: The aim of this research was to study the influence of different drying methods (pulsed fluid bed dryer, spray dryer and freeze dryer) of passion fruit pulp in the quality of final powder product. All these processes have the purpose to obtain a powder product, showing advantages and disadvantages in terms of handmade and costs. Nonetheless, the literature has a few studies related to comparison between these processes, focusing final product quality. Skim milk was utilized to evaluate its efficiency on partial substitution of additives commonly used in this kind of processes like maltodextrin and Arabic gum, which have the function of minimizing technologies difficulties in drying fruit juices and other products with high quantity of sugar due to hygroscopicity and termoplasticity in high temperatures and humidity. Initially, physicochemical analysis of passion fruit pulp and skim milk showed values close to those described by literature, with little differences due to passion fruit varieties used, time and place of culture. For skim milk the differences can be explain due to existence of a lot of commercial marks. Drying of passion fruit pulp without additives in many operations conditions had a small yield, becoming process impracticable in these conditions. It was obtained powdered passion fruit and additives in spray dryer. Using maltodextrin the greater yield (23.32%) was obtained for 1:4:4 ratio (passion fruit pulp:maltodextrin:skim milk) that is 50% of solids from additives substituted by skim milk. To Arabic gum the best results (21.96%) was obtained to 1:2:6 (passion fruit pulp:Arabic gum:skim milk) with 75% of solids from additives substituted by skim milk. The total substitution of additives (maltodextrin or Arabic gum) by skim milk also showed good results (15.5%). Texte intégralThis research proposes a reflection on the convergence between fashion, music and consumption, emphasizing the influence of \"pop culture\" in contemporary capitalist society. Is a parallel with the Brazilian rock of the 1980s and 1990s, more specifically with the literary-musical discourse as oppositional discursive practice that will give us support to discuss the consumption of fashion and other products that serve as a passport to social inclusion of the subject. It is intended to clarify that the discourse undertaken in rock lyrics of these decades have criticisms of the \"pop culture, the mass media and consumerism. It is intended to clarify that the discursive practice of rock lyrics of this decade can be considered as a meaningful production process, as well as a symbol of identity, highlighting trends and transformations, incorporating the broad sense of fashion. It is shown here, the importance of both the fashion, the music, not just an agglomeration of products of consumer goods and, more so, as a social and artistic production, since these means of expression can reproduce everyday events in a broader political, sociological, historical and cultural context. In this context we will discuss issues that prompt us about consumer society, consumerism and the close relationship established with the media. Peptídeos antimicrobianos são componentes importantes do sistema de defesa de diversos organismos contra possíveis invasores. Em geral, são pequenos (até 100 aminoácidos), catiônicos e anfipáticos. Eles têm despertado o interesse da comunidade científica por sua capacidade de atuação contra micróbios, que não conseguem desenvolver resistência a esses peptídeos. Ou seja, eles emergem como complemento e/ou alternativa ao uso dos antibióticos convencionais. Este trabalho desenvolveu um modelo computacional de um peptídeo híbrido de pediocina A (N-terminal) e plantaricina 149a (C-terminal), dois peptídeos bactericidas. Dados experimentais obtidos pelo grupo da prof. Dra. Rosângela Itri do IFUSP foram utilizados para modelagem e comparação dos resultados. Foram feitas simulações de MD do peptídeo interagindo com membranas puras e mistas de POPC e POPG utilizando os parâmetros do campo de força GROMOS 53A6 e 54A7. As simulações com uma unidade do peptídeo revelaram a atualização 54A7 era a mais adequado para modelagem desses sistemas. Os mapas de estrutura secundária mostraram que o peptídeo adquire configuração mais ordenada quando interage com membranas com maior quantidade de POPG em sua composição. As simulações com duas unidades do peptídeo sugeririam que o peptídeo interage e penetra na camada de POPG através da região Cterminal. Na simulação com membrana de POPC, nenhuma das porções terminais ficou estável no interior da membrana. O efeito do aumento da concentração de peptídeos foi examinado colocando cinco e dez unidades do peptídeo para interagir com as membranas. Na membrana de POPC, os peptídeos não formam um único aglomerado e causam pouca perturbação na bicamada. Já na membrana de POPG, o efeito da interação do aglomerado de peptídeos é acentuado, provocando grandes deformações na bicamada lipídica, praticamente a destruindo. Esse fenômeno sugere um possível mecanismo carpete para ação do peptídeo sobre a membrana fosfolipídica de bactérias.
Antimicrobial peptides are important components of defense system in various organizations against possible invaders. They are generally small (100 aminoacids), cationic and amphipathic. They have stimulated the interest of the scientific community for its ability to act against microbes that cannot develop resistance to these peptides. That is, they emerge as complement and/or alternative to the use of conventional antibiotics. This study developed a computational model of a hybrid peptide pediocin A (Nterminal) and plantaricin 149a (C-terminal), two bactericidal peptides. Experimental data obtained by the group of prof. Dr. Rosângela Itri (IFUSP) were used for modeling and compare the results. MD simulations were made of the peptide interacting with pure and mixed POPC and POPG membranes. These simulations were performed using the parameters of the force field GROMOS 53A6 and 54A7. Simulations with a single copy of the peptide revealed that the force field 54A7 was the most appropriate for modeling these systems. The secondary structure maps showed that the peptide acquires a more ordered configuration when interacting with membranes with higher amounts of POPG in its composition. The simulations with two copies of the peptide suggest that the peptide interacts and penetrates the POPG layer via the C-terminal part. In the simulation with POPC membrane, none of the end portions remained stable within the membrane. The effect of increasing the peptide concentration of was examined by placing five and ten copies of the peptide to interact with the membranes. In the POPC membrane, the peptides do not form a single cluster and they cause little disturbance in the bilayer. In the POPG membrane, the interaction of peptides cluster is enhanced, causing large deformation and practically destroying the lipid bilayer. This phenomenon suggests a possible carpet mechanism of action of the peptide on the phospholipid membrane of bacteria.

Texte intégralIn Part 1 of this thesis, we construct “Okounkov bodies” for an arbitrary pseudo-effective (1,1-class on a Kähler manifold. We prove the differentiability formula of volumes of big classes for Kähler manifolds on which modified nef cones and nef cones coincide. As a consequence we prove Demailly’s transcendental Morse inequality for these particular Kähler manifolds; this includes Kähler surfaces. Then we construct the generalized Okounkov body for any big (1,1)-class, and give a complete characterization of generalized Okounkov bodies on surfaces. We show that this relates the standard Euclidean volume of the body to the volume of the corresponding big class as defined by Boucksom; this solves a problem raised by Lazarsfeld and Mustaţă in the case of surfaces. We also study the behavior of the generalized Okounkov bodies on theboundary of the big cone.Part 2 deals with Kobayashi hyperbolicity-related problems. Chapter 2’s goal is to study the degeneracy of leaves of the one-dimensional foliations on higher dimensional manifolds. The first part of Chapter 2 generalizes McQuillan’s Diophantine approximations for one-dimensional foliations with absolutely isolated singularities, on higher dimensional manifolds. As an application, we give a new proof of Brunella’s hyperbolicity theorem, that is, all the leaves of a generic foliation of degree larger than 2 in CP 6n is hyperbolic. In the second part of Chapter 2 we introduce the so-called weakly reduced singularities for one-dimensional foliations on higher dimensional manifolds. The “weakly reduced singularities” assumption is less demanding than the one required for “reduced singularities”, but play the same role in studying the Green-Griffiths-Lang conjecture. Finally we discuss a strategy to prove the Green-Griffiths-Lang conjecture for complex surfaces.In Chapter 3, assuming that the canonical sheaf is big in the sense of Demailly, we prove theKobayashi volume-hyperbolicity for any (possibly singular) directed variety.In Chapter 4, our first goal is to deal with effective questions related to the Kobayashi and Debarre conjectures, relying on the work of Brotbek and his joint work with Darondeau. We then combine these techniques to study the conjecture on the ampleness of the Demailly-Semple bundles raised by Diverio and Trapani, and also obtain some effective estimates related to this problem. Our result integrates both the Kobayashi and Debarre conjectures, with some effective estimates.The purpose of Chapter 5 is twofold: on the one hand we study a Fujita-type conjecture by Popa and Schnell, and give an effective (linear) bound on the generic global generation of the direct image of the twisted pluricanonical bundle. We also point out the relation between the Seshadri constant and the optimal bound. On the other hand, we give an affirmative answer to a question by Demailly-Peternell-Schneider in a more general setting. As applications, we generalize the theorems by Fujino and Gongyo on images of weak Fano manifolds to the Kawamata log terminal cases, and refine a result by Broustet and Pacienza on the rational connectedness of the image.In Chapter 6, we give a concrete and constructive proof of the equivalence between the category of semistable Higgs bundles with vanishing Chern classes and the category of all representations of the fundamental groups on smooth Kähler manifolds. This chapter is written for the complex geometers who are not familiar with the language of differential graded category used by Simpson to prove the above equivalence on smooth projective manifolds, and for those who would like to see an elementary proof of Corlette-Simpson correspondence for semistable Higgs bundles

Neste trabalho, a mÃsica contemporÃnea, especificamente o pop rock nacional, enquanto um discurso constituinte (COSTA, 2001; MAINGUENEAU, 2006), representa os fenÃmenos de superficialidade, fluidez, fragmentaÃÃo e crise de identidade, tÃo debatidos nos estudos sociolÃgicos apresentados por autores como Hall (2005), Harvey (2005), Kumar (1997) e Bauman (1998, 2005). Partiu-se, assim, do pressuposto de que as canÃÃes fornecem sentidos, identificaÃÃes e modelos de identidade que corroboram esses fenÃmenos. Esta dissertação parte da concepção de que o discurso literomusical (COSTA, 2001) é um discurso que serve de referência para os sujeitos, como legitimador de verdades e disseminador de maneiras de como eles se relacionam com a realidade, com os outros sujeitos e com a própria identidade. Neste trabalho, a música contemporânea, especificamente o pop rock nacional, enquanto um discurso constituinte (COSTA, 2001; MAINGUENEAU, 2006), representa os fenômenos de superficialidade, fluidez, fragmentação e crise de identidade, tão debatidos nos estudos sociológicos apresentados por autores como Hall (2005), Harvey (2005), Kumar (1997) e Bauman (1998, 2005). Partiu-se, assim, do pressuposto de que as canções fornecem sentidos, identificações e modelos de identidade que corroboram esses fenômenos. A principal hipótese defendida nesta dissertação é a de que o sujeito encarnado nas canções do pop rock nacional contemporâneo apresenta características de um sujeito pós-moderno. O estudo contou com a análise das canções das bandas Engenheiros do Hawaii, Titãs, Detonautas e CPM22, e a dos cantores/compositores Cazuza e Pitty, contemplando, dessa maneira, dois períodos do pop rock nacional contemporâneo: as décadas de 80-90 e a década de 2000.

Texte intégralDifferent theoretical approaches have been used in the studies of biomolecular systems aiming to contribute with the treatment of several diseases. For neuropathic pain, for example, the study of compounds that interact with sigma-1 receptor (Sig-1R) can elucidate the main factors associated to their biological activities. For this purpose, studies of Quantitative Structure-Activity Relationships (QSAR) using Partial Least Squares (PLS) and Artificial Neural Network (ANN) methods were applied to 64 Sig-1R antagonists belong to 1-arylpyrazole class. PLS and ANN models were used in order to describe linear and nonlinear behavior, respectively, between a set of descriptors and the biological activity of the selected compounds. The PLS model was obtained with 51 compounds in the training set and 13 compounds in the test set (r² = 0.768, q² = 0.684 and r²test = 0.785). Leave-N-out tests, biological activity randomization and outliers detection confirmed the robustness and stability of the models and showed that they were not obtained by chance correlations. Models were also generated from Multilayer Perceptron Artificial Neural Network (MLP-ANN) and the 6-12-1 architecture, trained by tansig-tansig transfer functions, showed the best result for the biological activity prediction of the compounds (r²training = 0.891, r²validation = 0.852 and r²test = 0.793). Another approach was used to simulate synaptic membranes environment using lipid bilayers composed by POPC, DOPE, POPS and cholesterol. Performed molecular dynamics studies showed that high cholesterol concentration induces decrease of area per lipid and lateral diffusion and increase of membrane thickness and order parameter caused by ordering of phospholipids acil chains. The obtained lipid bilayers can be used to simulate interactions between lipids and small molecules or proteins contributing for researches associated to Alzheimer and Parkinson diseases. The approaches used in this thesis are essential for the development of new researches in Computational Medicinal Chemistry.

Texte intégralSome advanced oxidation processes (AOP) were applied after an anaerobic process for the treatment of bleaching effluents from a kraft pulp mill, aiming the increase of the effluent biodegradability and the removal of toxic compounds, in a way that a further biological treatment could be more efficient to attain the environmental regulations. A horizontal anaerobic imobilizaded sludge reactor (HAIS) was used as the anaerobic treatment in order to reduce the effluent initial organic load. The AOP used were 'O IND.3', 'O IND.3'/'H IND.2'O IND.2', 'O IND.3'/UV, UV, 'H IND.2'O IND.2', 'H IND.2'O IND.2'/UV and 'O IND.3'/'H IND.2'O IND.2'/UV. These AOP were not applied only to promote a further degradation of the remaining COD from the HAIS reactor, but in order to increase the effluent biodegradability and eliminate AOX or other toxic compounds from this effluent. In this way, the AOP would provide an effluent with characteristics to be easily treated by a further and final biological process. In the biological process, the anaerobic biomass was fast adapted to the wastewater. The HAIS reactor reached the stability and had efficiency in COD and AOX removals of approximately 50%, satisfying the aim of promoting an initial reduction in the easy-to-degrade organic matter presented in the bleaching effluents. All the tested AOP promoted modifications in de HAIS effluent biodegradability. The 'O IND.3'/UV at pH 8 was the more efficient process, achieving an increase of 160% on the biodegradability of the HAIS effluent after 45 minutes of oxidation and an ozone concentration of 1155 mg/L. By the AOX analyses, it was observed average degradations of the organochlorine compounds higher than 95% for all the AOP tested. The general results show the importance and the advantage of using an AOP as a complement of a biological treatment process for the degradation of effluents with low biodegradability, due to the presence of recalcitrant and toxic compounds.

Texte intégralThe present study consisted of (1) selection of pre-treatment adequate temperature/time binomial applied to the raw material, (2) optimization of fruit and vegetables pulps formulation, (3) selection of pasteurization conditions to be applied to the end product, (4) evaluation of untreated pulp (ST) and pasteurized pulp (TT) modifications, with utilization of transparent (T) and opaque (O) packaging, and maintained in refrigerated or in frozen conditions. Pulps were monitored for physicochemical, biochemical, microbiological, rheological and sensory parameters. The optimized pulp consists in an apple base (41 %) and pineapple (31 %), to which cabbage (8%), pumpkin (10%) and banana (10%) were introduced. The temperature/time binomial of the thermal pre-treatments are specific for each raw-material (100 °C/ 5 minutes for cabbage, 90 °C/ 6 minutes for pumpkin and 100 °C/ 1 minute for “Fuji” apple and 1,5 minutes for the Granny Smith variety). The established pasteurization temperature/time binomial was 75 °C/ 90 seconds. The developed pulp in transparent and opaque packaging under refrigeration showed a shelf-life of 7 and 14 days, respectively. In frozen storage, quality maintenance is more dependent of storage time and no significant changes were observed for 60 days.

Resumo: Currículos-Hipertextos-Pops assume, em sua escrita como tese, a banalidade, o ordinário e o comum aos quais remetem as culturas escolares, em seus ritmos espaço-temporais. Enfrenta esta condição de práticas educativas que se movimentam em três escolas, com professoras e professor de distintas experiências, com projetos educativos em multiplicidades. É tese que se inverte no caos. E nele se efetua. O trabalho com conceitos de Gilles Deleuze, na tese, coloca-os em fluxos para o campo dos estudos de currículo, invadindo-o e com ele seatritando. Não é uma conversa tranqüila, com canais abertos ao diálogo, à hospitalidade. Trabalhou-se na barbárie, foi preciso fazer do currículo passagem para a multidão, desconstrutora e violenta. A opção foi deixar vazar os fluxos pelo que há de mais comum e poderoso no pensamento curricular sobre a escola: sua organização em espaços e tempos da modernidade, sua centralidade nos conteúdos, sua expectativa em comunicar para ensinar. As pontencialidades do encontro do currículo com os tempos ¿deleuzianos¿ são, nesta tese, o desassossego das relações entre escola e culturas. Também são os traçados, linhas do ¿conto¿ para se considerar a política curricular como estética artística
Abstract: Curriculum-hypertexts-pops assumes, in its writing as a thesis, the banality, the ordinary and the common to which the scholastic cultures are sent, in their space-temporal rhythms? It faces this condition of educational practices that are set in motion in three schools,with teachers with different experiences, with educational projects in multiplicities. It is a thesis that inverts in the chaos. And it is made effective on it. The work with concepts of Gilles Deleuze, in the thesis, puts them into fluxes to the curriculum field studies, invading it with attrition. It is not a relaxed chatting, with open channels to dialogue, to the hospitality. It was worked under barbarism, it was necessary to make from the curriculum passage to a violent and deconstructive multitude. The option was to let empty the fluxes of the most common and powerful in the curricular thinking of the school: its organization into spaces and times of the modernity, its centrality in contents, its expectation to communicate to teach. The potentialities of the meeting of the curriculum with the ¿Deleuzian¿ times are, in this thesis, the disturbance of the relations between school and cultures. They are also the draw, lines of ¿story¿ to let curricular politics be considered artistic esthetics

Since Pope Francis’ election, the mass media has focused heavily on his statements and actions. Previous literature attests to the notion that Pope Francis is unique among his predecessors when it comes to media coverage, primarily due to his propensity to make statements or moral recommendations that seemingly oppose or deviate from traditional Catholic values. This thesis explores the relationship between Pope Francis and the American mass media by analyzing 226 articles published by The New York Times within the first six years of his Papacy. The findings of this thesis point to notions related to the valence of Papal coverage, which began as primarily positive but have shifted in nature from the beginning of Francis’ Papacy. The findings of this thesis also point towards the religious nature of Papal coverage and also the potential for sexscandal coverage to shift representation of the Pope.

Texte intégralRuminant is exposed to POPs deposited on grasslands. This thesis study the transfer of PCDD/Fs, PCBs and PAHs from fodder and soil to milk. PHE, PYR and B[a]P contaminated grass was incubated in the rumen of cows in order to determine the in sacco disappearance of PAHs, cuticular n-alkanes and DM. The 3 PAHs disappeared (83%) in the 1st hour of incubation while the cuticular n-alkanes and DM disappearance was slower. The transfer of PAHs from grass and soil towards milk has been studied in vivo in goats. The transfer to milk of native PAHs is minimal whatever the matrix ingested. The transfer of PCDD/Fs and PCBs from contaminated hay to milk has been also studied. The CORs of PCBs varied from 5% to 90% and for PCDD/Fs from 1% to 40%. The risk of human exposure via milk could become meanly with PCDD/Fs an PCBs. The current safety threshold values in milk as well as toxicity of 1-OH-PYR must be reviewed

Texte intégralThe objective of this study was evaluate despectinated citrus pulp (Braspolpa®) replacing 30% of pelleted citrus pulp in feedlot beef cattle finishing diets. In the first trial, was used despectinated citrus pulp in natura. Forty eight yearling bulls (24 Nelore and 24 Canchim), 18 months old and averaging 404 and 439 kg initial live weight for Nelore and Canchim, respectively. The animals were assigned in a randomized block design with a 2x2 factorial scheme (two breeds and two treatments) and grouped by breed and initial live weight. Experimental period lasted 92 days, with three periods. Diets contained sugar cane bagasse in natura, dried citrus pulp, soybean meal, urea, mineralvitamin mix, sodium bicarbonate, sodium monensin, and the energy sources tested (despectinated citrus pulp). In the second trial, the despectinated citrus pulp was ensiled with or without sodium benzoate (0.18% of GM). Fifty four yearling bulls (Nelore/Angus or Nelore Santa Gertrudis x Braunvieh), 16.5 months old and averaging 303 kg initial live weight. The animals were assigned in a complete randomized block design, and grouped by initial live weight. Experimental period lasted 91 days, with three periods. Diets contained sugar cane bagasse in natura, dried citrus pulp, cotton meal, urea, mineral-vitamin mix, sodium bicarbonate, sodium monensin, and the energy sources tested (despectinated citrus pulp with and without sodium benzoate). For these two trials, the rations were composed of 85% of the concentrate and 15% of the sugar cane bagasse in natura, and were balanced according to NRC (1996) level 1, to reach isonitrogeneous with positive rumen degradable protein and metabolizable protein balances. Variables evaluated were daily weight gain (DWG, kg/day), dry matter intake (DMI, kg DM/day), feed conversion (FC, kg DMI/kg DWG), net energy for maintenance and gain values, and the ratio between calculated net energy (based on experimental data) and net energy predicted by NRC (1996), level 1 (observed/predicted). In both trials, the substitution of dried citrus pulp for despectinated citrus pulp increased the NDF and ADF intakes (P<0.01) and did not affect (P>0.05) DMI (8.05 vs. 8.13 and 7.76 vs. 7.78 kg/day for the first and second trials, respectively), DWG (1.11 vs. 1.16 e 1.27 vs. 1.36 kg/day for the first and second trials, respectively), neither FC (7.82 vs. 7.29 e 6.62 vs. 5.96 kg of DM/kg for the first and second trials, respectively). Observed values for net energies of maintenance and gain did not differ among treatments (P>0.05). According to the experimental data, the net energy relation (observed/predicted) for the PCP treatment was lower (P<0.05) than PCUD treatment (1.15; 1.39; 1.22 vs. 1.23; 1.51; 1.28 and 1.10; 1.31; 1.18 vs. 1.18; 1.45; 1.24 Mcal/kg DM of NEm, NEg and ME for the first and second trials, respectively). The partial replacing of PCP for PCUD in natura or ensiling did not change the animal performance.

Resumo: Biorrefinaria é um novo conceito que abrange as instalações industriais que utilizam a biomassa como matéria-prima para a produção de intermediários químicos, combustíveis e energia, analogamente às refinarias de petróleo. Uma matéria prima que apresenta oportunidades para ser utilizada no Brasil é a polpa cítrica, que é o resíduo do processamento do suco de laranja. Este resíduo é produzido em grandes quantidades no Brasil, que é o maior produtor mundial de sucos. Atualmente, a polpa cítrica não tem valor comercial importante, já que os custos de secagem comprometem as margens de revenda como ração animal, que é a destinação típica para este resíduo. Por outro lado, a polpa cítrica é uma fonte de limoneno e pectina, que já têm processos de extração estabelecidos, e os carboidratos da polpa podem ser transformadas em etanol e alguns ácidos com alto valor agregado, como o ácido D-galacturônico, o ácido L-galactônico, o ácido múcico e o ácido ascórbico. O objetivo geral desta tese de doutorado foi propor soluções para três problemas das biorrefinarias de polpa cítrica, a falta de conhecimento sobre a hidrólise enzimática da pectina, o custo alto das pectinases que são usadas na hidrólise enzimática da pectina e a incapacidade de Saccharomyces cerevisiae produzir etanol a partir do ácido galacturônico liberado. Na primeira parte do trabalho, a hidrólise enzimática da pectina foi caracterizada, com intuito de entender a lentidão na liberação dos açúcares. Ficou demonstrado, pela primeira vez, que existe uma desaceleração significativa na liberação de açúcares redutores já nos primeiros minutos de hidrólise de pectina por complexos pectinolíticos. Foi mostrado que, com esta falta de linearidade no início da reação, o tempo de incubação escolhido para um ensaio tem grande efeito na atividade medida. De fato, é destacada a necessidade de estabelecer um protocolo padrão para a determinação da atividade pectinolítica, em termos da temperatura, da concentração de pectina e do tempo de incubação. Somente com esta padronização vai ser possível comparar processos de produção de pectinases realizados por grupos diferentes. Na segunda parte do trabalho, foi desenvolvido um processo, em escala piloto, para a produção de pectinases em fermentação em estado sólido, com intuito de baixar o custo destas enzimas. O processo foi realizado com 15 Kg (base seca) de um substrato composto por polpa cítrica e bagaço de cana, na razão de 51,6 a 48,4, (m/m). O uso de uma proporção alta de bagaço propiciou um leito de alta porosidade e, desta maneira, o problema de superaquecimento do leito, que é comum em processos de fermentação em estado sólido, foi completamente evitado. Também foi demonstrado que, no final da fermentação, o sólido pode ser secado e adicionado diretamente a uma solução de pectina para efetuar sua hidrólise; desta maneira, os custos de recuperação e concentração das pectinases são evitados. Na terceira parte do trabalho, foi construída uma linhagem de S. cerevisiae capaz de consumir o ácido D-galacturônico. Isto foi conseguido pela integração de uma via heteróloga, proveniente de fungos filamentosos, de catabolismo de ácido D-galacturônico. Este estudo representa o primeiro passo na construção de uma cepa de S. cerevisiae capaz de produzir etanol a partir de ácido D-galacturônico, sendo que este açúcar chega a compor cerca de 18% do total de açúcares em hidrolisados de polpa cítrica. Abstract: Biorefineries are industrial installations that utilize biomass as a raw material for the production of chemicals, fuels and energy, in a manner analogous to petroleum refineries. One raw material that has the potential to be utilized in Brazil is citrus pulp, the residue of the processing of orange juice. This residue is produced in large quantities in Brazil, which is the world's largest producer of orange juice. Currently, citrus pulp has no significant commercial value; it is typically used as a supplement for cattle feed, but the drying costs mean that the return is minimal. However, citrus pulp is a source of limonene and pectin, for which extraction processes are already established. Further, the carbohydrates in the pulp can be processed into ethanol and some acids with high added value, such as D-galacturonic acid, L-galactonic acid, mucic acid and ascorbic acid. The overall objective of this thesis was to propose solutions to three problems faced by citrus pulp biorefineries: the lack of knowledge about enzymatic hydrolysis, the high cost of the pectinases that are used in the enzymatic hydrolysis of pectin and the inability of Saccharomyces cerevisiae to produce ethanol from the galacturonic acid released in such hydrolysis processes. In the first part of the work, the enzymatic hydrolysis of pectin was characterized, with the aim of understanding why the release of sugars is so slow. For the first time, it was demonstrated that there is a significant deceleration in the release of reducing sugars within the first minutes of pectin hydrolysis by pectinases. It was shown that, with this lack of linearity at the start of the reaction, the incubation time that is chosen for an assay of pectinase activity has a large effect on the activity that is measured. In fact, the thesis raises the need to establish a standard protocol for the determination of pectinolytic activity, in terms of temperature, pectin concentration and time of incubation. Only with such standardization will it be possible to compare pectinase production processes proposed by different research groups. In the second part of the work, a pilot-scale process was developed for the production of pectinases in solid-state fermentation, with the aim of lowering the cost of these enzymes. The process was conducted with 15 kg (dry basis) of a substrate composed of citrus pulp and sugarcane bagasse, in a ratio of 51.6 to 48.4 (by mass). The use of a high proportion of bagasse ensured the bed had a high porosity and, consequently, the problem of overheating of the bed, which is common in solid-state fermentation processes, was completely avoided. It was also shown that, at the end of the fermentation, the solid can be dried and added directly to a pectin solution to effect its hydrolysis; thereby avoiding the need to recover and concentrate the pectinases. In the third part of the work, a strain of S. cerevisiae was constructed that is capable of consuming D-galacturonic acid. This was achieved by integration of a heterologous pathway for the catabolism of D-galacturonic acid from filamentous fungi. This study represents the first step in the construction of a strain of S. cerevisiae capable of producing ethanol from D-galacturonic acid, which represents about 18% of the total sugars in citrus pulp hydrolysates. Resumo: O presente trabalho teve como objetivo determinar as principais propriedades termofísicas envolvidas no processamento térmico de polpa de abacaxi, em especial no congelamento. Em adição, foi investigada a hipótese de que o comportamento das propriedades termofísicas da polpa de abacaxi pudessem ser previstas através de um sistema modelo constituído de solução de sacarose. Os sistemas constituídos de polpa de abacaxi ou solução de sacarose tiveram suas propriedades medidas a duas concentrações: 15 e 25 °Brix. Sua caracterização foi feita determinando-se o teor de sólidos totais e solúveis (°Brix), umidade e açúcares totais e redutores. A polpa de abacaxi também foi caracterizada através do conteúdo de fibras. A densidade foi medida a 25°C, utilizando-se picnômetros. A difusividade térmica foi medida com auxilio de uma cápsula metálica de geometria de cilindro infinito, acima do ponto de início de congelamento. Um calorímetro de mistura foi utilizado para a construção de curvas de entalpia-temperatura. A partir dessas curvas foram obtidas curvas de calor específico-temperatura e determinados o ponto de início de congelamento e a fração de água ligada. Uma determinação experimental de ponto de início de congelamento também foi realizada. Os resultados obtidos foram coerentes com propriedades termofísícas de sucos e polpas de frutas, encontrados em literatura. Das propriedades estudadas, apenas a densidade e algumas faixas de entalpia puderam ser previstas através do sistema modelo de solução de sacarose
Abstract: The objective of the present work was to determine die main thermophysical properties related to thermal processing of pineapple pulp, especially in freezing process. In addition, the hypothesis of utilizing a model system constituted of sucrose solution to predict thermophysical properties of pineapple pulp, was investigated. The pineapple pulp and sucrose solution systems were evaluated in two concentrations: 15 and 25 "Brix. These systems were characterized on basis of total and soluble solids (°Brix), moisture and amount of total and reducing sugars. Pineapple pulp was also characterized by fiber contents. Density was measured at 25°C, utilizing pycnometers. Thermal diffusivity was measured using a metallic capsule with infinity cylinder geometry, above the initial freezing point. The method of mixtures was used to construct enthalpy-temperature curves. Using these curves, specific heat-temperature curves was obtained and the initial freezing point and the bound water fraction were determined. An experimental measure of the initial freezing point was also realized. The acquired results were consistent with thermophysical properties of juice and pulp fruit found in literature. Of all properties studied, only density and some enthalpy ranges could have been predicted successfully by sucrose solution model systems

Resumo: Neste trabalho, o objetivo foi determinar as principais propriedades termofísicas envolvidas no processamento térmico da polpa de manga de variedade Tommy Atkíns. Para esta determinação, utilizou-se quatro amostras de polpa de manga: a integral, a peneirada, a centrifugada e a concentrada. A polpa integral (12,7 °Brix) foi produzida era laboratório e refinada em um despolpador de malha 1.8 mm. A polpa integral foi penetrada em uma malha 0.149 mm, centrifugada e concentrada até 30 °Brix. A caracterização da polpa foi feita determinando-se o teor de sólidos solúveis e insolúveis, umidade, cinzas, açúcares totais e redutores, pH, acidez total e pectina. A densidade foi medida por picnometria nas temperaturas de 10°C, 20°C 30°C. 40°C e 50°C. A difiisividade térmica foi medida acima do ponto de inicio de congelamento utilizando uma cápsula cilíndrica. O calor específico foi medido através de um Calorímetro Diferencial de Varredura (DSC) de -40°C a 40°C. e também com o auxílio do DSC foi possível determinar algumas propriedades associadas ao congelamento, como a temperatura de início e de pico de fusão, a entalpia de fusão e o conteúdo de água não congelada. Alguns modelos encontrados na literatura foram utilizados para o cálculo da densidade, da difusrvidade térmica, do calor específico e da condutividade térmica, e os valores obtidos foram comparados aos dados experimentais das propriedades termofísicas. Observou-se que os modelos utilizados podem predizer as propriedades termofísicas da polpa de manga
Abstract: The objective of this work was to identify the main thermophysical properties involved in the thermal processing of mango pulp (variety Tommy Atkins). Four different types of mango pulp were used in order to identify these properties: whole, screened, centrifuged and concentrated pulp. In order to obtain these four types of pulp, the whole pulp (12,7º Brix), obtained in the laboratory, was refined using a finisher with a 1.8 mm mesh,screened using a 0.149 mm mesh, centrifuged and concentrated to 30º Brix. The characterization of the pulp was effected by the determination of the soluble and insoluble solids in the pulp, moisture, ash, total and reducing sugar, pH, total acids and pectin. The density was determined using pycnometers at 10 °C, 20 °C, 30 WC, 40 °C and 50 °C The thermal diffusivity was determined above the freezing point using a cylindrical capsule. A Differential Scanning Calorimeter was used to determine the specific heat, in the range from -40 °C to 40 "C, and also properties such as the initial temperature of melting and melting point, enthalpy of melting and unfrozen water content of the pulp. Some of the thermophysical properties measured in this work, such as density, specific heat, thermal diffusivity and thermal conductivity were also calculated by some models found in the literature. A comparison of the results showed that these models could reasonably predict the thermophysical properties of mango pulp
