Résumé :
The present work deals with a theoretical model of catalysis by enzymes entrapped in reverse micelles. Three aspects of the enzyme-reverse-micelle system have been considered: structure, dynamics and enzyme distribution and catalysis in reverse micelles. A proposed structural model of reverse micelles [El Seoud (1984) in Reverse Micelles (Luisi, P. L. & Straub, B. E., eds.), p. 81, Plenum Press, New York] consists of three domains: surfactant apolar tails, bound water and free water. Dynamics are based on a dynamic equilibrium of association-dissociation that lead one to consider the dispersed polar phase as a pseudo-continuous phase [Luisi, Giomini, Pileni & Robinson (1988) Biochim. Biophys. Acta 947, 207-246]. Enzyme is distributed among the reverse-micelle domains and it expresses a catalytic constant for each one of them. The overall activity is calculated taking into account the volume in which enzyme is solubilized, and expressed as a function of the whole volume (V). The characteristic parameters of reverse micelles, omega 0 (= [H2O]/[surfactant]) and theta (= % water, v/v), were investigated as modulators of enzymic activity. Three basic patterns of modulation by omega 0 were found depending on which domain the enzyme expressed the highest catalytic constant. Combinations of those basic patterns lead to other modulation types that can be found experimentally, such as superactivation. Other combinations predict behaviour patterns not described to date, such as superinhibition. Dependence of catalytic activity on theta was only stated at omega 0 values around a critical value, which coincides with the appearance of free water.

Résumé :
A new polypore in the family Hymenochaetaceae, Coltricia aridicola, is described and illustrated from China and the USA based on morphology and phylogenetic analysis. The species was found in northeast and southwest China and western USA, growing on soil in gymnosperm forests. It is characterized by an annual growth habit, centrally stipitate basidiocarps with regular pilei, pileal surface distinctly zonate, medium-sized pores as 3–4 per mm, a monomitic hyphal system, generative hyphae with simple septa, the presence of antler-like hyphal elements at the pileal surface, contextual hyphae almost twice the diameter of the tramal hyphae, thick-walled, yellowish, cyanophilous, oblong-ellipsoid basidiospores measuring 7–7.8 × 4.5–4.8 µm, and growth in dry environments such as burnt soil or open areas of ground in gymnosperm forests. The phylogenetic analysis based on ITS sequences showed that samples of C. aridicola nested in Coltricia, forming a monophyletic lineage with strong support (100 % BS, 100 % BP, 1.00 BPP). The differences between C. aridicola and its similar and related species in the genus are discussed.

Résumé :
In this present study, five new wood-inhabiting fungal taxa, Botryobasidium gossypirubiginosum, Botryobasidium incanum, Botryobasidium yunnanense, Coltricia zixishanensis, and Coltriciella yunnanensis are proposed. Botryobasidium gossypirubiginosum is distinguished by its slightly rubiginous hymenial surface, monomitic hyphal system, which branches at right angles, and subglobose, smooth basidiospores (14–17.5 × 13–15.5 µm); B. incanum is characterized by its white to incanus basidiomata having a hypochnoid hymenial surface, and ellipsoid, smooth basidiospores (6.5–8.5 × 3.5–5 µm); B. yunnanense is characterized by its buff to slightly yellowish hymenial surface, monomitic hyphal system, and broadly ellipsoid to globose, smooth, thick-walled basidiospores (11.5–14.5 × 9.5–10.5 µm); Coltricia zixishanensis differs in its rust brown pileal surface, and ellipsoid, thick-walled basidiospores (5–6.5 × 4–4.5 µm). Coltriciella yunnanensis is distinguished by its tiny pilei, short stipe, and navicular, verrucose basidiospores (10.5–12.5 × 6–7 µm). Sequences of ITS and nLSU genes were used for phylogenetic analyses using the maximum likelihood, maximum parsimony, and Bayesian inference methods. The phylogenetic results inferred from ITS sequences revealed that B. gossypirubiginosum was closely related to B. robustius; the species B. incanum was grouped with B. vagum; B. yunnanense was related to B. indicum. The species C. zixishanensis was grouped with C. confluens and C. perennis. ITS sequences revealed that C. zixishanensis was grouped into the genus Coltriciella, in which it was grouped with Co. globosa and Co. pseudodependens.

Résumé :
The maize pathogen Fusarium verticillioides and their mycotoxins cause damage to plants, animals, and human health. This work aimed to evaluate the effect of crude extracts (CEs) from Agaricus subrufescens, Lentinula edodes, and Pleurotus ostreatus fruiting bodies on in vitro production of biomass and mycotoxins by two strains of F. verticillioides. Stipes and pilei were separated before extraction for A. subrufescens and L. edodes. Comparative metabolomics and dereplication of phenolic compounds were used to analyze all CEs. Mushroom CEs did not significantly inhibit the production of mycelial biomass at concentrations of 2 mg mL−1. CEs from A. subrufescens (stipes and pilei) and L. edodes pilei inhibited the production of fumonisins B1 + B2 + B3 by 54% to 80%, whereas CE from P. ostreatus had no effect. In contrast, CE from L. edodes stipes dramatically increased the concentration of fumonisins in culture media. Fusaric acid concentration was decreased in cultures by all CEs except L. edodes stipes. Differences in phenolic composition of the extracts may explain the different effects of the CE treatments on the production of mycotoxins. The opposing activities of stipes and pilei from L. edodes offer an opportunity to search for active compounds to control the mycotoxin production by F. verticillioides.

Hashizume, Keiji. « A Prospective Comparison of CT-Epidurogram Between Th1-Transforaminal Epidural Injection and Th1/2-Parasagittal Interlaminar Epidural Injection for Cervical Upper Limb Pain ». Pain Physician 2, no 22.2 (11 mars 2019) : 165–76. http://dx.doi.org/10.36076/ppj/2019.22.165.

Texte intégral
Résumé :
Background: Cervical epidural injections for treating neck and upper limb pain are performed by 2 methods: transforaminal and interlaminar. Many serious complications caused by inadvertent intravascular injection have been reported with the use of cervical transforaminal epidural steroid injection through the anterior-lateral approach. Despite international practical guidelines that have been proposed, cervical transforaminal epidural injection is still less recommended than cervical interlaminar epidural injection. Objectives: The objective of this study is to introduce Th1-transforaminal epidural injection (Th1- TFEI) through the posterior-lateral approach, compare the injectate spread in Th1-TFEI with that of Th1/2-parasaggital interlaminar epidural injection (Th1/2-pILEI), and clarify the clinical characteristics of Th1-TFEI. Study Design: This research involved a prospective study of 30 patients receiving both Th1-TFEI and Th1/2-pILEI. Methods: Thirty patients with unilateral upper limb pain were enrolled for this prospective study. Th1-TFEI and Th1/2-pILEI were administered on each case in random order under fluoroscopy, and computed tomographic (CT) epidurograms were compared. Changes in circulatory dynamics, presence of Horner’s syndrome, changes in the Numerical Rating Scale (NRS-11), and adverse events were investigated. Results: Patients included 15 men and 15 women and included 24 cases of cervical spine disease and 6 cases with other upper limb pain. The Th1-TFEI group had significantly higher rates of “Th1 root filling” (100%), “ventral spread” (70.0%), and “lateral limitation” (26.7%) compared to the Th1/2-pILEI group. In the Th1-TFEI group, cephalad spread averaged 2.97 vertebral bodies, reaching approximately up to C6. The Th1/2-pILEI group had an average of 4.76 vertebral bodies, approximately up to C4. The 2 groups showed significant differences in cephalad spread. Horner’s syndrome appeared in the Th1-TFEI group at a rate of 56.7%, significantly higher than that in the Th1/2-pILEI group at 17.2%. The presence of Horner’s syndrome showed significant correlations with “ventral spread” and “spread up to C6.” There were no significant differences in NRS-11 improvement and changes in circulatory dynamics between the groups. There were no major complications. Limitations: The components of injectate were standardized; however, the needle gauge numbers were varied. In addition, interpretation of the CT-epidurogram was not blinded. The sample size was small; therefore, multivariate analysis was not possible. Conclusions: CT-epidurogram comparison revealed that Th1/2-pILEI was not localized on the injection side, and there was better dorsal spread – although ventral spread was small. Contrarily, Th1-TFEI was localized on the injection side, and better ventral spread was shown while cephalad spread was limited. We expected the addition of a sympathetic block effect suggested by the Horner’s syndrome as well as the merits of the ventral spread. However, short-term clinical effects were equal to those of Th1/2 pILEI. In future research, we need to standardize the diseases to include and to increase the number of cases to enable evaluation of clinical effectiveness.Key words: Epidural, cervical, transforaminal, interlaminar, fluoroscopy, CT-epidurogram, dorsal, ventral, cephalad, Horner’s syndrome

Résumé :
The composition of the substrate is one of the most critical factors influencing the quality as well as the nutritional value and bioactive content of mushrooms. Therefore, the effects of various substrates, such as barley and oat straw (BOS), beech wood shavings (BWS), coffee residue (CR), rice bark (RB) and wheat straw (WS, control substrate), on the biochemical properties (lipid, protein, polysaccharide, glucan, ash, and mineral content, fatty acids and tocopherols composition), total phenolic compounds and antioxidant activity of Pleurotus mushrooms, P. ostreatus (strains AMRL 144, 150) and P. eryngii (strains AMRL 166, 173-6), cultivated in ‘bag-logs’, was examined. Proximate analysis of A. bisporus and A. subrufescens grown on two different composts (C/N ratios of 10 and 13) was conducted, too. The whole carposomes, pilei and stipes were analyzed. Results showed that BOS, RB, BWSs and CR improved the antioxidant activity of Pleurotus species and their nutritional characteristics. Both pilei and stipes were rich in polysaccharides (27.51–67.37 and 22.46–39.08%, w/w, for Pleurotus and Agaricus spp., respectively), lipids (0.74–8.70 and 5.80–9.92%, w/w), proteins (6.52–37.04 and 25.40–44.26, w/w, for Pleurotus and Agaricus spp., respectively) and total phenolic compounds (10.41–70.67 and 7.85–16.89 mg gallic acid equivalent/g for Pleurotus and Agaricus spp., respectively), while they contained important quantities of unsaturated FAs of nutritional and medicinal importance. Pilei were richer in proteins, total phenolic compounds and enhanced antioxidant activity and reducing power than stipes, whereas stipes were richer in IPSs and glucans compared to the corresponding pilei. Thus, mushroom cultivation could upgrade rejected agro-industrial residues and wastes to new uses as substrates for the production of mushrooms with specific nutritional and medicinal attributes.

Résumé :
In this study, phylogenetic analyses of Phellodon from China were carried out based on sequences from the internal transcribed spacer (ITS) regions, the large subunit of nuclear ribosomal RNA gene (nLSU), the small subunit of nuclear ribosomal RNA gene (nSSU), the largest subunit of RNA polymerase II (RPB1), and the second largest subunit of RNA polymerase II (RPB2), combined with morphological characters of the collected specimens in China. The fruiting bodies of the specimens were used to observe their characteristics, and three new species of Phellodon are discovered. Phellodon crassipileatus is characterized by its pale brown to dark brown pileal surface, tomentose pileal margin, white spines, and the presence of clamp connections in generative hyphae of pileal surface, context, and stipe. Phellodon griseofuscus is characterized by its dark brown to black pileal surface, white to pale brown pileal margin, the presence of both simple septa and clamp connections in generative hyphae of spines, and moderately long basidia. Phellodon perchocolatus is characterized by its woody and broad pileus, brown to greyish brown pileal surface when fresh, tomentose pileal margin when young, which becomes glabrous with age, and the presence of both simple septa and clamp connections in the generative hyphae of the spines. This is the first time both single and multi-genes analysis is used in such a phylogenetic and taxonomic study on Phellodon, which can provide the basis for the phylogenetic study of the genus.

Résumé :
Fruit bodies (separately pilei and stems) of mushrooms <i>Pleurotus ostreatus</i> (four strains) and <i>Pleurotus eryngii</i> were characterised as a source of polysaccharides. The contents of glucans and dietary fibres were determined with using the respective Megazyme enzymatic kits. Enzymatic analysis of the fruit bodies confirmed significant differences in the contents of these components among the species and strains. The stems contained more insoluble dietary fibres than the pilei in all the cases and more β-glucans in most cases. However, relatively high contents of β-glucan (20–50% of dry matter) could be a result of incomplete enzymatic hydrolysis of insoluble α-1,3-glucans. Nevertheless, low food quality stems of mushrooms <i>Pleurotus</i> sp. could be a valuable source of cell wall glucans for the preparation of food supplements.

The sugar beet is one of the most important crops for both social and economic reasons, even though the area under sugar beet cultivation in the Red River valley of North Dakota and Minnesota is comparatively smaller that of corn and other crop lands. It generates a large economic activity in local and regional level with a greater impact on jobs and stimulation of agriculture, transportation, and farm economy. Sugar beet transportation takes place in two stages in Red River Valley: the first step is from farms to piling centers (pilers) and the second step from pilers to processing facilities. This study focuses on the problem of optimizing piler locations based on supply variation. Sugar beet supply and harvest varies significantly due to numerous reasons such as weather, water availability, and different maturity dates for the crop. This provides for a variable optimal harvesting time based on the plant maturity and sugar content. Sub-optimized pilers location result in the high transportation and utilization costs. The objective of this study is to minimize the sum of transportation costs to and from pilers and the pilers utilization cost. A two-step algorithm based on the geographical information system (GIS) with global optimization method is used to solve this problem. This method will also be useful for infrastructure decision makers such as planners and engineers to predict the truck volume on rural roads.

This article is an attempt to analyse the historical memory of Witold Pilecki functioning in the reference literature and collective consciousness. The author concentrates on their idealising and simplistic elements, which lead to mythologization of Pilecki, and asks about the genesis and purpose of creation of myths about the Captain. Basing on an analysis of the sources, Cuber questions the legitimacy of the two popular expressions used with regard to Pilecki, that is, “volunteer to Auschwitz” and “the author of the first report about the Holocaust.” In this way the author points out the necessity to correct and supplement Pilecki’s biography by means of a careful and cautious analysis of all the available sources.

Résumé :
ABSTRACT Pseudomonas stutzeri has type IV pili for which the pilA gene (here termed pilAI) provides the structural protein and which are required for DNA uptake and natural genetic transformation. Downstream of pilAIwe identified a gene, termed pilAII, coding for a deduced protein with a size similar to that of PilAI with 55% amino acid sequence identity and with a typical leader peptide including a leader peptidase cleavage site. Fusions to lacZ revealed that pilAII is expressed only about 10% compared topilAI, although the genes are cotranscribed as shown by reverse transcription-PCR. Surprisingly, insertional inactivation ofpilAII produced a hypertransformation phenotype giving about 16-fold-increased transformation frequencies. Hypertransformation also occurred in pilAI pilAII double mutants expressing heterologous pilA genes of nontransformable bacteria, like Pseudomonas aeruginosa or Dichelobacter nodosus. The overexpression of pilAII decreased transformation up to 5,000-fold compared to that of thepilAII mutant. However, neither inactivation ofpilAII nor its overexpression affected the amounts of [3H]thymidine-labeled DNA that were competence-specifically bound and taken up by the cells. In thepilAII mutant, the transformation by purified single-stranded DNA (which depends on comA andexbB, as does transformation by duplex DNA) was also increased 17-fold. It is concluded that PilAII suppresses a step in transformation after the uptake of duplex DNA into the cell and perhaps before its translocation into the cytoplasm. The idea that the degree of the transformability of cells could be permanently adjusted by the expression level of an antagonistic protein is discussed.

Résumé :
Abstract Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare disease of controversial origin recently recognized as a neoplasm deriving from plasmacytoid dendritic cells (pDCs). Nevertheless, it remains an orphan tumor with obscure biology and dismal prognosis. In this study, we aimed to: 1) molecularly define the cellular counterpart of BPDCN and its relationship with other leukemias; 2) identify genes and cellular programs deregulated in the tumor; and 3) delineate novel potential therapeutic targets. To address these issues we collected and studied by gene expression profile (GEP) 27 BPDCN cases as well as 8 samples of non neoplastic pDCs. Further, a panel of samples including, myeloid precursors (MPs, N=4), lymphoid precursors (LPs, N=9), acute myeloid leukemias (AMLs, N=132), and acute lymphoblastic leukemias (ALLs, N=155) was analyzed. Validation was performed by immunohistochemistry (IHC) on tissue-microarrays, while functional experiments were carried out by using the CAL-1 cell line (derived from a BPDCN case). First, we recognized the cellular derivation of BPDCN, which proved to originate from the myeloid lineage and in particular from resting pDCs. Second, by comparing the GEP of BPDCN and resting pDCs, we identified genes and cellular programs deregulated in the tumor. Following, based on an integrated bio-informatic approach, including four different tools, we uncovered the aberrant activation of the NF-kB pathway that was confirmed in independent assays. Interestingly, among other molecules, we identified BCL2 and IRF4, two well known NFkB targets, as aberrantly upregulated in neoplastic samples and confirmed this observation by IHC. We then tested whether NFkB inhibition could represent a potential therapeutic strategy in this setting. We treated BPDCN cells ex vivowith either the proteasome inhibitor bortezomib or the selective IKKB inhibitor BMS-345541 and found them to be effective in inducing cell cycle arrest and apoptosis at relatively low dosage. By contrast, BPDCN cells turned out to be virtually insensitive to cytarabine, one of the most used drug in this condition. GEP and immunocytochemistry were then successfully used to prove that cell death was accompanied by NFkB shut-off. In conclusion, we identified a molecular signature representative of the transcriptional abnormalities of BPDCN and developed a cellular model proposing the first molecular targeted therapeutic approach in the setting of this currently incurable disease.
