Introduction: We describe a rare tumor arising from the prostate gland: Perivascular Epithelioid Cells tumor (PEC-ome). A 54-years old was treated for acute urinary retention with alpha-blockers at presentation due to benign prostate enlargement (65 cc) with asymmetric middle lobe and regular PSA (0.92 ng/ml). After 5 months, patient developed a second acute urinary retention episode and nodules in the left lung; he was treated with transurethral resection of the prostate and left lobectomy. Results: Histological examination of prostate and lung tissue gave the same diagnosis: leiomyosarcoma with atypical morphological features and patient was observed for 4 months. Considering the uncommon diagnosis, pathological review by the uro-pathologist at our Hospital was done. Additional immunohistochemistry was done and both tumors showed similar and typical features of metastatic PEC-ome (T1b N0 M1). Therefore a new staging showed local and distant progression with prostatic mass and small lung metastasis. Three cycles of Gemcitabine and Pazopanib were administered, but 2 months later a new urinary retention occurred, despite chemotherapy. Patient referred to our Hospital for salvage pelvic surgery with lymph node dissection. Final pathological diagnosis was PECome of the prostate stage pT4 pN0 R0 M1. Conclusions: PEC-ome is a rare but rapidly invasive mesothelial tumor with early metastatic potential. When this tumors originates from the fibromuscular stroma of the prostate it mimics benign prostatic enlargement and causes LUTS. Expert pathology aided by immunoisthochemistry is the cornerstone of diagnosis. There are no pathognomonic imaging on ultrasound or symptoms suggesting the presence of PECome in early stage. A multidisciplinary approach is necessary and radical surgery should be done to treat this aggressive cancer.

Abstract Neoplasms of perivascular epithelioid cells (PEComas) have in common the coexpression of muscle and melanocytic immunohistochemical markers. Although this group includes entities with distinct clinical features, such as angiomyolipoma, clear cell sugar tumor of the lung, and lymphangioleiomyomatosis, similar tumors have been documented in an increasing diversity of locations. The term PEComa is now generally used in reference to these lesions that are not angiomyolipomas, clear cell sugar tumors, or lymphangioleiomyomatoses. While most reported PEComas have behaved in a benign fashion, malignant PEComas have occasionally been documented. We present a case of hepatic PEComa with benign histologic features, which nonetheless presented with metastases to multiple sites nearly 9 years later. This case represents the second documented malignant PEComa of the liver, as well as the longest follow-up of a surviving patient with a malignant PEComa, emphasizing both the need for criteria that more accurately predict the behavior of PEComas and the necessity of long-term follow-up of patients with PEComas.

Malignant PEComas are rare mesenchymal neoplasms. These tumors harbor distinct myomelanocytic phenotype. The PEComa family of tumors includes lymphangioleiomyomatosis, angiomyolipoma, clear cell sugar tumor of the lung, and myomelanocytic tumor of the falciparum ligament/ligamentum teres. PEComas have no known normal cell counterpart. Majority of PEComas are benign and occur predominantly in the middle-age women. These tumors are commonly encountered in the uterus. Herein, we report a 20-year-old woman with a left inguinal mass metastatic to orbit, brain, lumbar spine, and skin at presentation. To our knowledge, this is the first case of metastatic PEComa to the orbit. This is the third case of primary PEComa of the inguinal area.

Ce travail de thèse s’articule autour de trois projets indépendants dont le but est de mieux caractériser trois tumeurs mésenchymateuses grâce à une approche multi-omique intégrative.Les tumeurs thoraciques indifférenciées SMARCA4-déficientes (SMARCA4-UT), initialement « sarcomes » semblaient répondre aux inhibiteurs de point de contrôle immunitaire (ICIs) comme d’autres tumeurs SWI/SNF déficientes, sans qu’aucune caractérisation de leur microenvironnement tumoral (TME) ne soit faite pour le comprendre. Grâce à des immunomarquages et à une analyse transcriptomique, nous avons mis en évidence un TME désertique avec une efficacité limitée des ICIs, en lien avec la cellule d’origine. Les tumeurs des cellules épithélioïdes périvasculaires (PEComes) forment un ensemble hétérogène de tumeurs coexprimant des marqueurs mélanocytaires et musculaires lisses pour lesquelles deux types génomiques se distinguent. Grâce à notre analyse, nous avons démontré qu’il existait d’autres réarrangements que ceux impliquant TFE3 et qu’il existait une classification transcriptomique pronostique de quatre sous types de PEComes, chacun étant enrichi d’un profil génomique et présentant des vulnérabilités différentes sur le plan thérapeutique. Les tumeurs desmoïdes (TDs) sont des tumeurs bénignes localement agressives dont l’hétérogénéité dans l’évolution tumorale est peu comprise. Grâce à notre analyse, nous avons mis en évidence que plus de 50% des TDs avaient des mutations dans un des gènes remodelant la chromatine et que parmi les deux sous-types transcriptomiques identifiés, le type immuno-myogénique doté d’un programme transcriptomique proche de celui des muscles, activait les voies de l’immunité évoquant un potentiel thérapeutique des ICIs
This thesis work takes the form of three independent projects aimed at better characterizing three mesenchymal tumors through an integrative multi-omics approach.The thoracic undifferentiated SMARCA4-deficient tumors (SMARCA4-UT), initially classified as "sarcomas," appeared to respond to immune checkpoint inhibitors (ICIs) similarly to other SWI/SNF-deficient tumors, despite no characterization of their tumor microenvironment (TME) being done to understand this response. Through immunostaining and transcriptomic analysis, we highlighted a desert-like TME with limited ICI efficacy, linked to the tumor’s cell of origin. Perivascular epithelioid cell tumors (PEComas) form a heterogeneous group of tumors co-expressing melanocytic and smooth muscle markers, with two distinct molecular types identified. Our analysis demonstrated that there are additional rearrangements beyond those involving TFE3 and provided a prognostic transcriptomic classification of four PEComa subtypes, each enriched with a unique genomic profile and presenting different therapeutic vulnerabilities. Desmoid tumors (TDs) are benign, locally aggressive tumors with poorly understood heterogeneity in tumor evolution. Our analyses revealed that more than 50% of TDs had mutations in chromatin remodeling genes and that among the two identified transcriptomic subtypes, the immuno-myogenic subtype, with a transcriptomic program similar to muscles, activated immune pathways suggesting a potential therapeutic benefit from ICIs

La sclérose tubéreuse complexe (TSC) est une maladie génétique caractérisée par une croissance des hamartomes dans différents organes y compris le cerveau, les reins, les poumons, la peau et le cœur. Ces lésions sont des sources de morbidité et de mortalité chez les patients TSC, car ils peuvent provoquerl’ épilepsie, l’autisme, le retard de développement et l’insuffisance rénale et pulmonaire. Les causes connues de TSC sont la perte de la fonction et les mutations des gènes TSC1 et TSC2. La majorité des lésions TSC contient plusieurs types cellulaires de la lignée mésenchymateuse, comme dans le cas des angiomyolipomes, l’lymphangioleiomyomatose et les angiofibromes. Un type unique de cellules épithélioïdes périvasculaires nommé (PEC) est constamment présent dans les lésions de TSC mésenchymateuses, comme angiomyolipomes et lymphangioleiomyomatose, basant sur les caractérisations morphologiques et l'expression des marqueurs communs mélanocytaires et myogéniques. Par conséquent, ces lésions sont officiellement classées, ainsi que d'autres tumeurs, comme PEComes. Leur origine cellulaire et les mécanismes moléculaires impliqués dans la pathologie restent à élucider. Ici, nous avons généré un modèle souris mosaïque TSC1 knockout qui développe des lésions rénales mésenchymateuses récapitulant périvasculaire épithélioïde cellules tumorales humaines (Pecoma) observés chez les patients TSC. Nous avons identifié YAP, le co-activateur transcriptionnel de la voie Hippo, a été régulée d'une manière mTOR-dépendante dans les lésions rénales de notre TSC1 knockout souris et les échantillons de l’angiomyolipome humaines. L'inhibition de YAP avec des outils génétiques ou pharmacologiques atténue considérablement la prolifération et la survie des cellules nulles TSC1 in vivo et in vitro. En outre, l’accumulation de YAP dans les cellules déficientes TSC1 / TSC2 pourra être dû à la dégradation de la protéine altéré par le système de l’autophagosome / lysosome. Ainsi, la régulation de YAP par mTOR et l'autophagie est un nouveau mécanisme de contrôle de la croissance, l'activité de YAP correspondant à la disponibilité des éléments nutritifs dans les conditions de croissance permissives. Il pourra servir comme une cible thérapeutique potentielle pour TSC et d'autres maladies avec une activité de mTOR dérégulée
The Tuberous Sclerosis Complex (TSC) is a genetic disease characterized by growth of hamartomas in different organs including brain, kidney, lung, skin, and heart. These lesions are sources of morbidity and mortality in patients with TSC, as they may cause intractable epilepsy, autism, developmental delay, renal and pulmonary failure. Known causes of TSC are loss of function mutations in TSC1 and TSC2 genes. The majority of TSC lesions contain multiple cell types of the mesenchymal lineage, as in the case of angiomyolipomas, lymphangioleiomyomatosis and angiofibromas. A unique cell type named perivascular epithelioid cell (PEC) is constantly present in mesenchymal TSC lesions, such as angiomyolipomas and lymphangioleiomyomatosis, basing on morphological features and the common expression of melanocytic and myogenic markers. Therefore, these lesions are officially classified, along with other tumors, as PEComas. Their cell of origin and the molecular mechanisms underlying their pathogenesis remain poorly defined. Here we generated a novel mosaic Tsc1 knockout mouse model which develop renal mesenchymal lesions recapitulating human Perivascular Epithelioid Cell tumor (PEComa) observed in TSC patients. We identified YAP, the transcriptional coactivator of Hippo pathway, was upregulated in both renal lesions of TSC mouse model and human angiomyolipoma samples in a mTOR-dependent manner. Inhibition of YAP with genetic or pharmacological tools greatly attenuates the proliferation and survival of Tsc1 null cells in vivo and in vitro. Futhermore, we found YAP accumulation in TSC1/TSC2 deficient cells is due to impaired degradation of the protein through the autophagosome/lysosome system. Thus the regulation of YAP by mTOR and autophagy is a novel mechanism of growth control, matching YAP activity with nutrient availability under growth permissive conditions. It may serve as a potential therapeutical target for TSC and other diseases with dysregulated mTOR activity

Aux activites de peche proprement dites, s'ajoute un important secteur de transformation (21 conserveries, 9 unites de congelation et 9 fabriques de sous-produits). Sur le plab social, la filiere halieutique offre l'emploi a 10 000 pecheurs, et fait vivre indirectement environ 60 000 personnes. Les obstacles auxquels se heurte l'industrie des peches a agadir peuvent etre ramenes a deux goulots d'etranglement ; l'expatriation de la flotte hauturiere et la mauvaise valorisation des debarquements realises par la flottille cotiere
Agadir is the first moroccan fishing-port (181. 164 tons in 1985 worth 136 280 750 dh). Wealth in this harbour comes from the small pelagians, and among them the sardines. To the actual fishing activities, an important processing sector must be added (21 canneries, 9 freezing units and 9 factories of by-products). On a social point of view, the halieutic branch gives work to 10. 000 fishermen and so provides a living to about 60. 00 peoples. The obstacles met by the fishing industry en agadir can be summed up in two "bottlenecks" : the expatriation of the deep-sea fleet and the bad valorization of the unshipped cargo made by the inshore fishing fleet

This manual was written specifically as a guide for the beginner dissector to complete a prosected cadaver that will serve as an effective teaching and learning tool. The dissections are written in a manner that facilitates the preservation of more superfi- cial structures on the right side of the cadaver and deeper structures that otherwise would have been concealed on the left side of the cadaver. This format is ideal for students learning anatomy for the first time, as multiple planes can be compared on the same cadaver. The level of structures dissected and identified in these chapters was established with the understanding that the dissector’s knowledge of human anatomy is at the level of completion of a general undergraduate anatomy survey course.

La position phylogénétique des Cervidae dans l'infra-ordre des Pecora a longtemps fait l'objet de nombreuses controverses, dont la plus récente est la confrontation des données morphologiques aux données moléculaires. Dans le but d'élargir le débat, notre étude propose d'étudier le répertoire comportemental de douze espèces de Pecora dont neuf Cervidae, afin de préciser leurs relations de parentés. Les résultats de nos observations éthologiques ont ainsi permis d'identifier des « éthotypes hypothétiques ancestraux », qui montrent que les Cervidae forment un groupe monophylétique constitué par deux lignées : la sous-famille des Cervinae ou plésiométacarpaliens qui apparaît comme un groupe naturel et les Cervidae télémétacarpaliens, Hydropotinae et Capreolinae. Ces derniers présentent deux groupes, avec un clade qui correspond aux Odocoileini et aux Rangiferi, et un groupe paraphylétique, constitué par les Hydropotini, les Capreolini et les Alceini. Une origine eurasiatique pour l'ensemble des Cervidae a été avancé, ainsi que la perte secondaire des bois chez Hydropotes. D'autre part, les Moschidae apparaissent comme le groupe frère des Cervidae, et les Bovidae semblent plus apparentés aux Cervoidea que ne le sont les Giraffidae. Une analyse impliquant d'avantage de taxons serait nécessaire, non seulement pour affiner nos résultats et notre méthode d'utilisation des données éthologiques, mais aussi pour discuter des perspectives théoriques qu'engendre l'apport du comportement en systématique phylogénétique
The phylogenetic position of Cervidae in the Pecoran infraorder caused many controversies, of which the most recent is the confrontation of the morphological data to the molecular data. With an aim to enlarge the debate, our research proposes to study the behavioral repertory of twelve species of Pecora including nine Cervidae, in order to specify their interrelationships. The results of our ethologic observations thus allowed to identify "ancestral hypothetical éthotypes", which show that Cervidae form a monophyletic group made up by two lineages : plesiometacarpals Cervids or Cervinae which appear as a natural group and telemetacarpals Cervids, Hydropotinae and Capreolinae. The last one presents two groups, with a clade which corresponds to Odocoileini and Rangiferi, and a paraphyletic group, consisted by Hydropotini, Capreolini and Alceini. A eurasiatic origin for Cervidae was advanced, as well as Hydropotes is secondarily antlerless. On the other hand, Moschidae appear as the sister group of Cervidae, and Bovidae seem more related in Cervoidea than are to it Giraffidae. An analysis implying more taxa would be necessary, not only to refine our results and our method of using ethological data, but also to discuss of the theorical prospects generated by the behaviour content in phylogenetic systematics

This work was carried out in the northern portion of Industrial and Portuary Complex of Pecém, located in Fortaleza Metropolitan Region, Ceará State, Brazil, and aims to evaluate aquifer vulnerability through DRASTIC and GOD methods, and characterization of aquifer pollution hazard. In 2013, four steps of monitoring were made for 24 wells, from which it was possible to identify three main aquifer systems: Dunas, Barreiras and Fraturado. The monitored parameters were: pH, electrical conductivity, total dissolved solids, temperature, salinity, and water table. The pH results allowed classifying the groundwater as acids.Total dissolved solids concentrations are inside of the potability pattern and allowed classifying the groundwater as fresh water. The major acidity and salt concentrations were found during the dry season. The water table has average depth around 5 meters. The DRASTIC method showed indices between very low and very high, with predominance of high and very high vulnerability associated to the Dunas Aquifer, and moderate vulnerability associated to the Barreiras Aquifer. Areas with low and very low vulnerability occur in portions of the Barreiras Aquifer where the water table is most deep, and in the Fraturado Aquifer. Water table, recharge, and vadose zone were the parameters that most influenced these results. The GOD method showed indices between moderate and high, with high vulnerability associated to the Dunas Aquifer, and moderate vulnerability associated to the Barreiras and Fraturado aquifers. Water table was the parameter that most influenced the results of this method. In both methods, the vulnerability mapping showed contours very close to the ones of the geologic map, suggesting strong influence of the lithology in the evaluating process. Activities developed in the area were classified with subsurface contaminant load potential moderate and high. Aquifer pollution hazard obtained by GOD vulnerability and subsurface contaminant load potential indicates the dominance of areas with moderate pollution hazard. The results suggest that aquifer vulnerability and aquifer pollution hazard were underestimated during the construction of the industrial complex, regarding the area where each activity should be building. Comparing both methods, DRASTIC showed better results for studies that requires greater details, as EIAs and Master Plans. The GOD method showed better results for regional studies, as land zonings. The results also reaffirms the environmental fragility of great part of the area intended to the industrial complex, and reinforce the need for monitoring the industrial activities regarding pollution prevention.
Este trabalho foi realizado na porção norte do Complexo Industrial e Portuário do Pecém, localizado na Região Metropolitana de Fortaleza (CE), e teve por objetivo avaliar a vulnerabilidade dos aquíferos por meio dos métodos DRASTIC e GOD e caracterização do perigo de contaminação. Realizou-se quatro etapas de monitoramento em 24 poços ao longo de 2013, onde verificou-se a presença de três sistemas aquíferos: Dunas, Barreiras e Fissural. Os parâmetros monitorados foram: pH, temperatura, condutividade elétrica, sólidos totais dissolvidos, salinidade e nível estático. A medidas de pH permitiram classificar as águas subterrâneas como ácidas. As concentrações de Sólidos Totais Dissolvidos encontram-se dentro dos padrões de potabilidade e permitiram classificar as águas como “águas doces”. O método DRASTIC apresentou índices entre muito baixo a muito alto, com predomínio de vulnerabilidade alta a muito alta, associadas ao Aquífero Dunas, e vulnerabilidade moderada associada ao Aquífero Barreiras. As áreas de baixa e muito baixa vulnerabilidade ocorrem em porções do Barreiras com nível estático mais profundo e no Aquífero Fissural. O nível estático, a recarga e zona vadosa foram os parâmetros que mais influenciaram estes resultados. O método GOD apresentou índices entre moderado e alto, com alta vulnerabilidade associada ao Aquífero Dunas, e vulnerabilidade moderada associada aos Aquíferos Barreiras e Fissural. O nível estático foi o parâmetro que mais influenciou os resultados deste método. Nos dois métodos, o mapeamento da vulnerabilidade apresentou contornos muito próximos aos do mapeamento geológico, o que sugere forte influência da litologia no processo de avaliação. As atividades presentes na área foram classificadas com potencial de carga contaminante entre moderado e elevado. O perigo de contaminação obtido pelo método GOD e pelo potencial de carga contaminante indicou o predomínio de áreas de perigo moderado. Os resultados sugerem que a vulnerabilidade dos aquíferos e o perigo de contaminação foram subestimados na instalação do complexo, no que concerne à área onde cada atividade industrial poderia ser instalada. Na comparação entre os métodos, o método DRASTIC apresentou melhores resultados para estudos que requeiram maior detalhamento, como EIA/Rimas e Planos Diretores. O método GOD apresentou resultados mais satisfatórios para estudos regionais, como zoneamentos territoriais. Os resultados também reiteram a fragilidade ambiental de grande parte da área destinada à instalação do complexo industrial, e reforçam a necessidade de monitoramento das atividades industriais no que diz respeito a prevenção de contaminações.

This master´s thesis deals with issues of tube furnaces in the process industries, especially with a design of a transfer pipeline, which is one of the main parts of the furnace. According to the requirements, designed pipeline is stress analyzed using the two pipeline programs Japar and Caesar II, that are briefly described. In addition to this design several variants of analysis are performed and the results are compared. Comparison helps to explain the diversity of the programs. For example, the effect of elbows on the stress analysis. The master´s thesis also describes the factors that affect the working life of the furnace.

La comparaison de l'amenagement et l'organisation entre des ports de peche indonisiens, francais, belges et allemands met a jour trois modes de gestion differents, soit d'etat, semi-public ou municipal. L'etude de ces trois modes de gestion illustre les differences qui existent dans le domaine socio-culturel, economique, technique ou politique entre un pays developpe et un pays en voie de developpement. La prise en compte de la surface des eaux indonesiennes nous permet de constater que la potentialites sont importantes et que l'amenagement des ports de peche indonesiens doit etre developpe alors qu'aujourd'hui 87,5% des ports sont de type cotier. La presence de ports industriels de type oceanique comme le port de jakarta est indispensable dans un contexte de developpement d'un veritable arriere-pays a l'image de ceux rencontres autour des ports europeens. Ce developpement est favorise par la concentration d'usines de transformation et d'industries annexes. Il ne pourra se faire sans l'organisation des pecheurs et des armateurs, des mareyeurs-conserveurs et des petits commercants, ou bien encore l'amelioration des voies et des moyens de transport. .
The comparative study of arrangement and organization of the fishing ports in indonesia, france, belgium and german shows the existence of three different modes of management : management by state; management by state and private; and management by municipality. Intensive study of these modes of management bring forward the differences that exist in social and cultural lives, economy, technology and politic between the developed and the developing countries. Taking account of the vastness of indonesian water surface, we see that indonesia has a fremendous potentialities and that the arrangement of the fishing ports have to improve in the near future; as right now 87,5% of the indonesian ports is the type of coastal ports. The presence of industrial fishing port of oceanic type like that of jakarta is essential in the contex of the favorable development of fishing industry. The existence of the big port can stimulate the distribution network and the development of real hinterland activities in the same manner like that encountered in the european ports. This development is propelled by the concentration of the fish processing facilities and its related industries. .

Le monde de la peche est en crise, surtout depuis les annees 90. Pour remedier a ce probleme, differents systemes de gestion ont ete instaures a l'echelle mondiale, soit des mesures visant a diminuer l'effort de peche, soit la creation d'espaces proteges permettant la reconstitution des stocks. Confrontee a une baisse importante des rendements de la peche langoustiere, la corse a ete une des pionnieres en ma matiere en instaurant a la fin des annees 70 la mise en place de 9 cantonnements de peche sur le pourtour insulaire. La creation de ces cantonnements repose sur des mesures prises en etroite collaboration avec les differents pecheurs. Ils se differencient des autres systemes de protection generalement utilises par l'absence de surveillance, leur existence reposant essentiellement sur le respect de chacun. L'etude entreprise ici vise, par le suivi regulier du cantonnement de peche de calvi (de mai 1994 a decembre 1999), a etudier l'influence d'un tel outil de gestion sur un ensemble d'especes cibles. La premiere partie de cette etude a consiste a mettre en evidence, par la technique du comptage visuel sur transects fixes, l'existence d'un effet reserve. Dans les grandes lignes ce dernier c'est revele semblable a celui observe dans les autres systemes de protection plus generalement utilises (parcs nationaux, regionaux, reserves naturelles). Nos resultats ont mis en evidence pour la majorite des especes cibles des changements : ' de structure demographique, avec des proportions de grands individus plus elevees dans la reserve qu'au niveau des zones de peche, ' de densite et de biomasse, avec des valeurs egalement plus elevees au sein du cantonnement de peche, ' de repartition spatiale des individus, avec une remontee vers les faibles profondeurs au sein du cantonnement de peche. La seconde partie de ce travail avait un double but. ' dans un premier temps ; evaluer l'influence des mesures de protection sur les variations saisonnieres naturelles des parametres etudies en premiere partie. Notre etude a permis de mettre en evidence pour les especes cibles une accentuation des variations saisonnieres au sein du cantonnement de peche, avec des regroupements de reproducteurs plus importants durant les periodes de reproduction et un nombre de petits individus plus eleve durant les periodes faisant suite aux phases de recrutement. Ces remarques suggerent qu'en comparaison a un certain nombre de reserve marine, l'effet cascade au sein du cantonnement de peche de calvi est attenue. ' dans un deuxieme temps ; observer sur six annees d'etude l'evolution temporelle de l'effet reserve. A l'exception de la richesse specifique, qui a tendance a diminuer plus rapidement au niveau des zones de peche que dans le cantonnement, l'ensemble des parametres etudies reste relativement constant dans la reserve comme au sin des zones de peche. Une stabilite qui reposerait sur une acceleration de la migration vers les zones de peche des predateurs cotiers (grands individus des especes cibles), rapidement devenus trop nombreux dans un espace protege restreint. Malheureusement ces explications sont encore sous forme d'hypotheses qu'il conviendrait de verifier.
