PASTORIZIA ANTICA E MODERNA NELL'ITALIA CENTRALE: UNO STUDIO INTERDISCIPLINARE NEI MONTI DEL CICOLANOQuest'articolo è un contributo al dibattito attuale, tra archeologi, di come sia possibile ricostruire la pastorizia antica attraverso i dati archeologici. I monti del Cicolano oggi vengono sfruttati ogni estate dalle gente dei villaggi circostanti per tre scopi diversi: agricoltura, pastorizia stanziale, pastorizia transumante. Il progetto comprende uno studio geomorfologico, una ricognizione archeologica, una ricerca documentaria, e studi etnoarcheologici. Il regolare insediamento stagionale cominciò nella tarda preistoria e nel periodo romano i monti furono usati da agricoltori e da pastori forse transumanti. L'agricoltura arativa e la pastorizia risultano ambedue nei documenti medioevali e postmedioevali e si possono riconoscere nel materiale archeologico raccolto. Maè assai difficile identificare differenti tipi di pastorizia nei dati archeologici forniti dalla ricognizione. L'archeologia dell'insediamento, la cultura materiale portatile, la fauna, i residui botanici creati dagli agricolturi e pastori di oggi nel Cicolano mostrano le difficoltà nel distinguere i diversi tipi di uso dei terreni alti. Ma, almeno, ci danno utili suggerimenti per sviluppare le future metodologie.

ABSTRACTThe yeastPichia pastorisis a widely used host for recombinant protein production. Understanding its physiology at extremely low growth rates is a first step in the direction of decoupling product formation from cellular growth and therefore of biotechnological relevance. Retentostat cultivation is an excellent tool for studying microbes at extremely low specific growth rates but has so far not been implemented forP. pastoris. Retentostat feeding regimes were based on the maintenance energy requirement (mS) and maximum biomass yield on glucose (YX/Smax) estimated from steady-state glucose-limited chemostat cultures. Aerobic retentostat cultivation enabled reproducible, smooth transitions from a specific growth rate (μ) of 0.025 h−1to near-zero specific growth rates (μ < 0.001 h−1). At these near-zero specific growth rates, viability remained at least 97%. The value ofmSat near-zero growth rates was 3.1 ± 0.1 mg glucose per g biomass and h, which was 3-fold lower than themSestimated from faster-growing chemostat cultures. This difference indicated thatP. pastorisreduces its maintenance energy requirement at extremely low μ, a phenomenon not previously observed in eukaryotes. Intracellular levels of glycogen and trehalose increased, while μ progressively declined during retentostat cultivation. Transcriptional reprogramming toward zero growth included the upregulation of many transcription factors as well as stress-related genes and the downregulation of cell cycle genes. This study underlines the relevance of comparative analysis of maintenance energy metabolism, which has an important impact on large-scale industrial processes.IMPORTANCEThe yeastPichia pastorisnaturally lives on trees and can utilize different carbon sources, among them glucose, glycerol, and methanol. In biotechnology, it is widely used for the production of recombinant proteins. For both the understanding of life in its natural habitat and optimized production processes, a better understanding of cell physiology at an extremely low growth rate would be of extraordinary value. Therefore, we have grownP. pastorisin a retentostat, which allows the cultivation of metabolically active cells even at zero growth. Here we reached doubling times as long as 38 days and found thatP. pastorisdecreases its maintenance energy demand 3-fold during very slow growth, which enables it to survive with a much lower substrate supply than baker's yeast.

On the basis of preliminary examination of the model of Seyfert galaxies and quasars we have concluded that their gravitational field may have an important influence on the shape of spectral lines when the emission cloud is in gravitational field of massive nuclei (mass about 107–108 M⊙ for Seyfert galaxies, see e.g. Padovani et al. 1990), and the emission cloud is large enough (for example about 1014–1015 m for Broad Line Region (BLR) of NGC 7469 (Bonatto & Pastoriza 1990) or about 1021 m for quasar 3C 257 (Forbes et al. 1990)), so that the emitters are in very different gravitational field along the line of sight.

Summary We present radio-continuum observations made at high angular resolution with the VIA at 20, 6 and 2 cm of the central region of the Sersic-Pastoriza galaxy NGC 1808. These observations reveal a population of compact radio sources, reminiscent of those found in the archetypal starburst galaxies M82 and NGC 253. The bulk of these compact features are not coincident with the optical hot-spots and are likely to be individual or unresolved groups of SNRs. We have also made H i observations of NGC 1808 with the VLA (very large array). Although this was primarily to search for unusual motions which may enable us to understand the nuclear activity, we also obtained information on the large-scale distribution and dynamics of gas in this system. The neutral hydrogen is largely concentrated in a central bar, with weak emission from the spiral arms. An absorption profile against the radio emission from the nuclear region shows two features straddling the systemic velocity. The velocity field of the galaxy is largely consistent with rotation although there is evidence of significant non-circular motions in the bar. We discuss any possible relationship between these non-circular motions and the starburst/nuclear activity in the central region of NGC 1808.

A series of strategies were applied to improve expression level of recombinant endo-β-1,4-xylanase fromAspergillus usamii(A. usamii) inPichia pastoris(P. pastoris). Firstly, the endo-β-1,4-xylanase (xynB) gene fromA. usamiiwas optimized forP. pastorisand expressed inP. pastoris. The maximum xylanase activity of optimized (xynB-opt) gene was 33500 U/mL after methanol induction for 144 h in 50 L bioreactor, which was 59% higher than that by wild-type (xynB) gene. To further increase the expression ofxynB-opt, theVitreoscilla hemoglobin(VHb) gene was transformed to the recombinant strain containingxynB-opt. The results showed that recombinant strain harboring thexynB-optandVHb(named X33/xynB-opt-VHb) displayed higher biomass, cell viability, and xylanase activity. The maximum xylanase activity of X33/xynB-opt-VHbin 50 L bioreactor was 45225 U/mL, which was 35% and 115% higher than that by optimized (xynB-opt) gene and wild-type (xynB) gene. Finally, the induction temperature of X33/xynB-opt-VHbwas optimized in 50 L bioreactor. The maximum xylanase activity of X33/xynB-opt-VHbreached 58792 U/mL when the induction temperature was 22°C. The results presented here will greatly contribute to improving the production of recombinant proteins inP. pastoris.

This study focuses on the characterization of the common pottery productions present in Roman contexts from different deposits in the surroundings of the city of Lucus Augusti, with the main objective of interpretate the transformations that occurred in the zone after the integration of this territory in the Roman world. Through an approach that bets on the analysis of cultural exchange that gives rise to a hybrid culture that has its echo in the pottery, we studied the productions of the hill fort of Saa (A Pastoriza), Agra dos Castros (Lugo) and Viladonga (Castro de Rei). These are different places from each other, which are at different distances from the city and with occupations in periods between the s. I BC and the s. V AD, which has allowed to observe the different realities present in the territory

Esta investigación estudia el caso de Nahir Galarza, una joven argentina condenada a cadena perpetua en 2018 por el asesinato de su ex pareja Fernando Pastorizzo. La propuesta metodológica supone un análisis sobre la circulación mediática del caso, desde que el mismo toma estado público hasta la condena; en intersección con una reflexión teórica sobre las violencias contra las mujeres, el género, los medios masivos y los medios digitales. Los resultados muestran que este acontecimiento estuvo atravesado por múltiples factores, evidenciados en discursos que construyeron sentidos predominantes, en el marco de un sistema hetero – patriarcal sostenido en medios, instituciones y sus violencias cotidianas.

El periodismo cultural difunde, fomenta, recrea y documenta la producción simbólica. Pastoriza (2006) lo define como una forma de cultura que ayuda a definir, dar voz o excluir relatos y sujetos que forman la cultura de una época. Este estudio aborda la construcción de la cultura en la labor periodística cotidiana. Para ello, parte de una investigación comparada sobre el peso de la representación de la cultura en medios locales en Sevilla (España) y Porto Alegre (Brasil). La metodología utilizada es el análisis de contenido, aplicado a 3.709 piezas publicadas entre 2013 y 2018 en la sección de cultura de los periódicos españoles Diario de Sevilla y ABC de Sevilla y brasileños Zero Hora y Correio do Povo. Se concluye que este periodismo tiene poca autonomía frente a la lógica económica de las industrias culturales, pues comparten la representación de una cultura comercial, en detrimento de la identidad local.

Para tanto, se propõe algumas reflexões em torno dos principais aspectos conceituais de “questão social”, reforma do Estado e redes de proteção; fundamentando-se em Netto (2001), Pastorini (2010), Telles (1999), Amaro (2014), entre outros. A construção deste texto possibilitou compreender a relação da “questão social” com o capitalismo e as contradições envolvidas nesse processo e como o avanço do neoliberalismo vem empreendendo retrocessos nos direitos conquistados que afetam diretamente a proteção de crianças e adolescentes, bem como os direitos da classe trabalhadora de modo geral.

Este artículo se propone analizar el tratamiento de las noticias policiales en los programas de la televisión de aire argentina en los que predomina el panelismo. Para ello, se seleccionó una noticia puntual que puede catalogarse como un caso conmocionante: el asesinato de Fernando Pastorizzo por parte de Nahir Galarza, una joven con la que mantenía una relación. Para analizar la cobertura televisiva de este caso se articularon conceptos provenientes de perspectivas no coincidentes: el análisis crítico del discurso, la teoría de la noticia y la teoría del encuadre o framing. Entre los principales hallazgos se destaca la uniformidad de perspectivas que mostraron los panelistas en los programas analizados y las tensiones visibles al pensar el caso desde una perspectiva de género.

El presente trabajo se inscribe en una investigación más amplia que tiene por objetivo, por un lado, analizar las condiciones de producción de los programas de la televisión abierta, con especial atención en los casos policiales, el discurso político y la vida de las celebrities, entre otros de resonancia mediática; por el otro, realizar un estudio en recepción con el propósito de indagar en las percepciones de los televidentes y las televidentes en torno a los programas y el tratamiento sobre casos específicos. Aquí se presenta un análisis en recepción sobre el tratamiento mediático del asesinato de Fernando Pastorizzo, cometido por Nahir Galarza; para ello se hicieron entrevistas y se utilizó una metodología cualitativa. Los resultados obtenidos indican que entre la instancia de producción y recepción hubo concordancia en varios aspectos; entre ellos se evidenció desconocimiento o confusión sobre qué significa la violencia de género.

La presente investigación tiene como objetivo fundamental, la elaboración de un diseño instruccional digital que contribuya al fortalecimiento de la identidad regional de los estudiantes. El estudio se apoya en los postulados teóricos de: UNESCO (2004), CBN (1997), Ausubel (1983), Vygotsky, Rodríguez (2001), Careaga (2001), Galvis (1992), Pastorini, (2000), Salazar (2008), Rosario (2005), Bernice McCarthy (1987), entre otros. La misma adoptó el tipo de investigación descriptiva, con un diseño de campo, enmarcado en la modalidad de proyecto factible. La población está constituida por 12 docentes, y 40 estudiantes de sexto grado. Para la fase diagnóstica se aplicará un cuestionario con 14 ítems para el personal docente y estudiantes, la validación será de contenido a través del juicio de expertos. Se llegó a la conclusión de la debilidad existente en cuanto a los elementos constitutivos de la memoria histórica como parte de la identidad regional.

Mucopolysaccharidosis type II is a human recessive disease linked to the X chromosome caused by deficiency of lysosomal enzyme Iduronate 2-Sulfate Sulfatase (IDS), which leads to accumulation of glycosaminoglycans in tissues and organs. The human enzyme has been expressed inEscherichia coliandPichia pastorisin attempt to develop more successful expression systems that allow the production of recombinant IDS for Enzyme Replacement Therapy (ERT). However, the preservation of native signal peptide in the sequence has caused conflicts in processing and recognition in the past, which led to problems in expression and enzyme activity. With the main object being the improvement of the expression system, we eliminate the native signal peptide of human recombinant IDS. The resulting sequence showed two modified codons, thus, our study aimed to analyze computationally the nucleotide sequence of theIDSnhwithout signal peptide in order to determine the 3D structure and other biochemical properties to compare them with the native human IDS (IDSnh). Results showed that there are no significant differences between both molecules in spite of the two-codon modifications detected in the recombinant DNA sequence.

Cu toate că mișcarea QAnon este privită ca fiind una de extremă dreapta, ai cărei membri și-au însușit teorii ale conspirației care aparent nu au nicio legătură cu religia creștină, cercetările științifice și investigațiile jurnalistice ale fenomenului au evidențiat faptul că o parte dintre adepții mișcării sunt persoane religioase, membri ai unor parohii sau congregații creștine, din Statele Unite ale Americii ori din alte state, în care acest fenomen s-a extins. În prezentul articol va fi evidențiată însușirea teoriilor conspirației QAnon de către membrii unor biserici sau congregații creștine, convingeri privite cu îngrijorare de pastorii respectivelor comunități religioase, aceștia trăgând un semnal de alarmă referitor la dezvoltarea unor sentimente religioase bazate pe precepte biblice îmbinate cu teoriile conspirației QAnon. Utilizarea unor instrumente științifice, precum realizarea unor sondaje de opinie în evidențierea trendului fenomenului, a reliefat faptul că numărul celor care își însușesc teoriile conspiraționiste QAnon este în creștere, doar în patru luni, din februarie până în iunie 2022, numărul americanilor care cred în aceste teorii crescând cu peste trei milioane.

Resumo Reunindo uma diversidade de fontes arquivísticas e imprensa aprofunda-se o papel exercido pela Santa Sé com relação à assistência e cúrias pastoriais da emigração italiana no exterior, no período que vai da segunda metade do século 19 até o fim do Concílio Ecumênico Vaticano II e a época pós-conciliar. O texto documenta como, até os anos 1880, as intervenções promovidas pela Igreja italiana para a tutela dos emigrantes foram limitadas e revestidas, no conjunto, por um caráter episódico e marginal. A situação mudou sensivelmente no curso dos pontificados de Leão XIII e Pio X. Este último, em particular, se empenhou tanto na intensificação de iniciativas de obras de assistência e na centralização das políticas para apoio às curias pastorais dos migrantes, quanto no recrutamento e formação cultural e espiritual do clero destinado a animar a vida religiosa das comunidades de emigrantes italianos no exterior. O empenho a favor dos refugiados e dos prisioneiros de guerra exercido pela Santa Sé, nos anos da Segunda Guerra Mundial, contribuiu para amadurecer, no interior da Igreja, uma sensibilidade maior e mudar, progressivamente, a atenção do problema da emigração italiana para aquele, mais complexo e de alcance universal, de todos que, não apenas por motivos econômicos, mas também por causas ligadas aos conflitos armados, às catástrofes naturais e perseguições, foram, e são ainda hoje, forçadas a abandonar os lugares de origem e a viverem distantes do próprio país: perseguidos, prisioneiros de guerra, refugiados, entre outros.

ABSTRACTA library of engineered promoters of various strengths is a useful genetic tool that enables the fine-tuning and precise control of gene expression across a continuum of broad expression levels. The methylotrophic yeastPichia pastorisis a well-established expression host with a large academic and industrial user base. To facilitate manipulation of gene expression spanning a wide dynamic range inP. pastoris, we created a functional promoter library through mutagenesis of the constitutiveGAPpromoter. Using yeast-enhanced green fluorescent protein (yEGFP) as the reporter, 33 mutants were chosen to form the functional promoter library. The 33 mutants spanned an activity range between ∼0.6% and 19.6-fold of the wild-type promoter activity with an almost linear fluorescence intensity distribution. After an extensive characterization of the library, the broader applicability of the results obtained with the yEGFP reporter was confirmed using two additional reporters (β-galactosidase and methionine adenosyltransferase [MAT]) at the transcription and enzyme activity levels. Furthermore, the utility of the promoter library was tested by investigating the influence of heterologous MAT gene expression levels on cell growth andS-adenosylmethionine (SAM) production. The extensive characterization of the promoter strength enabled identification of the optimal MAT activity (around 1.05 U/mg of protein) to obtain maximal volumetric SAM production. The promoter library permits precise control of gene expression and quantitative assessment that correlates gene expression level with physiologic parameters. Thus, it is a useful toolbox for both basic and applied research inP. pastoris.

ABSTRACTIn a recent vaccine trial performed with African children, immunization with a recombinant protein based onPlasmodium falciparumapical membrane antigen 1 (AMA-1) conferred a significant degree of strain-specific resistance against malaria. To contribute to the efforts of generating a vaccine againstPlasmodium vivaxmalaria, we expressed the ectodomain ofP. vivaxAMA-1 (PvAMA-1) as a secreted soluble protein in the methylotrophic yeastPichia pastoris. Recognized by a high percentage of sera from individuals infected byP. vivax, this recombinant protein was found to have maintained its antigenicity. The immunogenicity of this protein was evaluated in mice using immunization protocols that included homologous and heterologous prime-boost strategies with plasmid DNA and recombinant protein. We used the following formulations containing different adjuvants: aluminum salts (Alum),Bordetella pertussismonophosphoryl lipid A (MPLA), flagellin FliC fromSalmonella entericaserovar Typhimurium, saponin Quil A, or incomplete Freund's adjuvant (IFA). The formulations containing the adjuvants Quil A or IFA elicited the highest IgG antibody titers. Significant antibody titers were also obtained using a formulation developed for human use containing MPLA or Alum plus MPLA. Recombinant PvAMA-1 produced under “conditions of good laboratory practice” provided a good yield, high purity, low endotoxin levels, and no microbial contaminants and reproduced the experimental immunizations. Most relevant for vaccine development was the fact that immunization with PvAMA-1 elicited invasion-inhibitory antibodies against different Asian isolates ofP. vivax. Our results show that AMA-1 expressed inP. pastorisis a promising antigen for use in future preclinical and clinical studies.

Observational evidences of dust in the nuclear region of AGNs are substantial (Rudy 1984, ApJ, 284, 33; Jones et al. 1984, PASP, 96, 692). The ionization cones observed in several Seyfert galaxies has been interpreted as shadowing effects by a dust obscuring torus which hides the broad emission line region (BLR) and the central source (Wilson 1992; Storchi-Bergmann, Mulchaey and Wilson 1992, ApJ 395, L73). A large sample of optical and far-IR data for IRAS Seyfert galaxies has been analysed together with dust emission models (Bonatto and Pastoriza 1993), where it has been concluded that the same dust emission model can be applied to both Seyfert types. In order to further study the effects of dust in the spectra of active galactic nuclei, we have obtained spectrophotometry of 21 IRAS Seyfert galaxies in the range 3500–7200 Å and analyse them in conjuction with their IRAS fluxes. The stellar population type is derived from comparisons with normal galaxy templates using dilution effects in the K CaII line as discriminator. For 55% of the sample the population is of late type. For the rest, blue continua due to recent star formation and/or power-law may amount up to 30% at 4000Å. We conclude that the bulge stellar populations of IRAS Seyfert galaxies are similar to those of normal spirals, except that they are more reddened by E(B-V)i ∼ 0.20. Population-subtracted emission line ratios indicate on average stronger reddening for the narrow-line region (E(B-V)l ∼ 0.8. From photoionization models a power-law index for the ionizing continuum α=1.5, and a metallicity larger than solar are obtained. The most luminous IRAS galaxy of the sample (IRAS555) is discuss in detail: in order to be compatible with the observed IRAS fluxes and the optical stellar continuum, the ionizing continuum must be reddened by AV > 10 magnitudes. Consequently a dust structure in this galaxy appears to be increasingly affecting stars and gas towards the galaxy center.

We investigate two current problems in active galactic nuclei – the mode of fueling the putative black hole, and the question whether the circumnuclear regions have experienced unusual chemical processing – by studying the kinematics and chemical abundance of the gas in the nuclear region of galaxies with sites of ongoing star formation near the active nucleus. We discuss the results for three galaxies with nuclear rings: NGC1097 – for which we recently discovered broad double peaked Hα and Hβ emission from its LINER nucleus (Storchi-Bergmann, Baldwin & Wilson 1993, ApJ 410, L11); NGC1672, which also presents a LINER nucleus; and NGC5248, a galaxy with a ring but no nuclear activity, used as a comparison. Narrow–band images obtained with the CTIO 1.5m telescope were used to map the emitting gas. Longslit spectroscopy obtained with the 4m telescope at high spectral resolution (at several positions over the nuclear region) was used to obtain the gas velocity field. In the two galaxies with LINER nucleus, the starforming rings are located in the turnover of the rotation curves, which show that the gas is rotating faster at the rings than farther out. We conclude that the rings are associated with inner Lindblad ressonances, which may be particularly effective in forcing gas inwards and fuelling the black hole (Wilson et al. 1986, ApJ 310, 121). From the emission line ratios the gas excitation is maped, and it is found that even in NGC5248, the nucleus presents a different excitation, suggesting a very mild LINER activity. Low-dispersion spectroscopy was also obtained in order to calculate the chemical abundances and compare the values obtained for the nuclear gas with those obtained for the HII regions in the ring and beyond the ring (when present). The goal is to check the results of recent studies (Storchi-Bergmann & Pastoriza 1989, ApJ 347, 195; 1990, PASP 102, 1359), based on spectroscopy of the nucleus, which indicate an enhanced abundance of nitrogen, up to 5 times solar for the gas of the narrow line region of LINER and Seyfert 2 nuclei.

Artikkel käsitleb Salomo Heinrich Vestringi eesti-saksa sõnaraamatut „Lexicon Esthonico Germanicum”, mis on koostatud 18. sajandi alguses ja ilmus trükituna esmakordselt alles 1998. aastal. Vestring sündis Pärnus ja töötas enamiku oma elust seal pastorina. Sellepärast on ootuspärane, et sõnaraamat kajastab suuresti tollast Pärnu ümbruse ja laiemalt Edela-Eesti keelepruuki. Artiklis käsitletaksegi sõnaraamatus esivaid Edela-Eesti kohanimesid, muud sõnavara ning fonoloogilisi ja morfoloogilisi erijooni. Ilmneb, et suurel osal sellest sõnavarast ja grammatilistest erijoontest on vasted ka liivi keeles, eriti Põhja-Lätis kõneldud Salatsi liivi keeles. Vestringi esitatud ainesest tuleb esile selliseidki keelejooni, mis on hiljem tuntud üksnes liivi keelest. See annab alust arvata, et kunagi on Edela-Eesti murded olnud liivi keelele veel lähedasemad kui 20. sajandil, ning toetab seisukohta, et Edela-Eesti murded ja liivi keel on varem moodustanud ühtse edelaläänemeresoome murdekontiinumi.South-western Estonian places and language in Salomo Heinrich Vestring’s dictionary. The article concerns the Estonian- German dictionary „Lexicon Esthonico Germanicum” by Salomo Heinrich Vestring which was compiled in early 18th century and first published in 1998. Vestring was born in Pärnu and spent most of his life working there as a pastor. Therefore it is not surprising that the dictionary largely reflects contemporary language use of Pärnu area and south-western Estonia in general. The article discusses south-western Estonian place names, other vocabulary and characteristic phonological and morphological features occurring in the dictionary. It appears that a large part of the vocabulary and characteristic features of grammar have cognates in Livonian, particularly in Salaca Livonian spoken in northern Latvia. Vestring’s material even includes language features that are later only known to exist in Livonian. This gives a reason to believe that south-western Estonian dialects have earlier been closer to Livonian than in the 20th century and supports the view that south-western Estonian dialects and Livonian used to form a single south-western Finnic dialect continuum.

Abstract Background: Black, compared to White women with localized breast cancer have higher mortality and worse distant recurrence free survival (DRFS). This has been attributed to social determinants of health and higher prevalence of triple negative breast cancer (TNBC) in Black compared to White women. Recent studies indicate that racial disparity in outcome is present in patients with estrogen receptor-positive (ER+), but not ER- disease, in particular in patients with residual disease after neoadjuvant chemotherapy (NAC). It has been shown that in some patients NAC may induce pro-metastatic changes in tumor microenvironment, such as increased density of tumor associated macrophages and portals for cancer cell dissemination to distant sites called TMEM doorways (TMEM score). TMEM score correlates with metastasis in patients with ER+/HER2- breast cancer. We hypothesized that racial disparity in DRFS in patients with residual ER+/HER2- disease is due to enhanced pro-metastatic components (macrophage and TMEM doorway density) in the tumor microenvironment post-chemotherapy in Black compared to White women. Methods: We performed a retrospective, multi-institutional study of TMEM score and macrophage density in the residual disease after NAC from 196 patients diagnosed with unilateral invasive ductal cancer of breast between 2004 and 2014. 99 patients self-identified as Black and 97 as White. TMEM doorways were visualized by triple immunohistochemistry for macrophages (CD68), tumor cells (panMena), and endothelial cells (CD31). The evaluation of TMEM score and macrophage density was done using automated image analysis. Tumor characteristics and patient survival were compared between Black and White patients. The relationship between TMEM score, macrophage density and DRFS was examined by log-rank test and multivariate Cox regression model. The covariates in Cox model included TMEM score, age (continuous), race (Black vs White), surgery type (mastectomy vs lumpectomy), tumor stage (T3 vs T1; T2 vs T1), lymph node status (positive vs negative), and tumor subtype (triple negative [TN] vs ER+/HER2-; other vs ER+/HER2-). Results: Black compared to White women were more likely to develop distant recurrence (49.5% vs 34%, p=0.04), receive mastectomy (69.7% vs 51.5%, p=0.014), and have higher grade (p=0.001). Tumors from Black patients had more macrophages and a higher TMEM score in the entire cohort (p=0.004; p=0.001 respectively) and in the ER+/HER2- subset (p=0.008; p=0.008 respectively), but not in the TNBC subset. High TMEM score was associated with worse DRFS in all patients (p=0.004) and in the ER+/HER2- (p=0.03), but not in TNBC. In multivariate Cox model, TMEM score was an independent prognostic factor in the entire cohort (HR, 1.92; 95%CI, 1.15-3.22; p=0.01) and trended towards significance in ER+/HER2- disease (HR, 2.13; 95%CI, 0.96-4.71; p=0.06). TN, compared to ER+/HER2- cancers had higher TMEM score (p=0.01), and macrophage density (p=0.001). Conclusion: Racial disparity in outcome in patients with localized breast cancer may be due to a more pronounced pro-metastatic response to chemotherapy in Black, compared to White patients with ER+/HER2- disease. Thus, higher prevalence of TNBC in Black patients may not be the controlling factor in racial disparity.
