This study aims to: 1) determine the difference in the effect of Circuit body weight and Tabata Workout on the physical fitness of Pasis Dikreg Seskoal, 2) determine the difference in the effect of exercise intensity (high and low intensity) on the physical fitness of Pasis Dikreg Seskoal, 3) determine the interaction between Circuit Body Weight and Tabata Work Out with exercise intensity (high and low intensity) on the physical fitness of Pasis Dikreg. This study used factorial design research, comparing experimental classes using the circuit body weight method and the tabata workout method by considering the level of intensity as a moderator variable. The population in this study were all Seskoal Dikreg Pasis T.A. 2023 61st batch with a total of 90 people. The sample technique used in this study was total sampling. So the number of samples in this study were 90 Pasis Dikreg Seskoal 61st batch of 2023 academic year. The research techniques used are tests and measurements, in this study using the battery A and battery B test systems that have been commonly used by the TNI. The data analysis technique in this study used the MANOVA (Multivariate Analysis of Variance) analysis method with the help of the SPSS version 23.00 data processing program. Based on the analysis of research results and discussion: 1) There is a significant effect of Circuit body weight training and Tabata Workout on the physical fitness of Pasis Dikreg Seskoal, with a significance value of 0.001 < 0.05, 2) There is a significant effect of training intensity on the physical fitness of Pasis Dikreg Seskoal, with multivariate analysis of F 677 values, and a significance value of 0.001 < 0.05, and a decrease in body fat percentage of 6.80%, 3) High intensity circuit body weight training is more effective than Tabata Work Out training, this is evident from the significance value of 0.001 < 0.05, namely the results of Circuit Body Weight Training training increased by 5.14% while high intensity Tabata Work Out training increased by 0.63%.

This research examines psychometric properties of the 10-item Perceived Academic Social Influence Scale (PASIS) across gender groups with samples from Finland and Taiwan (Total N = 4727, 50.2% female adolescents). Results of composite scores, factor loadings, item-total correlations, and values of square root of Average Variance Extracted (AVE) confirmed strong reliability and validity of the scale. A Multi-Group Confirmatory Factor Analysis (MGCFA) revealed that measurement was equivalent at the level of scalar invariance across gender groups. Among three subcomponents of PASIS, latent mean score of teacher factor turned out to be higher than peer and parent factors regarding math achievement for female and male adolescents from both countries. Findings indicate that the PASIS is a reliable and valid measure to assess the perceptions of academic social influence for math learning, which can be utilized in different cultural contexts.

Penelitian ini bertujuan untuk menghasilkan produk pengembangan modul Hybrid termodinamika yang terdiri dari e-modul dan video pembelajaran pada Mata Kuliah Temodinamika yang digunakan oleh perwira siswa di Politeknik Pelayaran Surabaya dengan berdasar konsep Self Directed Learning (SDL) sehingga pasis dapat belajar secara mandiri. Pengembangan produk ini menggunakan metode pengembangan ADDIE. Hasil produk divalidasi oleh ahli desain pengembangan, ahli isi dan ahli media pembelajaran. Analisis yang dilakukan menggunakan analisis deskriptif evaluatif dari penilaian para ahli, teman sejawat dan uji coba lapangan. Hasil perhitungan dari ahli desain pengembangan sebesar 89,5%, ahli isi 88,5%, ahli media pembelajaran 88,8%, penilaian teman sejawat 89,2%, uji kelompok kecil 85,9%, dan uji coba lapangan 81,3%. Secara umum hasil perhitungan menunjukkan produk pengembangan sangat baik dan layak digunakan. AbstractThis study aims to produce development products in the form of E-Modules and learning videos on the Themodynamics Course used by student officers (pasis) at the Surabaya Merchant Polytechnic based on the concept of Self Directed Learning (SDL) so that the pasis can learn independently. This product development uses the ADDIE development method. Product results are validated by development design experts, content experts and learning media experts. The analysis was conducted using evaluative descriptive analysis from the assessment of experts, colleagues and field trials. The results of calculations from development design experts are 89.5%, content experts are 88.5%, learning media experts are 88.8%, peer assessments are 89.2%, small group tests are 85.9%, and field trials are 81.3 %. In general, the calculation results show that product development is very good and suitable for use.

Background: Interleukin (IL)-23 antagonists have shown great efficacy with minimal side effect profile in clinical trial data for the treatment of moderate to severe plaque psoriasis. As of yet, there have been no published data regarding real-world patients who have received tildrakizumab therapy. Objectives: To analyze real-world efficacy and safety of tildrakizumab in patients with moderate to severe plaque psoriasis. Methods: A retrospective chart review was performed at a large urban academic medical center for all patients treated with tildrakizumab for moderate to severe plaque psoriasis. Demographic information, Psoriasis Area and Severity Indexs(PASIs) from initial presentation and 12-month follow-up, comorbidities, and any possible adverse events were collected and analyzed statistically. Results: 30 patients on tildrakizumab therapy were included in the analysis. Overall, the mean ± standard deviation of the PASIs was 15.8 ± 11.8 at initial visit and 1.5 ± 2.9 at 12-month follow-up (P < .001). No serious adverse events were reported. Conclusions: Tildrakizumab has shown efficacy in clinical trials and this real-world cohort for the treatment of moderate to severe plaque psoriasis with a good safety profile. Future studies should be done to assess the efficacy of tildrakizumab compared with other IL-23 inhibitors.

AbstractThe lower Var valley is the only large outcropping zone of Plio-Quaternary terrains throughout the southwestern Alps. In order to assess the seismic hazard for the Alps – Ligurian basin junction, we investigated this area to provide a record of earthquakes that have recently occurred near the city of Nice. Although no historical seismicity has been indicated for the lower Var valley, our main objective was to identify traces of recent faulting and to discuss the seismogenic potential of any active faults. We organized multidisciplinary observations as a microseismic investigation (the PASIS survey), with morphotectonic mapping and imagery, and subsurface geophysical investigations. The results of the PASIS dense recording survey were disappointing, as no present-day intense microseismic activity was recorded. From the morphotectonic investigation of the lower Var valley, we revealed several morphological anomalies, such as drainage perturbations and extended linear anomalies that are unrelated to the lithology. These anomalies strike mainly NE-SW, with the major Saint-Sauveur – Donareo lineament, clearly related to faulting of the Plio-Pleistocene sedimentary series. Sub-surface geophysical investigation (electrical resistivity tomography profiling) imaged these faults in the shallow crust, and together with the microtectonic data, allow us to propose the timing of recent faulting in this area. Normal and left-lateral strike-slip faulting occurred several times during the Pliocene. From fault-slip data, the last episode of faulting was left-lateral strike-slip and was related to a NNW-SSE direction of compression. This direction of compression is consistent with the present-day state of stress and the Saint-Sauveur–Donareo fault might have been reactivated several times as a left-lateral fault during the Quaternary. At a regional scale, in the Nice fold-and-thrust belt, these data lead to a reappraisal of the NE-SW structural trends as the major potentially active fault system. We propose that the Saint-Sauveur–Donareo fault belongs to a larger system of faults that runs from near Villeneuve-Loubet to the southwest to the Vésubie valley to the north-east. The question of a structural connection between the Vésubie – Mt Férion fault, the Saint-Sauveur–Donareo fault and its possible extension offshore through the northern Ligurian margin is discussed.The Saint-Sauveur–Donareo fault shows two en-échelon segments that extend for about 8 km. Taking into account the regional seismogenic depth (about 10 km), this fault could produce M ~6 earthquakes if activated entirely during one event. Although a moderate magnitude generally yields a moderate seismic hazard, we suggest that this contribution to the local seismic risk is high, taking into account the possible shallow focal depth and the high vulnerability of Nice and the surrounding urban areas.

Procyanidins (PAs) is the important flavonoids components in strawberry young fruits, whichis synthesizedthrough direct polymerizationof flavanone-3-ol units. Numerous studies have beendone on the synthesis of PAs, while the oxidation/polymerization processes have not been fully explored. AtTT10, a laccasel-like polyphenol oxidase,was recently reported to be participated in the oxidative polymerization of proanthocyanidins. Whether FaTT10hasapositiveeffect onoxidative polymerization of strawberry fruit PAsis unclear. In this study, FaTT10 in strawberries were isolated and analyzed. Transient overexpression of FaTT10 decreased the accumulation of PAs monomer in strawberry fruit,whereas RNA interference increased the level of anthocyanin and PAs monomer. 4-Dimethylaminocinamaldehyde (DMACA)staining results also showed that lighter blue color in the FaTT10-RNAi fruit and concentrated blue color in the FaTT10-overexpression fruit. Meanwhile, PAs monomer showed a drop-and-rise tendency during strawberry fruit development and ripening.Furthermore, the expression of several flavonoids metabolism-related geneswere affected by the altered FaTT10 gene expression levels, such as L-phenylalanine ammonia-lyase(PAL); dihydroflavonol reductase(DFR);chalcone isomerase(CHI); flavanone 3-hydroxylase(F3H); 4-coumarate:CoA ligase(4CL); Cinnamate 4-hydroxylase(C4H); chalcone synthase(CHS); anthocyanin reductase(ANR); lecoanthocyanidins reductase(LAR) andanthocyanin synthase(ANS). In conclusion, these results revealed that FaTT10 is specifically involved in oxidative polymerization of PAsby regulating the expression of flavonoids metabolism-related genes in strawberry fruits. © 2021 Friends Science Publishers

Implementation according to the rules of occupational safety and health is needed in all jobs that are useful to avoid things that are not desirable, such as work accidents. The workforce consists of educated and trained people. One of the educated people referred to is the D-IV and DP-III graduates of the Makassar Shipping Polytechnic, because one of the fillers in the world of work is the Polytechnic graduate. The learning process while in the laboratory must be a serious concern in order to get qualified prospective workers, especially understanding about the application of occupational safety and health (K3) especially the use of personal protective equipment so that there are no direct or indirect obstacles in the work, such as work accidents occur, damage to machinery, cessation of production processes, environmental damage and expenses related to work accidents. The purpose of this research is to find out the availability and analyze the application of personal protective equipment to occupational safety and health in the workshop. This research includes the type of survey research with quantitative methods. This design seeks to determine the PPE facilities available at the Workshop, symbols / cues about occupational safety and health, the conditions of the practice room and what percentage of PPE use by cadets during practice. The data collection method is done by using a questionnaire and documentation used to determine PPE facilities available in the Workshop, symbols / cues about occupational safety and health, the conditions of the practice room and what percentage of the use of PPE cadets during practice. The conclusion is that the personal protective equipment (PPE) facilities in the workshop have been provided by the workshop and can be used by cadets or pasis and safely in accordance with government regulations, and the knowledge, understanding and application of cadets towards PPE have reached an average of 95%

Ce travail est une continuation des travaux antérieurs sur l'architecture iSOA et le modèle de service intentionnel MiS. Il s'agit de rendre accessible les services proposés par MiS en complétant la plateforme iSOA avec des mécanismes pour la publication et la recherche de services intentionnels dans un annuaire étendu. Ces mécanismes feront appel aux techniques des services web sémantiques et à la modélisation de la connaissance sous forme d'ontologies. Le propos de ce travail de recherche porte sur les lacunes identifiées sur le processus de publication d'un service, l'approche de recherche adéquate pour sélectionner les services intentionnels les plus adaptés aux besoins de l'utilisateur et la structure du descripteur d'un service intentionnel à publier dans l'annuaire. Nous proposons l'approche PASiS (Publishing And Searching intentional Services). Dans cette approche, l'input d'un fournisseur lors de la publication d'un service s'exprime en termes métier sous forme de buts. Nous proposons un processus de guidage de publication, pour d'une part, aider le concepteur dans cette tâche, et d'autre part garantir la complétude de la description d'un service publié. Ce processus utilise les ontologies pour capturer la sémantique du descripteur de service intentionnel. Ces ontologies sont : iSOnto (ontologie technique de services intentionnels), vOnto (ontologie générique des verbes) et pOnto (ontologie de domaine des produits). Enfin, notre approche prend en compte au niveau du descripteur et du processus de publication, les agrégations de services intentionnels telles que définies dans le modèle de MiS. Dans PASiS, l'utilisateur exprime ses besoins sous forme de but. Grâce à un modèle du processus de recherche et d'interrogation, nous proposons le guidage pour d'une part, aider l'utilisateur à tirer profit de toutes les possibilités qu'offre le modèle de requête, et d'autre part pour l'accompagner dans sa recherche et l'aider à trouver ce qu'il souhaite. L'appariement se fait entre les concepts associés à la requête et ceux associés aux descripteurs de services en utilisant une fonction de calcul de distance sémantique qui exploite les liens sémantiques (généralisation, spécialisation). En exploitant les concepts de services intentionnels agrégés, l'utilisateur peut parcourir l'arborescence de services composites et à variation et explorer ainsi l'espace des services intentionnels disponibles. Enfin, l'usage d'ontologies est la base d'un processus de reformulation intelligente de la requête de l'utilisateur. L'utilisation de l'ontologie générique des verbes et des liens sémantiques entre les verbes et les noms propre, nous pouvons raisonner sur la signification d'un but et d'introduire ainsi intelligence et guidage dans les processus de publication et de recherche de service. Ce travail est consacré également, à la spécification et à l'implémentation du descripteur intentionnel de service où l'annotation est utilisée comme une technique d'extension sémantique vers les trois ontologies : iSOnto, vOnto et pOnto. Dans ce travail, nous proposons d'étendre le langage SAWSDL pour décrire les services intentionnels. Cette extension vient combler un manque au niveau de la spécification de la nature de l'annotation sémantique dans SAWSDL. Pour cette raison, nous allons utiliser l'ontologie iSOnto pour expliciter non seulement les concepts d'un domaine mais également la nature de ces concepts. Enfin, l'approche proposée est illustrée au moyen d'une étude de cas d'application. A la lumière de ce cas d'étude, nous avons montré que les hypothèses de recherche sont valides.

Le but de cette thèse est d'étudier la torsion analytique dans deux contextes différents. Dans le premier contexte, on étudie l'asymptotique de la torsion analytique, quand un fibré vectoriel holomorphe hermitien est tordué par une puissance croissant du fibré en droites positif. Dans le deuxième contexte, on généralise la théorie de la torsion analytique pour des surfaces de Riemann avec des pointes hyperboliques. Motivé par des singularités de la métrique complète de courbure scalaire constante -1 sur des surfaces de Riemann stables épointées, on demande que la métrique sur la surface de Riemann soit lisse seulement en dehors d'un nombre fini des points au voisinage auxquelles elle peut avoir des singularités comme la métrique de Poincaré sur un disque épointé. On fixe un fibré vectoriel holomorphe hermitien qui peut avoir au pire des singularités logarithmiques au voisinage des points marqués. Pour ces données, en renormalisant la trace de l'opérateur de la chaleur, on construit la torsion analytique et on étudie ces propriétés<br>In the first context, we study the asymptotics of the analytic torsion, when a Hermitian holomorphic vector bundle is twisted by an increasing power of a positive line bundle. In the second context, we generalize the theory of analytic torsion for surfaces with hyperbolic cusps. Motivated by singularities appearing in complete metrics of constant scalar curvature -1 on stable Riemann surfaces, we suppose that the metric on the surface is smooth outside a finite number points in the neighborhood of which it can to have singularities like Poincaré metric has on a punctured disc. We fix a Hermitian holomorphic vector bundle which has at worst logarithmic singularities in the neighborhood of the marked points. For these data, by renormalizing the trace of the heat operator, we construct the analytic torsion and study its properties. Then we study the properties of the analytic torsion in family setting: we prove the curvature theorem, we study the behavior of the analytic torsion when the cusps are created by degeneration and we give some applications to the moduli spaces of pointed curves

Les apports nutritionnels périnataux en acide gras polyinsaturés n-3 et n-6 (AGPI n-3 et n-6), communément appelés oméga-3 et oméga-6, sont essentiels pour le développement cérébral. Ces lipides ne peuvent être synthétisés par notre organisme et doivent donc être apportés par l’alimentation selon un ratio d’un AGPI n-3 pour quatre AGPI n-6. L’alimentation occidentale a drastiquement évolué au cours des 70 dernières années, vers une consommation excessive d’oméga-6 et une diminution des apports en oméga-3. Ceci est corrélé à un accroissement du nombre d’enfants atteints de pathologies neurodéveloppementales. Cependant, le lien entre nutrition périnatale et neurodéveloppement reste peu compris. L’objectif principal de ma thèse a été d’étudier les mécanismes cellulaires et moléculaires par lesquels la réduction des apports périnataux en oméga-3 altèrent la mise en place des réseaux neuronaux, en me focalisant sur les interactions entre cellules gliales, microglie et oligodendrocytes, et neurones. Nos travaux montrent que la carence périnatale en AGPI n-3 entraîne 1) une altération des fonctions microgliales et oligodendrocytaires au cours du développement cérébral; 2) une augmentation de l'élagage des épines dendritiques par la microglie et des déficits dans le processus de myélinisation; 3) l'établissement de réseaux neuronaux dysfonctionnels dans l'hippocampe et le cortex préfrontal; 4) des déficits d'apprentissage, de sociabilité et l'apparition de comportements anxieux. Par ailleurs, nous avons montré que la carence en AGPI n-3, pendant la période périnatale, aggrave les effets délétères de l'inflammation prénatale. Les faibles apports en AGPI n-3 entrainent 1) une exacerbation de la réaction inflammatoire au challenge prénatal; 2) des altérations plus marquées et plus durables du renouvellement synaptique dans l'hippocampe; 3) des changements de la réactivité inflammatoire des lymphocytes intestinaux persistant à l'âge adulte 4) des déficits de mémoire et de l'hyperactivité chez la descendance à l'âge adulte. Dans l'ensemble, le présent travail a précisé certains des mécanismes par lesquels la carence en AGPI n-3 affecte le cerveau en développement en soulignant son effet néfaste sur la fonction microgliale et oligodendrocytes ainsi qu'en montrant comment elle sensibilise le cerveau en développement à d'autres événements négatifs<br>The perinatal dietary intake in n-3 and n-6 polyunsaturated fatty acids (PUFAs n-3 and n-6), also known as ‘omega-3’ and ‘omega-6’, is essential for brain development. As these lipids cannot be de novo synthesized by the body, they must be provided by the diet according to a ratio of one n-3 PUFA for four n-6 PUFAs. Western diet has dramatically evolved over the past 70 years, towards excessive omega-6 consumption and reduction in omega-3 intake. This correlated with an increasing number of children with neurodevelopmental pathologies. However, the link between perinatal nutrition and neurodevelopment remains poorly understood. The main objective of my thesis was to study the cellular and molecular mechanisms by which a reduction in perinatal n-3 PUFA dietary intake alters neural networks shaping, focusing on the interactions between glial cells (namely microglia and oligodendrocytes) and neurons. Our results show that perinatal n-3 PUFA deficiency leads to 1) an alteration of microglial and oligodendrocytes functions during brain development; 2) an increase in microglia-mediated dendritic spines pruning and deficits in myelination process; 3) the establishment of dysfunctional neural networks in the hippocampus and prefrontal cortex; 4) deficits in learning, sociability and occurrence of anxiety behaviors. Moreover, n-3 PUFA deficiency during the perinatal period exacerbates the deleterious effects of a prenatal maternal immune activation (MIA). Low n-3 PUFA intake 1) increases the maternal and fetal inflammatory response to MIA; 2) increases the duration and extent of MIA effects on neuronal morphology and microglia-neuron interactions; 3) alters the inflammatory reactivity of intestinal lymphocytes, that persists at adulthood 4) induces memory deficits and hyperactivity in offspring in adulthood. Overall, the present work specified some of the mechanisms by which n-3 PUFA deficiency affects the developing brain by highlighting its detrimental effect on microglia and oligodendrocytes function and showing how its sensitizes the brain to other developmental insults

L’allogreffe de cellules souches hématopoïétiques est un traitement curatif pour de nombreuses maladies hématologiques. Cependant, le succès de cette procédure est entravé par la maladie du Greffon-contre-l’Hôte (GVH), une complication potentiellement fatale induite par l’attaque des tissues de l’hôte par les cellules T alloreactives du donneur. La GVH a une prévalence de 40 à 80%, selon les caractéristiques de la transplantation. La GVH est la cause principale de mortalité post-greffe en dehors des rechutes et, en dépit des nombreuses avancées dans le domaine de l’allogreffe, la physiopathologie de cette maladie est encore mal comprise à ce jour, en particulier chez l’homme. Dans cette étude, nous nous sommes concentrés sur la caractérisation phénotypique et moléculaire du système immunitaire des patients greffés et de leurs donneurs. L’objectif de l’étude est de déterminer les facteurs liés à la reconstitution du système immunitaire et au développement de la GVH aigüe. Nous avons collecté le sang de 101 couples donneur /receveur apparentés HLA-identique provenant de trois cohortes indépendantes : avant transplantation pour les donneurs, à l’apparition des premiers symptômes chez les patients atteint de la GVH aigüe et 30 ou 90 jours post-greffe pour les patients non atteints. Nous avons caractérisé le profil cellulaire de ces échantillons par cytométrie spectrale et analysé leur profil transcriptomique.L’immunophenotypage des patients nous a permis de montrer que la reconstitution du compartiment des cellules T est incomplète, avec une inversion du rapport CD4/CD8 après la greffe. De plus, le compartiment cellulaire T est enrichi en cellules possédant un phénotype effecteur/mémoire tandis que le réservoir des cellules T naïves est appauvri. La reconstitution des cellules NK est caractérisée par un enrichissement du sous-type CD56bright et celle des monocytes par l’enrichissement des cellules CD16+. A l’apparition de la GVH, une augmentation de la fréquence des cellules possédant un phénotype semblable aux cellules souches mémoire est observée, par rapport aux patients non-atteints. Ces cellules pourraient représenter un réservoir cellulaire de la maladie, maintenant la production de cellules T alloréactives. Le profilage transcriptomique montre que les cellules T du donneur réagissent à l'environnement de l’hôte en acquérant un phénotype activé, avec une surexpression des gènes associés à l’activation, l’adhésion, la migration et les fonctions effectrices. Au commencement de la maladie, on observe que les cellules T surexpriment des gènes médiateurs de l’inflammation ainsi que des gènes impliqués dans la transduction du signal cytokinique et la migration cellulaire. Nos données démontrent que l’analyse de la distribution des sous-types cellulaires par cytométrie associée au profilage transcriptomique peut contribuer à élucider les mécanismes impliqués dans la reconstitution immunitaire et dans le développement de la GVH humaine<br>Allogeneic hematopoietic stem cell transplantation (HSCT) is a curative treatment for many hematologic malignancies. However, its success is hindered by graft-versus-host disease (GVHD), a potentially fatal complication deriving from alloreactive donor T cells attacking recipient tissues. Acute GVHD (aGVHD) prevalence lies between 40 and 80% depending on transplant characteristics. GVHD is the main cause of non-relapse morbidity and mortality after HSCT and despite the advances in the field, disease processes in humans remain poorly understood.In this study we investigated the phenotypic and molecular characteristics of immune cells in patients after HSCT and in their HLA-identical sibling donors, with the goal of defining immune parameters associated with the recovery of donor-derived immunity and with the development of acute GVHD. We analysed 101 donor-recipient pairs in three independent cohorts for which blood was collected from the donors before transplantation and for the recipients either at aGVHD onset, before any treatment, or at day 30 or 90 post-HSCT for recipients that did not develop GVHD. On the donors’ and recipients’ samples we performed cellular profiling using spectral flow cytometry as well as gene expression analysis.Immunophenotyping reveals an incomplete reconstitution of the T cell compartment in the recipients, with an inversion of the CD4/CD8 ratio, both at one and three months after HSCT. Moreover, the reconstituting T cell compartment is characterized by a shift in the effector/memory phenotype of these cells, with a parallel depletion of the naïve T cell pool. NK cell reconstitution is characterized by an expansion of the CD56bright subset, while monocytes undergo an expansion of CD16+ cells. At aGVHD onset recipients have an increase of cells with a T stem cell memory-like (TSCM-like) phenotype compared to recipients without aGVHD. These cells may represent a cellular reservoir for GVHD, maintaining the production of alloreactive T cells in the presence of host persistent antigens. Molecular profiling shows that donor T cells react to the environment of the host by acquiring an activated phenotype, with upregulation of genes associated with T cell activation, adhesion, migration and effector functions. T cell transcriptome profiling at aGVHD onset shows upregulation of inflammatory mediators as well as genes involved in cytokine signal transduction, cell migration and cell trafficking.Our data demonstrate that comprehensive analysis of the distribution of different immune cell subsets with flow cytometry together with gene expression profiling can contribute to elucidate the processes involved in immune reconstitution and acute GVHD development in humans. In the future, studies with new technologies will hopefully bring insights into the mechanisms underlying GVHD development that will help design new preventive and therapeutic strategies to be applied in the clinics

L’ouverture chez Lacan n’est pas un concept de la psychanalyse. Par ce terme, nous voulons rassembler plusieurs expressions et usages de Lacan qui peuvent s’approximer à un trou, à un écart ou bien à une distance indépassable. Au début des années cinquante, Lacan utilise le mot « béance » dans le cadre de la première hypothèse de l’absence symbolique. C’est ainsi que la « béance » est le premier mot qui participe de l’ouverture chez Lacan. Dans les séminaires suivants, d’autres formes d’ouverture apparaissent. D’abord, l’espace de la tombe, concept abordé dans la lecture que fait Lacan de la mort d’OEdipe, ainsi que dans son analyse d’Antigone et dans sa lecture d’Hamlet. Dans ces deux tragédies de Sophocle et dans l’oeuvre de Shakespeare, Lacan trouve une ouverture qui lui permet d’envisager certaines modifications à la théorie du sujet et de l’objet en psychanalyse. Mais c’est par la suite de l’analyse sur le tombeau que l’espace vide prend une place centrale dans les considérations de Lacan. Notamment dans son analyse d’Antigone, lorsque Lacan aborde la « Chose », qu’il emprunte à Freud, et à propos des premières considérations de l’objet « a », la question de l’ouverture est traitée à partir de l’histoire de l’art. Dans cette recherche nous suivons les rapports entre la psychanalyse et l’histoire de l’art afin d’étudier l’espace qui se trouve entre le corps du sujet et l’objet de son désir. Nous poursuivons ainsi l’analyse entamée par Lacan à la fin des années cinquante, notamment sur la naissance de la peinture en perspective et celle des Vanités. Dans cette dernière partie, il s’agit donc de montrer les approximations de Lacan sur la thématique de l’ouverture en psychanalyse avant qu’il ne la travaille avec la topologie. Cette recherche peut se comprendre comme un regroupement des antécédents sur l’ouverture chez Lacan, avant leur formalisation mathématique<br>For Lacan, the notion of “the opening” is not a psychoanalytic concept. This term encompasses several different expressions and ideas, which, in Lacan’s thought, relate to holes, gaps, and unsurpassable distances. In the early nineteen-fifties, Lacan uses the word béance—a gaping hole—when he articulates his first hypothesis regarding symbolic absence. Thus, béance is the first word that contributes to Lacan’s lexicon concerning openings. In following seminars, other forms of openings appear. First, the space of the tomb, a concept Lacan addresses in his reading of the death of Oedipus, in his analysis of Antigone, and in his treatment of Hamlet. In these two tragedies by Sophocles, and in Shakespeare’s play, Lacan discoversan opening that allows him to envision modifications to the theories of subject and object in psychoanalysis. After this analysis of the grave, empty space occupies a central place in Lacan’s thought. In his discussion of Antigone, where Lacan broaches the subject of Freud’s “Thing”, and in his first treatment of the objet petit a, the question of openings is approached through art history. This project explores the intersections of psychoanalysis, literature, and art history and examines the space between the subject and the object of its desire. In doing so, the study furthers the work Lacan began in the late nineteen-fifties on the birth of linear perspective and the vanitas in painting. These preliminary explorations of the theme of the opening in psychoanalysis led Lacan, in his later work, to approach the idea of the opening through topology. The present study can be understood as a regrouping of the preliminary instances where openings and their mathematical formalizations appear in Lacan’s work

Cette thèse analyse les changements produits dans l'enseignement d'espagnol et de portugais en tant que langues étrangères dans les systèmes éducatifs du Brésil, de l'Argentine et de l'Uruguay pendant la Décennie Dorée latino-américaine, comprise entre 2003 et 2012. Pendant cette période un certain nombre de lois ont été promulguées, comme la Loi 11 161 / 2005 au Brésil, qui obligeait tous les lycées du pays à proposer l'espagnol comme matière d'étude ; la Loi 26 468 / 2009 en Argentine, qui prévoyait la même obligation pour le portugais ; et la Loi 18 437 / 2008 en Uruguay, qui reconnaissait les dialectes de base portugaise du nord du pays. Ces lois supposent une transformation historique des rapports entre ces deux langues ibériques, jusqu'alors caractérisés par une mutuelle marginalité dans le domaine scolaire. Pourtant, les projets dessinés par ces lois constituent vont se heurter à des réalités d'ordre budgétaire, logistique, idéologique et géopolitique entre autres, qui vont compromettre leur réalisation finale. Tout au long de ce travail nous allons essayer d'identifier les tensions, les acteurs, les forces et les obstacles qui se sont alignés autour de ces langues dans ces trois pays. En partant des schémas théoriques habituels intégrant la notion de politique linguistique, nous proposons un modèle en trois plans, corpus, statut et gravitation, qui permet d'appréhender la spécificité de ce cas d'étude : ces deux langues sont les seules au monde à présenter à la fois un tel degré d'intelligibilité et d'importance sociolinguistique, et jamais on n'avait implémenté leur enseignement réciproque de manière généralisée. Ainsi, nous avons aussi essayé de répondre à un certain nombre d'interrogations : (1) Comment le fait d'être des langues proches détermine leur relation glottopolitique et leur insertion dans le système d'enseignement ; (2) quels discours accompagnent celles-ci comme d'autres langues étrangères scolaires et quels projets politiques elles représentent ; enfin, (3) quels sont leurs traits gravitationnels internes, leur projection externe et vers quelles autres langues elles affichent des préférences dans leurs flux de traduction. Nous avons pu ainsi tirer des conclusions sur les vicissitudes gravitationnelles de l'espagnol et du portugais, ainsi que donner des pistes sur les écueils et les ressources rencontrés par les langues super-centrales en général dans leur quête d'espaces internationaux. Cette analyse permet une meilleure caractérisation de la dichotomie entre deux modèles de globalisation linguistique en tension, hyper-centraliste et super-centraliste : l'un unipolaire avec un anglais hégémonique et des langues super-centrales reléguées à des sphères locales, l'autre pluripolaire où ces langues partagent avec l'anglais leur présence internationale et où prévaudrait l'horizontalité glottopolitique<br>This thesis focuses on changes that took place in the teaching of Spanish and Portuguese as foreign languages in the education systems of Brazil, Argentina and Uruguay during the so-called Golden Decade spanning approximately from 2003 to 2012. Measures like the Law 11.161/2005 in Brazil, which compelled high schools to offer Spanish as an optional subject; the Law 26.468/2009 in Argentina, which established the same requirement for Portuguese; and the Law 18.437/2008 in Uruguay, which recognises Portuguese-based dialects spoken in the north of the country, imply a historical transformation of the relationship between these two Iberian languages, which had thus far been characterised by mutual marginality in school systems. However, the projects embodied in these laws clash with several budgetary, logistic, ideological and geopolitical realities which compromise their final achievement. Throughout this work we will try to identify which actors, forces and obstacles align around these languages in these three countries. Based on standard theoretical schemes integrating the notion of Language Planning, we propose a three-plane model where corpus, status and gravitation will allow us to apprehend the specificities of this case study: No other pair of languages in the world presents this level of both intelligibility and sociolinguistic importance, and never before had a generalised reciprocal teaching of them been so seriously attempted. Therefore this inquiry will focus on : (1) How does the fact that they are closely related languages determine determine their glottopolitical relationship and their insertion into education systems; (2) Which discourses accompany these and other foreign languages taught in schools and which political projects do they represent; (3) What are their internal gravitational characteristics, their projection and for which languages do they show preference in their translation flows. We thus hope to draw valid conclusions on gravitational vicissitudes of Spanish and Portuguese, as well as to shed some light on the challenges faced and resources found by supercentral languages in their quest for international spaces.This analysis shoud allow a better characterisation of the dichotomy between two models of linguistic globalisation: hypercentralist and supercentralist. One is unipolar, with English as the hegemonic language and supercentral languages relegated to local levels; the other is pluripolar, with these languages sharing their international presence under a prevailing glottopolitical horizontality

La sénescence cellulaire (SC) qui correspond à un destin cellulaire est caractérisée par un arrêt stable de son cycle dont les cellules présentent un dysfonctionnement. La SC joue un rôle complexe dans la physiologie et la physio-pathologie. Les cellules sénescentes sont extrêmement impliquées dans la suppression de tumeurs, le développement embryonnaire et la cicatrisation des plaies, mais aussi dans de nombreuses pathologies liées à l’âge, y compris, paradoxalement, dans le développement de tumeurs. Mon travail de thèse représente une analyse exhaustive, en temps différé, des différentes couches de l’épigénome, du transcriptome et du métabolome des cellules qui entrent en SC. Il est divisé en trois sous-projets.Tout d’abord, j’ai étudié la dynamique de la liaison des facteurs de transcription(FT) aux activateurs dans la sénescence induite par oncogène (SIO). Les facteurs de transcription s’organisent en un réseau hiérarchisé, les ”pioneers” façonnant le paysage des activateurs en recrutant des ”settlers” et des ”migrants” pour affiner la régulation de l’expression des gènes. Plus précisément, j’ai découvert que les membres de la famille des AP1 précèdent la majorité des autres FT, amorçant la chromatine afin d’initier et coordonner la réponse transcriptionnelle de la SC.Ensuite, j’ai effectué une analyse approfondie des changements métaboliques associés à la SC, en intégrant les résultats de fibroblastes qui entrent en sénescence réplicative, induite par oncogènes et par dommages à l’ADN, en parallèle d’une caractérisation de la SIO dans les myoblastes primaires. J’ai identifié plusieurs métabolites qui s’accumulent ou diminuent dans les cellules sénescentes. Ceux-ci sont associés à des modifications post-traductionnelles, à la synthèse des protéines, àla biosynthèse et à l’oxydation des lipides, ainsi qu’à la production d’énergie. En particulier, l’alpha-cétoglutarate (aKG) et l’uridine diphosphate N-acétylglucosamine(UDP-GlcNAc) agissent comme des substrats pour les modificateurs de la chroma-tine, ce qui suggère un implication dans la régulation des gènes.Troisièmement, j’ai défini un modèle mathématique décrivant l’évolution tran-scriptionnelle des cellules entrant en SIO. J’ai généré ce modèle en me servant de l’algorithme Sparse Identification of Nonlinear Dynamics (SINDy) dans unenvironnement de calcul haute performance. J’ai validé le modèle avec des données de transcriptomiques provenant des expériences d’inhibition de JUN et RELA. Lorsde l’inhibition de JUN, un membre de la famille des AP1, la simulation du modèle s’est davantage rapprochée de la sénescence cellulaire que de RELA. Ce qui suggère que le rang des FT dans la hiérarchie de la liaison à la chromatine pourrait être déterminée par la prévisibilité de sa réponse transcriptionnelle.Dans son ensemble, mon analyse intégrative permet une meilleure compréhension de la SC et possède le potentiel de révéler des vulnérabilités jusqu’alors inconnues des cellules sénescentes et qui pourraient être utilisées en vue de traiter le cancer et les maladies liées à l’âge, favorisant ainsi une plus longue et meilleure qualité de vie<br>Cellular senescence (CS) is a cell fate characterized by a stable cell cyclearrest of dysfunctional cells. CS has an intricate role in physiology and patho-physiology. Senescent cells play a vital role in tumor suppression, embryonicdevelopment, and wound healing, but also in many age-related pathologies,including paradoxically, tumor development. My thesis work represents acomprehensive time-resolved analysis of the epigenome, transcriptome, andmetabolome layers of cells undergoing CS and is divided into three subprojects.First, I investigated the dynamics of transcription factor (TF) binding toenhancers in oncogene-induced senescence (OIS). TFs organize in a hierar-chical network, with pioneers shaping the enhancer landscape by recruitingsettlers and migrants to fine-tune gene expression. Specifically, I discoveredthat the AP1 family members precede the majority of other TFs, primingchromatin to initiate and coordinate the CS transcriptional response.Second, I performed an extensive analysis of the metabolic changes asso-ciated with CS, integrating results from fibroblasts undergoing replicative,oncogene-induced, and DNA damage-induced senescence, in addition to acharacterization of OIS in primary myoblasts. I identified several metabolitesthat accumulate or diminish in senescent cells, and those are associated withpost-translational modifications, protein synthesis, lipid biosynthesis and oxi-dation, and energy production. In particular, alpha-ketoglutarate (aKG) anduridine diphosphate N-acetylglucosamine (UDP-GlcNAc) act as substratesfor chromatin modifiers, suggesting roles in gene regulation.Third, I defined a mathematical model describing the transcriptionalevolution of cells undergoing OIS. I generated this model using the SparseIdentification of Nonlinear Dynamics (SINDy) algorithm in a high-performancecomputing environment. I validated the model with transcriptome dataderived from JUN and RELA depletion experiments. On inhibition of JUN, amember of the AP1 family, the model simulation behaved closer to senescentcells than on RELA, suggesting that TF rank in the chromatin bindinghierarchy may determine the predictability of its transcriptional response.Together, my integrative analysis provides a deeper understanding of CSand has the potential to reveal previously unknown vulnerabilities of senescentcells that may be exploited to treat cancer and age-related diseases, promotinga longer healthspan

Les infections nosocomiales représentent la première cause de mortalité et le premier poste de prescription d’antibiothérapies. Les pneumopathies nosocomiales représentent la première cause de morbi-mortalité en réanimation. Dans un contexte de ressources antibiotiques limitées et une augmentation de l’incidence des germes résistants, nous avons étudié:1°) Les facteurs de risque de pneumopathies acquises sous ventilation mécanique (PAVM) en tenant compte de la durée de la ventilation mécanique2°) La différence entre les pneumopathies associées aux soins en réanimation chez les patients non ventilés et les pneumonies acquises sous ventilation mécanique en terme d’étiologie et de conséquences pronostiques.3°) L’impact de l’adéquation de l’antibiothérapie initiale et les facteurs de risque d’avoir un traitement inadéquat du fait de la résistance bactérienne dans les PAVM à germes gram négatifs.Nos résultats montrent que : Les pneumonies acquises en réanimation en dehors de la ventilation mécanique invasive (ICU-HAP) et les PAVM sont dues aux mêmes germes avec un risque de décès plus important chez les ICU-HAP. Les PAVM précoces et tardives sont associées à des facteurs de risques différents pouvant justifier de politiques de préventions différentes. L’antibiothérapie préalable et la colonisation préalable à des bactéries multi résistantes expliquent la survenue de PAVM à Gram négatifs résistants. La durée de ventilation invasive avant la survenue de la pneumopathie n’est pas un facteur de risque indépendant de survenue de PAVM à gram négatifs résistants. Un pourcentage d’infection ou de colonisation à bacille gram négatif (BGN) résistant supérieur à 10% dans le centre est associé à la survenue d’une PAVM à gram négatif résistant, indépendamment des facteurs individuels. Les PAVM à S. maltophilia surviennent très tardivement, sont très largement associées à une consommation d’antibiotiques élevée, en particulier les pénèmes. Elles sont plus souvent associées à une antibiothérapie initiale inadéquate.Notre travail confirme la nécessité d’un effort particulier afin d’optimiser le diagnostic et l’adéquation précoce des antibiothérapies, en particulier pour les pneumonies acquises en dehors de la ventilation mécanique et les PAVM à BGN potentiellement résistants aux antibiotiques<br>Healthcare-associated infections are the leading cause of death and the first prescription of antibiotics. Healthcare-associated pneumonia represents the first cause of morbidity mortality in ICU. In the context of limited antibiotic resources and an increase in the incidence of resistant germs, we studied:1) Risk factors for ventilator acquired pneumonia (VAP) taking into account the duration of mechanical ventilation (MV).2) If there is a difference between nonventilated and mechanically ventilated pneumonia in terms of etiology and prognostic?3) What is the impact of the adequacy of the initial antibiotic treatment, and what are the risk factors for inadequate treatment in gram-negative resistant VAP?Our results show that: Pneumonia acquired by no ventilated patient in the ICU (ICU-HAP) and VAP are due to the same germs with a higher risk of death in ICU-HAP. Early and late VAP are associated with different risk factors that may need different prevention policies. Previous-antibiotic therapy and prior-colonization with multiresistant bacteria explain the occurrence of resistant Gram Negative VAP. Duration of MV before the onset of pneumonia is no longer an independent risk factor for the occurrence of resistant gram-negative VAP. A percentage of infection or colonization with resistant BGN upper to 10% in the centre is associated with the occurrence of a gram-negative, resistant PAVM, regardless of individual risk factors. S. maltophilia VAP occurs belatedly and are very widely associated with high antibiotic consumption, especially penems. They are more often associated with inadequate initial antibiotic therapy.Our work confirms the need for further research to optimize the diagnosis and early adequacy of antibiotics, in particular in ICU-HAP and VAP caused by potentially antibiotic-resistant BGN

Plusieurs hypothèses sont présentées par les médecins grecs anciens et ceux de l’époque islamique quant aux facteurs qui déterminent le sexe du fœtus. Hippocrate, Aristote et Galien présentent chacun des hypothèses différentes dans leurs œuvres à propos de la formation du fœtus mâle ou femelle. Ces hypothèses développent des facteurs internes comme la force, la dominance de la semence mâle ou femelle, la position du fœtus dans l’utérus et des facteurs externes comme la chaleur, le froid, les vents qui soufflent dans la région. Dans cette recherche, nous analysons les avis des médecins grecs anciens par l’examen de leurs œuvres ou à travers les traductions de celle-ci et par les citations dans les textes médicaux de l’époque islamique tels que Firdaws al-hikma fī Ṭibb et al-Ḥāwī fi Ṭibb. Nous considérons ensuite le point de vue du médecin Rāzī (IXe-Xe) en nous fondant sur un traité indépendant intitulé al-ubnah. Nous comparons ses points de vue avec ceux des médecins grecs. Grâce à ses avis sur la détermination du sexe du fœtus, Rāzī a tenté d’expliciter certains comportements d’un homme majeur attiré par un autre homme majeur. Dans cette recherche les troubles mentionnés et leurs causes sont étudiées du point de vue de Rāzī ainsi que les méthodes de traitement que proposent ce dernier en la matière<br>Among ancient Greek and Islamic period physicians there are several theories regarding the determinants of sex of the fetus. Hippocrates, Aristotle and Galen each proposed different theories for the sex determination of male or female embryos.In these theories, various factors such as strength and dominancy of male or female semen to each other, external factors like heat and cold, the location of the fetus in the uterus, the kind of winds blowing in that area and have been mentioned.In the current research, views of ancient Greek physicians on this subject will be discussed based on the transmission of their writings to the current time or translations or citations of their works in medical texts of the Islamic period such as Firdaws al-hikma fi Ṭibb and al-Ḥāwī fi Ṭibb. Afterwards Razi’s (IXe-Xe) view will be analyzed according to his independent treatise titled al-ubnah, the theories mentioned in al-Ḥāwī fi Ṭibb, and then it will be compared with the opinions of ancient Greek physicians. Rāzī, based on his point of view on the sex determination of the fetus, has explained sexual disorders like attraction between men. In this research, such attraction and his treatments proposed will be discussed
