Résumé :
Komunikasi matematika ialah sesuatu wujud keahlian yang begitu berarti dalam matematika. Tujuan dari penelitian ini ialah untuk mempelajari profil komunikasi matematika siswa dalam menuntaskan soal PISA ditinjau dari kemampuan matematika. Penelitian ini ialah penelitian deskriptif dengan pendekatan kualitatif. Instrumen yang digunakan dalam penelitian ini antara lain uji kemampuan matematika, tugas komunikasi matematika tulis siswa dalam menuntaskan soal PISA, serta pedoman wawancara. Subjek dalam penelitian ini ialah 3 siswa kelas IX dengan tiap-tiap jenis ialah, satu siswa dengan keahlian matematika tinggi, satu siswa dengan keahlian matematika sedang, serta satu siswa dengan keahlian matematika rendah. Hasil dari penelitian ialah siswa dengan keahlian matematika tinggi a) menuliskan perihal yang dikenal serta ditanyakan b) menuliskan statment dalam soal ke dalam model matematika c) menuliskan ide untuk menuntaskan soal d) menuliskan langkah-langkah penyelesaian serta pemecahan dengan akurat, lengkap serta mudah/lancar. Sebaliknya siswa dengan keahlian matematika rendah dalam a) menuliskan/ melaporkan apa saja yang dikenal serta ditanyakan secara akurat, lengkap, serta mudah/lancar. Berikutnya tidak akurat, tidak lengkap serta tidak lancar dalam b) menuliskan statment dari soal ke dalam model matematika c) menuliskan ide untuk menuntaskan soal d) menuliskan langkah- langkah penyelesaian serta pemecahan. Penelitian ini mencerminkan bahwa keahlian/kemampuan matematika pengaruhi siswa untuk memproses data, salah satunya pada saat mengomunikasikan ide atau gagasan dalam menuntaskan soal PISA. Kata Kunci: Komunikasi Matematika, Soal PISA, Kemampuan Matematika.

Abstract Human T-cell leukemia virus type 1 (HTLV-1) is an oncogenic retrovirus that is etiologically associated with adult T-cell leukemia. The HTLV-1 bZIP factor (HBZ), which is encoded by the minus strand of the provirus, is involved in both regulation of viral gene transcription and T-cell proliferation. We showed in this report that HBZ interacted with Smad2/3, and enhanced transforming growth factor-β (TGF-β)/Smad transcriptional responses in a p300-dependent manner. The N-terminal LXXLL motif of HBZ was responsible for HBZ-mediated TGF-β signaling activation. In a serial immunoprecipitation assay, HBZ, Smad3, and p300 formed a ternary complex, and the association between Smad3 and p300 was markedly enhanced in the presence of HBZ. In addition, HBZ could overcome the repression of the TGF-β response by Tax. Finally, HBZ expression resulted in enhanced transcription of Pdgfb, Sox4, Ctgf, Foxp3, Runx1, and Tsc22d1 genes and suppression of the Id2 gene; such effects were similar to those by TGF-β. In particular, HBZ induced Foxp3 expression in naive T cells through Smad3-dependent TGF-β signaling. Our results suggest that HBZ, by enhancing TGF-β signaling and Foxp3 expression, enables HTLV-1 to convert infected T cells into regulatory T cells, which is thought to be a critical strategy for virus persistence.

Education insurance provides services in the field of education. In education insurance, the insured not only gets protection benefits but also education funds. These benefits will be received if they have paid premiums. Insurance companies also need to set the exact amount of policy value. The purpose of this study is to determine the premium and policy value of education insurance by taking into account the child's life chances. In this study, used secondary data from the 2011 Indonesian Mortality Table and illustrated data in the form of education fund data. Premium is obtained using the equivalence principle and policy value is obtained using the prospective method. In the calculation of premiums and policy values for education insurance premiums by taking into account the child's life chances, modifications are made, the amount of education funds multiplied by the child's life chances. The results given in this study are the amount of education insurance premium by taking into account the child's life chances is Rp 6.946.456,00. Policy value increases during the disbursement of education funds and decreases at the end of coverage.

Bali has an increasing tourism potential. This is evidenced by the increasing number of foreign tourist visits to Bali Province each year. Although Bali's tourism trends have continued to increase over the past few years, efforts to improve the quality of Bali tourism need to be made. One way is to do forecasting. To support improvement efforts in Bali's tourism sector, the author created a forecasting system for foreign tourists to Bali province using artificial neural network methods with back propagation algorithms. Artificial Neural Networks with back propagation algorithms are neural network algorithms by finding optimal weight values. The forecast results using the binary sigmoid activation function were obtained by 489,862 foreign tourists in November 2019 with MAPE at 1.62% and 487,342 foreign tourists in December 2019 with MAPE of 11.78%. The forecast results using the bipolar sigmoid activation function were obtained by 493,200 foreign tourists in November 2019 with MAPE of 0.95% and 484,090 foreign tourists in December 2019 with MAPE of 12.37%.

Influence of radiation and non-uniform heat source on unsteady, magneto-hydrodynamic free convection flow of viscous incompressible fluid past an infinite vertical heated plate embedded in porous medium of an optically thin environment with time dependent suction and viscous dissipation is investigated in this paper. Analytical solutions of the coupled non-linear equations are obtained for the velocity field and temperature distribution using oscillating time-dependent perturbation technique. Expressions for skin-friction and heat transfer rate are also derived. The effects of the material parameters on velocity, temperature, skin-friction, and rate of heat transfer are discussed quantitatively.

The population data in Indonesia that is closest to the actual condition is only the data from the population census conducted by the Central Statistics Agency (BPS). The calculation of population projections in BPS throughout Indonesia uses the component method to see the projection of the population at the provincial level while the district level uses the geometric method with the assumption that the population will begin to increase geometrically with a basic reference for calculating compound interest. This study aims to determine population projections in the province of East Java using exponential and logistical models based on growth rates and carrying capacity. The data used in this study is secondary data, namely the population of the Central Statistics Agency (BPS) of East Java Province in 2012-2019. The research method used is to determine the research subject, collect data, analyze data and draw conclusions. The results of this research are the carrying capacity value of East Java Province of 43,997,165.5 and the logistic model population growth rate of 0.05111. The conclusion of this research is that the most accurate model for estimating the population of East Java is the V logistic model because it has the smallest Mean Absolute Percentage Error (MAPE) value. This research is expected to be useful for users of population data in calculating future population projections

The aim of this research is to determine the dynamic model equation of autoregressive distributed lag by using koyck method, to find out the effect of log US dollar exchange rate and log inflation on log stock price in 20142018, and to forecast value of log stock price on January 2019August 2019. The data used in 20142018. The data was transformed into logarithm format. Time series plot of log US dollar exchange rate, log inflation, and log stock price suggest that the fluctuation in the data, for instance, both upward and downward trends, during the period. We obtained that the Koyck transformation could changed the lag distribution model into autoregressive distributed lag (ARDL) dynamic model. Furthermore, the log of US dollar exchange rate and log inflation have negative effect on log stock price in particular period. We measured forecasting accuracy using mean absolute prediction error (MAPE) and concluded that ARDL forecasting using Koyck model shows a significant increase in stock price.

The high number of critical illness cases and the rising medical cost affect the number of insurance claimed. It can be a problem for the insurance company in estimating future claim trend to decide the risk premium cost, so that we need a method to solve the problem which is called Empirical Bayes Credibility Theory (EBCT). Based on that information, the goal of this research is estimating the risk premium cost for critical illness cases using EBCT method. In this research, a large simulation of the expected claim data of critical illness policyholders is used, which are grouped by age and gender. In processing data by applying the EBCT method, the credibility factor is determined first, then the size of the risk premium can be estimated, with the calculation results obtained by a different estimate of the risk premium for each age group based on gender.

Agricultural insurance protects farmers who experience crop failure. This study aims to calculate the value of agricultural insurance premium by applying simulated rainfall index-based using stochastic weather generator on soybean commodities in Negara sub-district. This study are used rainfall data to determine the probability of the transition, then perform rainfall simulations using the Stochastic Weather Generator method to obtain trigger values and continued with the calculation of agricultural insurance premiums. Results of this study provide the value that higher trigger is taken, the greater the insurance premium that must be paid. The value of insurance premiums to be paid is 4,18% - 5,66% of insurance costs Rp2.605.000,00.

Current technological advances cause information to be obtained quickly. Such information can be obtained through printed media or electronic media. Television is included in the category of electronic media. The infotainment program is one of the television programs. Almost all television stations have infotainment programs running every day. This proves that people love the infotainment program, so the infotainment program still running until now. People’s as watchers/audiens of the infotainment program have the right to provide an assessment of the quality of infotainment programs related to satisfaction and preference to assist in the development of infotainment programs. To understand people's preferences for infotainment programs, it can be done using conjoint analysis. In this research, it was found that the attributes that influence the people’s in watching infotainment program are presenter, air time, and program duration. In general, the infotainment program that desired by the people was an infotainment program that has a paired presenter, running at night with a half hour run time

In this paper, it is discussed about giving control variable to dynamic systems to process the spread of viruses on computers. The dynamic model describes the process of spreading viruses on the computer, where susceptible can be instantly removed and infected can instantly become susceptible. And through qualitative analysis on the model is given a control and completed using the Pontryagin Maximum Principle (PMP) method on infected interactions so that the infected computer can be minimized. Dynamic models that have been given control are simulated using MATLAB software and the results show the infected computer is reduced enough significan.

Consumption of cigarettes in large quantities by the public is one of the main concerns in every country because cigarettes contain harmful ingredients that can trigger various diseases. This journal will explain the mathematical model of the number of smokers affected by rising prices of cigarettes with square root dynamics. The population is divided into four, composed of potential smokers, occasionally smokers, heavy smokers, and ex-smokers. The results of the model analysis are that there is a single point of smoker’s endemic equilibrium. If conditions are met, then the endemic equilibrium point of smokers will be asymptotically stable, and over a long period of time there will always be a spread of smokers.

One example of transportation problems is the Shvan Art Shop, which is engaged in exporting 20 cm rattan bags. In distributing rattan bags, Shvan Art Shop does not directly send goods to their destination, but still through intermediaries. To overcome this problem, a transshipment model can be used to get a distribution line so that the outgoing costs are minimal. The purpose of this study is to compare distribution costs before applying the transshipment model with after applying the transshipment model using the ASM Repair and Revised Distribution methods. The results showed that the Revised Distribution method resulted in a distribution cost that was less than the Revised ASM method, which was Rp. 42,884,288.00.

Abstract Abstract 2480 Acute myeloid leukemia (AML) represents one of the most genetically heterogeneous malignancies; however, some processes are commonly dysregulated. One of the most frequently dysregulated processes in AML is Wnt signaling. In solid cancers, aberrant Wnt signaling has been shown to promote cancer by increasing nuclear accumulation of β-catenin and with consequent activation of target genes. In AML, overexpression of β-catenin is also common; in addition however, patient studies and genetic models indicate that other components of the Wnt pathway are also commonly dysregulated and may mediate transcriptional changes independently of β-catenin. The aim of this study was to identify aberrantly regulated Wnt components and target genes in AML by interactome analysis of the AML Affymetrix GeneChip® 3` expression microarray datasets; a network building algorithm used to understand relationships between genes. Analysis and interpretation of microarray data is still both biologically and computationally challenging. To address this, we performed batch adjustment to the large scale AML dataset by merging gene expression profile (GEP) data derived from different database sources (including different array platforms). GEPs data were generated from our AML patients enrolled in two different AML NCRI-MRC UK clinical trials using two different Affymetrix platforms, HG-U133A (n=216) and HG-U133Plus2.0 (n=139). GEPs from normal CD34+ bone marrow samples were downloaded from ArrayExpress (n=26). In order to compare AML vs. normal haematopoietic GEP, all data were merged into a single dataset. Individual. CEL files were imported into Partek® Genomics Suite™ and GC-RMA normalization was applied. Linear contrasts, mixed model analysis of variance with false discovery rate correction (P<0.05) and threshold analysis (>1.5 or <1.5 fold-change) were applied to the adjusted data followed by gene enrichment analysis using MetaCore™ (GeneGo Inc). Batch adjustment was performed using Distance Weight Discrimination (DWD) method to the merged GEPs. Prior to further inferential and gene ontology testing, the DWD merged datasets showed significant reduction in the source of data bias with GEP clustered according to their biological variation rather than technical variation. As a result, we present a final list of 58 significant changes in the expression of Wnt related genes in AML. Enrichment by protein function analysis highlighted 8 Wnt transcription factors to be dysregulated (TCF7L2/TCF4, MYC, NANOG, WT1, RUNX2, p300, TCF7, SMAD2), along with 5 receptors (CD44, FZD3, FZD4, FZD5, LDLR), 3 types of phosphatases (B56G, PR61-β, PPP2R5A) and other categories of Wnt related objects (n=33). Consistent findings were seen with previously established Wnt-associated genes specific to AML (CD44, WT1, MYC) showing that data sources from DWD adjustment was effective. We sought to evaluate the significant biological and functional relationships within the genes in the final dataset by transcription factor target modeling using MetaCore™ Interactome tools. Direct network interaction uncovered TCF7L2/TCF4 as the most significantly upregulated Wnt transcription factor with concurrent high expression of its downstream Wnt responsive genes CD44, AXIN1, ID2 that were also present in the final list. Importantly, β-catenin is unlikely to contribute to this transcriptional activation due to the fact that our data showed increased transcription of β-catenin degradation complexes (or negative regulation of Wnt signalling). Specifically, RUVBL1, that directly increases β-catenin activity was significantly downregulated, whereas the other significantly overexpressed upstream genes (APC, CSNK1E, AXIN1, WT1) are known to have inhibitory effect on β-catenin-mediated transcription. In summary, by using multiple GEP data from a large AML cohort in conjunction with robust statistical adjustments, we have identified TCF7L2/TCF4 mediated transcription as the most significant Wnt-regulated process to be altered in AML compared with normal blasts. We also predict that transcription of TCF7L2/TCF4 regulated genes is likely to be independent of β-catenin, supporting observations in genetic models which indicate that β-catenin (and γ-catenin) are redundant for normal haematopoiesis and are not required for TCF-mediated transcription. Abstract Pancreatic ductal adenocarcinoma (PDAC) is one of the top five deadliest forms of cancer with very few treatment options. The 5-year survival rate for PDAC is 10% following diagnosis. Preclinical murine models have been developed that leverage key driver mutations and have significantly contributed to our understanding of PDAC. One such genetically engineered mouse model (GEMM) that has emerged as an important tool is the KPC mouse (LSL-KrasG12D/+;LSL-Trp53R172H/+; p48-Cre) that spontaneously develops pancreatic tumors at ~14-16 weeks of age. Cadherin-11 (Cdh11), a cell-to-cell adhesion molecule has been suggested to play a role in development of the desmoplastic stroma in PDAC, that leads to difficulties in drug accessibility and has been hypothesized to contribute to chemotherapeutic resistance and correlate with poor prognosis. However, the mechanisms by which Cdh11 deficiency in the stromal microenvironment of PDAC-bearing KPC mice influences tumor infiltrating immune cells, has yet to be fully understood. Single-cell RNA sequencing (scRNAseq) of the immune (CD45+) compartment of tumor bearing Cdh11 proficient (KPC/Cdh11+/+), tumor bearing Cdh11 deficient (KPC/Cdh11+/-), non-tumor bearing Cdh11 deficient (Cdh11+/-) and wildtype (Cdh11+/+) mice was performed. We observed a sharp decrease in the presence of myeloid/monocyte lineage cells (CD14+) in KPC/Cdh11+/- tumors and also an increase in T, B and plasma cells, compared to KPC/Cdh11+/+ tumors. Genes upregulated in infiltrating T- and NK cells specific to a Cdh11 deficient background include Cd8a, Nkg7, Maf. Additionally, genes found to be upregulated in B cell clusters in Cdh11 deficient mice include those related to B cell differentiation/activation such as Lgals1, Id2, Itgb1, Rgs1. The increase in B and T cell infiltration was specific to the Cdh11 deficient background, since both pancreata from KPC/Cdh11+/+and Cdh11+/- mice had elevated levels of infiltration. Immunohistochemical validation of these findings has confirmed these changes in tumor infiltrating immune cells. Additionally of note, an increase in antibody-producing plasma cells was observed specifically in a Cdh11 deficient background. Igkc, an immunoglobulin found to be enriched in plasma cells was highly expressed in this group of immune cells. We also observed that KPC/Cdh11+/-had significantly more Igkc expressing cells than KPC/Cdh11+/+. Future work is needed to clearly define the role of Cdh11 in modulating B, T and plasma cell behavior and subsequent contributions to PDAC outcome. This study received funding by LDRD 19-SI-003. Abstract We identified a subset of BCP-ALL with switch towards the monocytic lineage within the first month of treatment (swALL)[Slámová et al Leukemia 2014]. During the switch cells gradually lose CD19 and CD34 expression and acquire CD33 and CD14 positivity. We proved clonal relatedness of switched monocytic blasts with the diagnostic leukemic cells based on identical Ig-TCR rearrangements. SwALL cases are not associated with MLL or BCR/ABL1 aberrancies and lack any known genetic markers of lineage ambiguity (detected by FISH or MLPA). We analyzed transcriptomes of swALL samples at diagnosis (n=4) and at d8 (n=4) where the immunophenotypic switching was already apparent as well as control BCP-ALL (n=4). RNA was isolated form either FACS sorted cells or whole BM when blasts constituted >80% of cells. For RNA-Seq we used Illumina HiSeq 2000 paired-end or single end sequencing. Raw sequencing data were analyzed using adapted protocol from Anders at al [Anders et al Nature Protocols 2013] and custom scripts. For methylome analysis we used Enhanced Reduced Representation Bisulfite Sequencing (ERRBS)[Akalin et al PLoS Genetics 2012]. ERRBS quantitatively measures DNA methylation at ~3M CpGs genome-wide. Samples from swALL at diagnosis (n=7) and at d8 (n=4) and control BCP-ALL (n=4) were processed. Analysis was performed according to [Akalin et al Genome Biology 2012] and followed with custom analysis in R statistical language. Comparison (generalized exact binomial test) of transcriptomes of B-lineage blasts from diagnosis between swALLs and control BCP-ALLs revealed a number of differentially expressed genes. Among 300 most significantly differentially expressed were KLF4, CEBPD, CLEC12A and CLEC12B (upregulated in swALL) and ANXA5, VPREB1, CD9 and IGHG3 (downregulated in swALL). Hierarchical clustering separated not only swALL and control BCP-ALL, but also swALL cells before and during the monocytic switch. Changes in gene expression during lineage switch included downregulation of ITGA6, Id2, EBF1, CD19, CD34, FLT3, MYB, CD79a, BCR, PAX5, GATA3 and TCF3 genes and upregulation of S100A10, AIF1, CD14, CD33, LGALS1, RNF130 and MNDA. When comparing all three cell types (swALL B cell and monocytic blasts and control BCP-ALL blasts) we concentrated on 1) immunophenotype switch markers and 2) lineage related transcription factors (TF): 1) Both markers typical for B cell blasts (CD19, CD34) decreased during the switch. However while CD19 was expressed in swALL at diagnosis at same levels as in control BCP-ALL, CD34 was overexpressed in swALL compared to BCP-ALL at diagnosis. Both monocytic markers (CD33, CD14) increased their expression during the switch. CD14 showed no difference between swALL and control BCP-ALL at diagnosis. However CD33 was interestingly upregulated in swALL already at diagnosis and continued to rise during the switch. SwALL had therefore deregulated expression of lineage commitment markers already at diagnosis favoring stemness marker CD34 and myeloid marker CD33. 2) B lineage commitment related TFs (EBF1, TCF3, PAX5) were expressed in B lineage blasts in both swALL and control BCP-ALL. However they were all downregulated during the switch. On the other hand myeloid lineage related transcription factor CEBPA is overexpressed in diagnostic B lineage blasts in swALL compared to control BCP-ALL cases. Similarly CEBPD is overexpressed in swALL and its expression further rises during the switch. Other hematopoietic TFs upregulated in swALL cases include KLF4, NANOG and GATA3. To confirm some of the epigenetic markers of swALL cases (demethylation of CEBPA promoter) and to widen epigenetic screening we used ERRBS. While some of the upregulated genes had expectedly hypomethylated promoters in swALL (CEBPA, GATA3) other genes (TCF3, PAX5) had demethylated promoters in all cases. While the whole DNA methylation picture is still a challenge to draw both omics method could clearly separate swALL cases from control BCP-ALL using principal component analysis. In summary we show that immunophenotypic shift is associated with gene expression changes of surface markers, lineage specific transcription factors and other genes. Some of the genes have altered expression already at diagnosis. Expression of some key lineage genes is differentially regulated by DNA methylation. Introduction: Iron Deficiency Anaemia (IDA) is a globally prevalent nutritional disorder and an important risk factor for the development of Mild Cognitive Impairment (MCI). A manifestation of IDA is altered electrogenesis in the central nervous system. As women of reproductive age are more susceptible to this form of anaemia it is important to assess their cognitive function. Auditory cognitive evoked potentials/P300 are sensitive in detecting MCI which is indicated by prolonged latency and reduced amplitude. Aim: To investigate the effect of IDA on cognitive function using cognitive evoked potentials/P300 in neurologically intact women. Materials and Methods: A cross-sectional study was conducted at the Central Neuro Physiology lab, Acharya Vinobha Bhave Rural hospital (AVBRH) attached to Jawaharlal Nehru Medical College (JNMC), Wardha, Maharashtra, India, from January 2018 to February 2022. A total of 260 women were recruited for the study. Based on their blood haemoglobin and serum ferritin levels, 130 women were grouped as anaemic and 130 as non anaemic. The P300 was used as an objective tool to assess cognitive function. Haematological parameters like blood haemoglobin and serum ferritin levels were compared (‘t’ test) and correlated (Spearman correlation) with the latency and amplitude of the P300 wave in the two groups. Results: The mean age (years) and Body Mass Index (BMI) (kg/m2 ) of anaemic women were 23.88±3.67 and 20.98±1.45, respectively; and that of non anaemic women were 24.09±3.41, and 21.25±1.27 (p>0.05) respectively. The blood haemoglobin (mg/dL) and serum ferritin (ng/mL) were significantly (p<0.001) lower in anaemic group (10.37±0.95, 8.55±3.78) compared to non anaemic group (13.02±0.70, 27.61±10.52). The latency of P300 wave (ms) was significantly prolonged (p<0.001) in anaemic women (317.75±7.34) in comparison to non anaemic women (311.71±9.02), while the P300 amplitude did not differ between the two groups (p>0.05). A highly significant low negative correlation of P300 latency with haemoglobin (r= -0.48, p<0.001) and highly significant moderate negative correlation with serum ferritin (r= -0.55, p<0.001) was observed. And a negligible positive correlation of P300 amplitude with haemoglobin (r= 0.26, p<0.05) and serum ferritin (r=0.24, p<0.05) was observed. Conclusion: Cognitive evoked potential is an objective method that aids in the early detection of cognitive impairment. Evaluating the cognitive function in anaemic women and ensuring adequate iron treatment can prevent MCI from progressing to severe forms like dementia and other neuropsychological disorders.

Background: Iron deficiency (ID) is the most common trace element deficiency in childhood. More than 25% of the world’s population is affected by anemia, of which more than 50% suffers from iron deficiency anemia (IDA). Iron deficiency can cause changes in neurotransmitter homeostasis, decrease myelin production, impair synaptogenesis, and decline the function of the basal ganglia. Therefore, IDA adversely affects cognitive functions and psychomotor development. Aim: is to study the effect of iron deficiency on behavior and cognitive function in children using conners parent rating scales and P300 event-related potentials (ERPs). Patients and Methods: This is a cross sectional comparative study included 20 children with iron deficiency, 20 children ​with iron deficiency anemia,and 20 healthy children who attended the pediatric out patient’s clinic and inpatient department of Al-Zahraa hospital –Alazhar University for Girls we include children with iron deficiency ,Children with chronic medical conditions,Children with co-morbid psychiatric diagnoses ,children with any form of psychiatric, physical, developmental or Intellectual disability, chronic underlying disease (e.g. inborn error of metabolism, genetic, endocrine or liver disease, vision or hearing impairment) or neurologic disorders were excluded from the study.

Elektronik Sistem Aplikasi pendaftaran (E-SIAP) merupakan inovasi yang dihadirkan oleh Dinas Kependudukan dan Pencatatan Sipil Kabupaten Blitar, inovasi ini dihadirkan dengan tujuan mengatasi permasalahan dimana antrian yang selalu membludak setiap harinya dan memudahkan masyarakat dalam pengurusan dokumen kependudukan tanpa harus datang ke kantor. Dengan adanya inovasi maka dapat meningkatkan efisien dan efektifitas dalam pemberian pelayanan. Dengan begitu tingkat kepuasan masyarakat bisa meningkat. Tujuan penelitian ini untuk mendeskripsikan dan menganalisis implementasi inovasi E-SIAP. Penelitian ini menggunakan pendekatan deskriptif kualitatif. Teknik pengumpulan data terdiri dari observasi, wawancara, dan dokumentasi. Teknik analisis data dilakukan dengan mengumpulkan data, mereduksi data, menyajikan data, dan penarikan kesimpulan. Hasil penelitian menunjukkan bahwa inovasi E-SIAP ini telah berjalan dengan baik sesuai arahan dari Dirjendukcapil Kemendagri yang mencanangkan go-digital dan Undang-Undang Administrasi Kependudukan. Sumber ide inovasi diperoleh dari gayung bersambut antara masyarakat dan pemerintah Dispendukcapil Kabupaten Blitar. Budaya inovasi sampai saat ini masih ada beberapa masyarakat yang mengurus secara manual, yaitu mereka yang tidak memiliki akses secara layak. Kemampuan dan alat yang terdapat pada inovasi E-SIAP sudah bisa menunjang kelancaran dalam pemberian pelayanan kepada masyarakat. Tujuan dan hasil dinilai sudah baik dengan memudahkan pelayanan. Kemudian adanya respon yang baik dari masyarakat dan hambatan yang paling mendasar yaitu masih ada beberapa generasi yang belum melek teknologi. Dalam mengumpulkan data inovasi untuk inovasi tunggal, pihak Dispendukcapil tidak hanya melibatkan pihak internal organisasi namun juga dengan stakeholder seperti bekerjasama dengan seluruh desa di Kabupaten Blitar dengan adanya Salam Sak Jangkah. AbstractResistance to next-generation anti-androgen enzalutamide (ENZ) constitutes a major challenge for the treatment of castration-resistant prostate cancer (CRPC). By performing genome-wide ChIP-seq profiling in ENZ-resistant CRPC cells we identify a set of androgen receptor (AR) binding sites with increased AR binding intensity (ARBS-gained). While ARBS-gained loci lack the canonical androgen response elements (ARE) and pioneer factor FOXA1 binding motifs, they are highly enriched with CpG islands and the binding sites of unmethylated CpG dinucleotide-binding protein CXXC5 and the partner TET2. RNA-seq analysis reveals that both CXXC5 and its regulated genes including ID1 are upregulated in ENZ-resistant cell lines and these results are further confirmed in patient-derived xenografts (PDXs) and patient specimens. Consistent with the finding that ARBS-gained loci are highly enriched with H3K27ac modification, ENZ-resistant PCa cells, organoids, xenografts and PDXs are hyper-sensitive to NEO2734, a dual inhibitor of BET and CBP/p300 proteins. These results not only reveal a noncanonical AR function in acquisition of ENZ resistance, but also posit a treatment strategy to target this vulnerability in ENZ-resistant CRPC.
