Texte intégralIn this thesis, we discuss vertically oriented mesoporous silica and their characteristic properties, synthesis procedures and applications. It is of particular interest as it provides direct access to underlying substrate through the pores and is suitable for a variety of applications such as molecular sieving, electrode protection, enhanced detection of cations through electrochemistry and electrochemiluminescence, deposition of isolated metallic and polymeric nanowires, DNA biosensor, etc. We then discuss the electrochemically mediated oxidation of natural aminothiols (cysteine, glutathione and homocysteine) by a ferrocene derivative following an EC’ mechanism and the rate kinetics at a bare indium-tin oxide (ITO) electrode. Further, mesoporous silica thin films exhibiting vertical nanochannels were grown by electrochemically assisted self-assembly method onto ITO electrode. When using the silica film deposited electrodes and ferrocenedimethanol (Fc(MeOH)2) as mediator in solution, both cysteine and glutathione can be oxidized by Fc(MeOH)2+ generated at the electrode surface, following an EC’ mechanism. Electron transfer rates were three times faster with cysteine than glutathione, suggesting that selectivity for cysteine could be achieved using mesoporous films. This was exploited for the reagent-free selective detection of cysteine over glutathione by using a ferrocene-functionalized mesoporous silica film on ITO electrode, based on a combination of charge transfer kinetic and mass transport limitations through the oriented nanochannels. This has been demonstrated by cyclic voltammetry and amperometry in a flow injection analysis mode. Experimentally, the lowest concentration of cysteine detected was 3 µM in flow injection analysis mode. Further, microelectrodes made of ITO and carbon were fabricated and modified with mesoporous silica. Carbon microelectrodes were further functionalized with ferrocene and characterized electrochemically. Overall, silica modified electrodes show a good potential in electroanalytical applications for a variety of reagents through their size and charge selective properties

Texte intégralIt has been known for decades that ultra-weak visible and UV light is emitted by most living tissues due to their oxidative metabolism, as well as by some simple reactions in aqueous medium. However, these phenomena are not well known due to the extreme weakness of the light emission, hence called "ultra-weak" luminescence. In this thesis, we propose an original experimental setup to make extremely sensitive measurements of the luminescence produced by a sample per unit volume.We reached a detection limit of ≈ 1 photon s−1cm−3 which corresponds to ≈ 2.10−21 M.s−1. To obtain this result, we combined: 1) the darkest environment possible, 2) an integrating sphere with extreme reflectivity which brings the maximum amount of photons onto the detector, 3) an amplified camera in binary mode (visible : 400 − 900) and a statistical model which optimizes detectivity, and 4) a semi-automatic procedure to operate in darkness. This setup enables the following: 1) the detection limit is ≈ 9.2 photons s−1cm−2 on the camera, which accounts for ≈ 1% of its dark current, 2) ≈ 12% of the photons emitted isotropically by the sample are detected, 3) measurement of light intensity variations is optimized by maximizing the detectivity, rather than by maximiz- ing the signal-to-noise ratio.Then, we studied the luminescence produced by the disproportionation of H2O2 in water. This reaction catalysed in cells by peroxidases, is important for the metabolism, and is critical to the understanding chemiluminescence in general and and luminol-based applications in particular. We quantified for the first time without a catalyst, the dose-dependent luminescence of the disproportionation reaction in pure water, with ≈ 15photonss−1cm−3 for [H2O2 = 90] mM. Using a well-known biological model, Saccharomyces cerevisiae (baker’s yeast), cultured in liquid, we showed a peak in the luminescence associated with the beginning of the exponential growth phase, followed by a sustained decrease over 10 hours. This light production represents approximately 10−5 photons per second per cell.We hope our work leads to better quantitative study of the ultra-weak luminescence in chemistry and biology. This quantification is necessary for understanding the fundamental underlying mechanisms behind luminescence and to develop chemical and biomedical applications

La réaction photocatalytique est un processus hétérogène qui nécessite l'adsorption des polluants à la surface du catalyseur. L'adsorption de phénol (polluant modèle de l'étude) à la surface de catalyseurs à base de TiO2 en suspension ou supporté a donc été étudiée. L'étude de la dégradation du phénol par différents « médias » photocatalytiques dans un réacteur de laboratoire, de volume 1L, parfaitement agité a ensuite été effectuée. L'activité photocatalytique des catalyseurs supportés a été quantifiée en présence ou non d'un perturbateur organique. Enfin, la conception et la mise en œuvre d'un nouveau type de photoréacteur pilote a été présentée. Il a été caractérisé en termes de cinétique de dégradation du phénol et de rendement énergétique.

Texte intégralIn this study, genes expression from four central receptors (GABAA, 5-HT1A, m-opioïdes and a1-adrenergic) involved in the modulation of anxiety was assessed. The impact of anxiety on the cellular and humoral immunity and on the oxidative status at the SNC (neurons and glial cells) and peripheral (lymphocytes, granulocytes and monocytes) level was highlighted. At the same time, the effect of anxiety coupled with an anxiogenic restraint stress, on the cellular and humoral immunity was also evaluated. Indeed, the high level of anxiety induced firstly, a significant depressive effect on cellular (total lymphocytes, TCD4+ and TCD8+) and humoral (IgA and IgE) immunity, and secondly, a significant increase of the level of intracellular reactive oxygen species (ROS) of neurons and glial cells in the cerebral cortex, the cerebellum and the hippocampus and in the peripheral blood granulocytes, monocytes and lymphocytes. In the same way, the anxiety coupled to acute and chronic restraint stress provoked, a depression of some parameters of cellular (total lymphocytes, TCD4+, TCD8+ and NK) and humoral (IgA, E and G) immunity, and a stimulation of others (granulocytes and monocytes). These works thus made it possible to validate scientifically the anxiogenic character of the model of restraint stress, to establish in a valid and reproducible way the bond and the correlation between the high level of anxiety in animals and their oxidative status inductive of a cytotoxicity as well as the role of the expression of coding genes of 4 receptors in the expression of this high anxiety and of a significant oxidative status at the level of the peripheral cells of the immune system and the neurons and glial cells at the central level

Texte intégralParkinson's disease (PD) is characterized by progressive motor disorders resulting in dopaminergic neurons degeneration in the substantia nigra. In this pathology, aging is a major predisposing factor. During my thesis, I examined initially, in vivo models used in the laboratory to study the PD in Drosophila, namely the expression of α-synuclein and paraquat. I have contributed to the identification of dopaminergic neurons subpopulation involved in locomotor effects of α-synuclein and highlighting the role of a dopamine D1-like receptor in neurotoxicity of paraquat. I then shown that the activity of dopaminergic neurons accelerated senescence and decreases Drosophila lifespan by contributing significantly to the production of ROS in the brain. Finally, we identified and characterized Drosophila homologous of LAMP-2A protein, involves in autophagy-mediated chaperone (AMC), and demonstrated that the increase in neuronal clearance of cytosolic proteins by the expression human LAMP-2A or its Drosophila homologue has positive effects on the locomotor decline associated with aging and resistance against PD factors, but it does not increase longevity. These results suggest that the CMA is a conserved mechanism in Drosophila and its activation protects against oxidative stress and neuronal aging
