Aim. The increasing of riboflavin accumulation by chemical mutagenesis of Bacillus subtilis IMB B-7797 strain producer was the aim of this work. Methods. We used the method of treating the strain producer with a chemical mutagen N-methyl-N-nitro-N-nitrosoguanidine to achieve this goal and followed by culturing the resulting clone and determining the accumulation of riboflavin. Results. B. subtilis IFBG NTG2 mutant strain producer which differed in morphological characteristics (color, size, colony shape) and riboflavin accumulation from the original culture as a result of chemical mutagen action on B. subtilis IMB B-7797 was obtained. Conclusions. B. subtilis IFBG NTG2 mutant strain producer as a result of action on B. subtilis IMB B-7797 by chemical mutagen and which differed in morphological features (color, size, colony shape) and riboflavin accumulation from the original culture. was obtained, B. subtilis IFBG NTG2 strain producer, which produced riboflavin amount of 14.8 g/dm3, which is 9% more than produced by the original B. subtilis IMB B-7797 strain producer was obtained by chemical mutagenesis. B. subtilis IFBG NTG2 strain produser is further proposed for use in industrial technology of riboflavin and creation of a recombinant strain produser for synthesis of riboflavin.

The relationship between aflatoxin B1 and G1 was examined in samples from 199 aflatoxin contaminated lots of inshell Brazil nuts imported to Europe. In most of the samples, the relationship between B1 and G1 were approximately 50/50 indicating that the major responsible aflatoxin producing fungi cannot be Aspergillus flavus, which produces solely B aflatoxins. Fungal strains were isolated from two batches of Brazil nuts and isolates of both A. nomius and A. flavus could be identified. The A. nomius isolates were good producers of both B and G aflatoxins, while the A. flavus strains only produced B aflatoxins. In conclusion, this study suggests that A. nomius is an important producer of aflatoxins in Brazil nuts and that its occurrence, and possibly other B and G aflatoxin producers, should be further examined since this may influence strategies for prevention and control of aflatoxins in Brazil nuts.

The study was conducted, in Pratapgarh district of Uttar Pradesh. Random sampling technique was used for the selection of blocks, villages and proportionate random sampling for selection of growers. From the list, 200 growers were selected, using proportionate sampling method i.e. 90 small, 70 medium and 40 large farmers respectively. The primary data were collected from the respondents by using interview schedule, while secondary data were collected from the official records, published data, magazines etc. The marketable surplus for Aonla in the area was found to be 140, 160 and 180 quintals per farm which constituting (99.10%), (99.48%) and (99.48%) to their total Aonla production. Channel-I, Marketing cost when producers sold their produce to consumer in the market was Rs.90/quintal. Net price received by the producer is 410/quintal. Producer share in consumer price was 82 per cent. Price spread is Rs 90. Marketing efficiency was 5.55 per cent. Channel-II, Marketing cost when producers sold their produce to retailers was Rs.105/quintal. Among these cost transportation charges was most important which accounted for Rs.15/quintal, followed by loading and unloading cost Rs.10/quintal, market cost Rs.10/quintal, labour cost was Rs.10/quintal and miscellaneous cost Rs.50/quintal respectively. Sale price of the producer to retailer was Rs.500/quintals inn different farms size group. Channel-III, this is identified as the longest channel. The producer sells his produce to the commission agents, who in turn sell it to retailer in the market. Finally, the produce reaches to the consumer after collecting margin. Average marketing cost when producer sold their produce to commission agents, in the market was Rs.165. Among these grading, cleaning etc. was Rs. 10 and 10 per Qts. loading and unloading cost Rs. 10 per Qtl. Transportation cost Rs. 20per Qts, Miscellaneous charges Rs. 25/qts, respectively.

We study production competition between two electricity producers, where one of them is subject to a nationalization decision and the other is a private producer that chooses managerial incentives to counter governmental actions. The government wants to maximize a modified form of social welfare and chooses partial nationalization, which still has a serious impact on the rival private producer. We find, that by offering managerial incentives the private producer recovers its lost profit and induces even less nationalization. We also find that such equilibrium might produce the same level of social welfare than one without incentives.

Willingness-to-pay (WTP) estimates help agribusinesses estimate whether a new product is likely to be profitable. For produce, new products, such as new fruit varieties, need to be adopted by producers before they can be sold to consumers. The study of ex ante fruit and vegetable producer preferences is relatively new. This study uses meta-regression analysis to compare the estimated WTP premium between U.S. producers and consumers to determine whether they differ. After controlling for differences in study methods, product attributes, and potential publication bias, the producer WTP was between 14.16 and 27.73 percentage points higher. Subject to several caveats and limitations, this suggests that consumer WTP can be a sufficient metric for the profitability of new produce products.

Allelopathic interactions mediated by bacteriocins production serve microorganisms in the never-ending battle for resources and living space. Competition between the bacteriocin producer and sensitive populations results in the exclusion of one or the other depending on their initial frequencies, the structure of their habitat, their community density and their nutrient availability. These interactions were extensively studied in bacteriocins produced by Escherichia coli, the colicins. In spatially structured environments where interactions are local, colicin production has been shown to be advantageous to the producer population, allowing them to compete even when initially rare. Yet, in a well-mixed, unstructured environment where interactions are global, rare producer populations cannot invade a common sensitive population. Here we are showing, through an experimental model, that colicin-producers can outcompete sensitive and producer populations when the colicin production rates are enhanced. In fact, colicin production rates were proportional to the producer competitive fitness and their overall success in out-competing opponents when invading at very low initial frequencies. This ability of rare populations to invade established communities maintains diversity and allows the dispersal of beneficial traits.

Abstract Studies on the social-psychological framework known as farmer identity have been conducted in the US, EU, and Australia. The focus of these studies is on understanding how farmer beliefs (i.e. Identities) translate into on-farm practices. For example, in 2015, Iowa farmers were surveyed and four identities emerged when asked which items were important to being a “Good Farmer.” Those identities were used to predict the support of soil and/or water policies in the state. Here, for the first time, we explore the identities of livestock producers. Specifically, Pennsylvania swine producer identities. Data were collected as part of a state-wide biosecurity survey. Eighty-four respondents answered some part of the survey, while 50 respondents completed all 31 items of the “Good Producer” question. Principal components analysis (PCA) was used to explore producer perception of a good producer (i.e. producer identities). Using the Kaiser-Meyer-Okin Measure (KMO=0.532) and Barrtlett’s Test of Sphericity (sig. = 0.000) we determined PCA results to be acceptable for exploratory purposes. Five producer identities (friendly conservationist, civically-savvy, willing naturalist, productivist, and appearance-minded) were identified, explaining 58.15% of the variance in these data. Reliability analysis confirmed the strength of items defining each component (i.e. identity). These identities are being used in discussion with PA swine producers to understand how producers self-identify. These identities will be used to predict whether a producer has a formal biosecurity plan written.

This article focuses on a profession of key importance for understanding today’s screen industries in Central-Eastern Europe: the independent producer. Using the approach of critical production studies, the article focuses on producers’ ‘reflexivity’ to reveal how their professional identity is being constructed and how they are positioning themselves within the broader ecology of the media industry. By analysing a set of semi-structured interviews with Czech producers of all kinds, this article identifies five recurrent tropes related to their ‘self-conceptions’. The tropes demonstrate how the producers perform their identities differently from their UK or US counterparts: as largely disempowered, dependent on public support and on the powerful public service broadcaster, desperately looking for more stability, autonomy and recognition.

Counts of lactic acid bacteria (LAB), total anaerobes and enterococci were determined in the course of ripening in the edge part (E) and the core part (C) of Dutch-type semi-hard cheese produced with different fat content (30 and 45 %) by two different producers (H and R) using two different starter cultures (L and Y). Counts of LAB at the beginning of ripening (day 0) in H producer´s samples were higher (P < 0,01) in comparison with the R producer´s ones. Count of enterococci was the highest (P < 0,05) at the end of the ripening (176th day) in sample R30YE. Higher (P < 0,01) enterococci counts were in R producer´s cheeses (in comparison with the H producer´s ones). Enterococci contamination was higher (P < 0,05) in E-samples than C-samples. Content of the sum of all BA in cheese was negatively correlated (P < 0,05) with counts of lactic acid bacteria (r = –0,24) and counts of total anaerobes (r = –0,23). No correlation between the sum of BA content and enterococci counts was found.

The effect of bed height and density on producer gas combustion quality from house hold scale gasification.Increased energy demand caused the crude oil to be expensive and limited so that renewable energy could be a solution of the crisis energy in the future. Rice husk that produced from rice mill had potential as energy in the rural area due to had energy content around 3000-3500 kcal/kg. Rice husk had proximate analysis for fixed carbon 15%, volatile matter 50%, ash 20%, and moisture 15% so that rice husk could be converted to the gas by gasification. Gas producer that was produced by gasification can be used for household fuel. The aim of this research was to observe influence of density and height of rice husk bed to the flame of the gas producer combustion in the small scale gasification. Small scale gasification was done in gasification stove that had capacity 1000 g rice husk. Variations in this research were density of rice husk bed (85, 95, and 105 kg/m3) and height of rice husk bed (25, 40, and 55 cm). Parameter of quality of gas producer combustion was holding time of the flame temperature above 500 oC. The results showed the combustion quality of the gas producer was influenced by density and height of rice husk bed.Keywords: rice husk, gasification, gas producer, bed height, bed density AbstrakPeningkatan kebutuhan energi menyebabkan minyak bumi menjadi mahal dan terbatas sehingga energi terbarukan dapat menjadi solusi untuk menggantikan minyak bumi pada masa mendatang. Salah satu sumber energi terbarukan adalah sekam padi yang dihasilkan dari pengolahan padi dengan kandungan energi 3000-3500 kcal/kg sekam padi kering. Sekam padi memiliki komposisi karbon sekitar 15%, volatile matter 50%, abu 20%, dan kandungan air 25% sehingga sekam padi dapat dikonversi melalui proses gasifikasi. Dengan metode gasifikasi skala rumah tangga, gas produser yang dihasilkan dapat dipergunakan sebagai bahan bakar rumah tangga. Tujuan penelitian ini adalah mengamati pengaruh tinggi dan kerapatan unggun terhadap kualitas pembakaran gas produser hasil gasifikasi sekam padi. Parameter kualitas pembakaran adalah pengukuran temperatur lidah api dengan posisi tetap. Waktu tahan temperatur di atas 500 oC menjadi standar kualitas proses gasifikasi sekam padi yang dipengaruhi oleh kerapatan dan tinggi unggun sekam padi. Gasifikasi dilakukan pada kompor gasifikasi skala rumah tangga yang mampu memuat sekam padi 1000 g. Variasi tinggi unggun adalah 25, 40, dan 55 cm serta kerapatan unggun 85, 95, dan 105 kg/m3. Hasil penelitian menunjukkan bahwa kualitas pembakaran gas produser dipengaruhi oleh tinggi dan kerapatan unggun.Kata kunci: sekam padi, gasifikasi, gas produser, tinggi unggun, kerapatan unggun

Objectives: This cross-sectional study was designed to detect the carbapenemase producing K. pneumoniae along with biofilm producers from different clinical specimens and to compare antibiotic susceptibility pattern of biofilm producing carbapenem resistant Klebsiella pneumoniae and biofilm non-producing carbapenem resistant Klebsiella pneumoniae. Methods: A total of 1475 non-repetitive clinical samples were included on this study. Antibiotic Sensitivity Testing (AST), Modified Hodge Test (MHT) and Modified Carbapenem inactivation method (mCIM) were performed for detection of carbapenemase production and Congo red agar method (CRA) along with Microtitre plate method were performed for detecting biofilm production. Results: Among the clinical specimens cultured, growth positivity was 62.71%. E. coli was most predominant organism followed by K. pneumoniae (17.89%). Among the 110 K. pneumoniae, 57 were found to be carbapenemase producer. Majority of the carbapenemase producing K. pneumoniae were isolated from sputum (45.61%), in the specimen collected from age group 61-70 (28.07%) and in out-patient department (50.88%). Similarly, 65.45% K. pneumoniae out of 110 were found to be biofilm producer by Congo red agar method while among those 72, 73.59% isolates were found to be quantitatively biofilm producer in Microtitre plate assay. Out of 57 carbapenemase producer, 35.08% were strongly biofilm producer while among 53 carbapenemase non-producer 30.18% were strongly biofilm producer from Congo red agar method. Moreover, Microtitre plate assay evidenced that, out of 57 carbapenemase producer, 40.35% were highly biofilm producing and among the 15 carbapenemase nonproducer 66.66% were highly biofilm producer. Conclusion: Biofilm formation is highly prevalent with varying degree of resistance among different antibiotics including carbapenems that further augments antibiotic resistance. The study showed carbapenemase producers are stronger biofilm producer than the non-carbapenemase producer. Therefore, it is recommended to identify biofilm formation among carbapenemase producers for effective choice of antibiotics.

The volume of hot dog sausages plays an important role in meat production. Due to their desirable eating properties (softness, juicy, mild taste and smell) they form a group of sausages acceptable for all consumer categories. They are produced according to producer’s specification, which are not the same. Therefore, the differences in quality may be expected. The objective of this paper was to make a comparative analyses of three parameters in five different products present on Novi Sad market: the content of water, fat and protein, and to determine the average content of these parameters in hot dogs produced by different producers. The average content of water ranged from 56.29±3.30% to 61.44±4.08%. The differences between the average content of water in the product of producer A and C were with no statistic significance (p=0.05), and there was no difference between the average water content in other products. The average total fat in hot dogs ranged from 17.60±3.07% to 20.66±4.65%. The average fat content in the products of D and B producer, as well as Cand B, was without statistically significant difference (p=0.05). The average fat content in the products of producer E and D differed p=0.01. In all other comparisons the difference between the average fat content was p=0.001. The average meat protein content ranged from 11.77±1.08% to 15.41±1.82%. Between the products from producer A and C there was no statistical significant difference (p=0.05). The average meat protein content in the products of D and E producers showed statistic difference of p=0.01, but the average content in other producers was p=0.001. The content of meat protein in all hot dogs were within limits of regulations (10%). Statistically significant difference of the average water content in five different producers was discovered in one case (out of ten), the average fat content in two, and the average protein content in one of the examined products.

Equol is a metabolite of isoflavone daidzein and has an affinity to estrogen receptors. Although equol is produced by intestinal bacteria, the association between the status of equol production and the gut microbiota has not been fully investigated. The aim of this study was to compare the intestinal bacteria responsible for equol production in gut microbiota between equol producer and non-producer subjects regarding the intake of daidzein. A total of 1044 adult subjects who participated in a health survey in Hirosaki city were examined. The concentration of equol in urine was measured by high-performance liquid chromatography. The relative abundances of 8 bacterial species responsible for equol production in the gut microbiota was assessed using 16S rRNA amplification. There were 458 subjects identified as equol producers. The proportion of equol production status and the intake of daidzein increased with age. Daily intake of daidzein was larger in equol-producer. The intestinal bacteria, which convert daidzein to equol were present in both equol producers and non-producers. However, the relative abundance and the prevalence of Asaccharobacter celatus and Slackia isoflavoniconvertens were significantly higher in equol producers than those in equol non-producers. The intestinal bacteria that convert daidzein to equol are present in not only the equol producers but also in the non-producers. The daidzein intake is associated with the equol production status through an increase of A. celatus and S. isoflavoniconvertens in the gut microbiota.

MPA (mycophenolic acid) is an immunosuppressive drug produced by several fungi in Penicillium subgenus Penicillium. This toxic metabolite is an inhibitor of IMPDH (IMP dehydrogenase). The MPA-biosynthetic cluster of Penicillium brevicompactum contains a gene encoding a B-type IMPDH, IMPDH-B, which confers MPA resistance. Surprisingly, all members of the subgenus Penicillium contain genes encoding IMPDHs of both the A and B types, regardless of their ability to produce MPA. Duplication of the IMPDH gene occurred before and independently of the acquisition of the MPAbiosynthetic cluster. Both P. brevicompactum IMPDHs are MPA-resistant, whereas the IMPDHs from a non-producer are MPA-sensitive. Resistance comes with a catalytic cost: whereas P. brevicompactum IMPDH-B is >1000-fold more resistant to MPA than a typical eukaryotic IMPDH, its kcat/Km value is 0.5% of ‘normal’. Curiously, IMPDH-B of Penicillium chrysogenum, which does not produce MPA, is also a very poor enzyme. The MPA-binding site is completely conserved among sensitive and resistant IMPDHs. Mutational analysis shows that the C-terminal segment is a major structural determinant of resistance. These observations suggest that the duplication of the IMPDH gene in the subgenus Penicillium was permissive for MPA production and that MPA production created a selective pressure on IMPDH evolution. Perhaps MPA production rescued IMPDH-B from deleterious genetic drift.

Selorejo Village, Blitar Regency is a village where some of the residents produce Opak Gambir snacks which are still managed using traditional methods. This causes the management in the production to be less professional and less organized. This program aims to improve education and training, especially in the fields of entrepreneurship, finance, and marketing, so that the management of this home industry will be more organized and professional. The method of implementing the program is by providing education, training, and coaching for producers. The results of the program show that the producer is very well educated about the principles of entrepreneurship. Opak Gambir producers are also able to make financial plans both for administration and also the selling price of their products. The producer is also able to determine marketing plans, which include starting from the process of determining prices, determining distribution locations, and how the promotion process is carried out. Through this community service, it is hoped that the producer will be able to manage their business in a professional manner, apart from being a source of income, is also a destination for the Opak Gambir industry.

Potatoes are producing over 280 million in whole world and Turkey is 6th potato producer. Turkey produces 5250000 tonne of potatoes. Approximately 20% of potatoes are waste in Niğde. Our study aimed to produce bioethanol from Solanum tuberosum by using the yeast Saccharomyces cerevisiae. As a result renewable energy sources can be produced from natural wastes.

In their struggle for life, bacteria frequently produce antagonistic substances against competitors. Antimicrobial peptides produced by bacteria (known as bacteriocins) are active against other bacteria, but harmless to their producer due to an associated immunity gene that prevents self-inhibition. However, knowledge of cross-resistance between different types of bacteriocin producer remains very limited. The immune function of certain bacteriocins produced by the Enterococcus genus (known as enterocins) is mediated by an ABC transporter. This is the case for enterocin AS-48, a gene cluster that includes two ABC transporter-like systems (Transporter-1 and 2) and an immunity protein. Transporter-2 in this cluster shows a high similarity to the ABC transporter-like system in MR10A and MR10B enterocin gene clusters. The aim of our study was to determine the possible role of this ABC transporter in cross-resistance between these two different types of enterocin. To this end, we designed different mutants (Tn5 derivative and deletion mutants) of the as-48 gene cluster in Enterococcus faecalis and cloned them into the pAM401 shuttle vector. Antimicrobial activity assays showed that enterocin AS-48 Transporter-2 is responsible for cross-resistance between AS-48 and MR10A/B enterocin producers and allowed identification of the MR10A/B immunity gene system. These findings open the way to the investigation of resistance beyond homologous bacteriocins.

Amycolatopsis coloradensis ATCC 53629 is the producer of the glycopeptide antibiotic avoparcin. While setting up the production of the avoparcin complex, in view of its use as analytical standard, we uncovered the production of a to-date not described ristosamynil-avoparcin. Ristosamynil-avoparcin is produced together with α- and β-avoparcin (overall indicated as the avoparcin complex). Selection of one high producer morphological variant within the A. coloradensis population, together with the use of a new fermentation medium, allowed to increase productivity of the avoparcin complex up to 9 g/L in flask fermentations. The selected high producer displayed a non-spore forming phenotype. All the selected phenotypes, as well as the original unselected population, displayed invariably the ability to produce a complex rich in ristosamynil-avoparcin. This suggested that the original strain deposited was not conforming to the description or that long term storage of the lyovials has selected mutants from the original population.

Musty and earthy odor are present in the source water of Feng-Shen waterworks (FSW) in south Taiwan all year round. Although 2-methylisoborneol (2-MIB) and geosmin are responsible for the musty and earthy odor, respectively, the possible odor producers remained unknown. In this study, actinomycetes that produce 2-MIB and geosmin were studied. Based on the 16S rDNA sequence, three species were identified after comparison with the GenBank database at the NCBI. The three species are Streptomyces malaysiensis as a 2-MIB and geosmin producer, Streptomyces caelestis as a 2-MIB producer, and Streptomyces roseoflavus as a geosmin producer. For S. malaysiensis and S. caelestis, the production of MIB and/or geosmin was increased with incubation temperature (20–30 °C) in starch–glutamate broth medium. The optimal MIB, geosmin and biomass occurred at 30 °C. The maximal geosmin/biomass (G/B) and/or MIB/biomass (M/B) also occurred at 30 °C, and sporulating cultures contained more geosmin and/or MIB than nonsporulatig cultures.

This study proposed a dual fuel operation of a mix of gasoline and producer gas from coal gasification on the spark ignition engine. The experiment was carried out on a constant load with variations in speed for single fuel operation of gasoline and dual fuel operation of a mix of gasoline and producer gas to see the influence on speed, torque, power, and braking (effective pressure). The power produced was compared to power produced by the single fuel of producer gas that has been reported in the literature. The result shows an increase of speed would increase torque, power, and braking (effective pressure) for single fuel operation of gasoline and dual fuel operation of a mix of gasoline and producer gas. The power operation of dual fuel of a mix of and gasoline and producer gas will decrease by about 10.9% compared to operation of single fuel of gasoline, and the power operation of the single fuel of producer gas will decrease by about 20% compared to the operation of the single fuel of gasoline. The maximum shaft power produced by dual fuel operation is 1.49 kW at a load of 5 kg and a speed of about 3,500 rpm.

ABSTRACT Bacillus subtilis strain 168 produces the extremely stable lantibiotic sublancin 168, which has a broad spectrum of bactericidal activity. Both sublancin 168 production and producer immunity are determined by the SPβ prophage. While the sunA and sunT genes for sublancin 168 production have been known for several years, the genetic basis for sublancin 168 producer immunity has remained elusive. Therefore, the present studies were aimed at identifying an SPβ gene(s) for sublancin 168 immunity. By systematic deletion analysis, we were able to pinpoint one gene, named yolF, as the sublancin 168 producer immunity gene. Growth inhibition assays performed using plates and liquid cultures revealed that YolF is both required and sufficient for sublancin 168 immunity even when heterologously produced in the sublancin-sensitive bacterium Staphylococcus aureus. Accordingly, we propose to rename yolF to sunI (for sublancin immunity). Subcellular localization studies indicate that the SunI protein is anchored to the membrane with a single N-terminal membrane-spanning domain that has an Nout-Cin topology. Thus, the bulk of the protein faces the cytoplasm of B. subtilis. This topology has not yet been reported for known bacteriocin producer immunity proteins, which implies that SunI belongs to a novel class of bacteriocin antagonists.

This study aims to calculate the additional amount of housewife income in Ujong Tanoh Darat Village, Meureubo District, West Aceh Regency as a producer of sun-dried bilimbi. Respondents in this study were ten producers in Ujong Tanoh Darat Village. Data collection techniques by observing and interviewing in detail related to the analysis of sun-dried bilimbi producer income. The data analysis method uses cost analysis, production revenue, production income, and revenue cost ratio (R / C). From the results of data processing in this study, it found that the average income earned by producers was Rp.300,550 per cycle, the average producer income was Rp. 602,000 per cycle and the average production cost incurred by producers is Rp.301,499 per cycle, and the R per C ratio obtained is 1.9, greater than 1 meaning the sun-dried bilimbi producer business in Ujong Tanoh Darat Village, Meureubo District West Aceh is feasible.
