Texte intégralThe aim of the present research was to evaluate the biology and fertility life table of Diaphorina citri (Kuwayama, 1908) (Hemiptera: Liviidae) comparatives in the two species of Murraya [Murraya paniculata (L.) Jack e Murraya koenigii (L.) Sprengel], and to evaluate the biology of Tamarixia radiata Waterston, 1922 (Hymenoptera: Eulophidae) in the two species studied in confinement, aiming to provide subsidies for pest control in Integrated Pest Management (IPM). Experiments were carried out in confinement (no choice) to evaluate the effect of the two species of Murraya on the feeding and fertility of D. citri, as well as the biology of Asian Citrus Psyllid (ACP) in two hosts. Biology and the effect of host on parasitism of T. radiata on nymphs of D. citri was evaluated. It can be concluded that: D. citri preferred to feed on Murraya koenigii (L.) Sprengel, but for oviposition the preferred host was Murraya paniculata (L.) Jack. However, the other parameters observed indicate that M. paniculata is the most suitable host for the development of the psyllid. According to the fertility life table, the reproduction net rate (R0) of D. citri was 3.1 times higher if reared on M. paniculata; the other parameters, except the mean time of each generation (T), also supported results when it was reared on M. paniculata. The development and parasitism of T. radiata on nymphs of D. citri were affected when reared on different hosts, and better results were observed when T. radiata reared on M. koenigii. The results obtained may provide subsidies for the control and management of the HLB.

Texte intégralThe plant Murraya paniculata is native in Asia where its root barks have produced the contraceptive and hormonal alkaloid yuehchukene. In Brazilian specimens no alkaloid was detected. This had inspired new researches about the microorganisms that interact with it in Brazil, including the present work. Therefore, 1711 endophytic microorganisms were isolated from four host plants, in São Paulo State, and the seasonal colonization indexes, the microbial frequencies and the specificities of the identified groups per tissues (microhabitats) were verified. Among the obtained endophytes, 1061 were filamentous fungi and 650 were bacteria, with 62% and 38% of frequency and 33% and 20% of colonization index, respectively. The colonization indexes of the bacteria in comparison to the of fungi were lower in all gatherings and hosts, principally for leaves that, had shown the bigger frequency for the fungi and the smaller for the bacteria. Fungi were clustered in 20 morphological groups, 15 identified to genus level and five unknown, including two Mycelia sterilia. The recognized genera were Fusarium, Colletotrichum and its teleomorphs Giberella and Glomerella respectively, Trichoderma, Phomopsis, Paecilomyces, Penicillium, Aspergillus, Epicoccum, Pestalotiopsis, Alternaria, Xylaria, Nigrospora and Guignardia and the unknown were named A-E groups. The more frequent genera were rich in diversity, according to morphological and molecular data. A more diversity of fungi genera were verified in the winter (17) and a less in the spring (9) as the endophytic fungi were smaller in quantity in the summer (44.7%) and greater in the autumn (77.7%). From a sample of 59 isolates and six fungi as prototypes genomic DNAs were obtained and their ITS-1 regions were sequenced. The unknown strain B (Fed) had sequenced its 5.8s and ITS-2 regions. 21 species were identified by their ITS-1 regions and two others were hypothesized as novelty: one Trichoderma and the Fed that all morphological and chemotaxonomic studies appoint as a new species. Trichoderma associated to M. paniculata were in majority from barks and had showed great diversity of species (9). It was evaluated too the potential of the endophytes from host for bioactive metabolite production. The more promising groups for extracellular substances were the fungi Penicillium, with bactericidal and antiprotozoal actions, Pestalotiopsis, active against Candida, and Trichoderma, with fungicidal and bactericidal activities. For Fed it was noted its intracellular metabolites as strong bactericidal and its volatiles as anti-fungi. By looking for the active principles of those microorganisms, from Penicillium citrinum the citrinin and an undetermined alkaloid were obtained and from Trichoderma koningii 13 metabolites, including four koninginins related to those biological activities observed were identified. From Fed-3, 10 substances were obtained, seven fixed and three volatile, including a novel manosil guaiane type sesquiterpene and the new, as natural substances, 3-Hydroxy-5-phenylmethyl- (3S,5R)-b10-O-tetrahydrofuran-2-one and chloroacetonitrile. The chloroacetonitrile was suggested as the active principle of the Fed s volatiles. This work was important to reveal the diversity of the endophytes from Brasilian M. paniculata, the high variability of their more frequent genera, two possible novel species and the potential of them to produce bioactive and new metabolites. This work is hoped to contribute for understanding the endophytic microorganism biodiversity as well as their chemical and biotechnological potential.
A planta Murraya paniculata é de origem asiática e em seu continente de origem foi isolado, das cascas das raízes, o alcalóide yuehchukeno que apresenta atividade anticoncepcional e hormonal. Em espécimes brasileiros nenhum alcalóide foi detectado, o que inspirou novas pesquisas envolvendo o estudo dos microrganismos que interagem com esta, incluindo o presente trabalho. Assim, foram isolados 1.711 microrganismos endofíticos de quatro hospedeiras, no estado de São Paulo, verificando-se os índices de colonização sazonal, a freqüência e a especificidade dos grupos identificados por tecidos (micro-habitat). Dos isolados, 1.061 eram fungos filamentosos e 650 eram bactérias, com freqüências de 62% e 38% e índices de colonização de 33% e 20%, respectivamente. Os menores índices de colonização foram observados para as bactérias em todas as coletas, principalmente nas folhas que apresentaram a maior freqüência para os fungos. Os fungos endofíticos foram classificados em 20 grupos morfológicos, 15 identificados ao nível de gênero e cinco desconhecidos, sendo dois Micelia sterilia. Os gêneros identificados foram Fusarium, Colletotrichum e seus telemorfos Giberella e Glomerella rescpectivamente, Trichoderma, Phomopsis, Paecilomyces, Penicillium, Aspergillus, Epicoccum, Pestalotiopsis, Alternaria, Xylaria, Nigrospora e Guignardia e os não identificados foram denominados como grupos A-E. Foi também detectada grande diversidade dentro dos gêneros mais freqüentes, pela morfologia e por marcadores moleculares. Foi verificada maior diversidade de gêneros de fungos no inverno (17) que na primavera (9). A população de fungos endofíticos foi menor no verão (44,7%) em contraste com o outono (77,7%). De uma amostragem de 59 isolados e seis controles foram extraídos os DNAs genômicos e seqüenciadas as regiões ITS-1. Para a linhagem indeterminada B (Fed) foi sequenciada a região 5,8s e a ITS-2. Foram identificadas 21 espécies pela região ITS-1 e detectadas duas possíveis espécies novas: uma de Trichoderma e o Fed cujos estudos morfológicos e quimiotaxonômicos apóiam esta hipótese. Os Trichoderma associados a M. paniculata foram isolados principalmente das cascas e apresentaram 9 espécies, sendo uma indeterminada. Foi também avaliado o potencial para produção de substâncias bioativas de endófitos. Os grupos mais promissores para a produção de substâncias extracelulares foram os dos fungos Penicillium com ação bactericida e anti-protozoária, Pestalotiopsis candicida e Trichoderma fungicida e bactericida. Fed com substâncias intracelulares de forte ação bactericida e de voláteis fungicidas. Buscou-se ainda o isolamento dos princípios ativos tendo sido isolados de Penicillium citrinum a citrinina e um alcalóide indeterminado, de Trichoderma koningii 13 substâncias, das quais quatro koningininas relacionadas às atividades biológicas detectadas. De Fed-3 foram identificadas 10 substâncias, sete fixas e três voláteis, incluindo um sesquiterpeno guaiano manosilado inédito e as substâncias 3-Hidróxi- 5-fenilmetil-(3S,5R)-b10-O-tetrahidrofuran-2-ona e cloroacetonitrila, isoladas pela primeira vez de fonte natural. A esta última é sugerida a ação fungicida dos voláteis. Esta pesquisa foi oportuna ao revelar a diversidade de microrganismos associados à hospedeira, a alta variabilidade entre e dentre os gêneros, incluindo duas possíveis novas espécies, e o potencial na produção de moléculas bioativas e inéditas. Espera-se ter contribuído para o conhecimento da biodiversidade dos microrganismos endofíticos e do potencial químico e biotecnológicos que eles representam.

Texte intégralCurrently, the control of the disease Huanglongbing (HLB) or greening is based mainly on the chemical control of the insect vector Diaphorina citri, through intensive applications of insecticides, which has been causing secondary pest outbreaks, pest resurgence, selection of population resistant and elimination of natural enemies. The goal of this work was to evaluate the possibility of using the curry leaf (Murraya koenigii) as a trap crop for the management of the D. citri. For this, the following tests have been performed: i) Transmission test of \'Candidatus Liberibacter asiaticus\' to determine if the curry leaf is not a host for the bacterium; ii) test of choice behavior, to verify the host preference of D. citri to curry leaves in relation to citrus plants; iii) test of probing behavior in curry leaf using the Electrical Penetration Graphs (EPG) technique, in order to understand the feeding of the insect on this plant; iv) selection of systemic insecticides for use in curry leaf, determining the residual period and efficiency; and v) test with trap crop in a citrus orchard, in which curry plants were installed in front of the citrus plants and the psyllid was monitored with the yellow stick traps. As a result, it has been observed that the curry leaf is not a host to \'Ca. L. asiaticus\' by the D. citri inoculation and, in a free choice and olfactometer test, is more attractive to the psyllid than the citrus plant under laboratory conditions. The probing behavior of the psyllid in curry leaf is different from citrus and orange jasmine, being the ingestion of xylem sap in curry leaf higher. In addition, the insecticides applied by \'drench\' such as thiamethoxam and imidacloprid in these plants are efficient against D. citri for more than 70 days after application and, in the field, the capture of psyllids in the plots with the curry leaf was higher and could therefore conclude that, the curry leaf M. koenigii is a potencial plant to be used as a trap crop for the management of D. citri and consequently, the disease HLB.

The Penicillium sp. A, Penicillium sp. B (M. paniculata) and P. herquei (M. azedarach) fungus were grown in liquid media (Czapeck s) and also in solid media (rice). After the inoculation of the fungus, their grownth were interrupted by adition of methanol. The extracts of the biomass were obtained by several maceration technics using organic solvents. The crude extracts were fractionated by liquid-liquid partition and also several chromatography technics. The pure compounds were obtained by refractionation of previous fractions by column chromatography, thin layer chromatography and HPLC. The structures of the compounds were elucidated by several spectroscopic technics as NMR, MS and IR. Several classes of compounds were isolated as nucleosides, esteroids, carbohydrates, polyketides among many others. Antimicrobial assays were carried out with purified compounds, where the polyketides citrinine, citrinine-D, citrinine H1, emodine, citreoroseine and janthinone showed promising results. Leishmanicidal assays were also carried out using emodine, citreoroseine and janthionone, where only the former showed a reasonable activity. The fragmentation by MS and also the biogenesis proposal of some isolated compounds are discussed in the present work.
Os fungos Penicillium sp A, Penicillium sp B (M. paniculata) e P. herquei (M. azedarach) foram cultivados em meios de cultura líquido (Czapeck s) e meio sólido (Arroz) para crescimento das colônias. Após o período de cultivo os crescimentos dos fungos foram interrompidos com metanol, posteriormente foram obtidos os extratos das biomassas produzidas utilizando-se técnicas de maceração com solventes orgânicos. Os extratos brutos foram fracionados através de partições liquido-liquido e cromatografia em coluna a vácuo. As frações obtidas foram então refracionadas utilizando-se cromatografia em coluna, cromatografia em camada delgada preparativa e HPLC para isolamento dos constituintes químicos. As substâncias tiveram as suas estruturas elucidadas através da utilização de técnicas espectrométricas de RMN, EM e IV. Onde foram isoladas substâncias das classes dos nucleosideos, esteróides, carboidratos, policetídeos, entre outras. Foram realizados ensaios antimicrobianos com as substâncias puras, sendo que os policetídeos citrinina, citrinina-D, citrinina H1, emodina, citreoroseina e janthinona apresentaram bons resultados. Realizaram-se, ainda, ensaios leishmanicidas com as substâncias emodina, citreoroseina e janthinona, somente a primeira apresentou atividade considerável. As propostas de fragmentação por massas e de biogênese de alguns constituintes químicos são discutidas no trabalho.

Texte intégralResumo: Murraya paniculata L. Jack (Rutaceae) é uma árvore amplamente utilizada nas Américas como ornamental. Porém, esta espécie vegetal nativa da Índia é considerada medicinal na Ásia e reconhecida pelas propriedades adstringentes, estimulantes, analgésicas e anti-inflamatórias. Diversos estudos revelam a presença de alcaloides, flavonoides, cumarinas e componentes do óleo essencial, fundamentalmente, e as propriedades antibacterianas, antifúngicas, anti-inflamatórias e analgésicas foram observadas para extratos de M. paniculata. Fitoterápicos derivados da planta estão disponíveis no Sistema Nacional de Saúde cubano, como anti-inflamatórios, porém é limitado o conhecimento dos metabólitos secundários majoritários da espécie coletada em Cuba e sua relação com a atividade farmacológica. Neste contexto, foi desenvolvido este trabalho que teve como objetivo realizar o estudo químico do extrato etanólico de folhas de M. paniculata, a fim de contribuir com sua padronização e com o estabelecimento de especificações de qualidade com base na composição química e atividade anti-inflamatória. O fracionamento do extrato etanólico foi iniciado por extração em fase sólida fornecendo 4 frações. A fração EFSMp4 foi fracionada utilizando-se cromatografia em coluna. A purificação final das substâncias majoritárias obtidas a partir de EFSMp4 foi realizada por cromatografia líquida de alta eficiência preparativa de fase reversa. Texte intégralThe urban forestry offers several benefits for the population and their need is increasing with the growth of cities. But like other urban equipment, there is cost of implementation, maintenance, and also losses. Among the potential damage of urban forestry, are paving the damage to structures and construction. This study aimed to identify the parameters that influence the damage and assist in the definition of public policies for urban forestry. The study was based on an inventory of urban forestry of the city of Piracicaba, in 15 districts. For the entire inventory, the units were identified and depths based on soil survey Pedológica semidetailed the city of Piracicaba. Was used for analysis of cracks in the sidewalk, a tree species, the sibipiruna (Caesalpinia rainy Dc.), And a bush, the falsa-murta (Murraya paniculata (L.) Jacq.), There is a concept widely widespread, that the shrub species, do not cause damage to the paving, while the trees are known to damage the sidewalks. In collecting field and focuses on the following: address, sort, tree removed or replaced, CAP, bed size, lap paved, pruning of shoots, damaged urban structure, and material conditions of the pavement of the floor. The rate of elimination of falsa-murta was equal to the sibipiruna (1.7%), whereas 89.2% of falsa-murta and 92.6% of sibipiruna were not replaced by other species. For the falsemyrtle, the concentration of individuals, was in the range of CAP 21 to 80 cm (82.9%), 70.2% had cracks in the sidewalk and 29.8% no broken; 44.6% of individuals had bed tree to 0.20 m2, 92.3% of individuals have suffered as bad pruning and 75.1% of individuals were in sidewalk cement. For sibipiruna, the concentration of individuals was in the range of 101 to 160 CAP cm (57.0%), 99.4% had cracks in the sidewalk and 0.6% absence of cracks, 44.6% of individuals were planted in locations classified as uncertain beds trees, 94.6% of individuals have suffered as bad pruning and sidewalk was the most common cement with 63.1% of individuals. The depth of soil had no influence on whether or not cause cracks in the two species. With the statistical analysis (Chi-square) could be countered some concepts available in urban forestry, concluding that the damage also shrubs and pruning influence the damage to sidewalks, among others. There is no simple solution to eliminate the conflict between the trees and road equipment and urban structures. The people need and should be informed about all parameters, options, benefits and harm of each action that stimulate or inhibit the planting of a tree in the sidewalk.

Texte intégralThe agro-ecosystems consist of complex trophic relationships between plants, herbivores and their natural enemies. It is known that the majority of plants can produce volatiles compounds used as chemical signals by different groups of insects. These compounds can be produced constitutively in healthy plants, i.e., without induction. In other hand, the production of induced volatiles occurs from the contact of secretions released by phytophagous with injuries caused by feeding or oviposition in plant tissue. For phytophagous, these volatile compounds may signal the presence of the host plant, as well as the presence of conspecifics and the sexual partner. Although, natural enemies, predators and parasitoids, the induced volatiles can signal the presence of phytophagous insects (prey / host) in the plant. In this context, the behavioral responses of Diaphorina citri Kuwayama (Hemiptera: Psyllidae) to plant volatiles of jasmine, Murraya paniculata (L.) Jack (Rutaceae) infested or not by conspecifics, were studied. It was also investigated the responses of psyllids to volatiles of Citrus sinensis infected by bacteria that cause the huanglongbing or HLB, one of the most serious diseases of citrus. Given the studies that aim to identify repellent compounds to D. citri, it was tested plant volatiles of guava, Psidium guajava L. (Myrtaceae) and their impact on plant location of Citrus limonia (L.) Osbeck (Rutaceae) by psyllids. Behavioral responses were measured by Y-tube and four-way olfactometers. Before establishing the olfactometry assays, studies of D. citri sexual behavior were performed. The obtained results showed that the beginning of D. citri sexual activities occurred between the second and third days after emergence, and the psyllids were more active during the photophase. In regard to the effects of plant volatiles, males and females of D. citri differently responded to the same olfactory stimuli. Thus, males were attracted only to odors associated with females. Females were attracted to plant odors, although they avoided odors associated with males, including plants previously infested by them. It was also verified that D. citri adults distinguished volatiles citrus with HLB from volatiles released by healthy plants. Given that, it was clear that volatiles from infected plants were attractive to both males and females psyllids. In search of repellent compounds, also was possible demonstrated that P. guajava volatiles not only hindered the location of plants of C. limonia by D. citri, but also provided repellent effect to psyllids. The findings presented here may help the development of new tactics for the behavioral management of D. citri.
