AbstractIn Caenorhabditis elegans, the Ras/Raf/MEK/ERK signal transduction pathway controls multiple processes including excretory system development, P12 fate specification, and vulval cell fate specification. To identify positive regulators of Ras signaling, we conducted a genetic screen for mutations that enhance the excretory system and egg-laying defects of hypomorphic lin-45 raf mutants. This screen identified unusual alleles of several known Ras pathway genes, including a mutation removing the second SH3 domain of the sem-5/Grb2 adaptor, a temperature-sensitive mutation in the helical hairpin of let-341/Sos, a gain-of-function mutation affecting a potential phosphorylation site of the lin-1 Ets domain transcription factor, a dominantnegative allele of ksr-1, and hypomorphic alleles of sur-6/PP2A-B, sur-2/Mediator, and lin-25. In addition, this screen identified multiple alleles of two newly identified genes, eor-1 and eor-2, that play a relatively weak role in vulval fate specification but positively regulate Ras signaling during excretory system development and P12 fate specification. The spectrum of identified mutations argues strongly for the specificity of the enhancer screen and for a close involvement of eor-1 and eor-2 in Ras signaling.

Establishment and maintenance of proper gene expression is a requirement for normal growth and development. The DREAM complex in Caenorhabditis elegans functions as a transcriptional repressor of germline genes in somatic cells. At 26°, DREAM complex mutants show increased misexpression of germline genes in somatic cells and High Temperature Arrest (HTA) of worms at the first larval stage. To identify transcription factors required for the ectopic expression of germline genes in DREAM complex mutants, we conducted an RNA interference screen against 123 transcription factors capable of binding DREAM target promoter loci for suppression of the HTA phenotype in lin-54 mutants. We found that knock-down of 15 embryonically expressed transcription factors suppress the HTA phenotype in lin-54 mutants. Five of the transcription factors found in the initial screen have associations with Wnt signaling pathways. In a subsequent RNAi suppression screen of Wnt signaling factors we found that knock-down of the non-canonical Wnt/PCP pathway factors vang-1, prkl-1 and fmi-1 in a lin-54 mutant background resulted in strong suppression of the HTA phenotype. Animals mutant for both lin-54 and vang-1 showed almost complete suppression of the HTA phenotype, pgl-1 misexpression, and fertility defects associated with lin-54 single mutants at 26°. We propose a model whereby a set of embryonically expressed transcription factors, and the Wnt/PCP pathway, act opportunistically to activate DREAM complex target genes in somatic cells of DREAM complex mutants at 26°.

Abstract The heterochronic genes lin-4, lin-14, lin-28, and lin-29 specify the timing of lateral hypodermal seam cell terminal differentiation in Caenorhabditis elegans. We devised a screen to identify additional genes involved in this developmental timing mechanism based on identification of mutants that exhibit temporal misexpression from the col-19 promoter, a downstream target of the heterochronic gene pathway. We fused the col-19 promoter to the green fluorescent protein gene (gfp) and demonstrated that hypodermal expression of the fusion gene is adult-specific in wild-type animals and temporally regulated by the heterochronic gene pathway. We generated a transgenic strain in which the col-19::gfp fusion construct is not expressed because of mutation of lin-4, which prevents seam cell terminal differentiation. We have identified and characterized 26 mutations that restore col-19::gfp expression in the lin-4 mutant background. Most of the mutations also restore other aspects of the seam cell terminal differentiation program that are defective in lin-4 mutant animals. Twelve mutations are alleles of three previously identified genes known to be required for proper timing of hypodermal terminal differentiation. Among these are four new alleles of lin-42, a heterochronic gene for which a single allele had been described previously. Two mutations define a new gene, lin-58. When separated from lin-4, the lin-58 mutations cause precocious seam cell terminal differentiation and thus define a new member of the heterochronic gene pathway.

ABSTRACTActivation of a canonical EGFR-Ras-Raf-ERK cascade initiates patterning of multipotent vulval precursor cells (VPCs) of Caenorhabditis elegans. We have previously shown that this pathway includes a negative-feedback component in which MPK-1/ERK activity targets the upstream kinase LIN-45/Raf for degradation by the SEL-10/FBXW7 E3 ubiquitin ligase. This regulation requires a Cdc4 phosphodegron (CPD) in LIN-45 that is conserved in BRAF. Here, we identify and characterize the minimal degron that encompasses the CPD and is sufficient for SEL-10-mediated, MPK-1-dependent protein degradation. A targeted screen of conserved protein kinase-encoding genes yielded gsk-3 (an ortholog of human GSK3B) and cdk-2 (a CDK2-related kinase) as required for LIN-45 degron-mediated turnover. Genetic analysis revealed that LIN-45 degradation is blocked at the second larval stage due to cell cycle quiescence, and that relief of this block during the third larval stage relies on activation of CDKs. Additionally, activation of MPK-1 provides spatial pattern to LIN-45 degradation but does not bypass the requirement for gsk-3 and cdk-2. This analysis supports a model whereby MPK-1/ERK, GSK-3/GSK3 and CDK-2/CDK2, along with SEL-10/FBXW7, constitute a regulatory network that exerts spatial and temporal control of LIN-45/Raf degradation during VPC patterning.

We have investigated the regulation of cell-cycle entry in C. elegans, taking advantage of its largely invariant and completely described pattern of somatic cell divisions. In a genetic screen, we identified mutations in cyd-1 cyclin D and cdk-4 Cdk4/6. Recent results indicated that during Drosophila development, cyclin D-dependent kinases regulate cell growth rather than cell division. However, our data indicate that C. elegans cyd-1 primarily controls G1 progression. To investigate whether cyd-1 and cdk-4 solely act to overcome G1 inhibition by retinoblastoma family members, we constructed double mutants that completely eliminate the function of the retinoblastoma family and cyclin D-Cdk4/6 kinases. Inactivation of lin-35 Rb, the single Rb-related gene in C. elegans, substantially reduced the DNA replication and cell-division defects in cyd-1 and cdk-4 mutant animals. These results demonstrate that lin-35 Rb is an important negative regulator of G1/S progression and probably a downstream target for cyd-1 and cdk-4. However, as the suppression by lin-35 Rb is not complete, cyd-1 and cdk-4 probably have additional targets. An additional level of control over G1 progression is provided by Cip/Kip kinase inhibitors. We demonstrate that lin-35 Rb and cki-1 Cip/Kip contribute non-overlapping levels of G1/S inhibition in C. elegans. Surprisingly, loss of cki-1, but not lin-35, results in precocious entry into S phase. We suggest that a rate limiting role for cki-1 Cip/Kip rather than lin-35 Rb explains the lack of cell-cycle phenotype of lin-35 mutant animals.

In Caenorhabditis elegans, mutations in the lin-2 gene inactivate the LET-23 receptor tyrosine kinase/Ras/MAP kinase pathway required for vulval cell differentiation. One function of LIN-2 is to localize LET-23 to the basal membrane domain of vulval precursor cells. LIN-2 belongs to the membrane-associated guanylate kinase family of proteins. We have cloned and characterized the human homolog of LIN-2, termed hCASK, and Northern and Western blot analyses reveal that it is ubiquitously expressed. Indirect immunofluorescence localizes CASK to distinct lateral and/or basal plasma membrane domains in different epithelial cell types. We detect in a yeast two-hybrid screen that the PDZ domain of hCASK binds to the heparan sulfate proteoglycan syndecan-2. This interaction is confirmed using in vitro binding assays and immunofluorescent colocalization. Furthermore, we demonstrate that hCASK binds the actin-binding protein 4.1. Syndecans are known to bind extracellular matrix, and to form coreceptor complexes with receptor tyrosine kinases. We speculate that CASK mediates a link between the extracellular matrix and the actin cytoskeleton via its interaction with syndecan and with protein 4.1. Like other membrane-associated guanylate kinases, its multidomain structure enables it to act as a scaffold at the membrane, potentially recruiting multiple proteins and coordinating signal transduction.

A screen was conducted for lethal mutations in the nematode Caenorhabditis elegans in a strain containing the mutator mut-4(st700)I to examine the nature of mutator-induced lethal mutations within two large chromosomal regions comprising a total of 49 map units (linkage group IV (right) and linkage group V (left)). The genetic analysis of 28 lethal mutations has revealed that the mutator locus mut-4(st700)I causes both putative single-gene mutations and deficiencies. We have identified lethal mutations in three different genes, in addition to seven deficiencies. There is a mutational hot spot on linkage group V (left) around the lin-40 locus. Six mutations appear to be alleles of lin-40. In addition, 5 of 7 deficiencies have breakpoints at or very near lin-40. All seven deficiencies delete the left-most known gene on linkage group V (left) and thus appear to delete the tip of the chromosome. This is in contrast to gamma ray and formaldehyde induced deficiencies, which infrequently delete the closest known gene to the tip of a chromosome.Key words: Caenorhabditis elegans, mutator, deficiencies, lethal mutations.

Background Falls and the risk of falling in older people pose a high risk for losing independence. As the risk of falling progresses over time, it is often not adequately diagnosed due to the long intervals between contacts with health care professionals. This leads to the risk of falling being not properly detected until the first fall. App-based software able to screen fall risks of older adults and to monitor the progress and presence of fall risk factors could detect a developing fall risk at an early stage prior to the first fall. As smartphones become more common in the elderly population, this approach is easily available and feasible. Objective The aim of the study is to evaluate the app Lindera Mobility Analysis (LIN). The reference standards determined the risk of falling and validated functional assessments of mobility. Methods The LIN app was utilized in home- and community-dwelling older adults aged 65 years or more. The Berg Balance Scale (BBS), the Tinetti Test (TIN), and the Timed Up & Go Test (TUG) were used as reference standards. In addition to descriptive statistics, data correlation and the comparison of the mean difference of analog measures (reference standards) and digital measures were tested. Spearman rank correlation analysis was performed and Bland-Altman (B-A) plots drawn. Results Data of 42 participants could be obtained (n=25, 59.5%, women). There was a significant correlation between the LIN app and the BBS (r=–0.587, P<.001), TUG (r=0.474, P=.002), and TIN (r=–0.464, P=.002). B-A plots showed only few data points outside the predefined limits of agreement (LOA) when combining functional tests and results of LIN. Conclusions The digital app LIN has the potential to detect the risk of falling in older people. Further steps in establishing the validity of the LIN app should include its clinical applicability. This thesis focuses on the popularization on film of William Shakespeare’s plays. If the Shakespearean pictures shot in the silent era were more similar to filmed theatre performances than to adaptations, in the 1920s the Bard’s works start to be adapted for the cinema in order to reach a new and wider audience, thus triggering the phenomenon of the popularization of his plays. This process achieves a significant phase in the 1990s thanks to Kenneth Branagh’s adaptations, which cause a revival of the genre. Branagh’s experience is crucial in that he begins his artistic career in the theatre and afterwards takes on the role of actor and film director. I present two case-studies: In the Bleak Midwinter, directed by Branagh in 1995, and William Shakespeare’s Hamlet, directed and interpreted by Branagh in 1996. They both rework the tragedy of Hamlet but from different perspectives: the former appropriates the anxieties of the Shakespearean characters and transfers them to the members of an eccentric, contemporary theatre company, while the latter adapts the integral version of Shakespeare’s play for the screen. That is why In the Bleak Midwinter is defined as an “appropriation” of Hamlet whereas William Shakespeare’s Hamlet is defined as an “adaptation”. The relationship between the two screen versions is investigated. The analysis is based on the exploration of their intertextual connections and on the factors which in the two films contribute to the popularization of the source text. Not only does Branagh interpret the popularization of Shakespeare’s Hamlet as a compromise with Hollywood commercial values but he also includes a deep political awareness epitomized by the persistent presence of the Norwegian prince Fortinbras.

Esamina gli effetti del rapporto padre-figlia, ma anche i cambiamenti in corso nell'America al tempo di James e come essi hanno influito sulla nazione riguardo l'aspetto economico, sociale e psicologico. Malgrado non sia l'opera più famosa di James, Washington Square più avanti ha ispirato adattamenti teatrali, cinematografici e altri lavori di altri autori. Uno dei più famosi adattamenti di Washington Square è il film "L'Ereditiera" diretto dal celebre William Wyler nel 1949 e che riprende l'opera teatrale del 1947 scritta da Ruth e Augustus Goetz. La protagonista del romanzo, Catherine, è stata interpretata dalla brillante Olivia de Havilland che, per quel ruolo, vinse anche un Oscar come Miglior Attrice Protagonista.

Der Gen-Knockdown mittels RNAi hat sich als essentiell erwiesen, um Inhibitoren der induzierten Transdifferenzierung in C. elegans zu identifizieren (Tursun et al., 2011). Bakterienstämme, die dsRNA exprimieren, das die Expression spezifischer Gene mindert, können dem Wurm direkt zugefüttert werden, um einen genomweiten RNAi-screen der insgesamt 20.000 Gene in C. elegans durchzuführen. Allerdings werden die meisten biologischen Prozese durch mehr als ein Gen reguliert, was den Bedarf nach einer Methode generiert, die es erlaubt, zwei oder mehr Gene gleichzeitig herunter zu regulieren, um die Steuerung biologischer Prozesse studieren zu können. Die derzeitig vorhandenen Methoden liefern entweder nicht reproduzierbare Ergebnisse oder sind nicht skalierbar. Wir nutzen baktierelle Konjugation, die es durch ein konjugatives Plasmid ermöglicht Bakterienzellen zu generieren, die zwei verschiedene RNAi-Plasmide enthalten. Das Ziel war es, modifizierte RNAi-Donor-Plasmide mittels bakterieller Konjugation an eine Vielzahl anderer Bakterienzellen zu übertragen, die bereits ein anderes RNAi-Plasmid enthalten und dies dann im Hochdurchsatzverfahren durchführen zu können. Um Enhancer induzierter Expression von unc-25::gfp in der Keimbahn, ermöglicht durch den Knockdown des Histonchaperons LIN-53 (RbAp46/48 in Menschen), zu finden, wurden RNAi-Klone generiert, die gleichzeitig lin-53 als auch eines von insgesamt 800 verschiedenen Chromatin-bezogenen Gene herunter regulieren. Dabei identifizierten wir RBBP-5, Mitglied des Set1/ MLL-Methyltransferase-Komplexes, als neuen Barrierefaktor der induzierten Transdifferenzierung. RBBP-5 agiert dabei mutmaßlich parallel zu LIN-53. Doppelte RNAi, ermöglicht durch bakterielle Konjugation, erlaubt den simultanen Knockdown zweier oder mehr Gene, um genetische Interaktionen studieren zu können und erweitert damit die Einsatzmöglichkeiten von RNAi-Screens, um untereinander verbundene biologische Prozesse zu studieren.
The knock down of genes by RNAi has been fundamental to identify inhibitors of induced cell transdifferentiation in C. elegans (Tursun et al., 2011). Bacteria strains expressing dsRNA that target specific genes can be fed to the worm allowing straightforward whole-genome RNAi screens of the 20,000 genes in theC. elegans genome. However, many biological processes are regulated by more than one gene raising the need for simultaneous knock down of two or more genes to more fully interrogate the regulation of complex biological processes. Two approaches are currently available for double RNAi knockdown, − two bacteria strains expressing specific dsRNA can be mixed and grown together and fed simultaneously, which gives highly variable results. Alternatively, a new bacterial clone can be generated carrying a plasmid on which two RNAi targets of interest are 'stitched' together, which is not scalable. To address this challenge, we have developed a protocol using bacterial conjugation mediated by the 'Fertility Factor' (F) Episome in order to combine two different RNAi plasmids in a single bacterium. The objective was to be able to transfer a single RNAi plasmid to a large number of bacterial cells carrying different RNAi clones in one step in a high-throughput manner for large scale 'double' or even 'triple' RNAi screens. To find enhancers of induced unc-25::gfp expression in the germ line enabled by the depletion of histone chaperone LIN-53 (RbAp46/48 in humans), double RNAi clones targeting lin-53 and a total of 800 chromatin-related genes were generated and screened. We identified the Set1/MLL methyltransferase complex member RBBP-5 as a novel reprogramming barrier that putatively acts in a parallel pathway to LIN-53. Double RNAi by conjugation permits to reliably knock down two genes simultaneously in order to study genetic interactions at a genome-wide level, thus further increasing the versatility of RNAi screens to investigate interconnected biological processes.

This thesis analyses how four directors, Orson Welles, Akira Kurosawa, Roman Polanski and Justin Kurzel, have produced a screen adaptation of Shakespeare’s Macbeth. It starts with a general overview of the play, focusing on the characters and on Shakespeare’s sources and it attempts to reconstruct the general atmosphere in England in the 17th century. After a quick overview on the first stage adaptations in Chapter One, the thesis turns to analyse in detail the four movies. In Chapter Two it discusses Orson Welles’s adaptation of 1948 and his dreamlike representation of the story.The third chapter focuses on Japanese director Akira Kurosawa’s Throne of Blood, also known as The Castle of The Spider’s Web, produced in Japan in 1957.The fourth and fifth chapters are dedicated respectively to Roman Polanski’s 1971 adaptation and to the latest screen version made by Justin Kurzel in 2015. A conclusion, in the end, shows how all the considerations made on all the different screen versions contribute to re-invent and reproduce the greatness of Shakespeare’s characters and that the appeal his stories have on audience demands for artists to develop always new forms of engagement with Shakespeare’s work.

The present study aims at analyzing how dark humour as a cinematic genre travels cross-culturally through a specific mode of audiovisual translation, i.e. dubbing. In particular, it takes into consideration the processes involved in dubbing humour from English into Italian as observed in the English- and Italian-language versions of ten British and American dark comedies from the 1940s to the 2000s. In an attempt to identify some of the main mechanisms of the dark humour genre, the humorous content of the films was analyzed in terms of the elements on which specific scenes are based, mainly the non-verbal and verbal components. In the cases in which verbal elements were involved, i.e. the examples of verbally expressed humour, the analysis was concerned with whether they were adapted into Italian and to what effect. Quantification of the different kinds of dark humour revealed that in the sample of dark comedies verbal dark humour had a higher frequency (85.3%) than non-verbal dark humour (14.7%), which partially disconfirmed the first part of the research hypothesis. However, the significance of contextual elements in the conveying of dark humour, both in the form of Nsp VEH (54.31%) and V-V (V+VE) (21.68%), provided support for the hypothesis that, even when expressed verbally, dark humour is more closely linked to context-based rather than purely linguistic humour (4.9%). The second part of the analysis was concerned with an investigation of the strategies adopted for the translation of verbal dark humour elements from the SL (English) into the TL (Italian) through the filter of dubbing. Four translational strategies were identified as far as the rendering of verbal dark humour is concerned: i) complete omission; ii) weakening; iii) close rendering; and iv) increased effect. Complete omission was found to be the most common among these strategies, with 80.9% of dark humour examples being transposed in a way that kept the ST’s function substantially intact. Weakening of darkly humorous lines was applied in 12% of cases, whereas increased effect accounted for 4.6% and complete omission for 2.5%. The fact that for most examples of Nsp VEH (84.9%) and V-AC (V+VE) (91.4%) a close rendering effect was observed and that 12 out of 21 examples of V-AC (PL) (a combined 57%) were either omitted or weakened seemed to confirm, on the one hand, the complexity of the translation process required by cases of V-AC (PL) and V-AC (CS). On the other hand, as suggested in the second part of the research hypothesis, the data might be interpreted as indicating that lesser effort on the translator/adaptor’s part is involved in the adaptation of V-AC (Nsp VEH) and V-V (V+VE). The issue of the possible censorial intervention undergone by examples of verbal dark humour in the sample still remains unclear.

The present study aims at analyzing how dark humour as a cinematic genre travels cross-culturally through a specific mode of audiovisual translation, i.e. dubbing. In particular, it takes into consideration the processes involved in dubbing humour from English into Italian as observed in the English- and Italian-language versions of ten British and American dark comedies from the 1940s to the 2000s. In an attempt to identify some of the main mechanisms of the dark humour genre, the humorous content of the films was analyzed in terms of the elements on which specific scenes are based, mainly the non-verbal and verbal components. Lo scopo della tesi è quello di analizzare tutti gli elementi tipici del palcoscenico comico.. Il titolo è un gioco di parole e quella principale è Earnest, il cui significato non indica solo una persona onesta, ma è anche un nome di persona, Ernesto. Essere Earnest rende “la commedia per persone serie banale”, come suggerisce anche il sottotitolo dell’opera stessa. La trama è semplice ed è una farsa poiché soltanto alla fine capiamo chi è davvero Earnest mostrandoci qual è la vera identità che Wilde condanna all’interno della società Vittoriana. Il mio lavoro di tesi è diviso in tre capitoli: il primo riguardante la biografia di Oscar Wilde; il secondo basato sull’analisi dell’opera teatrale, includendo le sue numerose versioni, i personaggi e il modo in cui la società l’ha recepita —focalizzandoci in particolare sulla ragione per la quale essa è l’ultima opera teatrale e non, di Wilde e quali sono le conseguenze che ne sono scaturite; l’ultimo, invece, è sulle differenza tra la commedia e il cinema, con particolare riguardo a due adattamenti cinematografici: quello di Anthony Asquith del 1952 e quello di Oliver Parker del 2002.

The aim of this study is to investigate the cinematographic adaptation of the novel The Big Sleep by Raymond Chandler, and define whether the movie succeeded as a translation of the book or it failed as a flawed adaptation. The study will employ the adaptation theory and will try to explain the selection and re-construction of the scenes taken from the novel and transformed into celluloid. It will focus on Chandler’s values, traced in the book, and the consequent translation of them into the homonym movie. An historical introduction will describe the life of Raymond Chandler and the arising of the hard-boiled genre from the birth of crime fiction and detective novel, in order to obtain a better understanding of Chandler’s writing and his realization of the though private detective formula. In the second chapter, a preface will outline an overview regarding the intersemiotic translation along with the assumptions to classify novel-based films. It will be followed by the analysis of the novel in relation with the plot, the characters and the dialogues, and the way they are reflected into the movie. In conclusion, the dissertation tries to provide a contribution in the field of adaptation, offering a point of view on Chandler’s highest literary achievement and its adapting into a film noir. In the process, it considers screenwriters and director’s choices in recreating the novel, the type of translation that has been operated by them – if literal, traditional or radical – what is preserved or lost regarding Chandler’s core ideas, and whether the movie The Big Sleep does succeed as an adaptation and reaches some artistical value.

This thesis is aimed at analyzing ‘Strong Black Lead’ movement, a Netflix content production project started in 2018, as a form of Black Activism and resistance on screen. This visual activism is examined through its main actions of occupying screen and opposing mainstream narratives in USA. The latter, represents a pivotal role in reshaping the imagery associated to African American identity. This is achieved first through the humanization process of the characters presented and the emancipation of black identity from its mainstream depiction such as criminals and other perpetuated stereotyped versions of Blackness through Hollywood and mass media. The first section provides a background to the movement by analyzing its objectives and the correlation with traditional representation of Blacks. Additionally, it gives a brief insight to the socio-political from Barack Obama’s election to that of Donald Trump.It discusses as well the importance of being in charge of storytelling and why this shift has a political meaning. The second section focuses on the analysis of two Netflix series produced by ‘Strong Black Lead’: When They See Us by Ava DuVernay and Dear White People by Justin Simien.The first one is employed as an example of the humanization of Black identity while exposing the criminal justice system as a racial oppression force. While, the second series represents the challenges of emancipating Black identity from the associated stereotypes when unraveling racial discrimination.

The most important period of human being is during the child development. Consequently, natural environment will establsih the individual body, spirit and success in life. The original thought of this study is the initial and the first step basic considerations on the environmental aspects of the child development in the natural healing sciences.     This study was based on the co-operational studies between the local in Dalin Town and our hospital before. We tried to do the overall screen and research about the development conditions of the infants (< 1 year old) in Dalin Town during 2003, 07-2004, 07. The initial screen tests were Simplified Child Development Screening Test and the curve figure of infant body weight in Taiwan. The telephone visiting was undergone with every infants living in Dalin Town were rolled in.     This study was carried out on the infants at 4, 6, 9 and 12 months old. Thus, we can try to search the reduction for the underlying factors bothering the development of our infants. In the children with overweight potential, the incidence of total developmental retardation potential was higher than the overall children.(P=0.039) In addition, comparing with the incidence of non-ideal bodyweight gain, the children who were born by the mother coming from foreign were higher than the children born by local mothers.(P=0.045) However, in our studies, sex, breast-feeding, main caregiver, minor perinatal events did not show the dominant influence yet.     The new underlying problems bothering the development condition of the infants is presented in our study.

For remote communication, it is very hard to convey emotional message, or atmosphere of conversation to the opponent people who are located in a geographically difference place. For Email messages, emoticon, or face mark symbols are often used to express emotion, which works well to show emotion in an explicit way. Even though, the opponent face is show on the screen in remote video communication, emotional expression over the network is not similar to face-to-face meeting. In a face-to-face meeting, atmosphere of the conversation is shared in an implicit manner as opposed to the explicit explanation using words, sign, or pictures. This research proposes an idea of implicit representation of emotional messages using light illumination [Lin, T.I. 2004; Vandewalle, G. 2010] with 1/f-fluctuation. It is reported that the 1/f-fluctuation makes a relaxed mental state. Therefore, ambient lighting with 1/f-fluctuation could provide a comfortable (and/or uncomfortable) atmosphere. In addition to this, this study aims to present emotional message in the illumination. In order to do so, four different types of 1/f-fluctuation pattern were created to represent four different types of typical human emotion, including joy, anger, pathos, and humor. The paper explains how the idea was implemented, using light illumination of four types of emotion based on the user experiments. According to some feasibility experiments, these four types of illumination were recognized to express each type of emotion. Applying emotional message representation with 1/f-fluctuation in the illumination of video conference speaker, implicit emotional message was presented during the conversation. Then the paper discusses the feasibility of the idea for ambient human interface.
