Résumé :
On the moment, there is a lack of available clinical personality assessment instruments in Latvia, so in the frame of the National Research Programme (2014-2017, project No. 5.8.2.) a new multi-item multi-scale self-report measure - the Latvian Clinical Personality Inventory (LCPI) is currently being developed. LCPI structure includes three parts: clinical scales, pathological personality scales and functioning scales and will provide a possibility to perform a combined analysis of mental disorders symptoms and functioning aspects using one clinical personality inventory. The main aim of this article to describe procedures and stages of the development of the preliminary version of the LCPI v2.2. item pool.

Background and Aim: Cubitus varus deformity is the most common late complication aftersupracondylar fracture of the distal humerus in children, incidence varying from 4% to 58%. Thepresent study was done to evaluate the results of dome osteotomy. Material and Methods: Thisprospective study was conducted in a tertiary care hospital over 16 months. In all patients humerus-elbow wrist angle was measured on both sides and the correction needed was calculated. The lateralcondyle prominence index (LCPI) was calculated by anteroposterior view radiographs of thedeformed and the normal elbow in full extension by (AB-BC)/AC. Dome osteotomy with para tricepsapproach was used. Pre and post-operative carrying angle of elbow, range of motion and lateralprominence indices were compared. Results: The age of patients ranged from 3 to 15 years with amean age of 8.47±3.14 years. Preoperative carrying angle of normal side ranged from 80 to 140and that of effected side ranged from -23 to -13 and the difference was statistically significant(p<0.05). LCPI ranged from -8.4 to 5.9%. The majority of cases had LCPI >2.7%. As compared to,an improvement in carrying angle at defect side was observed to be 28.41±2.15 which wassignificant (p<0.05). At baseline mean LCPI was 0.39±3.87% which changed to -0.86±3.47%, themean change of this was significant (p=0.01). baseline Conclusion: Dome osteotomy is a relativelytechnically demanding technique for correction of cubitus varus deformity but with a betterfunctional outcome without being associated with lateral condyle prominence.

As the world’s top energy consumer and carbon emitter, China’s carbon emissions policies, including the low-carbon pilot initiative (LCPI) implemented in July 2010, have important effects on global climate change. Therefore, accurately assessing the effect of this policy has become extremely important for low-carbon development. This article analyses the impact of implementing LCPI on regional carbon emissions by using Guangdong Province as the study area, which has the largest economic scale, population size and carbon emissions amongst China’s low-carbon pilot provinces. The results suggest that for the entire 2010–2015 period, Guangdong’s carbon emissions were reduced by about 10% due to the implementation of LCPI. This policy produced a significant impact on the carbon emissions from manufacturing industries but showed minimal impact on the carbon emissions from energy production. Unlike previous researchers who relied on estimations, the authors of this work obtained unified carbon emissions data for 1997–2015 from the China Emission Accounts and Datasets and then constructed comparison groups by using the synthetic control method instead of performing a subjective selection. The authors also examined the impact of LCPI on carbon emissions from different sources. This article proposes that policy support and low-carbon action are necessary for reducing regional carbon emissions and that the policies must be constantly adjusted during their implementation. The successful experiences in low-carbon pilots are also worth exploring and promoting in other regions.

A 36-kb genomic DNA segment of the Drosophila melanogaster genome containing 12 clustered cuticle genes has been mapped and partially sequenced. The cluster maps at 65A 5-6 on the left arm of the third chromosome, in agreement with the previously determined location of a putative cluster encompassing the genes for the third instar larval cuticle proteins LCP5, LCP6 and LCP8. This cluster is the largest cuticle gene cluster discovered to date and shows a number of surprising features that explain in part the genetic complexity of the LCP5, LCP6 and LCP8 loci. The genes encoding LCP5 and LCP8 are multiple copy genes and the presence of extensive similarity in their coding regions gives the first evidence for gene conversion in cuticle genes. In addition, five genes in the cluster are intronless. Four of these five have arisen by retroposition. The other genes in the cluster have a single intron located at an unusual location for insect cuticle genes.

Objective: To assess the influence of reduction of crude protein (CP) by 5% and 10% with supplementation of crystalline synthetic essential amino acids (EAAs) on performance and carcass traits of broilers. Design: Randomized controlled study Animals: A total of 320 one-day-old broiler chicks, (Cobb 500) reared up to 35 days of age were randomly allocated into eight experimental groups with four replicates (10 each). Procedures: The treatments as follow; the first group of chicks were fed on control protein to meet the CP requirement and total AAs according to Cobb 500 recommendation; the second group were fed idle EAAs (Id EAAs) to meet the recommended CP and the digestible EAAs. The third, fourth, and fifth groups were fed on diet 5% reduction of CP with EAAs supplementation exceed the digestible EAAs requirement of Cobb by 110, 120, and 130% (LCP3+110EAAs, LCP4+120EAAs, LCP5+130EAAs). However, the sixth, seventh, eighth groups of chicks were fed on diet 10% reduction of CP with AAs supplementation exceed the digestible EAAs requirement of Cobb by 110, 120, and 130% (LCP6+110EAAs, LCP7+120EAAs, LCP8+130EAAs). Results: Broilers of group four, five, six, seven and eight showed a significant decrease final body weight (BW), body weight gain (BWG) and feed intake at 21 and 35 days of age. However, the same variables were significantly enhanced in broilers of group one, two, and three (p <0.05). Additionally, there was a significant decrease of relative weight of abdominal fat pad of broilers in groups three, four, five compared to other treatment groups. Serum uric acid and cholesterol were linearly decreased with reduction of CP and supplementation with synthetic essential amino acids. Conclusion and clinical relevance: The present results indicate that reduction of CP by 5% or 10% with crystalline EAAs supplementation by 110, 120 and 130% more than the digestible EAAs of broiler recommendation did not support the growth performance.

Introduction: Late chronic periprosthetic infections (LCPIs) are worrisome complications of primary hip arthroplasties. The gold standard procedure is the 2-stage revision. 1-stage exchange is gaining popularity in order to reduce the invasivity of the former technique. A partial 2-stage exchange technique, retaining fixed components, may overcome some of the drawbacks of the previous techniques, allowing a much easier reconstruction step. Methods: 28 patients with a LCPI after a primary total hip arthroplasty underwent a first removal stage: the loosened component was removed (23 cups and 5 stems) and the fixed component, with no local signs of infection, was retained. An antibiotic hand molded spacer was positioned in 16 cases. After a mean time of 8 months and a tailored antimicrobial therapy, the spacer was removed and the implant was revised. Results: The mean follow-up was 5 years. The HHS score was 82.7. 4 cases failed (2 patients presenting a septic relapse after revision and 2 patients undergoing Girdlestone arthroplasty), achieving a survival rate of 83.4% at 5 years. 2 patients were unwilling to perform a further procedure and did not proceed to the second stage. All the other patients had no clinical, radiological, laboratory signs of septic relapse. Conclusions: The partial 2-stage approach seems a promising technique for LCPI in selected cases, with good infection control. It allows an easier revision by sparing the fixed components. Larger case series and longer follow-ups are needed to confirm the results and identify the limits of this approach.

The purpose of this article is to present a theoretical integrative model, which reflects contemporary tendencies in the understanding of mental disorders and functional impairment, and which is used as a theoretical frame for the development of the Latvian Clinical Personality Inventory (LCPI). This article, based on the latest research findings in the field, supports the necessity of a combined analysis of mental disorders and functional impairment. Due to scientific findings and deeper understanding of the interrelation between mental disorders and functioning impairment, it has become possible to develop psychological instruments for valid assessment of the disturbances in an individual's cognition, emotion regulation, and behaviour combining with analysis of most essential and relevant aspects of their functioning. An integrative theoretical model of LCPI, developed on the selected criteria from Diagnostic and Statistical Manual of Mental Disorders (DSM – 5; APA, 2013) and WHO International Classification of Functioning, Disability and Health (ICF; WHO, 2001/2015) is presented in the article. Conducted literature analysis permits to conclude, that in a context of clinical personality assessment, a combined analysis of symptoms of mental disorders and relevant functioning criteria is very promising and will be useful in many assessment contexts. Based on such an integrative approach the Latvian Clinical Personality Inventory is currently being developed. This project is a part of the National Research Programme (No. 5.8.2.).

The energy loss of heavy quarks traversing color dense matter is calculated with an analytical formula derived within the light-cone path integral (LCPI) approach. We find that the energy loss mechanism is dominated by gluon radiation and induces a suppression pattern of charm quark different from the suppression of light quarks. We find also that this radiative energy-loss is proportional to L2 for energetic quarks, but changes to a L dependance with decreasing quark energy, where L is the length of the traversed medium.

The main aim of this study was to evaluate the criterion validity and to estimate the cut-off score of the Depression scale (DS) and short Depression scale (DSs) for a new self-report measure – Latvian Clinical Personality Inventory (LCPI). Usefulness of DS and DSs for identifying patients with major depression were analysed based on psychometric analysis of data acquired from psychiatric inpatient sample with depressive disorder (n = 37) in comparison to randomised stratified community subsample (n = 176) selected from the overall test development sample (N = 888). The present study was carried within the framework of the National Research Program (BIOMEDICINE) 2014 – 2017 (sub-project Nr.5.8.2.). It was shown that all 24 item of DS show good to excellent discrimination power. Cronbach’s alpha was 0.97 for DS and 0.95 for DSs in test development sample. For DS, the optimal cut-off score was 26 points (sensitivity 95%, specificity 91%, and positive predicted value of 69%). For DSs, the optimal cut-off was 12 points (sensitivity 92%, specificity 89%, and positive predicted value 63%). DS and DSs of LCPI is proved to have good criterion validity in detecting depression and to be a reliable and valid instrument for assessment of depression symptoms in patients with depression and in general population. Subjects scoring at least 26 on DS or 12 points on DSs constitute a target group for further diagnostic assessment in order to determine appropriate treatment.

Abstract The aim of this study was to evaluate the utility of the pathological personality traits in predicting suicidal ideation, especially in combination with other risk factors, such as the level of depression, prior attempts of suicide, low self-esteem, low level of perceived social support and self-esteem-by-social support interaction, both in general and in psychiatric inpatient samples. Data were analysed within two samples: non-clinical general sample (n = 461) and psychiatric inpatient sample (n = 131). Latvian Clinical Personality Inventory (LCPI) was used as the instrument for data collection. LCPI is a comprehensive multi-scale multi-item inventory, which consists of nine clinical scales (including Depression Symptom Scale), 33 pathological personality scales, five functioning scales, and five additional scales, including a Suicidal Ideation Scale, Low Self-esteem Scale and Lack of Perceived Social Support Scale. Results of the hierarchical regression analysis showed that several facet-level pathological personality traits (depressivity, self-harm, dissociation proneness, submissiveness, and suspiciousness) added significant incremental variance to the prediction of suicidal ideation above and beyond the well-known main risk factors of suicidal ideation, such as depression and prior suicide attempt. This effect remained stable even after taking into account additional interpersonal risk factors, such as low self-esteem, low level of perceived social support and self-esteem-by-perceived social support interaction. The incremental effect of personality traits was medium in the psychiatric inpatient sample and small in the general sample. Findings of the study may assist in early screening for persons with suicide risk and for developing prevention programmes in different settings.

Die Arbeit behandelt ausgewählte Aspekte der Ausbildung strömungsinduzierter Orientierungen und deren Auswirkung auf die mechanischen Eigenschaften in spritzgegossenen LCP-Formteilen. Es werden die Grundlagen zum rheologischen Verhalten von LCP, zum Füllvorgang und zur Orientierungsausbildung beim Spritzgießen sowie zu den Methoden der Orientierungsuntersuchung zusammengefaßt. Rheologische Untersuchungen werden aus der Sicht der praktischen Kennwertermittlung durchgeführt. Dabei wird die Abhängigkeit der Viskosität von der Düsenlänge und von der thermo-rheologischen Vorgeschichte analysiert sowie eine Viskosimeter-Vorrichtung für die Spritzgießmaschine vorgestellt und erprobt. Die umfangreiche Analyse verarbeitungsinduzierter Formteileigenschaften behandelt schwerpunktmäßig Orientierungsphänomene in der Wanddickenrichtung. Die Orientierungseffekte werden durch Grauwertmessungen erfaßt. Ein Meßaufbau mit einer kalibrierten Mikroskop-Digitalkamera-Kombination ermöglicht die reproduzierbare, effektive Messung der Grauwerte in hoher Ortsauflösung und damit eine über die bisher bekannten qualitativen Vergleiche hinausgehende quantitative Auswertung. Anhand der Grauwertverläufe wird der Einfluss technologischer und konstruktiver Parameter auf die Orientierungsstruktur dargelegt und mit Ergebnissen der Computersimulation verglichen. Durch parallele Messung anisotroper mechanischer Kennwerte und Orientierungsmerkmale werden richtungsabhängige Struktur-Eigenschafts-Beziehungen bestimmt.

Syftet med denna studie var att se om en strukturerad vårdform vid livets slut såsom Liverpool Care Pathway for the dying, LCP, har någon inverkan på omvårdnaden av de palliativa patienterna jämfört med ordinarie palliativ vård samt att beskriva vårdpersonalens upplevelser av LCP. Studien genomfördes som en systematisk litteraturstudie där underlag inhämtades genom sökning av veteskapliga artiklar i databaser samt att artiklar söktes manuellt på Högskolan Dalarnas bibliotek. Artiklar har även sökts från referenser på andra artiklar direkt på titeln och vid sökningarna användes sökorden både enskilt och i kombination varvid 10 artiklar valdes ut till resultatet. Resultatet visade att efter införandet av LCP förbättrades dokumentationen, patienterna hade inte onödiga livsförlängande behandlingar. Symtomen i form av smärta, oro/ångest och slemproduktion var mindre, samt att kommunikationen mellan vårdpersonal, närstående och patient förbättrades. Sjuksköterskorna upplevde att tiden för dokumentation förkortades och mer tid kunde läggas på patienterna, sjuksköterskor ansåg också att den gamla rutinmässiga behandlingen och omvårdnaden förbättrades. LCP dokumenten gjorde även att anhöriga kunde följa omvårdnaden vilket gjorde dem tryggare i vad som skedde med deras närstående och vad som hände i vården runt denne.

La presente investigación tiene como objeto el análisis de la protección dispensada a los deudores y los demás sujetos pasivos de la relación jurídica crediticia por la Ley 5/2019, de 15 de marzo, de Contratos de Crédito Inmobiliario. Aprobada con la finalidad de transponer la Directiva comunitaria 2014/17, de 4 de febrero, esta Ley supera este objetivo inicial para introducir en el Ordenamiento jurídico un conjunto de obligaciones para el acreedor y unas limitaciones de carácter imperativo en las cláusulas de los contratos concertados con las personas físicas que recaigan sobre bienes inmuebles de uso residencial y a los consumidores que destinen el crédito a adquirir o conservar derechos de propiedad sobre inmuebles o terrenos construidos o por construir. Este análisis se realiza a través del estudio del ámbito de aplicación de la Ley en sus cuatro aspectos: subjetivo, objetivo, espacial y temporal, precedido en los dos primeros casos por una breve exposición de la forma en que la Directiva y otros Ordenamientos jurídicos de los países de la Unión Europea aborda estos aspectos, junto con los pronunciamientos del Tribunal de Justicia de la Unión Europea.
This paper analyzes the protection of people who celebrate credit agreements by the Law 5/2019, of 15 March 2019, regulating real estate credit contracts, that transposes into our legal system the Directive 2014/17, EU of the European Parliament and of the of 4 February 2014, on credit agreements for consumers relating to residential immovable property. The exposition starts with the main aspects of european regulation and the jurisprudence of the Judgment of the Court regarding to the delimitation and scope of consumer protection in credit agreements, the subjective scope, from the active and passive point of view and doubtful cases not regulated. The same analysis is carried out in relation to the object of protection and its scope in Chapter II. The last Chapter ends with the problems motivated by contracts with a foreign element and by the concurrence of territorial provisions and temporal delimitation of the LCCI. Finally, some conclusions are presented as possible solutions to the problems generated by the new legal regime implemented by the LCCI.

Syftet med denna uppsats är att undersöka hur undersköterskor i fyra olika verksamheter upplever att deras arbetssituation har förändrats sedan man införde LCP (Liverpool Care Pathway). Vidare belyses hur undersköterskorna menar att samarbete, kommunikation och relation har påverkats och på vilket sätt och i så fall hur. Uppsatsen är skriven utifrån samverkan och ett professionellt förhållningssätt i socialt arbete. Ett annat perspektiv som uppsatsen belyser är på vilket sätt metoden blev introducerad i organisationen, detta utifrån implementeringsprocesser och kommunikationsteorin. Resultatet visar att undersköterskorna upplever en större delaktighet och trygghet i deras arbete idag jämfört med tidigare. Vidare har samarbetet med andra samverkanspartners blivit mycket bättre sedan LCP- metoden infördes. Med hjälp av metoden känner undersköterskorna en ökad trygghet, delaktighet, klarhet, tydlighet och öppenhet. Detta genom de checklistor som LCP- metoden bygger på. Det framkom i uppsatsen att undersköterskorna känner att de kan prata mer öppet om döden och döendet med anhöriga efter införandet av LCP - metoden .Nyckelord: Kommunikation,
The purpose of this essay is to explore how staff nurses in four different businesses feel that their work situation has changed since the introduction of LCP (Liverpool Care Pathway). Further highlights is how the nurse believes that cooperation, communication and relationships have been affected, in what way and if so how. The essay is written from collaboration and a professional approach in social work. Another perspective the essay illustrates is the way in which the method was introduced in the organization, this according to implementation processes and communication theory. The result shows that the staff nurses experience a greater involvement and confidence in their work than before. Furthermore has the cooperating with other collaboration partners turn out to be much better since the LCP-method was introduced. By using the method the staff nurses feel an increased security, participation, transparency, clarity and transparency. The LCP-method with its checklists contributes to these feelings. It appeared in the essay that the staff nurses feel they can talk more openly about death and dying with relatives after the introduction of the LCP-method.

Introduktion: Liverpool Care Pathway (LCP) är en vårdplan framtagen för palliativ vård vars syfte är att överföra det bästa av hospicevården till den övriga sjukvården. LCP ger vägledning för vårdpersonal i vården av patienter i livets slutskede för att säkerställa god vård för patient och närstående. Syfte: Syftet var att beskriva hur sjuksköterskan upplever användandet av vårdplanen Liverpool Care Pathway (LCP) i livet slutskede. Metod: Metoden som användes var litteraturstudie. Sökningar gjordes i CINAHL och PubMed där elva vetenskapliga artiklar, som svarade på studiens syfte, framkom och granskades i sin helhet. Resultat: Resultatet visade att LCP förbättrade kommunikationen, ökade kunskap och självförtroende, ökade delaktigheten för närstående i vården, förbättrade arbetsmiljön och ökade patientsäkerheten. Dock sågs en risk med att använda LCP som en standardiserad vårdplan vilket kunde medföra att patient och närstående kunde känna sig negligerade. Slutsats: Sjuksköterskans upplevelse av användandet av LCP har i litteraturstudien beskrivits positiv både för sjuksköterskan och annan vårdpersonal. Användandet av LCP har även resulterat i förbättrad vård för patient och närstående.

Ce travail a pour objectif la mise au point d'un ensemble d'outils de modélisation directe et inverse en utilisant le code d'éléments finis CESAR-LCPC. Ces outils sont adaptés aux données électriques collectées sur des structures 3D à géométrie complexe. Pour le problème direct, un programme utilitaire servant d'interface avec le solveur CESAR a été créé afin de modéliser des séquences de mesures électriques (tomographies). Afin de pouvoir inverser un nombre important de données sur des modèles de grandes dimensions, une fonction objectif est minimisée en utilisant la technique de l'état adjoint. Cette approche est originale car elle vise le calcul direct de la variation à apporter aux paramètres du modèle, sans évaluation explicite de la matrice de sensibilité. Des données synthétiques ont été utilisées pour valider cet algorithme d'inversion. La fiabilité des modèles inversés est testée en utilisant un méthode de calcul de l'indice ROI (Region Of Investigation)
This work presents the adaptation and the use of the CESAR-LCPC finite element code for the forward and inverse modelling of 3D resistivity data. A forward modelling tool was created to simulate tomographies with CESAR and an inversion code was also presented for the processing of resistivity tomographies on complex 3D structures using any electrode arrangement. This algorithm is well suited for the processing of large data sets with a lot of unknown model parameters. The inversion code uses an original strategy to avoid the explicit calculation of a sensitivity matrix. The adjoint-state of the potential field is used to minimize an objective function for the electrical inverse problem. Finally, a ROI (Region Of Investigation) index method is used to assess whether features in the model are caused by the data or are artefacts of the inversion process

En el primer trabajo de esta tesis se dilucida el camino de plegamiento oxidativo del LCI. El LCI reducido y desnaturalizado se repliega a través de un flujo secuencial y rápido de intermediarios de uno y dos puentes disulfuro, y llega a una etapa limitante en la cual la mezcla de tres especies mayoritarias que contienen 3 puentes disulfuro y una población heterogénea de isómeros de cuatro puentes disulfuro no nativos (scrambled) coexisten. Los intermediarios de tres puentes disulfuro se han identificado como trampas cinéticas que se hallan a lo largo del camino de plegamiento del LCI, y se han caracterizado sus estructuras: dos de ellos contienen sólo puentes disulfuro nativos. El camino de plegamiento del LCI comprende propiedades exhibidas tanto por el BPTI como por la hirudina, dos modelos divergentes con características de plegamiento opuestas. Los resultados obtenidos en el estudio de las vías de plegamiento del LCI confirman el enorme espectro de diversidad de los caminos de plegamiento de las proteínas.
En el segundo trabajo de esta tesis se dilucidan las curvas de desplegamiento y desnaturalización del LCI. En presencia de un agente iniciador de la formación de tioles y un agente desnaturalizante se despliega el LCI nativo por el intercambio de sus puentes disulfuro, y la molécula se transforma en una mezcla de especies scrambled. Podemos distinguir claramente y aislar, entre los 104 posibles isómeros scrambled del LCI, a nueve de ellos que representan el 90% del total del LCI desplegado. La concentración de tiocianato de guanidinio e hidrocloruro de guanidinio requeridas para alcanzar el 50% de la desnaturalización son de 2,4 y 3,6 M, respectivamente. El LCI nativo es resistente a la desnaturalización por urea, incluso a concentraciones elevadas (8M). El camino de desplegamiento del LCI se ha podido definir en base a la evolución de la concentración relativa de los isómeros scrambled de la molécula a lo largo de su desnaturalización.
Existen dos poblaciones de especies scrambled que sufren variaciones a lo largo del camino de desplegamiento. Una población se acumula como intermediarios bajo condiciones desnaturalizantes fuertes (incluye al isómero denominado beads-form). La otra población muestra una correlación inversa entre su abundancia relativa y las condiciones desnaturalizantes y debería poseer otro tipo de estructuras no nativas. Los resultados que se presentan en el segundo trabajo muestran que el LCI, una molécula con potenciales aplicaciones biotecnológicas, tiene cinéticas bajas de desplegamiento y es altamente estable.
En el tercer trabajo de esta tesis se estudia el efecto del LCI en la fibrinólisis in vitro. Se ha descrito, tanto por estudios in vitro como in vivo, que el PCI (potato carboxypeptidase inhibitor) incrementa la tasa de lisis de los coágulos de fibrina. Con este mismo propósito, hemos evaluado y comparado la capacidad profibrinolítica del LCI con la del PCI. Todos los resultados han sido obtenidos en ensayos con plasma humano, en un sistema in vitro.
Hemos demostrado que ambos inhibidores, el LCI y el PCI, aceleran de forma substancial la lisis de los coágulos tratados con t-PA, esta aceleración es dependiente de la concentración de inhibidor. El LCI se ha mostrado unas 10 veces más potente que el PCI como acelerador de la fibrinólisis, y también muestra una mayor capacidad inhibidora de TAFI al realizar ensayos de actividad enzimática en plasma.
Se ha estudiado el posible efecto de estos dos inhibidores sobre la estructura del coágulo de fibrina mediante microscopía electrónica de transmisión y de barrido. Se ha observado un incremento significativo en el grosor de las fibras y una disminución simultánea en la densidad de su entrecruzamiento: el LCI puede tener una aplicación en las terapias trombolíticas basadas en t-PA.
The pathway of oxidative folding of LCI has been elucidated in the present work, using structural and kinetic analysis of the folding intermediates trapped by acid quenching. Reduced and denatured LCI refolds through a rapid, sequential flow of 1- and 2-disulfide intermediates and reaches a rate limiting step in which a mixture of three major 3-disulfide species and a heterogeneous population of non-native 4-disulfide (scrambled) isomers co-exist. The three 3-disulfide intermediates have been identified as major kinetic traps along the folding pathway of LCI, and their disulfide structures have been elucidated in this work. Two of them contain only native disulfide pairings, and one contains one native and two non-native disulfide bonds. The folding pathway of LCI features properties exhibited by both bovine pancreatic trypsin inhibitor and hirudin, two diverse models with extreme folding characteristics. The results further demonstrate the large diversity of disulfide folding pathways.
The unfolding and denaturation curves of leech carboxypeptidase inhibitor (LCI) were elucidated using the technique of disulfide scrambling. In the presence of thiol initiator and denaturant, the native LCI denatures by shuffling its native disulfide bonds and transforms into a mixture of scrambled species. Nine out of 104 possible scrambled isomers of LCI, amounting to 90% of total denatured LCI, can be distinguished. The denaturation curve, shows that the concentration of guanidine thiocyanate and guanidine hydrochloride required to reach 50% of denaturation is 2.4M and 3.6M, respectively. Native LCI is resistant to urea denaturation, even at high concentration (8M). The LCI unfolding pathway was defined based on the evolution of the relative concentration of scrambled isoforms of LCI upon denaturation. Two populations of scrambled species suffer variations along the unfolding pathway. One accumulates as intermediates under strong denaturating conditions, and corresponds to open or relaxed structures, one of which turned out to be the bead-form isomer. The other population shows an inverse correlation between their relative abundances and the denaturing conditions, and should have another kind of non-native structure, more compact than the unfolded state. The rate constants of unfolding of LCI are low when compared to other disulfide containing proteins. The results presented in this study show unequivocally the high stability and low kinetics of unfolding of the LCI fold, that guarantee the appropriate industrial and therapeutical handling of the molecule.
The potato carboxypeptidase inhibitor (PCI) has recently been reported to enhance the rate of fibrin clot lysis induced by tissue plasminogen activator (t-PA). Here, we have studied the fibrinolytic activity of LCI (leech carboxypeptidase inhibitor), and compared its effectiveness with that of PCI in vitro. Both LCI and PCI markedly accelerate the t-PA-induced lysis of human plasma clots in a concentration-dependent manner. Besides being approximately 10-fold more potent in accelerating fibrinolysis, LCI also showed a greater TAFIa inhibitory activity than PCI in enzymatic assays. The effects of both TAFIa inhibitors on the fibrin clot structure were compared by scanning and transmission electron microscopy. A significant increase in fiber thickness and a simultaneous decrease in the density of fiber entanglement were observed when clotting of human plasma was induced in the presence of either LCI or PCI. These alterations of clot structure correlated with the increased lysis rate determined in turbidimetric assays. In inhibitor-treated samples, formation of large pores within the fibrin gel occurs earlier than in controls, supporting the role of LCI as enhancer of clot lysis. These results suggest that LCI may be useful to improve the t-PA based therapy of thrombosis.

Microstructure-property relationship is being examined in a polymer matrix composite system consisting of vapor grown carbon nanofibers (VGCF) mixed in a thermotropic liquid crystalline polymer (LCP) matrix. These nanocomposites show an inherent hierarchical structuring, which we hope to utilize in the development of multifunctional structure-conduction composites with improved performance. Among unfilled polymers, extruded LCPs show relatively high strength and high stiffness that have been attributed in the literature to the preferential molecular alignment along the extrusion direction and the hierarchical nature of LCPs. Further, as is typical for polymers, LCPs have poor thermal and electrical conductivity relative to metals. By contrast, carbon nanofibers are known to possess high strength, high stiffness and high conductivity in the axial direction. It is expected that the combination of the extrusion process and the similarity of the length-scales of LCP fibrils and carbon nanofibers will lead to improved axial alignment of both phases within the nanocomposite filaments. This simultaneous alignment of the LCP matrix and that of the carbon nanofibers is expected to lead to interesting mechanical and conductive behavior in the nanocomposite filaments through hierarchical interactions at the nanometer to micrometer scale levels. Carbon nanofibers, 60-150 nm in diameter, were mixed with Vectra A950 LCP and the mixture was extruded as 0.5–2 mm diameter filaments. Nanocomposite filaments with 1%, 2%, 5% and 10 wt.% VGCF were characterized via tensile testing and fractography. The tensile modulus, failure strength and strain-to-failure were found to be sensitive to filament diameter, carbon nanofiber content and extrusion process. There was a noticeable increase in mechanical performance with decreasing filament diameter irrespective of carbon nanofiber content. Fracture surfaces showed hierarchical features from nanometer to micrometer scale and processing defects in the form of voids. The results of this research will be used to fabricate composite components that exploit structural hierarchy from nano-to macro-scale.

Abstract Liquid crystal polymers (LCPs) form a class of high-performance plastics that exhibit comparable mechanical, chemical, and electrical characteristics to engineering metals and ceramics arising from their mesoscopic ordering. The unique hierarchal LCP microstructure leads to anisotropic bulk behavior and an understanding of the development of this morphology during manufacturing, as well as the subsequent effect on polymer properties, is essential to the design of isotropic material manufacturing processes. In this investigation, the preferred orientation in injection molded LCP plaque samples was measured using wide-angle x-ray scattering (WAXS). The direction of preferred alignment was observed from the WAXS scattering patterns and the degree of orientation in the material was quantified using an anisotropy factor. In addition, the mechanical, thermal, and dielectric bulk behavior was measured with respect to the mold direction (MD) and transverse direction (TD). To investigate the effects of processing geometry on microstructural development, and the resulting macroscopic properties, plaques of three different thicknesses were analyzed. In addition, the influence of melt rheology was probed through the comparison of two different commercial LCP resins. It is shown that a strong correlation exists between material performance and both the bulk polymer texture and the individual regimes of the hierarchal structure. The effects of processing geometry and polymer rheology also demonstrate the structure-property-processing dynamics at work in injection molded LCPs.

It is known that liquid crystalline polymer (LCP) melts have a high elasticity which can be measured from its effect on the rheology on the cessation of shear. On the other hand, LCPs show very limited die swell after extrusion. In this paper, the results of experimental measurements of the die swell for a liquid crystalline material and polypropylene (PP), an amorphous polymer, are presented. The extrudate thickness 5 cm below the die lip is optically measured and the results are analyzed using ImageJ software. A numerical simulation of the die swell based on the capillary rheometry data and oscillatory rheometry is performed for LCP materials using ANSYS ® POLYFLOW ®. Different viscoelastic properties are used to model the LCP and optimum properties to model the die swell for the base volume flow rate are determined. Results show similarity between die swell modeling for the LCP at the base volume flow rate but increasing the die swell results in some deviation from the experimental results.

The dynamics of multibody systems with many contacts are frequently formulated as a Linear Complementarity Problem (LCP), for which several direct or iterative algorithms are available to solve it efficiently. These formulations rely on discretized friction models that approximate the friction cone of the Coulomb model to a pyramid. However, they produce rank-deficient LCPs even though the physical problem does not have constraint redundancy and has a unique solution. Here, a new discretized friction model is presented which results in an LCP formulation with a full-rank lead matrix. This model relies on an inertial term to couple the equations of the model, which behaves as close to the Coulomb model as the other discretized models. Moreover, it is shown through some simulations that some algorithms can be used with this formulation, which could not be used with the other rank-deficient LCP formulations.

Carbon nanofibers, such as single walled carbon nanotubes (SWNT), multiwalled carbon nanotubes (MWNT) and vapor-grown carbon nanofibers (VGCF or VGCNF) are routinely compounded with polymers to create thermally and electrically conductive polymer nanocomposites. Our group is interested in combining the conduction with structural functionality by reinforcing a high-performance thermotropic liquid crystal polymer (LCP) matrix with vapor-grown carbon nanofibers and single walled carbon nanotubes. High strength and stiffness can be achieved in LCPs through the alignment of molecular domains during high-shear mixing and extrusion. Further strength and stiffness enhancements are potentially possible if the carbon nanofibers could also be aligned, perhaps, with the assistance of the aligned domains of the LCP matrix. However, the geometrical structure of VGCF is quite different and the diameter is one to two orders of magnitude larger than that of SWNT. Therefore, the processing conditions and the interactions between the LCP domains and the nanofibers are expected to lead to different dispersion and alignment characteristics of VGCF and SWNT within the LCP matrix. In this work, twin-screw and Maxwell-type mixer-extruders were used to produce neat LCP filaments and LCP-nanofiber composite filaments with various concentrations of VGCF and SWNT. The dispersion and orientation of the VGCF and SWNT reinforcements were determined by X-ray diffraction and electron microscopy. The filaments were loaded in quasi-static uniaxial tension until fracture to determine the tensile modulus, strength and strain-to-failure. The mechanical properties showed a strong dependence on the filament diameter, nanofiber concentration and processing parameters. A significant increase in mechanical performance was observed with decreasing filament diameter irrespective of the carbon nanofiber concentration. Fracture surfaces examined under electron microscopy revealed hierarchical features at multiple length scales. At the macroscopic scale, a skin-core configuration was observed in the filament cross-section with the skin possessing a greater degree of LCP molecular alignment and nanofiber alignment than the core. The mechanical and electrical properties of the LCP, LCP-VGCF and LCP-SWNT nanocomposite filaments will be described and related to processing parameters, the type of carbon nanofibers (VGCF or SWNT) and the resulting composite microstructure.

This paper presents a new molecular model to define the interactions of a liquid crystalline polymer (LCP) flowing between two serrated walls. The wall is modeled by a rough atomic serrated wall. The roughness characteristics are given by the space and height of the serrated wall. Molecular model of the liquid crystalline polymer is described by an improved model that consists of GB (Gay-Berne) sites as rigid segments and LJ (Lennard-Jones) sites. There are two nonlinear springs each connecting from a GB site to a LJ site that situate between two GB sites as flexible segments. This improved model is newly developed to reduce the computational cost from that of the hybrid GB/LJ model, which has provided an effective way to investigate the boundary problems and flowing behaviors of LCPs at nano-scale. The molecular dynamics (MD) simulation using this reduced computational cost method to study the effect of boundary conditions on alignment and rheological properties of the LCP is shown in the result.

Liquid crystalline polymers (LCPs) are among a high-performance class of materials, which derive unique mechanical, chemical, and electrical characteristics from their long-range molecular order. The evolution of anisotropic orientation in the LCP microstructure during processing, however, can adversely affect the macroscopic polymer behavior. Simulation of this anisotropy is crucial to the design of manufacturing processes producing the desired material properties, and the ability to quantify the polymer directionality is a necessary metric of the model. Using a Monte-Carlo approach introduced by Goldbeck-Wood et al., a practical method for simulating LCP orientation is used to model the polymer flow, and the directionality results are then used to calculate a quantitative molecular degree of order. This metric, known as the order parameter, is an ideal candidate for measuring the LCP orientation, ranging from zero to unity between the isotropic and perfectly aligned states, respectively, as it is sensitive to both the direction of the average molecular orientation, as well as to the distribution of crystals around the average orientation. The effects of varying process parameters in the directionality model on the order parameter are shown. Understanding of these relationships will ultimately drive the design of manufacturing processes for more isotropic materials.

Abstract The performance properties of many polymers are dominated by the molecular alignment induced during processing by the elongational flow component, that is, the diagonal component of the velocity gradient tensor. Here we discuss two idealizations of this flow-microstructure interaction. We first summarize our recent studies of biaxial nematic patterns and phase transitions (Forest and Wang, 1998a) of lyotropic liquid crystalline polymers (LCPs) in response to imposed elongational flows. We show axisymmetric biaxial nematic patterns coexist with homogeneous biaxial patterns characterized by Rey (Rey, 1995). All biaxial patterns in unidirectional stretching flows are unstable, whereas planar stretching induces candidate stable biaxial nematic patterns. Non-homogeneous biaxial patterns have potential core defects, and two special director alignments are selected by imposing a finite-pressure condition along the axis of flow symmetry. The role of director dynamics in response to flow perturbations is described, and in particular new, elongation-induced, director instabilities are revealed, extending seminal stability and phase transition results (Khokhlov and Semenov, 1982; See et al., 1992; Bhave et al, 1993; Rey, 1995; Hu and Ryskin, 1992). We then examine two special solution families of the governing equations for free surface, axisymmetric, cylindrical LCP fiber flows, following analogous Newtonian results of Segur et al. (Segur et al., 1997). The cylindrical free surface only accommodates uniaxial orientation, and from these ideal transient solutions we analyze the linearized stability of the coupled flow and microstructure in the LCP thin-filament approximation (Forest et al, 1997).

Abstract Liquid crystalline polymers (LCPs) are a relatively new class of polymers with excellent high temperature properties. They are being considered for electronic packaging applications, such as molded multichip modules, where high dimensional tolerances are critical for component operation. High stresses and temperatures can be generated in molded leadframes during surface mount soldering operations and component service. Under these conditions, time dependent deformation or creep could result in warpage and failure of molded leadframes. In order to aid designers in estimating component lifetimes, the creep properties of Xydar G930 were investigated. Xydar G930 is a LCP with 30 wt.% glass filler. A series of short term (24 h) uniaxial creep tests were conducted at several temperatures and stress levels ranging from room temperature to 350°F, and 0.3 UTS to 0.8 UTS, respectively. Values of steady state creep rate were calculated from the creep curves and fit to an Arrhenius analog as a function of temperature and a power law relation as a function of stress. Results showed linear viscoelastic behavior at low stresses and significant dependence of creep on temperature and stress.
