Six bovins de race Boran ont été infectés par la souche chimiorésistante IL 3338 de Trypanosoma congolense. Au premier pic de parasitémie, 200 Glossina morsitans centralis ténérales organisées en plusieurs groupes ont été nourries sur chaque animal, juste avant leur traitement à l'acéturate de diminazène à la dose de 3,5 mg/kg de poids corporel (p.c.). Les animaux ont ensuite étaient suivis par prélèvement sanguin trois fois par semaine et ont été traitées comme précédemment chaque fois que l'hématocrite baissait dans trois prélèvements consécutifs, pour au moins un des animaux. Après huit traitements à approximativement deux semaines d'intervalle, la longueur moyenne de la période prépatente après chaque traitement n'a pas augmenté, restant à 7,8 +- 1,1 jours. La valeur moyenne de l'hématocrite est descendue de 33,2 +- 0,6 % lors du premier traitement, à 23,7 +- 2,6 % entre les huitième et neuvième traitements. En conséquence, à la suite du huitième traitement, l'acéturate de diminazène a été administré comme précédemment, mais à la dose de 7,0 mg/kg p.c. Résultant de l'augmentation de la dose, la valeur moyenne de l'hématocrite entre deux traitements est montée de 25,4 +- 2,4 % sitôt après le premier traitement à 32,9 +- 1,7 pour les deux mois qui ont suivi le cinquième traitement.

La dépression, deuxième source de handicap, constitue le principal facteur de risque de suicide. Il a été proposé à juste titre que le traitement de la dépression soit une stratégie efficace de prévention du suicide. Pourtant la dernière décennie a été marquée par les controverses sur le risque suicidaire potentiel des antidépresseurs, qui ont eu des effets néfastes sur la prise en charge des patients. Puisque les alertes sur ce risque ont été proposées à partir des résultats d’études qui n’avaient pas comme objectif de s’intéresser au processus suicidaire, nous avons réalisé une étude destinée à évaluer la survenue d’idées et de tentatives de suicide chez 4357 patients déprimés ambulatoires traités par antidépresseur et suivis 6 semaines en condition naturelles. Une idéation suicidaire est apparue de novo chez 9 % des patients qui n’en avaient pas à l’inclusion. Les critères prédictifs étaient l’aggravation de la symptomatologie anxieuse ou dépressive et l’instauration d’un nouveau traitement après échec antérieur. Deux pour cent des patients ont réalisé une TS au cours des 6 semaines de suivi. Les critères prédictifs étaient l’aggravation de la symptomatologie dépressive ou du désespoir, l’instauration d’un nouveau traitement après échec antérieur, et l’existence de mésusage d’alcool et d’antécédents de TS. Ces résultats suggèrent que le risque suicidaire en début de traitement soit lié aux facteurs de risque suicidaires, dont l’inefficacité du traitement antidépresseur. Par ailleurs, nous avons montré que l’initiation d’un traitement par non ISRS ou d’un traitement ISRS à forte dose augmente le risque d’aggravation des idées de suicide. L’ensemble de ces données, confortées par la littérature, nous indique que les médecins possèdent les outils de prévention en évaluant précisément et régulièrement la dépression et le risque suicidaire au cours du traitement antidépresseur.

Abstract Conventional pharmacotherapy in oncology centers on target-centric silencing of aberrant protein function through small molecule inhibitors, degraders, or CRISPR/RNAi. In contrast, here we leverage induced proximity to develop pharmacology that activates dominant signaling pathways for a therapeutic effect. To develop this concept, we describe a new class of bivalent small molecules that reprogram cancer-driving gene expression to activate cell death signaling in diffuse large B cell lymphomas (DLBCL). These molecules, called Transcriptional Chemical Inducers of Proximity (TCIPs), operate at fractional occupancy of their protein targets, without genomic modifications to the cell or organism. The first TCIPs transiently re-localized elongation factors such as BRD4 (TCIP1, PMID 37495688) to loci regulated by the transcription factor BCL6, inducing the expression of pro-apoptotic genes regulated by BCL6. In new, unpublished work, CDK-TCIPs have been made to re-direct the activity of transcriptional elongation kinases such as CDK9, CDK12, or CDK13 to induce the expression of pro-apoptotic BCL6-target genes. Profiling in ∼900 barcoded cancer cell lines show that the molecules exhibit potent cell killing potency in a highly DLBCL lineage-restricted manner. Cytotoxicity to healthy lymphocytes is 100- to 10,000-fold lower, although germinal center B cells are ablated in a dose-dependent manner consistent with the role of BCL6 in the germinal center. Biochemical, cell biological, proteomic, and genomic studies demonstrate that CDK-TCIPs operate via a gain-of-function mechanism reliant on the ternary complex formed by the bivalent molecule. Instead of global enzyme inhibition or degradation, CDK-TCIPs borrow and re-localize a fraction of total CDK activity to BCL6-bound chromatin, producing pharmacology from the induction of pro-apoptotic gene transcription. The findings suggest a path to rational design of new gain-of-function chemotherapeutic strategies to rewire cell signaling using induced proximity. New research data for low-dose, low- linear energy transfer (LET) radiation-induced, stochastic effects (mutations and neoplastic transformations) are modeled using the recently published NEOTRANS3 model. The model incorporates a protective, stochastic threshold (StoThresh) at low doses for activating cooperative protective processes considered to include presumptive p53-dependent, high-fidelity repair of nuclear DNA damage in competition with presumptive p53-dependent apoptosis and a novel presumptive p53-independent protective apoptosis mediated (PAM) process which selectively removes genomically compromised cells (mutants, neoplastic transformants, micronucleated cells, etc.). The protective StoThresh are considered to fall in a relatively narrow low-dose zone (Transition Zone A). Below Transition Zone A is the ultra-low-dose region where it is assumed that only low-fidelity DNA repair is activated along with presumably apoptosis. For this zone there is evidence for an increase in mutations with increases in dose. Just above Transition Zone A, a Zone of Maximal Protection (suppression of stochastic effects) arises and is attributed to maximal cooperation of high-fidelity, DNA repair/apoptosis and the PAM process. The width of the Zone of Maximal Protection depends on low-LET radiation dose rate and appears to depend on photon radiation energy. Just above the Zone of Maximal Protection is Transition Zone B, where deleterious StoThresh for preventing the PAM process fall. Just above Transition Zone B is a zone of moderate doses where complete inhibition of the PAM process appears to occur. However, for both Transition Zone B and the zone of complete inhibition of the PAM process, high-fidelity DNA repair/apoptosis are presumed to still operate. The indicated protective and deleterious StoThresh lead to nonlinear, hormetic-type dose-response relationships for low-LET radiation-induced mutations, neoplastic transformation and, presumably, also for cancer.

The development and use of immunotherapy in the last decade have led to a drastic improvement in results in the onco-haematological field. This has implied, on the one hand, the need for clinicians to manage a new type of adverse event and, on the other hand, a significant increase in costs. However, emerging scientific evidence suggests that, as with other drugs in the recent past, the registry dosage can be drastically reduced for immunotherapies without penalizing their effectiveness. This would also lead to an important reduction in costs, expanding the audience of cancer patients who could access immunotherapy-based treatments. In this “Commentary”, we analyze the available evidence of pharmacokinetics and pharmacodynamics and the most recent literature in favor of low-dose immunotherapy.

Objective. This study aims to assess the impact of adaptive statistical iterative reconstruction (ASIR) on CT imaging quality, diagnostic interpretability, and radiation dose reduction for a proven CT acquisition protocol for total body trauma.Methods. 18 patients with multiple trauma (ISS≥16) were examined either with a routine protocol (n=6), 30% (n=6), or 40% (n=6) of iterative reconstruction (IR) modification in the raw data domain of the routine protocol (140 kV, collimation: 40, noise index: 15). Study groups were matched by scan range and maximal abdominal diameter. Image noise was quantitatively measured. Image contrast, image noise, and overall interpretability were evaluated by two experienced and blinded readers. The amount of radiation dose reductions was evaluated.Results. No statistically significant differences between routine and IR protocols regarding image noise, contrast, and interpretability were present. Mean effective dose for the routine protocol was25.3±2.9 mSv,19.7±5.8 mSv for the IR 30, and17.5±4.2 mSv for the IR 40 protocol, that is, 22.1% effective dose reduction for IR 30 (P=0.093) and 30.8% effective dose reduction for IR 40 (P=0.0203).Conclusions. IR does not reduce study interpretability in total body trauma protocols while providing a significant reduction in effective radiation dose.

The experiment was conducted at Jute Agriculture Experimental Station (JAES), Manikganj and Jute Research Regional Station, Faridpur during 2019 to find out the effect of weeding and herbicide management on fibre yield and yield attributes of Tossa jute (Var. O-9897) cultivation. Two herbicide treatments, such as; W1 = Fenoxaprop-p-ethyl (Whip Super 9 EC) and W2 = Ethoxysulfuron (Sunrice ® 150WG) were tested. There were 14 treatments. T1- T6 treatments did not include hand weeding which was done 20 days after the herbicide spray [T1 = 100% recommended doses (RD) of both W1and W2 where W2 was sprayed one day after that of W1, T 2 = 50% dose both of W1and W2 where W2 was sprayed one day after that of W1, T3 = 100% dose of both W1 and W2 where both of the herbicide mixed together and sprayed on the same day, T4 = 50% dose of both W1 and W2 where mixture of both were sprayed on the same day, T5 = only 100% dose of W1, T6 = only 100% dose of W2]. The treatments T7 - T12 included one had weeding following the application of herbicides [T7 = 100% dose both of W1 and W2 where W2 was sprayed 1 day after that of W1, T8 = 50% dose both of W1 and W2 where W2 was sprayed 1 day after of W1, T9 = 100% dose both of W1 and W2 were mixed and sprayed together on the same day, T10 = 50% dose both of W1 and W2 mixed together and sprayed same day, T11 = only 100% dose of W1, T12 = only 100% dose of W2]. The treatments T13 and T14 did not include herbicide application [T13 = Two hand weeding (20-25 and 45-50 DAS) and T14 = control with no herbicide application]. The experiment was laid out in RCBD with three replications. Results revealed that Khudesama (Echinochloa colonum) and Angulighash (Digitaria sanguinalis), Khudesama and Mutha (Cyperus rotundus) were best controlled by herbicide application. T3 gave highest fibre yield of 1.89 tha-1 when W1 and W2 sprayed at the same day with 100% RD. In case of one weeding 20 days after spray fibre yield and stick yield were differed significantly as compared to those of control at Manikganj. Treatment T7 gave highest fibre yield (3.11 t ha-1) when W2 sprayed after 1 day of W1 with 100% RD. At Faridpur, T3 gave the highest fibre yield (2.68 t ha-1) when W1 and W2 sprayed at the same day with 100% RD. The probiotic definition requires the administration of an ‘adequate amount’ in order to obtain a health benefit. What that amount should be is not indicated. Here, an overview is given of studies that investigated the dose-response relation of probiotics in human interventions. Studies were divided in; meta-analyses, meta-analyses on specific probiotic strains, and studies testing two or more doses of a probiotic (combination) in the same study. Meta-analyses on the effect of probiotics on antibiotic associated diarrhoea (AAD) suggest a dose-response effect; for Clostridium difficile-associated diarrhoea on the other hand no dose-response was observed. For other end-points; such as necrotising enterocolitis, prevention of atopic dermatitis and slow intestinal transit, no dose-response relation was identified in meta-analyses. For prophylaxis in colorectal cancer and relief of irritable bowel syndrome, no dose-response relation was determined. However, for blood pressure, a meta-analysis observed that higher doses (greater than 1011 cfu) were more effective than lower doses. Meta-analyses of specific strains suggest a break-point for effectiveness of Lactobacillus rhamnosus GG in the treatment of acute gastroenteritis in children; no dose-response was observed for two other probiotics assessed. Studies comparing two or more doses indicate that faecal recovery and risk reduction of AAD follow a positive dose-response relationship. Other end-points such as immune markers, general health, and bowel function did not exhibit clear dose-response relations. For AAD, the findings are very compelling; both meta-analyses and dedicated dose-response studies observe a positive correlation between dose and AAD risk. These findings do not allow for extrapolation, but suggest that studying higher doses for this end-point would be worthwhile. The lack of a clear dose-response for other end-points, does not mean it does not exist; present data does just not allow drawing any conclusions.

The article proposes an approach to the types of functioning and mechanisms of change intervening in the rise of the discourse marker comme de raison in French, associated with evidentiality. The diachronic analysis provides the opportunity to study the relationship between comme de raison and the meanings of raison, but also the type of structure at the origin of the discourse marker. The status of the construction will be questioned, which is intertwined with the discussion about the mechanism of change (lexicalization, grammaticalization / pragmaticalization, cooptation). The semantic evolution could be described as follows: from « what is normal, right to do, what should be done » to « what is normal, right to say, what should be said », since this could be inferred from a previous sentence or a more general context.

Radiation-induced conductivity (RIC) of polystyrene has been studied experimentally and numerically in a broad time range from milliseconds to seconds at 79 K, which seems to have been done for the first time. It has been established that the hole dispersive transport is still operative but unlike the room temperature behavior it features an extremely small frequency factor (4 × 103 s−1) and accordingly low dispersion parameter (0.09). It has been shown that prompt conductivity per unit dose rate does not depend on temperature (290 or 79 K) and is equal to [Formula: see text] (±20%) at 4 × 107 V/m. At 79 K, current–voltage characteristics of both prompt and delayed components are close enough and non-linear. Due to the contribution of the thermalized charge carriers, the RIC even at 79 K can be satisfactorily described by the semi-empirical Rose–Fowler–Vaisberg model based on the quasi-band multiple trapping mechanism just as it happens at room temperature.

Multilingualism is one of the pillars of the European Union (EU), enshrined in its treaties and celebrated in its motto, “united in diversity.” Yet multilingualism no longer has its own portfolio in the Commission, having been systematically downgraded and now being under the auspices of the Directorate-General for Translation, a directorate which nevertheless does not have a unit dedicated to multilingualism. Moreover, with the rise of English as the EU’s unofficial lingua franca, increasingly more material is produced in English and not translated at all. Therefore, we should ask ourselves whether the EU’s de facto linguistic and translation regimes are not at odds with the treaties. Drawing together transdisciplinary threads chiefly from linguistics, political science and political philosophy, this paper assesses the EU’s current linguistic regime, while looking at different models of linguistic justice, language rights, and the value of language to propose a new linguisticmodus operandi for the EU, grounded in (1) a language, (2) a translation, and (3) a cultural turn.

Fixed expression and idioms are part of the translated discourse. They are ready -made constructions that summarize an idea or a concept by using outstanding formulas. Translate these expressions is a big challenge for the translator. Thus, a well done translation does not mean mastering the linguistic skills of both languages, since the text does not contain just linguistic facts. It is also about culture, emotions, connotations, style and the author’s genius. Formulaic language often presents significant issue in translation just as well as finding a fixed expression as equivalent.

The relationship between outcome and dose intensity was analyzed for first-line chemotherapy of advanced ovarian cancer using a particular CHAP (cyclophosphamide, hexamethylmelamine, Adriamycin [Adria Laboratories, Columbus, OH], cisplatin) regimen as the standard. Previously described techniques were used to calculate the average dose intensity of regimens containing one, two, three, or all four drugs of CHAP, relative to the standard. The average relative dose intensity, especially the relative dose intensity of cisplatin, correlated significantly with clinical response and with median survival time (MST) of the entire group (not just the remitters). There was a distinct advantage for multiagent regimens over single alkylating agents and especially for multiagent regimens containing cisplatin. Survival correlated with response rate (of multiagent regimens). This analysis suggests that dose intensity is a determinant of treatment outcome. Prospective randomized trials would be required to test whether, and to what extent, dose intensity determines outcome independently of total amount of drug given, performance status, or other factors. If dose intensity does determine outcome, methods of increasing it should be tested in an attempt to improve treatment results.

Unlike other officially multilingual states, Switzerland is often seen as a glottophile country, where linguistic minorities have few complaints. Its success is often attributed to the principle of territoriality (one official language per territory), but the principle of personality (free choice of by citizens of the language in State-citizen communications) is also important in bi/plurilingual cantons, in what appears to be a differentiated territoriality. The legal framework is based on cooperation between the cantons, which govern their own linguistic matters, and the federal State. The officialization of the three main languages (German, French, and Italian) was recently supplemented by that of Romansh, a vulnerable language from a demographic perspective. At the same time, the Swiss-German dialects have remained very much alive, despite their absence from constitutional texts. However, one should be wary of total acceptance of this idealised image of Switzerland, when the legislator seems indifferent to languages of immigration and, moreover, does not grant recognition to Franco-Provençal. This Romance language that is still spoken in parts of Francophone Switzerland is not protected by any policies aimed at ensuring its survival. This article aims to reflect on the applicability of the notion of glottophilia to Switzerland by focusing on the texts that constitute the architecture of language legislation in the country.

<p>Sound speed is an important acoustic parameter for tissue characterization. Herein we developed an ultrasound computed tomography (USCT) system for <i>ex vivo</i> sound speed imaging and evaluation of small animal organs. The proposed USCT system employs a 256-element ring array transducer and allows simultaneous signal transmission and reception for all channels. The method does not require complicated sample preparation procedures and can yield accurate measurement results. Experimental results show that sound speeds of excised rat brain, heart, liver, spleen, and kidney measured by the method are close to published data. This work demonstrates a new method for sound speed imaging and holds potential for <i>in vivo</i> applications.</p>

A 499.8 MHz SOLEIL-type superconducting cavity was simulated and designed for the first time in this paper. The higher-order mode (HOM) properties of the cavity were investigated. Two kinds of coaxial HOM couplers were designed. Using 4 L-type and 4 T-type HOM couplers, the longitudinal impedance was suppressed to 3 kΩ, and the transverse impedance below 30 kΩ/m. The HOM damping requirements of Hefei Advanced Light Facility (HALF) were satisfied. This paper conducts an in-depth study on the radio frequency (RF) design, multipacting optimization, and thermal analysis of these coaxial couplers. Simulation results indicated that under operating acceleration voltage, the optimized couplers does not exhibit multiplicating or thermal breakdown phenomena. The cavity has the potential to reach a higher acceleration gradient.

In March and April of 2021, my co-investigators and I conducted semi-structured interviews with academic librarians across Canada about their work during the COVID-19 pandemic, which included their thoughts about going “back to normal.” Most participants were resistant to returning to the “old normal” without myriad changes inspired by the COVID-necessitated adaptations. However, there were concerns raised about whether or not their ideas would be implemented or even heard by their administrations. Additionally, many participants felt caught between proving their value through productive (and measurable) labour and the care-work that felt necessary and pressing but was not externally validated. This paper highlights the need for refocusing on building library collegial governance structures that include all library workers. As well, there is indication that the COVID-19 pandemic presents a unique opportunity to do so, as, removed from the “sacred space” (Ettarh 2018) of the library building, participants showed resistance to the austerity narratives typically invoked during a crisis. Embodying our values starts with establishing and building on shared library governance structures. If the changes inspired by COVID are to come to pass, then our vision of care and relationship-building must be inclusive to our own workers, to harness our collective power to build a future that works for everyone.
