In this work the decomposition aspects of the Staurastrum iversenii Nygaard var. americanum phytoplankton species, isolated from the Barra Bonita Reservoir (22 degrees 29 minutes S e 48 degrees 34 minutes W), were approached; as resource of the decomposition were used: cells (ruptured and whole) and polysaccharide of higher and lower molecular masses. Were approached, still, the effects of the following variables: temperature and concentration of dissolved oxygen. Therefore, decomposition chambers were set up in laboratory with cells (ruptured and whole) and polysaccharides of higher and lower molecular masses deriving from S. iversenii. Based on the quantifications of the organic carbon concentrations, the decomposition processes of the resources were described and the kinetic coefficients evaluated. These descriptions constituted the base of the models proposed for the resource decomposition. These models allowed to infer that the used substrata constituted from a structural point of view, of composed heterogeneous, presenting labile and refracting fractions. In relation to the consumption of oxygen, the decomposition of the ruptured cells was observed to be faster than that of the whole cells; for the polysaccharides, the consumptions of oxygen were faster in the mineralization of the fractions of high molecular mass. The temperature influenced the speeds of the processes involved with the mineralization of the refracting fractions, besides altering the mineralization routes of the resources; it, also, induced the occurrence of alterations of the stoichiometric (O/C) relationships of the processes. It was possible to verify that the sugars which form these polysaccharides contributed with the supply of energy to the microorganisms during a long period. The proposed mathematical models were shown to be sensitive to the experimental results. The degradation of S. iversenii can contribute to the maintenance of the processes connected to the maintenance of the Barra Bonita Reservoir microbial loop.
Neste trabalho foram abordados aspectos da decomposição da espécie fitoplanctônica Staurastrumive iversenii Nygaard var. americanum, isolada do reservatório de Barra Bonita (22 graus 29 minutos S e 48 graus 34 minutos W); como recurso da decomposição utilizaram-se: células (rompidas e íntegras) e polissacarídeos de alta e baixa massas moleculares. Foram abordados, ainda, os efeitos das seguintes variáveis: temperatura e concentração oxigênio dissolvido. Para tanto, montou-se em laboratório, câmaras de decomposição com células (rompidas e íntegras) e polissacarídeos (de alta e baixa massas moleculares) provenientes de S. iversenii. Com base nas quantificações das concentrações de carbono orgânico descreveu-se os processos de decomposição dos recursos e avaliou-se os coeficientes cinéticos. Estas descrições constituíram a base dos modelos propostos para a decomposição dos recursos. Esses modelos permitiram inferir que os substratos utilizados constituíram-se, do ponto de vista estrutural, de compostos heterogêneos, apresentando frações lábeis e refratárias. Em relação ao consumo de oxigênio, observou-se que foi mais rápido na decomposição das células rompidas que das células íntegras; para os polissacarídeos, os consumos de oxigênio foram mais rápidos nas mineralizações das frações de alta massa molecular. A temperatura influenciou as velocidades dos processos envolvidos com a mineralização das frações refratárias, além de alterar as rotas das mineralizações dos recursos; induziu, também, a ocorrência de alterações das relações estequiométricas (O/C) dos processos. Foi possível verificar que os açucares que formam estes polissacarídeos contribuíram com o fornecimento de energia para os microrganismos durante um longo período. Os modelos matemáticos propostos mostraram-se sensíveis aos resultados experimentais. A degradação de S. iversenii pode contribuir para a manutenção dos processos ligados a manutenção do elo microbiano do reservatório de Barra Bonita.

Texte intégralA new strategy has been developed to facilitate the total synthesis of genes. The approach involves the synthesis of segments of the gene which are cloned into a vector for amplification and proof-reading. The construction of the gene proceeds from both ends toward the middle. In the synthesis, a segment of the gene is cloned into the vector and amplified; the resulting vector now contains a newly cloned segment which importantly includes unique internal restriction sites. The vector can be opened at these sites to allow insertion of an additional segment of the structural gene. A combination of three procedures: opening, cloning, amplification, constitutes a "step" of the synthesis, and a sequence of such "steps" leads to the synthesis of the total gene.
Many unique restriction sites have been designed into the gene thusly synthesized, as their presence will greatly facilitate the use of cassette mutagenesis in subsequent structure/function studies of the protein encoded by the synthetic gene.
This synthetic strategy has several attractive features. In principle it can be used for the construction of genes of essentially any length and, allows proof-reading and correction of errors at intermediate stages in synthesis. This approach thereby avoids some of the problems inherent in the more traditional strategy in which many segments of synthetic DNA are annealed, ligated together and inserted into a vector, followed by a hoped for isolation from the many species inevitably present in the resulting mixture of synthetic genetic material of a clone containing DNA with the desired sequence.
This stepwise approach has been applied to the synthesis of a gene for the blue copper protein, plastocyanin, of poplar leaf. The synthetic gene has been expressed in a fusion with a region of protein A under control of the λR promoter.
