We previously demonstrated that neural stem/progenitor cells (NSPCs) were induced within and around the ischemic areas in a mouse model of ischemic stroke. These injury/ischemia-induced NSPCs (iNSPCs) differentiated to electrophysiologically functional neurons in vitro, indicating the presence of a self-repair system following injury. However, during the healing process after stroke, ischemic areas were gradually occupied by inflammatory cells, mainly microglial cells/macrophages (MGs/MΦs), and neurogenesis rarely occurred within and around the ischemic areas. Therefore, to achieve neural regeneration by utilizing endogenous iNSPCs, regulation of MGs/MΦs after an ischemic stroke might be necessary. To test this hypothesis, we used iNSPCs isolated from the ischemic areas after a stroke in our mouse model to investigate the role of MGs/MΦs in iNSPC regulation. In coculture experiments, we show that the presence of MGs/MΦs significantly reduces not only the proliferation but also the differentiation of iNSPCs toward neuronal cells, thereby preventing neurogenesis. These effects, however, are mitigated by MG/MΦ depletion using clodronate encapsulated in liposomes. Additionally, gene ontology analysis reveals that proliferation and neuronal differentiation are negatively regulated in iNSPCs cocultured with MGs/MΦs. These results indicate that MGs/MΦs negatively impact neurogenesis via iNSPCs, suggesting that the regulation of MGs/MΦs is essential to achieve iNSPC-based neural regeneration following an ischemic stroke.

An inositol-requiring strain of Neurospora crassa was labelled during growth in liquid medium with [3H]inositol, and the levels of inositol phosphates and phosphoinositides were determined under inositol-sufficient and inositol-starved conditions. Because the mutant has an absolute requirement for inositol, the total mass of inositol-containing compounds could be determined. Inositol-containing lipids were identified by deacylation and co-migration with standards on h.p.l.c.; PtdIns3P, PtdIns4P, and PtdIns(4,5)P2 were found in approximately equal amounts, in addition to large amounts of PtdIns. Inositol starvation decreased the level of PtdIns to 10% of the sufficient level, and decreased the levels of the other phosphoinositides to about 25%. A number of inositol phosphates were found, including several InsP3s, InsP4s and InsP5s and phytic acid. Ins(1,4,5)P3 was identified by co-migration with standards on h.p.l.c. and by digestion with inositol phosphomonoesterase. High concentrations of all inositol phosphates were found in the extracellular medium in inositol-starved cultures. Inositol starvation on both liquid and solid agar media decreased the intracellular levels of some inositol phosphates, but increased the levels of phytic acid and several other inositol phosphates which may be its precursors and/or breakdown products. These results may indicate that inositol starvation induces phytic acid synthesis as a protection against the free-radical production and lipid peroxidation characteristic of inositol-less death.

Arthropod antimicrobial peptides (AMPs) offer a promising source of new leads to address the declining number of novel antibiotics and the increasing prevalence of multidrug-resistant bacterial pathogens. AMPs with potent activity against Gram-negative bacteria and distinct modes of action have been identified in insects and scorpions, allowing the discovery of AMP combinations with additive and/or synergistic effects. Here, we tested the synergistic activity of two AMPs, from the dung beetle Copris tripartitus (CopA3) and the scorpion Heterometrus petersii (Hp1090), against two strains of Escherichia coli. We also tested the antibacterial activity of two hybrid peptides generated by joining CopA3 and Hp1090 with linkers comprising two (InSco2) or six (InSco6) glycine residues. We found that CopA3 and Hp1090 acted synergistically against both bacterial strains, and the hybrid peptide InSco2 showed more potent bactericidal activity than the parental AMPs or InSco6. Molecular dynamics simulations revealed that the short linker stabilizes an N-terminal 310-helix in the hybrid peptide InSco2. This secondary structure forms from a coil region that interacts with phosphatidylethanolamine in the membrane bilayer model. The highest concentration of the hybrid peptides used in this study was associated with stronger hemolytic activity than equivalent concentrations of the parental AMPs. As observed for CopA3, the increasing concentration of InSco2 was also cytotoxic to BHK-21 cells. We conclude that AMP hybrids linked by glycine spacers display potent antibacterial activity and that the cytotoxic activity can be modulated by adjusting the nature of the linker peptide, thus offering a strategy to produce hybrid peptides as safe replacements or adjuncts for conventional antibiotic therapy.

The neonatal brain is substantially more resistant to various forms of injury than the mature brain. For instance, the prognosis following ischemic stroke is generally poor in the elderly but favorable in neonates. Identifying the cellular and molecular mechanisms underlying reparative activities in the neonatal brain after ischemic injury may provide feasible targets for therapeutic interventions in adults. To this end, we compared the reparative activities in postnatal day 13 and adult (8–12-week-old) mouse brain following middle cerebral artery occlusion. Immunohistochemistry revealed considerably greater generation of ischemia-induced neural stem/progenitor cells (iNSPCs) expressing nestin or Sox2 in ischemic areas of the neonatal brain. The iNSPCs isolated from the neonatal brain also demonstrated greater proliferative activity than those isolated from adult mice. In addition, genes associated with neuronal differentiation were enriched in iNSPCs isolated from the neonatal brain according to microarray and gene ontogeny analyses. Immunohistochemistry further revealed considerably greater production of newborn doublecortin+ neurons at the sites of ischemic injury in the neonatal brain compared to the adult brain. These findings suggest that greater iNSPC generation and neurogenic differentiation capacities contribute to the superior regeneration of the neonatal brain following ischemia. Together, our findings may help identify therapeutic targets for enhancing the reparative potential of the adult brain following stroke.

Lean Construction is one of the methods used to improve control over construction projects by eliminating waste in time and materials. Lean Construction is an adaptation from the Lean Manufacturing principles to the construction industry. The purpose of this article was to review the case studies published in 2018 in the Inspec database to find out where Lean is being implemented and how. The article approach by setting the search criteria first and then inspect the result to find the non-related results and eliminate it. After that, the article was reviewed and summarized. The article verifies each article finding; however Lean Construction and Building Information Modeling (BIM) are relatively new for the construction industry and they still need more time to be adopted widely and to be applied on a lower-cost budget. However, the discussed cases show a promising future for these technologies.

Inositol phosphates (IPs) are important signalling molecules with various biological roles in a cell. One such role it is often associated with is DNA repair. The DNA repair process following DNA insult is considered crucial for the genomic integrity and stability. Failure to perform this task will result in mutations and possibly disease. Thus, it is important that we expand our knowledge on how these repair processes occur and identify the key factors involved in its regulation. The aim of this project was to investigate whether DNA repair was mediated by inositol diphosphates (IDPs). Using a family of yeast knockout mutants with modulated levels of IPs, it was found that IDPs are crucial in repair of DNA following insult with bleomycin and 5-fluorouracil. The observed sensitivity of the mutants was thought to be due to lack of functional repair protein, UDG-like or APE-like, in the absence of essential cofactor such as IDPs. Experiments conducted revealed that the hypersensitive kcs1Δ contain both the repair proteins required to process the DNA lesions. However, extreme extraction methods were required to access these proteins, suggesting that the proteins are mislocalised and unavailable to access the damage site and perform DNA repair. GFP-tagging the proteins Ung1, Apn1 and Rad52 in kcs1Δ proved to be of little use as it failed to show exact localisation, movement and functionality status of these proteins following bleomycin insult. The enzymes accountable for the dephosphorylation of the IDPs in vivo are the diphosphoinositol polyphosphate phosphohydrolases (DIPPs). Little is known regarding the Michaelis-Menten kinetics parameters for Ddp1p/DIPPs. In this study, using improved methods for the enzymatic synthesis and electrophoretic purification of 1-InsP7, 5-InsP7 and InsP8, the DIPP family has been kinetically characterised. Each DIPP was found to ii display similar Km values for every substrate tested (range: 35-148 nM). The rank order of Kcat values (1-InsP7 > 5-InsP7 = InsP8) was identical for each enzyme, although DIPP-1 activity was observed to be 10- to 60-fold more than DIPP-2α/β and DIPP-3α/β, irrespective of the substrate. This study reveals that Ddp1, the yeast DIPP, is capable of hydrolysing not only 5-InsP7 but also 1-InsP7 and InsP8 to a single product, InsP6. The HPLC data found InsP7 accumulation to be relatively little during InsP8 breakdown by DIPPs. Such low build-up was found to be due to rapid conversion of InsP7 to InsP6. Through this study it is also clear that InsP8 prefers to dephosphorylate through 1-InsP7. In contrary, metabolically and functionally significant steady-state route of InsP8 synthesis was observed to be via 5-InsP7. Oesophageal cancer is considered as one of the deadliest cancers worldwide because of its aggressive nature and low survival rate. Epidemiologic studies have shown that low-dose daily intake of aspirin can decrease the incidence of oesophageal cancer. The data presented in this study show the effects of a number of in-house synthesized novel aspirin analogues on oesophageal cancer cell lines, squamous cell carcinoma (SSC) and adenocarcinoma (ADC). The aspirin analogues, fumaryldiaspirin (PN517) and benzoylsalicylates (PN524, PN528 and PN529), were observed to be more potent against the oesophageal cell lines than aspirin itself. Both, quantitative and qualitative apoptosis experiments conducted revealed that these compounds largely induced apoptosis, although some necrosis was evident with PN528 and PN529. Failure to recover following the treatment with these analogues emphasized that these drugs are largely cytotoxic in nature. The SSC cells (oe21) displayed increased sensitivity to the aspirin analogues compared to the ADC cell lines (flo-1 and oe33). The anticancer properties of these novel aspirin compounds appear to not involve the COX-enzymes at the tested concentrations. These initial findings support further studies into the potential of these aspirin analogues as chemotherapeutic agents against oesophageal cancer.

L’étude des inositols hautement phosphorylés est un domaine en pleine expansion. Leurs essors ne datent que d’une dizaine d’années, mais de nombreuses fonctions y sont déjà associées bien qu’ils en restent sans doute encore à découvrir. Les inositols phosphates (incluant les inositols hautement phosphorylés) s’inscrivent dans un cycle dont le représentant le plus connu est inositol 1,4,5-trisphosphate (Ins(1,4,5)P3). De ce fait, chaque inositol phosphate influence directement ou indirectement les autres membres de ce cycle.

Au cours de la thèse, nous avons pu éclaircir une controverse de la littérature sur la voie de synthèse des inositols hautement phosphorylés. Grâce à un modèle de cellules MEF (mouse embryonic fibroblast) n’exprimant aucune des trois isoformes de l’inositol 1,4,5-trisphosphate 3-kinase (ITPK) et à l’aide des cellules souches déficientes pour l’inositol polyphosphate multikinase (IPMK), nous avons pu révéler le rôle majeur de cette dernière dans la génération de l’InsP5 et l’InsP6.

Dans un second temps, nous avons comparé la neurogenèse de ces cellules souches IPMK+/+ et IPMK-/- mises dans un milieu de différenciation par défaut (DDM). Les cellules dépourvues de l’IPMK entrent en apoptose et se différencient très difficilement en progéniteurs neuronaux et en neurones. Afin de comprendre le mécanisme sous-jacent pouvant expliquer ce phénomène, nous avons réalisé des PCRs quantitatives qui ont montré une sous expression des gènes du neuroectoderme ainsi qu’une augmentation de l’expression de gènes du mésoderme dans les cellules IPMK-/- par rapport aux cellules IPMK+/+. De plus, nous avons découvert que le phénomène d’apoptose observé au cours de la différenciation en DDM était spécifique à ce milieu. En effet, nous n’avons pas pu mettre en évidence un tel phénomène au cours de la différenciation en corps embryoïdes.

Durant la thèse, nous avons également développé des anticorps dirigés contre l’isoforme B de l’inositol 1,4,5-trisphosphate 3-kinase et contre la forme native de l’IPMK. Ceci nous a permis de mener à bien nos expériences et d’ouvrir de futures perspectives de recherche.

En conclusion, nous avons démontré le rôle majeur de l’IPMK dans la voie de synthèse des inositols hautement phosphorylés. Nous avons également découvert que l’IPMK est très importante pour la survie de ces cellules souches en cours de différenciation et nous avons également introduit une nouvelle fonction pour l’IPMK dans la neurogenèse.

The ability for an organism to sense and respond appropriately to its environment is often critical for survival. One mechanism for this is the inositol phosphate (InsP) signaling pathway. This work focuses on the role of InsP signaling in maintaining energy homeostasis in the plant. InsP signaling is connected to energy sensing in plants via a protein complex containing both the inositol polyphosphate 5-phosphatases (5PTase13) and the Sucrose non-Fermenting Related Kinase 1 (SnRK1). SnRK1 is considered a fuel gauge for the plant cell that senses energy status and reprograms growth appropriately. While the SnRK1.1 gene has been well studied, the role other SnRK1 isoforms play in energy or stress signaling is less well understood. This work examined the role of 3 SnRK1 isoforms in energy signaling, finding that SnRK1.1 and SnRK1.2 are regulated and function differently in Arabidopsis. The second part of this work focuses on the inositol pyrophosphates, which are a novel group of InsP signaling molecules containing diphosphate or triphosphate chains (i.e. PPx) attached to the inositol ring. These PPx-InsPs are emerging as critical players in the integration of cellular metabolism and stress signaling in non-plant eukaryotes. Most eukaryotes synthesize the precursor molecule, myo-inositol (1,2,3,4,5,6)-hexakisphosphate (InsP6), which can serve as a signaling molecule or as storage compound of inositol, phosphorus, and minerals. Even though plants produce huge amounts of InsP6 in seeds, almost no attention has been paid to whether PPx-InsPs exist in plants, and if so, what roles these molecules play. This work details the presence of PPx-InsPs in plants and delineates two Arabidopsis gene products (AtVip1 and AtVip2) capable of PP-InsP5 synthesis. We further examined the subcellular location of enzymes connected to PPx-InsP synthesis as well as the developmental and tissue specific patterns of expression of the genes that encode these enzymes. We localized the enzymes involved in InsP6 and PPx-InsP production to the nucleus and endoplasmic reticulum (ER). The subcellular compartmentalization of PPx-InsP signaling may be unique to plants. An increased understanding in the pathways involved in energy sensing and metabolic response may reveal novel strategies to improve crops for yield and viability in the future.

Today, we spend various types of spaces in a day. It would be private spaces or public spaces. Public spaces have more variety function than private one. One type is for acting with public on purpose which is represented parks, public transportations, and libraries. The other is kind of waiting area, that is taken to go any public space. My project is about the creation of a familiar experience and mood in public spaces in order to enable people to feel more comfortable in unfamiliar public environments. I focus my explorations on waiting areas, like in airports or train stations, where being exposed to unfamiliar space could cause aggravation or even distress while one has to spend time in these unfamiliar spaces.  I try to transfer home feeling, one of the most private spaces, to the public spaces, by using sense of sight, hear, and smell, determinant of atmospheres in certain spaces. Among the sense, I more investigate light and shadows which are related with sense of sight. Unlike common used method in interior design, changing construction, colours, and so on, I try to make combination of real and unreal with light and shadows in one space.

This thesis deals with a more work-efficient and time-efficient method of calibration of standard glass scales, with practical use in the Czech Metrology Institute Regional Inspectorate in Brno. The desired streamlining of calibration were achieved in the use of a 3D coordinate measuring machine Micro-Vu Excel 4520. In the service software InSpec, six measuring programs were designed in the use of a standard glass scale brand SIP. The measurement uncertainties of this calibration were presented and calculated. This thesis draws up a draft proposal of the calibration procedure and drafts a formalized document of the calibration.

Inom geodetisk mätning har den tekniska utvecklingen inom terrester laserskanning (TLS) gått fort framåt sedan millennieskiftet. Tack vare dess framgång har TLS etablerat sig väl på marknaden som skanningsmetod och användningsområdena har därefter utvidgats. Leica Geosystems AB är ett internationellt mätningsteniskt företag, vilka har lanserat sin nya multistation Leica Nova MS60 med fältmjukvaran Leica Captivate. Till mjukvaran hör också applikationen Inspect a Surface, som möjliggör mätningsingenjör att kunna utvärdera erhållna skanningsdata ute i fält. Applikationen är ämnad att användas vid kontroll av ett skannat objekt genom att testa punktmolnet mot en definierad referens. Företaget har intresse av att en studie av denna karaktär utförs på deras produkter då det inte gjorts tidigare. Undersökningens primära syfte är att utröna i vilken grad fältmjukvaran Leica Captivate med applikationen Inspect a Surface i en Leica Nova MS60 står sig i jämförelse med traditionell efterbearbetning i extern mjukvara. Skanningar utfördes både i utom- och inomhusmiljö mot objekt som vanligen går att finna på byggarbetsplatser, såsom fasad, vägg, golv och cylindriska objekt med varierande radie och materialsegment. De horisontella och vertikala ytorna testades mot inmätta referenspunkter respektive interpolerade referenspunkter. Skanningsdata för cylindrar erhölls av MS60 respektive Leica Scan Station C10 för att visuellt undersöka fenomenet ”mixed pixels”. En cylinderinpassning i Leica Cyclone gjordes mot respektive segment för att se dess radie-avvikelse från referens och jämföra resultaten för respektive instrument. Även signifikanstest utfördes. Resultatet av undersökningen visar att utvärdering av ytor ger likvärdiga resultat i samtliga mjukvaror. Felkällan mixed pixels influerade i varierande grad för punktmolnen erhållet från MS60 och C10. Felkällan var som mest förekommande på materialet aluminium och tapetsegmentet som var närmast golvet. Cylinderpassning mot punktmoln från respektive instrument avvek på millimeternivå från varandra och referensen. Enstaka större avvikelser förekom bland annat på aluminiumsegmentet. Den slutsats som drogs var att applikationen är tillfredsställande att användas för efterbearbetning direkt ute i fält. Skanning av cylindrarna ledde till slutsatsen att skanningsdata från MS60 drabbas av felkällan mixed pixel i större grad än C10 bara om en yta är nära bakom cylindern. Samtidigt drabbades punktmolnet från C10 av mixed pixels om det var fritt bakom cylindrarna. Att använda sig av MS60 för skanning av cylindriska objekt visade sig vara tillfredsställande.
The technical development of terrestrial laser scanning has quickly proceeded in geodetic measurement since the millennium shift. Its success has made TLS well established on the market as a method of scanning and its field of applications has expanded. Leica Geosystems is an international company in the field of surveying and measurement instruments. They have developed a new multistation, Leica Nova MS60 with the field software Leica Captivate. In addition to the software the application Inspect a Surface allows surveyors to evaluate obtained point clouds in the field. The application serves the purpose of controlling an object by comparing a pointcloud with a defined reference. The company is interested of such a study to be made on their products, as it has not been conducted before. The main purpose of the study is to determine to what extent the field software Leica Captivate with Inspect a Surface in a Leica Nova MS60 manages with traditional post processing in external software. Scanning in indoor and outdoor environment was performed on objects common on construction sites, such asfacade, wall, floor, and cylindrical objects with varying radius and material segments. The horizontal and vertical surfaces were tested against measured reference points and interpolated reference points. Scanning data for cylinders was obtained by MS60 and Leica Scan Station C10 for a visual control of the phenomenon “mixed pixels”. In Leica Cyclone, a cylinder was fitted on each segment in order to determine radius deviation from reference and compare the results from each instrument. A hypothesis test was also conducted. The study shows that surfaces had equivalent results in all softwares. The error source mixed pixels influenced pointclouds from MS60 and C10 in varying degree. The error source was occurred most frequenlty on the aluminum and wallpaper segment which were close to the floor. Cylinder fitting on point cloud from, MS60 and C10 deviates millimeters from each other and the reference, apart from a few bigger deviations, the aluminum-segment for instance. The conclusion is that the application is satisfactory for post processing in the field. Scanning of cylinders leads to the conclusion thad mixed pixels occurs for MS60 more than C10 only if a surface is behind the cylinder. On the contrary, mixed pixels occur on C10 if there is free space behind the cylinder. Using MS60 for scanning of cylindrical object turns out to be satisfactory.

Detailed observations of geosynchronous satellites from earth are very limited. To better inspect these high altitude satellites, the use of small, refuelable satellites is proposed. The small satellites are stationed on a carrier platform in an orbit near the population of geosynchronous satellites. A carrier platform equipped with deployable, refuelable SmallSats is a viable option to inspect geosynchronous satellites. The propellant requirement to transfer to a targeted geosynchronous satellite, perform a proximity inspection mission, and transfer back to the carrier platform in a nearby orbit is determined. Convex optimization and traditional optimization techniques are explored, determining minimum propellant trajectories. Propellant is measured by the total required change in velocity, delta-v. The trajectories were modeled in a relative reference frame using the Clohessy-Wiltshire equations. Mass estimations for the carrier platform and the SmallSat were determined by using the rocket equation. The mass estimates were compared to the mass of a single, non-refuelable satellite performing the same geosynchronous satellite inspection missions. From the minimum delta-v trajectories and the mass analysis, it is determined that using refuelable SmallSats and a carrier platform in a nearby orbit can be more effcient than using a single non-refuelable satellite to perform multiple geosynchronous satellite inspections.

A significant portion of the pipeline infrastructure cannot be assessed for integrity using in-line inspection. There may also be reasons why some segments cannot be readily assessed by hydrostatic testing or by direct assessment. New standards and regulations for integrity management provide for using other technology that can be demonstrated to be as effective in assessing for the presence of integrity threats as the standard techniques. The first part of the report discusses a survey of PRCI member pipeline operating companies investigating the reasons why their systems are not piggable. The results are intended to be an aid to determining the direction of future PRCI research and development efforts. The results revealed that 62% of the pipe surveyed is presently piggable, and those members answering the survey plan to make another 21% of the system piggable. The survey further indicated that members plan to use direct assessment as the primary choice for inspecting nonpiggable segments. Consequently, the survey suggests that the primary goal of the Committee in addressing nonpiggable lines should be to validate direct assessment, followed by developing new above ground direct assessment and metal loss detection tools. The second part of the report discusses an analysis to establish suitable detection criteria for alternative assessment methods. Criteria for metal loss due to corrosion were established for 2 NPS to 24 NPS pipe of Grades B, X42, and X52 in all standard class locations so as to assure the same margin of safety afforded by the minimum hydrostatic test required per class. Criteria were also proposed for stress-corrosion cracking, selective corrosion in ERW seams, and mechanical damage.

Pipeline operators apply inspection methods and technologies for the purposes of assessing, or providing confidence in, the operational integrity of pipeline systems. In-line inspection (ILI), hydrostatic testing, and Direct Assessment (DA) methods are well-established and heavily relied upon by the industry; however, as additional technologies and methods continue to be developed and advanced, a structured process is needed for guidance in application of other methods. This document was developed for use by pipeline operators and service companies as a technology independent framework in the planning and execution of pipeline integrity assessments. This guidance presents requirements for any inspection process including qualification, validation, and development of a performance specification. It should be noted that these elements are required in application of any inspection method; in this regard, any established inspection method could be applied using the principles in this guideline. This guideline was thus created with many similar components as established standards and procedures for inspections such as API 1163 In-Line Inspection Systems Qualification and NACE SP-0502-2010 Pipeline External Corrosion Direct Assessment Methodology. This work was accomplished through collaboration with PRCI pipeline operating members to learn the steps each use to justify equivalent understanding of line pipe for other technologies. The criteria used to justify the established pipeline integrity technologies in their development (e.g. ILI, DA, RSTRENG) were also reviewed. This project reports the instances in the public record of pipeline operator notifications to PHMSA for application of other technologies and the criteria, both current and past, used to justify equivalent understanding.
