ABSTRACT We describe a genetic instability found in natural wine yeasts but not in the common laboratory strains of Saccharomyces cerevisiae. Spontaneous cyh2R /cyh2R mutants resistant to high levels of cycloheximide can be directly isolated from cyh2S /cyh2S wine yeasts. Heterozygous cyh2R/cyh2S hybrid clones vary in genetic instability as measured by loss of heterozygosity at cyh2. There were two main classes of hybrids. The lawn hybrids have high genetic instability and generally become cyh2R /cyh2R homozygotes and lose the killer phenotype under nonselective conditions. The papilla hybrids have a much lower rate of loss of heterozygosity and maintain the killer phenotype. The genetic instability in lawn hybrids is 3 to 5 orders of magnitude greater than the highest loss-of-heterozygosity rates previously reported. Molecular mechanisms such as DNA repair by break-induced replication might account for the asymmetrical loss of heterozygosity. This loss-of-heterozygosity phenomenon could be economically important if it causes sudden phenotype changes in industrial or pathogenic yeasts and of more basic importance to the degree that it influences the evolution of naturally occurring yeast populations.

Improved crop tolerance and weed suppressive ability are tactics that may reduce the negative effect of weeds on crop yield. Irrigated field experiments were conducted to compare leaf area index (LAI), intercepted photosynthetic photon flux (PPF), and relative tolerance and velvetleaf suppressive ability among two old (circa 1940) and two modern corn hybrids. Each hybrid was grown in monoculture and in mixture with velvetleaf at 1, 4, 16, and 40 plants m−1row. Plants were periodically harvested in monoculture plots to obtain estimates of corn LAI, and PPF interception was measured. Variation in hybrid tolerance to velvetleaf competition for light was evaluated by comparing among hybrids the coefficients of a regression of corn yield loss on velvetleaf density. Velvetleaf seed capsule production in the presence of each hybrid was compared to evaluate variation in velvetleaf suppressive ability among hybrids. Maximum corn yield loss was 32% lower for the two old hybrids, and velvetleaf capsule production was reduced by 62% at low velvetleaf densities in 1995 compared to the modern hybrids. In 1996, yield loss of the modern hybrid 3394 was 74% lower than that of the other three hybrids at low velvetleaf densities, whereas maximum yield loss of the old hybrid 336 was 44% lower at high densities. Velvetleaf capsule production did not vary among hybrids at any velvetleaf density in 1996. Hybrids with greater tolerance and velvetleaf suppressive ability also had greater LAI and PPF interception, suggesting optimized corn LAI and PPF interception may be useful in an integrated weed management program.

Abstract A growing body of evidence suggests that mutation rates exhibit intra-species specific variation. We estimated genome-wide loss of heterozygosity (LOH), gross chromosomal changes, and single nucleotide mutation rates to determine intra-species specific differences in hybrid and homozygous strains of Saccharomyces cerevisiae. The mutation accumulation lines of the S. cerevisiae hybrid backgrounds - S288c/YJM789 (S/Y) and S288c/RM11-1a (S/R) were analyzed along with the homozygous diploids RM11, S288c, and YJM145. LOH was extensive in both S/Y and S/R hybrid backgrounds. The S/Y background also showed longer LOH tracts, gross chromosomal changes, and aneuploidy. Short copy number aberrations were observed in the S/R background. LOH data from the S/Y and S/R hybrids were used to construct a LOH map for S288c to identify hotspots. Further, we observe up to a sixfold difference in single nucleotide mutation rates among the S. cerevisiae S/Y and S/R genetic backgrounds. Our results demonstrate LOH is common during mitotic divisions in S. cerevisiae hybrids and also highlight genome-wide differences in LOH patterns and rates of single nucleotide mutations between commonly used S. cerevisiae hybrid genetic backgrounds.

Tests were made on the water dynamics of water loss in maize at two locations at major maize production areas, Debrecen and Szeged, as a function of the heat sum required up to physiological ripening. The maximum water content was found to vary according to the hybrid and the year. No significant differences were found between the effective heat sums associated with maximum values within a year, the difference being 2-4 calendar days. There was a close correlation between the maximum grain water content and the length of the generative phase for a given hybrid in a given year. Better water supplies (1998) resulted in higher maximum water content values, which indirectly promoted longer and more intensive dry matter incorporation. Of all the environmental parameters, the drying down rate is primarily determined by the temperature. The changes in grain moisture content in a specific hybrid can be characterised by a joint linear graph. The drying down rate of the individual hybrids was determined by examining the b term of the linear equation.

This thesis sets to understand key limiting steps in organic solar cells (OSCs) and hybrid organic-inorganic light-emitting diodes (HyLEDs) and to provide routes to more environmentally friendly and lower cost devices. As well as studying organic optoelectronic devices, hybrid organic-inorganic approaches are also explored from both a fundamental point of view (quantum dot/organic solar cells) to a more technological one (air-stable, low cost light emitting diodes). The origin of the opencircuit voltage in bilayer solar cells is studied using transient optoelectronic techniques to probe the charge distribution in the active layer and the lifetime of charge carriers under working conditions. Furthermore, these techniques are applied to hybrid quantum dot/organic bilayer solar cells and provide a quantitative analysis of the non-geminate recombination in the device and its influence on the open-circuit voltage. The thesis also explores the use of water-soluble donors for OSCs and led to first true water-soluble donor.
Este trabajo de tesis proporciona las bases para el análisis detallado de los pasos limitantes del funcionamiento de celdas solares orgánicas (OSCs) y de los lightemitting diodos híbridos orgánico-inorgánicos (HyLED); además de procurar las rutas para la preparación de nuevos tipos de dispositivos económicos y seguros para el medio ambiente. El origen del voltaje de circuito abierto en celdas solares de doble capa ha sido estudiado mediante técnicas de optoelectrónica transientes para investigar la distribución de cargas en el estado activo y los tiempos de vida de las cargas bajo las condiciones de trabajo. Además, estas técnicas han sido aplicadas a celdas solares híbridas quantum dots/doble capa orgánica consiguiendo un análisis cuantitativo de la recombinación no-geminial en el dispositivo y su influencia sobre el voltaje de circuito abierto. La tesis también explora el uso de donantes solubles en agua para OSCs y llevó al primer verdadero donante soluble en agua.

Defining an operation strategy for a Split Parallel Architecture (SPA) Hybrid Electric Vehicle (HEV) is accomplished through calculating powertrain component losses. The results of these calculations define how the vehicle can decrease fuel consumption while maintaining low vehicle emissions. For a HEV, simply operating the vehicle's engine in its regions of high efficiency does not guarantee the most efficient vehicle operation. The results presented are meant only to define a literal strategy; that is, an understanding as to why the vehicle should operate in a certain way under the given conditions. The literature review gives a background of hybrid vehicle control publications, and without the SPA HEV addressed or a hybrid analysis based on loss calculations between engine only and hybrid modes, there is a need for this paper. Once the REVLSE architecture and components are understood, the hybrid modes are explained. Then the losses for each hybrid mode are calculated, and both the conversion and assist efficiencies are detailed. The conversion efficiency represents the amount of additional fuel required to store a certain amount of energy in the battery, and this marginal efficiency can be higher than peak engine efficiency itself. This allows electric only propulsion to be evaluated against the engine only mode, and at low torques the electric motor is more efficient despite the roundtrip losses of the hybrid system.

Multiview video is one of the emerging research areas especially among the video coding community. Transmission of multiview video over an error prone network is possible with efficient compression of these videos. But along with the studies for efficiently compressing the multiview video, new error concealment and error protection methods are also necessary to overcome the problems due to erroneous channel conditions in practical applications. In packet switching networks, packet losses may lead to block losses in a frame or the loss of an entire frame in an encoded video sequence. In recent years several algorithms are proposed to handle the loss of an entire frame efficiently. However methods for full frame losses in stereoscopic or multiview videos are limited in the literature. In this thesis a stereoscopic approach for full frame loss concealment of multiview video is proposed. In the proposed methods, the redundancy and disparity between the views and motion information between the previously decoded frames are used to estimate the lost frame. Even though multiview video can be composed of more than two views, at most three view are utilized for concealment. The performance of the proposed algorithms are tested against monoscopic methods and the conditions under which the proposed methods are superior are investigated. The proposed algorithms are applied to both stereoscopic and multiview video.

Robust ion-exchanged glass waveguides exhibit low optical losses in a broad spectral range and they allow integration of several devices on the same chip due to their planar structure. Consequently, they can be a low cost alternative to semiconductors for fabricating various integrated optical devices. Two high performance photonic devices were designed and realized, demonstrating the potential of glass waveguides. The well-controlled silver-film ion-exchange process allowed the fabrication of: i) a highly sensitive biosensor based on optical absorption and, ii) a low loss hybrid electro-optic (EO) polymer modulator with a narrow coplanar electrode gap. The single-mode, channel integrated optical ion-exchange waveguide on borosilicate glass (Corning 0211) is described for broad spectral band (400-650 nm) detection and analysis of heme-containing protein films at a glass/water interface. The evanescent wave interaction is improved significantly by fabricating ion-exchange waveguides with a step-like index profile. Silver nano-particle formation is reduced in order to achieve low loss in the Soret-band (~400 nm). Unlike other surface-specific techniques (e.g. SPR, interferometry) that probe local refractive-index changes and therefore are susceptible to temperature fluctuations, the integrated optical waveguide absorption technique probes molecular-specific transition bands and is expected to be less vulnerable to environmental perturbations. The hybrid integration of phosphate glass (IOG-1) and EO polymer is realized for the first time. The critical alignment steps which are typically required for hybrid optoelectronic devices are eliminated with a simple alignment-free fabrication technique. The low loss adiabatic transition from glass to EO polymer waveguide is enabled by gray scale patterning of the novel EO polymer, AJLY. Total insertion loss of 5 dB and electrode gap of 8 μm is obtained for an optimized device design. EO polymer poling at 135 ºC and 75 V/μm is enabled by the sol-gel buffer layer.

This collection reveals how, in Manchukuo (1932-1945), literature both furthered national aims while contesting them, as writers of varied ethnicities engaged in multivalent strategies to continue cultural production amidst difficult political circumstances. Studies of their work by transnational scholars today demonstrate that these writers faced factors influencing outcomes of their production, such as censorship, the Japanese puppet regime's propaganda aims, and even the market. In addition, particular hybrid language practices emerged, with writers engaging in transnational practices in a border region. This volume examines what we call "Manchukuo perspectives" unique to cultural producers in a state transformed by Japanese interests, but later shaped by more inclusive multivalent aims, reflected in the writings of Chinese, Korean, and Russian intellectuals who felt a keen loss of nation, which also included Japanese converted leftists who transformed their antipathy towards imperialist capitalism into support for a fascist state offering the utopian promises of a "right-wing proletarianism".

The key features of glomerular diseases—haematuria, proteinuria, loss of glomerular filtration rate, and hypertension—were recognized in the nineteenth century, and some earlier, but Richard Bright is usually given credit for synthesizing the concepts of renal disease, and glomerulonephritis came under the heading of Bright’s disease for almost a century. Separation into different types was based on first clinical syndromes, but in the early twentieth century, pathological description was improving and with the introduction of percutaneous renal biopsies in the 1950s, in the 1960s histopathological definitions assumed the ascendancy. A unifying classification of glomerular disease remains work in progress. Current classifications are pathologically based but increasingly include the results of other investigations (including genotype and a variety of immunological and other tests). This chapter follows this pragmatic, hybrid approach, categorizing glomerular disease by pattern on renal biopsy except where aetiological factors are clearly identified (e.g. HIV nephropathy), or associated multisystem disease is defined (e.g. lupus nephritis), or the immunopathogenesis is well characterized (e.g. antiglomerular basement membrane disease).

Chapter 5, “Bringing Back the Dead: Shakespeare’s Alcestis,” argues that, after incorporating Greek tragic women into comedies, Shakespeare increasingly drew on these figures to merge tragic and comic structures in plays featuring miraculous recoveries from apparent deaths. Plays such as Much Ado About Nothing, Pericles, and The Winter’s Tale not only dramatize women’s miraculous return to life from apparent death, but also link these recoveries with the performance of female lament, which elicits sympathies and melts audiences into supportive alliances. Drawing on sources shaped by Greek texts, these plays reconfigure tragedy with a happy ending, a hybrid genre identified with Euripides. In particular, they recreate the ending of Alcestis, in which a grieving man encounters a veiled woman who is eventually revealed to be his lost wife returned from death.

Greece developed a pension-heavy, clientelist, hybrid Mediterranean welfare state with many gaps in coverage. The global financial crisis of 2008 triggered a severe sovereign debt crisis, compelling the country to accept three bailout packages with stringent conditions as to spending cuts, privatization, and openness to international competition. Severe austerity has caused a protracted recession: the economy lost more than a quarter of its GDP between 2008 and 2015. The Mediterranean refugee crisis impacted severely on the country. New parties of the extreme left (SYRIZA) and extreme right (Golden Dawn) have gained support. SYRIZA was elected on an anti-austerity platform but failed to deliver and a fourth rescue package is under negotiation. The more likely future direction consists in an ever-tighter austerity programme with the immizeration of large sections of the population. A move towards neo-Keynesian intervention and social investment seems unlikely, given the level of debt and the bailout conditions.

This chapter introduces the “biohybrid systems” section of the Handbook of Living Machines and briefly reviews some important examples of systems formed by coupling biological to engineered components. These include brain–machine interfaces, both non-invasive, using different external measurement and scanning devices, and invasive approaches focusing on implantable probes. Next we consider fabrication methods for micro- and nanobiohybrid systems and an example of a biohybrid system at the organism level, in the form of a robot–animal biohybrid, developed using methods from synthetic biology. There are many application for biohybrid systems in healthcare: we include exemplar chapters describing intelligent prostheses such as artificial hands with tactile sensing capabilities, sensory organ–chip hybrids in the form of cochlear implants, and artificial implants designed to replace damaged neural tissue and restore lost memory function.

AbstractFreeze–thaw damage in cold areas is an important reason for the deterioration of durability of concrete structures, and the addition of fiber can effectively improve the frost resistance of concrete. To study the effect of steel fiber and polyvinyl alcohol (PVA) fiber on the frost resistance of concrete, an orthogonal experimental method was applied to analyze the frost resistance of steel-PVA hybrid fiber concrete. The effect of the steel fiber content, PVA fiber content, and steel fiber types were investigated on the mass-loss rate and relative dynamic elastic modulus of fiber concrete, to explore the mechanism of high frost resistance of it. The results show that steel fiber content has a significant effect on the mass-loss rate and relative dynamic elastic modulus of fiber concrete. The influence of PVA fiber content on the relative dynamic elastic modulus is relatively great, and the influence of steel fiber types of the mass-loss rate is the same. The frost resistance of concrete can be improved by adding hybrid fiber in concrete. When 1.0% End-hook steel fibers and 0.3% PVA fibers are added, the concrete specimen has the best frost resistance.

Barbados, through its Barbados National Energy Policy (BNEP) 2019-2030, announced its commitment to achieving 100 percent renewable energy and carbon neutrality by 2030. This commitment creates an opportunity for the GoB to manage the impact of the transition toward renewable clean energy by introducing measures to transform the way revenue from energy is collected thereby avoiding unnecessary fiscal costs. The purpose of this study is to calculate the revenue gap derived from Barbados 2030 energy transition goal of having a revenue-neutral transition and propose and evaluate various policy measures that could help seize opportunities to close that gap. The simulation model suggests that the energy transition would result in an estimated BBD$105 million in revenue losses a year by following the BNEP. Such a reduction would create a significant fiscal gap that would need to be addressed through the introduction of new forms of taxes or changes to current taxes in order to adapt tax collection to revenue creation from the new clean energy economy. A wide range of tax policy options and issues surrounding their effective implementation were discussed such as: increased taxes on fossil fuels, a change in the VAT rate, mileage taxes on electric and hybrid vehicles, and taxes on renewable energy production. Each of these new tax approaches can help address the fiscal gap estimated above.

Research activities in both our laboratories were directed toward development of control of the FLP/frt recombination system for plants. As described in the text of the research proposal, the US lab has been engaged in developing regulatory strategies such as tissue-specific promoters and the steroid-inducible activation of the FLP enzyme while the main research activities in Israel have been directed toward the development and testing of a copper-regulated expression of flp recombinase in tobacco (this is an example of a promoter activation by metal ions). The Israeli lab hat additionally completed experiments of previous studies regarding factors affecting the efficiency of recombinase activity using both a gain-of-function assay (excisional-activation of a gusA marker) and loss of function assay (excision of a rolC marker) in tobacco. Site-specific recombinase systems, in particular the FLP/frt and R/RS systems of yeast and the Cre/lox system of bacteriophage P1, have become an essential component of targeted genetic transformation procedures both in animal and plant organisms. To provide more flexibility in transgene excisions by the recombinase systems as well as gene targeting, and to widen possible applications, the development of controlled or regulated recombination systems is highly desirable and was therefore the subject of this research proposal. There are a few possible mechanisms to regulate expression of a recombinase system. They include: 1) control of the recombination system by having the target sites (e.g. frt) in one plant and the flp recombinase gene in another, and bringing the two together by cross fertilization. 2) regulation of promoter activities by external stimuli such as temperature, chemicals, metal ions, etc. 3) regulation of promoter activities by internal signals, i.e. cell- or tissue-specific, or developmental regulation. 4) regulation of enzyme activity by providing cofactors essential for biochemical reactions to take place such as steroid molecules in conjunction with a steroid ligand-binding protein (domains). During the course of this research our major emphasis have been focused toward studying the feasibility of hybrid seed production in Arabidopsis, using FLP/frt. Male-sterility was induced using the antisence of a pollen- and tapetum-specific gene, bcp1, isolated from Arabidopsis. The sterility inducing gene was flanked by frt sites. Upon cross pollination of flowers of male-sterile plants with pollen from FLP-containing plants, viable seeds were produced, and the progeny hybrid plants developed normally. The major achievement from this work is the first demonstration of using a site-specific recombinase to restore fertility in male-sterile plants (see attached paper, Luo et al., Plant J 2000; 23:423-430). The implication from this finding is that site-specific recombination systems can be applied in crop plants as a useful alternative method for hybrid seed production.

Infectious disease is one of the most serious causes of economic loss in all sectors of aquaculture. There is a critical need to understand the molecular basis for protection against infectious disease so that safer, more reliable and more cost-effective strategies can be designed for their control. As part of this effort, the major goal of our BARD project was to determine the importance of endobiotics as a defense against protozoan ectoparasites in fish. Endobiotics, or antimicrobial polypeptides, are peptides and small proteins that are increasingly recognized as having a vital role in the innate defense of virtually all animals. One objective of our BARD project was to determine the antiparasitic potency of one specific group of endobiotics that were isolated from hybrid striped bass (Morone saxatilis x M chrysops). We found that these endobiotics, which we had previously named histone-like proteins (HLPs), exhibited potent activity against Amyloodinium and that the putative levels of HLPs in the skin were well within the levels that we found to be lethal to the parasite in vitro. We also found evidence for the presence of similar antibiotics in sea bream (Sparus aurata) and Mediterranean sea bass (Dicentrarchus labrax). We also examined the effect of chronic stress on the expression of HLP in fish and found that HLP levels were dramatically decreased after only one week of a crowding/high ammonia sublethal stress. We also began to explore the feasibility of upregulating endobiotics via immunostimulation. However, we did not pursue this objective as fully as we originally intended because we spent a much larger effort than originally anticipated on the last objective, the attempted isolation of novel endobiotics from hybrid striped bass. In this regard, we purified and identified four new peptide endobiotics. These endobiotics, which we have named piscidins (from "Pisces" meaning fish), have potent, broad-spectrum activity against a number of both fish and human pathogens. This includes not only parasites but also bacteria. We also demonstrated that these peptides are present in the mast cell. This was the first time that the mast cell, the most common tissue granulocyte in vertebrates, was shown to possess any type of endobiotic. This finding has important implications in explaining the possible function of mast cells in the immune response of vertebrates. In summary, the research we have accomplished in this BARD project has demonstrated that endobiotics in fish have potent activity against many serious pathogens in aquaculture and that there is considerable potential to use these compounds as stress indicators in aquaculture. There is also considerable potential to use some of these compounds in other areas of medicine, including treatment of serious infectious diseases of humans and animals.

Potyviruses form one of the largest and most economically important groups of plant viruses. Individual potyviruses and their isolates vary in symptom expression, host range, and ability to overcome host resistance genes. Understanding factors influencing these biological characteristics is of agricultural importance for epidemiology and deployment of resistance strategies. Cucurbit crops are subject to severe losses by several potyviruses including the highly aggressive and variable zucchini yellow mosaic virus (ZYMV). In this project we sought to investigate protein domains in ZYMV that influence systemic infection and host range. Particular emphasis was on coat protein (CP), because of known functions in both cell to cell and long distance movement, and helper component-protease (HC-Pro), which has been implicated to play a role in symptom development and long distance movement. These two genes are also essential for aphid mediated transmission, and domains that influence disease development may also influence transmissibility. The objectives of the approved BARD project were to test roles of specific domains in the CP and HC-Pro by making sequence alterations or switches between different isolates and viruses, and testing for infectivity, host range, and aphid transmissibility. These objectives were largely achieved as described below. Finally, we also initiated new research to identify host factors interacting with potyviral proteins and demonstrated interaction between the ZYMV RNA dependent RNA polymerase and host poly-(A)-binding protein (Wang et al., in press). The focus of the CP studies (MSU) was to investigate the role of the highly variable amino terminus (NT) in host range determination and systemic infection. Hybrid ZYMV infectious clones were produced by substituting the CP-NT of ZYMV with either the CP-NT from watermelon mosaic virus (overlapping, but broader host range) or tobacco etch virus (TEV) (non- overlapping host range) (Grumet et al., 2000; Ullah ct al., in prep). Although both hybrid viruses initially established systemic infection, indicating that even the non-cucurbit adapted TEV CP-NT could facilitate long distance transport in cucurbits, after approximately 4-6, the plants inoculated with the TEV-CPNT hybrid exhibited a distinct recovery of reduced symptoms, virus titer, and virus specific protection against secondary infection. These results suggest that the plant recognizes the presence of the TEV CP-NT, which has not been adapted to infection of cucurbits, and initiates defense responses. The CP-NT also appears to play a role in naturally occurring resistance conferred by the zym locus in the cucumber line 'Dina-1'. Patterns of virus accumulation indicated that expression of resistance is developmentally controlled and is due to a block in virus movement. Switches between the core and NT domains of ZYMV-NAA (does not cause veinal chlorosis on 'Dina-1'), and ZYMV-Ct (causes veinal chlorosis), indicated that the resistance response likely involves interaction with the CP-NT (Ullah and Grumet, submitted). At the Volcani Center the main thrust was to identify domains in the HC-Pro that affect symptom expression or aphid transmissibility. From the data reported in the first and second year report and in the attached publications (Peng et al. 1998; Kadouri et al. 1998; Raccah et al. 2000: it was shown that: 1. The mutation from PTK to PAK resulted in milder symptoms of the virus on squash, 2. Two mutations, PAK and ATK, resulted in total loss of helper activity, 3. It was established for the first time that the PTK domain is involved in binding of the HC-Pro to the potyvirus particle, and 4. Some of these experiments required greater amount of HC-Pro, therefore a simpler and more efficient purification method was developed based on Ni2+ resin.

The COP9 signalosome (CSN) is an eight-subunit protein complex that is highly conserved among eukaryotes. Genetic analysis of the signalosome in the plant model species Arabidopsis thaliana has shown that the signalosome is a repressor of light dependent seedling development as mutant Arabidopsis seedlings that lack this complex develop in complete darkness as if exposed to light. These mutant plants die following the seedling stage, even when exposed to light, indicating that the COP9 signalosome also has a central role in the regulation of normal photomorphogenic development. The biochemical mode of action of the signalosome and its position in eukaryotic cell signaling pathways is a matter of controversy and ongoing investigation, and recent results place the CSN at the juncture of kinase signaling pathways and ubiquitin-mediated protein degradation. We have shown that one of the many CSN functions may relate to the regulation of translation through the interaction of the CSN with its related complex, eukaryotic initiation factor (eIF3). While we have established a physical connection between eIF3 subunits and CSN subunits, the physiological and developmental significance of this interaction is still unknown. In an effort to understand the biochemical activity of the signalosome, and its role in regulating translation, we originally proposed to dissect the contribution of "h" subunit of eIF3 (eIF3h) along the following specific aims: (i) Isolation and phenotypic characterization of an Arabidopsis loss-of-function allele for eIF3h from insertional mutagenesis libraries; (ii) Creation of designed gain and loss of function alleles for eIF3h on the basis of its nucleocytoplasmic distribution and its yeast-two-hybrid interactions with other eIF3 and signalosome partner proteins; (iii) Determining the contribution of eIF3h and its interaction with the signalosome by expressing specific mutants of eIF3h in the eIF3h- loss-of function background. During the course of the research, these goals were modified to include examining the genetic interaction between csn and eif3h mutations. More importantly, we extended our effort toward the genetic analysis of mutations in the eIF3e subunit, which also interacts with the CSN. Through the course of this research program we have made several critical scientific discoveries, all concerned with the apparent diametrically opposed roles of eIF3h and eIF3e. We showed that: 1) While eIF3e is essential for growth and development, eIF3h is not essential for growth or basal translation; 2) While eIF3e has a negative role in translational regulation, eIF3h is positively required for efficient translation of transcripts with complex 5' UTR sequences; 3) Over-accumulation of eIF3e and loss-of-function of eIF3h both lead to cop phenotypes in dark-grown seedlings. These results were published in one publication (Kim et al., Plant Cell 2004) and in a second manuscript currently in revision for Embo J. Are results have led to a paradigm shift in translation research – eIF3 is now viewed in all systems as a dynamic entity that contains regulatory subuits that affect translational efficiency. In the long-term agronomic outlook, the proposed research has implications that may be far reaching. Many important plant processes, including developmental and physiological responses to light, abiotic stress, photosynthate, and hormones operate in part by modulating protein translation [23, 24, 40, 75]. Translational regulation is slowly coming of age as a mechanism for regulating foreign gene expression in plants, beginning with translational enhancers [84, 85] and more recently, coordinating the expression of multiple transgenes using internal ribosome entry sites. Our contribution to understanding the molecular mode of action of a protein complex as fundamental as eIF3 is likely to lead to advances that will be applicable in the foreseeable future.

• Renewable energy (RE) projects in Indonesia usually have IRR between 10% and 15% and PP around 6 to 30 years • Attractive return usually could be found in large scale RE projects, although there are numerous other factors involved including technology developments, capacity scale, power purchasing price agreements, project locations, as well as interest rates and applied incentives. • Crowdfunding (CF) has big potential to contribute to the financing of RE projects especially financing small scale RE projects. • P2P lending usually targeted short-term loans with high interest rates. Therefore, it cannot be employed as an alternative financing for RE projects in Indonesia. • Three types of CF that can be employed as an alternative for RE project funding in Indonesia. Namely, securities, reward, and donation-based CF. In addition, hybrid models such as securities-reward and reward-donation could also be explored according to the project profitability. • Several benefits offer by securities crowdfunding (SCF) compared to conventional banking and P2P lending, as follows: (1) issuer do not need to pledge assets as collateral; (2) do not require to pay instalment each month; (3) issuer share risks with investors with no obligation to cover the investor’s loss; (4) applicable for micro, small, medium, enterprises (MSMEs) with no complex requirements; and (5) there is possibility to attract investors with bring specific value. • Several challenges that need to be tackled such as the uncertainty of RE regulations; (1) issuer’s inability in managing the system and business; (2) the absence of third parties in bridging between CF platform and potential issuer from RE project owner; (3) the lack of financial literacy of the potential funders; and (4) lastly the inadequacy of study regarding potential funders in escalating the RE utilisation in Indonesia.

Tomato yellow leaf curl geminivirus (TYLCV) is a major pathogen of cultivated tomato, causing up to 100% crop loss in many parts of the world. In Israel, where TYLCV epidemics have been recorded since the 1960' s, this viral disease is well known and has been of economic significance ever since. In recent years, TYLCV outbreaks also occurred in the "New World" - Cuba, The Dominican Republic, and in the USA, in Florida, Georgia and Louisiana. Thus, TYLCV substantially hinders tomato growth throughout the world. Surprisingly, however, little is known about the molecular mechanisms of TYLCV interaction with the host tomato cells. The present proposal, a continuation of the project supported by BARD from 1994, expanded our understanding of the molecular mechanisms by which TYLCV enters the host cell nucleus for replication and transcription and exits it for the subsequent cell-to-cell spread. Our project sought two objectives: I. To study the roles of the viral capsid protein (CP) and host cell factors in TYLCV nuclear import. II. To study the roles of CP and host cell factors in TYLCV nuclear export. Our research toward these goals have produced the following major achievements: . Developed a one-hybrid assay for protein nuclear export and import (#3 in the List of Publications). . Identified a functional nuclear export signal (NES) in the capsid protein (CP) of TYLCV (#3 in the List of Publications). . Discovered homotypic interactions between intact TYLCV CP molecules and analyzed these interactions using deletion mutagenesis of TYLCV CP (#5 in the List of Publications). . Showed developmental and tissue-specific expression of the host factor required for nuclear import of TYLCV CP, tomato karyopherin alpha 1, in transgenic tomato plants (#14 in the List of Publications). . By analogy to nuclear import of TYLCV ,identified an Arabidopsis VIPI protein that participates in nuclear import of Agrobacterium T -complexes via the karyopherin alpha pathway (#4,6, and 8 in the List of Publications). These research findings provided significant insights into (i) the molecular pathway of TYLCV entry into the host cell nucleus, and (ii) the mechanism by which TYLCV is exported from the nucleus for the cell-to-cell spread of infection. Furthermore, the obtained knowledge will help to develop specific strategies to attenuate TYLCV infection, for example, by blocking viral entry into and/or exit out of the host cell nucleus. Also, as much of our findings is relevant to all geminiviruses, new anti- TYLCV approaches developed based on the results of our research will be useful to combat other members of the Geminivirus family. Finally, in addition to the study of TYLCV nuclear import and export, our research contributed to our understanding of general mechanisms for nucleocytoplasmic shuttling of proteins and nucleic acids in plant cells.

Original objectives and revisions-The proposed research included these original objectives: field validation of diagnostic tests (PCR), the development and evaluation of new sensitive tools (LC-PCR/TaqManPCR, antibody detection by ELISA) including their use to study the ecology and the epidemiology of KHV (virus distribution in the environment and native cyprinids) and the carrier status of fish exposed experimentally or naturally to KHV (sites of virus replication and potential persistence or latency). In the course of the study we completed the genome sequence of KHV and developed a DNA array to study the expression of KHV genes in different conditions. Background to the topics-Mass mortality of koi or common carp has been observed in Israel, USA, Europe and Asia. These outbreaks have reduced exports of koi from Israel and have created fear about production, import, and movements of koi and have raised concerns about potential impacts on native cyprinid populations in the U.S.A. Major conclusions-A suite of new diagnostic tools was developed that included 3 PCR assays for detection of KHV DNA in cell culture and fish tissues and an ELISA assay capable of detecting anti-KHV antibodies in the serum of koi and common carp. The TKPCR assay developed during the grant has become an internationally accepted gold standard for detection of viral DNA. Additionally, the ELISA developed for detecting serum anti-KHV antibodies is now in wide use as a major nonlethal screening tool for evaluating virus status of koi and common carp populations. Real time PCR assays have been able to detect viral DNA in the internal organs of survivors of natural and wild type vaccine exposures at 1 and 10³ genome equivalents at 7 months after exposure. In addition, vaccinated fish were able to transmit the virus to naive fish. Potential control utilizing hybrids of goldfish and common carp for production demonstrated they were considerably more resistant than pure common carp or koi to both KHV (CyHV-3). There was no evidence that goldfish or other tested endemic cyprinids species were susceptible to KHV. The complete genomic sequencing of 3 strains from Japan, the USA, and Israel revealed a 295 kbp genome containing a 22 kbp terminal direct repeat encoding clear gene homologs to other fish herpesviruses in the family Herpesviridae. The genome encodes156 unique protein-coding genes, eight of which are duplicated in the terminal repeat. Four to seven genes are fragmented and the loss of these genes may be associated with the high virulence of the virus. Viral gene expression was studies by a newly developed chip which has allowed verification of transcription of most all hypothetical genes (ORFs) as well as their kinetics. Implications, both scientific and agricultural- The results from this study have immediate application for the control and management of KHV. The proposal provides elements key to disease management with improved diagnostic tools. Studies on the ecology of the virus also provide insights into management of the virus at the farms that farmers will be able to apply immediately to reduce risks of infections. Lastly, critical issues that surround present procedures used to create “resistant fish” must be be resolved (e.g. carriers, risks, etc.). Currently stamping out may be effective in eradicating the disease. The emerging disease caused by KHV continues to spread. With the economic importance of koi and carp and the vast international movements of koi for the hobby, this disease has the potential for even further spread. The results from our studies form a critical component of a comprehensive program to curtail this emerging pathogen at the local, regional and international levels.
