Résumé :
Abstract Tactical ad-hoc networks are evolving today towards complex heterogeneous networks in terms of architecture, protocols and security. Due to the difference in network resources and reliability, end-to-end quality of service provisioning becomes very challenging. If we also take into account communication issues such as unpredictable connectivity, preferential forwarding for special traffic classes, intermittency due to node or communication link failure, the problem is further aggravated. In this article, we examine the major challenges that must be solved in order to provide efficient QoS provisioning in the heterogeneous network. Finally we describe QoS-aware mechanisms for inter-domain and intra-domain heterogeneous networks, also including real-time services provision in highly mobile environments.

Résumé :
Cyber physical systems (CPS's) are a kind of complex system with highly integrated interaction between computing resources and physical environment in a network environment. There are some challenges in modeling and simulation of heterogeneous CPS due to its hybrid and heterogenous characteristics. To address the issue, we propose an executable domain specific modeling language for co-simulation (DSML4CS) to model the co-simulation of CPS. According to the construction method of domain modeling language, we present the abstract syntax, concrete syntax and operational semantics of DSML4CS. We also propose a flexible co-simulation mechanism, which supports partial step revision of specific co-simulation process with the state event fault-tolerant mechanism. The co-simulation language for heterogeneous CPS is implemented based on the GEMOC platform. Our aim is to provide the co-simulation service in CPS. The usability of DSML4CS is illustrated with a case study of a temperature control system in an energy-aware building.

Résumé :
SummaryThe granule cell domain of the cochlear nuclear complex contains interneurons, which are the targets for nonprimary auditory inputs from the superior olivary complex, inferior colliculus, auditory cortex, cuneate and trigeminal nuclei of the somatosensory system. The cellular targets of the non-primary projections are unknown due to a lack of information regarding postsynaptic profiles in the granule cell areas. In the present paper, we examined the synaptic relationships between a heterogeneous class of large synaptic terminals, called mossy fibers and their targets within subdivisions of the granule cell domain. During the late stage of postnatal development, we observed heterogenous groups of complex synaptic glomeruli. Using electron microscopy, we provide evidence for ultrastructural features of dendrites that receive input from the mossy fibers. The distinct synaptic relations between mossy fibers and dendrites of microneurons further imply fundamentally separate roles in processing of acoustic signals.

Résumé :
Knowledge graph-based data integration is a practical methodology for heterogeneous legacy database-integrated service construction. However, it is neither efficient nor economical to build a new cross-domain knowledge graph on top of the schemas of each legacy database for the specific integration application rather than reusing the existing high-quality knowledge graphs. Consequently, a question arises as to whether the existing knowledge graph is compatible with cross-domain queries and with heterogenous schemas of the legacy systems. An effective criterion is urgently needed in order to evaluate such compatibility as it limits the quality upbound of the integration. This research studies the semantic similarity of the schemas from the aspect of properties. It provides a set of in-depth criteria, namely coverage and flexibility, to evaluate the pairwise compatibility between the schemas. It takes advantage of the properties of knowledge graphs to evaluate the overlaps between schemas and defines the weights of entity types in order to perform precise compatibility computation. The effectiveness of the criteria obtained to evaluate the compatibility between knowledge graphs and cross-domain queries is demonstrated using a case study.

Résumé :
AbstractBecause many networked systems can only be characterized with fractional-order dynamics in complex environments, fractional-order calculus has been studied deeply recently. When diverse individual features are shown in different agents of networked systems, heterogeneous fractional-order dynamics will be used to describe the complex systems. Based on the distinguishing properties of agents, heterogeneous fractional-order multi-agent systems (FOMAS) are presented. With the supposition of multiple leader agents in FOMAS, distributed containment control of FOMAS is studied in directed weighted topologies. By applying Laplace transformation and frequency domain theory of the fractional-order operator, an upper bound of delays is obtained to ensure containment consensus of delayed heterogenous FOMAS. Consensus results of delayed FOMAS in this paper can be extended to systems with integer-order models. Finally, numerical examples are used to verify our results.

Résumé :
We consider a variant of the mean-field model of coupled phase oscillators with uniform distribution of natural frequencies. By establishing correlations between the quenched disorder of intrinsic frequencies and coupling strength with both in- and out-coupling heterogeneities, we reveal a generic criterion for the onset of partial locking that takes place in a domain with the coexistence of phase-locked oscillators and drifters. The critical points manifesting the instability of the stationary states are obtained analytically. In particular, the bifurcation mechanism of the equilibrium states is uncovered by the use of frequency-dependent version of the Ott–Antonsen reduction consistently with the analysis based on the self-consistent approach. We demonstrate that both the manner of coupling heterogeneity and correlation exponent have influence on the emergent patterns of partial locking. Our research could find applicability in better understanding the phase transitions and related collective phenomena involving synchronization control in networked systems.

Résumé :
New 5 G and beyond services demand innovative solutions in optical transport to increase efficiency and flexibility and reduce capital (CAPEX) and operational (OPEX) expenditures to support heterogeneous and dynamic traffic. In this context, optical point-to-multipoint (P2MP) connectivity is seen as an alternative to provide connectivity to multiple sites from a single source, thus potentially both reducing CAPEX and OPEX. Digital subcarrier multiplexing (DSCM) has been shown as a feasible candidate for optical P2MP in view of its ability to generate multiple subcarriers (SC) in the frequency domain that can be used to serve several destinations. This paper proposes a different technology, named optical constellation slicing (OCS), that enables a source to communicate with multiple destinations by focusing on the time domain. OCS is described in detail and compared to DSCM by simulation, where the results show that both OCS and DSCM provide a good performance in terms of the bit error rate (BER) for access/metro applications. An exhaustive quantitative study is afterwards carried out to compare OCS and DSCM considering its support to dynamic packet layer P2P traffic only and mixed P2P and P2MP traffic; throughput, efficiency, and cost are used here as the metrics. As a baseline for comparison, the traditional optical P2P solution is also considered in this study. Numerical results show that OCS and DSCM provide a better efficiency and cost savings than traditional optical P2P connectivity. For P2P only traffic, OCS and DSCM are utmost 14.6% more efficient than the traditional lightpath solution, whereas for heterogeneous P2P + P2MP traffic, a 25% efficiency improvement is achieved, making OCS 12% more efficient than DSCM. Interestingly, the results show that for P2P only traffic, DSCM provides more savings of up to 12% than OCS, whereas for heterogeneous traffic, OCS can save up to 24.6% more than DSCM.

Résumé :
General practitioners (GPs) are increasingly expected to provide palliative care as ageing populations put pressure on specialist services. Some GPs, however, cite barriers to providing this care including prognostication challenges and lack of confidence. Palliative care content within clinical practice guidelines might serve as an opportunistic source of informational support to GPs. This review analysed palliative care content within Australian guidelines for life-limiting conditions to determine the extent to which it might satisfy GPs’ stated information needs and support them to provide quality end-of-life care. Six databases and guideline repositories were searched (2011–2018). Eligible guidelines were those for a GP audience and explicitly based on an appraisal of all available evidence. Content was mapped against an established palliative care domain framework (PEPSI-COLA) and quality was assessed using AGREE-II. The nine guidelines meeting inclusion criteria were heterogenous in scope and depth of palliative care domain coverage. The ‘communication’ needs domain was best addressed while patient physical and emotional needs were variably covered. Spiritual, out-of-hours, terminal care and aftercare content was scant. Few guidelines addressed areas GPs are known to find challenging or acknowledged useful decision-support tools. A template covering important domains might reduce content variability across guidelines.

Résumé :
Pressure vessels (PV) are designed to hold liquids, gases, or vapors at high pressures in various industries, but a ruptured pressure vessel can be incredibly dangerous if cracks are not detected in the early stage. This paper proposes a robust crack identification technique for pressure vessels using genetic algorithm (GA)-based feature selection and a deep neural network (DNN) in an acoustic emission (AE) examination. First, hybrid features are extracted from multiple AE sensors that represent diverse symptoms of pressure vessel faults. These features stem from various signal processing domains, such as the time domain, frequency domain, and time-frequency domain. Heterogenous features from various channels ensure a robust feature extraction process but are high-dimensional, so may contain irrelevant and redundant features. This can cause a degraded classification performance. Therefore, we use GA with a new objective function to select the most discriminant features that are highly effective for the DNN classifier when identifying crack types. The potency of the proposed method (GA + DNN) is demonstrated using AE data obtained from a self-designed pressure vessel. The experimental results indicate that the proposed method is highly effective at selecting discriminant features. These features are used as the input of the DNN classifier, achieving a 94.67% classification accuracy.

Résumé :
Abstract. By extracting bound water from the soil and lifting it to the canopy, root systems of vegetation perform work. Here we describe how the energetics involved in root water uptake can be quantified. The illustration is done using a simple, four-box model of the soil-root system to represent heterogeneity and a parameterization in which root water uptake is driven by the xylem potential of the plant with a fixed flux boundary condition. We use this approach to evaluate the effects of soil moisture heterogeneity and root system properties on the dissipative losses and export of energy involved in root water uptake. For this, we derive an expression that relates the energy export at the root collar to a sum of terms that reflect all fluxes and storage changes along the flow path in thermodynamic terms. We conclude that such a thermodynamic evaluation of root water uptake conveniently provides insights into the impediments of different processes along the entire flow path and explicitly accounting not only for the resistances along the flow path and those imposed by soil drying but especially the role of heterogenous soil water distribution. The results show that least energy needs to be exported and dissipative losses are minimized by a root system if it extracts water uniformly from the soil. This has implications for plant water relations in forests where canopies generate heterogenous input patterns. Our diagnostic in the energy domain should be useful in future model applications for quantifying how plants can evolve towards greater efficiency in their structure and function, particularly in heterogenous soil environments. Generally, this approach may help to better describe heterogeneous processes in the soil in a simple, yet physically-based way.

Résumé :
Abstract Many climate subsystems are thought to be susceptible to tipping—and some might be close to a tipping point. The general belief and intuition, based on simple conceptual models of tipping elements, is that tipping leads to reorganization of the full (sub)system. Here, we explore tipping in conceptual, but spatially extended and spatially heterogenous models. These are extensions of conceptual models taken from all sorts of climate system components on multiple spatial scales. By analysis of the bifurcation structure of such systems, special stable equilibrium states are revealed: coexistence states with part of the spatial domain in one state, and part in another, with a spatial interface between these regions. These coexistence states critically depend on the size and the spatial heterogeneity of the (sub)system. In particular, in these systems the crossing of a tipping point not necessarily leads to a full reorganization of the system. Instead, it might lead to a reorganization of only part of the spatial domain, limiting the impact of these events on the system’s functioning.

Résumé :
Carnival Corporation & plc identified the need for a cutting-edge revenue management system; however, existing solutions from the airline and hospitality industries were not compatible with the idiosyncrasies of the cruise domain. As such, the company partnered with revenue analytics to build a complete revenue and inventory management system to meet its requirements. Yield optimization and demand analytics (YODA) is a system that leverages a unique quadratic programming model to jointly determine cruise prices and allocate cabin inventory to multiple cruises (e.g., 14-day and 7-day lengths) offered simultaneously on a given ship. The optimization inputs come from several machine learning algorithms that predict demand. YODA combines these algorithms with an elasticity model derived from an exponential curve to represent the unique price-sensitivity behavior observed in the cruise industry. The system generates millions of price recommendations each day and has been used to price voyages on 65 Carnival ships, approximately one quarter of the ships in the entire cruise industry, since December 2017. During A/B testing, YODA generated a 1.5%–2.5% incremental uplift in net ticket revenue, which is a significant revenue increase because Carnival was a Fortune 300 company in 2019.

In this narrative review, we present the evidence on nucleotide-binding and oligomerization (NOD) domain-like receptor (NLR) family pyrin domain (PYD)-containing 3 (NLRP3) inflammasome activation for its putative roles in the elusive pathomechanism of aging-related cerebral small vessel disease (CSVD). Although NLRP3 inflammasome-interleukin (IL)-1β has been implicated in the pathophysiology of coronary artery disease, its roles in cerebral arteriothrombotic micro-circulation disease such as CSVD remains unexplored. Here, we elaborate on the current manifestations of CSVD and its’ complex pathogenesis and relate the array of activators and aberrant activation involving NLRP3 inflammasome with this condition. These neuroinflammatory insights would expand on our current understanding of CSVD clinical (and subclinical) heterogenous manifestations whilst highlighting plausible NLRP3-linked therapeutic targets.

Résumé :
Abstract The presence of heterogeneity/subgroups in infants and older populations against single-domain brain or behavioral measures has been previously characterized. However, few attempts have been made to explore heterogeneity at the brain–behavior relationship level. Such a hypothesis posits that different subgroups of infants may possess qualitatively different brain–behavior relationships that could ultimately contribute to divergent developmental outcomes even with relatively similar brain phenotypes. In this study, we aimed to explore such relationship-level heterogeneity and delineate the subgrouping structure of newborns with differential brain–behavior associations based on a typically developing sample of 81 infants with 3-week resting-state functional magnetic resonance imaging scans and 4-year intelligence quotient (IQ) measures. Our results not only confirmed the existence of relationship-level heterogeneity in newborns but also revealed divergent developmental outcomes associated with two subgroups showing similar brain functional connectivity but contrasting brain–behavior relationships. Importantly, further analyses unveiled an intriguing pattern that the subgroup with higher 4-year IQ outcomes possessed brain–behavior relationships that were congruent to their functional connectivity pattern in neonates while the subgroup with lower 4-year IQ not, providing potential explanations for the observed IQ differences. The characterization of heterogeneity at the brain–behavior relationship level may not only improve our understanding of the patterned intersubject variability during infancy but could also pave the way for future development of heterogeneity-inspired, personalized, subgroup-specific models for better prediction.

Résumé :
Geographic Information System (GIS) is a computer system designed to capture, store, manipulate, analyze, manage, and present all types of spatial data. Spatial data, whether captured through remote sensors or large scale simulations has always been big and heterogenous. The issue of real-time and heterogeneity have been extremely important for taking effective decision. Thus, heterogeneous real-time spatial data management has become a very active research domain. Existing research has principally focused on querying of real-time spatial data and their updates. But the unpredictability of access to data maintain the behavior of the real-time GIS unstable. In this paper, we propose the use of the real-time Spatial Big Data and we define a new architecture called FCSA-RTSBD (Feedback Control Scheduling Architecture for Real-Time Spatial Big Data). The main objectives of this architecture are the following: take in account the heterogeneity of data, guarantee the data freshness, enhance the deadline miss ratio even in the presence of conflicts and unpredictable workloads and finally satisfy the requirements of users by the improving of the quality of service (QoS).

Résumé :
The number of studies evaluating flux or concentration footprints has grown considerably in recent years. These footprints are vital to understand surface–atmosphere flux measurements, for example by eddy covariance. The newly developed backwards trajectory model LaStTraM (Lagrangian Stochastic Trajectory Model) is a post-processing tool, which uses simulation results of the holistic 3D microclimate model ENVI-met as input. The probability distribution of the particles is calculated using the Lagrangian Stochastic method. Combining LaStTraM with ENVI-met should allow us to simulate flux and concentration footprints in complex urban environments. Applications and evaluations were conducted through a comparison with the commonly used 2D models Kormann Meixner and Flux Footprint Predictions in two different meteorological cases (stable, unstable) and in three different detector heights. LaStTraM is capable of reproducing the results of the commonly used 2D models with high accuracy. In addition to the comparison with common footprint models, studies with a simple heterogeneous and a realistic, more complex model domain are presented. All examples show plausible results, thus demonstrating LaStTraM’s potential for the reliable calculation of footprints in homogeneous and heterogenous areas.

Résumé :
Abstract Background The population of older patients with Inflammatory Bowel Diseases (IBD) is expanding. Knowledge on the prevalence and impact of geriatric impairments is scarce in this heterogenous group. In other fields of medicine, it has been established that geriatric impairments associate with adverse outcomes. Therefore, the aims of our study were to assess the prevalence of geriatric impairments in older IBD patients and to evaluate the association between geriatric impairments and disease burden of IBD through the short Inflammatory Bowel Disease Questionnaire (sIBDQ). Methods Consecutive IBD patients aged ≥65 years were included at outpatient departments and infusion centres of four hospitals in The Netherlands. Comorbidity, polypharmacy and malnutrition (somatic domain), cognitive impairment and depressive symptoms (mental domain), handgrip strength and gait speed (physical domain) and impairments in (instrumental) activities of daily living ((I)ADL) (functional domain) were assessed. Disease activity was assessed through Harvey Bradshaw Index (HBI) or partial Mayo Score (pMS) (remission: HBI<5 or pMS<2); The sIBDQ was used to assess IBD disease burden. Association between geriatric impairments and sIBDQ was assessed using multiple linear regression, including confounders age, sex, IBD type, disease duration and disease activity. Results In total, 336 patients were included: median age 70.0 (IQR 67.0–73.0); 161 CD (47.9%); 71 active disease (21.1%); mean sIBDQ 59.7 (SD 8.1). Prevalence of geriatric impairments is tabulated (Table 1). Both active disease and the presence of two or more impaired domains were statistically significantly and independently associated with a higher disease burden (lower sIBDQ) (Table 2). Conclusion In an older IBD patient population, geriatric impairments are frequently encountered and associate with a higher disease burden. These results are a call for more research on the relationship between geriatric impairments and IBD disease burden to improve patient care in older IBD patients.

Résumé :
Background Hay fever is a highly prevalent, heterogenous, and multifactorial disease. Patients may benefit from longitudinal assessments using mobile health (mHealth) principles. We have previously attempted to establish an effective mHealth platform for patients with hay fever through AllerSearch, our in-house smartphone app that assesses electronic patient-reported outcomes through a questionnaire on hay fever and provides evidence-based advice. To be used by the public, an investigation on its reliability and validity is necessary. Objective The aim of this paper is to assess the reliability and validity of subjective symptom data on hay fever collected through our app, AllerSearch. Methods This study used a prospective observational design. The participants were patients aged ≥20 years recruited from a single university hospital between June 2, 2021, and January 26, 2022. We excluded patients who could not use smartphones as well as those with incomplete data records and outlier data. All participants answered the Japanese Allergic Conjunctival Disease Standard Quality of Life Questionnaire (JACQLQ), first in the paper-and-pencil format and subsequently on AllerSearch on the same day. The JACQLQ comprises the following three domains: Domain I, with 9 items on ocular or nasal symptoms; Domain II, with 17 items on daily activity and psychological well-being; and Domain III, with 3 items on overall condition by face score. The concordance rate of each domain between the 2 platforms was calculated. The internal consistency of Domains I and II of the 2 platforms was assessed using Cronbach alpha coefficients, the concurrent validity of Domains I and II was assessed by calculating Pearson correlation coefficients, and the mean differences between the 2 platforms were assessed using Bland-Altman analysis. Results In total, 22 participants were recruited; the data of 20 (91%) participants were analyzed. The average age was 65.4 (SD 12.8) years, and 80% (16/20) of the participants were women. The concordance rate of Domains I, II, and III between the paper-based and app-based JACQLQ was 0.78, 0.85, and 0.90, respectively. The internal consistency of Domains I and II between the 2 platforms was satisfactory (Cronbach alpha of .964 and .919, respectively). Pearson correlation analysis yielded a significant positive correlation between Domains I and II across the 2 platforms (r=0.920 and r=0.968, respectively). The mean difference in Domains I and II between the 2 platforms was 3.35 units (95% limits of agreement: –6.51 to 13.2). Conclusions Our findings indicate that AllerSearch is a valid and reliable tool for the collection of electronic patient-reported outcomes to assess hay fever, contributing to the advantages of the mHealth platform.

Résumé :
Background and Purpose: Cerebrovascular disease (CVD) pathologies including vessel disease (atherosclerosis, arteriolosclerosis, and cerebral amyloid angiopathy) and tissue injury (macroinfarcts and microinfarcts) each contribute to Alzheimer and other forms of dementia. CVD is often a complex mix of neuropathologies, with little known about the frequencies of differing combinations or their associations with cognition. Methods: We investigated 32 possible CVD combinations (3 types of vessel disease and 2 types of tissue injury) using autopsy data from 1474 decedents (≈88 years at death; 65% female) of Rush Alzheimer’s Disease Center studies. We determined frequencies of all 32 CVD combinations and their relationships with global and domain-specific cognitive decline using mixed-effect models adjusted for demographics, neuropathologies, time before death, and interactions of these variables with time. Results: Of the 1184 decedents with CVD neuropathology (80% of the total sample), 37% had a single CVD (67–148 decedents/group) while 63% had mixed CVD profiles (11–54 decedents/group). When considered as 2 distinct groups, the mixed CVD profile group (but not the single CVD profile group) showed a faster cognitive decline across all domains assessed compared with decedents without CVD neuropathology. Most mixed CVD profiles, especially those involving both atherosclerosis and arteriolosclerosis, showed faster cognitive decline than any single CVD profile considered alone; specific mixed CVD profiles differentially associated with individual cognitive domains. Conclusions: Mixed CVD, more common than single CVD, is associated with cognitive decline, and distinct mixed CVD profiles show domain-specific associations with cognitive decline. CVD is not monolithic but consists of heterogenous person-specific combinations with distinct contributions to cognitive decline.

AbstractPopular dances encapsulate the aliveness of Africa's young. Radiating an Africanist aesthetic of the cool, these moves enflesh popular music, saturating mass media platforms and everyday spaces with imageries of joyful transcendence. This essay understands scriptive dance fads as textual and choreographic calls for public embodiment. I explore how three Nigerian musicians, and their dances, have wielded scriptive prompts to elicit specific moved responses from dispersed, heterogenous, and transnational publics. Dance fads of this kind productively complicate musicological approaches that insist on divorcing contemporary African music cultures from the dancing bodies that they often conjure. Taken together, these movements enlist popular culture as a domain marked by telling contestations over musical ownership and embodied citizenship.

Résumé :
Distributed radar is applied extensively in marine environment monitoring. In the early days, the radar signals are identified inefficiently by operators. It is promising to replace manual radar signal identification with machine learning technique. However, the existing deep learning neural networks for radar signal identification consume a long time, owing to autonomous learning. Besides, the training of such networks requires lots of reliable time-frequency features of radar signals. This paper mainly analyzes the identification and classification of marine distributed radar signals with an improved deep neural network. Firstly, the time frequency features were extracted from signals based on short-time Fourier transform (STFT) theory. Then, a target detection algorithm was proposed, which weighs and fuses the heterogenous marine distributed radar signals, and four methods were provided for weight calculation. After that, the frequency-domain priori model feature assistive training was introduced to train the traditional deep convolutional neural network (DCNN), producing a CNN with feature splicing operation. The features of time- and frequency-domain signals were combined, laying the basis for radar signal classification. Our model was proved effective through experiments.

Résumé :
The RNA-binding protein Sam68 is involved in apoptosis, but its cellular mRNA targets and its mechanism of action remain unknown. We demonstrate that Sam68 binds the mRNA for Bcl-x and affects its alternative splicing. Depletion of Sam68 by RNA interference caused accumulation of antiapoptotic Bcl-x(L), whereas its up-regulation increased the levels of proapoptotic Bcl-x(s). Tyrosine phosphorylation of Sam68 by Fyn inverted this effect and favored the Bcl-x(L) splice site selection. A point mutation in the RNA-binding domain of Sam68 influenced its splicing activity and subnuclear localization. Moreover, coexpression of ASF/SF2 with Sam68, or fusion with an RS domain, counteracted Sam68 splicing activity toward Bcl-x. Finally, Sam68 interacted with heterogenous nuclear RNP (hnRNP) A1, and depletion of hnRNP A1 or mutations that impair this interaction attenuated Bcl-x(s) splicing. Our results indicate that Sam68 plays a role in the regulation of Bcl-x alternative splicing and that tyrosine phosphorylation of Sam68 by Src-like kinases can switch its role from proapoptotic to antiapoptotic in live cells.

Résumé :
Polyphenols such as epigallocatechin gallate (EGCg) may have roles in preventing some chronic diseases when they are ingested as components of plant-based foods and beverages. Human serum albumin (HSA) is a multi-domain protein that binds various ligands and aids in their transport, distribution, and metabolism in the circulatory system. In the present study, the HSA-EGCg interaction in the absence or presence of fatty acid has been investigated. Förster resonance energy transfer (FRET) was used to determine inter- and intra-domain distances in the protein with and without EGCg and palmitic acid (PA). By labeling Cys-34 with 7-(diethyl amino)-4-methylcoumarin 3-maleimide (CPM), the distance between Trp-214 at domain IIA and CPM-Cys-34 at domain IA could be established. A small amount of PA decreased the distance, while a large amount increased the distance up to 5.4 Å. EGCg increased the inter-domain distance in HSA and HSA-PA up to 2.8 and 7.6 Å, respectively. We concluded that PA affects protein conformation more significantly compared to EGCg. Circular dichroism (CD) established that EGCg affects protein secondary structure more significantly than PA. PA had little effect on the α-helix content of HSA, while EGCg decreased the α-helix content in a dose-dependent fashion. Moreover, EGCg decreased α-helix content in HSA and HSA-PA to the same level. Dynamic light scattering (DLS) data revealed that both PA and EGCg increased HSA aggregation. EGCg increased HSA aggregation more significantly and promoted formation of aggregates that were more heterogenous. Any of these effects could impact the ability of serum albumin to transport and stabilize ligands including EGCg and other polyphenols.

Résumé :
After the abolishment of fixed exchange regime under the Bretton Woods System in 1970s, the issue of foreign exchange risk expecially among multinational corporations operating under small and open economies of Malaysia remains relevant till today. According to Abdul Wahab et al. (2017) and Adler and Dumas (1984), exchange rate exposure refers to the extent of firm's sensitivity to exchange rate fluctuations. While there has been growing interest in examining foreign exchange risk incidence under Malaysia market, however limited studies have been focusing on single sector of consumer products and services in Malaysia. Under Bursa Malaysia, the sector consists of several sub-fields including agricultural products, automotive, food and beverages, household goods, personal goods, retailers and travel and leisure services. Given the wide range of products and services, the corporations under the sector have been involved in vigorous cross-border trading activities. The consumer products and services equities become the second largest sector in Bursa Malaysia which constitues 15.89% of total stocks in main market of Bursa Malaysia in July 2021 with a 265.02 billion of total market capitalization. In terms of methodological issue, the study addresses on the bias in exchange risk pricing in previous studies. Specifically, majority of past studies assume homogeneity of exchange risk across time domains which seems to be unrealistic. Capital market contains wide range of participants with heterogenous investment holdings and expectations. To address time domain bias, the study adopts Maximal Overlap Discrete Wavelet Transformation (MODWT) in foreign exchange risk pricing to decompose a single time domain series into multiple time domains. Given this, the primary objective of the study is to examine the extent of multi-horizon foreign exchange exposure of consumer products and services sector in Malaysia from January 2000 till December 2020. Ovarian failure is defined as cessation of ovarian function under the age of 40 and it is characterized by amenorrhea hypo estrogenism and elevated gonadotrophin serum levels leading to infertility. Ovarian failure affects 1% of women. Causes of ovarian failure are extremely heterogenous. Ovarian failure is caused either by accelerated loss of eggs or the follicles themselves become less responsive to hormonal stimulation. It is high levels of gonadotrophins (luteinizing hormone and follicle stimulating hormone) hormonal defects may cause severe cardio vascular consequences and leads to the early onset of osteoporosis. Vascular endothelial growth factor (VEGF) is a major growth factor for endothelial cells. VEGF is involved in the process of angiogenesis for the development of Graffian follicle an important feature of growing follicle which has been arrested in the primordial follicular stage .VEGF mediates the process of angiogenesis through its receptors I and II.. Receptor II is known as Kinase insert domain receptor, is a type III receptor tyrosine Kinase.KDR is located at 4q11-q12.The main objective of this study was to screen the mutations in Kinase insert domain by using the techniques DNA isolation, Polymerase chain reaction and restriction fragment length polymorphism.

AbstractA detailed reconstruction of the environment is a crucial component of mobile robotic systems and enables higher level scene understanding. To achieve information redundancy heterogenous sensors need to be used with each sensor having specific strengths and weaknesses. Therefore, the goal of this work is to fuse information from multiple lidars, radars, a stereo camera and semantic camera information into one common scene representation. In contrast to past publications, we focus on the combination of distance measurements and semantic estimates in the image domain in one common evidential framework. Grid maps are used as common fusion structure which enable efficient data processing. The approach is validated on an automated driving plattform in real traffic scenarios. Experiments show that the scene reconstruction precision increases while still retaining the real-time capability.

Résumé :
Plants evolved intracellular immune receptors that belong to the NOD-like receptor (NLR) family to recognize the presence of pathogen-derived effector proteins. NLRs possess an N-terminal Toll-like/IL-1 receptor (TIR) or a non-TIR domain [some of which contain coiled coils (CCs)], a central nucleotide-binding (NB-ARC) domain, and a C-terminal leucine-rich repeat (LRR). Activation of NLR proteins results in a rapid and high-amplitude immune response, eventually leading to host cell death at the infection site, the so-called hypersensitive response. Despite their important contribution to immunity, the exact mechanisms of NLR activation and signaling remain unknown and are likely heterogenous. We undertook a detailed structure-function analysis of the plasma membrane (PM)-localized CC NLR Resistance to Pseudomonas syringae pv. maculicola 1 (RPM1) using both stable transgenic Arabidopsis and transient expression in Nicotiana benthamiana. We report that immune signaling is induced only by activated full-length PM-localized RPM1. Our interaction analyses demonstrate the importance of a functional P-loop for in planta interaction of RPM1 with the small host protein RPM1-interacting protein 4 (RIN4), for constitutive preactivation and postactivation self-association of RPM1 and for proper PM localization. Our results reveal an additive effect of hydrophobic conserved residues in the CC domain for RPM1 function and RPM1 self-association and their necessity for RPM1–RIN4 interaction. Thus, our findings considerably extend our understanding of the mechanisms regulating NLR activation at, and signaling from, the PM.

Abstract Mendelian susceptibility to mycobacterial disease (MSMD) is a heterogenous syndrome. The clinical characteristics include repetitive local infection and in some cases dissemination. In this work we analyzed a 5 years old boy with an uncharacterized mycobacterial infection. His parents are first degree cousins. Blood samples were collected and cultured 48 hours with medium, BCG or BCG+IL-12. Supernatants were assayed for IFN-γ by ELISA. PBMC were cultured with PMA-Ionomicyn and IL12R was measured by flow cytometry. We found low production of IFN γ after stimulation with BCG+IL12 that seems to correlate with an IL-12R defect. Flow cytometry showed a decrease of IL-12RB1 expression. Sequencing revealed a point mutation (A182T), which generates E61G change in the extracellular domain. Furthermore, the uncommon clinical features in this case might suggest another primary immunodeficiency.

Résumé :
Abstract Intrinsically disordered proteins are crucial elements of chromatin heterogenous organization. While disorder in the histone tails enables a large variation of inter-nucleosome arrangements, disorder within the chromatin-binding proteins facilitates promiscuous binding to a wide range of different molecular targets, consistent with structural heterogeneity. Among the partially disordered chromatin-binding proteins, the H1 linker histone influences a myriad of chromatin characteristics including compaction, nucleosome spacing, transcription regulation, and the recruitment of other chromatin regulating proteins. Although it is now established that the long C-terminal domain (CTD) of H1 remains disordered upon nucleosome binding and that such disorder favours chromatin fluidity, the structural behaviour and thereby the role/function of the N-terminal domain (NTD) within chromatin is yet unresolved. On the basis of microsecond-long parallel-tempering metadynamics and temperature-replica exchange atomistic molecular dynamics simulations of different H1 NTD subtypes, we demonstrate that the NTD is completely unstructured in solution but undergoes an important disorder-to-order transition upon nucleosome binding: it forms a helix that enhances its DNA binding ability. Further, we show that the helical propensity of the H1 NTD is subtype-dependent and correlates with the experimentally observed binding affinity of H1 subtypes, suggesting an important functional implication of this disorder-to-order transition.

Résumé :
The security of images has gained great interest in modern communication systems. This is due to the massive critical applications that are based on images. Medical imaging is at the top of these applications. However, the rising number of heterogenous attacks push toward the development of securing algorithms and methods for imaging systems. To this end, this work considers developing a novel authentication, intellectual property protection, ownership, and security technique for imaging systems, mainly for medical imaging. The developed algorithm includes two security modules for safeguarding various picture kinds. The first unit is accomplished by applying watermarking authentication in the frequency domain. The singular value decomposition (SVD) is performed for the host image’s discrete cosine transform (DCT) coefficients. The singular values (S) are divided into 64 × 64 non-overlapping blocks, followed by embedding the watermark in each block to be robust to any attack. The second unit is made up of two encryption layers to provide double-layer security to the watermarked image. The double random phase encryption (DRPE) and chaotic encryption have been tested and examined in the encryption unit. The suggested approach is resistant to common image processing attacks, including rotation, cropping, and adding Gaussian noise, according to the findings of the experiments. The encryption of watermarked images in the spatial and DCT domains and fused watermarked images in the DCT domain are all discussed. The transparency and security of the method are assessed using various measurements. The proposed approach achieves high-quality reconstructed watermarks and high security by using encryption to images and achieves robustness against any obstructive attacks. The developed hybrid algorithm recovers the watermark even in the presence of an attack with a correlation near 0.8.

Résumé :
The accessory protein Nef of human and simian immunodeficiency viruses (HIV and SIV) is an important pathogenicity factor known to interact with cellular protein kinases and other signaling proteins. A canonical SH3 domain binding motif in Nef is required for most of these interactions. For example, HIV-1 Nef activates the tyrosine kinase Hck by tightly binding to its SH3 domain. An archetypal contact between a negatively charged SH3 residue and a highly conserved arginine in Nef (Arg77) plays a key role here. Combining structural analyses with functional assays, we here show that Nef proteins have also developed a distinct structural strategy—termed the "R-clamp”—that favors the formation of this salt bridge via buttressing Arg77. Comparison of evolutionarily diverse Nef proteins revealed that several distinct R-clamps have evolved that are functionally equivalent but differ in the side chain compositions of Nef residues 83 and 120. Whereas a similar R-clamp design is shared by Nef proteins of HIV-1 groups M, O, and P, as well as SIVgor, the Nef proteins of SIV from the Eastern chimpanzee subspecies (SIVcpzP.t.s.) exclusively utilize another type of R-clamp. By contrast, SIV of Central chimpanzees (SIVcpzP.t.t.) and HIV-1 group N strains show more heterogenous R-clamp design principles, including a non-functional evolutionary intermediate of the aforementioned two classes. These data add to our understanding of the structural basis of SH3 binding and kinase deregulation by Nef, and provide an interesting example of primate lentiviral protein evolution.

Résumé :
The author explores a theoretic background and develops an integrative model of self-destruction of personality. The existing views on selfdestruction are local and isolated. The self-destruction has not been researched in the whole commonality of its many factors and sources. A new step in self-destruction research is to study it in terms of integration. The following research turns to the two ways on integration: inductive and deductive. In inductive approach, the author choses a certain set of self-destruction factors (their localization) and raises a question whether they have a common basis. In the deductive approach, on the contrary, the author offers a certain general concept that leads to a set of local factors of self-destruction. Within the framework of the integrative approach, these areas of self-destruction research are considered to be complementary. They form the basis of an integrative model of selfdestruction and develop from the standpoint of the conceptual model of the plural Self. Concerning the deformations of the plural Self, the self-destruction appears as its antipode. There are four domains in the self-destruction of a healthy personality: dissatisfaction with oneself, need to dominate, intolerance, detachment. This model concretizes the heterogenous picture of self-destruction in terms of its domain structure, largely overcomes the isolated notions and generalizes them.

To date, researchers who have done work life balance (WLB) have mainly focused on work and the family domain. However, the present-day workforce is heterogenous in nature and the workers might even value the other domains of non-working apart from family. These researchers mainly aim at investigating the significance of the other domains of non-working in work life balance with a special focus on the health of workers. Besides the meaning of the effect of balance between serious and fun activities and work well-being balance on representative fulfillment was evaluated. Likewise, these examinations investigate the impact of balance between serious and fun activities and work well-being balance on change in the degree of representative fulfillment according to the qualities of the laborers, including parental status, age, work capacity, and sex. The consequences of studies show that the specialists believe well-being to be a significant factor in the space of balance between fun and serious activities. Work well-being balance clarifies a greater amount of change in representative fulfillment when contrasted with balance between fun and serious activities. A sample of 311 respondents was surveyed to find the factors related to work life balance and their impact on job switching behavior. After applying factor analysis, a statistical tool, and analyzing the data, the study concludes that work health balance, job satisfaction, work environment, relation with management and co-workers, and work and personal life balance are the different factors related to work life balance that shows significant impact on job switching behavior.

Résumé :
To assess the potential contribution of the cysteine-rich domain of human platelet glycoprotein (GP) IIIa to the structure of the PlA1 epitope, we used site-directed mutagenesis to substitute alanines for cysteines at key positions potentially involved in PlA1 alloantigen formation. One of these GPIIIa isoforms in which alanine replaced Cys435 reacted normally with a series of well-characterized murine MoAbs directed against the 250 amino-terminal residues of GPIIIa, whereas binding of MoAbs specific for residues 348–692 was diminished or lost. Interestingly, of eight PlA1-specific antibodies tested, two recognized Ala435GPIIIa, two did not, and four bound to it less well than to wild-type (WT) GPIIIa. However, disruption of disulfide bonds located at or near the N-terminus of GPIIIa abolished the binding of all the anti-PlA1 alloantibodies tested. These findings provide evidence that the humoral response to the PlA1 antigen is (1) heterogenous, ie, the binding site on GPIIIa for human anti-PlA1 antibodies differs from one individual to another and (2) complex--although some anti-PlA1 alloantibodies bind to an epitope comprised solely of the amino-terminus of GPIIIa, others combine most efficiently with a more complex determinant that involves the cysteine-rich domain of GPIIIa. These findings may have implications for diagnostic and therapeutic use of synthetic or recombinant PlA1 mimetics.

Résumé :
Heterogenous nuclear ribonucleoproteins (hnRNPs) bind pre-mRNAs and facilitate their processing into mRNAs. Many of the hnRNPs undergo extensive posttranslational modifications including methylation on arginine residues. hnRNPs contain about 65% of the total NG,NG-dimethylarginine found in the cell nucleus. The role of this modification is not known. Here we identify the hnRNPs that are methylated in HeLa cells and demonstrate that most of the pre-mRNA-binding proteins receive this modification. Using recombinant human hnRNP A1 as a substrate, we have partially purified and characterized a protein-arginine N-methyltransferase specific for hnRNPs from HeLa cells. This methyltransferase can methylate the same subset of hnRNPs in vitro as are methylated in vivo. Furthermore, it can also methylate other RNA-binding proteins that contain the RGG motif RNA-binding domain. This activity is evolutionarily conserved from lower eukaryotes to mammals, suggesting that methylation has a significant role in the function of RNA-binding proteins.

Résumé :
Innovation Concept: Assessment of residents' Point of Care Ultrasound (PoCUS) competency currently relies on heterogenous and unvalidated methods, such as the completion of a number of proctored studies. Although number of performed studies may be associated with ability, it is not necessarily a surrogate for competence. Our goal was to create a single Ultrasound Competency Assessment Tool (UCAT) using domain-anchored entrustment scoring. Methods: The UCAT was developed as an anchored global assessment score, building on a previously validated simulation-based assessment tool. It was designed to measure performance across the domains of Preparation, Image Acquisition, Image Optimization, and Clinical Integration, in addition to providing a final entrustment score (i.e., OSCORE). A modified Delphi method was used to establish national expert consensus on anchors for each domain. Three surveys were distributed to the CAEP Ultrasound Committee between July-November 2018. The first survey asked members to appraise and modify a list of anchor options created by the authors. Next, collated responses from the first survey were redistributed for a re-appraisal. Finally, anchors obtaining >65% approval from the second survey were condensed and redistributed for final consensus. Curriculum, Tool or Material: Twenty-two, 26, and 22 members responded to the surveys, respectively. Each anchor achieved >90% final agreement. The final anchors for the domains were: Preparation – positioning, initial settings, ensures clean transducer, probe selection, appropriate clinical indication; Image Acquisition – appropriate measurements, hand position, identifies landmarks, visualization of target, efficiency of probe motion, troubleshoots technical limitations; Image Optimization – centers area of interest, overall image quality, troubleshoots patient obstacles, optimizes settings; Clinical Integration – appropriate interpretation, understands limitations, utilizes information appropriately, performs multiple scans if needed, communicates findings, considers false positive and negative causes of findings. Conclusion: The UCAT is a novel assessment tool that has the potential to play a central role in the training and evaluation of residents. Our use of a modified Delphi method, involving key stakeholders in PoCUS education, ensures that the UCAT has a high degree of process and content validity. An important next step in determining its construct validity is to evaluate the use of the UCAT in a multi-centered examination setting.

Résumé :
One of the most promising means of reducing carbon content in the atmosphere, which is aimed at tackling the threats of global warming, is injecting carbon dioxide (CO2) into deep saline aquifers (DSAs). Keeping this in mind, this research aims to investigate the effects of various injection schemes/scenarios and aquifer characteristics with a particular view to enhance the current understanding of the key permanent sequestration mechanisms, namely, residual and solubility trapping of CO2. The paper also aims to study the influence of different injection scenarios and flow conditions on the CO2 storage capacity and efficiency of DSAs. Furthermore, a specific term of the permanent capacity and efficiency factor of CO2 immobilization in sedimentary formations is introduced to help facilitate the above analysis. Analyses for the effects of various injection schemes/scenarios and aquifer characteristics on enhancing the key permanent sequestration mechanisms is examined through a series of numerical simulations employed on 3D homogeneous and heterogeneous aquifers based on the geological settings for Sleipner Vest Field, which is located in the Norwegian part of the North Sea. The simulation results highlight the effects of heterogeneity, permeability isotropy, injection orientation and methodology, and domain-grid refinement on the capillary pressure–saturation relationships and the amounts of integrated CO2 throughout the timeline of the simulation via different trapping mechanisms (solubility, residual and structural) and accordingly affect the efficiency of CO2 sequestration. The results have shown that heterogeneity increases the residual trapping of CO2, while homogeneous formations promote more CO2 dissolution because fluid flows faster in homogeneous porous media, inducing more contact with fresh brine, leading to higher dissolution rates of CO2 compared to those in heterogeneous porous medium, which limits fluid seepage. Cyclic injection has been shown to have more influence on heterogenous domains as it increases the capillary pressure, which forces more CO2 into smaller-sized pores to be trapped and exposed to dissolution in the brine at later stages of storage. Storage efficiency increases proportionally with the vertical-to-horizontal permeability ratio of geological formations because higher ratios facilitate the further extent of the gas plume and increases the solubility trapping of the integrated gas. The developed methodology and the presented results are expected to play key roles in providing further insights for assessing the feasibility of various geological formations for CO2 storage.

Résumé :
Cognitive radio has received considerable amount of attention as a promising technique to provide dynamic spectrum allocation. Spectrum sensing is one of the basic functions in the cognitive radio and is crucial to all other functions. Software- defined radios (SDRs) are considered due to its very high flexibility and have become a common platform for CR implementation replacing expensive spectrum analysers. The most popular among various SDR platforms is the universal software-defined radio peripheral (USRP). This paper presents a real-time swept spectrum sensing solution based on USRP B210. It also presents a detailed explanation of the concept of energy detection and the methodology for wide-band sensing. Finally, the performance of the proposed sensing solution is analysed through FFT graphs and spectrogram plot taken for 8 hours. The results showed that the proposed sensing solution was capable of achieving high resolution in the frequency domain of the wide band measured which implies that wide bands with heterogenous signals like the ISM band can be accurately resolved and analysed.

Résumé :
The future network slicing enabled mobile ecosystem is expected to support a wide set of heterogenous vertical services over a common infrastructure. The service robustness and their intrinsic requirements, together with the heterogeneity of mobile infrastructure and resources in both the technological and the spatial domain, significantly increase the complexity and create new challenges regarding network management and orchestration. High degree of automation, flexibility and programmability are becoming the fundamental architectural features to enable seamless support for the modern telco-based services. In this paper, we present a novel management and orchestration platform for network slices, which has been devised by the Horizon 2020 MonB5G project. The proposed framework is a highly scalable solution for network slicing management and orchestration that implements a distributed and programmable AI-driven management architecture. The cognitive capabilities are provided at different levels of management hierarchy by adopting necessary data abstractions. Moreover, the framework leverages intent-based operations to improve its modularity and genericity. The mentioned features enhance the management automation, making the architecture a significant step towards self-managed network slices.

Résumé :
e18050 Background: Adenoid cystic carcinoma (ACC) is a rare malignancy of glandular tissue with a high rate of local recurrence and metastatic disease for which clinical behavior varies widely. Currently, no standard therapy exists, and available therapies have low response rates. Therefore, prognostic biomarkers to determine optimal treatment strategies are urgently needed. Here we investigate the molecular landscape and therapeutic targets of ACC. Methods: ACC samples were analyzed using next generation sequencing (NGS) (NextSeq, 592 gene panel) or whole exome sequencing (NovaSeq) (Caris Life Sciences, Phoenix, AZ). Pathogenic fusion events were detected using whole transcriptome sequencing (NovaSeq). Statistical significance was determined using the Chi-square test and adjusted for multiple comparisons. Tumor infiltrating Immune cell fractions were determined using the QuanTIseq deconvolution algorithm and WTS data. Results: 327 ACC patients were identified, median age of 58 years, 62% female and 62% metastatic disease. The most frequent mutations (excluding indeterminate results) involved genes in the NOTCH pathway ( NOTCH1 16% [41 of 298 cases]), chromatin remodeling ( ARID1A 24.3% [27 of 138 cases], KDM6A 12.5% [28 of 224 cases], KMT2C 10.7% [21 of 197 cases]) and TP53 10.2% (25 of 246 cases). Chromatin remodeling genes were co-mutated with NOTCH1. NOTCH1 mutations were found in 41 cases, 18 primary and 23 metastatic were associated with reduced median overall survival (mOS 1.8 years vs 3.8 years, p = 0.01). Mutations were most frequent in the PEST (n = 35), NRR (n = 18), PEST+NRR (n = 16) and EGF-like domains (n = 11). PEST domain mutations were found exclusively in ACC originating from the head and neck (H&N). GSEA showed that MYC, E2F and G2M target pathways were enriched in NOTCH1 mutated ACC regardless of the presence of MYB fusions. All three pathways were enriched in PEST domain mutations whereas only MYC targets were enriched in NRR and NRR+PEST domain mutations indicating that PEST domain mutations drive transcriptomic changes in NOTCH1 mutated ACC. MYC gene expression was significantly upregulated in NOTCH1 mutated ACC. Fusion events were detected in 45% (104/231). MYB:NFIB was the most common fusion (40%) and was more frequent in H&N origin than other sites. There was no difference in the mOS between MYB fusion positive and negative ACC (4.4 years vs 5.5 years, p = 0.14). M2-polarized macrophages, myeloid dendritic cells and neutrophils were significantly higher in metastatic compared to primary ACC while NK cells were more abundant in primary tumor. Conclusions: ACC is a genetically heterogenous disease with an immune-excluded microenvironment. NOTCH1 mutations were associated with worse, while MYB:NFIB fusion was not associated with prognosis. Our study shows that M2 macrophages and MYC are potential novel therapeutic targets in ACC.

Résumé :
Heterogeneous Transfer Learning (HTL) aims to solve transfer learning problems where a source domain and a target domain are of heterogeneous types of features. Most existing HTL approaches either explicitly learn feature mappings between the heterogeneous domains or implicitly reconstruct heterogeneous cross-domain features based on matrix completion techniques. In this paper, we propose a new HTL method based on a deep matrix completion framework, where kernel embedding of distributions is trained in an adversarial manner for learning heterogeneous features across domains. We conduct extensive experiments on two different vision tasks to demonstrate the effectiveness of our proposed method compared with a number of baseline methods.

Résumé :
SummaryBernard-Soulier syndrome (BSS) is a rare inherited bleeding disorder, which is caused by deficiency or decrease of the platelet GPIb/IX/V complex. Analysis of two patients with BSS by How cytometry of the blood revealed different expression patterns of the components of the GPIb/IX/V complex. In case 1, GPIX was completely absent but residual amounts of GPIbα and GPV were detectable; in case 2, GPIbα was completely absent. We amplified the coding regions of GPIbα, GPIbß, GPV, and GPIX from the patients’ genomic DNA with the polymerase chain reaction (PCR) and sequenced the PCR products. In case 1, we identified a point mutation in the GPIX coding region that changes the codon for tryptophan-126 (TGG) to a nonsense codon (TGA). In case 2, we found a deletion of nucleotide within seven adenine repeats at the position of 1932 to 1938 in the coding region of GPIbα, which causes a frame shift that results in 58 altered amino acids and a premature stop codon. These genetic changes alter the transmembrane domain of GPIX or GPIbα and, therefore, would prevent proper insertion of the proteins in the plasma membrane. Thus, abnormality of a single component protein (GPIX or GPIbα) alters the assembly of the GPIb/IX/V complex and causes heterogenous surface expression of GPIbα, GPV and GPIX.

Résumé :
The tetrameric extracellular superoxide dismutase (EC-SOD) in human tissues and plasma has previously been found to be heterogenous with regard to heparin affinity and could be divided into at least three classes: A, lacking heparin affinity; B, with weak affinity; and C, with strong affinity. Using rigorous extraction conditions and an extensive set of anti-proteolytic agents, tissue EC-SOD is now shown to be almost exclusively of native homotetrameric C-class. Plasma EC-SOD on the other hand is shown to be mainly composed of a complex mixture of heterotetramers with modifications probably residing in the C-terminal heparin-binding domain. Proteolytic truncations appear to be a major cause of this heterogeneity. The findings suggest that, since 99% of the EC-SOD in the human body exists in the extravascular space of tissue, EC-SOD is primarily synthesized in tissues and secreted as homotetrameric native EC-SOD C. This tissue EC-SOD C should exist almost completely sequestered by heparin sulphate proteoglycans. C-terminal modifications subsequently occurring in the EC-SOD C would weaken the binding to heparan sulphate proteoglycan, facilitate entrance to the vasculature through capillaries and lymph flow, and finally result in the heterogeneous plasma EC-SOD pattern. With the new extraction and analysis procedure, the tissue content of EC-SOD is found to be higher than previously reported. It is found, for example, when compared with Mn-SOD, to be higher in umbilical cord and uterus, about equal in placenta and testis and as high as that of CuZn-SOD in umbilical cord. The findings suggest that the protection level against superoxide radicals provided by EC-SOD in the tissue interstitial space, given the small distribution volume, is not much less prominent than that bestowed on the intracellular space by CuZn-SOD and Mn-SOD.

Résumé :
BackgroundResearchers have identified about 40 genes with mutations that result in the most common cause of CKD in children, congenital anomalies of the kidney and urinary tract (CAKUT), but approximately 85% of patients with CAKUT lack mutations in these genes. The anomalies that comprise CAKUT are clinically heterogenous, and thought to be caused by disturbances at different points in kidney development. However, identification of novel CAKUT-causing genes remains difficult because of their variable expressivity, incomplete penetrance, and heterogeneity.MethodsWe investigated two generations of a family that included two siblings with CAKUT. Although the parents and another child were healthy, the two affected siblings presented the same manifestations, unilateral renal agenesis and contralateral renal hypoplasia. To search for a novel causative gene of CAKUT, we performed whole-exome and whole-genome sequencing of DNA from the family members. We also generated two lines of genetically modified mice with a gene deletion present only in the affected siblings, and performed immunohistochemical and phenotypic analyses of these mice.ResultsWe found that the affected siblings, but not healthy family members, had a homozygous deletion in the Cobalamin Synthetase W Domain–Containing Protein 1 (CBWD1) gene. Whole-genome sequencing uncovered genomic breakpoints, which involved exon 1 of CBWD1, harboring the initiating codon. Immunohistochemical analysis revealed high expression of Cbwd1 in the nuclei of the ureteric bud cells in the developing kidneys. Cbwd1-deficient mice showed CAKUT phenotypes, including hydronephrosis, hydroureters, and duplicated ureters.ConclusionsThe identification of a deletion in CBWD1 gene in two siblings with CAKUT implies a role for CBWD1 in the etiology of some cases of CAKUT.

Résumé :
Multi domain networks can be used in several scenarios including military, enterprize networks, emergency networks and many other cases. In such networks, each domain might be under its own administration. Therefore, the cooperation among domains is conditioned by individual domain policies regarding sharing information, such as network topology, connectivity, mobility, security, various service availability and so on. We propose a new architecture for Heterogeneous Multi Domain (HMD) networks, in which one the operations are subject to specific domain policies. We propose a hierarchical architecture, with an infrastructure of gateways at highest-control level that enables policy based interconnection, mobility and other services among domains. Gateways are responsible for translation among different communication protocols, including routing, signalling, and security. Besides the architecture, we discuss in more details the mobility and adaptive capacity of services in HMD. We discuss the HMD scalability and other advantages compared to existing architectural and mobility solutions. Furthermore, we analyze the dynamic availability at the control level of the hierarchy.
