Introduction. The existence of human cardiac stem cells (hCSC) and their regenerative capacity are not fully defined. The aim of this study was to identify and analyse the distribution of hCSCs by flow cytometry (FCM). Methods. Tissue samples from the left ventricle (LV) and the appendages of the right atrium (RA) and left atrium (LA) were taken during cardiac surgery. Mononuclear cells (MNCs) were isolated, labelled for the stem-cell-marker c-kit and hematopoietic-lineage markers and analysed by FCM. Results. HCSCs could be isolated from the RA, LA, and LV without significant quantitative difference between both atria (A) (RA 4.80 ± 1.76% versus LA 4.99 ± 1.69% of isolated MNCs, P=0.922). The number of hCSCs was significantly higher in both atria compared to the left ventricle (A 4.90 ± 1.29% versus LV 0.62 ± 0.14% of isolated MNCs, P=0.035). Conclusion. The atria contain a higher concentration of hCSC than the left ventricle. HCSCs located in the atria could serve as an endogenous source for heart regeneration.

PurposeInventory management systems in health-care supply chains (HCSC) have been pushed to breaking point by the COVID-19 pandemic. Unanticipated demand shocks due to stockpiling of medical supplies caused stockouts, and the stockouts triggered systematic supply chain (SC) disruptions inconceivable for risk managers working individually with limited information about the pandemic. The purpose of this paper is to respond to calls from the United Nations (UN) and World Health Organization (WHO) for coordinated global action by proposing a research agenda based on a review of current knowledge and knowledge gaps on the role of collaboration in HCSCs in maintaining optimal stock levels and reinforcing resilience against stockout disruptions during pandemics.Design/methodology/approachA systematic review was conducted, and a total of 752 articles were analyzed.FindingsCollaborative planning, forecasting and replenishment practices are under-researched in the HCSC literature. Similarly, a fragmented application of extant SC collaborative risk management capabilities undermines efforts to enhance resilience against systematic disruptions from medical stockouts. The paucity of HCSC articles in humanitarian logistics and SC journals indicates a need for more research interlinking two interdependent yet critical fields in responding to pandemics.Research limitations/implicationsAlthough based on an exhaustive search of academic articles addressing HCSCs, there is a possibility of having overlooked other studies due to search variations in language controls, differences in publication cycle time and database search engines.Originality/valueThe paper relies on COVID-19's uniqueness to highlight the limitations in optimization and individualistic approaches to managing medical inventory and stockout risks in HCSCs. The paper proposes a shift from a fragmented to holistic application of relevant collaboration practices and capabilities to enhance the resilience of HCSCs against stockout ripple effects during future pandemics. The study propositions and suggestion for an SC learning curve provide an interdisciplinary research agenda to trigger early preparation of a coordinated HCSC and humanitarian logistics response to future pandemics.

AbstractStem cell-based therapies with clinical applications require millions of cells. Therefore, repeated subculture is essential for cellular expansion, which is often complicated by replicative senescence. Cellular senescence contributes to reduced stem cell regenerative potential as it inhibits stem cell proliferation and differentiation as well as the activation of the senescence-associated secretory phenotype (SASP). In this study, we employed MHY-1685, a novel mammalian target of rapamycin (mTOR) inhibitor, and examined its long-term priming effect on the activities of senile human cardiac stem cells (hCSCs) and the functional benefits of primed hCSCs after transplantation. In vitro experiments showed that the MHY-1685‒primed hCSCs exhibited higher viability in response to oxidative stress and an enhanced proliferation potential compared to that of the unprimed senile hCSCs. Interestingly, priming MHY-1685 enhanced the expression of stemness-related markers in senile hCSCs and provided the differentiation potential of hCSCs into vascular lineages. In vivo experiment with echocardiography showed that transplantation of MHY-1685‒primed hCSCs improved cardiac function than that of the unprimed senile hCSCs at 4 weeks post-MI. In addition, hearts transplanted with MHY-1685-primed hCSCs exhibited significantly lower cardiac fibrosis and higher capillary density than that of the unprimed senile hCSCs. In confocal fluorescence imaging, MHY-1685‒primed hCSCs survived for longer durations than that of the unprimed senile hCSCs and had a higher potential to differentiate into endothelial cells (ECs) within the infarcted hearts. These findings suggest that MHY-1685 can rejuvenate senile hCSCs by modulating autophagy and that as a senescence inhibitor, MHY-1685 can provide opportunities to improve hCSC-based myocardial regeneration.

Background: The present study investigated the effectiveness of stimulation applied at cervical levels on pain and Parkinson’s disease (PD) symptoms using either tonic or burst stimulation mode. Methods: Tonic high cervical spinal cord stimulation (T-HCSCS) was applied on six PD patients suffering from low back pain and failed back surgery syndrome, while burst HCSCS (B-HCSCS) was applied in twelve PD patients to treat primarily motor deficits. Stimulation was applied percutaneously with quadripolar or octapolar electrodes. Clinical evaluation was assessed by the Unified Parkinson’s Disease Rating Scale (UPDRS) and the Hoehn and Yahr (H&Y) scale. Pain was evaluated by a visual analog scale. Evaluations of gait and of performance in a cognitive motor task were performed in some patients subjected to B-HCSCS. One patient who also suffered from severe autonomic cardiovascular dysfunction was investigated to evaluate the effectiveness of B-HCSCS on autonomic functions. Results: B-HCSCS was more effective and had more consistent effects than T-HCSCS in reducing pain. In addition, B-HCSCS improved UPDRS scores, including motor sub-items and tremor and H&Y score. Motor benefits appeared quickly after the beginning of B-HCSCS, in contrast to long latency improvements induced by T-HCSCS. A slight decrease of effectiveness was observed 12 months after implantation. B-HCSCS also improved gait and ability of patients to correctly perform a cognitive–motor task requiring inhibition of a prepared movement. Finally, B-HCSCS ameliorated autonomic control in the investigated patient. Conclusions: The results support a better usefulness of B-HCSCS compared to T-HCSCS in controlling pain and specific aspects of PD motor and non-motor deficits for at least one year.

ObjectivesWe compared the care services use and medical institutional deaths among older adults across four home care facility types.DesignThis was a retrospective cohort study.SettingWe used administrative claims data from April 2014 to March 2017.ParticipantsWe included 18 347 residents of Fukuoka Prefecture, Japan, who received home care during the period, and aged ≥75 years with certified care needs of at least level 3. Participants were categorised based on home care facility use (ie, general clinics, Home Care Support Clinics/Hospitals (HCSCs), enhanced HCSCs with beds and enhanced HCSCs without beds).Primary and secondary outcome measuresWe used generalised linear models (GLMs) to estimate care utilisation and the incidence of medical institutional death, as well as the potential influence of sex, age, care needs level and Charlson comorbidity index as risk factors.ResultsThe results of GLMs showed the inpatient days were 54.3, 69.9, 64.7 and 75.0 for users of enhanced HCSCs with beds, enhanced HCSCs without beds, HCSCs and general clinics, respectively. Correspondingly, the numbers of home care days were 63.8, 51.0, 57.8 and 29.0. Our multivariable logistic regression model estimated medical institutional death rate among participants who died during the study period (n=9919) was 2.32 times higher (p<0.001) for general clinic users than enhanced HCSCs with beds users (relative risks=1.69, p<0.001).ConclusionsParticipants who used enhanced HCSCs with beds had a relatively low inpatient utilisation, medical institutional deaths, and a high utilisation of home care and home-based end-of-life care. Findings suggest enhanced HCSCs with beds could reduce hospitalisation days and medical institutional deaths. Our study warrants further investigations of home care as part of community-based integrated care.

As one of the key technologies to solve the problem of high short-circuit current, the fault current limiter (FCL) has become a research hotspot in China and abroad. The overvoltage and protection measures of the FCL are the key technologies for its application. Therefore, this paper studies the lightning intruding overvoltage and protection measures for a 500 kV FCL based on a high coupled split reactor (HCSR). Firstly, according to the main topology of the system and the 500 kV HCSR-FCL structure, the lightning intruding overvoltage simulation model of the 500 kV station, including the nearby transmission lines, is established on the PSCAD (Power Systems Computer Aided Design) program. Secondly, the lightning overvoltage of the equipment in the station and the components of the HCSR-FCL are simulated and analyzed when the transmission lines nearby are subjected to lightning shielding failure and back flashover. Meanwhile, the influence of the HCSR-FCL on the lightning overvoltage of the equipment in the station are compared and analyzed before and after the HCSR-FCL is installed. The simulation results show that the overvoltage of the equipment in the station and the components of the HCSR-FCL is more serious when the shielding failure occurs in the transmission lines nearby. The HCSR-FCL can reduce the lightning overvoltage of the equipment in the station, but the maximum inter-terminal and inter-arm lightning overvoltage of the HCSR can reach 1064 kV and 790 kV, respectively, under the current limiting state and the current sharing state. Finally, methods of increasing the arresters on the transmission lines side of the HCSR-FCL and shunt capacitor between each module of the HCSR-FCL are proposed to reduce the lightning overvoltage. The lightning impulse withstand voltage of each component of the HCSR is also proposed: The inter-terminal lightning impulse withstand voltage of HCSR is 170 kV. The inter-arm lightning impulse withstand voltage of HCSR is 200 kV. The terminal-to-ground lightning impulse withstand voltage of the HCSR-FCL is 1550 kV.

Serum alpha fetoprotein (AFP) is the most widely used tumor marker in detecting patients with hepatocellular carcinoma (HCC). However, it has been indicated that HCCR-1 (human cervical cancer oncogene 1) might be supplementary to AFP in the detection. We conducted a prospective study in 120 normal and 524 liver disease patients to evaluate the significance of simultaneous measurement of 2 tumor markers (AFP and HCCR-1) in the diagnosis of HCC through the cohort study in Korea and China. We also performed immunohistochemical studies using 25 normal subjects (N), 32 liver cirrhosis (LC) and 116 HCC tissues. The sensitivities of AFP (20 ng/mL) and HCCR-1 (10 ng/mL) in HCC were 55.8% (164/294) and 44.2% (130/294), respectively. When AFP was combined with HCCR-1, sensitivities increased to 4.2% (N), 12.7% (chronic hepatitis; CH), 50.0% (LC), and 77.2% (HCC), respectively. Although there was no significant difference in the diagnostic rate for HCC between AFP and HCCR-1, many cases for AFP-negative HCC were positive for HCCR-1 and vice versa. Moreover, the combined use of AFP and HCCR-1 improved the diagnostic rate to 70.8% in small HCC (< 2 cm) and 81.6% in large HCC (≥ 2 cm), respectively. AFP and HCCR-1 are independent markers. Our result suggests that the HCCR-1 could be an useful biomarker for HCC while the diagnostic rate could be significantly improved in the combined use of HCCR-1 and AFP.

Hepatocellular carcinoma (HCC) is the fifth most common cancer worldwide malignancy and the third leading cause of cancer death in patients. Several studies demonstrated that hepatic cancer stem cells (HCSCs), also called tumor-initiating cells, are involved in regulation of HCC initiation, tumor progression, metastasis development, and drug resistance. Despite the extensive research, the underlying mechanisms by which HCSCs are regulated remain still unclear. MicroRNAs (miRNAs) are able to regulate a lot of biological processes such as self-renewal and pluripotency of HCSCs, representing a new promising strategy for treatment of HCC chemotherapy-resistant tumors. In this review, we synthesize the latest findings on therapeutic regulation of HCSCs by miRNAs, in order to highlight the perspective of novel miRNA-based anticancer therapies for HCC treatment.

High cervical spinal cord stimulation (HCSCS) was found to have therapeutic effects on Parkinsonian gait disturbance. However, the results were inconsistent and confounded with symptoms of pain. This study aimed to reveal the gait and dysarthric effects of HCSCS in PD (Parkinson’s disease) and MSA-P (Parkinson variant of multiple system atrophy) patients without pain. Three PD and five MSA-P patients without painful comorbidities were assessed for gait performance and speech before SCS surgery and at 3- and 6-month follow-up. Stride length and the time spent in the Timed Up-and-Go task showed little change after HCSCS surgery. Overall voice quality (measured by dysphonia severity index) and perceptual speech intelligence improved significantly at 3 months, but improvements slightly diminished at 6 months postoperatively. Change in quality of life (measured by 8-item Parkinson’s disease questionnaire) was also notable at 3 months but narrowed over time following HCSCS. In conclusion, HCSCS showed therapeutic effects in improving the dysarthria but not gait disturbance in pain-free PD and MSA-P patients.

Hepatocellular carcinoma (HCC) is one of the most frequent malignant tumors in the world. The only serological marker widely used for the diagnosis of HCC is alpha-fetoprotein (AFP). Despite that AFP is widely used for the diagnosis of HCC, it has a limit as a serological marker due to its low sensitivity and specificity. The human cervical cancer proto-oncogene 1 (HCCR-1) was previously reported as a new biomarker for HCC. To further evaluate the HCCR-1 as a biomarker for HCC, we conducted the prospective cohort study. We evaluated the significance of simultaneous measurement of 2 tumor markers in the diagnosis of HCC in China, Japan and Korea. Two markers for HCC, AFP and HCCR-1, were measured in the sera obtained from 1,338 patients at the time of initial diagnosis of HCC. Of the 1338 HCC patients, 616 (46%) and 686 (51.3%) were sero-positive for AFP and HCCR-1, respectively. The positive rate for HCC was increased up to 74.1% in combined use of AFP and HCCR-1. Many cases (54%) for AFP-negative HCC were positive for HCCR-1 and vice versa. More importantly, the diagnostic rate for small HCC (< 2 cm) was significantly improved in the combined analysis of AFP and HCCR-1 to 56.9% although it was only 40.1% and 23.4% in the single analysis of HCCR-1 and AFP, respectively. Our result suggests that the HCCR-1 could be an useful biomarker for HCC while the diagnostic rate could be significantly improved in the combined use of HCCR-1 and AFP.

Aldehyde dehydrogenase 1 (ALDH1) is a cytosolic marker of cancer stem cells (CSCs), which are a sub-population within heterogeneous tumor cells. CSCs associate with therapy-resistance, self-renewal, malignancy, tumor-relapse, and reduced patient-survival window. ALDH1-mediated aldehyde scavenging helps CSCs to survive a higher level of oxidative stress than regular cancer cells. Cruciferous vegetable-derived phenethyl isothiocyanate (PEITC) selectively induces reactive oxygen species (ROS), leading to apoptosis of cancer cells, but not healthy cells. However, this pro-oxidant role of PEITC in CSCs is poorly understood and is investigated here. In a HeLa CSCs model (hCSCs), the sphere-culture and tumorsphere assay showed significantly enriched ALDHhi CSCs from HeLa parental cells (p < 0.05). Aldefluor assay and cell proliferation assay revealed that PEITC treatments resulted in a reduced number of ALDHhi hCSCs in a concentration-dependent manner (p < 0.05). In the ROS assay, PEITC promoted oxidative stress in hCSCs (p ≤ 0.001). Using immunoblotting and flow cytometry techniques, we reported that PEITC suppressed the cancer-associated transcription factor (Sp1) and a downstream multidrug resistance protein (P-glycoprotein) (both, p < 0.05). Furthermore, PEITC-treatment of hCSCs, prior to xenotransplantation in mice, lowered the in vivo tumor-initiating potential of hCSCs. In summary, PEITC treatment suppressed the proliferation of ALDH1 expressing cancer stem cells as well as key factors that are involved with drug-resistance, while promoting oxidative stress and apoptosis in hCSCs.

ABSTRACT Numerical relativity predicts that the coalescence of a black hole (BH) binary causes the newly formed BH to recoil, and evidence for such recoils has been found in the gravitational waves observed during the merger of stellar-mass BHs. Recoiling (super)massive BHs are expected to reside in hypercompact stellar clusters (HCSCs). Simulations of galaxy assembly predict that hundreds of HCSCs should be present in the halo of a Milky Way (MW)-type galaxy, and a fraction of those around the MW should have magnitudes within the sensitivity limit of existing surveys. However, recoiling BHs and their HCSCs are still waiting to be securely identified. With the goal of enabling searches through recent and forthcoming data bases, we improve over existing literature to produce realistic renditions of HCSCs bound to BHs with a mass of 105 M⊙. Including the effects of a population of blue stragglers, we simulate their appearance in Pan-STARRS and in forthcoming Euclid images. We also derive broad-band spectra and the corresponding multiwavelength colours, finding that the great majority of the simulated HCSCs fall on the colour–colour loci defined by stars and galaxies, with their spectra resembling those of giant K-type stars. We discuss the clusters properties, search strategies, and possible interlopers.

Activation of the cellular signaling pathways can induce sorafenib-resistant hepatocellular carcinoma (HCCR). In this work, the PI3K/mTOR inhibitor GSK1059615 inhibited the proliferation and invasion of HCCR cells. PLGA-PEG-mal diblock copolymer was used to load GSK1059615 and sorafenib, and the vector was further modified with GPC3 antibody (hGC33) to obtain hGC33-modified GSK1059615 and sorafenib-loaded nanoparticles (Ab-G/S-NP). Ab-G/S-NP regulated the activation of cellular signaling pathways in HCCR cells by inhibiting the expression and activation of NF-κB and downregulating the level of programmed cell death 1 ligand 1(PD-L1) to reverse drug resistance of HCCR cells to sorafenib. In this paper, we develop a Hermite cubic spline collocation method (HCSCM) for solving variable-order nonlinear fractional differential equations, which apply C1-continuous nodal basis functions to an approximate problem. We also verify that the order of convergence of the HCSCM is about O(hmin{4−α,p}) while the interpolating function belongs to Cp(p≥1), where h is the mesh size and α the order of the fractional derivative. Many numerical tests are performed to confirm the effectiveness of the HCSCM for fractional differential equations, which include Helmholtz equations and the fractional Burgers equation of constant-order and variable-order with Riemann-Liouville, Caputo and Patie-Simon sense as well as two-sided cases.

Background. Lung cancer has a poor prognosis and a high mortality rate, and patients may develop multidrug resistance. Sparganii Rhizoma-Curcumae Rhizoma (HCSC), the classic herbal drug combination of traditional Chinese medicine (TCM), is commonly used in treating tumors, but its molecular mechanism is still unclear. Method. We explored the possible mechanisms underlying the antitumor effect of HCSC using network pharmacology. The bioactive components of HCSC and their targets were collected from the TCM Systems Pharmacology (TCMSP) database and PharmMapper. Gene Ontology (GO) and KEGG enrichment analyses were performed; the GeneMANIA platform was used for the functional enrichment analysis of the core targets and their neighboring genes. Molecular docking was performed between the bioactive components and core targets. HCSC freeze-dried powder was prepared, and the bioactive components were verified by liquid chromatography- (LC-) mass spectrometry (MS). Human lung adenocarcinoma H1975 cells were cultured to verify in vitro the molecular mechanism of action of HCSC in treating lung cancer, as predicted by network pharmacology. Finally, we used the Symmap database to predict the relationship between the herb and TCM syndrome. Result. A total of seven bioactive components were identified by network pharmacological analysis. Through enrichment analyses, it was found that the mechanism of action mainly involved mitochondrial-mediated caspase-dependent cell apoptosis signaling pathways. The results of molecular docking showed that the bioactive components in HCSC have a good affinity with the target proteins (ALB, BCL2L1, ESR1, HRAS, MAP2K1, MAPK14, and SIRT1). LC-MS confirmed that formononetin and bisdemethoxycurcumin were present in the HCSC freeze-dried powder, consistent with the prediction. The results of in vitro experiments on NCI-H1975 cells confirmed that HCSC can upregulate the mitochondrial-mediated caspase-dependent apoptosis signaling pathway by inducing the cleavage of caspase-3, caspase-9, and PARP, consistent with the network pharmacology prediction. Further, the qi deficiency and blood stasis associated with TCM syndrome can be treated with HCSC.

Abstract Deep Neural Networks (DNNs) have shown their powerful performance in classification; however, the robustness issue of DNNs has arisen as one primary concern, e.g., adversarial attack. So far as we know, there is not any reported work about the adversarial attack on handwritten Chinese character recognition (HCCR). To this end, the classical adversarial attack method (i.e., Projection Gradient Descent: PGD) is adopted to generate adversarial examples to evaluate the robustness of the HCCR model. Furthermore, in the training process, we use adversarial examples to improve the robustness of the HCCR model. In the experiments, we utilize a frequently-used DNN model on HCCR and evaluate its robustness on the benchmark dataset CASIA-HWDB. The experimental results show that its recognition accuracy is decreased severely on the adversarial examples, demonstrating the vulnerability of the current HCCR model. In addition, we can improve the recognition accuracy significantly after the adversarial training, demonstrating its effectiveness.

The implementation of hydraulic calcium silicate-based endodontic cements (HCSCs) in biologically based endodontic procedures for the primary dentition has been recently investigated, focusing on the biological response of stem cells from human exfoliated deciduous teeth (SHEDs) towards them. The present systematic review aimed to present a qualitative synthesis of the available literature consisting of in vitro assays, which assessed the cytocompatibility and bioactive properties of HCSCs in direct contact with SHEDs. Following the PRISMA statement, an electronic database search was carried out in Medline, Scopus, Embase, Web of Science, and SciELO on March 31st and updated on November 16th, 2020. In vitro studies evaluating the biological response of SHEDs to the treatment with HCSCs were eligible. Within the term biological response, assays assessing the cytocompatibility (i.e., cell viability, migration, proliferation), cell plasticity or differentiation (i.e., osteo/odontogenic marker expression), and bioactivity or biomineralization (i.e., mineralized nodule formation) were included. A total of seven studies were included after the selection process. The study sample comprised an extensive range of cell viability, migration, proliferation, adhesion, and bioactivity assays regarding the biological response of SHEDs towards five different commercially available HCSCs (MTA, ProRoot MTA, Biodentine, iRoot BP Plus, and Theracal LC). Biodentine, MTA, and iRoot BP Plus showed significant positive results in cytocompatibility and bioactivity assays when cultured with SHEDs. The results from in vitro assays assessing the cytocompatibility and bioactivity of the HCSCs MTA, Biodentine, and iRoot BP Plus towards SHEDs support their use in vital pulp treatment for the primary dentition.

ABSTRACT Infection of CD4-positive cells by human immunodeficiency virus type 1 (HIV-1) requires functional interaction of the viral envelope protein with a coreceptor belonging to the chemokine receptor family of seven-membrane-spanning receptors. For the majority of macrophage-tropic HIV-1 isolates, the physiologically relevant coreceptor is the human CCR-5 (hCCR-5) receptor. Although the murine homolog of CCR-5 (mCCR-5) is unable to mediate HIV-1 infection, chimeric hCCR-5/mCCR-5 molecules containing single extracellular domains derived from hCCR-5 are effective coreceptors for certain macrophage-tropic HIV-1 isolates. Here, we have sought to identify residues in hCCR-5 critical for HIV-1 infection by substitution of mCCR-5-derived residues into the context of functional chimeric hCCR-5/mCCR-5 receptor molecules. Using this strategy, we demonstrate that residues 7, 13, and 15 in the first extracellular domain and residue 180 in the third extracellular domain of CCR-5 are important for HIV-1 envelope-mediated membrane fusion. Of interest, certain substitutions, for example, at residues 184 and 185 in the third extracellular domain, have no phenotype when introduced individually but strongly inhibit hCCR-5 coreceptor function when present together. We hypothesize that these changes, which do not preclude chemokine receptor function, may inhibit a conformational transition in hCCR-5 that contributes to HIV-1 infection. Finally, we report that substitution of glycine for valine at residue 5 in CCR-5 can significantly enhance the level of envelope-dependent cell fusion by expressing cells. The diversity of the mutant phenotypes observed in this mutational analysis, combined with their wide distribution across the extracellular regions of CCR-5, emphasizes the complexity of the interaction between HIV-1 envelope and coreceptor.

With the continuous expansion of power system capacity, parallel high-voltage circuit breakers with a Highly Coupled Split Reactor (HCSR) have a potential application for limitation of high short-circuit current. However, as the parallel circuit breakers open at different time, the residual voltage of the split reactor on the post-open circuit breaker is higher and the stray capacitors of HCSR form high-order oscillating circuits, which may lead to serious transient recovery voltage (TRV). Therefore, it is necessary to analyze and simulate the transient characteristics of 500kV HCSR. Firstly, the equivalent model of HCSR circuit breakers is built in EMTP simulation plat-form, and its TRV generation is analyzed. Then, the TRV of parallel circuit breakers under different operating conditions and fault types is simulated and calculated. Finally, according to the statistical calculation results, the protection measure of 0.2uF shunt capacitance to limit rise rate of TRV is proposed.

Migratory capabilities of adult human stem cells are vital for assuring endogenous tissue regeneration and stem cell-based clinical applications. Although human blood serum has been shown to be beneficial for cell migration and proliferation, little is known about its impact on the migratory behavior of cardiac stem cells and underlying signaling pathways. Within this study, we investigated the effects of human blood serum on primary human cardiac stem cells (hCSCs) from the adult heart auricle. On a technical level, we took advantage of a microfluidic cultivation platform, which allowed us to characterize cell morphologies and track migration of single hCSCs via live cell imaging over a period of up to 48 h. Our findings showed a significantly increased migration distance and speed of hCSCs after treatment with human serum compared to control. Exposure of blood serum-stimulated hCSCs to the p38 mitogen-activated protein kinase (p38-MAPK) inhibitor SB239063 resulted in significantly decreased migration. Moreover, we revealed increased phosphorylation of heat shock protein 27 (Hsp27) upon serum treatment, which was diminished by p38-MAPK-inhibition. In summary, we demonstrate human blood serum as a strong inducer of adult human cardiac stem cell migration dependent on p38-MAPK/Hsp27-signalling. Our findings further emphasize the great potential of microfluidic cultivation devices for assessing spatio-temporal migration dynamics of adult human stem cells on a single-cell level.

Multimodal summarization with multimodal output (MSMO) generates a summary with both textual and visual content. Multimodal news report contains heterogeneous contents, which makes MSMO nontrivial. Moreover, it is observed that different modalities of data in the news report correlate hierarchically. Traditional MSMO methods indistinguishably handle different modalities of data by learning a representation for the whole data, which is not directly adaptable to the heterogeneous contents and hierarchical correlation. In this paper, we propose a hierarchical cross-modality semantic correlation learning model (HCSCL) to learn the intra- and inter-modal correlation existing in the multimodal data. HCSCL adopts a graph network to encode the intra-modal correlation. Then, a hierarchical fusion framework is proposed to learn the hierarchical correlation between text and images. Furthermore, we construct a new dataset with relevant image annotation and image object label information to provide the supervision information for the learning procedure. Extensive experiments on the dataset show that HCSCL significantly outperforms the baseline methods in automatic summarization metrics and fine-grained diversity tests.

We report the detection, for the first time in space, of cyano thioformaldehyde (HCSCN) and propynethial (HCSCCH) towards the starless core TMC-1. Cyano thioformaldehyde presents a series of prominent a- and b-type lines, which are the strongest previously unassigned features in our Q-band line survey of TMC-1. Remarkably, HCSCN is four times more abundant than cyano formaldehyde (HCOCN). On the other hand, HCSCCH is five times less abundant than propynal (HCOCCH). Surprisingly, we find an abundance ratio HCSCCH/HCSCN of ∼0.25, in contrast with most other ethynyl-cyanide pairs of molecules for which the CCH-bearing species is more abundant than the CN-bearing one. We discuss the formation of these molecules in terms of neutral-neutral reactions of S atoms with CH2CCH and CH2CN radicals as well as of CCH and CN radicals with H2CS. The calculated abundances for the sulphur-bearing species are, however, significantly below the observed values, which points to an underestimation of the abundance of atomic sulphur in the model or to missing formation reactions, such as ion-neutral reactions.

Kecelakaan tersebut terjadi karena belum adanya palang kereta, maupun kelalaian dari petugas operator palang pintu yang mengoprasikan palang pintu (human error). Untuk mengurangi hal tersebut sebaiknya palang pintu kereta di buat secara otomatis. Untuk itu perlu di kembangkan teknologi yang mampu mengatasi hal tersebut maka di buatlah prototipe berupa pintu perlintasan kereta otomatis. Komponen utama mikrokontroler dan sensor HCSR-04 berdasarkan uji coba alat ini berjalan lancar. Palang pintu yang membaca sensor HCSR-04 akan menutup dan sebaliknya jika sensor HCSR-04 tidak menbaca adanya kereta api yang lewat maka akan terbuka. A hiperplasia congênita das supra-renais (HCSR) representa um grupo de doenças genéticas que comprometem a síntese de cortisol devida à deficiência em uma das enzimas responsáveis pela esteroidogênese supra-renal. Na deficiência da 21-hidroxilase (D21OH), responsável por mais de 90% dos casos de HCSR, a secreção androgênica supra-renal está aumentada. A forma clássica de D21OH é tratada com glicocorticóide, repondo-se mineralocorticóide quando necessário. O uso continuado de corticosteróides pode comprometer o crescimento através de diferentes mecanismos. Por outro lado, a excessiva secreção adrenocortical dos esteróides sexuais pode levar à redução do tempo de crescimento, tanto por aceleração da idade óssea quanto pela possibilidade de indução de puberdade precoce central. A combinação dos efeitos da corticoterapia e do excesso de esteróides sexuais sobre o crescimento faz com que as crianças com HCSR estejam sob risco de baixa estatura. Daí a importância da cuidadosa avaliação e monitorização do crescimento e da evolução puberal desses pacientes. Igualmente importante é o emprego de glicocorticóides com menor capacidade em suprimir o crescimento. Desse modo, acreditamos que a melhor opção terapêutica para crianças com HCSR é representada pela hidrocortisona ou acetato de cortisona, empregadas na dose fisiológica e administrando-se a maior dose pela manhã.

Background.This is a single center oncological resume overlooking four decades of experience with liver transplantation (LT) for hepatocellular carcinoma (HCC).Methods.All 319 LT for HCC that were performed between 1975 and 2011 were included. Predictors for HCC recurrence (HCCR) and survival were identified by Cox regression, Kaplan-Meier analysis, Log Rank, andχ2-tests where appropriate.Results.HCCR was the single strongest hazard for survival (exp⁡B=10.156). Hazards for HCCR were tumor staging beyond the histologic MILAN (exp⁡B=3.645), bilateral tumor spreading (exp⁡B=14.505), tumor grading beyond G2 (exp⁡B=8.668), and vascular infiltration of small or large vessels (exp⁡B=11.612,exp⁡B=18.324, resp.). Grading beyond G2 (exp⁡B=10.498) as well as small and large vascular infiltrations (exp⁡B=13.337,exp⁡B=16.737, resp.) was associated with higher hazard ratios for long-term survival as compared to liver transplantation beyond histological MILAN (exp⁡B=4.533). Tumor dedifferentiation significantly correlated with vascular infiltration (χ2p=0.006) and intrahepatic tumor spreading (χ2p=0.016).Conclusion.LT enables survival from HCC. HCC dedifferentiation is associated with vascular infiltration and intrahepatic tumor spreading and is a strong hazard for HCCR and survival. Pretransplant tumor staging should include grading by biopsy, because grading is a reliable and easily accessible predictor of HCCR and survival. Detection of dedifferentiation should speed up the allocation process.

To examine factors influencing home death, an anonymous survey was mailed to 998 home care supporting clinics (HCSCs) in the 23 wards of Tokyo, Japan. We classified the HCSCs into two types (single physician practice and multiple physician practice) and identified factors of each type of practice that predict home death. The factors associated with a greater probability of dying at home were as follows: in the multiple physician practices, collaboration with hospitals and teaching coping skills to the family members and, in the single physician practices, collaboration with clinics. Our findings suggest that home end-of-life care services are unlikely to be achieved without cooperation among service providers and without improvement of the family members' coping skills.

PurposeThis paper aims to explore the drivers of sustainable healthcare supply chain (SHCSC) performance measurement through extensive literature review and experts' opinions. The drivers are then scrutinized and their priority vector is calculated to provide quality and cost-effective healthcare supply chain (HCSC) services.Design/methodology/approachThe drivers of the SHCSC performance measurement are validated using the grey-Delphi technique. After validating the drivers, they are prioritized using the grey-analytic hierarchy process (G-AHP), a multi-criteria decision-making tool.FindingsThe findings of the study highlight the prioritized drivers based on the preferences given by the experts. The findings of the study highlight the most prioritized drivers of healthcare (HC) by-product management system, coordinating and facilitating green suppliers in the HCSC and green packaging of pharmaceutical as well as other essential items.Practical implicationsThe HCSC managers should coordinate with all the stakeholders across the supply chain and involve them in the decision-making process to make products and services greener and become complicit in complying with the sustainable policy guidelines. The study highlights the strategic policy and managerial implications for implementing sustainability in the HCSC.Originality/valueThe validation and prioritization of the drivers of SHCSC in developing nations' contexts is the key contribution of the study. Grey-AHP enables a practical approach towards enhancing the sustainability of the HCSC and opening the doors for generalizing the study for future research works.

Objective To analyze the clinical and pathological manifestations of a hepatic carcinosarcoma case with rhabdomyosarcoma components (HCSR).Methods A case of HCSR was observed by macroscopy, microscopy, immunohistochemistry and electron microscopy, along with thorough review of correlated literatures.Results The tumor tends to occur in elder patients without differences on gender. Epigarstric sicknesses, AFP rising in serum, and a mass on the right liver shown in radiography are commonly initial signs. It was composed of both hepatic carcinoma and variously differentiated sarcoma components, with identifiable rhabdomyosarcoma. Immunohistochemistry showed that the hepatic carcinosarcoma was positive of epithelial markers and mesenchymatous component was diffuse positive of Vimentin, and mosaic positive of SDHB, CD117, while rhabdomyosarcoma was positive of muscular markers. Transmission electron microscopy showed the tumor had both epithelial and rhabdomyosarcoma ultra-microstructure.Conclusion HCSR is a very rare type and highly malignant tumor with a dismal prognosis, hardly demonstrating unique clinical manifestations. Diagnosis and differential diagnosis rely on combination of histomorphology, immunohistochemistry and ultra-microstructure observation.

During aging, senescent cells accumulate in various tissues accompanied by decreased regenerative capacities of quiescent stem cells, resulting in deteriorated organ function and overall degeneration. In this regard, the adult human heart with a generally low regenerative potential is of extreme interest as a target for rejuvenating strategies with blood borne factors that might be able to activate endogenous stem cell populations. Here, we investigated for the first time the effects of human blood plasma and serum on adult human cardiac stem cells (hCSCs) and showed significantly increased proliferation capacities and metabolism accompanied by a significant decrease of senescent cells, demonstrating a beneficial serum-mediated effect that seemed to be independent of age and sex. However, RNA-seq analysis of serum-treated hCSCs revealed profound effects on gene expression depending on the age and sex of the plasma donor. We further successfully identified key pathways that are affected by serum treatment with p38-MAPK playing a regulatory role in protection from senescence and in the promotion of proliferation in a serum-dependent manner. Inhibition of p38-MAPK resulted in a decline of these serum-mediated beneficial effects on hCSCs in terms of decreased proliferation and accelerated senescence. In summary, we provide new insights in the regulatory networks behind serum-mediated protective effects on adult human cardiac stem cells.

Home cancer care research (HCCR) has accelerated, as considerable attention has been placed on reducing cancer-related health costs and enhancing cancer patients’ quality of life. Understanding the current status of HCCR can help guide future research and support informed decision-making about new home cancer care (HCC) programs. However, most current studies mainly detail the research status of certain components, while failing to explore the knowledge domain of this research field as a whole, thereby limiting the overall understanding of home cancer care. We carried out bibliometric and visualization analyses of Scopus-indexed papers related to home cancer care published between 1990–2021, and used VOSviewer scientometric software to investigate the status and provide a structural overview of the knowledge domain of HCCR (social, intellectual, and conceptual structures). Our findings demonstrate that over the last three decades, the research on home cancer care has been increasing, with a constantly expanding stream of new papers built on a solid knowledge base and applied to a wide range of research themes.

Industries demand stringent requirements towards economical machining without hindering the surface quality while cutting high carbon high chromium (HcHcr) steel. Electrical discharge machining (EDM) of HcHcr steel aims at reducing machining cost (i.e., maximize material removal rate (MRR) and minimize tool wear rate (TWR)) with good surface quality (i.e., minimize surface roughness (SR)). A comparative study was carried out on EDM of HcHcr D2 steel (DIN EN ISO 4957) by applying Taguchi L18 experimental design considering different electrode materials (copper, graphite, and brass), dielectric fluids (distilled water and kerosene), peak current, and pulse-on-time. The process performances were analyzed with respect to material removal rate, surface roughness, and tool wear rate. Pareto analysis of variance was employed to estimate the significance of the process variables and their optimal levels for achieving lower SR and TWR and higher MRR. Hybrid Taguchi-CRITIC-Utility and Taguchi-PCA-Utility methods were implemented to determine the optimal EDM parameters. Higher MRR of 0.0632 g/min and lower SR of 1.68 µm and TWR of 0.012 g/min was attained by graphite electrode in presence of distilled water as dielectric fluid compared to the brass and copper. Additionally, a metallographic analysis was carried out to study the surface integrity on the machined surfaces. Micrographic analysis of the optimal conditions showed lower surface roughness and fewer imperfections (lesser impression, waviness surface, and micro-cracks) compared to worst conditions.

Abstract The purpose of the current study was to provide useful data, which may help neurosurgeons to manage the patient dose and image quality in spinal surgery procedures, utilising a phantom and a test object. The kerma area product, cumulative dose (CD) and entrance surface dose (ESD) rate on the phantom and image intensifier were measured, for selectable fields of view (FOVs), fluoroscopy modes, two geometric magnifications and various phantom thicknesses. The images were subjectively evaluated regarding low-contrast detectability and high-contrast resolution. Signal-to-noise ratio (SNR), contrast-to-noise ratio (CNR), high-contrast spatial resolution (HCSR) and figure of merit (FOM) values were also estimated. The ESD rates increased with increasing phantom thickness, when using electronic or geometric magnification, continuous or high-definition fluoroscopy (HDF). The observers’ evaluation showed relatively slight changes in image quality when pulsed fluoroscopy was used. SNR, CNR and HCSR values decreased with increasing phantom thicknesses, while remained almost constant when using pulsed fluoroscopy. SNR and HCSR improved in HDF, while the CNR remained almost constant only for the FOVs 23 and 17 cm. By applying electronic magnification, this resulted in improved HCSR. FOM values decreased in HDF, with increasing phantom thickness and using electronic magnification. For the ‘thinnest’ patients, CD may overestimate skin dose by 25% than the actual values. Geometric magnification resulted in improved FOM, especially for low-dose fluoroscopy and FOV 23 cm. The knowledge of the increments in dose values, image quality and FOM indices concerning phantom thickness may help neurosurgeons to optimise spinal surgery procedures by selecting the appropriate operational parameters, which could contribute toward the establishment of a radiation protection culture.

Bluetooth is a widespread wireless technology standard for limited wireless networks that permits Bluetooth devices to create a one-hop (piconet) or multi-hop (scatternet) network. During data transmission, a large number of links passing through a single master or bridge device may create congestion problems in a Bluetooth network. Therefore, routing in a multi-hop dynamic Bluetooth network, where a number of routing masters and routing bridges exist, sometimes create technical problems in a scatternet. Mobility and failure of routing devices disconnects the routing links and link reconstruction process consumes more resources that eventually decrease the performance. In this paper, Hybrid Congestion Sharing and Route Repairing protocol for Bluetooth networks” (HCSRR) is proposed. The objective of this paper is to provide an efficient technique for scatternet congestion avoidance and route maintenance. The proposed protocol is implemented and compared with most relevant protocols. From simulation results, it is observed that the HCSRR outperforms the existing protocols.

With the outstanding performance in 2014 at the ImageNet Large-Scale Visual Recognition Challenge 2014 (ILSVRC14), an effective convolutional neural network (CNN) model named GoogLeNet has drawn the attention of the mainstream machine learning field. In this paper we plan to take an insight into the application of the GoogLeNet in the Handwritten Chinese Character Recognition (HCCR) on the database HCL2000 and CASIA-HWDB with several necessary adjustments and also state-of-the-art improvement methods for this end-to-end approach. Through the experiments we have found that the application of the GoogLeNet for the Handwritten Chinese Character Recognition (HCCR) results into significant high accuracy, to be specific more than 99% for the final version, which is encouraging for us to further research.

A dosagem da 17-hidroxiprogesterona (17OHP) é usada para screening de hiperplasia congênita de adrenal (HCSR) por defeito da 21-hidroxilase e, nesta situação, se encontra muito aumentada. Pode estar alterada em mulheres com síndrome de ovários policísticos (SOP), porém a freqüência e a magnitude desta alteração foram pouco descritas. Analisamos retrospectivamente queixas, índice de massa corpórea (IMC), dosagens na fase folicular precoce de LH, FSH, testosterona livre, 17OHP, sulfato de deidroepiandrosterona (SDHEA) e volume ovariano ao ultra-som de 83 mulheres no menacme com SOP, antes de iniciar tratamento. HCSR foi excluída pela prova de ACTH nas que apresentaram valores basais de 17OHP maiores que 200ng/dL. A 17OHP esteve acima do valor de referência em 67,5% dos casos comparada a 38,6% para o LH; 10,8% para testosterona livre e 4,9% para SDHEA. A relação LH/FSH esteve maior que 1 em 50,6% das mulheres. O volume de pelo menos um dos ovários esteve aumentado em 53,5% das pacientes. O aumento médio da 17OHP foi de 30% acima do limite superior da normalidade (mediana 18%). Correlacionou-se inversamente com os valores de FSH e houve uma tendência de relação direta com os níveis de SDHEA. Das dosagens analisadas, a 17OHP foi a mais freqüentemente aumentada, sugerindo que além de funcionar como screening para HCSR, aumentos discretos desse hormônio possam apoiar o diagnóstico de SOP dentro de um quadro clínico compatível

For the identification of a stem cell population, the comparison of transcriptome data enables the simultaneous analysis of tens of thousands of molecular markers and thus enables the precise distinction of even closely related populations. Here, we utilized global gene expression profiling to compare two adult human stem cell populations, namely neural crest-derived inferior turbinate stem cells (ITSCs) of the nasal cavity and human cardiac stem cells (hCSCs) from the heart auricle. We detected high similarities between the transcriptomes of both stem cell populations, particularly including a range of neural crest-associated genes. However, global gene expression likewise reflected differences between the stem cell populations with regard to their niches of origin. In a broader analysis, we further identified clear similarities between ITSCs, hCSCs and other adherent stem cell populations compared to non-adherent hematopoietic progenitor cells. In summary, our observations reveal high similarities between adult human cardiac stem cells and neural crest-derived stem cells from the nasal cavity, which include a shared relation to the neural crest. The analyses provided here may help to understand underlying molecular regulators determining differences between adult human stem cell populations.

ArgumentWhat does “life” become at a moment when biological inquiry proceeds by manufacturing biological artifacts and systems? In this article, I juxtapose two radically different communities, synthetic biologists and Hyperbolic Crochet Coral Reef crafters (HCCR). Synthetic biology is a decade-old research initiative that seeks to merge biology with engineering and experimental research with manufacture. The HCCR is a distributed venture of three thousand craftspeople who cooperatively fabricate a series of yarn and plastic coral reefs to draw attention to the menace climate change poses to the Great Barrier and other reefs. Interpreting these two groups alongside one another, I suggest that for both, manufacturing biological artifacts advances their understandings of biology: in a rhetorical loop, they build new biological things in order to understand the things they are making. The resulting fabrications condense scientific and folk theories about “life” and also undo “life” as a coherent analytic object.

Background and Objectives: Symptomatic irreversible pulpitis in permanent mature teeth is a common indication for nonsurgical root canal treatment (NSRCT), but contemporary studies have reported on vital pulp therapy (VPT) applied in such teeth as a less invasive treatment. This systematic review assessed the outcomes of VPT, including partial and full pulpotomy performed with hydraulic calcium silicate cements (HCSCs) in permanent mature posterior teeth diagnosed with symptomatic irreversible pulpitis. Materials and Methods: The PRISMA guidelines were followed. The search strategy included PubMed®, EMBASE, Cochrane library and grey literature electronic databases. The quality assessment of the identified studies followed the Cochrane Collaboration Risk of Bias, ROBINS-I and Newcastle–Ottawa Scale tools. Results: The search of primary databases identified 142 articles, of which 9 randomized controlled trials and 3 prospective cohort studies were selected for review. The risk-of-bias was assessed as ‘high’ or ‘serious’, ‘fair’, and ‘low’ for three, seven and two articles, respectively. One to five years after VPT using HCSCs, the success rates mostly ranged from 78 to 90%. Based on two articles, the outcomes of the VPT and NSRCT were comparable at one and five years. Despite the necessity for the intra-operative pulp assessment in VPT procedures, the majority of the studies did not fully report on this step or on the time needed to achieve hemostasis. Small sample sizes, of under 23 teeth, were reported in three studies. Conclusions: The reviewed 12 articles reported favorable outcomes of the VPT performed with HCSCs in permanent mature posterior teeth with symptomatic irreversible pulpitis, with radiographic success in the range of 81 to 90%. Two articles suggested comparable outcomes of the VPT and root canal treatment. Universal case selection and outcome criteria needs to be established for VPT when considered as an alternative to NSRCT. This evidence supports the need for further research comparing longer-term outcomes of both of the treatment modalities.

This paper presents an effective hybrid optimization technique based on a chaotic sine cosine algorithm (CSCA) and pattern search (PS) for the coordinated design of power system stabilizers (PSSs) and static VAR compensator (SVC)-based controllers. For this purpose, the design problem is considered as an optimization problem whose decision variables are the controllers’ parameters. Due to the nonlinearities of large, interconnected power systems, methods capable of handling any nonlinearity of power networks are preferable. In this regard, a nonlinear time domain-based objective function was used. Then, the proposed hybrid chaotic sine cosine pattern search (hCSC-PS) algorithm was employed for solving this optimization problem. The proposed method employed the global search ability of SCA and the local search ability of PS. The performance of the new hCSC-PS was investigated using a set of benchmark functions, and then the results were compared with those of the standard SCA and some other methods from the literature. In addition, a case study from the literature is considered to evaluate the efficiency of the proposed hCSC-PS for the coordinated design of controllers in the power system. PSSs and additional SVC controllers are being considered to demonstrate the feasibility of the new technique. In order to ensure the robustness and performance of the proposed controller, the objective function is evaluated for various extreme loading conditions and system configurations. The numerical investigations show that the new approach may provide better optimal damping and outperforms previous methods. Nonlinear time-domain simulation shows the superiority of the proposed controller and its ability in providing efficient damping of electromechanical oscillations.

Hydrocarbon supply chain (HCSC) is a complex network that extends from oil and gas fields to demand nodes. Integrating operation and maintenance activities along this complex network is crucial since the hydrocarbon industry is the most influential sector in the world economy, and any disruptions or variations in hydrocarbon product supply will affect the whole world economy. Therefore, effective and thoughtful maintenance extends the life of an asset and enhances its reliability. To prevent huge losses in production and ultimately satisfy customer needs, the maintenance jobs are preferred to be performed during times of low demand. Thus, operation planning and maintenance scheduling decisions are dependent and should be optimized simultaneously. Therefore, the aim of this study is to develop an integrated mathematical model for the operation and maintenance planning of the oil and gas supply chain. The utility of the proposed model has been demonstrated using the Saudi Arabian HCSC. The proposed model effectively produces optimal operation and maintenance schedule decisions. A sensitivity analysis was conducted to study the effect of critical parameters on the obtained results.
