Résumé :
An environmental kindly technique, subcritical water extraction (SWE) are based on using water as extraction solvent at temperatures between 100 °C and 374 °C. Increasing the temperature at moderate pressure also reduces the surface tension and viscosity of water causes the polarity of subcritical water is comparable to organic solvents. Therefore, the subcritical water could be improved the competency for the extraction. The aim of this work was to study the flavonoid content of Emilia sonchifolia (L.) using different extraction procedures (SWE and the traditional extraction or ethanolic extraction). The results revealed that quercetin, a plant-derived flavonoid, was a major component in both extraction procedures. The use of SWE provided higher quercetin content and antioxidant activity. Quercetin content by SWE and traditional extraction were 45.92 mg/ml and 39.94 mg/ml, respectively. The EC50 (Effective Concentration, 50%) of SWE and traditional extraction were 496 and 555.67 mg/ml, respectively. Additionally, this work demonstrated that the traditional time-consuming techniques for 12 hours of the extraction of flavonoids could be substituted for the SWE technique within 1 hour. Consequently, the capability of SWE technique was elaborately evaluated and revealed on this work. Keywords: Subcritical water; Emilia sonchifolia (L.)

Résumé :
Suppliers would like to capitalize on buyers' willingness to pay a premium for goods that use environmentally friendly (green) production techniques, but they face a problem of asymmetric information: buyers cannot determine if a supplier is environmentally friendly. Since the green technique is generally more costly than environmentally unfriendly (brown) techniques, suppliers would be disinclined to choose a green technique, yielding larger pollution flows. One possible remedy is “eco-labeling,” where a third party certifies suppliers' products. This paper models certification as a noisy test, with green suppliers more likely to pass than brown suppliers. Two models are considered, one where suppliers' production techniques are exogenous (a short-run setting), and one where they can choose their technique (a long-run setting). Results show that eco-labeling can either raise or lower the volume of green units, and raise or lower net social surplus; increases in social surplus are more likely in the long-run.

Résumé :
As mentioned by Sardinia Region: ''Sustainable Development is what satisfies present's needs without shattering those of future generations, thanks to smart use of environmental resources and without waists''. This research aims to use a current resource available in high quantities in Sardinia to optimize the uses of the extracted materials and not to take just advantage of the Region. The mining activity in Sardinia, which is very important since ‘60s, during these years, has produced huge amounts of granite by-products. The ornamental use of granite is an important money source for Sardinia. Unfortunately, as the virgin material extracted must have high aesthetic qualities, many rock blocks are rejected. This research has the aim of making the most of the material stored in a quarry sites and of optimizing the uses of resource stone examined. The target is the use of granite by-products as material with high mechanical featured to be used for road pavements, from the foundation to surface. This research gives you the opportunity to make the most of regional resources, to minimize the thickness of asphalt, reducing the maintenance and realization costs. This is a good start for the Island to make money of something easy to export. Granite by-products will be used for the construction of innovative road pavement design. The Inverted Pavement Technique, studied and developed in South Africa since 1950, is going to be used for road infrastructure. In particular, I have focused the attention on the behavior of granular base layer. Thanks to this technique is actually possible to take advantage of mechanical features of base layer, creating base layers that assure high and long lasting performances with almost zero environmental costs. The Project is finalized to a sustainable design by using resources, considered as waste so far, present in the Sardinian Land and the minimal use of exhaustible and expensive row materials as asphalt layers. The increasing costs of petrol products and their limited availability leaded to find alternative solutions to flexible infrastructure everywhere in the world. Another target was to make sure that European and South African Regulation matched regarding granular aggregates. Through laboratory testing physical, chemical, and mechanical features of granite by-products were analyzed comparing to Dolerite, used in South Africa in the Inverted Pavement Technique.

Résumé :
The largest class of cell surface receptors in mammalian genomes is the superfamily of G protein-coupled receptors (GPCRs) which are activated by a wide range of extracellular responses such as hormones, pheromones, odorants, and neurotransmitters. Drugs which have therapeutic effects on a wide range of diseases are act on GPCRs. In contrast to traditional idea, it is recently getting accepted that G-protein coupled receptors can form homo- and hetero-dimers and this interaction could have important role on maturation, internalization, function or/and pharmacology. Bimolecular fluorescence complementation technique (BiFC)
is an innovative approach based on the reassembly of protein fragments which directly report interactions. In our study we implemented this technique for detecting and visualizing the GPCR interactions in yeast cells. The enhanced green fluorescent protein (EGFP) fractionated into two fragments at genetic level which does not possess fluorescent function. The target proteins which are going to be tested in terms of interaction are modified with the non-functional fragments, to produce the fusion proteins. The interaction between two target proteins, in this study Ste2p receptors which are alpha pheromone receptors from Saccharomyces cerevisiae, enable the fragments to come in a close proximity and reassemble. After reassembly, EGFP regains its fluorescent function which provides a direct read-out for the detection of interaction. Further studies are required to determine subcellular localization of the interaction. Moreover, by using the fusion protein partners constructed in this study, effects of agonist/antagonist binding and post-translational modifications such as glycosylation and phosphorylation can be examined. Apart from all, optimized conditions for BiFC technique will guide for revealing new protein-protein interactions.

Texte intégral
Résumé :

The challenge for the future was defined by the Brundtland Commission (1987) and by the Rio Declaration (1992), in which the fundamental principles for achieving a sustainable development were provided. Sustainable chemistry can be defined as the contribution of chemistry to the implementation of the Rio Declaration. This thesis shows how Pressurized Fluid Extraction (PFE) can be utilized in chemical analysis, and how this correlates to Green Chemistry.

The reliability and efficiency of the PFE technique was investigated for a variety of analytes and matrices. Applications discussed include: the extraction of the antioxidant Irganox 1076 from linear low density polyethylene, mobile forms of phosphorus in lake sediment, chlorinated paraffins from source-separated household waste, general analytical method for pesticide residues in rape seed, total lipid content in cod muscle, and squalene in olive biomass. Improved or comparable extraction yields were achieved with reduced time and solvent consumption. The decrease in use of organic solvents was 50-90%, resulting in minimal volatile organic compounds emissions and less health-work problem. Due to higher extraction temperatures and more efficient extractions, the selection of solvent is not as important as at lower temperatures, which makes it possible to choose less costly, more environmentally and health beneficial solvents. In general, extraction times are reduced to minutes compared to several hours. As a result of the very short extraction times, the amount of co-extracted material is relatively low, resulting in fewer clean-up step and much shorter analysis time. Selective extractions could be obtained by varying the solvent or solvent mixture and/or using adsorbents.

In this thesis, the PFE technique was compared to the twelve principles of Green Chemistry, and it was shown that it follows several of the principles, thus giving a major contribution to sustainable chemistry.

The basis of this psychoanalytic research is the comparative analysis between the works of D. W. Winnicott and André Green regarding the Theoretical-Clinical proposals presented by the authors to the treatment of the borderline patient, whose behavior is often characterized by a refractory dynamic to the classical treatment proposed by Sigmund Freud. Therefore, we aimed to draw a parallel between the psychoanalytic treatment sessions conducted by each of the authors, having as reference the instigating case of a borderline patient they had in common. Based on the clinical interventions that both authors report about the case, we attempt to establish which were the main theoretical aspects that supported them, as to investigate which innovations the authors bring to this type of patient, to whom they created clinical dispositives compatible with what they understand to be this psychopathology. To that end we intent to establish their main points of convergence and divergence. We concluded then that despite being quite close in some aspects, the authors propositions are, to a large extent, quite heterogeneous theories and present quite divergent clinical proposals. According to Winnicott, the regression to the dependency is the emphasis of the treatment, as a way to resume the normal process of personal maturation, amended by a traumatic situation and that needs to be lived again in analysis, this time with the support of a favorable environment to this return. To Green, the treatment is based on the creation of analytical environment favorable to the processes of verbalization and symbolization, as a possibility to give meaning to the vicissitudes of the drives as a way to the internalization of negative

Kiessling, Brittany L. « Ethnographic Investigations of Commercial Aquaculture as a Rural Development Technique in Tamil Nadu, India ». FIU Digital Commons, 2016. http://digitalcommons.fiu.edu/etd/2560.

Texte intégral
Résumé :
Since the 1960s, international aid organizations and governments have invested millions of dollars in promoting aquaculture as a way to stimulate local economies and improve food security. India is one such country, incorporating aquaculture research and extension programs as part of their development plans as early as 1971. India’s aquaculture promotion efforts gained momentum in 2004, following the Indian Ocean tsunami of 2004. The government sees aquaculture as a post-disaster development tool and a method to increase community resilience in rural areas of India. Aquaculture currently constitutes nearly half of global seafood production today. Due to this importance, and the attention such practices receive through funding and extension, many scholars have focused on the social impacts that aquaculture practices have on rural communities. In particular, scholars have investigated the effects of aquaculture on environmental conditions, food security, livelihoods, gender relations, and social conflict. However, more scholarship is needed concerning the historical legacies that have contributed to how aquaculture is promoted and practiced, particularly connections to the Green Revolution. Furthermore, there needs to be more research about commercial aquaculture as a post-disaster development strategy. My research – based on 9 months of ethnographic fieldwork and archival analysis in Tamil Nadu, India – contributes to this body of literature. I synthesized post-development theory with that of environmental risk and vulnerability, building upon the work of scholars such as James Ferguson, Tania Li, and Piers Blaikie. My analysis uncovers large disparities between the goals of aquaculture development programs and actual aquaculture outcomes. I attribute this to the technocratic governance structure of the aquaculture industry, which leads to a lack of engagement and participation between aquaculture managers, researchers, and practitioners. This lack of engagement ultimately makes the communities in which aquaculture is being practiced more vulnerable to anthropogenic and natural disturbances. Additionally, I found that aquaculture practices in the study site are causing significant changes to local agrarian structures, particularly through changes to labor. These changes have implications for social stratification and disempowerment of women. Overall, these findings contribute to the anthropological study of aquaculture as well as to theories of post-development.

Résumé :
We report a novel laser–assisted thermal poling technique for producing quasi–phase–matched (QPM) structures in planar x–cut Lithium Tantalate. As a proof of concept, low loss annealed proton exchanged (APE) waveguides were subsequently fabricated in these structures in order to demonstrate efficient second–harmonic generation (SHG) of blue light. Though others [1,2], have demonstrated higher QPM SHG conversion efficiencies in z–cut LiNbO3 and LiTaO3, we believe this process for x–cut material shows great promise for achieving higher SHG efficiencies. Furthermore, this process is compatible with the preferred crystallographic orientation for on-chip integration of IO (integrated–optic) devices such as high quality intensity modulators.

Assessment of intraoperative use of indocyanine green fluorescence imaging on the number of lymph node dissection during minimally invasive gastrectomy : a systematic review and meta-analysis. Résumé :
Review question / Objective: Whether is indocyanine green fluorescence imaging-guided lymphadenectomy feasible to improve the number of lymph node dissections during radical gastrectomy in patients with gastric cancer undergoing curative resection? Condition being studied: Gastric cancer was the sixth most common malignant tumor and the fourth leading cause of cancer-related death in the world. Radical lymphadenectomy was a standard procedure in radical gastrectomy for gastric cancer. The retrieval of more lymph nodes was beneficial for improving the accuracy of tumor staging and the long-term survival of patients with gastric cancer. Indocyanine green(ICG) near-infrared fluorescent imaging has been found to provide surgeons with effective visualization of the lymphatic anatomy. As a new surgical navigation technique, ICG near-infrared fluorescent imaging was a hot spot and had already demonstrated promising results in the localization of lymph nodes during surgery in patients with breast cancer, non–small cell lung cancer, and gastric cancer. In addition, ICG had increasingly been reported in the localization of tumor, lymph node dissection, and the evaluation of anastomotic blood supply during radical gastrectomy for gastric cancer. However, it remained unclear whether ICG fluorescence imaging would assist surgeons in performing safe and sufficient lymphadenectomy.

Résumé :
Conflicts between humans and birds likely have existed since agricultural practices began. Paintings from ancient Greek, Egyptian, and Roman civilizations depict birds attacking crops. In Great Britain, recording of efforts at reducing bird damage began in the 1400s, with books on bird control written in the 1600s. Even so, the problem persists. Avian damage to crops remains an issue today, but we also are concerned with damage to homes, businesses, and aircraft, and the possibility of disease transmission from birds to humans or livestock. Bird dispersal techniques are a vital part of safely and efficiently reducing bird conflicts with humans. The bird must perceive a technique as a threat if it is to be effective. No single technique can solve all bird conflicts, but an integrated use of multiple techniques, each enhancing the other, generally provides relief.

Résumé :
This technical note is part of a series collaboratively produced by the US Army Corps of Engineers (USACE)–Institute for Water Resources (IWR) and the US Army Engineer Research and Development Center (ERDC). It describes the funding process for Exploration Green, a largescale community initiative that transformed a former golf course into a multipurpose green space with flood detention, habitat, and recreation benefits. It is one in a series of technical notes that document successful examples of funding natural infrastructure projects. The research effort is a collaboration between the Engineering With Nature® (EWN®) and Systems Approach to Geomorphic Engineering (SAGE) programs of USACE. A key need for greater application of natural infrastructure approaches is information about obtaining funds to scope, design, construct, monitor, and adaptively manage these projects. As natural infrastructure techniques vary widely by location, purpose, and scale, there is no standard process for securing funds. The goal of this series is to share lessons learned about a variety of funding and financing methods to increase the implementation of natural infrastructure projects.

Résumé :
The fourth technological revolution has brought great advances in manufacturing processes and human communications. Although processors have become increasingly efficient, both in speed, capacity and energy consumption, their functionality regarding this last point has yet to improve. The latest innovations represent an opportunity to create "green computing" and not only more environmentally friendly electronics and software, but also to use their new efficiency to improve our daily activities, as well as the designs of our cities themselves to make them more environmentally sustainable. These new computerized systems must also be applied in accordance with the socioeconomic factors that must be taken into account in order to be modified in favor of sustainability and efficiency.

Résumé :
The pipeline workforce faces several challenges over the next few years. Unprecedented numbers of experienced staff are retiring. They are being replaced by a smaller, younger workforce tasked with more responsibilities and therefore unable to develop specific expertise while roving crews fill labor gaps resulting in less experience with each individual engine compressor. To maintain, if not improve reliability and reduce maintenance costs better analysis tools are needed to fill the experience and expertise gap and assist with troubleshooting and diagnosis. Concurrently, regulatory expectations appear to be shifting from Continuous Emissions Monitoring System (CEMS) like quantifying systems to Green-light/Red-light automotive like On Board Diagnostics (OBD). OBD approaches rely on the proper operation of already installed sensors and actuators to maintain emissions compliance and can be very cost effective. This study investigated diagnostic methods to address both opportunities based on experience in related industries.

Résumé :
In this proposal, we suggest novel ways to address food safety problems in the expanding sector of fresh, ready-to-eat, fresh-cut, minimally processed fruits and vegetables. These products are becoming increasingly popular with consumers because they provide an easy way to increase consumption of phytonutrient-rich fresh foods, as recommended by health experts. However, ready-to-eat fresh fruits and vegetables may be associated with two serious health hazards. The first hazard is microbiological; human pathogens, potentially present on raw fruits and vegetables, may survive the mild interventions of minimal processing and could be further spread by cross-contamination. The second hazard is chemical; typical decontamination techniques use chlorine derivatives and strong oxidizers, which can taint food products with potentially harmful and carcinogenic byproducts. The overall goal of this work is to provide consumers with healthy and safe ready-to-eat produce that is free of microbial and chemical hazards

Résumé :
This one year BARD project was funded to develop methods to monitor promoter activity a gene expression patterns in citrus fruit. To fulfill this goal, we divided the research tasks between both labs so that the Israeli side evaluated the use of microprojectile bombardment ; a tool to evaluate transient gene expression in various citrus fruit tissues, and the US side optimized technical parameters required for Agrobacterium-mediated transformation of various citrus cultivars. Microprojectile bombardment appeared to be a very efficient method for transient gene expression analysis in citrus leaf tissues but was somewhat less applicable in fruit tissues. Nevertheless, we did succeeded to achieve significant levels of 35S-GUS gene expression in young green flavedo tissue. However, only single random spots of 35S-GUS gene expression were detected mature flavedo and in juice sacs and albedo tissue. Overall, we assume that following some more technical improvements particle bombardment could provide a useful technique to rapidly analyze promoter activity at least in the flavedo tissue. For Agrobacterium-mediated transformation, we found that shoot cultures of 'Washington' navel oranges,'Fairchild' mandarins,'Eureca' lemons,'Troyer' citrange and various grapefruits provided a more reliable and consistent source of tissue for transformation than germinated seedlings. Moreover, various growth media's (McCown, Quoirin & Lepoivre, DCR) further improved shoot and root growth relative to MS mineral media, which is commonly used. Also pure white light (using bulbs which do not emit UV or blue light) improved shoot growth in various citrus varieties, and paromomycin appeared to be a more efficient antibiotic for the selection of transgenic plants than Kanamycin. Overall, these optimizations improve transformation efficacy and shoot growth and rooting capacity. In addition to the development of transformation methods, both Israeli and US labs achieved progress in the identification of citrus fruit-specific promoters. In Israel, we isolated a 3.6 kb promoter fragment of the thiamine biosynthesis c-thi gene, which is highly expressed in fruit peel tissue, whereas in the US we isolated a 1.5 kb promoter fragment of the citrus seed-specific cDNA CssH. The identification of more fruit-specific cDNAs and their corresponding promoter regions is currently in progress.
