ZusammenfassungMicroRNAs (miRNAs) sind einzelsträngige, nichtkodierende RNA-Elemente, die die Umsetzung von Genen zu Proteinen über die Transkription und Translation beeinflussen können. Durch ihre Bindung an mRNA greifen miRNAs als Regulatoren in die verschiedensten Signalwege im Organismus ein. Dabei beeinflussen miRNAs unterschiedlichste biologische Prozesse, darunter Zelldifferenzierung, Proliferation oder Organ-/Gewe-beentwicklung und sind auch in der Knochenhomöostase wichtig. Sie spielen eine Rolle bei Knochenformation, -resorption, -remodelling und Knochenzelldifferenzierung. Damit sind unterschiedliche Expressionen von miRNA ein wichtiger Pathomechanismus bei osteologischen Erkrankungen wie der Osteoporose, der renalen Osteodystrophie oder von Knochenmalignomen. In dieser Übersichts arbeit werden miRNAs definiert und charakterisiert sowie deren Rollen in der Osteologie und bei osteologischen Erkrankungen dargestellt.

Abstract Cystine-dense peptides (CDPs) are a class of drug-like miniproteins that marry many of the advantages of biologics (high affinity and specificity) and small molecule therapeutics (high tissue permeability and low immunogenicity). The beneficial properties of CDPs, and miniproteins in general, have driven interest in therapeutic applications. However, CDP diversity is vast from every clade of life, and properly interrogating “CDP space” requires specialized screening and modeling tools. With this in mind, we have created an optimized mammalian surface display platform to screen for CDPs of clinical interest using libraries of structurally-diverse native scaffolds optimized for stability. These native CDPs can be structurally modeled, which we did in determining the structures of over 4200 native CDPs. This modeling permits further selection in silico as well as targeted mutagenesis for favorable target-binding capabilities. Hits from these screens are routinely matured to sub-nM affinity. These CDPs can play numerous roles in a drug design pipeline, from an independent drug candidate to a delivery agent for tissue-targeting to a module in a polyspecific biologic. Recent novel CDP candidates have shown promise in immune-oncology space as part of a bispecific T-cell engager targeting PD-L1, where a single 2-week treatment was capable of eliminating subcutaneous PC3 prostate cancer xenograft tumors in 27/30 mice. Besides bispecifics, future directions for the platform include exploring targeted protein degradation. Additionally, we are expanding upon our previous work on CDPs to explore CNS or tumor delivery of therapeutic cargo. The versatility of CDPs and novel screening tools to rapidly identify and mature candidates of interest can facilitate rapid advancement of CDP therapeutics to address difficult targets in oncology. With the current global urge and mandates for gender equality, the African continent is re-awakening interest on gender issues and gender mainstreaming in public policy and planning. This paper addresses issues and prospects in embracing Gender Equality and Women Empowerment (GEWE) principles in a democratizing state such as Nigeria. Basic questions raised include – what are the ideals of GEWE? What relevance does the GEWE Agenda have for a democratising state? What different legislations, policies, and programmes are strategic to this process? What roles have been played by Civil Society Organizations in instituting a GEWE agenda in a nation state such as Nigeria? International and transnational commitments to gender equality require strategies that tackle root causes and prevailing attitudes that perpetuate disparities. In this article, we examine the role and impact of international development volunteers (IDV) as development actors who are well-placed for feminist transformational change, as they work in transnational spaces to influence, support, or reinforce changes in attitudes and behaviors towards gender equality and women’s empowerment (GEWE). This qualitative study analyses data collected from 45 interviews in three countries (Malawi, Kenya and Uganda) to document partner organization perspectives on relational dynamics emerging from interactions with IDVs. Partner organization staff highlighted several notable positive and negative contributions to GEWE outcomes arising from day-to-day interactions with IDVs. These interactions shaped their understandings of GEWE, enhanced confidence for GEWE programming, and provided exposure to role models who can shape alternative attitudes and behaviors to gender equality. While the study revealed varying degrees of challenges and benefits for partner organizations working with volunteers specifically on gender equality, partner organization staff highlighted contributions made by IDVs to transnational spatial relations, as well as the transformational interactions that shaped these relations. Insights provided by partner country staff members offer subaltern perspectives and rich insights into the contributions of IDVs in gender equality programming and shed new light on the challenges and opportunities for fostering transnational feminist spaces of knowledge sharing, relationship building, and alternative practices.

Abstract Background Effective measurement of Gender Equality and Women’s Empowerment (GEWE) is challenging in low and middle-income countries (LMICs), and even more so in humanitarian settings. Conflict, natural disasters, and epidemics may increase gender inequities, but also present an opportunity to address them. This scoping review describes and identifies gaps in the measurement tools, methods, and indicators used to measure GEWE in humanitarian settings, and presents a dashboard that can be used by researchers, organizations and governments to identify GEWE measurement tools. Methods Scientific articles published between January 2004 and November 2019 were identified using Embase, Medline, PsycInfo, CINAHL, Scopus, and PAIS index. Relevant non peer-reviewed literature was downloaded from the websites of humanitarian organizations. Publications on women and/or girls impacted by a humanitarian crisis in a LMIC, within 5 years of data collection, were included. Publications were double-screened in the title/abstract and full-text stages. We used a machine learning software during the title/abstract screening to increase the efficiency of the process. Measurement tools, sampling and data collection methods, gap areas (geographical, topical and contextual), and indicators were catalogued for easy access in an interactive Tableau dashboard. Results Our search yielded 27,197 publications and 2396 non peer-reviewed literature reports. One hundred and seventy publications were included in the final review. Extracted indicators were categorized into seven domains: economic, health, human development, leadership, psychological, security and justice, and sociocultural. The vast majority of studies were observational, and over 70% utilized a cross-sectional study design. Thirty-eight toolkits and questionnaires were identified in this review, of which 19 (50%) were designed specifically for humanitarian settings. Sociocultural was the largest domain in number of studies and indicators in this review, with gender-based violence indicators reported in 66% of studies. Indicators of economic, human development and leadership were uncommon in the peer-reviewed literature. Discussion While there has been some effort to measure GEWE in conflict-affected and other humanitarian settings, measurement has largely focused on violence and security issues. A more comprehensive framework for measuring GEWE in these settings is needed; objective measurement of women’s empowerment and gender equality should be prioritized by organizations providing humanitarian aid.

The paper examines triumphs, challenges, and paradigmatic shifts in terms of whether the African Union agenda 2063 has translated to any tangible change in Kenya’s quest for gender equality and development. The institutionalization of gender issues at different levels globally demonstrates the primacy of gender parity in development paradigms. AU’s agenda 2063 aspiration 6 explicitly talks of gender equality and development and provides Gender Equality and Women Empowerment (GEWE) strategy which looks toward women’s inclusion in Africa’s development. The Union outlines strategic pillars for implementation by member states. This paper provides a critical analysis of the integration of these pillars in Kenya’s development agenda. The study relied mainly on secondary data from journal articles, relevant documents from Kenya’s Gender Commission and Ministry, relevant legal instruments, and websites. It shows that no country has fully achieved gender equality but can only be categorized as either friendly or unfriendly towards it; Kenya has attained quite substantial strides in the integration of the GEWE strategy pillars; systemic and structural factors are the biggest challenges to the full realization of aspiration 6 in Kenya; quantity doesn’t necessarily translate to quality with reference to the presence of women in key institutions and positions of influence. Conclusively, the paper indicates the importance of feminization of development in Kenya; emphasizes the agency of both genders in the implementation of aspiration 6, and notes that society has failed gender equality. İşlevi, bir hücrenin veya organizmanın hayatta kalabilmesi veya üreme başarısı için vazgeçilmez olan genler, esaslı genler olarak kabul edilir. Esaslı genleri esaslı olmayanlardan ayırt etmek, bir organizmanın minimum fonksiyonel gereksinimlerinin anlaşılabilmesi için genetikte kilit bir sorudur. Esaslı genler küme bilgisi, ilaç tasarlanmasında da çok önemlidir. Literatürdeki, bir protein-protein etkileşim ağındaki gen konumunun, gen esaslılığı ile ilişkili olduğunu göstermiştir. Burada, bir protein-protein etkileşimi (PPI) ağının düğüm yerleştirmelerinin gen gerekliliğini tahmin etmeye yardımcı olup olamayacağını soruyoruz. İnsan geninin esaslığını tahmin etme konusundaki sonuçlarımız, düğüm gömülümlerinin tek başına %88'e kadar AUC skoruna ulaşabileceğini göstermektedir. Bu skor, gen özelliklerini karakterize etmek için topolojik özellikleri kullanılan modellerin başarımından ve önceki çalışma sonuçlarından daha iyidir. Ayrıca, türler arası homoloji bilgisi ile birleştiğinde, bu performansın %89 AUC skoruna ulaştığını gösteriyoruz.
